Opis:
In this study, 50 samples of air particulates collected from different places in- and outside the Al-Tuwaitha nuclear site, south of Baghdad were used to measure daily gross alpha and beta activity concentrations (AAC and BAC) for the period from 28 January 2015 to 13 April 2017. At the same time, several meteorological factors such as air temperature, wind speed, wind direction, air pressure, relative humidity, and solar radiation, were also measured. Air stability classes were also derived from wind speed and solar radiation. AAC/BAC variations in the surface air layer were discussed in relation to these factors. The results show that there are inverse relations between AAC/BAC and wind speed and temperature, linear relations between AAC/ABC and air pressure and weak relations between AAC/BAC and relative humidity and solar radiation. Lastly, AAC/BAC measurements in unstable air are as large as in neutral air.

In this study, after the pulse shape calibration of a liquid scintillation counting (LSC) spectrometer (Quantulus 1220), the efficiency was determined depending on sample quenching parameters. Then, gross alpha and beta activities in two spiked water samples obtained from International Atomic Energy Agency (IAEA) were used for the validation of the ASTM D7283-06 method, which is a standard test method for alpha and beta activity in water by LSC. Later, the drinking water samples (35 tap water and 9 bottled water) obtained from different districts of Ankara, Turkey, were measured. The maximum gross alpha activities are measured to be 0.08 Bq/L for tap waters and 0.13 Bq/L for bottled waters, whereas the maximum gross beta activities are found to be 0.18 Bq/L for tap waters and 0.16 Bq/L for bottled waters. These results indicate that these drinking water samples are below the required limits, which are 0.1 Bq/L for alpha emitting radionuclides and 1 Bq/L for beta emitting radionuclides. As a result, gross alpha and beta activities in drinking water of Ankara were determined accurately by this validated LSC method. It is also worth noting that LSC is a rapid and accurate method for the determination of gross alpha and beta activities without requiring a tedious sample preparation.

Opis:
Amaranthus caudatus L. seed germination was studied under different levels of water deficit induced by PEG 6000 in laboratory conditions. PEG at osmotic potentials -0.2 to -0.3 MPa at 24°C in darkness delayed germination and reduced final germination percentage. PEG solutions at osmotic potential lower than -0.3 MPa almost totally blocked seed germination. Ethephon was much more effective than GA3 in reversing PEG-caused inhibition of A. caudatus seed germination. PEG decreased α-amylase activity after 14 h incubation. It decreased β-amylase activity after 14 and 20 h, and caused an increase in total dehydrogenase activity only after 20 h of incubation. Unlike GA3, ethephon increased α-amylase activity in seeds after 12 and 14 h of incubation under water deficit. After 20 h of incubation there was no difference in α-amylase activity in any of the treatments. Neither ethephon nor GA3 affected the activity of β-amylase and dehydrogenase.

When the energy within the mass of the human brain that is associated with the intrinsic range in fluctuations of the Gravitational Constant (G) is set equal to the energy from a magnetic field (B) within the cerebral volume and solved for B, a value in the order of 20 to 50 nT is obtained. Quantitative Electroencephalographic (QEEG) and sLORETA (Low Resolution Electromagnetic Tomography) analyses of cerebral cortical activity during exposure to a range of applied rotating, frequency-modulated, transcerebral (between the two temporal lobes) magnetic fields between < 1 nT and 7000 nT while volunteers sat within a darkened, quiet chamber were completed. There was marked enhancement of power within the 4 Hz to 10 Hz band within the right caudal (cuneus) hemisphere while the ~5 to 20 nT averaged strength magnetic fields were applied but no significant responses at lesser or greater intensities. These results suggest that a physical process coupled to the source of the fluctuation (~10-15 m3kg-1s-2) in G may interact with right hemispheric activity within the range at which gravity waves have been estimated to interact with Schumann frequencies generated between the earth and ionosphere.

