We investigated the effect of daunorubicin on glutathione content and activity of GSH-related enzymes in cultured normal and diabetic human fibroblasts. Cells were incubated with 4 μM daunorubicin (DNR) for 2 h followed by culture in drug-free medium for up to 72 h. Treatment of diabetic cells with the drug caused a time-dependent depletion of intracellular GSH and a decrease of the GSH to total glutathione ratio. GSH depletion was accompanied by apoptotic changes in morphology of the nucleus. Analysis of GSH-related enzymes showed a significant increase of the activities of Se-dependent and Se-independent peroxidases and glutathione S-transferase. In contrast, glutathione reductase activity was reduced by 50%. Significant differences between normal and diabetic cells exposed to DNR were observed in the level of GST and Se-dependent glutathione peroxidase activities. These findings indicated that daunorubicin efficiently affects the GSH antioxidant defense system both in normal and diabetic fibroblasts leading to disturbances in glutathione content as well as in the activity of GSH-related enzymes.

We investigated glutathione level, activities of selenium independent GSH peroxidase, selenium dependent GSH peroxidase, GSH S-transferase, GSH reductase and the rate of lipid peroxidation expressed as the level of malondialdehyde in liver tissues obtained from patients diagnosed with cirrhosis or hepatocellular carcinoma. GSH level was found to be lower in malignant tissues compared to adjacent normal tissues and it was higher in cancer than in cirrhotic tissue. Non-Se-GSH-Px activity was lower in cancer tissue compared with adjacent normal liver or cirrhotic tissue, while Se-GSH-Px activity in cancer was found to be similar to its activity in cirrhotic tissue and lower compared to control tissue. An increase in GST activity was observed in cirrhotic tissue compared with cancer tissue, whereas the GST activity in cancer was lower than in adjacent normal tissue. The activity of GSH-R was similar in cirrhotic and cancer tissues, but higher in cancer tissue compared to control liver tissue. An increased level of MDA was found in cancer tissue in comparison with control tissue, besides its level was higher in cancer tissue than in cirrhotic tissue. Our results show that the antioxidant system of cirrhosis and hepatocellular carcinoma is severely impaired. This is associated with changes of glutathione level and activities of GSH-dependent enzymes in liver tissue. GSH and enzymes cooperating with it are important factors in the process of liver diseases development.

Male reproductive organs are extremely sensitive to the negative influence of toxic environmental factors as well as drugs, and until now not many attempts have been made at studying the detoxication enzymes and the relationship between the activity of those enzymes and spermatozoa fertility. In the present work we studied cytosolic glutathione-S-transferases (GST, EC 2.5.1.18) from different parts (head, corpus and tail) of bull and boar epididymis. We isolated two molecular forms of GST from each part of epididymis, characterized their biochemical properties and examined the mechanism of the catalyzed reaction. On the basis of their substrate specificity and isoelectric point, the isoforms were found to belong to the near neutral GST class mi. All examined GST forms exhibited higher affinity towards GSH than towards 1-chloro-2,4-dinitrobenzene (CDNB) and bull epididymis GST forms showed biphasic Lineweaver-Burk double reciprocal curves in the presence of GSH as a variable substrate. Boar epididymis anionic GST had the -SH groups both in the GSH and the CDNB binding place, whereas the cationic GST form - arginine residues in the CDNB binding place. Bull epididymis GST forms contained neither thiol nor arginine residues essential for catalytic activity.

Introduction. Oxidative stress induced by the generation of excess reactive oxygen species is one of the cause of atherosclerosis finally leading to coronary heart disease (CHD). Glutathione reductase (GR), a flavoprotein antioxidant enzyme, regenerates glutathione (GSH) from its oxidized form, which is essential for scavenging of hydrogen peroxide by glutathione peroxidase. The aim of this study was to analyze the activity of GR and GSH level in serum of patients with various stages of CHD. Materials and method. Sixty consecutive patients with diagnosis of stable angina (SA, n=20), unstable angina (UA, n=20) and non-fatal myocardial infarction (MI, n=20) together with 20 healthy control subjects were enrolled. Venous blood samples were collected just before the coronary catheterization procedure (in study patients). GR activities and GSH concentration was determined using commercially available colorimetric kits. Results. The highest GR activity and GSH serum level was noticed in UA patients. However, statistical evaluation showed that only glutathione reductase activity was significantly higher in serum obtained from myocardial infarction patients, compared to stable angina pectoris patients (19.52 ± 13.88 vs 11.63 ± 5.45 nmol/min/ml for MI and SA, respectively; p<0.048). There were no significant differences in GSH between the CHD patients and controls. Conclusion. The elevated activity of glutathione reductase in serum of patients with unstable angina pectoris and myocardial infarction suggests the role of antioxidant system acute coronary syndromes.

The aim of the study was the evaluation of cadmium effects on the activity of antioxidant enzymes in rat hepatocytes. The studies were conducted with isolated rat hepatocytes incubated for 1 or 2 hours in a modified (deprived of carbonates with phosphates) Williams’ E medium (MWE) in the presence of cadmium chloride (25, 50 and 200 μM). Hepatocytes incubated in the MWE medium without cadmium chloride were used as a control. The application of the modified Williams’ E medium allowed for the appearance of cadmium compounds in a soluble form that is indispensable for suitable estimation of its toxic action. There were evaluated markers of the oxidative stress such as: concentration of thiobarbiturate reactive substances (TBARS) – proportional to the level of lipid peroxidation, concentration of reduced glutathione (GSH), and the activity of antioxidant enzymes, including superoxide dismutase (SOD1 and SOD2), catalase (CAT), total glutathione peroxidase (GSHPx), selenium – dependent glutathione peroxidase (SeGSHPx), glutathione transferase (GST) and glutathione reductase (GSHR). Alterations of antioxidant enzymes activity, the level of TBARS and GSH in isolated rat hepatocytes caused by cadmium in vitro, were shown to depend on the concentration and time of exposure of cells to this metal. The increased level of TBARS and GSH was observed as well as changes in the activity of antioxidant enzymes. The activity of SOD isoenzymes and CAT was increased, whereas GSHPx and GST were decreased. These results indicate that cadmium induces oxidative stress followed by alterations in the cellular antioxidant enzyme system in isolated rat hepatocytes.

Opis:
Disturbances in the antioxidant system could play a role in pathogenesis of chronic liver disease. The aim of our study was to evaluate the levels/activities of antioxidants in blood of patients with chronic liver disease. We estimated selenium and glutathione concentrations and glutathione peroxidase activities in blood of 59 patients with chronic hepatitis B or C virus infection (group 1) and 64 patients with alcoholic, autoimmune or cryptogenic chronic liver disease (group 2). The results were compared with 50 healthy controls. Whole blood and plasma selenium and red cell glutathione concentrations were significantly lower in the patients compared with the controls. Red cell glutathione peroxidase activity was slightly reduced in both subgroups of group 1 and in group 2 with normal alanine aminotransferase values. Plasma glutathione peroxidase activity was slightly but significantly higher in patients with elevated aminotransferase values. The findings suggest that disturbances in antioxidant parameters in blood of patients with chronic liver disease may be the cause of the peroxidative damage of cells.

The active transport of oxidized glutathione and glutathione S-conjugates has been demonstrated for the first time in erythrocytes and this cell remained the main subject of research on the "glutathione S-conjugate pump" for years. Further studies identified the "glutathione S-conjugate pump" as multidrug resistance-associated protein (MRP). Even though cells overexpressing MRP and isolated MRP provide useful information on MRP structure and function, the erythrocyte remains an interesting model cell for studies of MRP1 in its natural environment, including the substrate specificity and ATPase activity of the protein.

In most cells, the major intracellular redox buffer is glutathione (GSH) and its disulfide-oxidized (GSSG) form. The GSH/GSSG system maintains the intracellular redox balance and the essential thiol status of proteins by thiol disulfide exchange. Topoisomerases are thiol proteins and are a target of thiol-reactive substances. In this study, the inhibitory effect of physiological concentration of GSH and GSSG on topoisomerase IIα activity in vitro was investigated. GSH (0-10 mM) inhibited topoisomerase IIα in a concentration-dependent manner while GSSG (1-100 µM) had no significant effect. These findings suggest that the GSH/GSSG system could have a potential in vivo role in regulating topoisomerase IIα activity.

In Escherichia coli, heterologous production of Schizosaccharomyces pombe phytochelatin synthase (PCS) along with overproduction of E. coli serine acetyltransferase (SAT) and γ-glutamylcysteine synthase (γECS) was achieved and resulted in the accumulation of phytochelatins in bacterial cells. Overproduction of either γECS alone or simultaneous production of all three proteins in bacterial cells were accompanied by reduced growth rate in liquid cultures. Interestingly, bacteria overproducing either γECS or both SAT and γECS (with elevated level of γ-glutamylcysteine but not of phytochelatins) were able to accumulate more cadmium per dry weight than the control. However, the most efficient cadmium accumulation was observed in bacteria with elevated levels of all three proteins: SAT, γECS and PCS. Therefore, "pushing" the entire pathway might be the most promising approach in modification of bacteria for potential bioremediation purposes because the level of intermediates, cysteine and glutathione, can limit the rate of production of phytochelatins. However, in such bacteria other metabolic process might become limiting for efficient growth.

Cadmium (Cd) stress significantly affects plant growth and development. Potentilla sericea is typically grown in gardens or as ground cover. In this study, the Cd response of P. sericea was analyzed based on physiological examinations and transcriptome analyses that uncovered the gene expression changes in P. sericea roots induced by a 7-day treatment with 90 μmol/L Cd2+. A total of 53,225 unigenes were identified, including 11,684 differentially expressed genes (DEGs; 8,083 upregulated and 3,601 downregulated). Additionally, 44 gene ontology terms and 127 Kyoto Encyclopedia of Genes and Genomes pathways were significantly enriched among these DEGs. Genes related to glutathione metabolism, plant hormone signal transduction, peroxisome processes, sulfur metabolism, and flavonoid biosynthesis pathways were confirmed as relevant to the Cd response of P. sericea. The molecular biology-related data described here may be useful for the future breeding of transgenic P. sericea plants with increased resistance to heavy metal stresses.

Background: Feature of modern existing hazards both environmental and occupational is cumulative exposure often leading to unexpected response of the organism resulting, among other things, in interactions with cytochrome P450 system involved in biotransformation of trichloroethylene and paracetamol. Hepatotoxity of paracetamol is closely connected with hepatic glutathione level. „In therapy of acute paracetamol poisoning application of N-acetylcysteine as a factor, which protects GSH level in cells, is recommended.” Materials and method: Tests were performed on rats which were treated with trichloroethylene, paracetamol and/or N-acetylcysteine. In rat liver total level of glutathione was determined i.e. reduced and oxidized form. Results: Paracetamol just after completion of the exposure affected the glutathione level. Trichloroethylene throughout the period of observation stimulated growth of glutathione level in liver. N-acetylcysteine didn’t have any influence on the level of investigated tripeptyde. Conclusions: N-acetylcysteine removes negative effect of paracetamol especially when it’s applied with 2-hour delay. After exposure for trichloethylene immediate application of N-acetylcysteine caused noticeable lowering of glutathione level. Cumulative exposure for three xenobiotics had positive influence for glutathione level in rat liver.
Wstęp: Cechą współcześnie występujących zagrożeń, zarówno środowiskowych jak i zawodowych jest narażenie łączne, wielokrotnie prowadzące do nieprzewidzianej odpowiedzi biologicznej organizmu, wynikającej między innymi z oddziaływań na układ cytochromu P450 biorący udział w biotransformacji trichloroetylenu i paracetamolu. Hepatotoksyczność paracetamolu jest między innymi ściśle związana z wątrobowym poziomem glutationu. W terapii zatruć ostrych paracetamolem zalecane jest podawanie N-acetylocysteiny jako czynnika ochraniającego poziom GSH w komórkach. Materiał i metody: Badania wykonano na szczurach, które traktowano trichloroetylenem, paracetamolem i/lub N-acetylocysteiną. W wątrobie szczura oznaczano poziom całkowity glutationu, tj. formę zredukowaną i utlenioną. Wyniki: Paracetamol tuż po zakończeniu ekspozycji negatywnie wpływał na poziom glutationu. Trichloroetylen przez cały czas obserwacji stymulował wzrost poziomu glutationu w wątrobie. N-acetylocysteina nie miała żadnego wpływu na poziom badanego tripeptydu. Wnioski: N-acetylocysteina usuwała negatywny wpływ paracetamolu, szczególnie wtedy, kiedy podano ją z 2-godzinnym opóźnieniem. Po narażeniu na trichloroetylen natychmiastowe podanie N-acetylocysteiny niosło za sobą wyraźnie obniżenie poziomu glutationu. Narażenie łączne na trzy oceniane ksenobiotyki.

Water is a renewable resource. However, with the human population growth, economic development and improved living standards, the world’s supply of fresh water is steadily decreasing and consequently water resources for agricultural production are limited and diminishing. Water deficiency is a significant problem in agriculture and increasing efforts are currently being made to understand plant tolerance mechanisms and to develop new tools (especially molecular) that could underpin plant breeding and cultivation. However, the biochemical and molecular mechanisms of plant water deficit tolerance are not fully understood, and the data available is incomplete. Here, we review the significance of glutathione and its related enzymes in plant responses to drought. Firstly, the roles of reduced glutathione and reduced/ oxidized glutathione ratio, are discussed, followed by an extensive discussion of glutathione related enzymes, which play an important role in plant responses to drought. Special attention is given to the S-glutathionylation of proteins, which is involved in cell metabolism regulation and redox signaling in photosynthetic organisms subjected to abiotic stress. The review concludes with a brief overview of future perspectives for the involvement of glutathione and related enzymes in drought stress responses.

