Biological heart valves have represented an important area of the tissue-derived biomaterials. Decellularization processes are considered to be useful for manufacturing of biodegradable scaffolds hich make it possible to create living and functioning tissues. These processes result in elimination of most disadvantages of GA-stabilized tissues. Acellular tissues may be obtained using various chemical, enzymatic and mechanical methods. Decellularization processes give the possibility of creating biomaterials for cell seeding which are not immunogenic, cytotoxic and calcifying.

Legal bases of the cells, tissues and organs transplantationsTransplantation is an operation consisting in treatment through the exchange of tissue or an organ. Transplantation became a revolution in medicine and forced the introduction of new legal regulations. The development of medicine means that transplantation of tissues and cells as an intervention saving health and life the patient has become increasingly common. Transplantations have to be carried out in accordance with medical knowledge and the law. Taking into account the donor, there are two most important sorts of transplantation, ex mortuo (from a corpse) and ex vivo (from a living donor). The necessity to harmonize Polish law with European law has led to passing on July the 1st 2005 a new law concerning collecting, storage, grafting cells and tissues and organs. The law came into force on December the 31st. The aim of the paper is the presentation and analysis of the new „transplantation” act.

In 2007 samples taken from carps (Cyprinus carpio L.) aged between one and two years with an average weight of 430 g were tested for selenium concentration. The fish was obtained from a 5 ha pond owned by the MILICZ Fish Breeding Farm, located in the Barycz River catchment. The following organs were taken from each 15 fishes: muscles, trunk kidneys, hepatopancrea, gills, intestine and spleen. Selenium concentrations were determined using the hydride vapour generation method. The highest selenium content was found in spleen (on average 0.848 mg Se kg–1), intestine (on average 0.651 mg Se kg–1) and kidney (on average 0.603 mg Se kg–1). The medium concentration, at 0.37 mg Se kg–1, was measured in hepatopancrea. Selenium was at its lowest concentration in gills and muscles.
W 2007 roku zbadano próbki pochodzące od kroczków karpia (Cyprinus carpio L.) o średniej masie 430 g. Ryby pozyskano z 5 ha stawu Gospodarstwa Rybackiego MILICZ, położonego w zlewni rzeki Baryczy. Z każdej z 15 ryb pobrano następujące narządy: mięśnie, nerkę tułowiową, wątrobotrzustkę, skrzela, jelito i śledzionę. Określono zawartość selenu metodą generacji par wodorków (HG AAS). W wyniku przeprowadzonych badań stwierdzono najwyższą zawartość selenu w śledzionie (średnio 0,848 mg Se kg-1), jelicie (średnio 0,651 mg Se kg-1 ) oraz nerce (średnio 0,603 mg Se kg-1). Średnią koncentracją selenu na poziomie 0,37 mg Se kg-1 charakteryzowała się wątrobotrzustka. Najmniej selenu stwierdzono w skrzelach i mięśniach ryb.

Extrapulmonary tuberculosis (EPTB) involves 10-15% of all tuberculosis (TB) cases. Recent statistical report showed that EPTB accounts for more than 50% of all cases of TB in HIV-positive patients. In spite of existing anti-tubercular drugs based treatment of EPTB, the ideal regimen and duration of treatment have not yet been established. In general, EPTB is a kind of TB infecting diversified tissues and organs of body other than lungs. In fact, people suffering from TB and co-infected with HIV are prone to develop EPTB much more frequently. The present chapter discusses on the general overviews of EPTB infecting distinct body sites other than lungs. Currently, the treatment of EPTB completely relies on existing anti-tubercular drugs. In addition, significant efforts, particularly close clinical monitoring would be an imperative step towards its therapeutic strategy.

Sepsis is a severe generalised infection caused usually by pathogenic bacteria. It is often the cause of hospitalization and death in patients treated in intensive care and other hospital wards. Latest research brought to better understanding of patomechanisms, took place significant development of therapy heading to improvement of general patients condition treated as a basis and additionally supported by local therapy. The aim of the study was to evaluate the possibility of using the solution Decasan in the comprehensive treatment of patients necrosis of soft tissues. Material and methods. The study included 192 patients (W/M 103/89; average -aged 53.35 ± 5.36 years). According to the classification of septic states (Chicago, 1991), patients were divided into three groups: first – patients to the local form of the infection, the second – with Systemic Inflammatory Response Syndrome (SIRS), which lasted up to 72 hours and the third – patients with various forms of sepsis, SIRS in which lasted 72 hours. Results. As a result of our studies carried out in patients where the wound was made decontamination solution Decasanu, received: pain reduction, decrease tissue swelling, early debridement of the wound and the appearance in the wound granulation, to reduce delays wound healing. Proposed algorithms treatment of various forms of sepsis, pointed out the essential elements, ie.: a comprehensive approach to the treatment of infection by early surgical intervention, intensive supportive therapy (fluid resuscitation), antibiotic therapy directed to microorganisms that cause infections and topical antiseptics therapy (solution Decasanu). Conclusions. The preparation antiseptic Decasan can be safely used for disinfection of skin, mucous membranes and wounds in the foci of infections caused by bacteria, fungi and protozoa.

