Trehalase (α, α-trehalose-1-C-glucohydrolase, EC 3.2.1.28) is a glycoside hydrolase enzyme that catalyzes the conversion of trehalose to glucose. Trehalose hydrolysis by trehalase enzyme is an important physiological process for various organisms, such as fungal spore germination, insect flight, and the resumption of growth in resting cells. So by studying biochemical properties of trehalase we can control the spore germination of harmful fungus, Apergillus niger. Till now trehalase has been reported from many other organisms including plants and animals, Trehalase has been purified and characterized from various organisms such as Saccharomyces cerevisiae, Lentinula edodes, Acidobacterium capsulatum, and Medicago sativa, as well as root nodules of Phaseolus vulgaris, soybean and from the from the seeds of Triticum aestivum. In this study, trehalase was purified from Aspergillus niger grown on cellulosic medium. And this purified trehalase was tested for various requirements it may need for enzymatic activity such as inorganic phosphate and Magnesium ions (Mg2+).

Papain activity was studied in water-organic solvent mixtures using the fluorogenic substrate Dabcyl-Lys-Phe-Gly-Gly-Ala-Ala-Edans. The increase of organic solvent (MeOH, EtOH, iPrOH, TFE, MeCN, (MeO)2Et and DMF) concentration in the mixture caused a substantial decrease the initial rate of papain-catalyzed hydrolysis. Moreover, the number of papain active sites decreased with the increase of DMF and MeOH concentration.

Applying pesticides to crops is one of the causes of water pollution by surface runoff, and chlorpyrifos, trifluralin and chlorothalonil are used respectively as insecticide, herbicide and fungicide for crop plants widely. To explore effects of three pesticides on aquatic organisms, superoxide dismutase (SOD) and glutathione S-transferase (GST) activities were determined after 24 h and 48 h exposure of D. magna with ages of 6–24 h to several low concentrations of chlorpyrifos (0.36, 0.72, 1.43, 2.86, 5.72 μg∙L−1), trifluralin (0.17, 0.33, 0.66, 1.33, 2.65 mg∙L−1) and chlorothalonil (0.09, 0.18, 0.36, 0.72, 1.43 mg∙L−1) respectively. Main reproductive parameters including first pregnancy time, first brood time, the number of first brood and total fecundity after 21 d exposures at the same concentrations of pesticides as described above were also measured. The results showed that the activities of GST increased in lower concentrations and decreased in higher concentrations after 24 h exposure to three pesticides, respectively. The activities of SOD showed the same changes after 48 h exposure. With the time prolonged, the activities of GST decreased while the activities of SOD increased. After 21 d exposure, the first pregnancy time and first brood time were delayed, while the number of the first brood and total fecundity per female decreased with increasing concentrations. These results corroborated that GST activity was more sensitive to those pesticides than SOD activity, and there was a significant relationship between total fecundity and pesticides-dose(r>0.94, n=6), GST activity after 48 h exposure and total fecundity after 21 d exposure (r>0.92, n=6).

The aim of the study was to determine the activity of dehydrogenases and urease in forest peat soils of different fertility. There were selected 23 experimental plots localised in central and northern Poland. The research was conducted on forest fens, transition bogs and raised bogs. The biggest differences in soil physical and chemical properties were detected between fen and raised bog soils while raised bog soils and transition bog soils differed the least. Statistically significant differences between particular subtypes of peat soils were observed for soil pH-H2O, pH-KCl, C/N ratio as well as the content of organic carbon, nitrogen, calcium and potassium. The highest average dehydrogenase activity in the soil surface level was observed in fen soils, and the lowest – in raised bogs soils. The results obtained on urease activity were similar. Differences in urease activity in the studied soil types were shown. Dehydrogenase activity did not reveal statistically significant diversity. The activity of urease was negatively correlated with the content of carbon, C/N ratio, hydrolytic acidity and moisture. Also the increase in enzymatic activity accompanied by the increase in pH has been observed.