The effects of long-term administration of a standardised extract of Curcuma longa on the explorative activity in aged 24-month-old male Wistar rats were estimated in a hole-board test. The animals received the extract orally for two months in rodent chow at doses of 10 and 50 mg/kg/day. The correlations between concentration of some neurotransmitters and amino acids in selected brain regions, as well as the level of corticosterone in plasma (described previously), and the parameters of exploration were calculated. The antioxidant processes (GSH, GST, CAT and MDA levels) in heart and skeletal muscles were also investigated. The results suggested that the explorative activity of plant extract-treated rats was enhanced; however, no correlations between brain neurotransmitter concentration or plasma corticosterone level and the parameters of explorative activity were observed. In biochemical investigation, chronic C. longa extract administration influenced antioxidant processes (CAT, GST and MDA levels) in skeletal muscles of aged rats, but not in the heart muscle.

We have recently shown that RLIP76, a ral-binding GTPase activating protein, mediates ATP-dependent transport of glutathione-conjugates (GS-E) and doxorubicin (DOX) (S. Awasthi et al., Biochemistry 39, 9327, 2000). Transport function of RLIP76 was found to be intact despite considerable proteolytic fragmentation in preparations used for those studies, suggesting either that the residual intact RLIP76 was responsible for transport activity, or that the transport activity could be reconstituted by fragments of RLIP76. If the former were true, intact RLIP76 would have a much higher specific activity for ATP-hydrolysis than the fragmented protein. We have addressed this question by comparing transport properties of recombinant RLIP76 and human erythrocyte membrane RLIP76 purified in buffers treated with either 100 or 500 μM serine protease inhibitor, PMSF. The purity and identity of recombinant and human erythrocyte RLIP76 was established by SDS/PAGE and Western-blot analysis. These studies confirmed the origin of the 38 kDa protein, previously referred to as DNP-SG ATPase, from RLIP76. Higher PMSF concentration resulted in lower yield of the 38 kDa band and higher yield of intact RLIP76 from both human and recombinant source. In contrast, the substrate-stimulated ATPase activity in presence of DNP-SG, doxorubicin, daunorubicin, or colchicine were unaffected by increased PMSF; similarly, ATP-dependent transport of doxorubicin in proteoliposomes reconstituted with RLIP76 was unaffected by higher PMSF. These results indicated that limited proteolysis by serine proteases does not abrogate the transport function of RLIP76. Comparison of transport kinetics for daunorubicin between recombinant vs human erythrocyte RLIP76 revealed higher specific activity of transport for tissue purified RLIP76, indicating that additional factors present in tissue purified RLIP76 can modulate its transport activity.

The last step of detoxification of both endogenous and environmental toxicants is typically a conjugation that produces a bulky hydrophilic molecule. The excretion of such conjugates out of cells is of sufficient biological importance to have led to the evolution of ATP-driven export pumps for this purpose. The substrate specificity of such transporters is broad, and in some cases it has been shown to include not only anionic conjugates but also neutral or weakly cationic drugs. In the present article, we review the molecular identity, functional and structural characteristics of these pumps, mainly on the example of human erythrocytes, and discuss their physiological role in detoxification and in the multidrug resistance phenotype of cancer cells.

The mechanisms underlying cartilage matrix degradation in joint diseases is not fully understood but reactive oxygen species are implicated as main causative factors. Comparative studies of glutathione reductase (GR) activity in synovial fluid from patients with rheumatoid arthritis (RA), reactive arthritis (ReA) and osteoarthritis (OA) as well as correlations between GR activity and concentration of the major cartilage components in synovial fluid are presented in this study. We found significantly higher activity of GR in RA (about three-fold) and ReA (about two-fold) than in OA. In RA and ReA patients, GR activity in synovial fluid correlates negatively with the concentrations of collagen and degradation products of sulfated glycosaminoglycans. In OA patients the activity of GR was significantly lower than in RA and ReA, which positively correlated with the concentration of collagen and showed a tendency for positive correlation with the degradation products of sulfated glycosaminoglycans. Our results suggest that in RA and ReA patients increased activity of GR does not prevent the increased degradation of collagen and proteoglycans by ROS.

Opis:
GST pi, the main glutathione S-transferase isoform present in the human brain, was isolated from various regions of the brain and the in vitro effect of tricyclic antidepressants on its activity was studied. The results indicated that amitripyline and doxepin - derivatives of dibenzcycloheptadiene, as well as imipramine and clomipramine - derivatives of dibenzazepine, inhibit the activity of GST pi from frontal and parietal cortex, hippocampus and brain stem. All these tricyclics are noncompetitive inhibitors of the enzyme with respect to reduced glutathione and noncompetitive (amitripyline, doxepin) or uncompetitive (imipramine, clomipramine) with respect to the electrophilic substrate. Their inhibitory effect is reversible and it depends on the chemical structure of the tricyclic antidepressants rather than on the brain localization of the enzyme. We conclude that the interaction between GST pi and the drugs may reduce their availability in the brain and thus affect their therapeutic activity. On the other hand, tricyclic antidepressants may decrease the efficiency of the enzymatic barrier formed by GST and increase the exposure of brain to toxic electrophiles. Reactive electrophiles not inactivated by GST may contribute in adverse effects caused by these drugs.

This study aimed to identify bacterial pathogens in milk samples from dairy cows with subclinical and clinical mastitis as well as to assess the concentrations of oxidant-antioxidant parameters [malondialdehyde (MDA), reduced glutathione (GSH), and total GSH levels] in both blood and milk samples. From a total of 200 dairy cows in 8 farms, 800 quarter milk samples obtained from each udder were tested in the laboratory for the presence of udder pathogens. Cultivated bacteria causing intramammary infection from milk samples were identified by Matrix-Assisted Laser Desorption/Ionization-Time of Flight (MALDI-TOF). In addition, from tested animals 60 cows were selected includıng 20 healthy cows that were CMT negative, 20 cows with subclinical mastitis (SM), and 20 cows with clinical mastitis (CM) for detection of MDA, GSH, and total GSH levels in blood and milk samples. Three hundred and eighty (47.5%; 380/800), 300 (37.5%; 300/800), and 120 (15%; 120/800) of milk samples, respectively were CMT positive or SM and CM, and those positives were cows from different farms. We observed that 87.4% (332/380), 25.3% (76/300), and 34.2% (41/120) of cows with CMT positive, CMT negative, and CM had bacterial growth. The most predominantly identified bacteria were Staphylococcus chromogenes (18.7%) obtained mainly from SM and Staphylococcus aureus (16.7%) as the most frequent cause of CM. According to our results, dairy cows with CM had the highest MDA levels, the lowest GSH, and total GSH levels in both blood and milk samples however, high MDA levels and low GSH levels in milk samples with SM were observed. Based on our results, lipid oxidant MDA and antioxidant GSH could be excellent biomarkers of cow’s milk for developing inflammation of the mammary gland. In addition, there was no link between nutrition and MDA and GSH levels.

A potentiometric procedure for cysteine thiol group concentration monitoring in media generating free radicals was developed using a thiol specific silver-mercury electrode. Electrolytic deposition of mercury on a silver wire and treatment with 20 mM cysteine in 0.5 M NaOH were used to produce the electrode. A silver-chloride electrode in saturated KCl was the reference. A glass capillary with 1 M KNO3 in 1% agarose gel was the liquid junction. The electrode responded to cysteine concentration in the range from 0.01 to 20 mM yielding a perfect linear relationship for the dependence of log [cysteine] versus electrode potential [mV], with b0 (constant) = -373.43 [mV], b1 (slope) = -53.82 and correlation coefficient r2 = 0.97. The electrode potential change per decade of cysteine concentration was 57 mV. The minimal measurable signal response was at a cysteine concentration of 0.01 mM. The signal CV amounted to 4-6% for cysteine concentrations of 0.01 to 0.05 mM and to less than 1% for cysteine concentrations of 0.5 to 20 mM. The response time ranged from about 100 s for cysteine concentrations of 0.01 to 0.1 mM to 30 s at higher cysteine concentrations. The standard curve reproducibility was the best at cysteine concentrations from 0.1 to 20 mM. In a reaction medium containing cysteine and copper(II)-histidine complex ([His-Cu]2+) solution in 55 mM phosphate buffer pH 7.4 the electrode adequately responded to changes in cysteine concentration. Beside cysteine, the silver-mercury electrode responded also to thiol groups of homocysteine and glutathione, however, the Nernst equation slope was about half of that for cysteine.

Introduction and aim. Oxidative stress is one of the complications that accompany defects in thyroid hormone levels. This study was designed to evaluate oxidative stress markers in patients with hypothyroidism. Material and methods. This case control study was comprised of forty-two hypothyroid patients aged 36–46 years and forty age matched (35–43 years) healthy control participants randomly selected from the Endocrine Clinic of Al-Yarmook Hospital in Iraq. Blood levels of thyroid hormones malondialdehyde, glutathione, and paraoxonase-1 were assessed. Body mass index was calculated for each patient and control participant. Results. Regarding the data of oxidative stress markers in hypothyroid patients compared to controls, a significant elevation was reported in blood levels of malondialdehyde and a significant reduction was found in blood levels of glutathione (p=0.031). On the other hand, the blood levels of paraoxonase-1 were significantly different in hypothyroid patients compared with the control. Conclusion. Elevated blood levels of malondialdehyde and declined blood levels of glutathione in hypothyroid patients are a signal of oxidative stress and consequently increase the risk of cardiovascular complications.

Zredukowana forma glutationu jest jednym z najważniejszych przeciwutleniaczy występujących w organizmie człowieka, a niedobór tego związku prowadzi często do poważnych zaburzeń. Glutation bierze również udział w modulowaniu odporności, procesach detoksykacji, a także reguluje aktywność innych białek i enzymów. Jest on czułym wskaźnikiem wrażliwości organizmu na stres oksydacyjny a jego stężenie w poszczególnych narządach i komórkach może wskazywać na stopień ich uszkodzenia. Oznaczanie stężenia glutationu z wykorzystaniem technik chromatograficznych stanowi uzupełnienie a niejednokrotnie alternatywę w stosunku do innych metod analitycznych: enzymatycznych, spektrofotometrycznych, spektrofluorymetrycznych, elektromigracyjnych, chemiluminescencyjnych, czy magnetycznego rezonansu jądrowego. Do najważniejszych etapów przygotowania próbki należą: zabezpieczenie aminotioli przed wysoką temperaturą i działaniem substancji utleniających pochodzących z matrycy, odbiałczanie, redukcja disulfidów, a także derywatyzacja (z wyjątkiem technik elektromigracyjnych i tandemowej spektrometrii mas).
Reduced form of glutathione is one of the most important antioxidants, which is found in human body. Deficiency of this compound can often lead to serious disorders. It plays a central role in immunity modulation, detoxification capacities and regulation of activity of another peptides and enzymes. In every live organism it acts as a sensitive rate of susceptibility to oxidative stress and its concentration in each organ and cell shows the level of their damage. Determination of glutathione using chromatographic methods has been a complement and many a time is also an alternative to different widely used methods, such as: enzymatic, spectrophotometric, spectrofluorometric, electromigrative, chemiluminescence, nuclear magnetic resonance. The most important steps in the sample preparation process for analysis are: amino thiols protection from high temperature and oxidation caused by substances from matrix, precipitation, reduction of disulfides and also derivatization (except for in vivo assays and those based on electrochemical and tandem mass spectrometry). Several methods have been presented in order to identify and quantify glutathione forms in human biological samples such as liquid chromatography with electrochemical, UV, fluorometric, mass spectrometric detection capillary electrophoresis and gas chromatography with mass spectrometry. This review presents different techniques for glutathione routine analysis in hospitals and also useful in clinical researches with large groups of patients.

Glutathione reductase (GR, E.C 1.6.4.2) is a flavoprotein that catalyzes NADPH-dependent reduction of oxidized glutathione (GSSG) to reduced glutathione (GSH). The aim of this study was to investigate in vitro effects of phenolic compounds isolated from Sideritis brevibracteata on bovine kidney GR. The Sideritis species are widely found in nature and commonly used as medicinal plants. 7-O-glycosides of 8-OH-flavones (hypolaetin, isoscutellarein and 3'-hydroxy-4'-O-methylisoscutellarein) were isolated from aerial parts of Sideritis brevibracteata. These compounds inhibited bovine kidney cortex GR in a concentration-dependent manner. Kinetic characterization of the inhibition was also performed.

We have recently demonstrated that a previously known Ral-binding GTPase activating protein, RLIP76, can also catalyze ATP-dependent transport of various structurally unrelated xeno- and endobiotics irrespective of their net charge (Awasthi et al., 2000, Biochemistry, 39: 9327). RLIP76 is a non-ATP binding cassette (ABC) protein but it has two ATP-binding sites and shows basal ATPase activity which is stimulated in the presence of its transport substrates (allocrites) such as doxorubicin (DOX) and S-(2,4-dinitrophenyl) glutathione (DNP-SG). Proteoliposomes reconstituted with purified RLIP76 catalyze ATP-dependent, saturable transport of DOX, as well as of glutathione-conjugates including leukotrienes (LTC4) and the GSH-conjugate of 4-hydroxynonenal (GS-HNE). In erythrocytes the majority of transport activity for DOX, GS-HNE, and LTC4 is accounted for by RLIP76. Cells exposed to mild oxidative stress show a rapid and transient induction of RLIP76 resulting in an increased efflux of GS-HNE and acquire resistance to oxidative stress mediated toxicity and apoptosis. Cells transfected with RLIP76 acquire resistance to DOX through increased efflux of the drug suggesting its possible role in the mechanisms of drug-resistance. In this article, we discuss the significance of transport functions of RLIP76 highlighting its role in the defense mechanisms against oxidative injury, and modulation of signaling mechanisms.

