A total of 94 pectolytic and 60 nonpectolytic Pseudomonas isolates were obtained from 250 samples of rotted vegetable specimens representing various economically important vegetables. The isolates were identified on the basis of standard biochemical tests. Pseudomonas fluorescens biovar V and II and Pseudomonas putida were the most abundant species among pectolytic isolates and Pseudomonas fluorescens biovar I among nonpectolytic ones. Only 3 Pseudomonas viridiflava isolates were identified and all of them were obtained from potato. Isolates of pectolytic phenotype were scattered among nonpectolytic ones irrespective of their taxonomical status. Isolates identified biochemically, as Pseudomonas marginalis were also present in nonpectolytic group. PCR method is unsuitable for identification and differentiation of bacteria belonging to pectolytic fluorescens Pseudomonas group due to great diversity of species. However, the results of PCR amplification of the genes encoding pectate lyase suggest that genes responsible for production of this enzyme may also be present in isolates of nonpectolytic phenotype.
Z 250 prób, z różnych gatunków roślin warzywnych z objawami mokrej zgnilizny wyodrębniono 94 izolaty pektynolitycznych i 60 izolatów nie pektynolitycznych bakterii z rodzaju Pseudomonas. Identyfikację i różnicowanie izolatów przeprowadzono przy zastosowaniu standardowych testów fizjologiczno-biochemicznych. Spośród izolatów z grupy pektolitycznych Pseudomonas najliczniej występował gatunek P. fluorescens, który był reprezentowany przez 4 biowary (oprócz czwartego), przy czym dominowały biowary II i V. Mniej licznie występował gatunek P. putida, a P. viridiflava był reprezentowany jedynie przez 3 izolaty, pochodzące z ziemniaka. Podobna była struktura populacji izolatów z grupy niepektolitycznych Pseudomonas. Metoda PCR okazała się być mało przydatna do wykrywania i różnicowania izolatów z grupy pektolitycznych Pseudomonas ze względu na duże ich zróżnicowanie. Amplifikacja DNA metodą PCR wykazała, że geny kodujące enzym liazy pektynowej występują także w izolatach niepektolitycznych rodzaju Pseudomonas.

Due to the importance of the biological control of plant diseases, testing and introducing new biocontrol-active microorganisms is a major concern among plant pathologists. The causal agent of cotton seedling damping-off disease is Rhizoctonia solani. In this regard, we tried to investigate the antagonistic activities of Pseudomonas aureofaciens (chlororaphis) 30–84 (phenazine producing wild type and non-phenazine producing mutant) strains on R. solani, in comparison with some isolates of P. fluorescent under both in vitro (laboratory) and in vivo (greenhouse) conditions. In the laboratory experiment, the inhibitory effects of all the bacteria, on the growth of R. solani, were evaluated using the dual culture procedure. Results showed that five isolates of P. fluorescent along with both strains of P. aureofaciens significantly inhibited the growth of R. solani. Effective bacterial antagonists were then evaluated in a greenhouse experiment where cotton seeds were coated with their suspensions and were sown in pasteurised field-soil. The soil had been pre-inoculated with a virulent isolate of R. solani. The efficacy of the bacterial antagonists was evaluated by counting the number of surviving seedlings in different treatments, at 15 and 60 days after sowing, for determining pre- and post-emergence damping-off incidence. According to the results of the greenhouse experiment, at both intervals, two isolates of P. fluorescens along with both strains of P. aureofaciens caused significant increases in the number of healthy seedlings, in comparison with the untreated control, and a commonly used fungicide (carboxin-thiram). The efficacy of phenazine producing a wild type strain of P. aureofaciens was higher than its non-phenazine producing mutant, indicating that phenazine plays an important role in the antagonistic activity of P. aureofaciens. Effective bacterial antagonists were then studied for their antagonistic mechanisms. The results showed that all four bacteria employed different mechanisms. The bacteria produced siderophore, and volatile metabolites and non-volatile metabolites, in their antagonistic activities. The results of this study suggest that P. auerofaciens may be a new biocontrol agent for controlling cotton seedling mortality disease.

An agar degrading bacterium Pseudomonas aeruginosa AG LSL-11 was acclimatized to alginate for the production of alginase. Production parameters such as pH, temperature, influence of simple carbohydrates and nitrogen sources, and effect of NaCl on growth and alginase production were carried out. Maximum growth was observed at pH 9.0 and 35 °C, while alginase was produced optimally at pH 9.0 and 30 °C. The alginase produced by Pseudomonas aeruginosa AG LSL-11 was inducible by alginate, and repressed by other simple sugar when supplemented along with alginate in the medium. The bacterium did not require NaCl for growth and production of alginase. The activity staining of partially purified culture supernatant after native PAGE revealed the presence of a single alginase.

W niekorzystnych warunkach środowiska, mikroorganizmy zasiedlają zarówno powierzchnie abiotyczne, jak i biotyczne takie jak tkanki zwierzęce czy roślinne, tworząc struktury biofilmu charakteryzujące się wysoką opornością. Adhezja mikroorganizmów, szczególnie patogenów oportunistycznych, niesie niebezpieczeństwo zasiedlania materiałów medycznych, co może doprowadzić do infekcji u osób z obniżoną odpornością. Chociaż dotychczasowe badania wskazują różne metody zapobiegania tworzeniu biofilmu, jego całkowita eliminacja ze środowiska jest nadal niemożliwa. Przedstawione opracowanie stanowi przegląd nowoczesnych metod usuwania dojrzałego biofilmu tworzonego przez patogeny oportunistyczne. Spośród wielu metod opisano m.in. zastosowanie: zimnej plazmy, ultradźwięków, pola elektrycznego, ozonowania wody, terapii fagowej, enzymów działających bezpośrednio na macierz biofilmu, bakteriocyn, środków chemicznych syntetycznych oraz pochodzenia naturalnego.
The inconvenient environmental conditions force microorganisms to colonize either abiotic surfaces or animal and plant tissues and, therefore, form more resistant structures – biofilms. The phenomenon of microbial adherence, opportunistic pathogens in particular, is of a great concern. Colonization of medical devices and biofilm formation on their surface, may lead to severe infections mainly in humans with impaired immune system. Although, current research consider various methods for prevention of microbial biofilms formation, still, once a biofilm is formed, its elimination is almost impossible. This study focuses on the overview of novel methods applied for eradication of mature opportunistic pathogens' biofilms. Among various techniques the following: cold plasma, electric field, ultrasounds, ozonated water treatment, phagotherapy, matrix targeting enzymes, bacteriocins, synthetic chemicals and natural origin compounds used for biofilm matrix disruption were briefly described.

The culture conditions for the production of carrageenase were optimized using one-factor-at-a-time method combined with orthogonal array design. With the one-factor-at-a-time method revealed optimal conditions for carrageenase production were 24 h of fermentation period, 28 °C incubation temperature at pH 8.0 with NaNO3 as nitrogen source and carrageenan as carbon source in MMS media. Further optimization of carrgeenase production by using orthogonal experimental design L9 (34) with four factors, temperature, pH, NH4NO3 and carrageenan with their relevant levels revealed optimised conditions for carrageenase production were temperature of 28 °C, pH 8.0, 2 g L-1 NaNO3 and 2 g L-1 carrageenan. The order of the factors affecting the fermentation process was found to be temperature > pH > NaNO3 > carrageenan. The temperature played a significant role on the carrageenase production. Higher carrageenase yield with activity of 0.542 ±0.045 U ml-1 was obtained in the optimised medium when compared to those of basal medium. Carrageenase hydrolysed products of carrageenan were identified by LC-ESI-MS as neocarrabiose, neocarrabiose-4 sulfate, neocarratetraose, neocarratetraose-4 sulfate, anhydrogalactose, galactose, galactose-4 sulphate and sulphate.

This study was conducted to investigate the degree of bioremediation that would occur in the samples of soil polluted with raw and treated crude petroleum (oil) with the aid of Aspergillus niger (fungi) and Pseudomonas aeruginosa (bacteria). This was achieved by monitoring the Organic carbon content in the soil over 45 days and pH over 25 days. Four systems of 500 g soil were polluted with 40 g treated crude petroleum, while four systems were contaminated with 40 g raw crude petroleum. The eight systems were labeled accordingly. Two systems for raw crude control and treated crude control (RCC and TCC) were left as control, two systems for raw crude Aspergillus niger and treated crude Aspergillus niger (RCA and TCA) were treated with Aspergillus niger only, two systems for raw crude Pseudomonas aeruginosa and treated crude Pseudomonas aeruginosa (RCP and TCP) were treated with Pseudomonas aeruginosa only and the last two systems for raw crude Aspergillus niger and Pseudomonas aeruginosa (RCAP and TCAP) were treated with both Pseudomonas aeruginosa and Aspergillus niger. At the end of the bioremediation period, the results obtained showed that the pH was not particularly a solid parameter to estimate the degree of bioremediation. This is because a proper trend in the results obtained could not be determined. Additionally, it was observed that Aspergillus niger (fungi) and Pseudomonas aeruginosa (bacteria) alone and separate remediate raw crude polluted soil better than treated crude polluted soil. This was determined by the TOC (Total Organic Carbon) values on the 45th day of the experiment.