Introduction and objective. The prevalence of Molar-Incisors Hypomineralization (MIH) and difficulties in diagnosing the disease make it is necessary to determine the exact diagnostic and therapeutic patterns of management. Because of the higher requirements of patients affected by MIH, restorative management of such teeth is a challenge, both for the physician and for the suffering patient. Brief description of state of knowledge. Many long-term randomized studies and meta-analyses clearly indicate that the etiology of occurrence of molar-incisor hypomineralization is multifactorial. Most often it is a combination of systemic and genetic factors. Currently, one of the widely-discussed factors is deficiency of vitamin D as a key factor in the process of enamel development. The study describes current information about Molar-Incisor Hypomineralization, and presents a clinical approach after diagnosis according to protocol which facilities planning of the subsequent stages. Conclusions. The preventive or treatment procedure of MIH should also engage the guardians / parents of the children. The disease undiagnosed in time, due to its pain implications, can lead to problems in dental treatment. Often, the lack of awareness of dentists having no knowledge about the prevalence of MIH, leads to drawing erroneous conclusions and determining therapeutic solutions harmful to the patient, including extraction of the teeth affected by the disease.

The positron annihilation lifetime spectroscopy was applied to the samples of the human uterine leiomyomas and the normal myometrium tissues taken from the selected place of the uterus during a surgery. The method indicated differences in values of the measured positron annihilation lifetime spectroscopy parameters (lifetimes and intensities) between healthy and diseased tissue samples. The additional measurements were performed either in darkness or in presence of visible light which influenced the free radicals present in both kind of tissues and, as a result, made changes in free annihilation and o-Ps decay lifetime and intensity values.

Neutrofilowe sieci zewnątrzkomórkowe (NET) to niedawno odkryty mechanizm, dzięki któremu neutrofile mogą skutecznie walczyć z patogenami. W wyniku aktywacji neutrofile wyrzucają zdekondensowany DNA, połączony z histonami i białkami pochodzącymi z ziarnistości. Te przestrzenne struktury mogą wyłapywać, ograniczać rozprzestrzenianie się, a w niektórych przypadkach zabijać patogeny. Pomimo korzystnych efektów, NET są także odpowiedzialne za patogenezę różnych chorób autoimmunizacyjnych, takich jak reumatoidalne zapalenie stawów, łuszczyca czy toczeń rumieniowaty układowy, a także przyczyniają się do uszkodzeń narządów w trakcie sepsy. Jak dotąd niewiele wiadomo o tym jak NET są usuwane z naczyń krwionośnych i narządów oraz przez które komórki. Istniejące, nieliczne doniesienia wskazują na udział makrofagów w usuwaniu NET, jednakże wyniki te pochodzą tylko z eksperymentów in vitro. Otwartym pozostaje zatem pytanie o mechanizmy tego procesu w skomplikowanym środowisku żywego organizmu. W związku z patologicznym aspektem tworzenia NET, ważnym będzie opracowanie skutecznego środka farmakologicznego zdolnego do usuwania tych struktur.
Neutrophil extracellular traps (NET) represent a recently discovered mechanism by which neutrophils can efficiently fight pathogens. Upon activation, neutrophils release decondensed extracellular DNA decorated with histones and granular proteins. These three-dimensional structures can trap pathogens, limit their spread and sometimes kill them. Despite their beneficial effects, NETs are also involved in pathogenesis of various autoimmune diseases, such as rheumatoid arthritis, psoriasis or systemic lupus erythematosus, and also contribute to organ damage during sepsis. So far, little is known on how NETs are removed from vasculature and tissues, and by which cells. Limited available studies indicate that macrophages might remove NETs, but these results were obtained in vitro. Thus it remains unknown how this process occurs in a complex milieu of the body. Due to the pathological aspect of NET formation, a challenge for the near future will be development of pharmacological agents capable of NET removal.

Objectives: The study aimed to identify the patients with pathological lesions in the maxillary sinuses in which the reported symptoms might be indicative of odontogenic origin, as well as to establish specific causative risk factors promoting their development. Methods and Materials: The study covered 44 patients with suspected odontogenic maxillary sinusitis. Dental examination and Cone Beam Computed Tomography were completed. The age of patients ranged between 19 and 69 years, and the mean age was 43 (SD = 13.9) years. Results: Out of 44 patients, 22 (50%) had non-odontogenic lesions in maxillary sinuses, while in 15 (34.1%) dental origin was established. In the remaining 7 (15.9%) patients, no pathological changes were found in the sinuses. The median of reported symptoms was 10 months (Q1 = 4, Q3 = 24). The reported complaints were not associated with the actual cause of pathological lesions. Conclusions: Odontogenic cause of the lesions in the sinuses should primarily be hypothesized, especially in patients with long-term disease symptoms, also regarding any unilateral inflammations.

