Opis:
Biological synthesis of metal nanoparticles gained worldwide interest due to their rapid, non-toxic, economical, single-step technique and eco-friendly alternative. Green-chemical approach of production of Zinc Oxide (ZnO) nanoparticles (NPs) has been exploited in the field of medicine, food packaging, nano-optical and electrical devices. Muntingia calabura is a multipotent medicinal plant with astounded biological activities and phytoconstituents. The nanoparticles obtained using aqueous extracts of various parts of M. calabura were characterized with UV-VIS spectroscopy to obtain information concerning the optical properties of synthesized ZnO nanoparticles. The proposed green and economical method could be used for large scale production of nanostructures because of its advantages over other physical and chemical methods.

The 6-oxopurine xanthine (Xan, neutral form 2,6-diketopurine) differs from the corresponding 6-oxopurines guanine (Gua) and hypoxanthine (Hyp) in that, at physiological pH, it consists of a ≈ 1:1 equilibrium mixture of the neutral and monoanionic forms, the latter due to ionization of N(3)-H, in striking contrast to dissociation of the N(1)-H in both Gua and Hyp at higher pH. In xanthosine (Xao) and its nucleotides the xanthine ring is predominantly, or exclusively, a similar monoanion at physiological pH. The foregoing has, somewhat surprisingly, been widely overlooked in studies on the properties of these compounds in various enzyme systems and metabolic pathways, including, amongst others, xanthine oxidase, purine phosphoribosyltransferases, IMP dehydrogenases, purine nucleoside phosphorylases, nucleoside hydrolases, the enzymes involved in the biosynthesis of caffeine, the development of xanthine nucleotide-directed G proteins, the pharmacological properties of alkylxanthines. We here review the acid/base properties of xanthine, its nucleosides and nucleotides, their N-alkyl derivatives and other analogues, and their relevance to studies on the foregoing. Included also is a survey of the pH-dependent helical forms of polyxanthylic acid, poly(X), its ability to form helical complexes with a broad range of other synthetic homopolynucleotides, the base pairing properties of xanthine in synthetic oligonucleotides, and in damaged DNA, as well as enzymes involved in circumventing the existence of xanthine in natural DNA.

W pracy badano wpływ dodatku naturalnych surowców pochodzenia roślinnego na biosyntezę kwasu cytrynowego, prowadzoną metodą hodowli w podłożu stałym z użyciem pleśni Aspergillus niger. W tym celu do głównego surowca, wysłodków buraczanych, dodawano różne ilości tapioki, skrobi kukurydzianej, otrąb pszennych i żytnich, wycierki ziemniaczanej oraz melasy buraczanej i trzcinowej. Wydajność kwasu cytrynowego wahała się, w zależności od stosowanego surowca, od 68 g/kg s.s., w przypadku otrąb pszennych do 160 g/kg s.s., przy zastosowaniu wycierki ziemniaczanej oraz do ponad 200 g/kg s.s., w przypadku dodania tapioki lub melasy trzcinowej.
A solid state fermentation method was developed for citric acid production from different substrates by culturing Aspergillus niger S. A Sugar beet pulp was applied as the main substrate; it was enriched by raw materials such as: tapioca, beet or cane molasses, corn starch, wheat bran, and potato waste pulp. The yield of citric acid varied depending on the type of different substrates added; it ranged from 68 g/kg d.s. for wheat bran added, to 160 g/kg d.s. as for potato waste pulp, and up to more than 200 g /kg d.s. as for the addition of tapioca and cane molasses.

α,α-Trehaloza jest naturalnie występującym disacharydem, zbudowanym z dwóch cząsteczek glukozy połączonych wiązaniem α,α- 1,1’-O-glikozydowym. Znana jest głównie ze zdolności do stabilizacji białek w warunkach stresu środowiskowego oraz hamowania ich agregacji. Po opracowaniu wielkotonażowej, biotechnologicznej metody produkcji, trehaloza znalazła liczne zastosowania w różnych gałęziach przemysłu, głównie w przemyśle spożywczym, kosmetycznym, farmaceutycznym i medycynie. Dowiedziono, że właściwości bioprotekcyjne trehalozy zostają zintensyfikowane w przypadku jej glikopolimerów.
α,α -Trehalose is naturally occurring disaccharide composed of two glucose molecules linked by α,α -1,1’-O-glycosydic bond. It is primarily known from its ability to protect proteins during environmental stresses and inhibition of their aggregation. The development of largescale bioprocess has allowed to use trehalose in various applications, mainly in the food, cosmetic and pharmaceutical industry as well as in medicine. It was reported that bioprotective properties are amplified when trehalose is incorporated in polymer chain as a pendant group.

