Microfluidic devices, such as lab-on-a-chip systems, are highly advantageous for cell engineering and cell based assays. It is a particularly useful approach for development of the in vitro cellular systems mimicking the in vivo environment. In this paper, a novel lab-on-a-chip device for three-dimensional human cell culture and anticancer drug testing is presented. Cells were cultured as Multicellular Tumor Spheroids (MCTS) — the best cancer tumor model developed so far. Diff erent designs were tested and novel technique of microfabrication in poly(dimethylsiloxane) was developed. MCTS were cultured in a system of polymeric microwells, with the network of microfluidic channels for culture medium flow. Design included optimal shear stress and proper nutrients supply for cultured cells. Final design provided MCTS culture for four weeks with the homeostasis-like state achievement, which is characteristic for the in vivo situation.

Opis:
Lab-on-a-chip systems are promising tools in the field of cell engineering. Microfluidic systems are integrated microlaboratories consisting of many microstructures such as microchannels and microchambers, which can be used for cell analysis and cell culture. Appropriately designed geometry of the chip allows to mimic in vivo conditions. Microsystems enables continuous culture medium perfusion. During cell culture, regulation of the flow rate of medium is possible, which allows to control conditions of the cultivation. In this paper we present a review of microfluidics systems which are used in cell engineering. We describe methods of microsystems fabrication, parameters which influence cell proliferation in microscale and examples of microsystems for cell analysis and cell culturing. Microfluidic systems for maintaining cell culture are mainly fabricated of poly(dimethylsiloxane) (PDMS) and glass, non-toxic materials for cells. The most commonly used method for fabrication of PDMS microsystems is photolithography and replica molding techniques. Cell culture in microsystems can be carried out in two ways: as a two-dimensional (2D) cell culture and three-dimensional (3D) cell culture. In two-dimensional culture cells grow as a monolayer on a flat surface of microchambers or microchannels. Microsystems for two-dimensional cell culture are widely described in the literature. They are mainly used for: (i) cell proliferation after exposure to external stimuli, (ii) testing the activity of cytotoxic drugs, (iii) interactions and cell migration and (iv) the evaluation of procedures applicable in tumor therapy e. g. photodynamic therapy. However, two-dimensional cell culture do not mimic fully in vivo conditions. In living organisms cells grow spatially creating three-dimensional structures like tissues. Therefore, nowadays microsystems for 3D cell culture are being developed intensively. Three-dimensional cell culture in microfluidic systems can be achieved in three ways: by the design of suitable geometry and topography of microchannels, by the use of hydrogels or by spheroids formation. Three-dimensional cell culture in microfluidic systems are much better experimental in vitro models than cell culture in traditional culture vessels. It is the main reason why microsystems should be still improved, as to become widely used research tools in cellular engineering.

Despite the extensive research efforts over the past 25 years that have focused on HIV, there is still no cure for AIDS. However, tremendous progress in the understanding of the structure and biology of the HIV virus led to the development of safe and potent HIV-based transgene delivery vectors. These genetic vehicles are referred to as lentiviral vectors. They appear to be better suited for particular applications, such as transgene delivery into stem cells, compared to other viral- and non-viral vectors. This is because Lentivirus-based vectors can efficiently infect nondividing and slowly dividing cells. In the present review article, the current state of understanding of HIV-1 is discussed and the main characteristics that had an impact on vector design are outlined. A historical view on the vector concept is presented to facilitate discussion of recent results in vector engineering in a broader context. Subsequently, a state of the art overview concerning vector construction and vector production is given. This review also touches upon the subject of lentiviral vector safety and related topics that can be helpful in addressing this issue are discussed. Finally, examples of Lentivirus-based gene delivery systems and their applications are presented, with emphasis on animal transgenesis and human gene therapy.

Industrial engineers gather knowledge during their bachelor studies through lectures and practical classes. The goal of practical class might be an extension of knowledge and/or a consolidation and application of already gathered knowledge. It is observed that there exists a gap between theory learnt during lectures and practical classes. If practical classes require holistic approach and solving complex tasks (problems), students strive with understanding relations and connections between parts of knowledge. The aim of this article is to show an example of a simple practical assignment that can serve as a bridge between lectures and practical classes through discussion of interactions and relations between parts of theoretical knowledge. It is an example of in-class simulating of a line and cellular layout considering discussion of elements impacting and impacted by the type of layout (e.g. learning curve, changeovers, etc.). In-class verification of the presented approach confirmed its usability for teaching industrial engineers and bridging the gap between theory delivered through lectures and more advanced practical classes.

