Introduction. Liver cirrhosis is a chronic disease in which progressive fibrosis is noted. This process leads to changed architectonics of the liver parenchyma and the appearance of regenerative nodules, all of which are caused by pathological activation of the hepatic stellate cells. This process is enhanced on a molecular level by many cytokines, with platelet-derived growth factors (PDGFs) playing the key role. Objective. The aim of the study was to assess serum concentrations of PDGFs active biodymers (PDGF-AA, PDGF-BB and PDGF-AB) in patients with alcoholic liver cirrhosis, and to correlate them with the stage of disease. Materials and method. 64 patients with alcoholic cirrhosis and a control group of 16 healthy individuals were analysed. Liver cirrhosis was determined based on clinical image, history of the patients’ alcohol consumption, laboratory findings and abdominal ultrasonography. The serum PDGF-AA, PDGF-BB and PDGF-AB concentrations were determined using ELISA kits. Results. Serum concentration of PDGF-AA and PDGF-BB homodimers increases in patients with alcoholic liver cirrhosis (p=0.034 and p<0.0001, respectively), unlike the serum concentration of PDGF-AB heterodimer (p>0.05). When the stage of the disease increases, the concentrations of PDGF-AA and PGFD-BB in blood also oncrease. Furthermore, the serum level of both PDGF-AA and PDGF-BB correlates significantly with the severity of alcoholic liver cirrhosis (measured by Pugh-Child’s scale), the correlation being stronger in the case of PDGF-BB levels than PDGF-AA (R=0.28; p=0.027 and R=0.26; p=0.038, respectively). Conclusions. The plasma levels of PDGF-AA and -BB may be indicators of alcohol-induced liver fibrosis process, and might be considered as future possible treatment targets, with PDGF-BB levels being an even better indicator than PDGF-AA levels.

Our earlier paper has reported that Wharton's jelly is a reservoir of several peptide growth factors, including acidic and basic fibroblast growth factors (aFGF and bFGF, respectively). Both can be extracted by buffered salts solutions in the form of high molecular mass complexes, probably with a component(s) of the extracellular matrix. Both aFGF and bFGF from such extracts hardly penetrate 10% polyacrylamide gels during electrophoresis. Pre-treatment of Wharton's jelly with hyaluronidase slightly increased the extractability of aFGF, but did not affect the extractability of bFGF. In contrast, the pre-treatment of tissue homogenate with bacterial collagenase (2000 U/ml, 37°C, 18 h) increased the extractability of bFGF. The presence of β-mercaptoethanol in the extracting solutions increased the extractability of both FGFs, but did not release FGFs in their free form, despite reducing the molecular mass of the FGF-containing complexes. We conclude that both aFGF and bFGF are bound through disulphide bonds to a protein component of Wharton's jelly. We propose that ground substance composed mainly of collagen fibrils and hyaluronate molecules, which surrounds the cells of Wharton's jelly, prevents the access of the extracting solution to aFGF and bFGF. Although hyaluronate and collagen do not bind aFGF or bFGF directly, they may constitute a barrier which prevents the dispersion of FGFs in Wharton's jelly. Thus, the high concentration of FGFs around the cells of Wharton's jelly may facilitate the interaction of these factors with membrane receptors, thereby resulting in stimulation of cell division and differentiation, as well as of the synthesis of extracellular matrix components.

Both epidermal growth factor (EGF) and transforming growth factor (TGF) play an important physiological role in the processes of proliferation and differentiation of several different cell types. However, the expression profiles of these factors in domestic bitches endometrium are still poorly recognized. The aim of the present study was to identify and analyze the differential expression of these factors in various stages of the estrus cycle. Endometrial tissue from proestrus (n=17), estrus (n=10), day 10 diestrus (n=15), day 35 diestrus (n=18) and anestrus (n=25) was collected soon after ovariohysterectomy. Total RNA was isolated from the endometrium by means of Chomczyński and Sacchi method, treated by DNase I, and reverse- transcribed into cDNA. Quantitative analysis of EGF, TGFβ1, TGFβ2, and TGFβ3 cDNA was performed by real-time quantitative polymerase chain reaction (RT-PCR). EGF expression in canine endometrium was increased in the estrus stage as compared to proestrus (P<0.05), day 10 diestrus (P<0.05), day 35 diestrus (P<0.01) and anestrus (P<0.001). We also found the differences in EGF expression between day 10 and day 35 of estrus as well as between day 35 of estrus with anestrus (P<0.05, P<0.01, respectively). The TGFf1 transcript contents were also higher in estrus as compared to other stages (P<0.01). The TGFβ2 and TGFβ3 in the estrus stage was increased compared to proestrus, day 10 diestrus, day 35 diestrus and anestrus (P<0.05). We proved that expression of EGF and TGFβ transcript isoforms is related to the phase of estrus in bitches and therefore may be regulated by specific hormone concentrations during these periods. Our results confirm the hypothesis that these growth factors play a role in the regulation of biochemical changes in the endometrial tissues during the estrus cycle.

Introduction and objective. Vitamin D deficiency is common in obesity; however, its contribution in the development of metabolic complications remains uncertain. The aim of the study was to examine the relationships between vitamin D status and metabolic complications. Materials and method. The results of blood pressure measurements, biochemical tests and ultrasound of the liver were compared in both groups. The study was conducted at the Children’s University Hospital in Krakow, Poland. 30 obese adolescents (mean 13.23y.o.); 18 with 25OHD levels <20ng/mL, 12 with 25OHD>20 ng/mL. Results. The vitamin D deficient group presented with significantly higher values of the diastolic blood pressure (125.9vs.115mmHg), uric acid level (384.7vs.301.5umol/L) and lower phosphorus level (1.4vs.1.65mmol/L), higher prevalence of arterial hypertension (44vs.8.3%), and liver steatosis (25vs.8.3%); lower, but not significantly, levels of fibroblast growth factor 23 and fibroblast growth factor 19. Conclusions. Hypovitaminosis D in obese adolescents is associated with higher prevalence of arterial hypertension, liver steatosis, elevated serum uric acid and low phosphorus levels. The potential contribution of the fibroblast growth factor 23 and fibroblast growth factor 19 in these complications development needs further investigation.

Purpose: Multiple cellular functions are stimulated by a Insulin-like Growth Factor-I (IGF-I). The biological activity of IGF-I is modulated by IGFbinding proteins (IGF-BPs) and at the same time, the availability of IGF-BPs may be regulated by the proteolytic activity of some metalloproteinases (MMPs). The aim of the present study was to compare the amounts of IGF-I and IGF-BPs in relation to the activity of MMP-9 in serum and knee synovial fluid from patients with Lyme arthritis (LA) and post-traumatic damage (PTD). Material and methods: Serum and synovial fluids were taken from knee joints of patients with PTD and LA. ELISA (for IGF-I assay), polyacrylamine gel electrophoresis following Western immuneblotting (for IGF-I and IGF-BPs expression), and zymography (for metalloproteinases detection), were used. Results: The concentration of IGF-I in serum and synovial fluid from LA patients were significantly lowered in comparison to PTD patients. Interesting, the synovial fluid /serum ratio of IGF-I concentrations was also lower in LA patients. Low expression IGF-BP3 and high activity of MMP-9 were detected in the LA synovial fluid. Conclusions: The high proteolytic activity of MMP- 9 results in a cleavage of both IGF-I and IGF-BP3 causing a decrease in content of these substances in LA synovial fluid. In addition, the reduction in IGF and IGF-BP amounts may affect the repair processes in joint tissues of LA patients. The low concentration of IGF-I and IGF-BP3 slows down the repair processes in the joint tissues of LA patients.

The hydrazine derivatives of benzopyrones remain an unexplored group of chemical compounds. This preliminary study investigates the influence of A-5, CH-3 and K-2 derivatives at concentrations of 1, 10, 100 nM and 1 μM on selected biochemical factors of a melanoma cell line WM-115, with regard to their potential angiogenic properties. The studied compounds were found to influence cell proliferation, as well as total protein, bFGF and FGFR1 concentration.