Opis:
The pyrokinin (PK) family plays a multifunctional role in an array of important physiological processes in a variety of insects. A PK active core analog containing an (E)-alkene, transPro isosteric component was evaluated in five disparate PK bioassays and/or in a recombinant PK receptor cell line, representing six different insect species. The assays included pheromone biosynthesis in the moth Heliothis peltigera, melanization in the larval Spodoptera littoralis, pupariation acceleration in the larval fly Neobellieria bullata, diapause termination in the moth Heliothis zea, and hindgut contraction in the cockroach Leucophaea maderae. This constrained analog demonstrated unselective agonist activity that approached, matched, or exceeded the activity of parent PK peptides of equal length in all six PK assays. The results provide strong evidence for the orientation of Pro and the core conformation adopted by PK neuropeptides during interaction with disparate PK receptors. A PK active core analog incorporating a second transPro motif, the dihydroimidazoline moiety, was found to demonstrate pure, selective agonism in the melanotropic bioassay, with no significant activity in three other PK bioassays. Both types of transPro isosteric analogs feature modification adjacent to the primary tissue-bound peptidase hydrolysis site that is expected to enhance biostability over natural PK peptides. The research further identifies two novel scaffolds with which to design either selective or non-selective mimetic PK analogs as potential leads in the development of environmentally favorable pest management agents capable of disrupting PK-regulated systems.

Aminoacyl-tRNA synthetases (AARS) are essential proteins found in all living organisms. They form a diverse group of enzymes that ensure the fidelity of transfer of genetic information from the DNA into the protein. AARS catalyse the attachment of amino acids to transfer RNAs and thereby establish the rules of the genetic code by virtue of matching the nucleotide triplet of the anticodon with its cognate amino acid. Here we summarise the effects of recent studies on this interesting family of multifunctional enzymes.

In laboratory condition phenolic compounds, such as scopoletin, the ferulic, p-coumaric and trans-cinnamic acid retarded growth of Phoma exigua var. foveata mycelium. Equal effect revealed inhibitors of polyamine biosynthesis, DFMA and DFMO. A role of tested substances in potato gangrene pathogenesis is discussed.
W kulturach in vitro związki fenolowe, takie jak skopoletyna, kwasy: ferulowy, p-kumarowy i trans-cynamonowy ograniczały wzrost grzybni Phoma exigua var. foveata, sprawcę gangreny ziemniaka. Podobne działanie wykazywały inhibitory biosyntezy poliamin, difluorometyloarginina (DFMA) oraz difluorometyloornityna (DFMO). Przeprowadzono dyskusję nad ewentualną rolą testowanych substancji w patogenezie gangreny ziemniaka.

Flax (Linum usitatissimum L.) is one of the promising oil crops in the north of Kazakhstan. Over the last 10 years, the total area under this crop in the region has increased fourteen-fold, since flax is a very plastic crop for steppe and dry steppe conditions, and oil seeds are in high demand in the world market. Flaxseed oil, due to the content of polyunsaturated fatty acids, occupies one of the first positions among other edible vegetable oils. Depending on the environmental conditions, the oil content of the crop may vary from 36.4% to 52.0%, while, as noted by many researchers, the change in the oil content depends on the genotypic characteristics. Therefore, the study aims to analyze the influence of the genotype and climatic conditions of cultivation on the fat biosynthesis in flax seeds in the context of the dry steppe zone of Northern Kazakhstan on dark chestnut soils of the Akmola region. Field experiments were conducted in 2015–2017 with the study of nine oil flaxseed cultivars (of mid-season varieties) sown on May 20th at the seeding rate of 7 million seeds/ha based on the traditional technology of cultivation recommended for the region. The climatic parameters were taken into account according to the meteorological service data. The fat content was determined by the extraction method using a Soxhlet apparatus, in accordance with GOST (All-Union State Standard) 10857–64, and the moisture content was determined by means of the thermogravimetric method, according to the National Standard of the Republic of Kazakhstan 2.195–2010, for the recalculation of the oil content for absolutely dry matter. It was found that the formation of oil depends on the climatic parameters and on the sum of active temperatures during the ripening period, in particular. The evaluation of plasticity and stability of the cultivars (genotypes) showed that VNIIMK 620, Lirina, Karabalyksky 7 can be considered highly valuable cultivars and Severny, Biryuza, Kazar, Ilyich – valuable cultivars in terms of the oil content for the dry steppe zone.

The in vitro protein biosynthesis has the potentials to become a powerful technology for biochemical research. Beside the determination of structure and function the in vitro evolution of proteins is also of great interest. The system described was used to produce bovine heart fatty acid binding protein (FABP) and bacterial chloramphenicol acetyltransferase (CAT) with and without fusion of the Strep-tag II affinity peptide. The proteins were purified after and during protein biosynthesis by using a StrepTactin Sepharose matrix. No significant influence of the Strep-tag and the conditions during the affinity chromatography on maturation or activity of the protein was observed. The in vitro evolution of proteins is feasible by means of ribosome display. The selection of a specific mRNA coding for a shortened FABP with a N-terminal His-tag via the accompanying protein property was shown. Goal of the selection was to bind the FABP via the His-tag on Ni(II)-IDA-agarose. After nine cycles of transcription, translation, affinity selection and RT-PCR the protein with the His-tag could be enriched 108-fold. In order to correlate a possible relationship between changes in protein population and biological function studies were initiated in which 2-dimensional protein patterns of the total in vitro system were compared after 0 and 2 h reaction time. The very interesting findings are that a number of proteins disappear, while others are newly formed during protein synthesis.