The aim of the study was to evaluate the effect of acetaminophen (APAP) and/or trichloroethylene (TRI) on the liver cytochrome P450-dependent monooxygenase system, CYP2E1 and CYP1A2 (two important P450 isoforms), and liver glutathione (GSH) content in rats. Rats were given three different doses of APAP (250, 500 and 1000 mg/kg b...) and then the above-mentioned parameters were measured for 48 h. The lowest APAP dose produced small changes in the cytochrome P450 content of liver. At 500 mg/kg APAP increased the cytochrome P450 content to 230% of the control. The inductive effect was seen at 1000 mg/kg dose but at 24 h and later. NADPH-cytochrome P450 reductase activity was the highest after the lowest dose of APAP, while after the highest dose it was equal to the control value. TRI increased both the cytochrome P450 content and the NADPH-cytochrome P450 reductase activity. When TRI was combined with APAP, both these parameters increased in the first hours of observation, but they returned to the control values at 24 h. When APAP was given at 250 mg/kg, GSH levels decreased to 55% of the control at 8 h and returned to the control values at 24 h. The higher doses of APAP decreased GSH levels more than the lowest dose, but after 24 h GSH levels did not differ from those of the control. When TRI was given at 250 mg/kg, the GSH levels decreased to 68% of the control at 2 h and then they increased gradually and tended to exceed the control values at 48 h. The effect of TRI combined with APAP on the level of GSH was virtually the same as that of APAP alone given at 500 mg/kg.

Sepsis is a serious medical problem and is one of the main causes of high mortality in intensive care units. Fifty percent of patients with severe sepsis will develop acute lung injury (ALI). Amentoflavone (AMF) is a polyphenolic compound possessing potent anti-inflammatory activities. The study aimed to explore the protective effects of AMF against ALI in cecal ligation and puncture (CLP)-induced septic rats. The results showed that AMF administration protected against septic ALI, as reflected by marked amelioration of histological injury of lung tissues and decrease of pulmonary edema in CLP-treated rats. AMF ameliorated CLP-induced increase of systemic and lung TNFα and IL-1β and binding activity of p65 NF-κB, indicating the inhibition of inflammation. Moreover, AMF prevented CLP-induced oxidative stress, as evidenced by increase of oxygen consumption rate, decrease of TBARS content, increase of SOD activity and GSH level in lung tissue of CLP-treated rats. CLP resulted in significant decrease of mRNA expression of Nrf2 and GCLc, which was inhibited by AMF. AMF-induced protective effects on ALI, inflammation, and oxidative stress were inhibited by lentivirus shRNA-mediated silence of Nrf2 and buthionine sulphoximine (BSO), an inhibitor of GSH synthesis. AMF increased Nrf2-binding activity in GCLc promoters in lung tissue of CLP-treated rats. The results suggested that AMF protected against ALI in septic rats through upregulation of Nrf2-GCLc signaling, enhancement of GSH antioxidant defense, reduction of oxidative stress and final amelioration of inflammation and histological injury of lung. The data provide new therapeutic options for the treatment of sepsis-associated ALI.

Introduction: Head and neck squamous cell carcinoma (HNSCC) accounts for about 6% of all malignant cancers. In the epidemiology of oral cavity neoplasm, important risk factors include: tobacco smoking, alcohol abuse, bad oral hygiene, papilloma virus infection, riboflavin and iron deficiency. Objective: The objective of the investigation was a synthesis of Cu(II) complex and the evaluation of antioxidative enzymatic barrier in red blood cells of patients with head and neck tumor as well as in the control group. Materials and methods: For the investigation conduction, a consent of Bioethics Committee number RNN/142/09/KB was obtained. Blood for the examination was obtained from the patients of the Dapartment of Head and Neck Neoplasms Surgery Medical University of Łódź. The experiment was conducted on the group of 40 patients with HNSCC and 40 healthy people, using spectrophotometric method, glutathione peroxidase was marked. Results: The investigation was conducted on the hemolysate obtained from the patients that were divided into two groups – a study group (1 and 2), which consisted of patients diagnosed with head and neck cancer and a control group (1 and 2) – healthy people. A significant statistical result for GPX occurred in control-1 and study-1 group with complex compound Cu(II) (p<0,001). Conclusions: Presented research prove, that complex compound Cis-dichlorobis(N1-hydroxymethyl-3methylpyrazole-κN2)copper (II) has an impact on the activity of the antioxidative GPX enzyme.

Zmiana warunków otoczenia jest stresem dla organizmu, który może prowadzić do wzmożonej generacji reaktywnych form tlenu i licznych uszkodzeń w obrębie komórek. Istotną rolę w przeciwdziałaniu tym uszkodzeniom pełnią enzymy antyoksydacyjne. Celem niniejszej pracy było określenie wpływu zmian ciśnienia otoczenia w warunkach nurkowania na aktywność peroksydazy glutationowej oraz katalazy. W badaniu wzięło udział 11 płetwonurków w wieku od 18 do 41 lat. Badani spędzili 40 min w jeziorze, w wodzie o temperaturze 13° C, schodząc maksymalnie na głębokość około 9 m. Podczas zanurzenia wszyscy oddychali powietrzem. Krew do badań pobrano z żyły odłokciowej dwukrotnie: przed zanurzeniem w wodzie (kontrola) oraz bezpośrednio po wynurzeniu. W pracy wykazano tendencję do wzrostu aktywności katalazy oraz tendencję do obniżania się aktywności peroksydazy glutationowej, ale zmiany te nie były istotne statystycznie. Ciśnienie otoczenia wywierane przez wodę podczas nurkowania nie ma istotnego wpływu na aktywność peroksydazy glutationowej oraz katalazy w erytrocytach. Może to sugerować, że nie doszło do zwiększonej generacji nadtlenku wodoru będącego substratem reakcji katalizowanych przez oba enzymy. Brak istotnych statystycznie zmian aktywności badanych enzymów antyoksydacyjnych może być efektem zmian przystosowawczych w organizmie osób nurkujących.
The changes in ambient conditions are usually stress factor for the organism, which may lead to increased generation of reactive oxygen species and numerous damages within the cells. The important role in counteracting of free radical damages to cells is played by antioxidant enzymes. The aim of this study was to determine the impact of changes in ambient pressure during diving on activity of selected antioxidant enzymes: glutathione peroxidase and catalase . The study involved eleven divers, aged from 18 to 41 years. Subjects spent 40 minutes in the lake, in water at 13° C, going down to a depth of about 9 m. All divers breathe with the air. Blood samples were taken from basilica vein twice: before immersion in water (control) and immediately after surfacing. The study showed an increasing trend in catalase activity and trend to decrease in glutathione peroxidase activity, but the changes were not statistically significant. Ambient pressure exerted by the water during diving has no significant effect on the activity of glutathione peroxidase and catalase in erythrocytes. This may suggest, that there was no increased generation of hydrogen peroxide which is a substrate of the reaction catalyzed by two enzymes. No statistically significant differences in the activity of antioxidant enzymes could be due to adaptive changes in the body of divers.

Sulfur is an essential macronutrient for all living organisms. Plants are able to assimilate inorganic sulfur and incorporate it into organic compounds, while animals rely entirely on organic sources of sulfur. In the last decades sulfate availability in soils has become the major limiting factor for plant production in many countries due to significant reduction of anthropogenic sulfur emission forced by introducing stringent environmental legislation. The sulfur flux after transferring plants from optimal conditions to sulfur deficiency is regulated on multiple levels including transcription, translation and activity of enzymes needed for sulfate assimilation and synthesis of sulfur-containing metabolites. Most of these regulatory steps are not yet fully characterized. Plant responses to sulfur limitation are complex and can be divided into phases depending on the degree of sulfur shortage. The initial responses are limited to adaptations within sulfur metabolic pathway, while multiple metabolic pathways and developmental process are affected when sulfur shortage becomes more severe. The major aim of this work is a comprehensive review of recent progress in understanding the regulation of plant adaptations to sulfur deficit.

The aim of the paper is the assessment of the efficiency of anti-oxidative system in spinach plants growing in the substratum polluted with zinc. The assessment was conducted on the basis of changes of reduced glutathione (GSH) form concentration in the plant aboveground organs. Spinach plants, ‘Matador’ c.v., were cultivated in soils contaminated with zinc in two pot experiments conducted in 2010 and 2011. The experimental substratum was light, slightly soil with granulometric composition of sandy silt loam. Zinc in the acetate form, was supplied to the soil in four doses corresponded to this metal critical concentrations in soil with 0, I, II and III degrees of pollution with this element according to IUNG classification. Simultaneously, the control with natural Zn content in soil was maintained. Zn concentrations in spinach ranged from 412.8 to 1722 mg kg-1 d.m. and increased with growing degree of substratum pollution with this element. Over the course of the vegetation period the content of Zn in plants was generally greater. GSH content in spinach grown in both years of experiments fluctuated from 31.70 to 238 μg g-1 f.m. The biggest content of this compound in spinach was stated in the initial phase of plants growth. The plants tolerated only the first two Zn doses supplied to the substratum. Spinach growing in the objects where zinc additions to the soil equalled II and III degree of substratum pollution died shortly after germination. The plants from these objects in the initial growth phase contained significantly less GSH than spinach from the objects with two first degrees (0 and I) of substratum pollution with zinc or from the control. The content of reduced glutathione form in spinach is a good biomarker of oxidative stress caused by zinc presence in plants. Synthesis of a bigger amount of GSH conditions spinach plant resistance to over the norm zinc content in soil. The efficiency of antioxidative system in spinach is bigger in the initial phase of this plant growth.

The aim of this study was to compare the effect of parenteral and oral supplementation of Selenium (Se) and vitamin E (VTE) on selected antioxidant parameters in blood and colostrum as well as their effect on the incidence of mastitis in dairy cows during the final phase of gravidity (6 weeks) and first two weeks after calving. For the practical part of the study 36 dairy cows of Slovak pied breed in the second to fourth lactation-gestation cycle were selected. The animals weredivided into three groups: the control (C) and two experimental groups (D1 and D2). The selected groups were treated as follows: in group D1 products containing Se (Selevit inj.) and vitamin E (Erevit sol. inj.) were administered intramuscularly twice, six and three weeks prior to parturition; in group D2 a vitamin-minerals supplement in the form of sodium selenite (Na2SeO3) and dl-α-tocopherol acetate were supplemented orally for six weeks calving. The blood samples were collected from the vena jugularis in dairy cows approximately 42 days before calving (control sampling), on parturition day, and the 14th day after calving. Higher concentrations of Se and VTE were found in the blood plasma samples of both experimental groups collected on the day of parturition. In addition, the orally supplemented group (D2) showed higher Se and α-tocopherol concentrations in blood plasma on the14th day after calving as well a reduction of occurrence of mastitis by about 25 % compared to the control group. The relationship between inflammatory response and oxidative stress was also confirmed. The concentrations of milk malondialdehyde indicating lipid peroxidation during mastitis were significantly higher in milk samples from infected cows than in milk samples from healthy animals in each monitored group. In order to prevent oxidative stress and moderate inflammatory response in dairy cows it is very important to optimally balance their nutritive needs with an appropriate ratio of Se and VTE supplements. Therefore we still recommend supplementation of the cows’ postpartum dietwith 0.5 mg of Se/kg dry matter (DM) and 102 mg of dl-α-tocopherol acetate/kg DM to stabilize their optimal blood levels, stimulate the activity of glutathione peroxidase and reduce the incidence of mastitis.

Background: Numerous authors have shown that selenium (Se) concentration and glutathione peroxidase (GSH-Px) activity in plasma of chronic kidney disease (CKD) patients are lower than in healthy subjects, but there are only few publications on the level of GSH-Px protein in those patients and no reports on the effect of Se supplementation to HD patients on the level of this enzyme. Subjects and Methods: Se concentration and GSH-Px protein level in plasma were measured in a group of 30 CKD patients on hemodialysis (HD) supplemented with 200 µg Se/day for 3 months, and 28 patients on HD administered with placebo. Se concentration was measured by graphite furnace atomic absorption spectrometry and plasma GSH-Px protein level by the sandwich ELISA method using polyclonal antibody specific for human plasma GSH-Px. Results: Se concentration in patients on placebo did not change throughout the 3-month study period, but increased significantly in Se supplemented group. Se supplementation to CKD patients on HD had no effect on the level of GSH-Px protein. Conclusions: The lack of GSH-Px protein in CKD patients on HD is not linked to Se deficiency since the level of this element increased after Se supplementation while enzyme protein level did not change. The damaged kidney of HD patients is unable to synthesize GSH-Px, even after induction with selenium.

