In the course of infections caused by pathogenic yeasts from the genus Candida, the fungal cell surface is the first line of contact with the human host. As the surface-exposed proteins are the key players in these interactions, their identification can significantly contribute to discovering the mechanisms of pathogenesis of two emerging pathogens from this genus, C. parapsilosis and C. tropicalis. Therefore, the aim of the present study was to identify the cell wall-attached proteins of these two species with the use of cell surface shaving and a shotgun proteomic approach. Different morphological forms of C. parapsilosis and C. tropicalis cells obtained after growth under various conditions were subjected to this treatment. This allowed to indicate the most abundant cell surface proteins on the basis of the normalized spectral abundance factors. In case of yeast-like forms these were, among others, proteins similar to a chitinase, glyceraldehyde-3-phosphate dehydrogenase and an inducible acid phosphatase for C. parapsilosis, and a constitutive acid phosphatase, pyruvate decarboxylase and glyceraldehyde-3-phosphate dehydrogenase for C. tropicalis. In case of pseudohyphal forms, proteins similar to a cell surface mannoprotein Mp65, chitinase and glycosylphosphatidylinositol-anchored transglycosylase Crh11 were identified at the cell surface of C. parapsilosis. The Rbt1 cell wall protein, a hyphally regulated cell wall protein and proteins from agglutinin-like sequence protein family were found as the most abundant on C. tropicalis pseudohyphae. Apart from the abovementioned proteins, several additional covalently bound and atypical cell wall proteins were also identified. These results extend the current knowledge regarding the molecular basis of virulence of these two non-albicans Candida species.

Using ultrastructural cytochemical techniques we have found differences in the distribution of surface coat components between the invasive protozoan parasite Entamoeba histolytica and the non-invasive Entamoeba dispar. Carbohydrate-containing components and anionic sites in the cell surface of both species were detected by staining with ruthenium red and cationized ferritin, respectively. Ruthenium red staining revealed a thicker surface coat in E. histolytica trophozoites, whereas trophozoites of E. dispar showed a higher concentration of cationized ferritin particles on its surface. Mannose or glucose residues were found at the plasma membrane of both parasites treated with Concanavalin A (Con A)-peroxidase; the surface reaction product was more evident in E. dispar, compared with E. histolytica. Con A rapidly produced surface caps in E. histolytica trophozoites, whereas E. dispar showed a much less efficient mobilization of surface Con A receptors. Agglutination with Con A produced much larger clumps in E. histolytica in comparison with E. dispar. In turn, biotinylation assays revealed striking differences in the composition of surface membrane proteins in both amebic species. Overall, these results further emphasize the phenotypic differences between these two common parasites of the human intestinal tract, once considered to be the same protozoan.

The phenomenon of blood coagulation on the surfaces of implantable devices such as artifi cial blood vessels, valves or hearts is the major problem that has to be taken into consideration during the construction of such devices. Since the main factor that has an infl uence on clotting of blood is the surface of the material the device is made of, numerous attempts has already been undertaken to modify surfaces of existing materials in order to provide necessary protection against clot formation, very often by coating the materials with bio-active substances. The aim of presented study was to create a coating of phosphatidylcholine that would mimic natural cell outer membrane on the surface of polyurethane — a material already in use in construction of cardiovascular implants. The obtained coated materials have been evaluated with the use of FT-IR/ATR spectroscopy. The change in contact angle of modifi ed material was also investigated. Finally in vitro cytotoxicity tests were performed in order to examine biocompatibility of obtained material.

Opis:
Noctiluca scintillans is an important member of the mesozooplankton in terms of biomass and production in the Seto Inland Sea, Japan. The densities and adenosine triphosphate (ATP) contents of N. scintillans cells were measured. Vertical profiles of N. scintillans cellular activity in the coastal water were determined and the ATP contents were high at middle layers, with a maximum depth of 10 m. ATP contents were low in the surface and lower layers. These results suggest that active N. scintillans cells in subsurface layers with low density play an important role in the coastal ecosystem, and high-density cells in the surface water are not active.