Biofilms are highly organized microbial communities displaying high resistance to disinfectants and other external environmental factors. Medical equipment, such as stents and catheters, can be colonized by a variety of bacteria including opportunistic pathogens circulating in the environment and dangerous to immunocompromised patients. Application of materials resistant to biofilm formation will minimize the risk of patients' infection. Hence, the aim of this research was to determine the biofilm growth of environmental bacteria isolates on polyvinyl chloride and styrene-acronitrile copolymer surfaces. Nine strains (Pseudomonas aeruginosa, Burkholderia cepacia and Serratia liquefacies) isolated from cosmetics, and a reference P. aeruginosa strain ATCC 15442, were tested. The ability and dynamics of biofilm formation on intubation catheters (30°C, up to 24 h) in bacterial growth cultures (107-108 CFU/ml) was investigated, with subsequent sonication and quantification by agar plate count method. The results indicated that all the tested bacteria expressed a strong ability for the polymer surface adhesion, reaching 4.6 to 6.7 log CFU/cm2 after 30 minutes. Moreover, for the majority of strains, the level of 24-hour biofilm production was from 6.67-7.61 log CFU/cm2. This research indicates that the environmental strains circulating between the cosmetics and patients may pose a threat of biofilm formation on medical equipment surfaces, and presumably in the clinical surroundings as well.

A study was conducted to determine the Production of cellulase and pectinase enzyme by using Plant growth promoting rhizobacteria like Pseudomonas fluorescence and Bacillus subtilis. These to micro organism are isolated by serial dilution method. One gram of soil sample was diluted in to 10 ml of sterile distilled water and 1 ml of sample solution was serially diluted in 9ml of sterile water up to 10 dilution. Each sample from dilution 10-5 and 10-6 were taken and streaked in to KB and NA medium and incubate at 24 hrs. After 24 hrs Pseudomonas fluorescence and Bacillus subtilis was observed in the medium of KB and NA medium. Both the culture was sub cultured and maintain in the same for the further work. CMCase medium was prepared and sterilized by autoclave for 121 ºC for 15 minutes after sterilization these medium contain petriplate was streaked by bacteria and incubates for 48h after incubation period a clear halo zone was produced by these bacteria among these bacteria Pseudomonas fluorescence are able to produce high amount of cellulose compare to Bacillus subtilis. Pectin agar medium was prepared and sterilized by autoclave for 121 ºC for 15 minutes after sterilization these medium contain petriplate was streaked by bacteria incubates for 48h after incubation period a clear halo zone was produced by these bacteria, among these bacteria Pseudomonas fluorescence are able to produce high amount of Pectinase compare to Bacillus subtilis. Plant growth promoting rhizobacteria (PGPR) are beneficial bacteria that colonize plant roots and enhance plant growth by a wide variety of mechanisms.

The suitability of low-cost impedance sensors for microbiological purposes and biofilm growth monitoring was evaluated. The sensors with interdigitated electrodes were fabricated in PCB and LTCC technologies. The electrodes were golden (LTCC) or gold-plated (PCB) to provide surface stability. The sensors were used for monitoring growth and degradation of the reference ATCC 15442 Pseudomonas aeruginosa strain biofilm in invitro setting. During the experiment, the impedance spectra of the sensors were measured and analysed using electrical equivalent circuit (EEC) modelling. Additionally, the process of adhesion and growth of bacteria on a sensor’s surface was assessed by means of the optical and SEM microscopy. EEC and SEM microscopic analysis revealed that the gold layer on copper electrodes was not tight, making the PCB sensors susceptible to corrosion while the LTCC sensors had good surface stability. It turned out that the LTCC sensors are suitable for monitoring pseudomonal biofilm and the PCB sensors are good detectors of ongoing stages of biofilm formation.

Celem badań było określenie, jakie metabolity, bioaktywne względem wybranych fitopatogenów zbóż, są syntetyzowane przez wyizolowane z ryzosfery pszenicy bakterie Pseudomonas. Do doświadczeń wybrano dwa różne szczepy bakterii Pseudomonas fluorescens, które charakteryzowały się w warunkach in vitro silnym antagonizmem w stosunku do fitopategenów zbóż z rodzaju Pyrenophora Alteria i Fusarium. Jak wykazały badania obydwa szczepy bakterii w warunkach doświadczeń intensywnie wydzielały cyjanowodór oraz siderofory, w tym piowerdynę. Kwas salicylowy lub jego pochodną wykryto w hodowli tylko jednego z badanych izolatów. Obydwa zastosowane w doświadczeniu szczepy P. fluorescens charakteryzowały się zdolnością do syntezy zewnątrzkomórkowych peptydaz o różnym poziomie aktywności. W hodowlach bekterii nie wykryto enzymów litycznych zdolnych do rozkładu wiązań glikozydowych - chitynazy i ß-1,3 glukanazy.
The study was aimed at estimating which metabolites (bioactive towards selected cereal plant phatogens) are synthesized by Pseudomonas isolated from wheat rhizosphere. Two different strains of Pseudomonas fluorescens characterised by the strong in vitro antagonism in relation to cereal plant phatogens of the genera Pyrenophora , Alterial and Fusarium were selected for experiments. Both bacterial strains under experimental conditions intensively released hydrogen cyanide and siderophores including pyoverdine. Salicylic acid and its derivative were found in the culture of only one of analysed isolates. Both strains of P. fluoroscens were able to synthesize extracellular peptidases of different activity. No hydrolysing enzymes able to decompose glycoside bonds (chitinase and ß-1.3 gluconase) were found bacterial cultures.

In the study we tested drug sensitivity to 3 carbapenems (doripenem, imipenem and meropenem) of Gram-negative clinical isolates from Southern Poland. Material and methods. 89 strains were examined: 42 from Pseudomonas genus, 16 Acinetobacter baumannii strains and 31 Enterobacteriaceae strains. Etests were used according to the producers instructions, MIC values were interpreted using EUCAST criteria. Results. Highest in vitro activity against Pseudomonas spp. was shown for doripenem, then meropenem and the lowest for imipenem (MIC values were definitely lower for doripenem; differences were statistically significant); A. baumannii strains showed similar sensitivity to doripenem, meropenem and imipenem (differences non-significant); all Enterobacteriaceae strains showed sensitivity to the tested antimicrobials. Conclusions. As a conclusion-doripenem, which has high in vitro activity (almost the same as imipenem and meropenem) as well as beneficial pharmacologic properties, may be an alternative solution in the treatment of multiresistant Gram-negative bacteria, especially in patients in severe status who require restrictive antibiotic regimens.

Pseudomonas aeruginosa infection is problematic in patients with cystic fibrosis (CF). P. aeruginosa secretes a diversity of pigments, such as pyocyanin and pyoverdine. The aim of this study was to evaluate the effects of complexes of nickel(II) ([Ni(iaa)2(H2O)2]·H2O (iaa = imidazole-4-acetate anion), [Ni(1-allim)6](NO3)2 (1-allim = 1-allylimidazole) and NiCl2 on pyocyanin and pyoverdine production by 23 strains of P. aeruginosa isolated from cystic fibrosis under growth conditions specific for the CF respiratory system. The antibacterial effects and biophysical properties of the tested substances were measured by spectrofluorometric techniques, as well as by laser interferometry, confocal and atomic force microscopy. The cytotoxic properties of all compounds were measured by Annexin/IP assay against A549 cells. All tested compounds have no effect on pyocyanin production and decrease the pyoverdine secretion in about 40% of tested P. aeruginosa strains at non-cytotoxic range of concentrations. Imidazole-4-acetate anion and 1-allylimidazole have good diffusion properties in the mature P. aeruginosa PAO1 biofilm. In conclusion, the tested nickel(II) complexes do not have clinical implications in P. aeruginosa eradication in cystic fibrosis. The diffusion properties of 1-allylimidazole and imidazole-4-acetate and their lack of effect on A549 cells suggest that they might be considered for chemical synthesis with other transition metals.

Phenol is one of the main pollutants that have a serious impact on the environment and can even be very critical to human health. The biodegradation of phenol can be considered an increasingly important pollution control process. In this study, the degradation of phenol by Pseudomonas fredriksbergsis was investigated for the first time under different growth conditions. Six different initial concentrations of phenol were used as the primary substrate. Culture conditions had an important effect on these cells' ability to biodegrade phenol. The best growth of this organism and its highest biodegradation level of phenol were noticed at pH 7, temperature 28 °C, and periods of 36 and 96 h, respectively. The highest biodegradation rate was perceived at 700 mg/L initial phenol concentration. Approximately 90% of the phenol (700 mg/L) was removed in less than 96 hours of incubation time. It was found that the Haldane model best fitted the relationship between the specific growth rate and the initial phenol concentration, whereas the phenol biodegradation profiles time could be adequately described by the modified Gompertz model. The parameters of the Haldane equation are: $0.062 h^{−1}$, 11 ppm, and 121 ppm for Haldane’s maximum specific growth rate, the half-saturation coefficient, and the Haldane’s growth kinetics inhibition coefficient, respectively. The Haldane equation fitted the experimental data by minimizing the sum of squared error (SSR) to $1.36×10^{-3}$.