Acquired drug resistance of tumor cells is frequently observed in cancer patients undergoing chemotherapy. We studied murine leukemia L1210 cells sensitive and resistant to the cytotoxic action of cisplatin and showed that cisplatin-resistant leukemia cells were also refractory to TGF β1-dependent growth inhibition and apoptosis. Addressing the question about the mechanisms responsible for the cross-resistance to cisplatin and TGF β1, we found that cisplatin- and TGF β1-resistant L1210 cells possessed a decreased expression of type I TGF β1 receptor, while the expression of type II TGF β1 receptor was not affected. Western blot analysis of Smad proteins 2, 3, 4, 6, and 7, which participate in signal transduction pathway down-stream of the TGF b1 receptors, revealed an increased expression of Smad 6, inhibiting TGF b1 action, only in cisplatin- and TGF β1-resistant L1210 cells. TGF β1 and especially the cytotoxic mistletoe agglutinin increased Smad 6 expression in TGF β1-sensitive but not in TGF β1-resistant L1210 cells. TGF β1-resistant L1210 cells also differed from TGF β1-sensitive cells by the lack of expression of the pro-apoptotic p53 protein and higher level of expression of the anti-apoptotic Bcl-2 protein. Thus, the described co-expression of tumor cell refractoriness to an anti-cancer drug and to the inhibitory cytokine TGF β1 is accompanied by multiple changes in the TGF β1 signal transduction pathway and in other regulatory systems of the target cells. Besides, we found that various anti-tumor drugs and cytotoxic plant lectins increased the level of TGF b1 expression in both TGF β1-sensitive and -resistant L1210 cells. A hypothesis is proposed that TGF β1 can at least partly mediate the effect of cell-stressing agents and, thus, the development of TGF β1 resistance may be responsible for the appearance of tumor cell refractoriness to the action of some anti-cancer drugs.

Mammary tumors in female dogs are usually malignant and tend to metastasize to distant organs, especially to regional lymph nodes and lungs. Radiography is the standard diagnostic method to detect pulmonary metastases in these animals. Magnesium (Mg), vascular endothelial growth factor (VEGF), and osteopontin (OPN) levels have been used in recent studies to make prognoses of human breast cancer. To the best of our knowledge, however, there are not many studies that have been performed on this subject, and there is no study on animals in which the three indicators are scrutinized together. The aim of this present study is to evaluate Mg, VEGF, and OPN levels in healthy dogs and in dogs with mammary tumors with/without pulmonary metastases, and to investigate the alterations of these parameters in the serum and tissue samples of dogs with mammary tumors in connection with the histological tumor type and tumor grade. Mammary tumor groups were designed according to the presence of pulmonary metastasis in radiography; group M1 consists of 20 dogs with metastatic mammary tumors to the lung, and group M0 consists of 20 dogs with nonmetastatic mammary tumors. Ten clinically healthy dogs composed group H. The dogs represented different breeds and ages. Threeview thoracic radiographs were taken to determine any metastasis in lungs by digital radiography. Magnesium, VEGF, and OPN were determined in the mammary gland samples and blood serum of 40 dogs with malignant mammary tumors and in 10 healthy dogs. The magnesium levels were measured by atomic absorption spectrophotometry, both in the tissue and serum samples. Also, the tissue and serum VEGF and OPN levels were determined by ELISA with commercially available kits. The tissue Mg levels in the M0 group were significantly (P<0.05) higher than in group H. However, the serum VEGF level was significantly associated with a tumor type. Additionally, the serum OPN levels exhibited a tendency to be higher in dogs with mammary tumors with pulmonary metastases, grade 3, and carcinosarcoma. It is concluded that Mg, VEGF, and OPN could have practical use for diagnosing and understanding the pathophysiology of CMT.

Two molecules - vascular endothelial growth factor involved in new vessels formation and survivin - antiapoptotic protein, reported to be associated with worse prognosis in various malignancies have been chosen for the study. Both are potential target for novel therapiesThe aim of the study was to determine the immunostaining of VEGF and survivin in gastric carcinoma and to analyse their relationship to the selected clinicopathological features and survival.Material and methods. Formalin-fixed, paraffin-embedded sections from 41 gastric adenocarcinomas were used for immunohistochemical reaction with monoclonal antibodies against vascular endothelial growth factor and survivin. The results were compared with selected clinicopathological features and survival.Results. Positive immunohistochemical reaction for vascular endothelial growth factor and survivin was revealed in 24 (58,53%) and 30 (73,17%), gastric carcinomas respectively. Vascular endothelial growth factor-negative gastric carcinomas were significantly more common in cases without metastases to regional lymph nodes and distant organs and in less advanced cases. Similar, distant metastases were also statistically less common in survivin-negative carcinomas. The differences in immunohistochemical reactions for survivin between less and more advanced cases almost reach statistical significance. The only factors significantly influenced 1, 2 and 3-year survival were vascular endothelial growth factor and survivin status. Statistically significant higher percentage of survival was noted in patients with vascular endothelial growth factor- and survivin-negative tumors.Conclusions. It seems that vascular endothelial growth factor and survivin play role in local invasion and spread of gastric adenocarcinoma and negatively influences survival. However, further studies are required to assess their true usefulness in the clinical practice.

The aim of this study was to evaluate the effects of steroidal saponin supplementation on blood concentration of T, GH and IGF-1. The research involved 14 men between the age of 45 and 60 years. The duration of the experiment was 12 weeks. There were two series of laboratory tests. Independent tests were conducted at the beginning and after 12 weeks of the intervention. A two-way repeated measures ANOVA revealed a statistically significant effect of the intervention on the following variables: T-Ch (η2 = 0.542), HDL-Ch (η2 = 0.522), LDL-Ch (η2 = 0.587), T (η2 = 0.603), IGF-1 (η2 = 0.512) and GH (η2 = 0.621). Thus, FFM significantly increased while TBF and BM decreased in comparison to pre-intervention levels. The analyzed results indicate that treatment or supplementation of individual hormone deficiencies can be a successful form of counteracting the aging process. Nevertheless, the effects of TT supplementation on the concentration of T as well as GH and IGF-1, requires further studies, especially in middle-aged and older subjects, along with different exercise programs. The analyzed results indicate that treatment or supplementation of individual hormone deficiencies can be a major form of counteracting the aging process.

The in vivo effects of some derivatives of aliphatic ketones (2-undecanone, 3-undecanone, 4-undecanone and their derivatives) on L-l sarcoma tumor angiogenesis and VEGF content were studied in Balb/c mice. Mice that inhaled 10% solution of 3-undecanone(3-on) or 1% solution of 2-undecanone propylene acetal (Acpr2) for 3 days after tumor cells implantation, presented lower neovascular response measured by tumor-induced cutaneous angiogenesis test (TIA) and lower tumor VEGF content in 5-days tumors, than non-inhaled controls. Other substances presented various effects on tumor VEGF concentration and angiogenesis. Histological examination of lesions collected from mice inhaled Acpr2, or non-inhaled controls, revealed small diffused areas of necrosis in the former group. In both groups, slight to moderate inflammatory infiltrations were seen at the tumor's margin. In Acpr2 group, there were less small blood vessels at tumor's margin than in the control group.

Opis:
Carbon monoxide (CO) is an odorless, tasteless and colorless gas which is generated by heme oxygenase enzymes (HOs). HOs degrade heme releasing equimolar amounts of CO, iron and biliverdin, which is subsequently reduced to bilirubin. CO shares many properties with nitric oxide (NO), an established cellular messenger. Both CO and NO are involved in neural transmission and modulation of blood vessel function, including their relaxation and inhibition of platelet aggregation. CO, like NO, binds to heme proteins, although CO binds only ferrous (FeII) heme, whereas NO binds both ferrous and ferric (FeIII). CO enhances the activity of guanylate cyclase although it is less potent than NO. In contrast, CO inhibits other heme proteins, such as catalase or cytochrome P450. The effects of CO on gene expression can be thus varied, depending on the cellular microenvironment and the metabolic pathway being influenced. In this review the regulation of gene expression by HO/CO in the cardiovascular system is discussed. Recent data, derived also from our studies, indicate that HO/CO are significant modulators of inflammatory reactions, influencing the underlying processes such as cell proliferation and production of cytokines and growth factors.