A simple theoretical model considering cell membrane mechanosensitivity can accurately describe published experimental data on membrane area creeping and recovery, and on osmotic expansion and rupture. The model to data fit reveals real values of membrane tension and elasticity modulus, and the parameters describing membrane organization and kinetics of mechanosensitive membrane traffic, including small solute transport, water permeability, endocytosis, exocytosis, and caveolae formation. This estimation allows for separation and quantitative analysis of the participation of different processes constituting the response of plasmalemma to short time-scale membrane load. The predicted properties of the model were verified for membrane stretching at different osmotic pressures. Finally, a simple hypothesis concerning stressed cell membrane breakdown is postulated.

Opis:
Izomery chlorotoluenu są stosowane w wielu dziedzinach przemysłu, a ich szerokie wykorzystanie wiąże się z niekontrolowanym uwalnianiem do środowiska. Stąd konieczne jest ich usuwanie przede wszystkim ze środowiska gruntowo-wodnego. Do usuwania zanieczyszczeń organicznych ze środowiska wykorzystuje się wiele metod fizykochemicznych, których podstawową wadą jest problem z dalszym ich zagospodarowaniem. Z tego względu coraz większym zainteresowaniem cieszą się metody biologiczne, wykorzystujące zdolności drobnoustrojów do degradacji związków organicznych. W badaniach laboratoryjnych określono wpływ długotrwałego kontaktu szczepu Raoultella planticola SA2 z izomerami chlorotoluenu na przebieg biodegradacji tych związków. Istotnym elementem badań była także ocena wpływu każdego z izomerów chlorotoluenu na właściwości powierzchniowe mikroorganizmów prowadzących ich biodegradację. W tym celu określono zmiany w przepuszczalności błony komórkowej, potencjału dzeta komórek bakteryjnych oraz hydrofobowości ich powierzchni. Sprawdzono również, czy podatność wybranych izomerów na biodegradację zmienia się po długotrwałym kontakcie mikroorganizmów z degradowanymi związkami (tzw. stres metaboliczny). Obserwowane różnice między biodegradacją poszczególnych izomerów chlorotoluenu wykazały, że związki te – mimo podobnych właściwości fizycznych – w różnym stopniu ulegały biodegradacji oraz bardzo różnie oddziaływały na komórki mikroorganizmów. Stwierdzono, że ważnym elementem podczas biodegradacji chlorowych związków aromatycznych był długotrwały kontakt mikroorganizmów z tymi związkami. Wyniki badań wykazały także istotny wpływ położenia atomu chloru w izomerach chlorotoluenu na ich biodegradację związków oraz właściwości powierzchniowe mikroorganizmów. Drobnoustroje poddane długotrwałemu oddziaływaniu z izomerami chlorotoluenów przeprowadzały ich biodegradację szybciej i bardziej skutecznie. Zmianom uległy również właściwości powierzchniowe komórek mikroorganizmów.
Chlorotoluene isomers are used in a number of industries and their wide application results in the uncontrolled release of these compounds to the environment. Therefore, their removal is required primarily from the soil-water environment. Numerous physicochemical methods are in use to remove organic compounds from the ecosystem. However, their primary disadvantage is a question of further utilization of substances removed. Therefore, biological methods that employ microbial ability to degrade toxic organic compounds become increasingly popular. In the study, effect of a long-term contact between the Raoultella planticola SA2 strain and chlorotoluene isomers on biodegradation of these compounds was determined. Impact evaluation of each isomer on surface characteristics of microorganisms involved in the biodegradation process was another significant component of the research. Therefore, changes in the membrane permeability, zeta potential and bacterial cell surface hydrophobicity were measured. Further, effect of prolonged contact with the microorganisms (so-called metabolic stress) on susceptibility of selected isomers to biodegradation was examined. The observed differences between biodegradation of individual chlorotoluene isomers demonstrated that despite similar physical characteristics, a degree of their biodegradation varied as well as their effects on the microbial cells. The long-term contact of chloroaromatic compounds with the microorganisms was an important factor during their biodegradation. Moreover, the results indicated that the chlorine atom position had a strong impact on the chlorotoluene isomer biodegradation and the bacterial surface properties. Bacteria under the long-term stress conditions were able to degrade the chlorotoluene isomers faster and more effectively. Their surface characteristics changed as well.