Chromium in wastewater is classified as one of the dangerous contaminants that require further treatment before being discharged to water body. The concentration of chromium in water body, especially river, has increased as many industries utilize chromium as raw material and then discharge their wastewater without any treatment. Biosorption is one of methods that are widely used to treat heavy metal containing wastewater. Bacteria are the most common microorganisms to be used as heavy metal treatment agent. Azotobacter s8, Bacillus subtilis and Pseudomonas putida had been proven to have a heavy metal resistant capability. The screening test showed that Minimum Inhibitory Concentration (MIC) value of chromium for all bacteria ranged from 100 to 250 mg/L of CrCl3. The chromium biosorption test by bacteria showed that Azotobacter s8 was able to remove 10.53%, and Bacillus subtilis was able to remove 5.68% chromium from 50 mg/L initial concentration, while Pseudomonas putida showed no chromium removal. The chromium biosorption capacity by Azotobacter s8 was 580.08 mg/g and 349.30 mg/g for Bacillus subtilis.

Zinc is a vital micronutrient for all life forms, and Zn-solubilizing bacteria (ZSB) present in the soil convert inorganic zinc into forms available for plants. This study assessed ZSB isolated from cow dung for their plant growth-promoting (PGP) characteristics and potential to enhance tomato plant growth. The experiment assayed a total of 30 bacteria from cow dung for Zn-solubilization using insoluble ZnO and ZnCO3. Atomic absorption spectroscopy quantitatively evaluated Zn-solubilization, and the isolates were further studied for Zn-solubilization and plant growth in Solanum lycopersicum. The CDS7 and CDS27 isolates were the most significant Zn solubilizing strains. CDS7 exhibited increased ZnO solubility (32.1 mg/l) compared to CDS21 (23.7 mg/l). PGP trait quantitative results revealed that the CDS7 and CDS21 bacterial strains solubilized insoluble phosphate (287.2 and 217.7 μg/ml, respectively) and produced indole acetic acid (22.1 and 14.8 μg/ml, respectively). Based on 16S rRNA gene sequencing, CDS7 and CDS21 were identified as Pseudomonas kilonensis and Pseudomonas chlororaphis, and 16S rDNA sequences were submitted to the GenBank database. Furthermore, ZSB strains were administered to tomato seeds under a pot study. The treatments with CDS7 inoculant and a consortium of both isolates were reported with maximum plant development (stem length 63.16 and 59.89 cm, respectively) and zinc content (3.13 and 2.36 mg/100 g, respectively) in tomato fruit compared to the control. In conclusion, microorganisms isolated from cow dung with PGP activity can improve Zn bioavailability and plant growth sustainably. They can be used as biofertilizers in agricultural fields to improve plant growth and production.

Aim: 1. To assess the prevalence of cross-infections with P. aeruginosa in order to evaluate the epidemiological situation of this infection in patients with cystic fibrosis attending our centre; 2. To correlate the clinical features of the patients carrying a potentially transmissible strain with the entire study group in order to determine the risk factors and possible effects of its acquisition. Material and methods: 170 Pseudomonas aeruginosa strains obtained from the respiratory tract of 75 cystic fibrosis patients attending the Warsaw Centre in 2011 and 2012 were typed using restriction enzyme analysis-pulsed field gel electrophoresis (Spe I restriction enzyme was used). Simultaneously, the information concerning contacts between patients, as well as several clinical data regarding the course of the disease were collected. Results: Twenty four clusters of strains were detected. The main cluster included 49 isolates derived from 21 patients. The other detected clusters included 2 to 12 isolates derived from 1 to 7 patients. Three clusters comprised the isolates derived from three pairs of siblings. There were 15 clusters containing 2 to 7 strains belonging to the same patient. The remaining 24 patients were infected with their own strains, not fitting any clonal group. Several clinical parameters showed that the 21 patients whose strains constituted the main cluster, were in worse clinical condition than the other patients in the study group. Moreover, the total duration of their hospitalizations in order to perform intravenous antibiotic treatment was longer. Conclusions: 1. Frequent hospitalizations of CF patients with a more severe course of the disease seem to be a risk factor of cross-infections with P. aeruginosa. 2. Intensification of measures to prevent cross-infection, such as hygienic precautions, patient segregation, introduction of home intravenous antibiotic therapy programme, as well as further education of patients and their parents should lead to the improvement of the epidemiological situation in our centre.
Cel: 1. Ocena częstości występowania zakażeń krzyżowych Pseudomonas aeruginosa i ich możliwych konsekwencji klinicznych u pacjentów chorujących na mukowiscydozę leczonych w naszym ośrodku; 2. Porównanie cech klinicznych grupy pacjentów, u których doszło do zakażenia krzyżowego z cechami klinicznymi całej grupy badanej, w celu określenia czynników ryzyka i możliwych skutków takiego zakażenia. Materiał i metody: 170 szczepów Pseudomonas aeruginosa wyizolowanych w latach 2011-2012 z dróg oddechowych 75 pacjentów z mukowiscydozą pozostających pod opieką Instytutu Matki i Dziecka typowano metodą makrogenomowej analizy restrykcyjnej z elektroforezą w zmiennym polu elektrycznym (z wykorzystaniem endonukleazy Spe I). Zbierano wywiady dotyczące najważniejszych parametrów klinicznych, a także kontaktów pomiędzy pacjentami. Wyniki: W toku analizy wyodrębniono 24 grupy klonalne. Największa grupa zawierała 49 izolatów pochodzących od 21 chorych. Inne grupy obejmowały od 2 do 12 izolatów pochodzących od 1 do 7 pacjentów. Trzy grupy zawierały szczepy pochodzące od trzech rodzeństw. Piętnaście grup klonalnych obejmowało od 2 do 7 szczepów należących do tego samego pacjenta. Pozostałych 24 pacjentów było zakażonych szczepami, nie należącymi do żadnej grupy klonalnej. Ocena parametrów klinicznych wykazała, że przebieg choroby u 21 pacjentów, których szczepy stanowiły największą grupę klonalną, był cięższy niż u pozostałych pacjentów w grupie badanej. Także łączny czas trwania ich hospitalizacji (antybiotykoterapii dożylnych) był dłuższy. Wnioski: 1. Częste hospitalizacje pacjentów z mukowiscydozą w gorszym stanie klinicznym wydają się być istotnym czynnikiem ryzyka zakażeń krzyżowych Pseudomonas aeruginosa. 2. Intensyfikacja działań zapobiegających zakażeniom krzyżowym, takich jak środki higieny, izolacja chorych, wprowadzenie w Polsce programu domowej antybiotykoterapii dożylnej, a także dalsza edukacja pacjentów i ich rodziców odnośnie dróg transmisji zakażeń bakteryjnych, prawdopodobnie przyczynią się do poprawy sytuacji epidemiologicznej w naszym ośrodku.

Quaternary ammonium salts (QAS) belong to surfactant commonly used both, in the household and in different branches of industry, primarily in the process of cleaning and disinfection. They have several positive features inter alia effectively limiting the development of microorganisms on many surfaces. In the present work, two compounds were used as biocides: hexamethylene-1,6-bis-(N,N-dimethyl-N-dodecylammonium bromide) that belongs to the gemini surfactant (GS), and its single analogue - dodecyl(trimethyl)ammonium bromide (DTAB). Two fold dilution method was used to determine the minimum concentration of compounds (MIC) which inhibit the growth of bacteria: Staphylococcus aureus (ATCC 6538 and an environmental strain), Pseudomonas aeruginosa (ATCC 85327 and an environmental strain), and yeast Candida albicans (ATCC 11509 and an environmental strain). The viability of cells in liquid cultures with addition of these substances at ¼ MIC, ½ MIC and MIC concentrations were also determined. The obtained results show that DTAB inhibits the growth of bacteria at the concentration of 0.126-1.010 µM/ml, and gemini surfactant is active at 0.036-0.029 µM/ml. Therefore, GS is active at more than 17-70-fold lower concentrations than its monomeric analogue. Strains isolated from natural environment are less sensitive upon testing biocides than the references strains. Both compounds at the MIC value reduced the number of cells of all strains. The use of too low concentration of biocides can limit the growth of microorganisms, but often only for a short period of time in case of special environmental strains. Later on, they can adapt to adverse environmental conditions and begin to evolve defence mechanisms.