Papain activity in a buffer containing Me2SO was studied using fluorogenic substrates. It was found that the number of active sites of papain decreases with increasing Me2SO concentration whereas the incubation time, in a buffer containing 3% Me2SO does not affect the number of active sites. However, an increase of papain incubation time in the buffer with 3% Me2SO decreased the initial rate of hydrolysis of Z-Phe-Arg-Amc as well as Dabcyl-Lys-Phe-Gly-Gly-Ala-Ala-Edans. Moreover, an increase of Me2SO concentration in working buffer decreased the initial rate of papain-catalysed hydrolysis of both substrates. A rapid decrease of the initial rate (by up to 30%) was observed between 1 and 2% Me2SO. Application of the Michaelis-Menten equation revealed that at the higher Me2SO concentrations the apparent values of kcat/Km decreased as a result of Km increase and kcat decrease. However, Me2SO changed the substrate binding process more effectively (Km) than the rate of catalysis kcat..

Taking into account the progressive degradation of soils it is important to assess their quality. Soil quality depends on a large number of physical, chemical, biological and biochemical properties. In the publications available, there are presented three approaches regarding the use of soil properties to estimate soil quality: (1) the use of individual properties, (2) the use of simple indexes and (3) the use of complex indexes derived from combinations of different properties. The purpose of this study was to investigate the possibility to use enzymes as indicators of forest soil quality. Experimental plots (43) were located in central Poland. The study was carried out in a number of diverse fresh forest sites. To assess the quality of forest soils dehydrogenase and urease activity and the degree of base saturation were used. One of the final conclusions point out that enzymatic activity indicates current site condition as well as the changes that occur in soil better than soil physical and chemical properties. In other words, in comparison to soil enzymatic activity, soil physico-chemical properties constitute a less sensitive indicator of soil changes.

Chloroplast protease AtDeg2 (an ATP-independent serine endopeptidase) is cytosolically synthesized as a precursor, which is imported into the chloroplast stroma and deprived of its transit peptide. Then the mature protein undergoes routing to its functional location at the stromal side of thylakoid membrane. In its linear structure AtDeg2 molecule contains the protease domain with catalytic triad (HDS) and two PDZ domains (PDZ1 and PDZ2). In vivo AtDeg2 most probably exists as a supposedly inactive haxamer, which may change its oligomeric stage to form active 12-mer, or 24-mer. AtDeg2 has recently been demonstrated to exhibit dual protease/chaperone function. This review is focused on the current awareness with regard to AtDeg2 structure and functional significance.

Heavy metals are a serious source of soil pollution, especially in the vicinity of metal smelters. His study examines the effect of selected heavy metals on the activity of soil enzymes (acid phosphatase, dehydrogenase, β-glucosidase, urease and protease) in topsoil (0–30 cm) in the vicinity of the zinc smelter “Miasteczko Slaskie”. Soil enzyme activity in the polluted site was compared with a non-polluted site in the outskirts of the Pazurek nature reserve (near Olkusz). Soil samples were collected in coniferous communities. The activity of soil enzymes were compared at distances of 0.5 km, 1 km and 1.5 km from the emitter. In the vicinity of the emitter (all distances) Cd, Pb and Zn levels in topsoil exceeded acceptable limits. Also, Pb content at a non-polluted site (Pazurek nature reserve) was higher than permissible levels. Zn (HNO 3 extracted) were the highest in the topsoil samples collected at a distance of 1.5 km from the Miasteczko Slaskie plant, Cd (HNO 3 and CaCl 2 extracted) and Pb (CaCl 2 extracted) contents were highest at the distance 1 km from the emitter. However, the pollution index in the superficial soil layer (0–10 cm) was similar at all three distances from the emmiter. The Zn, Cd and Pb bioavailable concentrations (CaCl 2 extracted) ranged: 15–220, 0.1–7.2, 2.8–55.7 mg kg –1 respectively. The greatest decreases were found for acid phosphatase, dehydrogenase and protease activity at the distance of 0.5 km from the emitter. Results show that soil enzymatic activity should accompany basic chemical analysis to track disorders and adverse effects in polluted soil.
Zanieczyszczenie gleby metalami ciężkim zwłaszcza w pobliżu hut metali to poważny problem. Oceniano wpływ metali ciężkich na aktywność enzymatyczną gleby. Celem badań była ocena aktywności enzymów glebowych: fosfatazy kwaśnej, dehydrogenaz, β-glukozydazy, ureazy i proteazy w wierzchniej warstwie gleby w najbliższym sąsiedztwie huty cynku Miasteczko Śląskie. Aktywność enzymatyczną gleby na stanowiskach zanieczyszczonych porównywano z aktywnością na stanowisku niezanieczyszczonym, próbki gleby pobierano również w pobliżu rezerwatu przyrody Pazurek koło Olkusza ze zbiorowisk borowych. Aktywność enzymatyczna w glebie najbliższego sąsiedztwa emitora była porównywana w odległości 0,5, 1, 1,5 km od emitora. Wykazane zawartości Cd, Pb i Zn przekraczały dopuszczalne normy przyjęte dla gleby. Zawartość Pb w próbkach gleby pobieranych w pobliżu rezerwatu Pazurek była wyższa niż dopuszczalne wartości dla gleby. Zawartość Zn (ekstrakcja HNO3) była najwyższa w odległości 1,5 km od emitora (0–10 cm), z kolei zawartości Cd (ekstrakcja HNO 3 i CaCl 2 ) i Pb (ekstrakcja CaCl 2 ) były najwyższe 1 km od emitora. Wskaźnik zanieczyszczenia wykazany dla wierzchniej warstwy gleby (0–10 cm) był na porównywalnym poziomie w glebie w sąsiedztwie emitora. Koncentracje biodostępnych form metali (ekstrakcja CaCl2) mieściły się w przedziałach odpowiednio: 15–220, 0,1–7,2, 2,8–55,7 mg kg –1 . Najniższe aktywności fosfatazy, dehydrogenazy i proteazy wykazano w próbkach gleby pobieranej w odległości 0.5 km. Badania aktywności enzymatycznej gleby obok analiz chemicznych powinny być prowadzone w celu śledzenia zmian w glebach zanieczyszczonych.

