Opis:
Oxidative stress, i.e., excessive production of oxygen free radicals and reactive oxygen species, leads to lipid peroxidation and to formation of reactive aldehydes which act as second messengers of free radicals. It has previously been shown that oxidative stress may be involved in the transcriptional regulation of cytomegalovirus (CMV) immediate early promoter, involved in viral reactivation from latency. In the current study we used a plasmid containing the yellow fluorescent protein (YFP) gene under the control of CMV-1 promoter to monitor the influence of hydrogen peroxide and reactive aldehydes, 4-hydroxy-2-nonenal (HNE) and acrolein, on CMV-1 promoter activation in human embryonic kidney cells (HEK293). While acrolein was ineffective, hydrogen peroxide slightly (50 %) stimulated the CMV promoter. In contrast, HNE had a strong, up to 3-fold, enhancing effect on the CMV-1 promoter within four as well as after 24h of treatment. The most effective was the treatment with 24 µM HNE. This effect of HNE suggests that stressful conditions associated with lipid peroxidation could lead to CMV activation.

Opis:
In the present study we have identified hydroxy-plastochromanol in plants for the first time. This compound was found both in low light and high light-grown Arabidopsis plants, however, under high light stress its level was considerably increased. Hydroxy-plastochromanol accumulated also during ageing of leaves of low light-grown plants, similarly as in the case of other prenyllipids. Our results indicate that hydroxy-plastochromanol found in leaves is probably formed as a result of plastochromanol oxidation by singlet oxygen generated in photosystem II during photosynthesis. These data also support the hypothesis that plastochromanol is an efficient antioxidant in vivo, similarly as tocopherols and plastoquinol.

Introduction/aim: The aims of this study were to develop a population pharmacokinetic (PPK) model for the clearance of 25-hydroxy vitamin D in non-elderly postmenopausal women and to identify the factors which have a significant influence on its clearance. Methods: The study population consisted of postmenopausal women who had been referred for evaluation of bone mineral density (BMD) by DEXA (dual-energy x-ray absorptiometry) scanner. The population pharmacokinetics modelling was conducted using the ADVAN 1 subroutine from a non-linear mixed effects (NONMEM) program, and thirty-two covariates were assessed. Results: A total of 75 serum concentrations were obtained from the same number of postmenopausal women and used for PPK analysis. The mean values of the participants’ age was 57.92±3.93 years and their body weight was 69.76±11.49kg. A wide range of 25-hydroxy vitamin D concentrations was observed (from 3.41 to 61.92ng/mL) with a mean value of 26.19±10.95ng/mL. A total of 32 covariates were examined and preliminary results suggested the influence of six covariates on 25-hydroxy vitamin D clearance. In the final PPK model however, only one covariate was shown to have a significant impact on the clearance value – the mean daily dietary intake dose of vitamin D (DD). Conclusions: These findings offer a preliminary basis on which to determine the level of vitamin D supplementation required by individual postmenopausal women. It could prove particularly important in achieving optimal serum levels of vitamin D in this vulnerable population.

The method has been successfully applied to synthetic and commercial samples.

Opis:
Five divalent transition metals Cu(II), Co(II), Ni(II), Mn(II) and Zn(II) complexes have been synthesized using 3-hydroxy-4-[N-(2-hydroxynaphthylidene)-amino]-naphthalene-1-sulfonic acid (H3L) Schiff base as a ligand derived from the condensation reaction between 4-amino-3-hydroxynaphthalene-1-sulfonic acid and 2-hydroxy-1-naphthalde-hyde. The synthesized complexes were characterized using microanalytical, conductivity, FTIR, electronic, magnetic, ESR, thermal, and SEM studies. The microanalytical values revealed that the metal-to-ligand stoichiometry is 1:1 with molecular formula [M2+(NaL)(H2O)x].nH2O (where x = 3 for all metal ions except of Zn(II) equal x = 1; n = 4, 10, 7, 4, and 6 for Cu(II), Co(II), Ni(II), Mn(II) and Zn(II), respectively). The molar conductivity result indicates that all these complexes are neutral in nature with non-electrolytic behavior. Dependently on the magnetic, electronic, and ESR spectral data, octahedral geometry is proposed for all the complexes except to zinc(II) complex is tetrahedral. Thermal assignments of the synthesized complexes indicates the coordinated and lattice water molecules are present in the complexes. SEM micrographs of the synthesized complexes have a different surface morphologies. The antimicrobial activity data show that metal complexes are more potent than the parent ligand.

Chitosan (Ch) is an attractive biopolymer with multiple reactive groups. However it is poorly soluble at neutral pH. Quaternization improves its solubility and permits the development of various positively charged drug delivery systems. The aim of this work was to study the solubility, toxicity, cell binding, and penetration of 20 kDa chitosan with 9, 40, 58 and 98% of quaternary ammonium group substitution (ChQ1 to ChQ4 accordingly). We showed that ChQ with substitution degree >40% was soluble in a wide pH range. Unexpectedly ChQ2 and ChQ3 were more toxic to cells than Ch, ChQ1 and ChQ4. Higher toxicity of ChQ was found against macrophage like cell line RAW264.7 than against epithelial cells MiaPaCa-2. All ChQ, in contrast to unmodified Ch, easily bound and penetrated the cells with the highest uptake by ChQ4. Thus, quaternized chitosan derivatives can be used for biomedical applications.

About nine (E)-(2,5-dinitrophenyl)-2-(substituted benzylidene)hydrazines have been synthesized by hydroxy apatite catalyzed condensation of 2,4-dinitrophenyl hydrazine and substituted benzaldehydes under microwave irradiation conditions. The obtained yields of these hydrazines are more than 85%. The purities of synthesized hydrazines are examined by the analytical and spectroscopic data reported earlier. The infrared spectral C=N, NH frequencies (v, cm-1) and the NMR spectral chemical shifts of CH, NH protons and carbons are assigned and are correlated with Hammett substituent constants, F and R parameter using single and multi-regression analysis. From the results of statistical analysis the effects of substituent on the group frequencies have been discussed. The antioxidant activities of these synthesized hydrazines were measured using DPPH radical scavenging method. The hydroxy- and methoxy- substituted hydrazines shows significant antioxidant activity.