The aim of the work was to elaborate new method of wool entrapping into silica alginate beads and checking of their adsorption properties. The purpose of the present study was also to determine water bioremediation efficiency using Pseudomonas aeruginosa cells immobilized into wool entrapped into silica-alginate beads. The results of study showed that obtained capsules are stable in saline water environment. Additionally neither silica adsorbers nor wool impacted negatively on viability of bacterial strains that were entrapped into these capsules. Moreover, it should be underline that 83,01% of adsorbed engine oil has been degraded by immobilized microorganisms after 3 months of biodegradation process. The results proved that properties and non-toxic character of these beads are suitable enough to use them in purification of water from petroleum hydrocarbons.

Bioremediation is based on microorganisms able to use pollutants either as a source of carbon or in co-metabolism, and is a promising strategy in cleaning the environment. Using soil contaminated with petroleum products from an industrial area in Saudi Arabia (Jubail), and after enrichment with the polycyclic aromatic hydrocarbon (PAH) naphthalene, a Methylobacterium radiotolerans strain (N7A0) was isolated that can grow in the presence of naphthalene as the sole source of carbon. M. radiotolerans is known to be resistant to gamma radiation, and this is the first documented report of a strain of this bacterium using a PAH as the sole source of carbon. The commonly reported Pseudomonas aeruginosa (strain N7B1) that biodegrades naphthalene was also identified, and gas chromatography analyses have shown that the biodegradation of naphthalene by M. radiotolerans and P. aeruginosa did follow both the salicylate and phthalate pathways.

Szczepy bakterii Stenotrophomonas maltophilia KB2 oraz Pseudomonas putida N6 odznaczają się zwiększoną zdolnością do degradacji związków aromatycznych. W badaniach stwierdzono całkowity rozkład fenolu (3 mmol/dm3) w ciągu pięciu godzin przez oba badane szczepy. U szczepu KB2 po indukcji fenolem wykazano obecność 2,3-dio-ksygenazy katecholowej odpowiedzialnej za meta-rozszczepienie związków aromatycznych, natomiast u szczepu Pseudomonas putida N6 wykazano obecność 1,2-dioksygenazy katecholowej, charakterystycznej dla szlaku orto rozszczepienia pierścienia aromatycznego. W wyniku badań nad wrażliwością tych enzymów na obecność jonów metali wykazano, że jony Zn2+ aktywowały 2,3-dioksygenazę katecholową szczepu KB2. Wszystkie pozostałe jony były inhibitorami tego enzymu. Spośród przebadanych jonów metali najsilniejszym inhibitorem obu wyizolowanych dioksygenaz okazał się jon Cu2+, natomiast w mniejszym stopniu aktywność 1,2-dioksygenazy katecholowej szczepu N6 hamowały Cd2+ i Zn2+. Wzrost aktywności tego enzymu zaobserwowano w obecności Co2+. Pozostałe jony metali nie wpłynęły znacząco na aktywność 1,2-dioksygenazy katecholowej szczepu N6. Stwierdzona w badaniach częściowa aktywność obu badanych dioksygenaz po zastosowaniu soli metali sugeruje możliwość wykorzystania szczepów bakterii Stenotrophomonas maltophilia KB2 oraz Pseudomonas putida N6 do oczyszczania środowisk skażonych związkami aromatycznymi.
The strains of Stenotrophomonas maltophilia KB2 and Pseudomonas putida N6 are characterized by an enhanced capacity for degrading aromatic compounds: within five hours of incubation both the strains were found to provide a complete degradation of phenol (3 mmol/dm3). Upon induction with phenol, catechol 2,3-dioxygenase, an enzyme responsible for the meta-cleavage of aromatic compounds, was detected in the Stenotrophomonas maltophilia KB2 strain, whereas in the Pseudomonas putida N6 strain the presence was revealed of catechol 1,2-dioxygenase, an enzyme characteristic of the pathway for the orthofission of the aromatic ring. Tests on the sensitivity of the enzymes to metal ions have demonstrated that Zn2+ ions activated catechol 2,3-dioxygenase in the KB2 strain. The other metal ions were found to be inhibitors of this enzyme. Among the metal ions tested, the Cu2+ ion was the strongest inhibitor of the two isolated dioxynases. Slightly weaker was the inhibition of catechol 1,2-dioxygenase induced by Cd2+ and Zn2+ ions in the N6 strain. The activity of this enzyme increased in the presence of Co2+ ions. The other ions had no significant influence on the activity of the catechol 1,2-dioxygenase isolated from the N6 strain. The partial activity of both dioxygenases observed upon the application of metal salts suggests that both the strains, Stenotrophomonas maltophilia KB2 and Pseudomonas putida N6, may contribute much to the remediation of an environment polluted with aromatic compounds.

Plant growth-promoting rhizobacteria (PGPR) are a collection of microorganisms often used to support and promote plant development and combat plant infectious diseases with various biological control methods. The most significant restricting factors for agricultural productivity worldwide are abiotic constraints. In the present study, seven bacterial isolates from the rice rhizosphere were selected for detailed tests based on results obtained in experiments determining the ACC deaminase synthesis and drought tolerance at !0.30 MPa PEG level. Screening results of the stress tolerance analysis of the seven isolates for elevated temperature (50°C), alkalinity (10% NaCl), and drought (-1.2 MPa) showed that abiotic stress resistance was less prevalent in DRO2, DRO13, and DRO43 isolates than in DRO17, DRO28, DRO35, and DRO51 isolates. During the study, it was observed that DRO17, DRO28, and DRO51 tended to maintain similar cell density at -0.73 MPa PEG level, as observed at -0.30 MPa stress condition. No bacterial growth was observed at higher PEG level (-1.12 MPa) for any bacterial isolate. Four strains of Pseudomonas (DRO17, DRO28, DRO35, and DRO51) exhibited salinity and temperature tolerance. Antifungal screening using the bangle method showed that DRO35 was highly antagonistic towards Rhizoctonia solani 4633, followed by Fusarium moniliforme 4223, with an inhibition of 64.3% and 48%, respectively. The DRO28 isolate exhibited 72.5% growth inhibition for Fusarium moniliforme 4223, while the DRO51 isolate showed 38.2% growth inhibition for Bipolaris hawaiiensis 2445. DRO17 reduced the growth of Rhizoctonia solani 4633, and Curvularia lunata 350 by 36% and 31%, respectively. In conclusion, the screening of bacterial strains with promising stress tolerance and antifungal characteristics could support farmers to achieve the required positive outcomes in the agriculture field.

Background: We present a case of combined Pseudomonas aeruginosa, Acanthamoeba and Candida keratitis and clinical course of treatment in two adolescents girls who did not follow the rules of proper use of soft contact lenses. Case reports: Two teenage contact lens users sought medical attention, reporting visual impairment, redness and pain of the eye. Slit lamp examination showed unilateral corneal ulcer. Confocal microscopy revealed images compatible with amoebic cysts and yeasts cells, whereas the results of conjunctival swabs confirmed the presence of Pseudomonas aeruginosa. After topical and general treatment in the first patient, the corneal defect was healed, and visual acuity improved. The girl remains under the constant supervision of an ophthalmological clinic. The medications were gradually reduced, but with time she developed crystalline keratopathy. Regrettably, the second patient, despite the applied treatment had recurrent inflammation and progressive corneal thinning, which required amniotic membrane transplantation to prevent perforation. Conclusion: The early identification of the pathogen as well as adequate treatment is crucial, as contact lens-associated keratitis are potentially sight threatening. Patient and caregiver education, adequate contact lens hygiene and consistent follow-up are important to prevent serious complications.

The culture conditions for the production of extracellular agarase by Pseudomonas aeruginosa ZSL-2 were optimized using One-Factor-At-A-Time combined with orthogonal array design. One-Factor-At-A-Time method investigates the effect of time, temperature, NaCl, carbon sources, nitrogen sources and pH on agarase production. The optimized culture conditions obtained from the statistical analysis were temperature of 30 °C, pH 8.5, NH4NO3 2 g L-1 and agar 3 g L-1. The L9 orthogonal array design was used to select the fermentation parameters influencing the yield of agarase. The order of the factors affecting the fermentation process was found to be NH4NO3 > pH > agar > temperature, with temperature playing a significant role on the agarase production (p < 0.10). The higher yields than those in basal media culture were obtained in the final optimized medium with activity of 0.439 ± 0.013 U ml-1. Extracellular agarase hydrolysed agar into a range of oligosaccharides which were analysed by LC-ESI-MS spectrometry as anhydrogalactose, galactose, agarobiose, agarotetrose and agarohexaose.