Human neutrophil peptides (HNPs, alpha-defensins) are a group of six defensins being considered as new antimicrobial drugs because of their multifunctional efficiency against bacteria, viruses and fungi. Regardless of the unique biological properties alpha-defensins are unstable compounds and their activity depends on many physical and chemical factors as well as on the kind of the used cultivation media. This leads to research difficulties and obstructions in their therapeutic application. The purpose of this study was to determine antibacterial activity of alpha-defensin 2 dissolved in pharmacopeia buffers and in parallel to develop selective and accurate analytical tests for identification and assay of alpha-defensin 2 in the course of study. The antibacterial effect of alpha-defensin 2 was determined against the strain Escherichia coli (ATCC 25922). It was found that 10 mg/l of Human neutrophil peptide-2 (HNP-2, alpha-defensin 2) dissolved in pH 9.0 buffer caused 90% inhibition of the bacterial respiratory activity. This buffer was considered as a suitable environment for deploying the antibacterial activity of the alpha-defensin. А selective MS analysis method for identification of alpha-defensin 2 in sample mixtures was developed. Also HPLC method with alternative selectivity was elaborated for identification and assay of mixtures containing alpha-defensin 2 and aminoacids in Mueller Hinton broth. The procedure includes development of system suitability test determination.

Background. This study investigates the indicators of electroencephalographic (EEG) oscillatory activity and processes that are correlated with manual movements executed by the subdominant (left) hand in women with a high or a low individual α-frequency. Material and methods. 113 healthy right-handed women from the ages of 19 to 21 were divided randomly into two experimental groups with high (n = 59, IαF > 10.25 Hz) and low (n = 54, IαF ≤ 10.25 Hz) individual EEG α-frequency (IαF). EEG power during flexion or extension of the subdominant hand fingers was evaluated. Results. Manual movements performed by women, especially those exhibiting high modal α-frequency, were accompanied by reduced α and β power in mid and posterior cortical areas. These changes occurred in combination with a local power increase in α1-oscillations in the frontal leads. A local increase of α3-activity in the frontal cortex areas was also revealed in women with low IαF. In this same group of women, generalized increases in EEG power of θ-, β- and γ-oscillations were observed in the cortex. Conclusions. These results revealed a greater redundancy of brain processes in women with low IαF power compared to women with high α-frequency.
Wprowadzenie. Niniejsza praca poświęcona jest badaniu wskaźników elektroencefalograficznej aktywności oscylacyjnej i procesów, które są skorelowane z ruchami manualnymi, u kobiet z wysoką lub niską indywidualną częstotliwością α określoną podczas ruchów manualnych wykonywanych przez mniej dominującą (lewą) rękę. Materiał i metody. 113 zdrowych praworęcznych kobiet w wieku od 19 do 21 lat zostało podzielonych na dwie grupy eksperymentalne z wysoką (n = 59, IαF ≥ 10,254 Hz) i niską (n = 54, IαF ≤ 10,25 Hz) indywidualną wartością częstotliwości EEG α (IαF). U tych kobiet oceniona została aktywność EEG podczas zginania lub prostowania mniej dominujących palców. Wyniki. Ruchom manualnym wykonywanym przez kobiety, zwłaszcza o wysokiej modalnej częstotliwości α, towarzyszyło pewne zmniejszenie mocy fal EEG α i β w środkowych i tylnych obszarach kory mózgowej. Takie zmiany połączono z lokalnym wzrostem mocy w oscylacjach α1 w przednich przewodach. Lokalny wzrost aktywności α3 w obszarach kory czołowej został również ujawniony u kobiet z niską wartością IαF. Pewne uogólnione zwiększenie mocy fal EEG oscylacji θ, β i γ zaobserwowano w korze kobiet należących do tej grupy. Wnioski. Wyniki ujawniają większą nadmiarowość procesów mózgowych u kobiet z małą mocą fal IαF w porównaniu z kobietami z wysoką częstotliwością α.

