Opis:
SNP-minisequencing has become common in forensic genetics, especially for analysing degraded or low copy number DNA (LCN DNA). The aim of this study was to examine the usefulness of five SNP (single nucleotide polymorphism) markers for analyzing degraded and LCN DNA recovered from archival samples. DNA extractions of eight formalin-fixed paraffin-embedded (FFPE) tissues were performed and DNA fragments were amplified in one multiplex PCR (polymerase chain reaction). SNPs were identified in a minisequencing reaction and a gel electrophoresis in ABI Prism® 377 Sequencer. The research confirmed the usefulness of SNP-minisequencing for analysing FFPE tissues.

Introduction and objective. The aim of the study is to evaluate the mineralization tooth tissues in the inhabitants of Melniki village Szackie, Lake District, Ukraine, with regard to silver content and chosen macro-elements in the soil, tap water, and the water in Pisoczno Lake located in the vicinity of village. Materials and method. The macro-elements, silver ions contained in the lake, tap water and soil were examined. 125 people aged 12–73, indigenous inhabitants of Melniki village, were qualified for dental examinations. The research took into account the distance between the place of residence and Pisoczno Lake as a source of silver ions, dividing the examined patients into 2 groups: A) living at a distance less than 2,500 m from the lake, and B) living at a distance more than 2.500 m from the lake. Results. In area A, significantly higher contents of phosphorus and potassium were detected, while in the soil of area B there was more Ca and Mg with 3 times higher number of Mg ions. A high concentration of Ag ions was observed in both Pisoczno Lake and tap water. The severity of caries expressed by D3MFT number was 4.18 in the group aged 12–16 living in area A, and 4.24 for the inhabitants of area B. The mean value of D3MFT for the group aged 32- 45 living in area A equalled 21.58, while for area B it was 21.19. The severity of caries expressed by the mean D3MFT value in the examined group of 46–73-year-olds was 24.60 for area A, and 23.80 for area B. The observed differences were not statistically significant. Conclusions. The high value of D3MFT recorded in the inhabitants of Melniki village indicates the poor condition of mineralized tooth tissues. The fact that the contents of macro-elements in the soil and drinking water increased, together with the distance from the Pisoczno Lake shoreline, did not affect the oral health of the indigenous inhabitants. No impact of silver ions contained in the lake and tap water on the condition of mineralized tooth tissue was detected in the examined group of inhabitants of Szackie Lake District.

Opis:
This paper deals with application of 3D scanning technology in medicine. Important properties of 3D scanners are discussed with emphasize on medical applications. Construction of medical 3D scanner according to these specifications is described and practical application of its use in medical volumetry is presented. Besides volumetry, such 3D scanner is usable for many other purposes, like monitoring of recovery process, ergonomic splint manufacturing or inflammation detection. 3D scanning introduces novel volumetric method, which is compared with standard methods. The new method is more accurate compared to present ones. Principles of this method are discussed in paper and its accuracy is evaluated and experimentally verified.

W pracy przedstawiono możliwość wykorzystania metody elementów skończonych do analizy stanu naprężenia w twardych tkankach zębów, odbudowanych wypełnieniem stomatologicznym klasy I, poddanych działaniu obciążenia mechanicznego. Badania prowadzono na modelu geometrycznym odwzorowującym rzeczywistą geometrię zęba przedtrzonowego, uzyskaną z wykorzystaniem techniki mikrotomografie rentgenowskiej oraz oprogramowania CAD. Analizie poddano rozkłady naprężenia zredukowanego wyznaczonego wg hipotezy Hubera-Misesa-Hencky'ego (H-M-H) w twardych tkankach zęba, jak również podjęto próbę oceny stopnia wytężenia warstwy adhezyjnej na granicy wypełnienie kompozytowe-tkanka biologiczna. Zastosowanie symulacji numerycznych (Abaqus) umożliwia dokonanie realnej oceny stopnia wytężenia tkanek zęba, co pozwala na ocenę ryzyka niepowodzenia leczenia stomatologicznego i pomaga w opracowaniu racjonalnych metod zapobiegania uszkodzeniom zębów na skutek działania obciążeń mechanicznych.
This paper presents the possibility of applying the finite element method for the analysis of stress level in hard dental tissues, restored with class I dental filling and exposed to mechanical load. The studies were made on a geometrical model imitating the real geometry of a premolar tooth obtained using the X-ray microtomography technique and CAD software. The distributions of reduced stress defined in accordance with the Huber-Mises-Hencky (H-M-H) hypothesis in hard dental tissues were analysed, and assessment of the degree of strength of the adhesive layer at the border of the composite filling and biological tissue was attempted. The application of numerical simulations (Abaqus) enables real assessment of the tooth tissue strength, which allows assessing the risk of unsuccessful dental treatment, and helps to prepare rational methods of preventing tooth damage resulting from mechanical load.