Farnesyl diphosphate synthase (FPPS) is a key enzyme in isoprenoid biosynthesis which supplies sesquiterpene precursors for several classes of essential metabolites including sterols, dolichols, ubiquinones and carotenoids as well as substrates for farnesylation and geranylgeranylation of proteins. It catalyzes the sequential head-to-tail condensation of two molecules of isopentenyl diphosphate with dimethylallyl diphosphate. The enzyme is a homodimer of subunits, typically having two aspartate-rich motifs with two sets of substrate binding sites for an allylic diphosphate and isopentenyl diphosphate per homodimer. The synthase amino-acid residues at the 4th and 5th positions before the first aspartate rich motif mainly determine product specificity. Hypothetically, type I (eukaryotic) and type II (eubacterial) FPPSs evolved from archeal geranylgeranyl diphosphate synthase by substitutions in the chain length determination region. FPPS belongs to enzymes encoded by gene families. In plants this offers the possibility of differential regulation in response to environmental changes or to herbivore or pathogen attack.

Opis:
The use of sewage sludge in agriculture is known to be environmentally friendly as it is a practical way of recycling municipal waste. A short-term experiment was carried out to study the changes in soil chemical and microbiological properties influenced through the addition of sewage sludge along with saline irrigation under a pearl millet-wheat crop rotation. There were three irrigation (canal water (0.35 dS m-1), 8 and 10 dS m-1 electrical conductivity saline water) and five fertilizer treatments (control-F1, sewage sludge (5 t ha-1)-F2, sewage sludge (5 t ha-1) +50% recommended dose of fertilizer (RDF)-F3, sewage sludge (5 t ha-1)+75% RDF-F4 and RDF-F5). The results showed that soil organic carbon, and available nitrogen, and phosphorus were reduced significantly under saline conditions but there was an increase in available potassium with the increasing salinity levels of the irrigation water. A significant reduction in soil microbial biomass carbon and enzyme activities was caused by 8 and 10 dS m-1 as compared to 0.35 dS m-1 at different growth stages of crops. Treatment F4 attained the highest soil microbial activity at each crop growth stage by a significant margin among all of the fertilizer treatments, which is associated with a substantial build-up of organic carbon and available NPK in the soil. Soil microbial activities followed a particular trend: at 35 and 75 days after sowing>harvest>sowing in pearl millet and wheat, respectively. Sewage sludge dumping is a major problem in India, and the present study suggests that by applying sewage sludge, soil biological health is improved; therefore its use in agriculture is recommended to farmers.