Introduction and objective. Liver cirrhosis is a disease involving the liver parenchyma, which is characterised by fibrosis. and impaired architectonics of the parenchyma with regenerative nodules. The aim of the study was to determine the relationship between stage of alcoholic liver cirrhosis, concentrations of selenium, zinc and profibrotic and proangiogenic cytokines (FGF-19, ENG). Materials and method. The study included 99 patients with alcoholic cirrhosis and 20 healthy subjects. Ion chromatography with UV/VIS detection was used for determination of zinc ions in the previously mineralized serum samples. The measurements of selenium were performed with the ContrAA700 high-resolution continuum source graphite tube atomic absorption spectrometer. ELISA was used to determine concentration of FGF-19 and ENG in serum samples. Results. Concentrations of zinc and selenium were significantly decreased in cirrhotic patients (p<0.001 for both). The highest concentration of FGF-19 was found in Child-Pugh stage C liver cirrhosis patients (806.9±650.3 pg/ml), and was significantly higher than observed in controls (p=0.005) and stage A patients (compensated cirrhosis) (p=0.02). The highest concentration of ENG was demonstrated in the control group (3.24±148 ng/ml) while the lowest in patients with decompensated cirrhosis (7.32±5.39 ng/ml and 7.92±4.18 ng/ml for stage B and C; p=0.03 and p=0.02, respectively). The use of the multiple-variable model demonstrated that the independent factors affecting the concentration of ENG were the concentration of bilirubin (p=0.02), INR (p=0.01) and duration of alcohol abuse (p=0.02). The independent determinants of FGF-19 concentrations were found to be the stage (severity) of liver cirrhosis (p=0.04) and INR (p=0.03). Conclusions. Concentrations of zinc and selenium in serum of patients with alcoholic liver cirrhosis are not independently related to concentrations of FGF-19 and ENG.

Aim: The aim of the current study was to assess the serum levels of insulin-like growth factor-1 (IGF-1) and ghrelin in hypertrophied adenoids in children suffering with or without otitis media with effusion before and after adenoidectomy. Material and methods: Serum IGF-1 and ghrelin concentrations were measured with specific enzyme-linked immunoassay (ELISA) methods. The study was carried out in 20 children with otitis media with effusion. The reference group comprised 24 children with hypertrophied adenoid, while control group included 19 children. Results: This mean values of IGF-1 in children with otitis media with effusion and children with hypertrophied adenoid before adenoidectomy were significantly lower than those found in healthy children. Serum levels of IGF-1 were higher after adenoidectomy. There was a significant difference of serum ghrelin levels between both examined groups and the control group. Conclusion: Our results suggest that adenoidectomy in children with hypertrophied adenoids and in children with otitis media with effusion significantly increases the level of IGF-1 in serum compared to before surgery through the effect of the GH-IGF-1 axis, which could contribute to children’s growth.

Introduction and aim. Angiogenesis, which is accomplished by capillary sprouting, is the process by which new vessels are created from pre-existing ones. In tumor, once their initial blood supply is depleted, a tumour is unable to grow without additional blood flow. Additionally, a tumor’s microvasculature, or microvessel density (MVD), increases along with its capacity to produce angiogenesis. We aimed to observe the relationship between the expression of vascular endothelial growth factor (VEGF) and MVD (using CD34) in oral squamous cell carcinoma (OSCC). Material and methods. The expression of VEGF and CD34 antibodies was analysed using immunohistochemistry method on 50 cases of histopathologically proved OSCC. The expression was correlated with clinicopathological parameters. Results. A significant correlation was observed between VEGF expression and gender, LVSI. No correlation between any other factors and the difference in VEGF expression was statistically significant. Similarly, the MVD expression was not found to be statistically significant in any of the pathological parameters. Conclusion. VEGF positivity as well as MVD were found to be independent of the tumor pathology. Tumor MVD was found to be independent of the expression of VEGF. Further studies in a larger study group may establish a significant association so that antiangiogenic targeted therapy may be initiated.

The process of wound healing consists of the following phases: inflammation, proliferation, remodeling. Non-steroidal antiinflammatory drugs may be important in this process, especially in a stage called angiogenesis. For this reason, it was decided to investigate the effect of selective COX-2 (cyclooxygenase 2) inhibitor (NS-398) on the proliferation of endothelial cells and their ability to secrete bFGF (fibroblast growth factor) for vascular endothelial cells (HMEC-1). For determination of the secretion of bFGF in a cell line HMEC-1 immunosorbent ELISA assays were used. In turn, the cell proliferation assay was performed using the MTT method. Using MTT method, it was found that NS-398 at 10 μM did not affect cell viability. Whereas selective COX-2 inhibitor at 100 μM decreased cell viability in a statistically significant manner and inhibited the proliferative effect of 100 μg/mL LPS at concentrations of 10 and 100 μM. In the further step, application of NS-398 (10 and 100 μM) with LPS (100 μg/mL; inflammatory environment) reduced the secretion of bFGF in a statistically significant manner. The investigations showed that NS-398 has an antiangiogenic effect which is based on reducing the proliferation of vascular endothelial cells and inhibiting the secretion of bFGF- factor responsible for angiogenesis during wound healing.

Heme oxygenase-1 (HO-1), an inducible enzyme degrading heme to biliverdin, iron and carbon monoxide, is involved in regulation of inflammation and angiogenesis. Tin protoporphyrin (SnPPIX) and zinc protoporphyrin (ZnPPIX) are commonly used as competitive inhibitors of HO-1. We aimed to compare the effects of SnPPIX and ZnPPIX on the production of vascular endothelial growth factor (VEGF), activity of inducible nitric oxide synthase (iNOS) and cell viability. All experiments were performed on rat vascular smooth muscle cells and murine RAW264.7 macrophages treated with 3-10 μM protoporphyrins. Some cells were additionally stimulated with IL-1β or with lipopolysaccharide. After a 24 h incubation period SnPPIX and ZnPPIX significantly reduced the generation of VEGF in vascular smooth muscle cells and RAW264.7, both in resting and stimulated cells. The inhibitory potentials of both protoporphyrins on VEGF synthesis were very similar. In contrast, analysis of iNOS activity revealed that results obtained with different HO-1 inhibitors are discrepant. Generation of nitric oxide by iNOS was significantly increased by SnPPIX but strongly decreased by ZnPPIX. Similar differences were observed when cell viability was compared. SnPPIX improved the cell survival rate, whereas the same doses of ZnPPIX exerted some cytotoxic effects. In summary, SnPPIX and ZnPPIX can be used as HO-1 inhibitors in some experimental models. However, these compounds produce also HO-independent effects, which can make the interpretation of experiments very uncertain. Thus the involvement of the HO-1 pathway should be always confirmed by more specific methods.

Nowadays non-alcoholic fatty liver disease (NAFLD) is becoming the most common chronic liver pathology both in adults and children. NAFLD manifestation ranges from a simple liver steatosis to steatohepatitis (nonalcoholic steatohepatitis - NASH), which may progress to advanced fibrosis, cirrhosis and end-stage liver disease. Due to the coexistence of visceral obesity, insulin resistance and dyslipidemia, NAFLD is considered to be the hepatic manifestation of metabolic syndrome. In recent years, in the pathogenesis of metabolic syndrome, type 2 diabetes mellitus, cardiovascular disease and also NAFLD, more and more attention has been paid to the so-called organokines, proteins with both paracrine or/and endocrine activities. These include most known adipokines (mainly produced by adipose tissue), myokines (mainly produced by skeletal muscles) and hepatokines exclusively or predominantly produced by the liver. It was shown that the liver may affect the lipids and glucose metabolism by hepatokines released into the blood and NAFLD seems to be associated with altered hepatokines production. Fetuin-A, fibroblast growth factor-21 (FGF-21), selenoprotein P, sex hormone-binding globulin (SHBG), angiopoietin-related growth factor (also known as angiopoietin-related protein 6) and leukocyte derived chemotaxin 2 (LECT2) are considered as the most important hepatokines. In this review, we provide an overview of the main hepatokines and we summarize the association of liver-derived proteins with the development and progression of NAFLD.