The objectives of the study are to compare the living and non-living cells of Pseudomonas sp. G1 in their removal capacity of Cu(II) and Zn(II). For these purposes, the removal capacity, desorption efficiency of living and non-living cells and various factors affecting the adsorption, such as reaction time, initial pH of the solution and metal concentration were determined. It was found that the optimum pH value for Cu(II) removal by living and nonliving cells was 5.0, while it was 6.0 and 5.0, respectively, for Zn(II) removal. The binding capacity by living cells is significantly higher than that of dead cells at tested conditions. Most of the metal adsorption occurred during the first 10 min. Moreover, desorption efficiency of Cu(II) and Zn(II) by living cells was 76.3 and 68.4% under 0.1 M HCl and it was 93.1 and 86.5% by non-living cells, respectively.
Celem prowadzonych badań było porównanie zdolności biosorpcji Cu(II) i Zn(II) ze ścieków przez żywe i martwe komórki Pseudomonas sp. G1. Zakres pracy obejmował ocenę efektywności sorpcji i desorpcji metali przez żywe i martwe komórki oraz wpływ na nią takich parametrów, jak: czas reakcji, wartość pH i stężenie metali w roztworze. Stwierdzono, że optymalne pH dla procesu biosorpcji Cu(II) przez żywne i martwe komórki wynosi 5,0, natomiast w przypadku Zn(II) odpowiednio 6,0 i 5,0. W warunkach doświadczenia komórki żywe wykazywały znacznie większą zdolność do usuwania jonów metali ciężkich niż komórki martwe. Większość jonów metali została zaadsorbowana w ciągu pierwszych 10 minut trwania doświadczenia. Ponadto efektywność procesu desorpcji Cu(II) i Zn(II) za pomocą 0.1 M HCl wynosiła dla żywych komórek 76,3 i 68,4%, a dla martwych odpowiednio 93,1 i 86,5%.

The purpose of this study was to investigate the effect of bacterially produced gluconic acid on the dissolution of pyromorphite and Pb remobilization. Pyromorphite Pb5(PO4)3Cl is formed as a product of the phosphate-induced treatment of Pb-contaminated sites. This very stable mineral greatly decreases the bioavailability of Pb. In this study, bacterial and abiotic batch experiments on the dissolution of pyromorphite were carried out. In the microbial experiments, the mineral was dissolved in the presence of the phosphate–solubilizing soil bacterium, Pseudomonas putida. The bacterial growth medium was supplemented with glucose, which under natural condi- tions can be supplied to microbes via symbiosis with plants. P. putida acquired P from pyromorphite and enhanced its dissolution. Elevated Pb concentrations were observed in the suspensions with bacteria. The bacterial secretion of 16.5 mM gluconic acid played a significant role in Pb remobilization; the pH of the solution dropped down from an initial 7.4 to 3.5. In the abiotic experiments, pyromorphite was dissolved at several concentrations of gluconic acid and at an acidic to neutral pH range. Both acidification and formation of stable Pb-gluconate ligands enhanced the dissolution of pyromorphite and caused Pb remobilization.

Ze względu na swoje potencjalne zastosowanie w medycynie i różnych gałęziach przemysłu siderofory syntetyzowane przez bakterie cieszą się w ostatnich latach bardzo dużym zainteresowaniem badaczy. Głównym sideroforem wytwarzanym przez bakterie z rodzaju Pseudomonas w warunkach niedoboru żelaza jest piowerdyna. W niniejszej pracy wyizolowano i oczyszczono, metodą chromatografii chelatującej, piowerdyny trzech szczepów bakterii P. fluorescens - 5N, G i D oraz piowerdynę szczepu P. putida A. Zbadano wymienialność sideroforów między tymi szczepami. Krzyżową wymienialność piowerdyn zaobserwowano między szczepami 5N i D, D i G oraz 5N i A. Po porównaniu profili elektroforetycznych białek zewnętrznej błony komórkowej wykazano, że bakterie w warunkach niedoboru żelaza wytwarzają dodatkowe białka, których synteza może być indukowana niedoborem żelaza w podłożu.
Interest in siderophores produced by bacteria seems to be increasing among researchers all over the world, mostly due to their potential applications in medicine and various branches of industry. Pyoverdine is a significant siderophore synthesized by bacteria under iron-deficient conditions. This paper describes isolation and purification of pyoverdines from three strains of Pseudomonas fluorescens (5N, G and D) and pyoverdine from P. putida A strain. The copper-chelate chromatography method was employed. Cross-reacting among Pseudomonas strains was analysed. Cross-reactivity was observed between the following strains: 5N and D, D and G, 5N and A. Moreover, we isolated outer membrane proteins from bacterial culture. The comparison of electrophoretic profiles of these proteins showed that bacteria produced additional proteins whose synthesis is probably induced by iron deficiency.

Optimization of biosurfactant has improved the value-chain, process development and cost of production associated with downstream synthesis. This study was designed to determine the optimal conditions for production of biosurfactant using hydrolyzed agroresidues under controlled conditions. Rhizobacterial isolate was obtained from Paspalum sp. growing on an aged crude oil impacted soil in Bodo, Rivers State, Nigeria. The bacterial isolates were identified using 16S rRNA molecular approach on a set of universal primers. One-Variable at a Time approach was applied for verification of pH, Carbon and nitrate sources respectively. Stat-ease Design-Expert version 12.0 was employed in the optimization of the variables while the operational conditions were fitted into a 20-run design matrix using α- level 2.0. Molecular identification confirmed the bacterial isolae to be Pseudomonas sp. with Accession number MH40927 with a gene molecular weight of 6.0kbp. Response for biomass, biosurfactant and Critical Miscelle Concentration (CMC) was observed to fit into a 2nd order Quadratic functions at p<0.05 with optimal conditions were pH = 7.0, Corn chaff = 2.0 g/L and Urea = 1.0 g/L. Biosurfactant = +51.98 + 6.79A + 4.30B 5.45C + 0.2975AB + 0.4975AC + 1.63BC-6.46A² - 3.51B² - 6.94C². This study further identified a directly proportional relationship between biosurfactant production and operational variables which represents a cheaper and feasible production roadmap for biosynthesis.

Malignant Otitis Externa is a rare infection in the external auditory canal, involving the temporal bone, that can be life-threatening for the patient. Immunocompromised elderly patients with diabetes are especially prone to the condition. Mortality rate used to be nearly 50%, whereas nowadays, due to long-term antibiotic therapy including fluoroquinolones, third and fourthgeneration cephalosporins, gentamicin and topical therapy, it has been reduced to about 15%. Diagnosis is based on laboratory tests, imaging studies and microbiological testing. The main etiological factor inducing the development of malignant external otitis is Pseudomonas aeruginosa. The paper presents a case report of a 71-year-old man with uncontrolled diabetes and malignant otitis externa. We describe the way an accurate diagnosis was established, together with the process of treatment, which was completed with clinical improvement, both systemic and local. The patient remains under care of the ENT, endocrine and neurological department. This report summarises information about the malignant otitis externa.
„Złośliwe” zapalenie ucha zewnętrznego (malignant external otitis) jest rzadko występującym zakażeniem przewodu słuchowego zewnętrznego i kości podstawy czaszki mogącym zagrażać życiu chorego. Schorzenie to występuje głównie u osób starszych z zaburzeniami odporności oraz cukrzycą. Śmiertelność wynosiła niegdyś około 50%, aktualnie dzięki stosowaniu wła- ściwej, długotrwałej antybiotykoterapii m.in fluorochinolonów, cefalosporyn III i IV generacji, gentamycyny i dzięki leczeniu miejscowemu wynosi około 15%. Rozpoznanie opiera się na badaniach laboratoryjnych, obrazowych i mikrobiologicznych. Czynnikiem etiologicznym izolowanym najczęściej jest Pseudomonas aeruginosa. Przedstawiono przypadek procesu diagnostycznego u 71-letniego pacjenta z niewyrównaną cukrzycą, ze złośliwym zapaleniem ucha zewnętrznego oraz proces terapeutyczny u tego chorego, zakończony poprawą stanu ogólnego i miejscowego. Pacjent pozostaje pod kontrolą w poradniach ambulatoryjnych: laryngologicznej, endokrynologicznej, neurologicznej. Niniejsze opracowanie będzie podsumowaniem wiadomości o złośliwym zapaleniu ucha zewnętrznego.