The metabolism of oxygen in aerobic organisms leads to generation of reactive oxygen species (ROS). These entities are able to oxidize almost all classes of macromolecules, including proteins, lipids and nucleic acids. The physiological level of ROS is usually regulated by antioxidant defense mechanisms. There are at least three groups of antioxidant enzymes: superoxide dismutases, catalases and glutathione peroxidases (GSH-Pxs) which neutralize ROS. The trace elements (copper, zinc and selenium) bound to the active sites of the above listed enzymes play an important role in the antioxidant defense system. In mammals, a major function of selenium (Se) and Se-dependent GSH-Pxs is to protect cells from oxidative stress. Selenium concentrations and GSH-Px activities are altered in blood components of chronic kidney disease (CKD) patients. The Se level is frequently lower than in healthy subjects and the concentration very often decreases gradually with advancing stage of the disease. Studies on red cell GSH-Px activity in CKD patients reported its values significantly lower, significantly higher and lower or higher, but not significantly as compared with healthy subjects. On the other hand, all authors who studied plasma GSH-Px activity have shown significantly lower values than in healthy subjects. The degree of the reduction decreases gradually with the progression of the disease. High inverse correlations were seen between plasma GSH-Px activity and creatinine level. A gradual decrease in plasma GSH-Px activity in CKD patients is due to the fact that this enzyme is synthesized predominantly in the kidney and thus the impairment of this organ is the cause of the enzyme's lower activity. Se supplementation to CKD patients has a slightly positive effect in the incipient stage of the disease, but usually no effect was observed in end-stage CKD. Presently, kidney transplantation is the only treatment that may restore plasma Se level and GSH-Px activity in patients suffering from end-stage CKD. A few studies have shown that in kidney recipients, plasma Se concentration and GSH-Px activity are restored to normal values within a period of 2 weeks to 3 months following surgery and thus it can be acknowledged that Se supplementation to those patients has a positive effect on plasma GSH-Px activity.

Main thiols and disulfides were determined in the hemolymph of the Jamaican field cricket Gryllus assimilis at various developmental stages. On the basis of these data, redox potentials of the glutathione, cysteine and homocysteine redox systems were calculated. The concentrations of all thiols studied decreased during development (at a stage of 6 molts) with respect to young crickets, and increased again in adult insects. Redox potentials of the glutathione and cysteine systems increased from values of -131.0±5.6 mV and -86.9±17.1 mV, respectively in young crickets to -58.0±3.6 mV and -36.1±4.2 mV, respectively, at the stage of 6 molts and decreased to values of -110.4±24.8 mV and -66.3±12.2 mV, respectively, in adult insects. Redox potentials of the glutathione and cysteine systems in the hemolymph of young and adult insects were similar to those reported for human plasma.

Particles generated from numerous anthropogenic and/or natural sources, such as crystalline α-Fe2O3 nanoparticles, have the potential to damage lung cells. In our study we investigated the effects of these nanoparticles (12.5 µg/ml) on lipid peroxidation and the antioxidative system in MRC-5 lung fibroblast cells following exposure for 24, 48 or 72h. Exposure to α-Fe2O3 nanoparticles increased lipid peroxidation by 81%, 189% and 110% after 24, 48 and 72h, respectively. Conversely, the reduced glutathione concentration decreased by 23.2% and 51.4% after 48 and 72h of treatment, respectively. In addition, an augmentation of the activities of superoxide dismutase, catalase, glutathione peroxidase, glutathione transferase and glutathione reductase within the interval between 48-72h was noticed. Taking into account that the reduced glutathione level decreased and the malondialdehyde level, a lipid peroxidation product, remained highly increased up to 72h of exposure, it would appear that the MRC-5 antioxidant defense mechanisms did not efficiently counteract the oxidative stress induced by exposure to hematite nanoparticles.

Glutathione-S-transferase (GST) a Phase-II drug detoxification enzyme, was detected in Setaria cervi, a bovine filarial parasite. In vitro effect of diethylcarbamazine, butylated hydroxyanisole and phenobarbitone on the GST of adult female S. cervi was assayed by the addition of these compounds in the maintenance medium. The specific activity of GST towards 1-chloro-2,4-dinitrobenzene was increased progressively 1.2-1.97, 1.3-2.4 and 1.2-2.7 times at 10-100 µM of diethylcarbamazine, butylated hydroxyanisole and phenobarbitone, respectively, after 5 h at 37°C. Substrate specificity studies showed a higher increase in specific activity with ethacrynic acid and no change with cumene hydroperoxide. Although the intensity of GST activity band was more in extract from diethylcarbamazine or butylated hydroxyanisole treated worms extract, an extra band of activity appeared in those worm extracts compared to control worm extract. SDS/PAGE showed increased thickness of the band corresponding to purified GST in extracts from diethylcarbamazine/butylated hydroxyanisole/phenobarbitone treated worms. Purification and quantification of GST from diethylcarbamazine and butylated hydroxyanisole treated worms indicated an increase in enzyme specific activity. The increase in GST protein by these agents was blocked by prior treatment with actinomycin D, indicative of a transcription dependent response. The role of this enzyme in motility and viability of microfilariae and adult female was tested in vitro using a range of known GST inhibitors. Of those tested, ethacrynic acid, ellagic acid, 1-chloro-2,4-dinitrobenzene, cibacron blue and butylated hydroxyanisole reduced the viability and motility of microfilariae and adult female worms at micromolar concentrations. These results suggest that S. cervi GST is inducible in response to the antifilarial drug diethylcarbamazine and may play an important role in parasite's survival, thus could be a potential drug target.

Activities of superoxide dismutase, catalase and glutathione peroxidase in erythrocytes of cystic fibrosis children were studied in order to estimate the severity of their deficiency. Our results point to increased susceptibility of erythrocytes of cystic fibrosis subjects to oxidative injury and indicate that the antioxidant status of patients should be carefully monitored.

The aim of the presented study was to evaluate the reductive activity of antioxidants in the presence of carbon manufactured by Radio Frequency Plasma Activated Chemical Vapor Deposition method (RF PACVD). 2,2-diphenyl-1-picrylhydrazyl (DPPH) method was employed to estimate antioxidants reductive potential. Based on the experimental results, the antioxidant activity of carbon colloid was not confirmed. Nevertheless the reductive activity of antioxidants measured in time manner, in the presence of carbon, was significantly elevated comparing to the antioxidants alone. The presented results suggest that the surface of carbon syn-thetized by RF PACVD method suspended in water, creates more friendly reductive environment for antioxidants with hydroxyl groups (ascorbic acid - AA and butylated hydroxyanisole - BHA) then with sulfhydryl groups (glutathione - GSH).
Węgiel wytworzony metodą RF PACVD zwiększa aktywność antyoksydantów. Celem doświadczenia było zbadanie aktywności redukującej wybranych antyoksydantów (kwasu askorbinowego, butylowanego hydroksyanizolu i glutationu) w obecności cząstek węgla wytworzonego metodą RF PACVD testem DPPH. Test ten polega na pomiarze kinetyki reakcji między przeciwutleniaczem a wolnymi rodnikami. Im skuteczniejszy przeciwutleniacz, tym stała szybkości powstawania zredukowanej formy difeny-lopikrylohydrazyny (DPPH-H) jest większa. Jakkolwiek nie stwierdzono aktywności redukującej hydrokoloidu węgla RF PACVD to jednak rozpuszczone w nim antyoksydanty istotnie zwiększyły swój potencjał redukujący w porównaniu do próbek zawierających czysty antyoksydant. Uzyskane wyniki sugerują, że cząstki węgla RF PACVD zawieszone w wodzie stwarzają korzystne środowisko dla aktywności związków redukujących z aktywną grupą hydroksylową (AA i BHA), ale nie dla związków z grupą sulfhydry-lową (GSH).

Cisplatyna jest cytostatykiem o wysokiej skuteczności w leczeniu nowotworów złośliwych, jednak powoduje poważne skutki uboczne, najczęściej w postaci uszkodzenia nerek (nefrotoksyczność), wątroby (hepatotoksyczność) i narządu słuchu (ototoksyczność). Jednym z ubocznych skutków chemioterapii przy zastosowaniu cisplatyny jest wywoływanie przez ten ksenobiotyk stresu oksydacyjnego w komórkach. Wolne rodniki powstające pod wpływem cisplatyny mogą być neutralizowane przez egzogenne i endogenne antyoksydanty (np. koenzym Q10). Koenzym Q10 (CoQ10) jest silnym antyoksydantem nieenzymatycznym, obecnym we wszystkich komórkach organizmu człowieka. W warunkach fizjologicznych, CoQ10 syntetyzowany jest w wystarczających ilościach do prawidłowej ochrony antyoksydacyjnej komórek, jednak w przebiegu chorób nowotworowych, a także podczas chemioterapii jego stężenie może znacznie się obniżyć. Celem przeprowadzonych badań była charakterystyka wpływu CoQ10 na stres oksydacyjny komórki indukowany cisplatyną. Za pomocą metod spektrofotometrycznych, oznaczano in vitro aktywność enzymów antyoksydacyjnych w erytrocytach – dysmutazy ponadtlenkowej (SOD) i peroksydazy glutationowej (GPx) oraz w mitochondriach – aktywność enzymu detoksykacyjnego II fazy biotransformacji – S-transferazy glutationowej (GST) po dodaniu cisplatyny oraz oceniano wpływ CoQ10 na aktywność tych enzymów. Na podstawie przeprowadzonych badań wykazano, że w stresie oksydacyjnym indukowanym cisplatyną, CoQ10 hamował spadek aktywności SOD oraz GPx w krwinkach czerwonych po dodaniu ksenobiotyku. Suplementacja CoQ10 u pacjentów przed rozpoczęciem chemioterapii może uzupełniać niedobory tego związku w organizmie oraz zwiększać odporność komórek na uszkodzenia oksydacyjne indukowane ksenobiotykiem.
Cisplatin is one of the most effective cytotoxic agents in the treatment of cancers. However, cisplatin has number of severe side effects, including such ones as nephrotoxicity, hepatotoxicity and ototoxicity. It is reported that cisplatin-induced oxidative stress in normal cells is one of the side effects of chemotherapy with cisplatin. Free radicals that are generated under the influence of cisplatin, are neutralized by endogenous and exogenous antioxidants (e.g. coenzyme Q10). Coenzyme Q10 (CoQ10) is a strong non-enzymatic antioxidant, presented in all cells of human body. Under physiological conditions, CoQ10 is synthesized in sufficient quantity to ensure proper antioxidant protection of cells. However, the concentration of CoQ10 may be decreased by cancers and chemotherapy. The purpose of this study was to characterize the influence of CoQ10 on the cisplatin-induced oxidative stress. Using spectroscopic methods, the activity of antioxidative enzymes in erythrocytes – superoxide dismutase (SOD) and glutathione peroxidase (GPx) and in the mitochondria – activity of II phase biotransformation enzyme – glutathione S-transferase (GST) after exposure to cisplatin was measured in vitro and the effect of CoQ10 on antioxidative enzymes activity was studied. CoQ10 inhibited the reduction in SOD and GPx activity after administration of cisplatin. However, the activity of GST, in samples with CoQ10, was reduced after exposure to cisplatin. The supplementation of CoQ10, before starting cisplatin chemotherapy, can minimalize the toxicity of cisplatin to normal cells.

Human glutathione S-transferase omega 1-1 (hGSTO1-1) is a newly identified member of the glutathione S-transferase (GST) family of genes, which also contains alpha, mu, pi, sigma, theta, and zeta members. hGSTO1-1 catalyzes the reduction of arsenate, monomethylarsenate (MMA(V)), and dimethylarsenate (DMA(V)) and exhibits thioltransferase and dehydroascorbate reductase activities. Recent evidence has show that cytokine release inhibitory drugs, which specifically inhibit interleukin-1b (IL-1b), directly target hGSTO1-1. We found that (+)-α-tocopherol phosphate and (+)-α-tocopherol succinate inhibit hGSTO1-1 in a concentration-dependent manner with IC50 values of 2 µM and 4 µM, respectively. A Lineweaver-Burk plot demonstrated the uncompetitive nature of this inhibition. The molecular mechanism behind the inhibition of hGSTO1-1 by α-tocopherol esters (vitamin E) is important for understanding neurodegenerative diseases, which are also influenced by vitamin E.

Two seed lots of Acer saccharinum (recalcitrant), with an initial moisture content of 50% and 55%, were stored at +3oC for 6 months. After this time, their viability (measured as germinability) reached 100% and 30%, respectively. In embryo axes and cotyledons extracted from seeds, two major low molecular antioxidants were assayed: ascorbate (ASA and DHA) and glutathione (GSH and GSSG); and activities of enzymes of the ascorbate-glutathione cycle were measured: ascorbate peroxidase (APO) (E.C. 1.11.1.11), monodehydroascorbate reductase (MR) (E.C. 1.6.5.4), dehydroascorbate reductase (DHAR) (E.C. 1.8.5.1), and glutathione reductase (GR) (E.C. 1.6.4.2.). GSH and GSSG contents of embryo axes of stored seeds decreased, as compared to the control (fresh, non-stored seeds), but a larger decrease was observed in seeds with 30% viability. In cotyledons, a particularly high increase in the GSH content in relation to the control was observed in seeds with 100% viability, while the GSSG content was significantly lower in both stored seed lots than in the control. The ASA level was twice as high in seeds with 30% viability as in the control, both in embryo axes and in cotyledons. The activity of enzymes of the ascorbate-glutathione cycle was higher in embryo axes than in cotyledons. In embryo axes of seeds with 100% viability, enzyme activities were slightly lower than in the control, while in those of seeds with 30% viability, their activities were higher than in the control. The observed changes in activities of enzymes of the ascorbate-glutathione cycle and in ascorbate and glutathione levels suggest that the stored seeds of A. saccharinum have an active antioxidant system, which plays an important role in maintaining their viability during storage.

