The digestive tract is nowadays conceived as a barrier organ constituted by a mucosal membrane separating the gut lumen from the inner milieu. The gut lumen is laden by a myriad of antigens brought about by the diet, but also pertaining to the overwhelming bacterial species of the gut microbiome. The mucosal cell population comprehends epithelial cells, and a variety of immune reactive cells. Of them, the mononuclear types effecting innate responses are endowed by membrane signaling receptors and, as a rule, are sensing the polysaccharides of bacterial cell walls; non-tolerated signals may then push the chain reaction on, to end in full activation of inflammation mediators. Acquired immunity is in turn mainly effected by T-cell types, some of them, behaving as autoreactive cells, may induce metastatic inflammation beyond bowel boundaries, partly explaining the so-called extra-intestinal manifestations of inflammatory bowel disease (IBD). The scenario is further complicated by the possible influence of epigenetic factors: diet, stress, smoking, drugs. Being IBD a low-penetrance disorder, for the full phenotype to develop, a critical mass of the above listed factors (typically, a disturbed membrane permeability, an immune stimulus, and an epigenetic factor) must occur. In the century since the full description of IBD, a variegated plethora of measures have been attempted. Some updated designs are now under scrutiny. Microbiota engineering, apoptosis modulation, and diet modification are just a few of the measures that we are arbitrarily describing here.

Opis:
Single cell morphotypes of the species Triadinium caudatum and Spirodinium equi, together with a representative of the genus Blepharocorys (Blepharocorys sp.) were used for phylogenetic analysis based on their 18S rRNA genes. Spirodinium equi clustered with sequences already described for the entodiniomorphs isolated from horses and the Blepharocorys sp. also grouped within the Entodiniomorphida clade, although both sequences were distinct from those described from rumen ciliates. Triadinium caudatum clustered within the Vestibuliferida, and most closely to that of Paraisotricha, only other member of this order which has been described in the horse. It was concluded that although members of the orders Entodiniomorphida and Vestibuliferida are present in the equine gut, and that they share an ancient linage with their rumen counterparts, they are ancestrally different groups.

The article discusses the effect of synbiotic therapy on the qualitative and quantitative changes in colonic microbiota in preschool children with community-acquired pneumonia. A total of 33 children aged from 1 to 3 years were included in the study. All patients were divided into two groups. The first group consisted of 17 children who received standard therapy. The second group consisted of 16 patients, whose treatment was supplemented with a synbiotic preparation at a dose of one sachet per day. The assessment of colonic microbiota was conducted in 33 patients with community-acquired pneumonia before treatment and on treatment days 12–14. The control group consisted of 20 healthy age and sex-matched children. Bacteriological study of gut microflora was conducted using the Epshtein-Lytvak method. The evaluation of the microflora composition changes in children suffering from community-acquired pneumonia showed an intensification of gut microflora imbalance after standard therapy. When studying the effectiveness of the synbiotic preparation in patients with community-acquired pneumonia, it was found that the state of colon microbiocenosis in patients who received complex therapy with synbiotic drug significantly improved compared to children treated with standard regimen. Thus, of the use of synbiotic therapy in patients with community-acquired pneumonia improves or fully restores gut microflora in some cases. This preparation can serve as a safe and convenient way to restore gut microflora balance and prevent the development of gastrointestinal pathologies.
W artykule omówiono wpływ terapii synbiotycznej na zmiany jakościowe i ilościowe w mikroflorze jelita grubego u dzieci w wieku przedszkolnym chorujących na pozaszpitalne zapalenie płuc. Badaniem objęto 33 dzieci w wieku od 1 do 3 lat. Pacjentów podzielono na dwie grupy. Pierwsza liczyła 17 dzieci otrzymujących standardowe leczenie. Do drugiej grupy włączono 16 pacjentów, u których leczenie uzupełniono podawaniem preparatu synbiotycznego w dawce jedna saszetka na dobę. Ocenę mikroflory jelita grubego u 33 pacjentów przeprowadzono przed rozpoczęciem leczenia oraz w dniach 12.–14. terapii. Do grupy kontrolnej włączono 20 zdrowych dzieci dobranych pod względem wieku i płci. Badanie bakteriologiczne mikroflory jelitowej przeprowadzono metodą Epshtein-Lytvak. Ocena zmian w składzie mikroflory u dzieci z pozaszpitalnym zapaleniem płuc wykazała nasilenie zaburzeń równowagi mikroflory jelitowej u dzieci otrzymujących standardowe leczenie. W odniesieniu do skuteczności preparatu synbiotycznego u pacjentów z pozaszpitalnym zapaleniem płuc wykazano istotną poprawę mikrobiocenozy jelitowej u dzieci otrzymujących kompleksowe leczenie uzupełnione terapią synbiotyczną w porównaniu z dziećmi leczonymi według standardowego schematu. Zatem zastosowanie terapii synbiotycznej u dzieci z pozaszpitalnym zapaleniem płuc poprawia lub w pełni przywraca prawidłową mikroflorę jelitową w pewnych przypadkach. Preparat ten można stosować jako bezpieczny i dogodny sposób przywracania równowagi mikroflory jelitowej oraz zapobiegania patologiom przewodu pokarmowego.