In this research antimicrobial activity of nanoparticles ZnO on perilous bacteria such as Pseudomonas aeruginosa was evaluated. P. aeruginosa is important pathogen that caused burn wound infections as it is multi-drug resistant and has several virulence factors. Fifteen samples of P. aeruginosa were collected from patients who suffering from Burn infections in Al-Hilla teaching hospital burn unit with the age range between (7-80) years old for both genders. After collecting burn samples, the diagnosis and characterization were performed by culturing and biochemical tests. ZnO NPs were synthesized by chemical method, Zinc oxide nanoparticles are well-known to be one of the multifunctional inorganic compounds which are widely used in medical applications. This study aims to prepare ZnO nanoparticles with particle size ranging from 23-29  nm. In the present study, surface modification of ZnO nanoparticles was performed, and influence of modification of the structure and morphological properties was investigated. The resulting nanoparticles were characterized by X-ray diffraction (XRD), scanning electron microscopy (SEM) and atomic force (AFM). Zinc oxide nanoparticles with the average diameter of about 29 nm were modified with an oleic acid to exert more compatibility. From the results obtained it is suggested that modified ZnO-nanoparticles could be used effectively in safety environmental and medical applications. Antibacterial activity for nanoparticle ZnO against P. aeruginosa isolates was measured by: Agar Diffusion Technique and Minimum Inhibitory Concentration (MIC)/Minimum bactericidal Concentration (MBC) with microdilution. The best zone of inhibition was (35.5mm) at a concentration of 40 μg/ml of nano-ZnO in one strain of P. aeruginosa while the lowest inhibition zone was (16 mm) at a concentration of 20 μg/ml of nano ZnO in one strain also. In addition, all P. aeruginosa isolates were completely inhibited at the concentration of 3.7 μg/ml of nano-ZnO (MIC) but no significant antibacterial activity was observed at concentrations less than 1.8 μg/ml of nano-ZnO and the (MBC) was same as MIC (3.7 μg/ml) for all P.aeruginosa isolates.

The aim of the study was to analyze the occurrence of Pseudomonas fluorescens in urban soil in the second year after fertilization with unprocessed sewage sludge from the wastewater treatment plant in Sokółka and processed sludge from wastewater treatment plant in Bialystok. The study was conducted on experimental plots located in the green belts along the main roads in Bialystok (Piastowska and Hetmańska streets). For the studied soil, two different types of sewage sludge were used: after-press dewatered sludge from the treatment plant in Sokółka and dry sludge in the form of pellets from the treatment plant in Bialystok. The experiment plots were fertilized with three doses of sewage sludge: 0-control, 14.5 and 29 Mg D.M./ha. In the second year after application of sewage sludge, microbiological tests of the rhizosphere area showed seasonal fluctuations in the number of Pseudomonas fluorescens bacteria. The highest number of Pseudomonas fluorescens bacteria was observed in April and in October, while the lowest number of bacteria occurred in July, which could have been conditioned by atmospheric factors. The analysis of the correlation indicates that in the urban soil mixed with sewage sludge the number of Pseudomonas fluorescens was significantly positively correlated with C:N ratio, organic carbon and content of phosphorus.

Pseudomonas spp. and their metabolites present environmentally friendly alternative to chemical products to improve plant growth in many different applications. Extensive studies have shown that these microorganisms and their metabolites could have an important role in agriculture and horticulture in improving crop productivity. The aim of the research was to assess a potential biological activity of Pseudomonas fluorescens against Fusarium oxysporum isolated from the spears of asparagus. The antagonistic properties of metabolites were assayed with a dual culture plate method on PDA and Czapek media for supernatants obtained from 6, 12, 24 and 48-hour culture of P. fluorescens. The culturing process was conducted at 26ºC for 8 days and the fungal linear growth was measured every 1-2 days and compared to control. The fungistatic activity of P. fluorescens was estimate on the basis the growth rate index. The highest inhibition of the linear growth of mycelium, reaching 61%, has been found for 48-hour supernatants at OD = 2.0 and lowest for 12-hour supernatants on Czapek medium compared with the control trial. Significantly weaker linear growth of mycelium within the range of 4.0-33.0% was observed on PDA medium with a maximum inhibition for 48-hour supernatants at OD = 2.0 (33.0%). Promising method to asparagus protection against Fusarium sp. may be the use of P. fluorescens as the biocontrol agents.
Pseudomonas spp. oraz ich metabolity mogą stanowić alternatywę dla związków chemicznych w celu poprawy wzrostu roślin. Mogą one odegrać istotną rolę w rolnictwie i ogrodnictwie w poprawie wydajności upraw. Celem przeprowadzonych badań była ocena właściwości przeciwgrzybowych Pseudomonas fluorescens wobec F. oxysporum, wyizolowanego z podstawy pędów szparaga. Ocenę właściwości antagonistycznych metabolitów bakteryjnych (6-, 12-, 24- i 48-godzinnych) przeprowadzono metodą hodowlano-płytkową z zastosowaniem podłoży Czapka i PDA. Hodowlę prowadzono w temp. 26ºC przez 8 dni, dokonując pomiarów co 1-2 dni i porównywano w stosunku do kontroli. Na podstawie indeksu tempa wzrostu określono aktywność fungistatyczną P. fluorescens. Największą inhibicję wzrostu liniowego grzybni wynoszącą 61% obserwowano na podłożu Czapka po zastosowaniu supernatantów z 48-godzinnej hodowli bakterii o gęstości 2.0, a najmniejszą dla 12-godzinnej hodowli. Znacznie słabszą inhibicję wzrostu grzybni, w zakresie od 3,98 do 33,2%, obserwowano na podłożu PDA. Maksimum aktywności (33,0%) stwierdzono dla supernatantów pozyskanych z 48-godzinnej hodowli bakterii o gęstości 2.0. Efektywną metodą zmniejszenia porażenia szparagów przez Fusarium sp. może być zastosowanie bakterii P. fluorescens jako czynnika biologicznej ochrony.

Improperly treated heavy metal wastewater discharged into water sources could cause a serious issue for the environment. The aim of this study was to bioaccumulate iron (Fe) using native bacteria isolated from the laboratory drainage water containing a high concentration of iron. The experiment was conducted in 250 mL conical flasks containing 150 mL Fe solution in concentrations of 25, 100, and 250 mg/L, respectively. Approximately 10% of bacteria inoculum was cultivated in each Fe concentration for 24 and 48 hours. The results showed that Pseudomonas hibiscicola was identified as an effective iron-accumulating species of bacteria. The species could remove Fe up to 82% (25 mg/L), 77.8% (100 mg/L) and 32% (250 mg/L). This promising result indicates that the native bacteria isolated from the environment pose a great potential for the remediation of wastewater containing iron.

The ability of six strains belonging to the genus Pseudomonas isolated from the rhizosphere of wheat to produce pyoverdin was examined. The studied strains demonstrated a varied level of production of the siderophore, depending on the culture conditions. The highest level of pyoverdin was determined after 72 hours of growth at 20- 25°C in iron-free medium supplemented with succinate. The synthesis of pyoverdin by all the strains studied was strongly repressed by the addition of iron ions (III) to the growth medium. Calcium, cadmium and magnesium ions stimulated the synthesis of the siderophore examined, whereas zinc and lead ions partially decreased its level. Enrichment of the growth medium in cobalt ions completely inhibited the synthesis of siderophores as well as growth of the bacteria.
Zbadano zdolność sześciu szczepów z rodzaju Pseudomonas wyizolowanych z ryzosfery pszenicy do produkcji piowerdyny (pyoverdin). Badane szczepy wykazywały zróżnicowany poziom produkcji tego sideroforu w zależności od warunków hodowli. Wykazano, że syntetyzowały one największą ilość piowerdyny w 72. godzinie hodowli i w temperaturze 20-25°C, na bezżelazowej bursztynianowej pożywce. Synteza piowerdyny u wszystkich badanych szczepów podlegała silnej represji po dodaniu do podłoża hodowlanego jonów żelaza (III). Jony wapnia, kadmu i magnezu w niewielkim stopniu stymulowały syntezę badanego sideroforu, natomiast jony cynku i ołowiu obniżały jej poziom. Wzbogacenie podłoża hodowlanego w jony kobaltu całkowicie zahamowało syntezę sideroforów i wzrost bakterii.

Endophytes are an important constituent of sustainable agriculture because of their ability to produce a large number of agriculturally important metabolites. A salt-tolerant fluorescent green pigment-producing endophytic bacterium was isolated on 2.5% NaCl-supplemented nutrient agar from the leaf samples of Zanthoxylum alatum Roxb. The isolate Z1B4 was identified as Pseudomonas fluorescens based on morphological features, fatty acid methyl ester analysis, biochemical tests, and 16S rRNA gene sequencing. P. fluorescens Z1B4 showed positive results for tricalcium phosphate solubilization; 1-aminocyclopropane-1-carboxylate (ACC) deaminase activity; and production of auxins, siderophores, hydrogen cyanide, and ammonia. P. fluorescens Z1B4 also showed strong antagonistic activity against Curvularia lunata (MTCC 283), Fusarium verticillioides (MTCC 3322), and Alternaria alternata (MTCC 1362) and exhibited stress tolerance to a wide range of temperature and pH and concentrations of NaCl and calcium salts. Under natural conditions, following inoculation with the isolate Z1B4, a significant increase in the growth of pea and maize test plants in pots was observed compared to that of uninoculated control plants. The rifampicin-resistant mutant Z1B4Rif was recovered from the roots, shoots, and leaves of the test plants, indicating that the isolated endophytic bacterium can grow well within different plant tissues. The present study indicated that the endophytic bacterium P. fluorescens Z1B4 can be used as a bacterial inoculant in stressed environments for sustainable agriculture.