Changes in blood leucocyte levels were investigated in Spraque-Dowley rats vaccinated with cDNA or protein of glutathione S-transferase (GST) of F. hepatica and subsequently challenged with metacercariae of the liver fluke. The analysis of the leucocyte responses measured in vaccinated rats suggests that the form of antigen used for vaccination intluenced dynamics of white blood cell response to the fluke infection. The most clear differences were observed in neutrophil and eosinophil levels. The weakest reaction of these cells to the challenge infection was observed in rats vaccinated twice with cDNA. In contrast, in rats which received the first antigen dose as cDNA and the second vaccination with GST protein, both neutrophil and eosinophil responses were much higher, especially at 5 and 9 WAI.
Badano reakcję leukocytów krwi szczurów Spraque-Dowley immunizowanych cDNA lub białkiem transferazy S-glutationowej (GST) F. hepatica na inwazję metacerkarii tej przywry. Zwierzęta grupy 3 otrzymały pierwszą dawkę antygenu w formie cDNA a drugą w postaci białka. Próbki krwi do badań pobrano w dniu zarażenia a następne w 1, 5 i 9 tygodniu po zarażeniu (tpz). U szczurów szczepionych cDNA zaobserwowano statystycznie istotne obniżenie liczby limfocytów poczynając od 1 tpz. Największe różnice zaobserwowano w reakcji granulocytów. Szczury immunizowane dwukrotnie cDNA wykazywały najmniejszą eozynofilię zaś u szczurów grupy 3, wystąpiła w 9 tpz eozynofilia i neutrofilia silniej wyrażona niż u szczurów nie immunizowanych.

Role of lipid peroxidation products, particularly 4-hydroxynonenal (4-HNE) in cell cycle signaling is becoming increasingly clear. In this article, recent studies suggesting an important role of 4-HNE in stress mediated signaling for apoptosis are critically evaluated. Evidence demonstrating the modulation of UV, oxidative stress, and chemical stress mediated apoptosis by blocking lipid peroxidation by the α-class glutathione S-transferases (GSTs) is presented which suggest an important role of these enzymes in protection against oxidative stress and a role of lipid peroxidation products in stress mediated signaling. Overexpression of 4-HNE metabolizing GSTs (mGSTA4-4, hGSTA4-4, or hGST5.8) protects cells against 4-HNE, oxidative stress (H2O2 or xanthine/xanthine oxidase), and UV-A mediated apoptosis by blocking JNK and caspase activation suggesting a role of 4-HNE in the mechanisms of apoptosis caused by these stress factors. The intracellular concentration of 4-HNE appears to be crucial for the nature of cell cycle signaling and may be a determinant for the signaling for differentiation, proliferation, transformation, or apoptosis. The intracellular concentrations of 4-HNE are regulated through a coordinated action of GSTs (GSTA4-4 and hGST5.8) which conjugate 4-HNE to GSH to form the conjugate (GS-HNE) and the transporter 76 kDa Ral-binding GTPase activating protein (RLIP76), which catalyze ATP-dependent transport of GS-HNE. A mild stress caused by heat, UV-A, or H2O2 with no apparent effect on the cells in culture causes a rapid, transient induction of hGST5.8 and RLIP76. These stress preconditioned cells acquire ability to metabolize and exclude 4-HNE at an accelerated pace and acquire relative resistance to apoptosis by UV and oxidative stress as compared to unconditioned control cells. This resistance of stress preconditioned cells can be abrogated by coating the cells with anti-RLIP76 antibodies which block the transport of GS-HNE. These studies and previous reports discussed in this article strongly suggest a key role of 4-HNE in stress mediated signaling.

Hydrogen sulfide (H2S) is endogenously synthesized from l-cysteine in reactions catalyzed by cystathionine beta-synthase (CBS, EC 4.2.1.22) and gamma-cystathionase (CSE, EC 4.4.1.1). The role of 3-mercaptopyruvate sulfurtransferase (MPST, EC 2.8.1.2) in H2S generation is also considered; it could be important for tissues with low CTH activity, e.g. cells of the nervous system. The expression and activity of CBS, CTH, and MPST were detected in the human glioblastoma-astrocytoma (U-87 MG) and neuroblastoma (SHSY5Y) cell lines. In both cell lines, the expression and activity of MPST were the highest among the investigated enzymes, suggesting its possible role in the generation of H2S. The RP-HPLC method was used to determine the concentration of cystathionine and alpha-ketobutyrate, products of the CBS- and CTH-catalyzed reactions. The difference in cystathionine levels between cell homogenates treated with totally CTH-inhibiting concentrations of dl-propargylglycine and without the inhibitor was used to evaluate the activity of CBS. The higher expression and activity of CBS, CTH and MPST in the neuroblastoma cells were associated with more intensive generation of H2S in the presence of 2 mM cysteine. A threefold higher level of sulfane sulfur, a potential source of hydrogen sulfide, was detected in the astrocytoma cells in comparison to the neuroblastoma cells.

Six-week old sunflower seedlings, cv. Ogrodowy, were treated with 0, 15, 45 and 60 mg Pb dm-3, and then the content of lead and selected physiological and biochemical parameters were measured. Photosynthesis efficiency, water relations (intensity of transpiration, relative water content (RWC)) and gene-encoding metallothionein were measured three times after 24, 48 and 72 hours of exposure to Pb. The content of glutathione and lead was analysed after 72 hours’ exposure to Pb. Most of the lead uptake was accumulated in the roots, then in the stems and leaves, but when re-calculated per plant dry weight, the uptake of the metal did not depend on the lead dose applied. The highest 60 mg Pb dm-3 treatment was accompanied by a significant decrease in dry weight content. Moreover, most of the lead taken up in these plants was transported to the stems and leaves (23.6% of total lead uptake). The lead doses used in this study did not affect the intensity of photosynthesis, but a decrease in transpiration and relative water content was observed. The glutathione level in the plants varied depending on the organ examined and the Pb concentration in the treatment. The expression of the metallothionein gene HaMT1 was observed in the stems only. These results indicate that the sunflower cultivar Ogrodowy is a promising plant for phytoremediation of lead-polluted soils.

The activity of GST in mouse skeletal muscles was measured during the muscular phase of trichinellosis, starting from the 3rd week post infection (w.p.i.) to the 11th w.p.i. Activity was determined spectrophotometrically by monitoring the formation of thioether (S-2,4-dinitrophenylglutathione) from the reduced form of glutathione and 1-chloro-2,4-dinitrobenzene used as a substrate and as an example of xenobiotics. The changes in the activity of GST were as follows: an increase in activity starts in the 4th w.p.i., peaks (up to 310% of the normal value) in the 6th w.p.i., decreases in the 8th week and a final, weak rise was observed in week 11. The statistically significant changes in GST activity in this phase of experimental trichinellosis suggest that this enzyme participates in the biochemical defence of the host against Trichinella infection.

Increasing pollution of the environment causedby heavy metals is becoming a significant problem in developing cities. Species andcultivars of plants for urban plantings shouldexhibit tolerance to these pollutants, andwhat is even more significant, through their absorption they shouldred uce the level of environmental contamination. The aim of the research was to determine whether Berberis thunbergii (DC.), which was grown in the immediate vicinity of roads, developed mechanisms limiting harmful effects of accumulating heavy metals. The mechanism for heavy metal resistance, involving the generation of phytochelatins (PCs), was investigatedin relation to As, Cd, Cr, Co, Cu, Hg, Ni, Pb andZn accumulation. Levels of thiols, i.e. glutathione (GSH) andphytochelatins (PCs), increasedin plants grown in pollutedareas in the city of Poznań in comparison to a residential site (control) and it was related to the activity of phytochelatin synthase (PC-synthase) andthe accumulation of metals. The results indicate that in Berberis thunbergii growing in the polluted urban environment a defense mechanism adapting the plant to potentially adverse conditions was initiated.

The paper attempted to assess the activity of antioxidative system in cells of spinach plant, ‘Matador’ c.v., growing in the soil contaminated with Ni. Plant material for analyses was obtained from two pot experiments conducted in 2010 and 2011 in the vegetation hall of the Experimental Station of the University of Agriculture in Krakow. Ni content in the plant aboveground parts was assessed by ICP-ES method, contents of reduced glutathione form by colorimetry and ascorbic acid by titrimetric method. Nickel content in spinach aboveground parts ranged from 2.00 to 204.5 mg kg–1 d.m. and increased with growing substratum pollution with this element and usually with plant age. The plants contained from 31 to 238 g GSH g–1 f.m. In the first three objects with 0o, Io and IIo degree of substratum pollution according to IUNG classification, this antioxidant contents were higher in comparison with its amount in plants from the control and objects with lower degree of pollution. In the object with the highest nickel dose application, GSH content in plants decreased significantly in comparison with plants from the other objects, while the plants on this object died shortly after germination. Ascorbic acid content in spinach in the both years of experiments ranged from 24.13 to 73.09 mg 100 g–1 f.m. and increased in plants from the successive objects with growing substratum contamination with nickel. In the first phase of growth spinach plants contained generally much more of GSH and AsA, which indicated much better efficiency of the antioxidative system at the initial period of growth.
W pracy podjęto próbę oceny aktywności systemu antyoksydacyjnego komórek roślin szpinaku odmiany ‘Matador’ rosnących w glebie zanieczyszczonej Ni. Materiał roślinny do analiz pozyskano w dwóch doświadczeniach wazonowych prowadzonych w latach 2010 i 2011 w hali wegetacyjnej stacji doświadczanej 996 Monika Arasimowicz et al Uniwersytetu Rolniczego w Krakowie. W częściach nadziemnych roślin oznaczono zawartość Ni metodą ICP-ES, zredukowanej formy glutationu metodą kolorymetryczną oraz kwasu askorbinowego metodą miareczkową. Zawartość niklu w częściach nadziemnych szpinaku wynosiła od 2,00 do 204,5 mg kg–1 s.m. i zwiększała się wraz ze stopniem zanieczyszczenia podłoża tym pierwiastkiem oraz na ogół wraz z wiekiem roślin. Rośliny zawierały od 31 do 238 g GSH g–1 ś.m. W roślinach z pierwszych trzech obiektów o zanieczyszczeniu podłoża odpowiadającym 0o, Io, IIo , według klasyfikacji IUNG, zawartość tego antyoksydantu była większa w porównaniu z zawartością GSH w roślinach z obiektu kontrolnego i obiektów o niższym stopniu zanieczyszczenia. W obiekcie z największym zastosowanym dodatkiem niklu zawartość GSH w roślinach zmniejszyła się istotnie w porównaniu z roślinami z pozostałych obiektów, a rośliny z tego obiektu obumarły niedługo po wschodach. Zawartość kwasu askorbinowego w szpinaku w obydwu latach doświadczeń mieściła się w przedziale od 24,13 do 73,09 mg 100 g–1 ś.m. i zwiększała się w roślinach z kolejnych obiektów o coraz większym zanieczyszczeniu podłoża niklem. Rośliny szpinaku w pierwszej fazie wzrostu zawierały na ogół znacznie więcej GSH i AsA, co wskazuje na znacznie wyższą sprawność systemu antyoksydacyjnego roślin w początkowym okresie wzrostu.

Opis:
Glutathione (GSH) is a endogenous , low molecular weight thiol compound. It has a wide spectrum of biological activity in the body. It is an important element of the antioxidant system that protects cells against the effects of oxidative stress. As an antioxidant, GSH inactivates free radicals and reactive forms of oxygen and nitrogen in enzymatic and non-enzymatic reactions, regenerates other antioxidants, e.g. vitamins C and E, maintains - SH groups in proteins in a reduced state, participates in the detoxification of xenobiotics. The decrease in GSH concentration occurs in many diseases and in the aging process. Increasing the level of GSH in the body's cells is possible by consuming dietary components containing GSH or amino acids (especially cysteine) for its endogenous synthesis and supplementation with GSH pharmaceutical preparations. The aim of the study was to characterize dietary supplements available in Poland containing glutathione (GSH). The analysis covered 39 supplements from GSH, which were available in Poland. Their characteristics include: place / country of production; type of preparations (single and multi-component); bioavailability of GSH contained therein - preparations in the liposome formula (lipophilic GSH) and in the non-liposome formula (non-lipophilic GSH); pharmaceutical form in which these preparations were available on the commercial offer and GSH content in supplements. Among 39 GSH supplements, there were 24 single-component and 15 multi-component preparations. The largest number of GSH supplements available in Poland came from the USA and Great Britain. Among GSH supplements, there were 15 liposome (lipophilic) formula preparations, of which 12 were lipophilic one-component and 3 multi-component and 24 non-liposome (non-lipophilic) formula preparations - 12 single and 12 multi-component. Capsules, gel, liquid and tablets were the most common pharmaceutical form among all analyzed GSH supplements. The rarest pharmaceutical form was lozenges, aerosol, powder and ampoule. Analysis of GSH supplements available in Poland showed that in one single dose of the preparation was from 18 mg to 750 mg GSH. The most common dose was 250, 450 and 500 mg GSH in a single dose. One-component supplements most often contained 450 mg, then 250 and 500 mg GSH, while multi-component supplements - 250 mg GSH.
Glutation (GSH) jest niskocząsteczkowym związkiem tiolowym, który wykazuje szerokie spektrum biologicznej aktywności. Jest ważnym elementem systemu antyoksydacyjnego chroniącego komórki przed skutkami stresu oksydacyjnego. Jako antyoksydant GSH unieczynnia wolne rodniki i reaktywne formy tlenu i azotu, regeneruje inne antyoksydanty np. witaminę C i E, utrzymuje grupy –SH w białkach w stanie zredukowanym, bierze udział w detoksykacji ksenobiotyków. Obniżenie stężenia GSH występuje w wielu schorzeniach. Zwiększenie poziomu GSH w komórkach organizmu jest możliwe poprzez spożywanie składników diety zawierających GSH, czy aminokwasy (szczególnie cysteina) do jego endogennej syntezy oraz suplementację preparatami farmaceutycznymi z GSH. Celem pracy była charakterystyka dostępnych w Polsce suplementów diety zawierających w swoim składzie glutation (GSH). Analizą objęto 39 suplementów z GSH. Ich charakterystyka uwzględnia: miejsce/kraj ich produkcji; typ preparatów (jedno- i wieloskładnikowe); biodostępność zawartego w nich GSH - preparaty w formule liposomowej (lipofilny GSH) i w formule nieliposomowej (nielipofilny GSH); postać farmaceutyczną preparatów oraz zawartość GSH w suplementach. Wśród 39 suplementów z GSH było 24 preparaty jedno- i 15 wieloskładnikowych. Największa liczba suplementów z GSH pochodziła z USA i Wielkiej Brytanii. Wśród suplementów z GSH było: 15 preparatów formuły liposomowej (lipofilnej ), z czego lipofilnych jednoskładnikowych było 12, a wieloskładnikowych 3 oraz 24 preparaty formuły nieliposomowej (nielipofilnej) - 12 jedno- i 12 wieloskładnikowych. Najczęściej występującą postacią farmaceutyczną wśród wszystkich analizowanych suplementów z GSH były kapsułki, żel, płyn i tabletki. W pojedynczej dawce preparatu znajdowało się od 18 mg do 750 mg GSH. Najczęściej znajdowało się 250, 450 i 500 mg GSH . Suplementy jednoskładnikowe najczęściej zawierały 450 mg GSH, natomiast wieloskładnikowe 250 mg GSH.