The binding of TMOF to its gut receptor was followed by incubating guts removed from male and female Aedes aegypti. TMOF at physiological concentrations, in the presence of [γ32P]ATP, causes phosphorylation and release of gut-membrane protein (45 kDa) that is further processed by proteolysis. In the presence of protease inhibitors only the 45 kDa protein was released. The phosphorylation and processing of the 45 kDa protein does not happen in the absence of TMOF. Both larvae and adult guts release the protein in the presence of TMOF. Male Ae. aegypti do not synthesize trypsin in their gut and do not release the 45 kDa protein in the presence of TMOF because a TMOF receptor is probably absent. Homogenized guts do not release the 45 kDa protein, indicating that the protease processing or the ecto-protein kinase activity is probably reduced after breaking the tissue. The 45 kDa phosphorylated protein can be dephosphorylated by alkaline phosphatase and protein phosphatase, indicating that the phosphate group is covalently linked to either a serine or a tyrosine moiety. This is the first report that shows that in insects, binding of a peptide hormone activates its receptor by proteolysis.

Crohn’s disease (CD) is a chronic, inflammatory illness of the digestive tract, characterized by alternating periods of remission and recurrence. The pathogenesis of CD is still unclear but probably is a result of a complex interaction between immunological, genetic and microbiological disorders. In recent years, there has been an increasing extent of evidence that gut microbiota plays a very important role in the pathogenesis of CD. Currently, the most effective treatment is biological therapy using anti-TNF monoclonal antibodies. It is interesting whether biological drugs resulting in fast remission, contributes to the normalization of the gut microbiota. Due to the fact that the children’s population is a significant percentage of all patients with CD, it is important to pay close attention to the problem of microbiological disorders in this age group. The aim of this study was to investigate whether there are quantitative changes of chosen bacteria species and fungi of the genus Candida in children with Crohn's disease relative to healthy children and assesment of quantitative changes in patients after biological treatment. In the group of children with Crohn’s disease, the numbers in Candida were significantly higher (9.74×1017 CFU/g) than in the control group (9.35×1010 CFU/g, p = 0.011). Biological therapy led to a significant reduction in the amount Candida (5.91×1011) and was comparable with the number in the control group. In the case of bacteria, we observed an increase in S. marcescens (3,4×108) in the patients group compared to the controls (1,85×108) and an increase in L. fermentum (2,34×1010) in relation to healthy children (3,31×108, p = 0,048) Biological treatment had an impact on the decrease in L. fermentum (4,76×109, p = 0.05).

Introduction and objective. Cardiovascular diseases are the leading cause of death in Europe and worldwide. One of the most important risk factors for atherosclerosis are lipid metabolism disorders, in particular hipercholesterolaemia. The aim of the study was to determine the correlation between gut microbiota composition and atherosclerosis risk factors, so in order that it might be used as a biomarker for coronary artery disease diagnosis. Material and method. The study involved middle-aged men in eastern Poland with central obesity (n=20), subjects with atherosclerosis (n=15) and those with no cardiovascular diseases (n=5). The gut microbiota composition was determined using tag-encoded 16S rRNA gene using Illuminal MiSeq. Data were analyzed with the use of t-test. Results. Firmicutes (49.26%) and Bacteroidetes (44.46%) were the dominant Phyla in the middle-aged men in eastern Poland. Subjects with improper levels of total cholesterol were enriched in Prevotella (p=0.03) and decreased level of Clostridium (p=0.02). They also showed a falling tendency in Faecalibacterium (p=0.07). An upward trend was observed in Prevotella (p=0.07) in subjects with improper LDL-C values. Conclusions. The study showed that intestinal microbiome is likely to play a role in the pathogenesis of atherosclerosis through its role in lipid metabolism. Bacterial genera of particular importance were Prevotella, Bacteroides, Clostridium, Faecalibacterium. However, further studies involving larger groups of subjects are required to confirm these observations.

Multiple sclerosis (MS) is a chronic inflammatory and neurodegenerative disease which affects the central nervous system. Currently, there are numerous disease-modifying therapies for this condition. Most of them address the inflammatory aspects of the disease and are most effective in the relapsing-remitting stages of multiple sclerosis. However, none of them can completely stop the progression of MS and they are usually associated with adverse effects. There is an ongoing search for novel approaches that involve different modes of action. Here, we discuss examples of new immunomodulating agents such as antigen-specific therapies, neuroprotectants, regenerative strategies and gut microbiota modification.