Soil contaminated with crude oil has negatively affected some of the communities in Ekiti State region in Nigeria. There is an increased interest in microbial lipases because of their enormous potential in industrial and biotechnological applications. This study investigated the characteristics of partially-purified lipase from Pseudomonas aeruginosa isolated from oil-contaminated soil in Ado-Ekiti, Ekiti State. The lipase was purified by gel-filtration, having a molecular weight of 39.11 kDa, with Km and Vmax values of 12.50 and 28.86, respectively. The enzyme had an optimal pH of 8.0 and exhibited its maximal activity at 50°C, whereas the relatively stable temperature and pH were 40°C and 5.0, respectively. The enzyme activity was enhanced by olive oil, which served as the carbon source. Sodium chloride enhanced lipase activity, while calcium chloride acted as mild inhibitor, and iron chloride acted a strong inhibitor. The lipase from Pseudomonas aeruginosa possessed properties of an industrial enzyme and will be useful for biodegradation and bioremediation studies.

Objectives To evaluate the possible occupational hazard of environmental strains of opportunistic Pseudomonas aeruginosa on hydrocarbon contaminated sites during remediation, 2 multidrugresistant isolates originating from environmental (soil and groundwater) samples were examined. Material and Methods Antibiotic resistance profiles of the examined 2 strains were determined by Etest® against 20 different agents. Virulence investigations included the hemolytic activity test, the detection of virulence-related gene sequences such as exoA, exoU, exoS, exoY, exoT and the determination of intraperitoneal LD 50 (the lethal dose, 50%) values in a mouse model. The hydrocarbon-degrading ability was evaluated in a gravimetric experiment, in vitro. The phylogenetic relationship of the isolates was investigated with a multilocus sequence typing scheme. Results Multidrug resistant environmental strains of P. aeruginosa are strongly related to isolates that have proven effects on the infection of patients who suffer from cystic fibrosis, have a notable hemolytic activity, carry important virulence markers (exoS or exoU, respectively) and retain their hydrocarbon degradation ability (87.4% and 62.8% hydrocarbon degradation rate, respectively). Conclusions Pseudomonas aeruginosa presumably raise considerable concerns for human health in the environment, already well known among nosocomial isolates, and the application of environmental strains of this species for environmental purposes is questionable.

Polyhydroxyalkanoates (PHAs) are natural polyesters that are synthesized by many bacteria as an intracellular carbon and energy compound. Medium-chain-length polyhydroxyalkanoates (mcl-PHAs) have gained much interest in research on microbial biopolymers because of their ease of chemical modification. Mcl-PHAs are naturally synthesized by Pseudomonas species by transformation of wide range of substrates. The physiological background of mcl-PHAs synthesis is known, and key genes engaged in this process are discovered already, but the knowledge about their molecular regulation is still limited. Especially, there is lack of information concerning the transcription termination of gene coding for PHA polymerase (phaC1). It is assumed that the main role could play Rho-independent termination, which is related to presence of palindromic sequences, which leads to formation of the hairpin structure and to dissociation of the ternary elongation complex (TEC). In this work, DNA sequences located after phaC1 gene belonging to nineteen Pseudomonas strains were investigated. Among all analyzed strains, five had palindromic sequences, typical for Rhoindependent terminators. Our results proved, that gene phaC1, coding for PHA polymerase, can be independently regulated only in some species.

The proteolytic activity of three Pseudomonas aeruginosa strains, ATCC 27853 - a reference strain, and two clinical isolates was tested. The activity was examined after culturing the bacteria in two different growth media: the minimal M9 medium and rich Luria-Bertani broth (LB). Based on zymograms and protease activity specific assays, it was concluded that the reference strain produced three proteolytic enzymes in the LB medium: protease IV, elastase B and elastase A, while alkaline protease was only produced in the M9 medium. The clinical isolates of P. aeruginosa produced elastase B and alkaline protease when grown in the LB medium and the minimal M9 medium, respectively. PCR analysis confirmed the presence of both the lasB gene encoding elastase B and aprA coding for alkaline protease in the genomes of the three P. aeruginosa strains analyzed. The expression of these genes coding for two important P. aeruginosa virulence factors was dependent on the growth conditions in all the strains studied. The contribution of the extracellular proteinases to the virulence of P. aeruginosa strains used in this study was investigated using an insect model, the greater wax moth Galleria mellonella.

The paper deals with an examination of possible application of biodegradation in the decontamination of soil samples from Leos Janacek's Airport in Ostrava. Soils samples were used for laboratory tests of biodegradation taken from oil interceptors. The laboratory biodegradation tests were carried out with a pure bacterial culture of Pseudomonas putida, a pure laboratory culture of Rhodococcus sp., their mixture and a mixture prepared combining their media free of bacteria. The results of the paper imply that for the given purpose, i.e. for biodegradation of airport pollutants, it is most suitable to apply a mixed bacterial culture of Pseudomonas putida and Rhodococcus sp. The results of the paper show that the biodegradation method is applicable for the pollution in question.
Artykuł jest poświęcony badaniom nad możliwością wykorzystania rozkładu biologicznego do oczyszczania próbek gleby pobranych z lotniska im Leosa Janacka w Ostravie. Próbki gleby zanieczyszczone substancjami ropopochodnymi między innymi w czasie tankowania samolotów, zostały przebadane w warunkach laboratoryjnych. Zbadano proces biodegradacji za pomocą czystych kultur bakterii Pseudomonas putida oraz Rhodococcus sp., ich mieszanin oraz medium wolnego od bakterii. Uzyskane wyniki pozwalają na stwierdzenie, że biodegradacja jest dobrą metodą dla oczyszczania gleby z substancji ropopochodnych.

For the study on Pseudomonas bacteria, samples of vegetables with soft rot symptoms were collected in a market place and storage-rooms. The samples were examined on selective media for Gram-negative bacteria of the genus Pseudomonas. Forty Pseudomonas strains were isolated from injured carrot roots, potato tubers, onions, leek leaves, beetroots, cabbages, radish. On the basis of biological tests the bacteria were ascribe to 10 Pseudomonas species: P. fluorescens, P. marginalis, P. putida, P. facilis, P. aeruginosa, P. delafieldii, P. cichorii , P. pseudoalcaligenes, P. cepacia (=Burkholderia cepacia), P. gladioli (=Burkholderia gladioli pv. aliicola). Five species isolated from potato tubes, carrot roots, onions, and beetroots – P. fluorescens, P. marginalis, P. cichorii, P. cepacia (= Burkholderia cepacia), and P. gladioli (=Burkholderia gladioli pv. aliicola) – were able to cause soft rot symptoms of various vegetables.
Z warzyw przechowywanych w różnych warunkach na pożywkach sztucznych wydzielono 40 patogenów rodzaju Pseudomonas. Po sprawdzeniu właściwości biologicznych bakterii stwierdzono, że s to gatunki P. fluorescens, P. marginalis, P. putida, P. facilis, P. aeruginosa, P. delafieldi, P. cepacia (=Burkholderia cepacia), P. cichorii, P. alcaligenes ir P. gladioli pv. aliicola (=Burkholderia gladioli pv. aliicola). W wyniku przeprowadzonych testów na określenie patogeniczności bakterii stwierdzono, że pięć spośród wyizolowanych okazało się patogenicznymi: P. fluorescens wyizolowana z bulw ziemniaka, korzeni marchwi i cebuli, P. marginalis wyizolowana z bulw ziemniaka, P. cepacia, z korzeni marchwi i cebuli, P. cichorii, wyizolowana z korzeni marchwi oraz P. gladioli wyizolowana z cebuli. Wykazano, że niektóre gatunki bakterii mogą być patogeniczne i powodować zgniliznę warzyw. Porażone warzywa mogą być pierwotnym źródłem zakażenia zdrowych warzyw. Nieliczne spośród tych bakterii są chorobotwórcze dla zwierząt i człowieka.

An estS9 gene, encoding an esterase of the psychrotolerant bacterium Pseudomonas sp. S9 was cloned and sequenced. The deduced sequence revealed a protein of 636 amino acid residues with a molecular mass of 69 kDa. Further amino acid sequence analysis revealed that the EstS9 enzyme contained a G-D-S-L motif centered at a catalytic serine, an N-terminal catalytic domain and a C-terminal autotransporter domain. Two recombinant E. coli strains for production of EstS9N (a two domain enzyme) and EstS9Δ (a one domain enzyme) proteins were constructed, respectively. Both recombinant proteins were successfully produced as inclusion bodies and then purified under denaturing conditions. However, because of the low enzymatic activity of the refolded EstS9Δ protein, only the EstS9N protein was further characterized. The purified and refolded EstS9N protein was active towards short-chain p-nitrophenyl esters (C2-C8), with optimal activity for the butyrate (C4) ester. With p-nitrophenyl butyrate as the substrate, the enzyme displayed optimal activity at 35°C and pH 9.0. Additionally, the EstS9N esterase retained ~90% of its activity from 25-40°C and ~40% of its activity at 10°C. Moreover, analysis of its kinetic parameters (Km, kcat, kcat/Km) toward p-nitrophenyl butyrate determined at 15°C and 25°C confirmed that the EstS9 enzyme is cold-adapted. To the best of our knowledge, EstS9 is the third characterized cold-active GDSL-esterase and the first one confirmed to contain an autotransporter domain characteristic for enzymes secreted by the type V secretion system.