Pre-eclampsia, the most common pregnancy associated syndrome, is connected with remodelling of extracellular matrix of the umbilical cord tissues. Since the fibroblast growth factor (FGF) is known to be a stimulator of collagen and glycosaminoglycan biosynthesis, one may expect that it plays an important role in such a remodelling. Studies performed on the umbilical cords of 10 control and 10 pre-eclamptic newborns demonstrated that both the umbilical cord arterial wall and Wharton's jelly contain FGF mainly in complexes with the components of different molecular mass. Pre-eclampsia is associated with a decrease of endogenous FGF-binding by soluble high molecular mass components of the umbilical cord. It is suggested that FGF released from these complexes may be actively bound by fibroblasts of the umbilical cord, stimulating them to produce collagen and sulphated glycosaminoglycans.

The trophic effect of continuous intraventricular infusion of nerve growth factor (NGF) on morphology of the basal forebrain (BF) cholinergic neurons was tested in 4- and 28-month-old male Wistar rats. All studies were conducted using behaviorally uncharacterized animals from the same breeding colony. Immunohistochemical procedure for choline acetyltransferase (ChAT) and p75NTR receptor has been applied to identify cholinergic cells in the structures of basal forebrain (BF). Using a quantitative image analyzer, morphometric and densitometric parameters of ChAT- and p75NTR-positive cells were measured immediately after cessation of NGF infusion. In 28-month-old non-treated rats the number of intensively ChAT-positive cells in all forebrain structures was reduced by 50-70% as compared with young animals. The remaining ChAT-positive cells appeared shrunken and the neuropil staining was markedly reduced. In contrast, the same neurons when stained for p75NTR were numerous and distinctly visible with perfect morphology. Analysis of Nissl stained sections also showed that 28-month-old rats did not display significant losses of neuronal cell bodies. NGF restored the number of intensely stained ChAT-positive cells to about 90% of that for young controls and caused a significant increase in size of those cells in 28-month-old rats as compared with the control, age-matched group. NGF did not influence the morphology of p75NTR-positive neurons, which were well labeled, irrespective of treatment and age of the rats. In 4-month-old rats, NGF infusion decreased the intensity of both ChAT and p75NTR immunostaining. These data provide some evidence for preservation of BF cholinergic neurons from atrophy during aging and indicate that senile impairment of the cholinergic system in rats concerns decrease in ChAT-protein expression rather than an acute degeneration of neuronal cell bodies. Treatment with NGF resulted in restoration of cholinergic phenotype in the BF neurons of aged rats. However, the present study also rises issue of possible detrimental effects of NGF in young normal animals.

Research was carried out on 432 Polish Lowland Sheep (405 ♀ and 27♂) of three varieties: Corriedale, Żelaźnieńska Sheep and Polish Lowland Sheep of Podlasie region. All animals were subjected to gene identifi cation factor insulin – IGF-1, in the assessment of alleles C and T. In conclusion it should be noted that in the three examined varieties of Polish Lowland Sheep showed no polymorphism in exon 3 of the insulin-like growth factor (IGF-1) gene, limiting its scope to determine the allele C, respectively genotype CC. This result indicates the need for further research in this area in “culture” sheep imported and adapted to Polish conditions and the production environment.
Polimorfizm genu czynnika insulinopodobnego IGF-1 u polskich owiec nizinnych utrzymywanych w województwie podlaskim. Badania przeprowadzono na materiale 432 polskich owiec nizinnych (405 ♀ i 27♂) trzech odmian: corriedale, owcy żelaźnieńskiej i polskiej owcy nizinnej regionu podlaskiego. Wszystkie zwierzęta były poddane identyfikacji genu czynnika insulinopodobnego IGF-1, w zakresie oceny występowania alleli C i T. Podsumowując, należy stwierdzić, iż u badanych trzech odmian polskich owiec nizinnych nie wykazano polimorfizmu występowania alleli i genotypów genu czynnika insulinopodobnego IGF-1, ograniczając jego zakres do ustalenie jedynie do allelu C i genotypu CC. Wynik ten wskazuje na potrzeby przeprowadzenia dalszych badań z tego zakresu u owiec kulturowych pochodzących z importu i adaptowanych do polskich warunkach środowiska produkcyjnego.

Introduction: Tyrosine kinase inhibitors (TKI) are the standard of treatment in patients with advanced non-small cell lung cancer (NSCLC) with EGFR (endothelial growth factor receptor) gene activating mutation. Objective: The evaluation of the efficacy and toxicity of TKI drugs in NSCLC patients treated in single centre. Material and methods: NSCLC patients treated with TKI (gefitinib, erlotynib, afatinib) between 2012– 2016 were retrospectively analysed. We evaluated: overall response rate (ORR) which is the sum of complete responses (CR) and partial remissions (PR), progression free survival (PFS), overall survival (OS) and adverse events (AE) according to CTCAE (Common Terminology Criteria for Adverse Events) scale. Results: The study group were 16 patients ORR was 50% (CR: 1, PR: 7). Median PFS and OS was 8,7 and 22,9 months respectively. Adverse events observed mainly in stage 1 and 2 were related to hyponatraemia, hyperbilirubinemia, skin toxicity and mucositis. There was one death reported due to infectious complications. Conclusion: The efficacy and toxicity of TKI in study group were found to be similar to those described in the literature.

Immunotherapy supports other therapeutic methods and is an important element in the fight against cancer. Its main tasks include stimulation and guidance of the immune system to fight cancer, inhibition of the mechanisms that block the immune system, and direct destruction of neoplastic cells. Different from chemotherapy or hormonal therapy, its mechanism of action is associated with a different profile of adverse events. HER2 receptor inhibitors may cause symptoms of heart failure, which usually recede, once the treatment has been discontinued. Bevacizumab, an anti-VEGF antibody, induces numerous cardiovascular complications, including arterial hypertension, arterial embolism, haemorrhage and haemoptysis as well as venous thromboembolism.

Physiologically, angiogenesis is tightly regulated, or otherwise it leads to pathological processes, such as tumors, inflammatory diseases, gynecological diseases and diabetic retinopathy. The vascular endothelial growth factor (VEGF) is a potent and critical inducer of angiogenesis. The VEGF gene expression is regulated by a variety of stimuli. Hypoxia is one of the most potent inducers of the VEGF expression. The hypoxia inducible factor 1 (HIF-1) plays as a key transcription factor in hypoxia-mediated VEGF gene upregulation. Nitric oxide (NO) as well as hypoxia is reported to upregulate the VEGF gene by enhancing HIF-1 activity. The Akt/protein kinase B (PKB) pathway may be involved in NO-mediated HIF-1 activation in limited cell lines. There are some reports of negative effects of NO on HIF-1 and VEGF activity. These conflicting data of NO effects may be attributed mainly to the amount of released NO. Indeed, NO can be a positive or negative modulator of the VEGF gene under the same conditions simply by changing its amounts. The VEGF-mediated angiogenesis requires NO production from activated endothelial NO synthase (eNOS). Activation of eNOS by VEGF involves several pathways including Akt/PKB, Ca2+/calmodulin, and protein kinase C. The NO-mediated VEGF expression can be regulated by HIF-1 and heme oxygenase 1 (HO-1) activity, and the VEGF-mediated NO production by eNOS can be also modulated by HIF-1 and HO-1 activity, depending upon the amount of produced NO. These reciprocal relations between NO and VEGF may contribute to regulated angiogenesis in normal tissues.