The human digestive system is colonized by a huge number of microorganisms, that are referred to collectively as the gut microbiota. The composition of intestinal microorganisms are shaped from an early life and undergoes constant changes depending on the influence of external factors, such as: type of delivery, feeding the young child, diet in subsequent years of life, pharmaceuticals use, stress, lifestyle or infections and previous inflammation within the digestive tract. Despite transient changes in microbiota composition, the intestinal ecosystem is constantly striving to maintain homeostasis, both qualitative and quantitative, which is fundamental to human health and human development. Microbes present in the intestines are responsible for sealing the intestinal barrier, mucin production, stimulation of the angiogenesis process, supporting digestive processes by fermentation and decomposition of undigested food residues, vitamin production or protection from pathogenic microorganisms. As shown by numerous studies carried out in recent years, intestinal dysbiosis plays a fundamental role in the development of many chronic diseases such as inflammatory bowel disease, diabetes, obesity, celiac disease, connective tissue diseases and others. Insightful understanding of the interactions between microorganisms and the host organisms can provide new information about pathogenesis of diseases as well as new ways to prevent and treat intestinal or systemic disorders. The aim of this work is to review the latest reports on the role of the gastrointestinal microbiome in selected chronic diseases.

The human microbiome is a complex collection of microorganisms, including their genes and the metabolites colonizing the human body, and playing various functions in health and disease. The arrival of culture-independent molecular techniques such as metagenomics, transcriptomics, proteomics, and metabolomics have removed the limitations imposed by culture-dependent techniques. These advanced techniques have also brought about some paradigm shifts in what is known about the structural and functional diversities of the human microbiome in health and disease. The dynamics of the human microbiome is implicated in a number of human gastrointestinal and non-gastrointestinal diseases. This makes it a contemporary issue in biological and medical sciences. Of interest, some applications have already emerged for the human microbiome. These include being the source of antimicrobial substances, faecal microbiome therapy, probiotics, prebiotics and phage therapy. Given that a number of factors can alter the host microbiome - such as environment, lifestyle, stage of life, occupation, mode of delivery, therapy and so on, there is a need for more human microbiome projects that will help to capture these diversities in various continents. Furthermore, for the full impact of the various applications (both potentials and current) of human microbiome to be felt, there is need for more studies that will fully elucidate their physiology in humans.

ObjectivesThe aim of this study was to observe the depression-like behavior changes of rats exposed to lead with or without probiotic intervention, and to investigate changes in the gut microbiota and fecal short-chain fatty acids (SCFAs) levels after lead exposure, and the possible functions of probiotics in this process.Material and MethodsAdult male Sprague Dawley rats were exposed to a 300 mg/l lead acetate solution for 24 weeks, with or without probiotic (freeze-dried powder containing Lactobacillus and Bifidobacterium: 6 billion live bacteria/2 g) intervention in weeks 17–24. The sucrose preference test (SPT), the forced swim test (FST), and the tail suspension test (TST) were preformed to study the depression-like behaviors of these rats. The alteration of rat gut microbiota induced by lead exposure was analyzed by 16S rRNA sequencing, and the levels of fecal SCFAs were detected using gas chromatography.ResultsNeurobehavioral tests showed that lead exposure induced depression-like behavior in rats, including reduced sucrose preference in the SPT, and increased immobility times in the FST and the TST. Sequencing and gas chromatography showed that lead exposure changed the structure and the phylogenetic diversity of the gut microbiota, as well as significantly altered the levels of SCFAs. Moreover, the depression-like behaviors, and the changes in both gut microbiota and SCFAs, could be mitigated by probiotic intervention.ConclusionsLead exposure not only changes the structure and diversity of the gut microbiome but also affects metabolic function. Już w trzecim roku życia była leczona, jako przetoka ropna pośladka prawego, która się okresowo uaktywniała. W 2016roku zgłosiła się do Poardni Chirurgicznej w Gorlicach z olbrzymim ropniakiem pośladka prawego z przetoką ropną . Na oddziale chirurgicznym w Gorlicach operacyjnie usunieto ropniak o ścianach dobrze ukształtowanych razem z przetoką. Torbiel zawierała ponad litr ropy. Po wygojeniu w bliźnie pojawiłą się przetoka śluzowa .Badaniem histologicznym rozpoznano retrocecal cyst hamartoma z komórkami mucinos adenocarcinoma. W czasie następonego pobytu chorej w szpitalu po wykonaniu rezonansu magnetycznego stwierdzono resztkowa zmianę w okolicy przedkrzyżowej sięgająca do trzeciego kręgu krzyżowego. Zmianę usunięto radykalnie potwierdzoną badaniem histopatologicznym. U chorej nawrotu nie zaobserwowano.

Opis:
The fruit processing industry generates huge amounts of waste annually, causing severe environmental problems. Mango processing produces around 20 million tons of wastes from the non-consumed fraction, mainly peels and seeds, which constitute 30–60% of the fruit weight. However, various phytochemicals found in these residues have been implicated in preventing cancer and also cardio-metabolic and gastrointestinal diseases. Particularly, phenolic compounds hold a promising potential to be utilized as modulatory agents of the human gut microbiota (prebiotics-like actions). This review article mainly discusses the effect of phenolics from mango wastes on gut microbiota modulation and its beneficial repercussions for human health. Moreover, it also discusses the importance of phenolic compounds of mango peel and seed kernel residues, their extraction, identification, and quantification.