Cell wall proteins of Candida albicans, besides their best known role in the adhesion of this fungal pathogen to host's tissues, also bind some soluble proteins, present in body fluids and involved in maintaining the biochemical homeostasis of the human organism. In particular, three plasma factors - high-molecular-mass kininogen (HK), factor XII (FXII) and prekallikrein (PPK) - have been shown to adhere to candidal cells. These proteins are involved in the surface-contact-catalyzed production of bradykinin-related peptides (kinins) that contribute to inflammatory states associated with microbial infections. We recently identified several proteins, associated with the candidal cell walls, and probably involved in the binding of HK. In our present study, a list of potential FXII- and PPK-binding proteins was proposed, using an affinity selection (on agarose-coupled FXII or PPK) from a whole mixture of β-1,3-glucanase-extrated cell wall-associated proteins and the mass-spectrometry protein identification. Five of these fungal proteins, including agglutinin-like sequence protein 3 (Als3), triosephosphate isomerase 1 (Tpi1), enolase 1 (Eno1), phosphoglycerate mutase 1 (Gpm1) and glucose-6-phosphate isomerase 1 (Gpi1), were purified and characterized in terms of affinities to the human contact factors, using the surface plasmon resonance measurements. Except Gpm1 that bound only PPK, and Als3 that exhibited an affinity to HK and FXII, the other isolated proteins interacted with all three contact factors. The determined dissociation constants for the identified protein complexes were of 10-7 M order, and the association rate constants were in a range of 104-105 M-1s-1. The identified fungal pathogen-host protein interactions are potential targets for novel anticandidal therapeutic approaches.

This article proposes the ideas of stochastic resonance and noise as devices with which to think of the body or self as plural and porous. Boundaries and surfaces are proposed as indefinite; cell switching and narratives of the self are discussed in relation to external forces, via Arendt’s inter-subjectivity and La Celca’s colonization as infection. The sonic artwork Ghost, which uses models of spiking neurons to materialize endogenous and exogenous composition in relation to noise and sonic memory is presented as an exploration of the boundary or limit of the notion of self. This paper, which serves as a cogitatum (a force) rather than cognitio (the result), articulates the human body as a complex and open system that steers towards chaos by adapting and accepting further complexity as, and within, constantly adaptive networks of creativity. We suggest that by focusing on the porosity of boundaries and the mechanisms that underlie their permeability, problems around identity and subjectivity might be seen in a new light.

This yields a reduction of the Whitehead asphericity conjecture (which asserts that every subpolyhedron of an aspherical 2-polyhedron is also aspherical) to the case of fake surfaces. The behaviour of glass beads and polyethylene pellets as the extra bacteria support in the fine black shale ore in the column bed bioreactor was compared. Capillary rise experiments enabled the calculation of the contact angle and the surface free energy of glass beads and polyethylene pellets. The results showed that the support material made up of polyethylene pellets yielded a higher copper extraction, which could be explained by conditions favourable to cell adhesion on the polyethylene surface.