This study investigated associations between the growth hormone/insulin-like growth factor-1 (GH/IGF-1) axis, adiponectin, resistin and metabolic profile in 47 GH-deficient children before and during 12 months of GH treatment. 23 short age-matched children without growth hormone deficiency (GHD) or any genetic or chronic disorders were recruited as controls at baseline. Metabolic evaluation included measurements of adiponectin, resistin, IGF-1, total cholesterol (total-C), low-density lipoprotein cholesterol (LDL-C), high-density lipoprotein cholesterol (HDL-C), triglycerides (TG), glucose, insulin, glycated haemoglobin (HbA1c), thyroid stimulating hormone (TSH) and free thyroxine (free T4) concentrations. The GH-deficient children had significantly higher adiponectin (p<0.05) and total cholesterol (p<0.05) levels, and a significantly lower level of resistin (p<0.05) than the controls. Resistin at 6 months of GH treatment significantly correlated with changes in height SDS in that period (r=0.35) and with the level of fasting insulin (r=0.50), the HOMA-IR (r=0.56) and the QUICKI (r=-0.53) at 12 months of therapy. Adiponectin level at 12 months of GH treatment was significantly associated with changes in HDL-C within the first 6 (r=0.73) and within 12 (r=0.56) months of therapy, while resistin significantly correlated with an increment in IGF-1 within 12 months of treatment (r=0.49) and with total-C at 12 months (r=0.56). Untreated GH-deficient children had higher adiponectin and lower resistin levels than healthy short children without GHD. Adiponectin and resistin levels did not change significantly during the first 12 months of GH therapy. Good responders to GH treatment had a tendency for higher resistin level during GH therapy, which positively correlates with the insulin resistance parameters.

Growth differentiation factor-9 (GDF-9), an oocyte-derived member of the TGF-β superfamily, plays an essential role in regulation of follicular development. This study aimed to determine the cyclic changes in serum GDF-9 concentration, compare its levels before and after ovariohysterectomy (OHE), and investigate its potential as a tool in ovarian remnant syndrome (ORS) diagnosis in cats. GDF-9 measurements were performed on 50 cats referred for routine OHE. The stage of the estrous cycle was determined by vaginal cytology and measurement of serum estradiol and progesterone levels was carried out to detect the cyclic changes in circulating GDF-9. One week after OHE, serum samples were collected again from 30 cats to reveal differences in GDF-9 levels. GDF-9 levels in the follicular phase were significantly higher than those in the interestrus (p<0.05). The postoperative analysis could be performed. GDF-9 levels slightly decreased one week after OHE (p=0.053). In conclusion, blood GDF-9 levels change during the estrous cycle, and may decrease with age in cats. However, further studies are needed to reveal the efficiency of GDF-9 in ORS diagnosis.

Integrins are the major receptors within the extracellular matrix (ECM) that mediate several functions connected with cell life and metabolism, such as cell adhesion, migration, cytoskeletal organization, proliferation, survival, and differentiation. A vascular endothelial growth factor (VEGF) is one of the most important angiogenic factors. It has been suggested that the expression of this gene may play crucial physiological roles in reproductive organs. All investigated endometrial tissues were isolated on day 10-12 after mating. Control bitches, used in this study, were in metestrus, which was determined according to the vaginal cytology and progesterone level in blood. Early pregnancy was verified by flushing the uterine horns with PBS. Total RNA was isolated from the bitches endometrium by means of the Chomczyński and Sacchi method, treated by DNase I, and reverse-transcribed into cDNA. A quantitative analysis of integrins α2b, β2 and β3, VEGF 164, 182 and 188 cDNA was performed by RT-PCR. In results we have shown an increased expression of all investigated genes (integrins α2b, β2 and β3, VEGF 164, 182, and 188) in pregnant bitches uterus as compared to non-pregnant females (P<0.001). Our results indicated that the expression of genes encoding integrins and vascular endothelial growth factors is different in relation to the time of the embryo implantation and it is increased in the first period of this process. This may be associated with the induction of specific mechanisms responsible for receptivity of uterus following the embryo attachment. In addition, all of investigated genes are up-regulated in a pregnancy-specific manner and the increased expression of these genes may regulate the uterus function during the implantation of canine embryos.

Transforming growth factor β1 (TGF-β1) is a cytokine affecting cell proliferation and development, which also has an immunomodulatory activity. Correlations between polymorphisms of the TGF-β1 gene and clinical parameters of inflammatory bowel disease (IBD) were reported previously in adults. Here, we tested whether such correlations occur in pediatric patients suffering from IBD. One hundred and four pediatric IBD patients were involved in this study. Among them, 36 were diagnosed with Crohn's Disease (CD) and 68 were diagnosed with ulcerative colitis (UC). The control group consisted of 103 children, in which IBD was excluded. TGF-β1 levels were determined in plasma and intestinal mucosa samples. The presence of the TGF β1 protein and the amount of TGF β1 mRNA were estimated in intestinal mucosa by immunohistochemistry and reverse transcription Real-Time PCR, respectively. Four common polymorphisms of the TGF-β1 gene were investigated: -800G/A, -509C/T, 869T/C and 915G/C. No significant correlation between TGF-β1 genotypes and (i) TGF-β1 levels in plasma and tissue samples, (ii) TGF-β1 gene expression efficiency in intestinal mucosa, (iii) IBD clinical parameters and (iv) inflammatory activity could be detected in children suffering from IBD. We conclude that, contrary to previous suggestions, the four common polymorphisms of the TGF-β1 gene do not influence the susceptibility to or clinical parameters of IBD in the tested population of children.

Bronchial asthma is characterized by persistent airway inflammation and airway wall remodeling. Among many different cells and growth factors triggering changes in bronchi structure, transforming growth factor β1-induced fibroblast to myofibroblast transition is believed to be very important. The aim of this study was to evaluate whether theophylline (used in asthma therapy) and two other methylxanthines (pentoxifylline and its active metabolite lisofylline), may affect transforming growth factor β1-induced fibroblast to myofibroblast transition in bronchial fibroblasts derived from asthmatic patients. We show here for the first time that selected methylxanthines effectively reduce transforming growth factor β1-induced myofibroblast formation in asthmatic bronchial fibroblast populations. PTX was found to be the most effective methylxanthine. The number of differentiated myofibroblasts after PTX, LSF and THEO administration was reduced at least twofold. Studies on the use of methylxanthines opens a new perspective in the development of novel strategies in asthma therapy through their two-pronged, anti-inflammatory and anti-fibrotic action. In the future they can be considered as promising anti-fibrotic drugs.

Alcohol abuse is one of the most significant factors in the development of liver fibrosis. The pathomechanism of liver fibrosis is the same regardless of its etiology. Fibrosis is a sign of an imbalance between the synthesis of the extracellular matrix components and their degradation. Among the many cytokines that affect hepatic stellate cell activation it seems that transforming growth factor beta (TGF-β) is the most significant, either as the direct factor stimulating polymerase chain reaction (HSC) proliferation and transformation into myofibroblasts, or as the direct factor causing an increase in the activity of genes responsible for the synthesis of extracellular matrix components. The aim of the study was to reveal possible dependencies and differences between the presence of certain alleles of the TGF-β1 gene and its blood level in the study and control group. Blood samples were obtained from 39 patients, the control group consisted of 21 patients. The results obtained in the course of this study showed no statistically significant differences between the frequencies of particular polymorphisms. In the case of haplotype frequencies, insignificant differences were found for the algorithm Excoffier-Laval-Balding predicted haplotypes while one significant difference between the study and control groups was detected in case of the TC haplotype frequency predicted using the Expectation-Maximization algorithm. However, the difference in frequency of TC haplotype predicted by both algorithms was not significant. Genetic analysis of two single nucleotide polymorphisms (SNPs) in exon I of the TGF-β1 gene did not show significant differences between the occurrence of particular polymorphisms and haplotypes in the populations under study.