Introduction and objective. The aim of the study is to evaluate the mineralization tooth tissues in the inhabitants of Melniki village Szackie, Lake District, Ukraine, with regard to silver content and chosen macro-elements in the soil, tap water, and the water in Pisoczno Lake located in the vicinity of village. Materials and method. The macro-elements, silver ions contained in the lake, tap water and soil were examined. 125 people aged 12–73, indigenous inhabitants of Melniki village, were qualified for dental examinations. The research took into account the distance between the place of residence and Pisoczno Lake as a source of silver ions, dividing the examined patients into 2 groups: A) living at a distance less than 2,500 m from the lake, and B) living at a distance more than 2.500 m from the lake. Results. In area A, significantly higher contents of phosphorus and potassium were detected, while in the soil of area B there was more Ca and Mg with 3 times higher number of Mg ions. A high concentration of Ag ions was observed in both Pisoczno Lake and tap water. The severity of caries expressed by D3MFT number was 4.18 in the group aged 12–16 living in area A, and 4.24 for the inhabitants of area B. The mean value of D3MFT for the group aged 32- 45 living in area A equalled 21.58, while for area B it was 21.19. The severity of caries expressed by the mean D3MFT value in the examined group of 46–73-year-olds was 24.60 for area A, and 23.80 for area B. The observed differences were not statistically significant. Conclusions. The high value of D3MFT recorded in the inhabitants of Melniki village indicates the poor condition of mineralized tooth tissues. The fact that the contents of macro-elements in the soil and drinking water increased, together with the distance from the Pisoczno Lake shoreline, did not affect the oral health of the indigenous inhabitants. No impact of silver ions contained in the lake and tap water on the condition of mineralized tooth tissue was detected in the examined group of inhabitants of Szackie Lake District.

SNP-minisequencing has become common in forensic genetics, especially for analysing degraded or low copy number DNA (LCN DNA). The aim of this study was to examine the usefulness of five SNP (single nucleotide polymorphism) markers for analyzing degraded and LCN DNA recovered from archival samples. DNA extractions of eight formalin-fixed paraffin-embedded (FFPE) tissues were performed and DNA fragments were amplified in one multiplex PCR (polymerase chain reaction). SNPs were identified in a minisequencing reaction and a gel electrophoresis in ABI Prism® 377 Sequencer. The research confirmed the usefulness of SNP-minisequencing for analysing FFPE tissues.

Opis:
Published information about anatomical studies of the lignocellulosic material derived from the rhizomes of Thaumatococcus daniellii is almost non-existent, thereby necessitating this current study. This is aimed at examining, microscopically, the anatomical features of the cell wall structure of this plant’s rhizomes. Consequently, rhizomes differentiated into sections, based on the mean height of stalks growing on each section, were harvested in the arboretum of the Department of Forest Resources and Wildlife Management, University of Benin campus, Benin City Nigeria, washed thoroughly with clean water to remove sand from the surface, placed in ethanol, taken to the laboratory and further fixed in increasingly concentrated ethanol for periods ranging from 1 to 3 hours. Using a microtome, thin slices of tissues 30μm thickness were obtained from the different sections along the rhizomes in the cross-sectional transverse and longitudinal planes, stained, mounted on glass slides and dried. Observation of tissues mounted on glass slides, including capturing of photomicrographs, were carried out by employing a photomicroscope at 40X magnification. On analysing the different photomicrographs, we saw that the anatomical features of the cell wall for lignocellulosic tissues from the different parts of the rhizome were similar in preponderance and shape in each of the planes. It is expected that the outcome of this research will contribute to the series of background information needed in utilisation and for further researches that will improve knowledge concerning this lignocellulosic material.

Opis:
Objectives Laser ablation inductively coupled plasma mass spectrometry (LA-ICP-MS) has considerable applicative potential for both qualitative and quantitative analyses of elemental spatial distribution and concentration. It provides high resolutions at pg-level detection limits. These qualities make it very useful for analyzing biological samples. The present study responds to the growing demand for adequate analytical methods which would allow to assess the distribution of nanostructured molybdenum(IV) disulfide (MoS₂) in organs. It was also motivated by an apparent lack of literature on the biological effects of MoS₂ in living organisms. The study was aimed at using LA-ICP-MS for comparing micro- and nanosized MoS2 ditribution in selected rat tissue samples (lung, liver, brain and spleen tissues) after the intratracheal instillation (7 administrations) of MoS₂ nano- and microparticles vs. controls. Material and Methods The experimental study, approved by the Ethics Committee for Animal Experiments was performed using albino Wistar rats. This was performed at 2-week intervals at a dose of 5 mg/kg b.w., followed by an analysis after 90 days of exposure. The MoS₂ levels in control tissues were determined with the laser ablation system at optimized operating conditions. The parameter optimization process for the LA system was conducted using The National Institute of Standards and Technology (NIST) glass standard reference materials. Results Instrument parameters were optimized. The study found that molybdenum (Mo) levels in the lungs of microparticle-exposed rats were higher compared to nanoparticle-exposed rats. The opposite results were found for liver and spleen tissues. Brain Mo concentrations were below the detection limit. Conclusions The LA-ICP-MS technique may be used as an important tool for visualizing the distribution of Mo on the surface of soft samples through quantitative and qualitative elemental mapping.