Pre-sowing seed dressing with a chemical preparation or a post-culture liquid of bacteria considerably improved the emergences, healthiness and yielding of bean plants. Despite the pre-sowing seed dressing, plants and seeds obtained after the harvest were infected by Alternaria alternata, Botrytis cinerea, Fusarium culmorum, Fusarium oxysporum f. sp. phaseoli, Fusarium solani, Phoma exigua, Pythium irregulare, Rhizoctonia solani and Sclerotinia sclerotiorum. The best protective effect against those plant pathogens was observed for the post-culture liquid of Pseudomonas sp. Psf 47.
Celem pracy było określenie skuteczności ochronnego działania Zaprawy Oxafun T (substancja aktywna: karboksyna 37.5 % + tiuram 37.5 %) oraz płynów pohodowlanych Bacillus sp. Bf 155 i Pseudomonas sp. Psf 47 przeciwko patogenicznym dla Phaseolus coccineus grzybom przeżywającym w glebie. Przedsiewne zaprawianie nasion, preparatem chemicznym lub płynem pohodowlanym bakterii, znacznie poprawiło wschody, zdrowotność oraz plonowanie roślin fasoli. Mimo przedsiewnego zaprawiania nasion, rośliny oraz uzyskane po zbiorze nasiona fasoli były porażane przez Alternaria alternata, Botrytis cinerea, Fusarium culmorum, Fusarium oxysporum f. sp. phaseoli, Fusarium solani, Phoma exigua, Pythium irregulare, Rhizoctonia solani i Sclerotinia sclerotiorum. Najskuteczniejszym ochronnym działaniem przed tymi fitopatogenami wyróżnił się płyn pohodowlany Pseudomonas sp. Psf 47.

The optimum conditions for biosorption of Pb(II) and Ni(II) from aqueous solution were investigated, by using living and nonliving Pseudomonas fluorescens and Bacillus pumilus isolated from wastewater treatment plant. It was found that the optimum pH for Pb(II) removal by living and nonliving cells was 6.0, while 7.0 for Ni(II) removal. At the optimal conditions, metal ion biosorption was increased as the initial metal concentration increased. The binding capacity by living cells is significantly higher than that of nonliving cells at tested conditions. The maximum biosorption capacities for lead and nickel by using Ps. fluo-rescens and B. pumilus were 77.6, 91.4 and 65.1, 73.9 mg/g, respectively. The results of bio-sorption time and desorption experiments suggested that Pb(II) and Ni(II) uptake by the living bacterial biomass might be enhanced by intracellular accumulation.

The present study investigates phytochemical screening and antibacterial efficacy of various solvent extracts of marine algae such as Sargassum swartzii against some selected human and fish pathogenic bacteria. The brown seaweed Sargassum swartzii was collected from Kudankulam, Tirunelveli district, Tamil Nadu, India. Soxhlet extraction method was used to get the extracts of different solvents like aqueous, ethanol, methanol, acetone and were tested for their qualitative phytochemical screening using common standard tests. Quantitative phytochemical analysis such as total phenolic content and total flavonoid content of S. swartzii and antibacterial activity against human pathogens viz., Escherichia coli, Pseudomonas aeruginosa and Stephylococcus aureus and fish pathogens viz., Aeromonas hydrophila and Vibrio vulnificus bacteria using disc diffusion methods. Preliminary phytochemical screening of sixteen different chemical compounds were carried out. The maximum biochemical compounds were present in the ethanol extract and the minimum compounds were present in aqueous extract. The ethanol extract was significantly higher in phenolic content (15.35 ±2.61 mg of GAE/g) and the methanol extract was significantly higher in flavonoides content (26.92±7.14 mg of QE/g). In human pathogen the highest antibacterial activity was present in Pseudomonas aeruginosa (18.00±0.00mm) and the fish pathogen was significantly higher in Aeromonas hydrophila. The present study showed that the ethanol extract of marine brown seaweed Sargassum swartzii contains bio active constituents with highest antibacterial activity against human and fish pathogen.

The soil sample was collected from the paddy field of Sriperumbudur, Tamilnadu which is having a history of repeated pesticide applications. The isolation of efficient pesticide degrading bacteria was identified as Pseudomonas aeruginosa, Staphylococcus aureus and Bacillus subtilis. The growth of the three pesticide degrading isolates was assessed in Minimal salt broth containing 25 ppm of pesticides. Two popularly used pesticides Metribuzin and Profenofos were selected for this study. Among the three bacterial isolates, the bacteria Bacillus subtilis utilized the pesticides effectively and showed maximum growth. The growth of the three pesticides degrading isolates Pseudomonas aeruginosa, Staphylococcus aureus and Bacillus subtilis was assessed in Minimal salt broth containing 25 ppm of pesticides at different temperature levels (25 °C, 30 °C, 35 °C, 40 °C, 45 °C, 50 °C & 55 °C) and pH levels (pH 4, pH 5, pH 6, pH 7 & pH 8) and carbon sources (Lactose, Dextrose, Fructose, Mannose & Galactose) and nitrogen sources Peptone, Yeast extract, Beef extract, Malt extract and Casein respectively. The maximum growth rate of bacteria was recorded at 35 °C and pH 6. The maximum growth of bacteria was in the presence of Dextrose followed by Fructose, Galactose and Mannose. The least growth was recorded in Lactose broth culture. The maximum growth of bacteria was in the presence of Malt extract followed by Peptone, Yeast extract and Casein. The least growth was recorded in Beef extract broth culture. The bacterial isolates showed maximum growth in the Minimal salt broth containing Profenofos followed by Metribuzin.

Występujące powszechnie i narastające zjawisko oporności na antybiotyki wśród bakterii chorobotwórczych jest jednym z największych wyzwań dzisiejszej medycyny zakażeń. Szczególne zagrożenie stanowią zakażenia szpitalne wywoływane przez wielooporne szczepy określonych gatunków, np. Pseudomonas aeruginosa. Jest to patogen oportunistyczny, odznaczający się opornością naturalną na kilka klas stosowanych antybiotyków. Dzięki wysokiej plastyczności genomu, obejmującej różnorodne mutacje funkcjonalne (strukturalne i regulacyjne) oraz pozyskiwanie obcego DNA, jest w stanie szybko adaptować się do niesprzyjających warunków środowiska. Szczególnie niepokoi zdolność nabywania przez P. aeruginosa dodatkowych cech oporności, co w połączeniu z naturalnymi mechanizmami czyni ten patogen wybitnie trudnym do zwalczania. Bakteria ta jest w stanie wywoływać m. in. ostre zapalenie płuc, zakażenia łożyska krwi, skóry i tkanek miękkich (w tym ran operacyjnych i oparzeniowych). Jest również czynnikiem etiologicznym zakażeń przewlekłych, towarzyszących np. mukowiscydozie. Antybiotykami stosowanymi obecnie przeciwko zakażeniom P. aeruginosa są najczęściej cefalosporyny III i IV generacji, karbapenemy, fluorochinolony i aminoglikozydy. W związku z malejącą liczbą dostępnych, skutecznych opcji terapeutycznych pracuje się nad nowymi terapeutykami lub nowatorskim wykorzystywaniem dotąd już poznanych.
The rapid spread of antibiotic resistance (AMR) in pathogenic bacteria is one of the greatest challenges of modern infectiology. In particular, the most threatening are nosocomial infections caused by multi-drug-resistant strains of several major species, such as Pseudomonas aeruginosa. This opportunistic pathogen exibits a broad-spectrum of natural resistance. Due to its high genome plasticity, comprising functional mutations and acquisition of foreign DNA, P. aeruginosa can easily adapt and persist in harsh environmental niches. The critical issue is its outstanding ability to acquire diverse AMR mechanisms, including those encoded by mobile genetic determinants. In addition to the intrinsic resistance, P. aeruginosa can be highly resistant to all of the currently available antipseudomonadal antimicrobials. P. aeruginosa is the etiological agent of a variety of infections, including acute pneumonia, bloodstream infections or skin and soft tissue infections (e. g. postoperative or burn wounds). It is responsible also for chronic infections, like those in cystic fibrosis (CF) patients. The major antimicrobials used in P. aeruginosa infections are newer-generation cephalosporins, carbapenems, fluoroquinolones or aminoglycosides. Owing to limitations of the effective therapeutic options against P. aeruginosa, new antimicrobials and novel indications and thus applications for older drugs are being developed.