Opis:
Covalent attachment of a thermoresponsive polymer to solid support leads to layers exhibiting temperature-dependent properties. Below the cloud point temperature (TCP) of the thermoresponsive polymer the layer is hydrophilic – it is hydrated and polymer chains adopt an expanded conformation. Above TCP, the polymer chains collapse due to dehydration and the surface becomes hydrophobic. This is a reversible process, lowering the temperature cause hydration and swelling of the layer. Such thermoresponsive layers can be obtained via reactions of entities present on the surface (e.g. functional groups, radicals etc.) with complementary functionalities in the polymer chains (grafting to) or with monomer subjected to polymerization (grafting from). Thermoresponsive layers may be used in many biomedical applications such as separation of molecules or cell sheet engineering. In this work, well-defined thermoresponsive (co)polymers of glycidol and ethyl glycidyl carbamate (mPGl), 2-ethyl and 2-nonyl-2-oxazoline (PENOx) as well as homopolymers of 2-isopropyl-2-oxazoline (PIPOx) were grafted to functionalized glass and silica substrates with the aim to obtain thermoresponsive layers for potential application in cell sheet engineering. Presence of polymers covalently bonded to substrates was confirmed by FT-IR and XPS studies. The polymer layers were 5-50 nm thick, depending on the molar mass and polymer concentration. Microscopic techniques indicated a smooth surface of mPGl layers, slightly rough texture of PENOx layers and fibrille-like fibers surface of PIPOx layers. Ellipsometry and contact angle studies revealed the response of layers to temperature changes. Biocompatibility of layers with dermal fibroblasts was confirmed by toxicity tests. Thermoresponsive surfaces were employed as substrates for skin cell culture and harvesting. Fibroblasts adhesion and proliferation on investigated surfaces was comparable with control sample. A confluent cell sheet was formed after 24 hours of culture. The influence of surface properties on cell adhesion and proliferation was examined. Detachment of cells from surfaces was controlled by variation of the temperature. An intact monolayer of cultured dermal fibroblasts was detached. No mechanical or enzymatic methods were required to harvest the cell sheets. Skin cell sheets, detached from thermoresponsive polymer layers may be applied in the cell sheet engineering that is highly desirable in tissue regeneration.

The work contains an analysis of the theoretical potential of the electric energy yield for a photovoltaic panel made of polycrystalline silicon. Calculations were based on monthly multiannual average values of solar radiation intensity available in solar conditions of Małopolska Region. For their purposes, the researchers have chosen a source with a power reaching 235 Wp, which is available in the market and may be used in farms located in this region.

Metal surfaces have become important over the last decade for potential surgical implants, and within this context we present here a study of the cell growth on modified gold surfaces. Gold films, deposited on glass plates and annealed with a hydrogen flame, were modified with four different carboranethiol derivatives: 1-(HS)-1,2-C2B10H11 (A), 1,2-(HS)2-1,2-C2B10H10 (B), 9,12-(HS)2-1,2-C2B10H10 (C) and 1,12-(HS)2-1,12- C2B10H10 (D). The materials engendered from these modifications were used to investigate the adhesion and growth of rat aortic smooth muscle cells cultured on these surfaces in a DMEM medium with 10% of fetal bovine serum. One day after seeding, the highest number of initially adhered cells was found on the surface of a bare gold film. However, three days after seeding, the number of cells on carboranethiol-modified gold samples B, C and D was significantly higher than the number on a bare gold film. After seven days, the number of cells on a bare gold film and on gold films modified with derivatives A, B and D was very similar, but the surface of a gold film modified with derivative C exhibited a significantly smaller number of cells. This may be explained by the exposure of the CH vertices of the carborane cluster, which are more acidic than the BH vertices exposed toward the cells in either A or B.

Since it is known that various cell lines may ex-press different behaviours on the scaffolds surface, a comprehensive analysis using various cellular mo-dels is needed to evaluate the biomedical potential of developed biomaterials under in vitro conditions. Thus, the aim of this work was to fabricate bone scaffolds composed of a chitosan-agarose matrix reinforced with nanohydroxyapatite and compare the biological response of two cell lines, i.e. mouse calvarial preosteoblasts (MC3T3-E1 Subclone 4) and human foetal osteoblasts (hFOB 1.19). Within this study, the osteoblasts number on the scaffold surface and the osteogenic markers level produced by MC3T3-E1 and hFOB 1.19 cells were determined. Furthermore, changes in calcium and phosphorous ions concentrations in the culture media dedicated for MC3T3-E1 and hFOB 1.19 were estimated after the biomaterial incubation. The obtained results proved that the fabricated biomaterial is characterized by biocompatibility and osteoconductivity since it favours osteoblasts attachment and growth. It also supports the production of osteogenic markers (collagen, bALP, osteocalcin) by MC3T3-E1 and hFOB 1.19 cells. Interestingly, the developed biomaterial exhibits different ion reactivity values in the two culture media dedicated for the mentioned cell lines. It was also revealed that mouse and human osteoblasts differ in the cellular response to the fabricated scaffold. Thus, the use of at least two various cellular models is recommended to carry out a reliable biological characterization of the novel biomaterial. These results demonstrate that the tested bone scaffold is a promising biomaterial for bone regeneration applications, however further biological and physicochemical experiments are essential to fully assess its biomedical potential.

