Clostridium perfringens (MTCC 1349) is a Gram-positive, anaerobic, endospore forming, and rod-shaped bacterium. This bacterium produces a variety of toxins under strict anaerobic environment. C. perfringens can grow at temperatures ranging between 20°C and 50°C. It is the major causetive agent for gas gangrene, cellulitis, septicemia, necrotic enteritis and food poisoning, which are common toxin induced conditions noted in human and animals. C. perfringens can produce produce four major types of toxins that are used for the classification of strains, classified under type A-E. Across the globe many countries, including the United States, are affected by C. perfringens food poisonings where it is ranked as one of the most common causes of food borne infections. To date, no direct one step assay for the detection of C. perfringens has been developed and only few methods are known for accurate detection of C. perfringens. Long detection and incubation time is the major consideration of these reporter assays. The prensent study proposes a rapid and reliable colorimetric assay for the detection of C. perfringens. In principale, this assay detects the para nitrophenyl (yellow colour end product) liberated due to the hydrolysis of paranitrophenyl phosphetidyl choline (PNPC) through phospholipase C (lecithinase). Constitutive secretion of phospholipase C is a charactristic feature of C. perfringens. This assay detects the presence of the extracellular lecithinse through the PNPC impragnated impregnated probe. The probe is impregnated with peranitrophenyl phosphotidyl choline ester, which is colourless substrate used by lecithinase. The designed assay is specific towards PNPC and detectes very small quantites of lecithinase under conditions used. The reaction is substrate specific, no cross reaction was observed upon incubation with other substrates. In addition, this assay gave negative results with other clostridium strains, no cross reactions were observed with other experimental strains like C. tetani, C. botulinum, C. acetobutyricum, Bacillus subtilis, and Escherichia coli. This assay is extramly rapid and provides reliable and reproducible results within one hour of incubation at 37°C.

Opis:
Ionic liquids (IL) are defined as compounds consisting only of ions with a melting point below 100°C. [1,2,4]. The first obtained ionic liquid was ethylammonium nitrate synthesized in 1914 by Paul Walden as a result of the protonation of ethylamine with nitric acid [4]. New group of IL are amino acid ionic liquids. They are non-toxic and bio- renewable ionic liquids of natural origin. The amino acids contain at least two functional groups, an amino group and a carboxyl group, which makes them both cationic and anionic in the amino acid ionic liquid. Amino acid ionic liquids attract more and more attention in the field of catalysis, as solvents in chemical transformations and cellulose solvents due to low production costs and their biodegradable. Compounds based on imidazolium, phosphonium, ammonium and choline ionic liquids found a wide application in biological sciences, modern chemistry and materials science [10]. One of their biggest advantages is the ability to absorb CO2 (especially their amino acid derivatives), which increases the chance of eliminating carbon dioxide from the environment. Amino acid ionic liquids have also found application in the field of catalysis.

Opis:
A reversible effect of pH on the ionization of amino-acid residues at the active center of choline oxidase was observed near the optimum pH (8). Inactivation of choline oxidase took place in the pH ranges 3-6 and 9-11, in which irreversible changes in the structure occur leading to the enzyme inactivation. The first order rate constants of the enzyme's inactivation at various pH values were estimated for the irreversible changes. The Arrhenius analysis revealed no significant changes in the activation enthalpy, while an increase in the activation entropy reflected an increase in the conformational freedom.

Opis:
Nematodes were found to synthesize phosphorylcholine-containing molecules not present in higher organisms, i.e. phosphorylcholine-substituted glycosphingolipids and (glyco)proteins. Investigations on the biosynthesis of these structures provided first biochemical evidence for the presence of the Kennedy and Bremer-Greenberg pathways in the model organism Caenorhabditis elegans.

