Opis:
In this contribution we deal with a new mathematical description of the response of short-term coleoptile/hypocotyl expansion growth to temperature. Although the interest of both the bio-mechanical basis of elongation growth and temperature responses has been studied in plant biology and biophysics for a long time, yet the question of the mode of actions of temperature is very relevant and still open. Here we introduce a simple idea that the normal distribution, due to the central limit theorem (CLT), is able to report on temperature-dependent elongation growth. The numerical fittings for temperature affected growth are in good agreement with empirical data. We suggest that the observation concerning a crossover effect occurring in temperature driven elongation together with CLT leads to the formulation of a hypothesis about the possible universal character of such a description, supposedly for many plant species and families. We conclude with the statement that properly constructed equations of temperature affected growth, should possibly include a specific term proportional to exp[-((T-T0)/T0)2] with T0 corresponding to the temperature of the optimum growth.

Opis:
The epidermis of Helianthus hypocotyl can be peeled off and, in the form of detached strips can be used as a model system to study the effect on cortical microtubule (cMTs) orientation of these factors, which are difficult to be manipulated in situ, such as apoplastic pH or applied stress. In the first step, however, the orientation and reorientation of cMTs in the epidermis in situ must be described. The cMTs under the epidermal wall in hypocotyl epidermis at different positions along the hypocotyl and on its opposite sides were studied by means of immunostaining, using epi-fluorescence microscopy. The angle λ that parallel array of cMTs makes with cell longitudinal axis was measured. The variation of λ in a population of cells was documented by λ-histogram (frequency of cells exhibiting a particular λą∆λ plotted against λ value). The histograms were of either transverse type (maximum at λ ~90o, denoted as type A) or oblique type (two maxima on both sides of the transverse direction, denoted as type B) in the apical part of the hypocotyl, and were either of B type or of longitudinal type (maximum at λ ~0o or 180o denoted as type C) in the basal part. The change from A or B to C basipetally may be considered as due to the developmental trend in cMT orientation. The occurrence of B above A in some hypocotyls in their apical part strengthens the hypothesis on the autonomous reorientation of cMTs. The intermingled occurrence of A and B reorientation in the upper part of hypocotyl is interpreted as a manifestation of a subtle control of cell growth in latitudinal direction. The majority of histograms were asymmetric showing predominance of cMT parallel arrays inclined as the middle part of the letter Z.

Opis:
Light and developmental processes affect the cortical microtubule (cMT) orientation. The cMT orientation with a special regard to its chirality was analyzed under the outer epidermal cell walls in different regions of sunflower hypocotyls kept in darkness and after irradiation with blue and red light. The results show that the cMT orientation depends on the cell position along hypocotyl, but generally cMTs are oblique. The oblique orientation has defined chirality: either of Z-form (right-handed) or S-form (left-handed). In the lower region of hypocotyls the Z-form dominates. After irradiation of hypocotyls with blue light this domination has been maintained and appeared also in the upper region. In contrast, after irradiation with red light the Z-form domination has not been apparent. It is proposed that in darkness, variations of cMT orientations in the epidermis along the hypocotyl are due to developmental processes, while blue and red light affect the cMT orientation via "shifting" these processes backward and forward, respectively.

The seedlings of the red pepper (Capsicum annuum L.) cv. Trapez grown in water culture for a period of 14 days with A1 (0, 10, 20 and 40 mg·dm⁻³ AlCl₃·6 H₂O). Some morphological and anatomical features of red pepper shoots were analyzed. Reduction in height and diameter of stems as well as decrease in fresh mass of shoots were observed after Al-treatment. In the hypocotyl the thickness of cortex parenchyma layer and the size of their cells were reduced. The aluminum treatment resulted in the increased in thickness of the epidermis outer cell wall. Under Al stress in the cotrex and the central cylinder parenchyma cells were present numerous enlarge plastids which contained large grains of starch and dark little bodies which were possible aluminum deposits. They weren't observed in control seedlings.

Opis:
We investigated direct and indirect formation of somatic embryogenesis in Brassica oleracea var. botrytis (cauliflower), a very important vegetable crop worldwide. Direct somatic embryogenesis, which is rather rare, was achieved in culture of 2-week-old hypocotyl explants of Brassica oleracea var. botrytis on MS medium supplemented with 1.0 mg/l 2,4-dichlorophenoxyacetic acid (2,4-D) and 0.5; 1.0; and 1.5 mg/l kinetin. Initial induction of embryogenic callus was achieved on MS supplemented with very low concentrations of 2,4-D (0.05 mg/l and 0.1 mg/l). Indirect somatic embryogenesis from leaf sections was obtained on MS supplemented with 0.05 or 0.1 mg/l 2,4-D. We examined various stages of somatic embryos (globular, heart, torpedo, cotyledonary). More embryos per explant were produced through the indirect pathway (23–25) than through the direct pathway (14–19). The number of embryos produced was high. There is a potential for recurrent, repeated or secondary somatic embryogenesis, possibly an unlimited source for mass propagation and ideal for synthetic seed production in this species. Plant regeneration was achieved on half-strength MS medium without any hormones.

Opis:
The effect of methyl jasmonate (MJ) vapors on content of phenolic compounds: free phenolic acids, total quercetin, and total phenolics in etiolated buckwheat seedlings were studied. The data presented show that low concentration of MJ (10-8 M) had no influence on trans-cinnamic acid (CA), but stimulated the accumulation of chlorogenic acid in hypocotyls and cotyledons of buckwheat seedlings. A moderate dose of MJ (10-6 M) did not change the level of chlorogenic acid in the hypocotyls and cotyledons, but CA synthesis was promoted in cotyledons, whereas in hypocotyls no significant effect was found. Highest concentration of MJ (10-4 M) caused small decline of CA in hypocotyls, but large stimulation of the acid production in cotyledons was noted. MJ had stimulatory effect on caffeic acid forming, but inhibited synthesis of vanillic acid in hypocotyls and cotyledons. Lowest concentration of MJ (10-8 M) elicited accumulation of quercetin glycosides in both studied tissues of buckwheat seedlings, however at higher doses (10-6 and 10-4 M) did not affect the flavonol level. The obtained results suggest that nonequivalent influence of methyl jasmonate on the phenolics composition can be a result of various mechanisms of MJ uptake, transforming and/or its translocation in buckwheat hypocotyls and cotyledons. Decline of anthocyanins level in buckwheat hypocotyls caused by MJ cannot be explained by enhanced accumulation of quercetin glycosides or free phenolic acids, but probably by synthesis of other unknown phenolic compounds.

Six seedling hypocotyl anatomical characters of sugar beet diploid lines and triploid hybrids were measured. Root yield and sugar content of these lines and hybrids were evaluated in replicated field trials. Some of the studied hypocotyl characters: the diameter of the central core, the diameter of parenchymatic cells outside the central core and the width of xylem, correlated negatively with sugar content and positively with root yield. This suggests that these parameters can be used in preliminary selection of sugar-beet breeding material. Introducing such criteria into the breeding process could speed up the selection and reduce the number of expensive field trials.