Changes in blood leucocyte levels were investigated in Spraque-Dowley rats vaccinated with cDNA or protein of glutathione S-transferase (GST) of F. hepatica and subsequently challenged with metacercariae of the liver fluke. The analysis of the leucocyte responses measured in vaccinated rats suggests that the form of antigen used for vaccination intluenced dynamics of white blood cell response to the fluke infection. The most clear differences were observed in neutrophil and eosinophil levels. The weakest reaction of these cells to the challenge infection was observed in rats vaccinated twice with cDNA. In contrast, in rats which received the first antigen dose as cDNA and the second vaccination with GST protein, both neutrophil and eosinophil responses were much higher, especially at 5 and 9 WAI.

The aim of the study was to investigate the concentration of chosen tracę elements, activity of catalase (CAT) and glutathione content (GSH) in human milk. The study subjects were recruited from among the lactating women from Małopolska province in Poland. Milk was taken from 23 mothers classified inlo three groups of age (20-25, 26-31 and 32-37 years) and two groups of lactation period (colostrums; 1-3 days; and transitional milk, over 4th day). CAT and GSH values were determined by the spectrophotometric method. Cu, Zn, Mg, Fe and Cd concentrations were determined by atomie absorption spectrophotometry (AAS). The results indicate no significant differences between activity of catalase taking into account the age of mothers and the day of lactation. Similarly glutathione level showed insignificant differences between sludicd groups. Our results indicate that human milk of women from Małopolska province, Poland, may contain significant amount of cadmium. We found statistically significant correlation between some metals in milk from women in different age and period of lactation. It concerned: Fe vs Cd and Cd vs Cu correlation. The result of our study indicate that the actiyity of antioxidant enzymes did not change during the course of lactation and in relation to mother's age.
Celem badań było określenie koncentracji metali ciężkich, a także zawartości enzymów antyoksy-dacyjnych: katalazy (CAT) i zredukowanego glutationu (GSH) w mleku ludzkim. Kobiety biorące udział w eksperymencie były rekrutowane spośród matek w różnych okresach laktacji, pochodzących z terenu województwa małopolskiego. Próbki mleka zostały pobrane od 23 zdrowych kobiet z klas wiekowych: 20-25, 26-31, 32-37 lat. Dodatkowo został dokonany podział na grupy ze względu na okres laktacji, pierwsza: 1-3 dzień laktacji - siara, druga: powyżej 4 dnia - mleko przejściowe. Aktywność katalazy była mierzona spektrofotometrycznie według metody Bartosza (1995), natomiast poziom glutationu oznaczano spektrofotometrycznie przy użyciu metody Ellmana (1965). Koncentrację pierwiastków, takich jak: Zn, Mg, Cu, Fe oraz Cd oznaczano za pomocą spektrofotometrii absorpcyjnej (AAS). Aktywność katalazy, a także poziom glutationu zredukowanego nie wykazały statystycznie istotnych różnic pomiędzy grupami wiekowymi oraz okresem laktacji. Wyniki naszych badań pokazują, że mleko kobiet z regionu Małopolski zawiera pewne ilości kadmu. Dodatkowo zostały wykazane statystycznie istotne korelacje pomiędzy zawartością metali w próbkach mleka od kobiet w różnym wieku i okresie karmienia, dotyczyło to związku pomiędzy: Fe i Cd oraz Cd i Cu. Pomiędzy pozostałymi badanymi pierwiastki nie zaobserwowano statystycznie istotnych korelacji.

The purpose of the present study was to evaluate the effects of soybean selenium proteinate on Se tissue retention and meat quality in pigs. In group A (n=11) the mixtures were supplemented with soybean selenium proteinate, in group B (n=11) with sodium selenite and in group C (n=11) with Se-enriched yeast (0.3 mg Se per kg in all groups). The use of soybean selenium proteinate resulted in lower retention of Se in tissues (liver, heart, muscle) compared to Se-enriched yeast. Selenium concentrations in tissues achieved by soybean selenium proteinate and sodium selenite were comparable. No differences in serum Se, serum GSH-Px and meat quality traits were found among the groups.

Microsomal glutathione transferase 1 (MGST1) possesses glutathione transferase and peroxidase activities and is active in biotransformation of xenobiotics and in defense against oxidative stress. To assess MGST1 role in the development and functioning of PC12 cells, we constructed a cell line with reduced MGST1 (PC12_M). Real-time PCR and immunoblot assays showed MGST1 expression lowered to 60 % and immunocytochemical analyses demonstrated an altered concentration and distribution of the enzyme. PC12_M cells revealed a larger tendency to grow in clusters, weaker adhesion, irregular shape of bodies, short neurite outgrowth and higher percentage of necrotic cells (34 %). The total GSTs activity determined with non-specific substrate CDNB (1-chloro-2,4-dinitrobenzene) decreased by 15-20 %, whereas that with DCNB (2,4-dichloro-1-nitrobenzene), a substrate more specific for cytosolic GSTs, was similar to the one in control cells. This suggests that reduction of MGST1 cannot be compensated by other glutathione transferases. In PC12_M cells the total glutathione content was higher by 15-20 %, whereas the GSSG/GSH ratio was lower than in control cells. Moreover, the laminin-dependent migration rate was much faster in control cells than in PC12_M, suggesting some alterations in the metastatic potential of the line with suppressed MGST1. The amount of MAP kinases (p38, JNK, ERK1/2) was elevated in PC12_M cells but their phosphorylation level declined. Microarray analysis showed changed expression of several genes, which may be linked with differentiation and necrosis of PC12_M cells. Our data suggest that MGST1 could be an important regulator of PC12 cells development and might have significant effects on cell growth and proliferation, probably through altered expression of genes with different biological function.

In this study we have evaluated the heavy metals accumulation in the leaves of Silene vulgaris and its influence on glutathione, free amino acids and anthocyanins concentrations. Glutathione is a tripeptide (gamma-Glu-Cys-Gly) which is involved in many metabolic processes of a plant cell. It is related to the sequestration of xenobiotics and heavy metals and it is also an essential component of the antioxidant system, which keeps reactive oxygen species under control. The accumulation of anthocyanin pigments in the leaves can be induced by environmental and anthropogenic stressors, such as pollution, osmotic stress, and nutrients deficiency. The positive role of anthocyanins as well as amino acids and organic acids in metal sequestration is suggested. Samples of Silene vulgaris leaves were collected in May and July 2003-2005 from the heavy metals contaminated sites - from the nearest vicinity of non-ferrous metal smelter Szopienice at the distance of 50, 250, 450 m, from zinc wastes heap in Katowice and from a former calamine site in Dąbrowa Górnicza (the South of Poland). In the previous study we noted that the nearest vicinity of non-ferrous metal smelter Szopienice was more polluted than the other areas. The highest Zn, Cd and Pb accumulation in the leaves of Silene vulgaris was noted in the plants collected in the nearest vicinity of the non-ferrous metal smelter Szopienice. The free amino acids concentrations have differed in the vegetative season. It was higher in the most polluted sites (50 and 250 m) in July. Higher content of the reduced form of glutathione was estimated in the plant leaves collected in the distance of 250 m from the emitter in July than at the beginning of the study. Also the highest anthocyanins accumulation in Silene vulgaris leaves was noticed in July and in plants from we most polluted area (250 m). The estimations, especially glutathione content, seem to be promising in ecophysiological research connected with heavy metals stress in plants growing in the postindustrial areas.
Przeprowadzono badania nad akumulacją Zn, Pb i Cd w liściach Silene vulgaris i wpływie tych metali na zawartość glutationu, wolnych aminokwasów i antocyjanów. Glutation jest tripeptydem (gamma-Glu-Cys-Gly). który jest zaangażowany w obronę przed ksenobiotykami i metalami ciężkimi i jest podstawowym komponentem komórkowej antyoksydacyjnej obrony, utrzymującym reaktywne formy tlenu (ROS) pod kontrolą. Akumulacja barwników antocyjanowych może być powodowana przez środowiskowe i antropogenne czynniki stresowe, takie jak zanieczyszczenie, stres osmotyczny, niedobór składników odżywczych . Sugeruje się pozytywną rolę antocyjan obok, wolnych aminokwasów i kwasów organicznych w oddzielaniu metali ciężkich. Liście roślin Silene vulgaris zbierano w maju i lipcu 2003-2005 z terenów zanieczyszczonych metalami ciężkimi (z najbliższego sąsiedztwa Huty Metali Nieżelaznych "Szopienice" w odległości 50, 250, 450 m, z hałdy pocynkowej w Katowicach Wełnowcu oraz terenu po eksploatacji galmanu w Dąbrowie Górniczej). W poprzednich badaniach wykazano największe zanieczyszczenie metalami ciężkich na terenie najbliżej położonym przy Hucie Metali Nieżelaznych "Szopienice". Największą akumulację Zn, Cd, Pb stwierdzono w liściach roślin Silene vulgaris zbieranych w najbliższym sąsiedztwie emitora (50, 250 m). Stężenie wolnych aminokwasów różniło się podczas sezonu wegetacyjnego. Zawartości były większe na najbardziej zanieczyszczonym terenie w lipcu niż na początku badań. Większą zawartość zredukowanej formy glutationu oznaczono w liściach roślin zbieranych w odległości 250 m od emitora w lipcu niż na początku badań. Zanotowano także największą akumulację antocyjanów w liściach roślin w lipcu i u roślin z najbardziej zanieczyszczonego terenu (250 m). Szczególnie oznaczanie zawartości glutationu wydaje się być obiecujące w ekofizjologicznych badaniach nad stresem wywoływanym przez metale ciężkie u roślin na terenach poprzemysłowych.