The dysregulation of the metabolism of glycosaminoglycan and protein components of extracellular matrix (ECM) is a typical feature of diabetic complications. High glucose-induced enrichment of ECM with hyaluronan (HA) not only affects tissue structural integrity, but influences cell metabolic response due to the variety of effects depending on the HA polymer molecular weight. TSP-1-dependent activation of TGFβ1 axis is known to mediate numerous matrix disorders in diabetes, but its role concerning HA has not been studied so far. In this work we demonstrated that 30 mM D-glucose increased the incorporation of [3H]glucosamine in high-molecular-weight (> 2000 kDa) HA of medium and matrix compartments of human mesangial cultures. Simultaneously, the synthesis of HA with lower molecular weight and HA degradation were not altered. The cause of the increased high-molecular-weight HA synthesis consisted in the up-regulation of hyaluronan synthase (HAS) 2 mRNA without alterations of the expression of HAS3, which generates HA of lower molecular weight. D-Glucose at 30 mM also stimulated the production of transforming growth factor β1 (TGFβ1), the excessive activation of which was determined by the up-regulation of thrombospondin-1 (TSP-1). The blockage of TGFβ1 action either by neutralizing anti-TGFβ1 antibodies or by quenching the TGFβ1 activation (with TSP-1-derived synthetic GGWSHW peptide) abolished the effect of high glucose on HAS2 mRNA expression and normalized the synthesis of HA. Exogenous human TGFβ1 had the same effect on HAS2 expression and HA synthesis as high glucose treatment. Therefore, we supposed that TSP-1-dependent TGFβ1 activation is involved in the observed high glucose effect on HA metabolism. Since high-molecular-weight HA polymers, unlike middle- and low-molecular weight HA oligosaccharides, are known to possess anti-inflammatory and anti-fibrotic functions, we suppose that the enrichment of mesangial matrix with high-molecular-weight HA may represent an endogenous mechanism to limit renal injury in diabetes.

RUNX2 is a member of the PEBP2/CBF transcription factors family controlling the expression of genes whose products are essential for bone formation. Mutations in the RUNX2 gene may be associated with cleidocranial dysplasia (CCD), a rare skeletal disease characterized by stature aberrations, delayed closure of the cranial sutures, hypoplastic or aplastic clavicles, and multiple dental abnormalities. As RUNX2 is involved in many signaling pathways, we hypothesize that CCD may be associated with their changes. We determined the expression of RUNX2 and its signaling partners TCF7, involved in canonical Wnt signaling, and fibroblast growth factor receptors, FGFR1 and FGFR2 in periodontum of CCD patients and control individuals. We did not observe any differences between the level of RUNX2, TCF7 and FGFR1/2 mRNA, determined by real-time PCR, in CDD patients and controls. Therefore, RUNX2 signaling pathways with their partners TCF7 and FGFR1/2 may not be involved in CCD pathogenesis.

Polymorphism of insulin like growth factor IGF-1gene in selected Polish sheep breeds. Research was carried out in 2009-2013 on 1751 sheep bred in Poland (1366 ♀; 385♂) - 4 meat-wool breeds: Polish Merino, Old type Polish Merino, Corriedale and Żelaźnieńska sheep and 3 meat breeds (Berrichone du cher, Suffolk, Charolaise) from 34 flocks selected randomly across the country. All animals were subjected to identification factor insulin-IGF-1 gene, in the assessment of C and T alleles. Summing up, it should be noted that in 4 meat-wool breeds and 2 meat breeds (Berrichone du cher , Suffolk) there were no polymorphism of alleles and genotypes of insulin-like growth factor (IGF-1) gene, limiting its scope to determine C allele and CC genotype. Only one Charolaise ewe (breed imported from France) had T allele and C:T genotype. That result indicates the need for further research about sheep imported and adapted in Polish production conditions and assess the adaptation process.
Polimorfizm genu insulinopodobnego czynnika wzrostu IGF-1 u wybranych polskich ras owiec. Badania przeprowadzono na 1751 owcach ras hodowanych w Polsce (1366♀ i 385♂) – 4 ras wełnisto-mięsnych: merynos polski, merynos polski starego typu, corriedale i owca żelaźnieńska oraz 3 ras mięsnych (berrichone du cher, suffolk i charolaise) w latach 2009-2013, pochodzących z 34 stad wybranych losowo na obszarze całego kraju. Wszystkie zwierzęta poddane były identyfikacji genu czynnika insulinopodobnego IGF-1, w zakresie oceny występowania alleli C i T. Podsumowując, stwierdzić należy iż u badanych 4 ras wełnisto-mięsnych oraz 2 ras mięsnych (berrichone du cher i suffolk) nie wykazano polimorfizmu występowania alleli i genotypów genu insulinopodobnego czynnika wzrostu IGF-1, ograniczając jego zakres do ustalenia jedynie do allelu C i genotypu CC. U importowanej z Francji rasy charolaise stwierdzono odstępstwo od tej reguły tylko u 1 maciorki posiadającej allel T i genotyp C:T. Wynik ten wskazuje na potrzeby prowadzenia dalszych badań z tego zakresu u owiec pochodzących z importu i adaptowanych w polskich warunkach środowiska produkcyjnego, na podstawie którego można będzie ocenić zakres procesów adaptacyjnych.

Opis:
Angiogeneza jest jedna z cech charakterystycznych raka. Lepsze ilosciowe zrozumienie tego procesu pozwoliłoby na opracowanie skuteczniejszych terapii antyangiogennych. Postawiono hipoteze, ze nie tylko liczba komórek sródbłonka, ale takze jakosc układu naczyniowego, odgrywa wazna role w sposobie, w jaki chemio- iradioterapie sa dostarczane do guza. W zwiazku z tym rozwazamy minimalnie sparametryzowany matematyczny model proliferacji i dojrzewania komórek sródbłonka. Komórki sródbłonka dziela sie na dwa rodzaje - dojrzałe i niedojrzałe (lub proliferujace). Zakłada sie, ze komórki ulegaja samoistnemu dojrzewaniu, podczas gdy utracie jakosci naczyn krwionosnych posredniczy zewnetrzny czynnik wzrostu (tutaj VEGF). Rozwazany model został dopasowany do danych eksperymentalnych. Pokazuje on, w jaki sposób hamowanie VEGF prowadzi do lepszej jakosci układu naczyniowego i wolniejszej proliferacji.
Blood vessel sprouting (angiogenesis) is one of the hallmarks of cancer. Better quantitative understanding of this process would allow more effective antiangiogenic therapies to be developed. It has been hypothesised that not only the number of endothelial cells, but also the quality of the vasculature play an important role in how chemo- and radiotherapies are delivered to tumour site. Hence in this study a minimally-parametrised mathematical model of endothelial cell proliferation and maturation is developed. Endothelial cells are subdivided into two compartments -- mature and immature (or proliferating). The cells are assumed to undergo a self-mediated maturation, while loss of blood vessel quality is mediated by an external growth factor (here VEGF). The model is fitted to experimental data. The model shows how inhibition of VEGF results in better quality vasculature and slower proliferation.

Peroxisome proliferator-activated receptors-γ (PPARγ) are ligand-inducible transcription factors of the nuclear hormone receptor superfamily. We examined the effect of PPARγ activation on the generation of vascular endothelial growth factor (VEGF), one of the major angiogenic agents. Rat vascular smooth muscle cells (VSMC) and murine macrophages RAW264.7 were incubated for 24 h with PPARγ activators: prostaglandin J2 and ciglitazone. PPARγ were expressed in VSMC and RAW cells and their activity was upregulated in the presence of PGJ2 and ciglitazone. Incubation of the cells with PPARγ activators significantly augmented the release of VEGF protein into the media, both in resting and in IL-1β- or LPS-stimulated cultures. The higher protein generation was connected with the increased expression of mRNA and transcriptional activation of VEGF promoter. We conclude that the activation of PPARγ upregulates the generation of VEGF and may be involved in the regulation of angiogenesis.

In the veterinary literature there are few data concerning the expression of insulin-like growth factor type I (IGF-IR) in the canine mammary gland tumors. The aim of the present study was the evaluation of IGF-IR expression and its correlation to the expression of estrogen receptor α (ERα) and progesterone receptor (PR), proteins: Bcl-2, Bax, p53 in canine mammary gland tumors, and also a correlation with other features: bitch’s age, tumor diameter, histologic type of tumor, degree of histologic malignancy, proliferate activity. The study was done on 112 epithelial neoplasms: 21 (19%) were adenoma, 38 (34%) complex carcinoma (adenocarcinoma), 47 (42%) simple carcinoma (adenocarcinoma) and 6 (5%) solid carcinoma. Histochemistry and immunohistochemistry methods were employed. It was shown that more common and/or higher IGF-IR expression in cells of canine mammary gland tumors was related to the histologic type of cancer of worse prognostic (solid and simple carcinoma), high histologic degree of malignancy (IIIo) but the statistical analysis did not reveal any significant differences. We observed the high degree of IGF-IR expression in tumors which displayed the high ERα and PR expression. These results suggest the involvement of IGF-IR in the development of hormonosensitive canine mammary tumors. Additionally, the significant positive correlation between expression of IGF-IR and p53, Bax was found. Our study provides some evidence that interactions exist between the IGF-IR and these apoptosis-associated proteins may contribute to the development and progression of canine mammary gland tumors. These results require further investigations.

