The goal of work was identification A1 variant of bovine beta casein which involves ischemic heart disease and diabetes mellitus in human. The digestion of A1beta casein can result in the production of bioactive beta casomorphin-7 (BCM-7); this is not the case with A2. This bioactive peptide has been linked to physiological traits that may elicit effects on components of the vascular and immune systems. The material involved 111 Slovak Spotted breed. Bovine genomic DNA was extracted from whole blood by using commercial kit, and used in order to estimate beta-casein genotypes by means of PCR-RFLP method. The PCR products were digested with DdeI restriction enzyme. In the population included in the study were detected all three genotypes, homozygote genotype A1A1 (14 animals), heterozygote genotype A1A2 (37 animals) and homozygote genotype A2A2 (60 animals). In the total population of cattle homozygotes A2A2-0.5405 were the most frequent, while homozygotes A1A1-0.1261 were the least frequent ones. This suggests a superiority of allele A2 (0.7072) which does not produce BCM-7, and thus is safe for human consumption. The expected homozygosity for gene CSN2 is in the population stated a slight increase in homozygosity (0.5858). This caused a slight decrease in the level of possible variability realization (41.80%), which corresponds to the effective number of alleles (1.7071).

Introduction. Giardia duodenalis (G. intestinalis) is a common protozoan causing gastrointestinal disorders in many species of mammals. The genus of Giardia has high molecular diversity. Dogs and cats, in addition to their typical infection with assemblages C, D and F, may be a reservoir of zoonotic assemblages (A and B). Objective. The aim of this study was a genetic characteristic of Giardia isolates of dogs and cats from the area of Wroclaw (Poland). Materials and method. A total of 128 and 33 faecal samples from dogs and cats, respectively, were analyzed by routine coprological methods. The animals were diagnosed on the presence of G. duodenalis antigens in faeces soluble with the use of SNAP Giardia (IDEXX Laboratories) immunosorbent assay. 27 DNA isolates of Giardia were subjected to molecular identification (PCR-RFLP). Results and conclusions. The prevalence of G. duodenalis was 21.1% (27/128) in dogs and 15.1% (5/33) in cats. In dogs, C assemblage was present in 18 (81%) positive stool samples, D assemblage in 2 (9%) samples, B assemblage present in one (4.5%), and mixed assemblages (C and D) occurred in one (4.5%) sample. F assemblage was found in 4 (80%) cats’ positive stool samples and A assemblage occurred in one case (20%). Confirmation of the presence of A and B zoonotic assemblages suggests that infected pets can be a threat to human health. This study describes for the first time the presence of mixed infections within host-specific C and D assemblages in dogs in Poland.

Background. The occurrence of clarithromycin resistance among Helicobacter pylori strains is a major cause of the treatment failure. Resistance to this drug is conferred by point mutations in 23S rRNA gene and the most prevalent mutations are A2143G and A2142G. The aim of the study was to evaluate the occurrence of A2143G and A2142G mutations in a group of H. pylori strains resistant to clarithromycin. Materials and Methods. The study included 21 clarithromycin-resistant H. pylori strains collected between 2006 and 2009 in southern Poland. Resistance to clarithromycin was quantitatively tested with the E-test to determine the minimal inhibitory concentration (MIC value). The point mutations of H. pylori isolates were detected by PCR followed by RFLP analysis. Results. The MIC values for clarithromycin for the analyzed strains ranged from 1.5 mg/L to 64 mg/L. Nine H. pylori strains exhibited A2143G mutation and A2142G mutation was found in 9 isolates as well. The results of RFLP analysis of 3 clarithromycin-resistant strains were negative for both mutations. The average MIC values for A2143G and A2142G mutants were 6 and 30 mg/L, respectively. Conclusions. Frequencies of A2143G and A2142G mutations were the same in all isolates tested. Strains with A2143G mutation exhibited lower MIC values than A2142G mutants. Application of PCR-RFLP method for detection of clarithromycin resistance allows for better and more efficient management of H. pylori infections.

Infections with human papillomavirus (HPV) are detected frequently. The occurrence of persistent infection with oncogenic HPV type associates with the development of cervical cancer. However, the part of HPV infections consist of the multiple viral types. The influence of HPV co-infections on the risk of the development of cervical cancer is not clear, but the occurrence of more than one viral type seems to increase the risk of advanced lesions of the uterine cervix. Additionally, possible interactions (both positive and negative) between particular HPV types in co-infections may influence the risk of the lesions progression. Although, the issue of HPV co-infections is not well known, the previous scientific reports make that the introduction of the molecular methods, allowing the identification not only single but also multiple HPV infections, in routine diagnostics seem to be important. Here, we present the PCR-RFLP and Real-Time PCR methods as the useful tools in the diagnostics of HPV infections.

