Opis:
Different designs of the plant tissue culture vessel, such as size, material, and shape, may alter its microenvironment atmosphere. The present study was conducted on protocorm-like bodies (PLBs) of Dendrobium Sabin Blue orchid to determine the development of PLBs on plastic and glass culture vessels of different sizes. PLBs were cultured in half-strength Murashige and Skoog (MS) medium with the same initial weight of 0.5 g in 10 replicates. The growth index of the PLBs was calculated after 11 weeks to study their growth in every vessel; additionally, biochemical analysis was performed to determine carbohydrate content, proline concentration, and photosynthesis pigments in the PLBs. Scanning electron microscopy (SEM) was performed to study stomata development on PLBs in each vessel, and histological analyses were conducted to study the cell structure. Overall, the PLBs cultured in a large 470 ml plastic vessel showed successful growth with a high growth index, high carbohydrate content, low-stress condition, and high chlorophyll content. SEM confirmed that the presence of trichome and rhizoid in PLBs cultured in the 470 ml plastic vessel. Histological analysis showed the formation of the shoot on the PLBs and the presence of starch granules. Thus, the use of plastic as a culture vessel provides a good impact for culturing PLBs and has low cost.

Opis:
The aim of this study was to determine the current status of the population of marsh helleborine at a locality in the Wielkopolska National Park. Variability in generative shoots was analysed. Recorded results were compared with a study published in the 1990's, in which this locality was described. Since that time the area covered by that population has decreased; nevertheless, it exhibits characteristics of being in a good condition. In that population three forms of that species were reported: f. palustris, f. longibracteata and f. ochroleuca. The greatest threat for the existence of that heliophilous orchid was connected with the overgrowing of its habitat, for this reason measures should be taken to prevent succession and encroachment of trees and shrubs to that area.

In an age of changes in species’ geographical ranges, compounded by climatic and anthropogenic impacts, it become important to know which processes and factors influence plant populations and their persistence in the long term. Here we investigated dynamic and fitness components in twelve populations of Malaxis monophyllos (L.) Sw., situated in different geographical (regions) and ecological (type of habitat) units. Although M. monophyllos is a rare species, characterized by highly fragmented, boreal-montane distribution range, in last few decades it successfully colonized secondary habitats in Polish uplands. Our results indicate that M. monophyllos is represented mainly by small populations, which annual spatial and temporal changes might be very high, what affects the ephemeral character of these populations, regardless of the region and type of habitat. This dynamic structure, in turn, is caused by intensive exchange of individuals in populations, as well as by their short above-ground life span. Despite the large range of variation in size and reproductive traits, we can distinguish some regional patterns, which indicate boreal region as the most optimal for M. monophyllos growth and persistence in the long term, and with montane and upland/anthropogenic populations, due to lower reproductive parameters, as the most threatened. Although it should be considered that anthropogenic populations, despite their lower reproductive parameters and instability in the long term, present an intermediate, geographical and ecological character, therefore they may be valuable in shaping, both M. monophyllos’ future range, as well as its potential for response on ongoing and future changes. In general, reproduction is the main factor differentiating of M. monophyllos populations in regions, and we can suspect that it may become the cause of the future differentiation and isolation of these populations, occurring with progressive range fragmentation.

The orchid flora of Puebla State represents 16% of the total species present in Mexico demonstrating rich biodiversity. However, several municipalities in the north-east of Puebla area are located within a conservation priority area for its tropical montane cloud forest and tropical rain forest due to decision of the Comisión Nacional para el Conocimiento y el Uso de la Biodiversidad of Mexico. We have recorded 93 species from Cuetzalan del Progreso with 25 new records of the flora of the state. These new dditions thereby currently increase the orchid flora of Puebla to 197 species. Of these, six species are classified as threatened, three as subject to special protection by the NOM-059-SEMARNAT-2010 and one classified as least concern by the International Union for the Conservation of Nature. Moreover, we recorded 37 species illegally traded in “tianguis” (informal markets) of Cuetzalan del Progreso. The orchid diversity in the fragmented forests can be harnessed in establishing a model for the sustainable management and to promote conservation action f species at some level of threat and in the restoration of areas with fragmented forests. Unfortunately, the orchid diversity has been seriously endangered by several anthropogenic factors. We have also recorded significant anthropogenic threats in this municipality for the long-term existence of the local orchid members and species with potential for commercial production. A viable approach will be to allow the introduction of in vitro germination program and organic production of orchids by local and fringe communities; this will reduce inhabitants’ dependence on the scarce forest resources, lessen illegal harvesting and trafficking of wild orchids, and at the same time provide economic development for the local communities.