Objectives: The study aimed to identify the patients with pathological lesions in the maxillary sinuses in which the reported symptoms might be indicative of odontogenic origin, as well as to establish specific causative risk factors promoting their development. Methods and Materials: The study covered 44 patients with suspected odontogenic maxillary sinusitis. Dental examination and Cone Beam Computed Tomography were completed. The age of patients ranged between 19 and 69 years, and the mean age was 43 (SD = 13.9) years. Results: Out of 44 patients, 22 (50%) had non-odontogenic lesions in maxillary sinuses, while in 15 (34.1%) dental origin was established. In the remaining 7 (15.9%) patients, no pathological changes were found in the sinuses. The median of reported symptoms was 10 months (Q1 = 4, Q3 = 24). The reported complaints were not associated with the actual cause of pathological lesions. Conclusions: Odontogenic cause of the lesions in the sinuses should primarily be hypothesized, especially in patients with long-term disease symptoms, also regarding any unilateral inflammations.

Abstract Streptococcus suis is a re-emerging zoonotic pathogen that may cause severe disease, mostly meningitis, in pigs and in humans having occupational contact with pigs and pork, such as farmers, slaughterhose workers and butchers. The first stage of the pathogenic process, similar in pigs and humans, is adherence to and colonisation of mucosal and/or epithelial surface(s) of the host. The second stage is invasion into deeper tissue and extracellular translocation of bacterium in the bloodstream, either free in circulation or attached to the surface of monocytes. If S. suis present in blood fails to cause fatal septicaemia, it is able to progress into the third stage comprising penetration into host’s organs, mostly by crossing the blood-brain barrier and/or blood–cerebrospinal fluid barrier to gain access to the central nervous system (CNS) and cause meningitis. The fourth stage is inflammation that plays a key role in the pathogenesis of both systemic and CNS infections caused by S. suis. The pathogen may induce the overproduction of pro-inflammatory cytokines that cause septic shock and/or the recruitment and activation of different leukocyte populations, causing acute inflammation of the CNS. Streptococcus suis can also evoke – through activation of microglial cells, astrocytes and possibly other cell types – a fulminant inflammatory reaction of the brain which leads to intracranial complications, including brain oedema, increased intracranial pressure, cerebrovascular insults, and deafness, as a result of cochlear sepsis. In all stages of the pathogenic process, S. suis interacts with many types of immunocompetent host’s cells, such as polymorphonuclear leukocytes, mononuclear macrophages, lymphocytes, dendritic cells and microglia, using a range of versatile virulence factors for evasion of the innate and adaptive immune defence of the host, and for overcoming environmental stress. It is estimated that S. suis produces more than 100 different virulence factors that could be classified into 4 groups: surface components or secreted elements, enzymes, transcription factors or regulatory systems and transporter factors or secretion systems. A major virulence factor is capsular polysaccharide (CPS) that protects bacteria from phagocytosis. However, it hampers adhesion to and invasion of host’s cells, release of inflammatory cytokines and formation of the resistant biofilm which, in many cases, is vital for the persistence of bacteria. It has been demonstrated that the arising by mutation unencapsulated S. suis clones, which are more successful in penetration to and propagation within the host’s cells, may coexist in the organism of a single host together with those that are encapsulated. Both ‘complementary’ clones assist each other in the successful colonization of host’s tissues and persistence therein. S. suis has an open pan-genome characterized by a frequent gene transfer and a large diversity. Of the genetic determinants of S. suis pathogenicity, the most important are pathogenicity islands (PAI), in particular, a novel DNA segment of 89 kb length with evident pathogenic traits that has been designated as 89K PAI. It has been estimated that more than one-third of the S. suis virulence factors is associated with this PAI. It has been proved that the virulent S. suis strains possess smaller genomes, compared to avirulent ones, but more genes associated with virulence. Overall, the evolution of the species most probably aims towards increased pathogenicity, and hence the most significant task of the current research is an elaboration of a vaccine, efficient both for humans and pigs.