In this work the physicochemical and biological properties of nanocrystalline TiO2 thin films were investigated. Thin films were prepared by magnetron sputtering method. Their properties were examined by X-ray diffraction, photoelectron spectroscopy, atomic force microscopy, optical transmission method and optical profiler. Moreover, surface wettability and scratch resistance were determined. It was found that as-deposited coatings were nanocrystalline and had TiO2-anatase structure, built from crystallites in size of 24 nm. The surface of the films was homogenous, composed of closely packed grains and hydrophilic. Due to nanocrystalline structure thin films exhibited good scratch resistance. The results were correlated to the biological activity (in vitro) of thin films. Morphological changes of mouse fibroblasts (L929 cell line) after contact with the surface of TiO2 films were evaluated with the use of a contrast-phase microscope, while their viability was tested by MTT colorimetric assay. The viability of cell line upon contact with the surface of nanocrystalline TiO2 film was comparable to the control sample. L929 cells had homogenous cytoplasm and were forming a confluent monofilm, while lysis and inhibition of cell growth was not observed. Moreover, the viability in contact with surface of examined films was high. This confirms non-cytotoxic effect of TiO2 film surface on mouse fibroblasts.

In this study, glass frit was coated uniformly on the surface of Al particles instead of adding glass frit to Al powder by simple mixing to form a nano-layer. The influence of the glass-frit coating on the formation of the back-surface field and electrical characteristics of the resulting Al electrode were investigated. Microstructural observations indicated that the glass components were uniformly distributed and the back-surface field layer thickness was more uniform compared to the simply mixed sample. Inaddition, the sheet resistance was ˂10 mΩ/□, much lower than the 23 mΩ/□ of the simply mixed Al electrode.

Opis:
Homocentrically grown SnO2 nanorods were synthesized by a hydrothermal method, whose diameter and length are around 20 nm and 100-200 nm, respectively. The photovoltaic properties of dye-sensitized SnO and MgO-treated SnO2 films were investigated. A light-to-electricity conversion yield of 0.8% was achieved by applying the nanorods as a thin film layer for the dye-sensitized solar cells. Treatment with MgO on SnO2 surface improves the photovoltage, fill factor and cell efficiency owing to preventing a recombination of the electrons injected into SnO with acceptors in the electrolyte.

Opis:
Niestabilne ceny ropy naftowej, czy też aspekty ekologiczne związane z redukcją emisji dwutlenku węgla do atmosfery przyczyniły się do poszukiwań nowych źródeł energii. Jedną z technologii nowych źródeł energii elektrycznej, gwałtownie rozwijających się w ostatnim czasie są ogniwa paliwowe, m.in. z membraną polimerową, charakteryzujące się wysoką gęstością energetyczną oraz sprawnością w porównaniu do klasycznych źródeł energii elektrycznej, czyli akumulatorów. Z uwagi na ekologiczną, efektywną i cichą pracę technologia ta znajduje zastosowanie w wielu różnych urządzeniach. W niniejszym artykule zajęto się problemem zastosowania technologii ogniw paliwowych z membraną polimerową PEMFC (ang. Polymer Electrolyte Membrane Fuel Cell) do zasilania elektrycznego bezzałogowego pojazdu nawodnego BPN w zastosowaniach militarnych. W ramach zrealizowanej pracy opracowano scenariusze misji rozpoznawczej dla BPN, opracowano symulator systemu zasilania BPN, bazujący na ogniwach paliwowych oraz przeprowadzono testy numeryczne.
Unstable oil prices, or ecological aspects associated with the reduction of carbon dioxide emissions to the atmosphere, have contributed to the search for new sources of energy. One of the technologies of new sources of electricity, rapidly developing in recent years are fuel cells, among others with a polymer membrane, characterized by high energy density and efficiency compared to conventional sources of electricity, i.e. batteries. Due to its ecological, efficient and quiet operation, this technology is used in many different devices. This article deals with the use of the Polymer Electrolyte Membrane Fuel Cell (PEMFC) fuel cell technology for the electrical supply of unmanned surface vehicle USV in military applications. As part of the work carried out, scenarios of reconnaissance missions were developed for USV, USV fuel cell simulator was developed, and numerical tests were performed.