Background. Insulin-like growth factor 1 (IGF-1) is known as somatomedin C. This polypeptide hormone is functionally and structurally similar to insulin. IGF-1 effects on tissue by the IGF-1R and the insulin-like growth factor-binding protein also known as IGFBP. Abnormal IGF-1 and IGFBP signaling are positively correlated with a high risk of selected civilization diseases development. Physical inactivity is a one of the main causes of majority of chronic diseases and it is associated with eg. IGF-1 and IGFBPs level. Objective. The aim of the study was to explanation the effect of physical activity on IGF-1 and its binding protein – IGFBPs concentration in the context of selected civilization diseases prevention. The review of clinical trial. Material and Methods. The review of articles had published in databases: MEDLINE, EMBASE, Scopus and Web of Science until December 2015. The selected prospective studies about the effect of exercise on IGF-1 level and its binding protein IGFBP in the context of selected civilization diseases prevention were collected. Results. The majority of the included studies indicate that mechanical loading is a key mechanism linking IGF-1/IGFBPs concentration and selected chronic diseases development. The duration and intensity of physical activity have a significant impact on IGF-1 and IGFBP serum. The highest concentration of IGF-1 in serum was after eccentric training. “Overtraining” increases unfavorable and unbound IGF-1 levels and contributes to the increased incidence of hormone-cancer and osteoarthritis. Conclusions. Irregularity of the GH/IGF-1 axis may affect on the development of rheumatic diseases, cardiovascular diseases (regulate cardiac growth and metabolism) and metabolic syndrome.
Wprowadzenie. Insulinopodobny czynnik wzrostu 1 (IGF-1, insulin-like growth factor 1), zwany inaczej somatomedyną C. IGF-1 to hormon polipeptydowy, który funkcjonalnie i strukturalnie jest podobny do insuliny. IGF-1 oddziałuje na tkanki za pomocą receptora IGF-1R i białek wiążących IGFBP. Nieprawidłowości w produkcji IGF-1 i IGFBP są związane z wysokim ryzykiem rozwoju wielu chorób cywilizacyjnych. Brak aktywności fizycznej i związany z tym nieprawidłowy poziom krążącego IGF-1 przyczyniają się do rozwoju wielu przewlekłych chorób. Cel. Celem badania była próba wyjaśnienia wpływu zmian stężenia IGF-1 i jego białka wiążącego IGFBP pod wpływem aktywności fizycznej na profilaktykę chorób cywilizacyjnych na podstawie analizy badań klinicznych. Materiał i metody. Dokonano przeglądu artykułów opublikowanych do grudnia 2015 i zamieszczonych w bazach: MEDLINE, EMBASE, Scopus i Web of Science. Zebrano najnowsze badania prospektywne dotyczące znaczenia wysiłku fizycznego dla utrzymania prawidłowego poziomu IGF-1 i jego białka wiążącego IGFBP w kontekście profilaktyki wybranych chorób cywilizacyjnych. Wyniki. Większość analizowanych badań wskazuje, że aktywność fizyczna regulując stężenie IGF-1 i IGFBPs wpływa na rozwój wybranych chorób cywilizacyjnych. Długość trwania i intensywność wysiłku fizycznego ma istotne znaczenie dla stężenia IGF-1 i IGFBPs. Największy wzrost IGF-1 obserwuje się w wyniku escentrycznej pracy mięśni. Stan „przetrenowania” powoduje wzrost niekorzystnego, niezwiązanego IGF-1 w surowicy. Wzrost poziomu IGF-1 niezwiązanego przyczynia się do zwiększonej zachorowalności na nowotwory hormonozależne i chorobę zwyrodnieniową stawów. Wnioski. Nieprawidłowości osi GH/IGF-1 mogą wpływać na rozwój chorób reumatycznych, chorób układu krążenia i zespołu metabolicznego.

Previously published studies on levels of the transforming growth factor-β1 (TGF-β1) protein and mRNA of the corresponding gene in patients suffering from inflammatory bowel diseases (IBD) gave varying results, leading to contradictory conclusions. To solve the contradictions, we aimed to assess longitudinally TGF-β1 protein and mRNA levels at different stages of the disease in children suffering from IBD. The study group consisted of 19 pediatric patients with IBD at the age between 3.5 and 18.4 years. The control group consisted of 42 children aged between 2.0 and 18.0 years. The plasma TGF-β1 concentration was measured with ELISA. mRNA levels of the TGF-β1 gene isolated from samples of the intestinal tissue were assessed by reverse transcription and real-time PCR. Levels of TGF-β1 protein in plasma and corresponding mRNA in intestinal tissue were significantly higher in IBD patients than in controls. TGF-β1 and corresponding transcripts were also more abundant in plasma and intestinal tissue, respectively, in patients at the active stage of the disease than during remission. In every single IBD patient, plasma TGF-β1 level and mRNA level in intestinal tissue was higher at the active stage of the disease than during remission. Levels of TGF-β1 and corresponding mRNA are elevated during the active stage of IBD but not during the remission. Longitudinal assessment of this cytokine in a single patient may help to monitor the clinical course of IBD.

Introduction. The biological activity of VEGF depends on the presence of its specific receptors on the endothelial surface: VEGFR-1, VEGFR-2, and on their soluble forms sVEGFR-1 and sVEGFR-2. The binding of the membrane-bound receptors with VEGF affects the permeability, proliferation and migration of vascular endothelial cells. This creates the necessary conditions for the vascularisation of solid tumours and for the spread of remote metastases. The sVEGFR-1 and sVEGFR-2 receptors are believed to be natural inhibitors of VEGF. Objective. To determine the clinical usefulness of VEGF and the sVEGFR-1 and sVEGFR-2 receptors level assay in women with primary breast cancer. The assessment also took into account: patient’s age, stage of the disease, histological grade, status of the axillary lymph nodes and size of the primary tumour. Material and methods. The concentrations of VEGF, sVEGFR-1 and sVEGFR-2 were ascertained in 103 women with primary breast cancer. The concentrations of VEGF in the plasma, and those of the soluble receptors sVEGFR-1 and sVEGFR-2 in the serum, were assessed by ELISA, R&D Systems. Results. The study found significantly raised concentrations of VEGF, sVEGFR-1 and sVEGFR-2 in the serum of women with breast cancer, relative to the values obtained from the control group. It was found that with increasing clinical stages of the disease, the levels of VEGF and concentrations of sVEGFR-1 and sVEGFR-2 also increased. Similar findings were noted when assessing the degree of the histological grade of the tumours. Significantly higher values of VEGF protein and the assessed receptors were obtained from women with metastases to the axillary lymph nodes. A positive relationship, though without statistical significance, was noted between the concentration of sVEGFR-2 and the size of the tumour. Conclusions. The high concentrations of the VEGF cytokine and the sVEGFR-1 and sVEGFR-2 receptors in women with breast cancer are responsible for giving rise to the processes of tumour angiogenesis. The concentrations of the VEGF protein and the soluble forms of the receptors sVEGFR-1 and sVEGFR-2 in the serum of breast cancer patients showed positive correlations with the clinical stage of the disease. These results point to the usefulness of VEGF assessment and its soluble receptors in the clinical evaluation of patients with breast cancer.

In this paper a novel application of latent factor growth models is applied to responses to the manufacturing industry tendency survey conducted by the Research Institute for Economic Development, Warsaw School of Economics. An approach based on a common factor was assumed to explain variation in time response to specific questions drawn from the survey questionnaire. It was demonstrated that responses to questions relating to general economic situation in Poland, inflation and employment were explained by a latent growth factor, which was confirmed by RMSEA. Using cross-correlation and an ARIMAX model, it was shown that slopes obtained from latent factor growth models could be applied to forecasting or at least nowcasting of GDP growth and unemployment rate. Survey data of the type described clearly offer potential for refinement of economic projections and it is hoped that this work might stimulate further discussion of the methodology based on latent factor growth modeling for forecasting main macroeconomic time series.