Wstęp: Wolne rodniki to atomy, cząsteczki lub ich fragmenty, których nadmiar prowadzi do rozwoju stresu oksydacyjnego, będącego przyczyną wielu chorób, m.in. nowotworowych, neurodegeneracyjnych i zapalnych, oraz starzenia się organizmu. Głównymi egzogennymi źródłami wolnych rodników są m.in. zanieczyszczenia przemysłowe, dym tytoniowy, promieniowanie jonizujące, ultradźwięki i pole magnetyczne. Pole magnetyczne niskiej częstotliwości powszechnie stosowane jest w terapii fizykalnej. Celem pracy była ocena wpływu pola magnetycznego niskiej częstotliwości (extremely low frequency magnetic field - ELF-MF), stosowanego w magnetoterapii, na zawartość glutationu całkowitego, utlenionego i zredukowanego oraz stanu redoks w komórce mięśnia poprzecznie prążkowanego w zależności od czasu działania pola. Materiał i metody: Materiał badawczy stanowiły szczury, samce o wadze 280-300 g. Zwierzęta podzielono na 3 grupy badawcze: I - grupa kontrolna, II grupa - poddana 30 min/dzień przez 2 tygodnie działaniu pola magnetycznego o parametrach: 7 mT i 40 Hz, III grupa - poddana 60 min/dzień przez 2 tygodnie działaniu pola magnetycznego o parametrach: 7 mT i 40 Hz. U zwierząt oznaczono glutation całkowity, utleniony i zredukowany oraz stan redoks w homogenatach tkanki mięśniowej po zakończeniu ekspozycji na działanie pola magnetycznego. Wyniki: Eksponowanie szczurów 30 min/dzień i 60 min/dzień przez 2 tygodnie na pole magnetyczne o parametrach 40 Hz i 7 mT powodowało istotny wzrost stężenia glutationu całkowitego w mięśniu poprzecznie prążkowanym w porównaniu z wynikami grupy kontrolnej (p < 0,001). Wnioski: Uzyskane wyniki sugerują, że glutation odgrywa ważną rolę w ochronie tkanek przed polem magnetycznym. Ekspozycja na pole magnetyczne stosowane w magnetoterapii prowadzi do rozwoju mechanizmów adaptacyjnych, których celem jest utrzymanie równowagi oksydacyjno-redukcyjnej organizmu, i zależy od czasu ekspozycji. Background: Free radicals (FR) are atoms, molecules or their fragments. Their excess leads to the development of oxidizing stress, the cause of many neoplastic, neurodegenerative and inflammatory diseases, and aging of the organism. Industrial pollution, tobacco smoke, ionizing radiation, ultrasound and magnetic field are the major FR exogenous sources. The low frequency magnetic field is still more commonly applied in the physical therapy. The aim of the presented study was to evaluate the effect of extremely low frequency magnetic field used in the magnetotherapy on the level of total glutathione, oxidized and reduced, and the redox state of the skeletal muscle cells, depending on the duration of exposure to magnetic field. Material and Methods: The male rats, weight of 280-300 g, were randomly devided into 3 experimental groups: controls (group I) and treatment groups exposed to extremely low frequency magnetic field (ELF-MF) (group II exposed to 40 Hz, 7 mT for 0.5 h/day for 14 days and group III exposed to 40 Hz, 7 mT for 1 h/day for 14 days). Control rats were kept in a separate room not exposed to extremely low frequency magnetic field. Immediately after the last exposure, part of muscles was taken under pentobarbital anesthesia. Total glutathione, oxidizedand reduced, and the redox state in the muscle tissue of animals were determined after exposure to magnetic fields. Results: Exposure to low magnetic field: 40 Hz, 7 mT for 30 min/day and 60 min/day for 2 weeks significantly increased the total glutathione levels in the skeletal muscle compared to the control group (p < 0.001). Conclusions: Exposure to magnetic fields used in the magnetic therapy plays an important role in the development of adaptive mechanisms responsible for maintaining the oxidation-reduction balance in the body and depends on exposure duration. Opis:
Pseudomonas aeruginosa is the most common cause of chronic and recurrent lung infections in patients with cystic fibrosis (CF) whose sputa contain copious quantities of P. aeruginosa toxin, pyocyanin. Pyocyanin triggers tissue damage mainly by its redox cycling and induction of reactive oxygen species (ROS). The reactions between reduced glutathione (GSH) and pyocyanin were observed using absorption spectra from spectrophotometry and the reaction products analysed by nuclear magnetic resonance imaging. Pyocyanin reacted with GSH non-enzymatically at 37°C resulting in the production of red-brown products, spectophotometrically visible as a 480 nm maximum absorption peak after 24 h of incubation. The reaction was concentration-dependent on reduced glutathione but not on pyocyanin. Minimizing the accessibility of oxygen to the reaction decreased its rate. The anti-oxidant enzyme catalase circumvented the reaction. Proton-NMR analysis demonstrated the persistence of the original aromatic ring and the methyl-group of pyocyanin in the red-brown products. Anti-oxidant agents having thiol groups produced similar spectophotometrically visible peaks. The presence of a previously unidentified non-enzymatic GSH-dependent metabolic pathway for pyocyanin has thus been identified. The reaction between pyocyanin and GSH is concentration-, time-, and O2-dependent. The formation of H2O2 as an intermediate and the thiol group in GSH seem to be important in this reaction.

Plant glutathione peroxidases are an important class of enzymes which play key roles in the stress adaptability of plants both in context of biotic and abiotic stress pathways. They have been over the years much studied in animals since the catalytic residues are comprised of selenocysteine a variant amino acid which is ribosomally encoded with the help of an RNA structural element known as SECIS. Various workers over the years have shown that plant glutathione peroxidases play active roles in ROS sequestration, lipid hydroperoxidation as well as regulate glutathione levels. However, each plant has various patterns of glutathione peroxidase expression and action and in some plants certain isoforms have not been detected at all. This work focuses on the prediction and identification of single nucleotide polymorphisms (SNPs) and INDELs in the coding regions of plant glutathione peroxidases, with the help of a Bayesian based algorithm subsequently validated. A large number of informative sites were detected 279 of which had variant frequency of ≥ 50 %. This data should be beneficial for future studies involving genetic manipulation and population based breeding experiments.

Understanding of the widespread biological importance of carotenoids is increasing. Accompanying this is the developing recognition that the interaction of carotenoids with other molecules, such as proteins, is also essential. Here the significance of carotenoid-protein interactions with respect to biological function is reviewed for three well characterised carotenoprotein complexes; crustacyanin, the orange carotenoid protein and glutathione-S-transferase P1. In addition a preliminary report is made on the recent partial purification of an echinenone-binding protein extracted from a New Zealand sea urchin, Evechinus chloroticus.

Earlier studies from our laboratories have shown that RLIP76, a previously described Ral-binding GTPase activating protein (Jullien-Flores et al., 1995, J. Biol. Chem. 270: 22473), is identical with the xenobiotic transporter DNP-SG ATPase, and can catalyze ATP-dependent transport of glutathione-conjugates as well as doxorubin (Awasthi et al., 2000, Biochemistry, 39: 9327). We have now reconstituted purified bacterially expressed RLIP76 in proteoliposomes, and have studied ATP-dependent uptake of the glutathione conjugate of 4-hydroxynonenal (GS-HNE) by these vesicles. Results of these studies show that RLIP76 reconstituted in proteoliposomes catalyzes ATP-dependent transport of GS-HNE against a concentration gradient. The transport of GS-HNE is saturable with respect to ATP as well as GS-HNE with Km values of 1.4 mM and 2.5 μM, respectively. These studies demonstrate that RLIP76 mediates active transport of GS-HNE, and are consistent with our previous work showing that RLIP76-mediated efflux of GS-HNE regulates the intracellular concentration of 4-HNE and thereby affects 4-HNE mediated signaling.

These facts lead to their increasing popularity in the human diet. Numerous studies suggest that their regular consumption has a positive effect on health and the well-being of individuals. Another bioactive food components found in cherries are enzymes. The interest in research about enzymes in cherries is not so significant as for other compounds like polyphenols or vitamins. However, number of studies were carried out to characterise enzymes and their function in cherries especially with relation to extending their shelf life. The aim of this work is to give a brief overview of latest research on browning enzymes, softening enzymes and glutathione S-transferase.

The study was designed to determine the possible relationship between Brassica oleracea var. italica seedlings stored at 2°C in the dark for seven and fourteen days, respectively, and the level of certain antioxidant parameters in particular organs. A parallel objective of the experiment was to determine if the reaction of seedlings to low temperature might be persistent in fully developed plants until harvest time. After 14 days of chilling a significant increase in the glutathione content was observed in the seedling leaves in comparison to the non-chilled plants. During vegetation in field conditions this effect was maintained in leaves up to the stage of formation of flower buds. At harvest the highest content of glutathione was demonstrated in broccoli heads, obtained from plants, which were previously chilled in the seedling phase for two weeks. Peroxidase activity in broccoli seedlings increased each year of the three-year study due to the duration of the cooling time, whereas in the case of catalase the changes were not so distinct. At harvest time the activity of both enzymes in the leaves and flower buds fluctuated according to the particular year of study.

Celem niniejszej pracy było określenie, czy krenoterapia (kuracja pitna) za pomocą wód siarczkowo- siarkowodorowych słonych (WSSS) pochodzących z ujęcia "Zuzanna" z okolic Buska-Zdroju ma wpływ na zmianę stężenia glutationu (GSH i GSSG) we krwi pełnej. WSSS zawierają nie mniej niż 1 g związków siarki ogólnej w kilogramie wody leczniczej, a ich działanie zależy nie tylko od zawartości związków siarki, ale także od rodzaju i zawartości innych biopierwiastków. Liczne badania potwierdzają korzystny wpływ H2S na parametry antyoksydacyjne organizmu. W analizie stężenia GSH i GSSG we krwi wykorzystano metodę elektroforezy kapilarnej z detektorem UV. Badaniu poddano grupę 40 ochotników, zarówno kobiet jak i mężczyzn, w różnych przedziałach wiekowych. Kuracja za pomocą WSSS trwała 2 tygodnie. Otrzymane wyniki badań potwierdzają, że H2S występujący w WSSS zwiększa stężenie glutationu we krwi, a także dają uzasadnienie dla wykorzystywania krenoterapii w lecznictwie.
The objective of the study was to agree whether crenotherapy (drinking therapy) with sulfide/hydrogen sulfide (SHS) waters from "Zuzanna" spring located in the area of Busko-Zdrój leads to increasing of glutathione (GSH and GSSG) content in human blood. SHS waters contain at least 1 g of total sulfur per kilogram of water and a treatment effect also depends on other bioelements. A lot of earlier experiments confirmed positive influence of H2S on antioxidative properties of organism. The method employing capillary electrophoresis with UV detector for the analysis of glutathione in human blood was developed. The group of 40 volunteers consisted of both women and men, in different age range. The therapy with SHS waters lasted 2 weeks. We recently demonstrated that the administration of H2S in SHS waters increases GSH concentration in blood, and therefore crenotherapy could be used in therapeutics.

The aim of the study was the electrophoretical identification of molecular forms of selected antioxidant enzymes in dog semen. Ejaculates to be studied were chosen from five dogs, aged from two to eight years. Polyacrylamide gel electrophoresis was carried out under non-denaturing conditions and then gels were stained for the activity of the following enzymes: superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPx). Sperm homogenates and all fractions (pre-spermatic, spermatic and post-spermatic) of dog ejaculate demonstrated one protein band with SOD activity characterized by low electrophoretic mobility. Based on the confirmed sensitivity to H₂O₂, it can be assumed that the detected SOD is an enzyme containing ions of Zn²⁺ and Cu²⁺ (Cu,Zn SOD). In sperm homogenates one protein band with GPx activity was characterized by high electrophoretic mobility, whereas in the spermatic and post-spermatic fractions of dog ejaculate three protein bands with different (low, medium and high) electrophoretic mobility were identified. CAT molecular forms were not found in either sperm homogenates or in the analyzed fractions of ejaculate.

Nanotechnology has been widely applied in agriculture, and understanding of the mechanisms of plant interaction with nanoparticles (NPs) as environmental contaminants is important. The aim of this study was to determine the effects of foliar application of cobalt oxide (Co 3O4) NPs on some morpho-physiological and biochemical changes of canola (Brassica napus L.) leaves. Seeds were sown in plastic pots and grown under controlled conditions. Fourteen-day-old seedlings were sprayed with different concentrations of Co3O4 NPs (0, 50, 100, 250, 500, 1000, 2000, and 4000 mg L -1 ) at weekly intervals for 5 weeks. Growth parameters of the shoot (length, fresh and dry weights) were stimulated by low concentrations of Co 3O4 NPs (50 and 100 mg L -1 ) and repressed by higher con- centrations. Similar trends were observed in photosynthetic pigment contents. The results indicated that high concentrations of Co3O4 NPs increased lipoxygenase (LOX) activity and the malondialdehyde (MDA), hydrogen per- oxide (H 2O2), and dehydroascorbate (DHA) contents, but reduced the membrane stability index (MSI), ascorbate (ASC), and glutathione (GSH). Despite the increase of antioxidant capacity (DPPH) and the accumulation of non- enzymatic antioxidants (total flavonoids and flavonols) and osmolytes (proline, glycine betaine (GB) and soluble sugars) at high concentrations of Co 3O4 NPs, the growth and photosynthesis were reduced. The defence system activity did not seem to be sufficient to detoxify reactive oxygen species (ROS). Altogether, high concentrations of Co3O4 NPs showed a phytotoxic potential for canola as an oilseed crop.

The present work was aimed to obtain information about age-dependent changes of γ-glutamyltransferase (GGT) activity and the levels of non-protein sulfhydryl compounds (NPSH) in rat kidneys. In addition, protein-bound cysteine (PB-Cys), sulfane sulfur compounds and reactive oxygen species (ROS) were estimated The results indicate that the activity of GGT and NPSH levels in the kidneys are reduced with age. At the same time, a significant increase in the level of protein-bound cysteine was observed. Simultaneously, the content of sulfane sulfur compounds was increased in the group of the oldest animals. These findings indicate that the capacity for extracellular glutathione degradation and, in consequence, the availability of cysteine for intracellular glutathione biosynthesis may be impaired. The increased PB-Cys level indicates potentiation of the thiolation reaction, i.e. development of protein-mixed disulfides. These results reveal age dependent disturbances in the thiol-disulfide equilibrium in the kidneys which leads to an imbalance between pro- and antioxidatory processes.