Despite of the growing interest in pike-perch for aquaculture and its economic importance in fisheries, knowledge on the population structure and phylogeography of the species is still limited. We report the discovery of two major cytochrome b lineages and describe a simple method for their detection based on PCR amplification followed by restriction digestion with Alw26I. Screening of 708 individuals showed that haplotype A was fixed or dominating in Central and East European countries, whereas haplotype B was mainly found in several French populations and Tunisian pike-perch introduced from Europe. Sequencing the complete cytochrome b cds of 17 representative individuals revealed that haplotypes A and B differed by five substitutions but also showed further differentiation of both haplotypes due to an additional substitution in a single haplotype A and B individual from France, respectively. Five partial pike-perch cytochrome b cds available from NCBI GenBank could also clearly be assigned to one or the other of the two major lineages. Therefore, this new mtDNA marker might be considered as suitable not only for studies on population structure and phylogeography of pikeperch but also to trace its introduction history and to assess the genetic composition of aquaculture brood stocks.

The ubiquitin-conjugating enzyme E2E 2 (UBE2E2) gene plays an important role in insulin synthesis and secretion under conditions in which stress to the endoplasmic reticulum is increased in β-cells. In this case-control study, we have selected rs7612462 polymorphism within UBE2E2 gene to identify in a Saudi population the type 2 diabetes mellitus (T2DM) subjects. In total, 376 subjects with T2DM and 380 controls were enrolled in this study. We have collected 5 mL of peripheral blood from each participant for biochemical and molecular analyses. PCR-RFLP was used to generate genotypes at rs7612462 in all of the study subjects. Mikroorganizmy glebowe, pod względem cech genomowych i fenotypowych, stanowią wysoce zróżnicowaną grupę organizmów żywych. Z powodu tak dużej różnorodności ważne jest dobranie odpowiednich metod, dających największy stopień różnicowania mikroorganizmów. Narzędziami umożliwiającym analizę zmienności genetycznej mikroorganizmów są techniki genetyczne, a wśród nich jedną z najważniejszych jest łańcuchowa reakcja polimerazy, czyli PCR (Polymerase Chain Reaction), technika opracowana w latach 1980. Niniejsza praca stanowi przegląd podstawowych zagadnień dotyczących badania zmienności genetycznej mikroorganizmów glebowych w oparciu o markery molekularne z wykorzystaniem technik bazujących na reakcji PCR tj. PCR-RFLP, TRFLP, ARDRA, RAPD.
Soil microorganisms represent a highly diverse group of living organisms in terms of genomic and phenotypic characteristics. Due to such a large diversity, it is important to select appropriate identification methods which would secure its most complete determination. Genetic techniques are proper tools of choice for analyzing genetic variability of microorganism, the most important of which is the polymerase chain reaction (PCR), developed in the 1980s. This work presents an overview of the basic issues concerning studies on genetic variability of soil microorganisms with help of molecular markers and application of PCR techniques such as PCR-RFLP, TRFLP, ARDRA, RAPD.