The objective of the study was to evaluate the effect of energy contents in a dry period diets in “7+1” feeding strategy of dry cow including 7 weeks of the dry period (far-off) and 1 week of a close-up period, on colostrum quality and the immune status of calves. Forty Holstein multiparous cows were dried at 56d before the expected date of calving and were assigned to the higher energy diet group (HE; 0.69 UFL/kg DM, NDF 52% DM), or the lower energy diet group (LE; 0.61 UFL/1kg DM, NDF 56% DM). From -7d to the expected calving date up to 21 d of lactation, all cows were fed the same fresh, lactation diet. Samples of colostrum were collected within 2h after parturition and its density, dry matter content and concentrations of immunoglobulins, fat, protein, lactose, urea and somatic cell counts were measured. Calves were weighed 2 h after calving and on the 21d of life. On 3d and 21d of calves’ life blood samples were collected and the concentration of IgG, IgA, IgM, insulin-like growth factor 1 (IGF-1) as well as total protein and albumin concentrations were determined. Treatments had no significant effect on composition of colostrum and serum immunoglobulins and IGF-1 concentration. In both groups the weight of calves at birth was similar, still daily body weight gain during 21 day of rearing period in HE group was higher than in the LE group (P=0.05).

A long-term sports training induces morphological and functional changes in the cardiovascular system, with the activation of angiogenesis being one of the most significant ones. Aim: The aim of the study was to assess the impact of an 8-month sports training on the serum levels of vascular endothelial growth factor and the physical performance in young athletes. Material and methods: A total of 28 sports middle school students (athletics faculty) aged 13 years, including 14 boys and 14 girls, were included in the study. All participants underwent clinical assessment at each stage of the study. Electrocardiographic and echocardiographic examinations were performed. Furthermore, the levels of vascular endothelial growth factor were measured and a cardiac stress test was performed, the outcome of which was used to calculate the physical working capacity (PWC170). Results: There was a statistically significant decrease (274.3 ± 195.7 vs. 193.8 ± 153.8 ng/ml, p < 0.001) in the serum levels of vascular endothelial growth factor during the 8-month sports training. The levels were significantly lower in male athletes both at baseline (196.2 ± 157.3 vs. 352.4 ± 204.0 ng/ml, p = 0.02), and at the end of the observation period (139.6 ± 110.9 vs. 247.9 ± 174.6 ng/ml, p = 0.003). In the same period, the average PWC170 value increased throughout the study group (108.6 ± 28.1 vs. 119.0 ± 34.1 W, p = 0.02), and sex-related statistically significant changes occurred only on the male group of athletes. No statistically significant correlations were found between the relative changes in the levels of vascular endothelial growth factor and the PWC170 index (Spearman’s rank correlation coefficient – Rs 0.158, p > 0.05). Conclusions: Long-term sports training results in a decrease in the levels of vascular endothelial growth factor. At the same time, physical efficiency improvement is observed, which may suggest the involvement of adaptive, exerciseinduced angiogenesis in the skeletal muscles. However, the observed changes show distinct differences depending on the sex.
Długotrwały trening sportowy prowadzi do morfologicznych i czynnościowych zmian w układzie sercowo-naczyniowym, wśród których ważne miejsce zajmuje aktywacja procesów angiogenezy. Cel pracy: Celem pracy była ocena wpływu 8-miesięcznego treningu sportowego na stężenie naczyniowo-śródbłonkowego czynnika wzrostu w surowicy oraz wydolność fizyczną u młodych lekkoatletów. Materiał i metody: Badaniem objęto 28 uczniów gimnazjum sportowego o profilu lekkoatletycznym w wieku 13 lat, w tym 14 chłopców i 14 dziewcząt. Na każdym etapie badania wszyscy uczestnicy zostali poddani ocenie klinicznej. Wykonano u nich badania elektrokardiograficzne oraz echokardiograficzne. Oznaczono także stężenie naczyniowo-śródbłonkowego czynnika wzrostu oraz przeprowadzono próbę wysiłkową, której wynik posłużył do wyliczenia wskaźnika wydolności tlenowej PWC170. Wyniki badań: Podczas 8-miesięcznego treningu stężenie naczyniowo- -śródbłonkowego czynnika wzrostu w surowicy zmniejszyło się istotnie statystycznie (274,3 ± 195,7 vs 193,8 ± 153,8 ng/ml, p < 0,001). Jego stężenie było też istotnie niższe w grupie sportowców płci męskiej, zarówno na początku (196,2 ± 157,3 vs 352,4 ± 204,0 ng/ml, p = 0,02), jak i na końcu okresu obserwacji (139,6 ± 110,9 vs 247,9 ± 174,6 ng/ml, p = 0,003). W tym samym okresie średnia wartość wskaźnika PWC170 zwiększyła się w całej badanej grupie (108,6 ± 28,1 vs 119,0 ± 34,1 W, p = 0,02), a w odniesieniu do płci zmiany istotne statystycznie wystąpiły tylko u sportowców płci męskiej. Nie stwierdzono istotnych statystycznie korelacji pomiędzy względnymi zmianami stężenia naczyniowo-śródbłonkowego czynnika wzrostu i wartości wskaźnika PWC170 (współczynnik korelacji rang Spearmana – Rs 0,158, p > 0,05). Wnioski: Podczas długotrwałego treningu sportowego dochodzi do obniżenia stężenia naczyniowo-śródbłonkowego czynnika wzrostu. Jednocześnie obserwowana poprawa wydolności fizycznej może sugerować, że odbywa się to przy udziale adaptacyjnej, indukowanej wysiłkiem fizycznym angiogenezy w mięśniach szkieletowych. Zmiany te wykazują jednak wyraźne różnice zależne od płci.

Opis:
Purpose: Growth differentiation factor-15 (GDF- 15) protein belongs to a transforming growth factor-β family which determines the growth and differentiation of cells. In cancers, GDF-15 influences on the proliferation, differentiation, viability, migration and invasiveness of cancer cells. The aim of our study was to evaluate the expression of GDF-15 in the tissue and its levels in sera of patients with colorectal cancer. Materials and methods: The level of GDF-15 in the sera of 55 patients diagnosed with colorectal cancer was determined using the ELISA method whereas expression of this protein was performed by immunohistochemical method. Results: The mean value of GDF-15 levels in the sera of patients with colorectal cancer was significantly higher than in healthy control group (p<0.001). The expression of GDF-15 in the tissue was weak, moderate and strong in 23.6%, 15.7% and 60.7% cases, respectively. Statistical analysis showed that the expression of GDF-15 correlated with patients’ age (p<0.005) and non-mucinous type of cancer (p<0.001). The high GDF-15 levels in the serum was associated with tumor size (p<0.01) and distant metastases (p<0.05). Conclusions: According to our results, we postulate that the level of GDF-15 in serum can be use to assess the metastatic behavior of colorectal cancer

Opis:
The aim of the present study was to define the effect of TGF-β1 on C2C12 myoblasts myogenesis. TGF-β1 together with its receptor is a negative auto-paracrine regulator of myogenesis, which influences the proliferation, differentiation, and functions of muscle cells. TGF-β1 exerts highly significant inhibitory effect on differentiation of C2C12 mouse myoblasts manifested by the impairment of cell fusion and very low expression of myosin heavy chain. The study of differentiating C2C12 mouse myoblasts treated with TGF-β1 revealed 502 genes (436 down-regulated and 66 up-regulated) with statistically different expression. TGF-β1-regulated genes were identified to be involved in 29 biological processes, 29 molecular functions groups and 59 pathways. The strongest inhibiting effect of TGF-β1 was observed in the cadherin and Wnt pathways. The key-genes that could play the role of TGF-β1 targets during myoblasts differentiation was identified such as: Max, Creb1, Ccna2, Bax, MdfI, Tef, Tubg1, Cxcl5, Rho, Calca and Lgals4.