Published information about anatomical studies of the lignocellulosic material derived from the rhizomes of Thaumatococcus daniellii is almost non-existent, thereby necessitating this current study. This is aimed at examining, microscopically, the anatomical features of the cell wall structure of this plant’s rhizomes. Consequently, rhizomes differentiated into sections, based on the mean height of stalks growing on each section, were harvested in the arboretum of the Department of Forest Resources and Wildlife Management, University of Benin campus, Benin City Nigeria, washed thoroughly with clean water to remove sand from the surface, placed in ethanol, taken to the laboratory and further fixed in increasingly concentrated ethanol for periods ranging from 1 to 3 hours. Using a microtome, thin slices of tissues 30μm thickness were obtained from the different sections along the rhizomes in the cross-sectional transverse and longitudinal planes, stained, mounted on glass slides and dried. Observation of tissues mounted on glass slides, including capturing of photomicrographs, were carried out by employing a photomicroscope at 40X magnification. On analysing the different photomicrographs, we saw that the anatomical features of the cell wall for lignocellulosic tissues from the different parts of the rhizome were similar in preponderance and shape in each of the planes. It is expected that the outcome of this research will contribute to the series of background information needed in utilisation and for further researches that will improve knowledge concerning this lignocellulosic material.

Sepsis is a severe generalised infection caused usually by pathogenic bacteria. It is often the cause of hospitalization and death in patients treated in intensive care and other hospital wards. Latest research brought to better understanding of patomechanisms, took place significant development of therapy heading to improvement of general patients condition treated as a basis and additionally supported by local therapy. The aim of the study was to evaluate the possibility of using the solution Decasan in the comprehensive treatment of patients necrosis of soft tissues. Material and methods. The study included 192 patients (W/M 103/89; average -aged 53.35 ± 5.36 years). According to the classification of septic states (Chicago, 1991), patients were divided into three groups: first – patients to the local form of the infection, the second – with Systemic Inflammatory Response Syndrome (SIRS), which lasted up to 72 hours and the third – patients with various forms of sepsis, SIRS in which lasted 72 hours. Results. As a result of our studies carried out in patients where the wound was made decontamination solution Decasanu, received: pain reduction, decrease tissue swelling, early debridement of the wound and the appearance in the wound granulation, to reduce delays wound healing. Proposed algorithms treatment of various forms of sepsis, pointed out the essential elements, ie.: a comprehensive approach to the treatment of infection by early surgical intervention, intensive supportive therapy (fluid resuscitation), antibiotic therapy directed to microorganisms that cause infections and topical antiseptics therapy (solution Decasanu). Conclusions. The preparation antiseptic Decasan can be safely used for disinfection of skin, mucous membranes and wounds in the foci of infections caused by bacteria, fungi and protozoa.

FT-Raman spectroscopy was used to investigate microstructural changes in the secondary protein structure of soft tissues subjected to increasing levels of macroscopic strain. Main protein bands at 938 cm-1 assigned as v(Cα–C), 1668 cm-1 — amide I and 1268 cm-1 — amide III are sensitive to applied strain and undergo wavenumber shifting. Other main vibrational modes at 1004 cm-1 assigned to the phenyl ring breathing mode and 2940 cm-1 (n(CH3,CH2)) remain unaltered. Spectroscopic results were compared with the mechanical relations obtained from the standard protocol of uniaxial tensile tests carried out in a testing machine. A clear correlation between Raman band shifting and the level of mechanical stress was established. Initially the load is transferred through elastin and then gradually also by collagen. It was proved that transferring loads by soft tissues involves changes in structural protein conformation. This process was described in detail for a tendon. It was also confirmed that mechanical properties of soft tissues depend on collagen and elastin fiers orientation.

The study focus on the occurrence and levels of polybrominated diphenyl ethers (PBDEs) as contaminants of emerging concern in Poland. In the article, their properties, primary uses and emission sources to the environment were discussed. The review of the available studies on the PBDEs content in the samples of different matrices (groundwaters, surface waters, sediments, air and dust, food, animal and human tissues) was conducted and the results exceeding the available limit values were indicated. It enabled to evaluate the problem of the PBDEs contamination in Poland.
W artykule przedstawiono dane dotyczące występowania i stężeń polibromowanych eterów difenylowych jako nowo pojawiających się zanieczyszczeń w Polsce. W artykule przedyskutowano ich właściwości, wykorzystanie oraz źródła emisji do środowiska. Przeprowadzono przegląd dostępnych badań dotyczących zawartości PBDEs w próbkach różnych matryc środowiskowych (wodach podziemnych, powierzchniowych, osadach, powietrzu i kurzu, żywności, tkanach zwierzących i ludzkich) oraz wskazano wyniki przekraczające ustalone wartości graniczne. Pozwoliło to na ocenę problemu zanieczyszczenia środowiska Polski przez PBDEs.