Background. In hospital patients suffering from adverse clinical and biochemical symptoms of malnutrition, it is often necessary to employ parenteral nutrition to avoid the body’s tissue becoming broken down by being metabolised. Thus, the patient’s welfare and survival can be supported throughout any periods of medical crisis. Two of the enzymes responsible for metabolising glycoconjugates are a-fucosidase (FUC) and p-glucuronidase (GLU), present in lysosomes. They release fucose or glucuronic acid from the non-reducing end of oligosaccharide chains. Objective. To determine the effect of parenteral nutrition administered to ill patients, on glycoconjugate metabolism, by measuring serum and urinary activities of FUC and GLU. Material and methods. Blood samples and the daily urine collection were taken from 23 patients’ who had been undergoing parenteral nutrition for either 5 or 10 days, as well as from a baseline sample. Enzyme activities in serum and urine were determined by the method of Zwierz et al. Results. Serum FUC activities were significantly lower after 10 days compared to 5, (p< 0.0172), whereas GLU activities were significantly lower after both 5 and 10 days, (p< 0.0007 and p< 0.0208 respectively), compared to levels before starting parenteral nutrition. GLU activities were however higher after 10 days than those after 5 days, (p< 0.0023). In urine, FUC activities were significantly decreased after 10 days compared to 5 days after starting parenteral nutrition, (p< 0.0245). Urine GLU activities were unaffected by parenteral nutrition nor was any effect seen on FUC or GLU activities when calculated per 1mg creatinine. Conclusions. Serum FUC and GLU activities can be used for assessing the effect of parenteral nutrition on glycoconjugate metabolism. The significant decreases of serum GLU activity observed after 5 and 10 days, may serve to indicate that the components of parental nutrition are appropriate and that the body has become suitably adapted to this form of nutrition.
Wprowadzenie. Kliniczne lub biochemiczne objawy niedoborów pokarmowych stwarzają konieczność wdrożenia żywienia pozajelitowego w celu ograniczenia katabolizmu własnych tkanek i stworzenia warunków umożliwiających choremu przetrwanie krytycznego okresu, a-fukozydaza (FUC) i P-glukuronidaza (GLU) są enzymami lizosomalnymi uczestniczącymi w katabolizmie glikokoniugatów. Odcinają fukozę (FUC) lub kwas glukuronowy (GLU) od nieredukującego końca łańcuchów oligosacharydowych. Cel . Celem badań było zbadanie wpływu żywienia pozajelitowego na katabolizm glikokoniugatów poprzez ocenę aktywności FUC i GLU w surowicy krwi i moczu chorych żywionych pozajelitowo. Materiał i metody. Krew z żyły łokciowej oraz mocz z dobowej zbiórki pobrano od 23 pacjentów żywionych pozajelitowo trzykrotnie: przed rozpoczęciem żywienia pozajelitowego, w piątej oraz dziesiątej dobie alimentacji dożylnej. Aktywność FUC i GLU w surowicy krwi i moczu oznaczano metodą kolorymetryczną Zwierza i wsp. Wyniki. W trakcie żywienia pozajelitowego, stężenie aktywności FUC w surowicy krwi uległo istotnemu obniżeniu w dziesiątej dobie (p<0,0172), w porównaniu do doby piątej, żywienia pozajelitowego. Stężenie aktywności GLU istotne obniżyło się (p<0,0007) w piątej oraz (p<0,0208) dziesiątej dobie, w porównaniu do aktywności przed zastosowaniem żywienia pozajelitowego. Stężenie aktywności GLU istotnie rosło (p<0,0023) w dziesiątej dobie, w porównaniu do piątej doby żywienia pozajelitowego. Stężenie aktywności FUC w moczu uległo istotnemu obniżeniu (p<0,0245) w dziesiątej dobie, w porównaniu do piątej doby żywienia pozajelitowego. Żywienie pozajelitowe nie wpływa istotnie na stężenie aktywności GLU w moczu oraz moczowe aktywności FUC i GLU przeliczane na 1 mg kreatyniny. Wnioski. Stężenia aktywności FUC i GLU w surowicy mogą być użyte do oceny wpływu żywienia pozajelitowego na katabolizm glikokoniugatów. Istotne obniżenie aktywności GLU w surowicy krwi w 5 i 10 dniu pozajelitowego żywienia, może świadczyć o prawidłowym doborze składników i adaptacji organizmu do żywienia pozajelitowego.