Opis:
W latach 1998-2005 prowadzono obserwacje i badania nad występowaniem fitoplazm w drzewach i krzewach ozdobnych w Polsce. Przedmiotem badań były krzewy z rodzaju Clematis sp., Magnolia spp. i Rosa sp. oraz drzewa z rodzaju Acer sp., Carpinus sp., Fagus sp., Fraxinus sp. i Quercus spp. wykazujące objawy chorobowe sugerujące porażenie przez fitoplazmy. Objawy te obejmowały zahamowanie wzrostu, zmiany w zabarwieniu a nawet nekrozę i deformację liści, proliferację i zamieranie pędów, czasem deformację i zielenienie lub brak kwiatów oraz zamieranie roślin. Do wykrywania i identyfikacji fitoplazm stosowano test biologiczny oparty na reakcji rośliny testowej Catharanthus roseus, zakażanej techniką szczepienia, oraz analizę PCR-RFLP. Stosując technikę szczepienia, fitoplazmy przeniesiono z chorych róży, magnolii i klonu na siewki C. roseus. Przy pomocy techniki łańcuchowej reakcji polimerazy (PCR) ze starterami amplifikującymi fragment 16Sr DNA fitoplazm, obecność fitoplazmatycznego DNA wykryto w roślinach Rosa spp., Magnolia spp., Clematis sp., Fraxinus excelsior, Carpinus sp. i Acer negundo oraz w eksperymentalnie zakażonych siewkach C. roseus. Analiza RFLP produktu amplifikacji z użyciem enzymów restrykcyjnych AluI, HhaII MseI i RsaI wykazała, że rośliny grabu, jesionu, klonu, magnolii, powojnika i róży były naturalnie porażone przez fitoplazmę z grupy żółtaczki astra (16SrI-B) - 'Ca. Phytoplasma asteris’. W niektórych roślinach róży i magnolii stwierdzono także fitoplazmę z grupy proliferacji jabłoni (16SrX-A) - ‘Ca. Phytoplasma mali’. W badanych roślinach fitoplazmy występowały nierównomiernie i w stosunkowo niskiej koncentracji, przez co mogły być wykrywalne dopiero w drugiej rundzie PCR. W roślinach buka i dębu, mimo wyraźnych symptomów, nie wykryto obecności fitoplazmatycznego DNA.
The presence of phytoplasmas in six plant species (Acer sp., Carpinus sp., Fagus sp, Fraxinus sp. i Quercus spp) with severe symptoms of leaf discoloration and malformation, leaf and shoot necrosis, abnormal production of secondary shoots and decline, as well as in periwinkle plants experimentally inoculated by grafting was demonstrated by PCR with primer pairs amplifying phytoplasma 16Sr DNA fragment. The RFLP analysis of the PCR product done with restriction enzymes AluI, HhaII MseI and RsaI showed that the tested rose, magnolia, clematis, ash-leaf maple, ash and hornbeam plants were naturally infected with aster yellows phytoplasma (16SrI-B), now reclassified as ‘Candidatus Phytoplasma asteris’. Apple proliferation phytoplasma (16SrX-A), now reclassified as 'Candidatus Phytoplasma mali’, was detected in diseased roses and magnolias. The phytoplasmas were stated using nested PCR in affected woody plants, indicating their law titre and uneven distribution. We failed to detect the phytoplasma presence in diseased Quercus and Fagus trees.

Thrips from the genus Frankliniella (Thysanoptera, Thripidae) are phytophagous on crops and wild plants. Some of them cause slight economic damage, however, others including F. occidentalis and F. intonsa are responsible for considerable losses in crop production. Moreover, they constitute a double threat for host plants by not only feeding on them but also vectoring viruses, some of which are on the quarantined list of the European Plant Protection Organization. The rapid detection and differentiation between more and less harmful Frankliniella species is, therefore, important in order to combat the pests at the time of their appearance. In this study, we have undertaken to develop a method of detecting F. occidentalis, F. intonsa, F. pallida, and F. tenuicornis. The protocol is based on PCR amplification of ITS1 rDNA fragments of these insects using universal primers pair giving products of slightly distinct length for studied insects. Restriction enzymes digestion which is easy to interpret, allows for visible differentiation of all these Frankliniella species. The method was shown to be species-specific and sensitive. Even single specimens in either the larvae or adult stage could be distinguished.

The breast cancer suppressor proteins BRCA1 and BRCA2 interact with RAD51, a protein essential for maintaining genomic stability by playing a central role in homology-dependent recombinational repair of the DNA double-strand breaks. Therefore, genetic variability in the RAD51 gene may contribute to the appearance and/or progression of breast cancer. A single nucleotide polymorphism in the 5'- untranslated region of RAD51 (a G to C substitution at position 135, the G/C polymorphism) is reported to modulate breast cancer risk. We investigated the distribution of genotypes and frequency of alleles of the G/C polymorphism in breast cancer. Tumor tissues were obtained from postmenopausal women with node-negative and node-positive breast carcinoma with uniform tumor size. Blood samples from age matched healthy women served as control. The G/C polymorphism was determined by PCR-based MvaI restriction fragment length polymorphism. The distribution of the genotypes of the G/C polymorphism did not differ significantly (P >0.05) from those predicted by the Hardy-Weinberg distribution. There were no differences in the genotype distribution and allele frequencies between node-positive and node-negative patients. There were no significant differences between distributions of the genotypes in subgroups assigned to histological grades according to Scarf-Bloom-Richardson criteria and the distribution predicted by Hardy-Weinberg equilibrium (P >0.05). Our study implies that the G/C polymorphism of the RAD51 gene may not be directly involved in the development and/or progression of breast cancer and so it may not be useful as an independent marker in this disease.

Genetic variability of three sympatric crucian carp ( Carassius carassius ) populations from NW Poland was studied within a research project aimed at assessing the utility of those populations for stocking in inland waters. DNA samples were collected from 65 individuals. Restriction analysis was performed using 4 enzymes (HaeIII, HinfI, FspBI, TasI) of known restriction sites. The restriction pro files obtained were classified as belonging to a single haplotype (H-1). Selected DNA products were sequenced; the subsequent comparison made it possible to detect the presence of substi- tutions in the genome fragment analysed.
