HNPP neuropathy
PMP22 gene mutations
Opis:
Hereditary neuropathy with liability to pressure palsies (HNPP) is manifested by a spectrum of phenotypes, from the classical HNPP course associated with intermittent nerve palsies to a neuropathy resembling Charcot-Marie-Tooth type 1 (CMT1) disease. The majority of HNPP cases are associated with submicroscopical deletions in the 17p11.2-p12 region containing the PMP22 gene, while PMP22 point mutations are rare, representing about 15% of HNPP cases. In this study, we present a patient manifesting with atypical HNPP phenotype associated with a new Thr99fsX110 mutation in the PMP22 gene. We conclude that all patients who fulfill the electrophysiological criteria of HNPP, even if they lack the typical HNPP phenotype, should be tested for point mutations in the PMP22 gene.

Hershfield’s method
Opis:
This paper presents an attempt to estimate the value of Probable Maximum Precipitation (PMP) over the Kielce Upland for one-day and multi-day intervals. Statistical methods were employed in the calculations. Precipitation data were obtained from 23 gauging stations. A regional frequency factor km (from Hershfield’s formula) was calculated for each station based on a series of maximum annual precipitation totals from 1961 to 2006. The calculated PMP values range from 120.7 mm to 228.4 mm (within the 1-day precipitation group), from 140.7 mm to 266.3 mm (2-day group), from 158.9 mm to 294.1 mm (3-day group), and from 175.2 mm to 294.8 mm (4-day group). These values became reference values used to show the spatial distribution of PMP throughout the Kielce Upland region.

gene dosage effect
Charcot-Marie-Tooth disease
PMP22 gene
Opis:
Little is known about the molecular background of clinical variability of Charcot-Marie-Tooth type 1A (CMT1A) disease and hereditary neuropathy with liability to pressure palsies (HNPP). The CMT1A and HNPP disorders result from duplication and deletion of the PMP22 gene respectively. In a series of studies performed on affected animal transgenic models of CMT1A disease, expression of the PMP22 gene (gene dosage) was shown to correlete with severity of CMT course (gene dosage effect). In this study we hypothesized that single nucleotide polymorphisms (SNPs) located within the 5' regulatory sequence of PMP22 gene may be responsible for the CMT1A/HNPP clinical variability. We have sequenced the PMP22 5' upstream regulatory sequence in a group of 45 CMT1A/HNPP patients harboring the PMP22 duplication (37) /deletion (8). We have identified five SNPs in the regulatory sequence of the PMP22 gene. Three of them i.e. -819C>T, -4785G>T, -4800C>T were detected both in the patients and in the control group. Thus, their pathogenic role in the regulation of the expression of the PMP22 gene seems not to be significant. Two SNPs i.e. -4210T>C and -4759T>A were found only in the CMT patients. Their role in the regulation of the PMP22 gene expression can not be excluded. Additionally we have detected the Thr118Met variant in exon 4 of the PMP22 gene, which was previously reported by other authors, in one patient. We conclude that the 5' regulatory sequence of the PMP22 gene is conserved at the nucleotiode level, however rarely occurring SNPs variant in the PMP22 regulatory sequence may be associated with the gene dosage effect.

Opis:
An on-line phase measurement profilometry based on improved Stoilov’s algorithm is proposed to measure the 3D shape of moving object. While only one frame sinusoidal grating is projected on the moving object, the equal phase-shifting step deformed patterns modulated by profile of the measured object can be captured at every equivalent moving distance of the measured object instead of digital phase-shifting. Stoilov’s algorithm is an equal phase-shifting step algorithm at an arbitrary phase-shifting step, which is suitable for on-line phase measurement profilometry. However, the arbitrary phase-shifting step of Stoilov’s algorithm depends on the captured deformed patterns, in which the digitized errors of digital light projector or CCD camera, and the disturbance of surrounding light could be introduced, it will lead to some abnormities in wrapped phase, such as the denominator in Stoilov’s algorithm could be zero, which could cause the reconstructed 3D profile of the measured object appear burr, distortion or aberration, even could not be reconstructed. So an on-line phase measurement profilometry based on improved Stoilov’s algorithm is proposed. The arbitrary phase-shifting step is retrieved by both pixel matching and fringe cycle calibration rather than the captured deformed patterns. Experiments verify the feasibility and effectiveness of the proposed on-line phase measurement profilometry.

Opis:
Physical scale models have a large range of application in studies of hydraulic works. In port engineering, they can be used to optimize the general layout of terminals, evaluation of protection structures, simulation of vessel maneuvers and investigation of mooring plans for vessels, among several subjects. Once physical modeling allows a high accuracy in the waves and currents representation as well as their interaction with the bottom and the vessels, the studies of mooring systems in coastal and estuarine ports based on physical modeling tests provide greater reliability in comparison with those grounded on distinct types of models. To highlight the importance of this kind of application, this article presents the case study of the Ponta da Madeira Port (PMP), located in the State of Maranhao, Brazil, developed with the support of the 1:170 scale reduced physical model conceived and calibrated for this area. This study analyzed several alternatives to improve the availability of the northern berth of the Pier III of PMP, including new mooring strategies and the construction of a new improvement structure. The results, which concerned on preliminary tests of the mooring lines tensions, evidenced structural intervention could substantially reduce the risk of mooring lines break, indicating that further investigations concerning different layouts for the improvement structure are promising in order to provide an increase of this berth availability.

Opis:
The solid particle number method was introduced in the European Union (EU) light-duty legislation for diesel vehicles to ensure the installation of the best-available technology for particles (i.e., wall-flow diesel particulate filters) without the uncertainties of the volatile nucleation mode and without the need of large investment for purchasing the equipment. Later it was extended to gasoline vehicles with direct injection engines, heavy-duty engines (both compression ignition and positive ignitions) and non-road mobile machinery engines. Real Driving Emissions (RDE) testing on the road with Portable Emissions Measurement Systems (PEMS) for particle number (and NOx) during type approval and in-service conformity testing was recently (in 2017) introduced for light-duty vehicles, and is under discussion for heavy-duty vehicles in-service conformity testing. This paper will summarize the existing legislation regarding solid particle number and discuss the on-going activities at EU level. The main focus at the moment is on improving the calibration procedures, and extending the lower detection size below 23 nm with interlaboratory exercises. In parallel, discussions are on-going to introduce testing at low ambient temperature, regeneration emissions in the light-duty regulation, a particle limit for other technologies such as gasoline port-fuel injection vehicles, and the feasibility of particle measurements to L-category vehicles (mopeds, motorcycles, tricycles and minicars). A short overview of periodical technical inspection investigations and the situation regarding non-exhaust traffic related sources with special focus on brakes and tyres will be described.