This work describes a dual band compact fully integrated rectenna circuit for implantable medical devices (IMDs). The implantable rectenna circuit consists of tunnel diode 10×10μm2 QW-ASPAT (Quantum Well Asymmetric Spacer Tunnel Layer diode) was used as the RF-DC rectifier due to its temperature insensitivity and nonlinearity compared with conventional SBD diode. SILVACO atlas software is used to design and simulate 100μm2 QW InGaAs ASPAT diode. A miniaturized dual band implantable folded dipole antenna with multiple L-shaped conducting sections is designed using CST microwave suits for operation in the WMTS band is 1.5GHz and ISM band of 5.8GHz. High dielectric constant material Gallium Arsenide (εr=12.94) and folded geometry helps to design compact antennas with a small footprint of 2.84mm3 (1×4.5×0.63) mm3. Four-layer human tissue model was used, where the antenna was implanted in the skin model at depth of 2mm. The 10-dB impedance bandwidth of the proposed compact antenna at 1.5GHz and 5.8GHz are 227MHz (1.4-1.63GHz) with S11 is -22.6dB and 540MHz (5.47-6.02GHz) with S11 is -23.1dB, whereas gains are -36.9dBi, and -24.3dBi, respectively. The output DC voltage and power of the rectenna using two stage voltage doubler rectifier (VDR) are twice that produced by the single stage at input RF power of 10dBm.

Opis:
Celem pracy jest scharakteryzowanie wybranych metod badań właściwości mechanicznych owoców i warzyw oraz wskazanie parametrów opisujących cechy reologiczne badanych materiałów. Badano właściwości mechaniczne jabłek, bananów, buraków ziemniaków w testach ściskania, relaksacji i cięcia. Stwierdzono, że testy wytrzymałościowe, m.in. ściskania i cięcia, odnoszące się bezpośrednio do właściwości takich jak twardość były bardzo dobrym wskaźnikiem tekstury. Uzyskane wartości naprężenia maksymalnego oraz sił cięcia oraz obrazy krzywych ściskania i cięcia wskazywały na wysoką twardość buraków i ziemniaków. Owoce takie jak jabłka i banany charakteryzowały się niższą odpornością na ściskanie i cięcie co wskazywało na większą miękkość tkanki. Materiały, takie jak, jabłko i ziemniak możemy opisać jako lepkosprężyste, ze szczególnym udziałem cech elastycznych w przypadku buraka, zaś banan wykazuje tendencje płynięcia.
The study aimed to characterize some methods used for testing mechanical properties of selected fruits and vegetables as well as to define the parameters describing rheological properties of tested materials. Mechanical properties of the apples, bananas, beet roots and potato tubers were tested by compression, relaxation and cutting tests. It was found that the strength tests, i.e. compression and cutting, related directly to the properties such as hardness, were adequate texture indicators. Obtained maximum stress and cutting force values as well as the compression and cutting curves showed high hardness of the beets and potatoes. The fruits such as apples and bananas were characterized by lower compression and cutting resistance what indicated higher softness of the tissues. The materials such as an apple or potato may be described at the viscoelastic ones, with particular share of elastic properties on case of the beet rots, whereas the bananas showed some tendencies to flowing.

Opis:
The positron annihilation lifetime spectroscopy was applied to the samples of the human uterine leiomyomas and the normal myometrium tissues taken from the selected place of the uterus during a surgery. The method indicated differences in values of the measured positron annihilation lifetime spectroscopy parameters (lifetimes and intensities) between healthy and diseased tissue samples. The additional measurements were performed either in darkness or in presence of visible light which influenced the free radicals present in both kind of tissues and, as a result, made changes in free annihilation and o-Ps decay lifetime and intensity values.

Opis:
Extrapulmonary tuberculosis (EPTB) involves 10-15% of all tuberculosis (TB) cases. Recent statistical report showed that EPTB accounts for more than 50% of all cases of TB in HIV-positive patients. In spite of existing anti-tubercular drugs based treatment of EPTB, the ideal regimen and duration of treatment have not yet been established. In general, EPTB is a kind of TB infecting diversified tissues and organs of body other than lungs. In fact, people suffering from TB and co-infected with HIV are prone to develop EPTB much more frequently. The present chapter discusses on the general overviews of EPTB infecting distinct body sites other than lungs. Currently, the treatment of EPTB completely relies on existing anti-tubercular drugs. In addition, significant efforts, particularly close clinical monitoring would be an imperative step towards its therapeutic strategy.