The study focused on the influence of an increased copper content in the Murashige and Skoog (1962) solid medium on the in vitro plant growth and development of Dendrobium kingianum Bidwill. Sterile explants of pseudobulbs were used for micropropagation of orchid plants on the MS regeneration medium supplemented with 0.5 mg dm-3 NAA and 1.0 mg dm-3 kinetin. Copper (as CuSO4 × 5H2O) was added to all the combinations in concentrations of 0.025 (control), 0.625, 1.25, 2.5 and 5.0 mg dm-3. The results showed that the treatments with 1.25 and 2.5 mg dm-3 stimulated the orchid growth and development in in vitro culture. After eight months of growing in in vitro culture, the highest number of shoots, the longest roots and the heaviest fresh weight of plantlets were obtained in these treatments. In medium with the highest copper concentration (5.0 mg dm-3), a negative influence of the metal on the length of roots and fresh weight of orchids was noted. Spectrophotometric analysis (ASA) showed that the copper and iron accumulation increased in both shoots and roots with the increase in the external Cu level, whereas the zinc and calcium accumulation in these organs decreased. The copper and zinc accumulation in the roots was about 1.5-2.5 times higher than in the shoots, but the iron accumulation was about 3-3.5 times higher. The calcium accumulation in roots was only 5-12% higher than in shoots.

The subject of the research was the population of orchid growing in the area of Pierzchno in Wielkopolska. This position was found in 2001 where two kinds of orchids were recorded – Orchis morio and Dactylorhiza majalis. Male orchid occurs in the particular position in this geographical region. Quite numerous population of Dactylorhiza majalis (broad-leaved marsh orchid) and counting only a few specimens of the population of Orchis morio were followed in 2013–2015. The research concerned both individual and group features. During this period, we observed a clear progression of broad-leaved marsh orchid population. It resulted in an increase in the number of ramets and the area occupied by it. In the first year of research two experimental plots were extracted and in the next growing season, a third surface was found, occupied by the species. Locating juveniles in the second season also confirms the population dynamics. In the last year of observation, orchids appeared only on the second surface, which substantially increased its dimensions. Unfortunately, in the case of populations of the male orchid clear regression was noted because fewer individuals grew from year to year. On the meadow nearby Pierzchno favourable living conditions were found for Dactylorhiza majalis and unfavourable ones for Orchis morio, which could lead to the disappearance of this population.

Oranżerie, palmiarnie i storczykarnie, osobliwe obiekty kultury materialnej, stanowiące zazwyczaj urozmaicenie pałacowej oprawy, z powodu kruchości ich materii, wymagają nadzwyczajnej i stałej pieczy. Pozbawione gospodarza, ulegają szybko nieuchronnej destrukcji. Stan storczykarni zespołu pałacowo-parkowego łańcuckiego zamku w okresie powojennym przedstawiał się katastrofalnie. Wydawało się, że ten niewielki obiekt – subtelnie kreujący wyjątkowe miejsce w pałacowym otoczeniu, dodający uroku historycznemu założeniu, stanowiący kiedyś szczególną miejscową atrakcję – jest już bezpowrotnie utracony. Współczesna rzeczywistość, po zakończonej w 2008 r. rewitalizacji obiektów łańcuckiego zabytkowego zespołu pałacowo-parkowego, przedstawia (obok budynków menażu i kasyna) obraz odrodzonej storczykarni, wzbogaconej przestrzennie i użytkowo. Storczykarnia wpisuje się na nowo w specyfikę niewielkiego Łańcuta, przywracając na tle dawnej rezydencji pełnię jego tożsamości.
Orangeries, palm houses and orchid houses, the peculiar cultural complexes usually consituting the variety of the palace setting, due to the assigned „brittle matter” to them, need the extraordinary and stable care. Deprived of the farmer they quickly undergo destruction. The condition of the orchid house of the palace-park compex of the Łańcut castle, was disastrous within the post-war period. It seemed that the tiny place, subtly creating the exceptional place, in a palace surrounding, adding the charm to the historic landscaping, being a local attraction in the past, is irrevocably lost. The modern reality after the revitalisation of the Łańcut historic palace-park complex after 2008 presents itself (apart from the mess and a casino house) and the orchid house as revived, and spatially and functionally enriched. The Orchid House has been set anew in the specificity of small Łańcut restoring its full identity on the background of the old residence.