Due to development of the renewable energy sources, the powering of fuel cells (FCs) with bio-fuels is very important. The one of this fuel is methyl alcohol. The use of fuel cells on a large scale is mainly limited by the high cost of catalysts - mainly platinum. Elimination of Pt as catalyst would allow for wider commercial application of FCs. The paper presents a study of methyl alcohol electrooxidation on electrode with NiCo alloy catalyst. Researches were done by the method of polarizing curves of electrooxidation of methanol in glass vessel. Conducted measurements show that there is a possibility of electrooxidation of methanol with Ni-Co catalyst. In any case, the process of electrooxidation of methanol occurs. A maximum current density was equal 50 mA/cm2 . So, the work shows possibility to use Ni-Co alloys as catalysts for fuel electrode to methyl alcohol electrooxidation.

Providing more and more energy is an essential task of today's energetic industry. In the last few years, addition to traditional methods of energy production, alternative energy sources have been fast developing. One of the devices that can use these sources is fuel cell. The fuel cells can be a power source of future mainly due to their high efficiency, low influence on environment and possibility of powering with different fuels. Most often fuel cells are powered by hydrogen. However, problems with the problems with its cheap production and storage are the reason for the search of new fuels for fuel cells. But it must be a fuel that will provide zero or low emission level. One of these fuels can be vegetable oil. The paper presents measurements of electrooxidation of coconut oil emulsion on a smooth platinum electrode in an aqueous solution of KOH. Electrochemical measurements were performed in a glass cell with AMEL System 5000 potentiostat. The obtained maximum current density is equal 25 mA/cm2. So, a fundamental possibility of using the coconut oil as fuel for fuel cell. But is necessary to keep the temperature of process above 303K.

Functional modeling of human genes and diseases requires suitable mammalian model organisms. For its genetic malleability, the mouse is likely to continue to play a major role in defining basic genetic traits and complex pathological disorders. Recently, gene targeting techniques have been extended towards developing new engineering strategies for generating extensive lesions and rearrangements in mouse chromosomes. While these advances create new opportunities to address similar aberrations observed in human diseases, they also open new ways of scaling-up mutagensis projects that try to catalogue and annotate cellular functions of mammalian genes.

As fuel for fuel cells can be used various substances, but mainly fuel cells are powered by clear hydrogen (or hydrogen obtained from organic substances by reforming process). However, problems with the storage of hydrogen are the reason for the search of new fuels for fuel cells. Due to development of the renewable energy sources, the powering of fuel cells with bio-fuels is very important. Vegetable oil is an alternative fuel for diesel engines and for heating oil burners. Powering high efficiency power sources like fuel cells with renewable fuels (like canola oil) will allow development of renewable energy sources and elimination or reduce of toxic substances emissions. The paper presents the possibility of using canola oil as fuel for direct electricity production. The work shows possible electrooxidation of canola oil emulsion on a smooth platinum electrode in an solution of H2SO4. The resulting current density of canola oil electrooxidation reached the maximum level of 8 mA/cm2. So, the possibility of using canola oil as fuel for direct electricity production has been proved.

Low density polyethylene (LDPE) was modified by an Ar plasma discharge and then grafted with glycine (Gly), bovine serum albumin (BSA) or polyethylene glykol (PEG). Some plasma-treated samples and samples grafted with BSA were exposed to a suspension of colloidal carbon particles (C, BSA+C). Pristine LDPE and tissue culture polystyrene dishes (PSC) were used as control samples. The materials were seeded with rat aortic smooth muscle cells and incubated in a medium DMEM with 10% of fetal bovine serum. On day 1 after seeding, the cells on LDPE modified with plasma only, Gly, BSA and BSA+C adhered in similar numbers as on PSC, while the values on non-modified and PEG-modified samples were significantly lower. On day 5, the highest cell numbers were found again on LDPE with Gly, BSA and BSA+C. On day 7, the highest number of cells was found on LDPE modified only with plasma. The latter cells also dis-played the largest cell spreading area. The increased cell colonization was probably due to the formation of oxygen-containing chemical functional groups after plasma irradiation, and also due to positive effects of grafted Gly, BSA and BSA in combination with colloidal C particles.

Opis:
Baker’s yeast Saccharomyces cerevisiae is quite commonly applied as a wholecell biocatalysts in biotransformations – reactions based on enzymatic transformations of chemical compounds. Yeast cells are easy in cultivation and use. They are usually used to catalyze such reactions as bioreduction or hydrolysis. The full sequencing of its genome accompanied with achievements of genetic engineering allowed to design new yeast strains characterized by high conversion yield and reaction selectivity. Genetically modified cells of Saccharomyces cerevisiae catalyze biotransformations, which lead to chiral building blocks important in pharmaceutical industry (especially those obtained by reduction of á- and â -oxoesters). „Designer yeast” is a new catalyst for Baeyer–Villiger oxidation. Recombinant yeast lipases have been discussed as useful means in biodiesel production because the microbiological method of producing of this kind of fuel has many advantages. There is a growing interest in application of modified yeast in biotransformation reactions. Modern directions to improve catalytic abilities of baker’s yeast include: the use of surface display technology of enzymes, optimization or increase in availability of cofactor required for bioreduction reactions or gene knock-out, which eliminates the activity of enzymes with conflicting and unwanted stereoselectivities. Commonly used technique is also overexpression of the desired protein or expression of heterologous enzymes in yeast cells.