The level of mineral utilization depends on many factors related to animals as well as to the chemical form of given nutrients. It has been experimentally demonstrated that animals utilize inorganic forms of minerals less efficiently than organic ones. Hence, an attempt has been made to evaluate the bioavailability of selenium bound in different compounds to lambs. Selenium supplementation in fodder mixtures was another aspect included in our tests. Thus, an experiment was conducted on 48 growing lambs, testing the level and chemical forms of selenium in fodder mixtures for animals. In group I (the control) lambs received fodder mixture without any selenium supplement. In the experimental groups, selenium was supplemented as sodium selenite in the amount 0.2 mg Se kg-1 d.m. feeds (II), or selenium enriched yeast (Se-yeast) in the amounts of 0.1 and 0.2 mg Se kg-1 d.m. of feed groups III and IV, respectively. At the end of the experiment, when animals had reached 35 kilos of weight, blood samples were taken. The activity of glutathione peroxidase was estimated in heparinized blood samples. Eight lambs were chosen from each group and killed to collect samples of soft tissues (liver, kidney, brain, muscle). The content of selenium was determined in the tissue samples. Supplementation of feeds for lambs with selenium, both inorganic (sodium selenite) and organic (Se-yeast), increased the content of the element in soft tissues of animals. The highest level of the element was found in the liver and kidneys: 4.65 and 4.90 and 2.10 and 2.30 mg kg-1 fresh tissue, of the lambs receiving sodium selenine and Se-yeast in the amount 0.2 mg Se kg-1 D.M. of feed (groups II and IV), respectively. Selenium compounds added to feeds given to lambs significantly (P£0.01) increased activity of GSH-Px in blood, especially in the case of animals which received yeast enriched with selenium. In blood of the lambs which received feeds with Se-yeast in the amount 0.2 mg Se kg-1 d.m. of feed (group IV), the activity of glutathione peroxidase was by 129.71 U gHb-1 higher, and of the animals which received Se-yeast in the amount 0.1 mg kg-1 d.m. or sodium selenite, the activity of the enzyme was higher by 86.33 and 86.35 U gHb-1, respectively, than the activity of this enzyme in blood of animals from the control group. Supplementation of lambs’ rations with Se in the form of selenite or yeast enriched with selenium forms increased the content of Se in soft tissues and gluthatione peroxidase activity in comparison with animals which did not receive additional doses of this nutrient in fodder mixtures. The availability of Se was more profoundly affected by the amount of the element added rather than its form.
Stopień wykorzystania składników mineralnych z paszy zależy od wielu czynników związanych zarówno ze zwierzęciem, jak i z formą chemiczną stosowanych związków. W badaniach wskazano, że związki nieorganiczne są najczęściej gorzej wykorzystywane niż ich organiczne odpowiedniki. Dlatego podjęto próbę oceny przyswajalności przez jagnięta różnych związków selenu, uwzględniając także poziom dodatku tego pierwiastka w mieszankach. W badaniach na 48 rosnących jagniętach czynnikiem różnicującym poszczególne grupy były różne poziomy i formy połączeń chemicznych selenu w mieszankach treściwych. W grupie I (kontrolnej) jagnięta otrzymywały mieszankę bez dodatku selenu. W grupach (II) doświadczalnych mieszanki uzupełniano seleninem sodu w ilości 0.2 mg Se kg-1 s.m. paszy, a w grupach III i IV – drożdżami wzbogaconymi w selen (drożdże-Se), odpowiednio w ilości: 0.1 i 0.2 mg Se kg-1 s.m. paszy. Na końcu doświadczenia, po osiągnięciu przez zwierzęta masy ciała ok. 35 kg, pobrano od nich krew. Następnie w pełnej heparynizowanej krwi oznaczono aktywność peroksydazy glutationowej (GSH-Px). Z każdej grupy wybrano losowo po 8 jagniąt, zwierzęta ubito i pobrano tkanki miękkie (wątroba, nerki, mózg, mięsień przywodziciel uda). W zliofilizowanym materiale biologicznym oznaczono zawartość selenu. Podane jagniętom w mieszankach treściwych zarówno nieorganiczne (selenin sodu), jak i organiczne (drożdże-Se) formy połączeń selenu zwiększyły zawartość tego pierwiastka w tkankach miękkich jagniąt. Najwięcej selenu znajdowało się w wątrobie i nerkach, odpowiednio 4.65 i 4.90 oraz 2.10 i 2.30 mg kg-1 świeżej tkanki jagniąt otrzymujących w paszy selenin sodu i drożdże–Se w ilości 0.2 mg Se kg-1 s.m. paszy (gr. II i IV). Stosowane w paszy jagniąt związki selenu wyraźnie (P≤0.01) zwiększyły aktywności GSH-Px we krwi owiec, zwłaszcza u zwierząt, które w mieszance otrzymywały drożdże wzbogacone w selen. U owiec otrzymujących w paszy drożdże-Se w ilości 0.2 mg Se kg-1 s.m. paszy (grupa IV) wykazano wyższą o 129.71 U gHb-1 aktywność peroksydazy glutationowej, a u zwierząt, którym w paszy podawano drożdże-Se w ilości 0.1 mg Se kg-1 s.m. paszy lub selenin sodu, aktywność tego enzymu we krwi była wyższa odpowiednio o 86.33 i 86.35 U gHb-1 w porównaniu z grupą kontrolną. Zastosowanie w dawkach dla jagniąt dodatku Se, zarówno w formie seleninu, jak i wzbogacanych drożdży, spowodowało wzrost jego zawartości w tkankach miękkich oraz wzrost aktywności peroksydazy glutationowej w porównaniu ze zwierzętami z grupy kontrolnej. Większy wpływ na dostępność Se miał jego poziom w dawce niż stosowana forma.

Robinia pseudoaccacia plants grown hydroponically were treated Pb(NO3)2 with 15, 45 mg Pb²⁺ ∙ dm⁻³. After 6, 12, 24, 72 hours of the metal treatment the plants were collected and dissected organs. The plants accumulated and transported to ground part 0.88% and 1.35% of total accumulated lead for the lower and higher dose of Pb²⁺ respectively. The level of GSH was differed and depended on organs, dose and time treatment of Pb²⁺. We investigate (different pattern of expression) expression of RpGSH1 and RpPCS genes in roots. The study showed that glutathione and genes encoded enzymes connected with synthesis of him, plays important role in the process of detoxification in plant.
Rośliny robinii akacjowej uprawianej w hydroponice traktowane były Pb(NO3)2 w ilościach: 0, 15, 45 mg Pb²⁺ ∙ dm⁻³. Materiał roślinny zbierano po 6, 12, 24 i 72 godzinach od traktowania ołowiem i rozdzielono na organy. Procent pobranego ołowiu w przeliczeniu na suchą masę roślin przetransportowany do części nadziemnej wynosił 0,88 dla najwyższej dawki traktowania oraz 1,35 dla niższej. Poziom glutationu był zróżnicowany i zależał od badanego organu, dawki i czasu od potraktowania ołowiem. Odnotowano zróżnicowaną ekspresję RpGSH1 i RpPCS w korzeniach roślin. Badania wykazały, że glutation i geny szlaku jego biosyntezy odgrywają ważną rolę w procesie detoksykacji ołowiu w roślinie.

Opis:
The concentration of non-enzymatic antioxidants such as ascorbate and glutathione in tissues is one of the major plant responses to biotic and many abiotic stresses, including metals. Therefore, it is crucial to develop the most effective methods for simultaneous quantitative analysis of these antioxidants. Capillary zone electrophoresis allows relatively fast and effective analysis. The aim of the paper was to apply and optimise the capillary electrophoresis conditions for simultaneous determination of glutathione, glutathione disulphide, ascorbate, and dehydroascorbate in small plant tissue samples exposed to copper and cadmium. The method ensures good linearity and reproducibility, with correlation coefficients 0.988 for ascorbate and 0.999 for glutathione and glutathione disulphide, and with detection limits approximately 2.50, 0.65 and 0.50 ppm, respectively. Cu stress was found to increase the ascorbate concentration and glutathione content in leaves, while Cd increased glutathione in the oldest leaf segments and root.
Zawartość nieenzymatycznych antyoksydantów takich jak kwas askorbinowy oraz glutation w roślinach jest jedną z głównych form odpowiedzi roślin na biotyczne i abiotyczne czynniki stresowe, w tym na metale ciężkie. Dlatego też niezbędne jest rozwijanie efektywnych metod jednoczesnego ilościowego oznaczania tych antyoksydantów. Elektroforeza kapilarna pozwala na takie relatywnie szybkie i efektywne analizy. Celem pracy było zastosowanie oraz optymalizacja metody jednoczesnego pomiaru zawartości glutationu, disiarczku glutationu, kwasu askorbinowego oraz kwasu dehydroaskorbinowego w małych próbkach roślinnych z użyciem elektroforezy kapilarnej. Opracowana metoda zapewnia dobrą liniowość i powtarzalność ze współczynnikami korelacji na poziomie 0,988 dla kwasu askorbinowego oraz 0,999 dla glutationu i disiarczku glutationu, z limitem detekcji odpowiednio 2,50; 0,65 oraz 0,50 ppm. Stwierdzono, że stres indukowany nadmiarem Cu powodował wzrost zawartości kwasu askorbinowego i glutationu w liściach, podczas gdy po zastosowaniu Cd obserwowano wzrost zawartości glutationu w starszej części liścia i korzeniu.

The high level of cadmium in the abdominal muscle of the brown shrimp Crangon crangon is due to the serious pollution of the water in the Gulf of Gdańsk. The inhibition of malic enzyme (ME) activity by cadmium, and in consequence the reduced formation of NADPH, could interfere with cellular mechanisms for detoxifying the organism and reducing oxidative stress. The reduced glutathione (GSH) concentration in the abdominal muscle of C. crangon was calculated to be 5.8 mM. The objective of this study was to evaluate the part played by GSH in the effect of cadmium on the activity of NADP-dependent malic enzyme from abdominal muscles of brown shrimps. This enzyme is activated by certain divalent cations (Mg, Mn). The results demonstrate that cadmium inhibits ME activity from shrimp muscle, and that GSH and albumin can reduce this cadmium-inhibited NADP-dependent malic enzyme activity.

The cotton leafhopper (Amrasca biguttula biguttula Ishida) is considered to be an alarming insect pest causing both quantitative and qualitative loss in cotton. In situ bioassay studies were done and the role of detoxifying enzymes in conferring resistance to neonicotinoid groups of insecticides in low (MUD), medium (DVG), high (HVR) and very high (GLB) pesticide usage areas of Karnataka were determined. Bioassay studies showed that imidacloprid, thiamethoxam, acetamiprid, thiacloprid and clothianidin registered varying levels of resistance for all the locations studied. The resistance ratio was high in imidacloprid (3.35, 8.57, 9.15 and 12.27 fold respectively) and the lowest in dinoferuran (1.86, 5.13, 6.71 and 9.88 fold respectively). Furthermore, the enzyme activity ratio (glutathione-S-transferase) was relatively greater, and corresponded to the higher LC50 values of neonicotinoids for very high, high, medium and low pesticide usage areas. Our study suggested that the higher activity of the detoxifying enzyme in the resistance population of cotton leafhopper apparently has a significant role in endowing resistance to neonicotinoid groups of insecticides. However, this study recommends using neonicotinoids in cotton growing areas with caution.

Opis:
The study was carried out on two highbush blueberry cultivars: Bluecrop and Darrow in 2001 and 2002 seasons. Analyses for phytochemical contents in berries were made during commercial maturity of fruit. Concentration of anthocyanins and low molecular weight thiol compounds was significantly higher in ´Darrow´ berries in comparison to Bluecrop cv., irrespective of the harvest year. Tested cultivars did not differ in the ratio of the reduced form of glutathione in its total quantity. As opposed to this, the higher proportion of the reduced form of ascorbate in its total concentration was noted for ´Bluecrop´, but it was statistically proven only in the first year of research. A very low activity of antioxidative enzymes was noted, however on the average, considerably higher activity of glutathione reductase showed ´Darrow´, whereas ´Bluecrop´ had higher catalase activity. The ascorbate peroxidase activity was not detected in the extracts of ´Bluecrop´s’ berries. Berries of ´Darrow´, which exhibited, in general, the higher antioxidants content in the first year of the study, were harvested at two different dates in 2002: in the middle of July and at the end of August. The harvest date had a significant effect on the level of some phytochemicals. The fruit harvested in August had lower content of cysteine, glutathione, phenolics, flavonols and anthocyanins than the ones harvested in July. Ascorbate content was similar at both harvest dates. Growing season also had the influence on antioxidant properties, especially on the content of ascorbic acid.
Badania przeprowadzono w latach 2001–2002. Ocenie poddano dwie odmiany borówki wysokiej: Bluecrop i Darrow. Analizy chemiczne dotyczące zawartości fitozwiązków w owocach wykonano w fazie dojrzałości zbiorczej. Stężenie antocyjanów oraz niskocząsteczkowych związków tiolowych było istotnie wyższe w owocach odmiany Darrow w porównaniu z odmianą Bluecrop, niezależnie od roku badań. Udział zredukowanego glutationu w jego całkowitej zawartości kształtował się na zbliżonym poziomie u obu ocenianych odmian. Przeciwnie, istotnie wyższą zawartością zredukowanego askorbinianu charakteryzowała się odmiana Bluecrop, ale tylko w pierwszym roku badań. Aktywność enzymów oksydacyjnych w dojrzałych owocach borówek była bardzo niska. Jakkolwiek przeciętnie, wyższą aktywnością reduktazy glutationowej charakteryzowała się odmiana Darrow, natomiast ekstrakt z owoców odmiany Bluecrop wykazywał wyższą aktywność katalazy. Ekstrakty z owoców odmiany Bluecrop nie wykazywały aktywności peroksydazy askorbinianowej. Owoce odmiany Darrow, które charakteryzowały się na ogół wyższym stężeniem antyoksydantów w pierwszym roku badań, w roku kolejnym poddano analizie w dwóch terminach: w połowie lipca i końcu sierpnia. Termin zbioru wpłynął istotnie na poziom fitozwiózków. W owocach zebranych w sierpniu stwierdzono niższą zawartość cysteiny, glutationu, związków fenolowych ogółem jak i flawonoli oraz antocyjanów w porównaniu do zbioru wcześniejszego. Poziom askorbinianu był zbliżony, niezależnie od terminu zbioru. Stwierdzono wyraźny wpływ sezonu wegetacyjnego na poziom analizowanych antyoksydantów, szczególnie dotyczyło to stężenia askorbinianu.