The genus Eulophia embraces over 230 species distributed through the tropical and subtropical Africa, Asia, Australia and the Americas. In Neotropics it is represented by a sole species – E. alta. The aim of the presented study was to evaluate the difference between ecological niches occupied by American and African populations of this species based on the ecological niche modeling. The similarity between the glacial and present niches occupied by E. alta was calculated and the factors limiting the species occurrence were identified. Areas of seasonal tropical forest, tropical savanna and woodland served as refugia for the studied species during last glacial maximum and they were more widespread in Neotropics than in Africa. No significant niche shift after last glacial maximum was observed. The distribution of E. alta in its whole range is restricted mainly by temperature seasonality. The differences in the niches occupied by African and Neotropical populations of E. alta suggest preglacial disjunction of the species range and independent adaptation of both groups. Despite the significant range disjunction of E. alta the species is characterized by relatively high degree of niche conservatism.

Opis:
The physiological response of Phalaenopsis × hybridum ‘Innocence’ to biotic stress caused by Pseudococcus longispinus feeding was investigated. The condition of the cytoplasmic membranes expressed by a value of electrolyte outflow (EL) and TBARS and the activity of antioxidative system enzymes: catalase and peroxidase, and the amount of non-enzymatic antioxidant – proline, were determined. The changes in all the analyzed physiological parameters depended on the duration of the pest feeding. The outflow of electrolytes, TBARS content and catalase activity was the highest in the first period of the experiment (after 24-hour of mealybug feeding). Significant increase of peroxidase activity and proline content was noted after 7 days of insects feeding. The values of all analyzed parameters (except EL) demonstrated a decreasing tendency after 14 days of P. longispinus feeding. The observed reaction of P. hybridum ‘Innocence’ testifies to mechanisms triggered with the aim of neutralizing the effects of biotic stress and enabling the normal functioning of the cells in the orchid plants colonized by longtailed mealybug.
Badano fizjologiczną reakcję P. × hybridum ‘Innocence’ na stres biotyczny wywołany żerowaniem P. longispinus. Określono stan błon cytoplazmatycznych wyrażony wartością wypływu elektrolitów (EL) i zawartością substancji reagujących z kwasem tiobarbiturowym (TBARS), a także aktywność enzymów systemu antyoksydacyjnego: katalazy i peroksydazy, oraz ilość nieenzymatycznego antyoksydanta – proliny. Zmiany w wartowniach wszystkich analizowanych parametrów zależały od długości żerowania szkodnika. Wartość EL, zawartość TBARS oraz aktywność katalazy były najwyższe w pierwszym terminie eksperymentu (24-godzinne żerowanie czerwca). W przypadku aktywności peroksydazy i zawartości proliny istotny wzrost notowano dopiero po 7 dniach Żerowania. Po 14 dniach od zasiedlenia storczyków wartości wszystkich analizowanych parametrów (wyjątek EL) wykazywały wyraźną tendencję spadkową. Obserwowana reakcja P. hybridum ‘Innocence’ świadczy o uruchomieniu przez rośliną mechanizmów, których zadaniem jest neutralizacja skutków stresu biotycznego i umożliwienie komórkom powrotu do normalnego funkcjonowania.

A series of studies on pollination biology of the Epipactis atrorubens (Hoffm.) Besser, E. purpurata Sm., and E. palustris (L.) Crantz populations was conducted in Poland, Lithuania and Czech Republic between 2003 and 2010. The research focused on pollinators and visitors to aforementioned orchid species as well as on the chemical analysis of orchids’ nectar which was done using the GC/MS method. It was found that: 1) the type of pollinators depends on the orchid population size and the surrounding environment, where the temperature and amount of precipitation during the vegetative season are the most vital factors; 2) pollinators and visitors to the examined orchids might differ in successive growing seasons; 3) the studied Epipactis species differ in the chemical composition of their nectar and its scent, which can influence their pollination biology; 4) the tendency to autogamy observed in E. purpurata might be due to lack of pollinators in its habitats.