The physicochemical properties of a deep eutectic solvent (DES) and thus its area of application are influenced by the nature salt or hydrogen bond acceptor (HBA), hydrogen bond donor (HBD), molar ratio, temperature and water content. The search for and discovery of new HBD would further widen the scope of DES versatility. In this study, the capacity of castor oil to serve as HBD and form DES with choline chloride (ChCl) was investigated. ChCl and castor oil were mixed in four molar ratios, namely 1:1, 1:2, 1:3, and 1:4. DES was formed at 1:3 and 1:4 molar ratios and code-named DES1 and DES2, respectively. The physicochemical properties of both DESs were measured as a function temperature from 313 K to 353 K. The results indicate that the densities of these castor oil-based DESs are uniquely low when compared to other DESs and ionic liquids. Their physicochemical properties generally follow the known trend for other DES, in terms of temperature variations. The viscosity, conductivity, surface tension and refractive index of castor oil based-based DES all fall within the known range for other choline chloride-based DESs. Based on its pH values, castor oil based DES is a Bronsted acidic deep eutectic solvent. This discovery offers significant potential to expand the possible combinations of salts and HBDs and further increase the areas of application of this solvent.

Two-component deep eutectic solvents (DES) based on choline dihydrogencitrate and glycerol or urea were tested as starch plasticizers. Thermal analysis techniques were applied to characterize the properties of starch/DES systems. The X-ray diffractometry measurements revealed a significant decrease in crystallinity indicating that used DES exhibited the ability to penetrate the ordered regions of potato starch, which is a necessary feature of a true plasticizer. However, the differential scanning calorimetry and dynamic thermal analysis results surprisingly showed an increase of Tg of starch materials indicating chemical crosslinking at elevated temperature. The eutectic solvents based on choline dihydrogencitrate could act as a plasticizer and a simultaneously crosslinking agent.

The aim of the study has been to assess the efficiency of choline-stabilized orthosilic acid (ch-OSA; bioavailabile form of silicon) application under increasing intensity of manganese stress on the chemical composition of plants and yielding of tomato (Lycopersicon esculentum Mill. cv. Alboney F1 and cv. Emotion F1). Plants were grown in rockwool with the application of a nutrient solution of the following chemical composition (mg dm-3): N-NH4 2.2, N-NO3 230, P 50, K 430, Ca 145, Mg 65, Cl 35, S-SO4 120, Fe 2.48, Zn 0.50, Cu 0.07; pH 5.50, EC 3.00 mS cm-1. The following manganese levels in the nutrient solution were tested (mg dm-3): 9.6 and 19.2. The effect of ch-OSA application (at a concentration of Si equal 0.3 mg dm-3 of the nutrient solution) was investigated at both Mn-levels. The ch-OSA application alleviated Mn toxicity by increasing the biomass production in the Mn-low variant (+ 8.2% for Alboney F1 and 16.8% for Emotion F1, the differences being significant for Emotion F1), whereas the ch-OSA application in the Mn-high variant did not influence the plant yielding. All the factors affected the plant nutrient status and the chemical composition of tomato fruits. The chemical composition of leaves depended on (means of all the studied combinations): Mn (K, Na) and ch-OSA nutrition (N, Mg, Na), cultivar (for P, K, Ca, Na), but in the case of fruits significant differences were found between Mn (N, P, Ca, Mg) and ch-OSA nutrition (N, Mg, Na) and cultivar (N, P). In both Mn-levels, visual symptoms of manganese toxicity appeared on plants – in the Mn-high variant, they were observed after 4 weeks, while in the case of the Mn-low variant - after 10 weeks of exposure to strong Mn-stress. Treatment with ch-OSA at both of the studied Mn levels did not prevent the development of visual toxicity symptoms on the plants.

Dokonano przeglądu potencjalnego wpływu wybranych składników żywności na aktywność umysłową oraz ryzyko wystąpienia dysforii, demencji i depresji. Przedstawiono dowody na kluczową rolę m.in. witamin z grupy B i witamin przeciwutleniających w ochronie przed niekorzystnymi zmianami w naczyniach krwionośnych, aterosklerozą i demencją. Wskazano równie na udział choliny, alkoholu i kofeiny w funkcjonowaniu centralnego układu nerwowego oraz diety na syntezę neuroaktywnego składnika – β-endorfiny.
In the paper presented, the authors review the potential impact of selected nutrients on mental activity, as well as the risk of dysphoria, dementia, and depression to occur in people. Evidence was provided that the B-complex and antioxidant vitamins play an important role in the protection against unfavorable changes in blood-vessels, as well as against arteriosclerosis and dementia diseases. It was demonstrated that such substances as choline, alcohol, and caffeine are significantly involved in the functioning of the central nerve system, and that a diet influences the synthesis of ß-endorphin, which is a neuro-active substance.