Phytosulfokine-α (PSK-α) (H-Tyr(SO3H)-Ile-Tyr(SO3H)-Thr-Gln- OH) (I), a sulfated growth factor universally found in both monocotyledons and dicotyledons, strongly promotes proliferation of plant cells in culture. The C-terminal truncated analog named PSK-β (Tyr(SO3H)-Ile-Tyr(SO3H)-Thr) (II) showed a 10-fold lower activity than that of the parent pentapeptide. Because PSK-α promotes proliferation and differentiation during the plant growth we undertook the studies on the influence of PSK-α on plant defense mechanisms in that period. In present studies on PSK-α (I), PSK-β (II), and its analogues, we performed a search of another biological properties. The aim of our investigation was evaluation of PSK-α, PSK-β and their selected analogues in relation to growth and development of plant pathogens, such as Phoma narcissi and Botrytis tulipae. In these studies we elaborated the synthesis of PSK-α or PSK-β and their 22 analogues modified by natural and non-natural amino acid residues. Peptides were synthesized by the solid phase method according to the Fmoc procedure on a Wang-resin. Free peptides were released from the resin by 95% TFA in the presence of EDT. Biological effect of these peptides was evaluated by test on the growth and development of pathogens of P. narcissi and B. tulipae.

Concentration of glycogen, maltose, and trehalose as well as the activity of hydrolases catabolising them: «-amylases, glucoamylase, maltase, and trehalase were studied during a 48 h infection of Galleria mellonella (Lepidoptera) instar larvae infected with infective Heterorhabditis zealandica (20 nematodes/insect). The content of trehalose in the infected insects during the first day of infection was higher (p < 0.05) than in the controls. The concentration of maltose and glycogen were similar in both groups despite the fact that the activity of all examined o-glycosidases was generally much higher in the infected insects than in the controls.

Genetic analysis of two rye interline crosses and a set of wheat/rye chromosomal addition lines was performed to reveal the mechanism underlying wide variation range of alpha-amylase activity in sound grain. The long arm of chromosome 6R was found to be responsible for increased enzyme synthesis during late stages of triticale grain maturation. Only nuclear genes seemed to control alpha-amylase activity, as reciprocal crosses between rye lines showed no maternal effects. Low enzyme activity showed complete dominance over high level of its synthesis. Segregation ratios, observed in F₂ and BC₁ crosses, indicated that recessive alleles at two independent duplicative loci underlie intensive alpha-amylase production.

Opis:
The albino Wistar rats were used to investigate the effect of thyroxine administration on the activity of citric acid, α-Amylase, creatine phosphokinase (CPK), creatinine, cholesterol and triglycerides level in blood serum and citric acid in liver and kidney. Two groups of rats were taken for the experiment. One group was injected subcutaneously with 200 μl of thyroxine/50 g of body weight, the other group served as control. The animals were vivisected with anesthesia after intervals of 24, 48, 72 and 96 hours α-Amylase and CPK levels in blood; citric acid level in liver and kidney of control and thyroxine treated rats. The results showed that the α-Amylase activity remained unaltered (100 g/h/l) throughout the experiment. However, citric acid level found to be 460 mg in the liver and 415 mg in kidney of control rats. The values slightly increased to 480 and 680 mg in respective tissues of thyroxine treated animals for 24 hr. The values decreased in 48 and 72 hours of treatment. Further the values found to be increased after 96 hours of treatment. Although the thyroxine treatment altered citric acid, Creatinine, Cholesterol, Triglycerides and CPK levels in the tissues, the serum α-Amylase was not influenced by the thyroxine. The study suggests that the thyroxine acts as a regulator all the above parameters in the cellular level.

Single and double-gene Rhtl, Rht2, Rht3, Rht1 + Rht2 and Rht3 + Rht2 isogenic lines of wheat in four parental rht varieties were grown in drilled yield trials at four sites in 1989. The same lines were also grown in 1988 together with hybrid genotypes from CHA (chemical hybridising agent) F₁ production plots. In the inbred lines shorter than one metre, Rht alleles reduced total shoot biomass by shortening the straw; mass of straw per unit plant height was unaffected. Highest grain yield was obtained from plant heights between 70 and 100 cm. The Rht genotype achieving this stature varied according to parent varietal height. The hybrids grown allowed comparisons between intra- and inter-varietal crosses over a range of Rht genotypes. In F₁ hybrids positive heterosis was observed for plant height, grain yield and mean grain weight. Highest yields were obtained from inter-varietal hybrids carrying one, two or three doses of Rht1 or Rht2 or one dose of Rht3. An Rht3lrht hybrid showed resistance to premature α-amylase production during grain ripening.
