Potato virus Y (PVY) is an important pathogen of to- bacco. Growing resistant cultivars is the best way to prevent si- gnificant losses of the crop caused by this virus. However, the protection given by the commonly used resistance factor, called va gene, can be overcome by the highly virulent PVY strains. Therefore, tobacco breeding for resistance will benefit from intro- ducing additional PVY resistance/tolerance factors. BPA is a to- bacco breeding line with introgressed PVY tolerance from a wild species N. africana. This trait is effective against a wide range of PVY isolates, including the ones that overcome va resistance. Here, we describe the inheritance of PVY tolerance of BPA. We obtained F1 and F2 plants from reciprocal crosses between BPA and a susceptible tobacco cultivar BP-210. Then we performed me- chanical inoculation tests using sap from PVY infected leaves on both generations of plants. Four weeks later we recorded disease symptoms and subjected all experimental plants to DAS-ELISA tests. All F1 plants developed vein necrosis which confirmed their susceptibility to the virus. The proportion of susceptible and tole- rant plants in the F2 fitted 3:1 ratio which was expected under the assumption that the tolerance is determined by a single, recessive gene. Moreover, the proportion of the susceptible and tolerant in- dividuals did not differ between two F2 populations derived from crosses where BPA was used as a maternal plant or a pollen do- nor, hence cytoplasmic factors do not influence the tolerance of that breeding line.

Near-amphihaploid hybrid plants of Nicotiana tabacwn x N. alata disomic for some N. alata chromosomes were nearly completely self- and cross-sterile but under open pollination conditions occasionally set poorly filled seed capsules containing very small numbers of seeds. Six plants from those seeds which survived to maturity resembled N. alata. Upon cytological examinations five plants were found to have the diploid chromosome complement of N. alata (2n=18), which suggested their androgenetic origin following outcrossing with paternal N. alata. The sixth plant had 9 bivalents and 11 univalents, which was indicative of its hybrid origin. Unlike the self-incompatible N. alata parent, N. alata-like hybrid derivatives were self-compatible. Surprisingly, the selfed progeny of the 18-chromosome plants showed sensitivity to tentoxin, a chloroplast trait characteristic of paternal N. alata but hardly compatible with its cytoplasmic lineage since the tentoxin-insensitive N. tabacum was the cytoplasmic parent of the hybrid. The progeny of the 29-chromosome plant was tentoxin-insensitive, which agreed with the parentage of the hybrid. Populations derived from the 18-chromosome plants were highly uniform but differed sharply from one another. The offspring of the 29-plant was highly heterogeneous mainly because of univalent segregation in that progeny.

The genus Nicotiana is one of the largest in the Solanaceae family and includes more than 80 species. The most well-known and widespread species of the genus Nicotiana is tobacco (Nicotiana tabacum), within which there are numerous cultivars. Tobacco is one of the most important industrial plants in Poland and worldwide. The great diversity within the genus makes it an excellent source of variation in a narrowing gene pool and can be used in breeding programmes. Studies of Nicotiana species also concern mechanisms of polyploidisation and evolution. There are also model species within the genus. However, in order to make full use of the collected germplasm resources, a detailed knowledge of the collection materials is necessary. While there are various reports in the international literature describing specific issues, the aim of this paper is to indicate the diversity of species in the genus Nicotiana as a whole on the basis of our own research and available studies. This review covers the characterisation of the genus Nicotiana in terms of origin and geographical distribution, as well as cytogenetic and molecular differences between species. An important aspect is the presentation of the morphological diversity of Nicotiana accessions and the variation in the most important tobacco alkaloids. A very important issue is the resistance of Nicotiana species to bacterial, fungal and viral diseases, which allows their use in resistance breeding.

The objective of this study was to investigate the effect of Selenium (Se) supply (0, 3, 6, 12, 24 mg kg−1) on the growth, photosynthetic characteristics, Se accumulation and distribution of flue-cured tobacco (Nicotiana tabacum L.). Results showed that low-dose Se treatments (≤6 mg kg−1) stimulated plant growth but high-dose Se treatments (≥12 mg kg−1) hindered plant growth. Optimal Se dose (6 mg kg−1) stimulated plant growth by reducing MDA content and improving photosynthetic capability. However, excess Se (24 mg kg−1) increased MDA content by 28%, decreased net photosynthetic rate and carboxylation efficiency by 34% and 39%, respectively. The Se concentration in the roots, stems, and leaves of the tobacco plants significantly increased with increasing Se application. A linear correlation (R = 0.95, P < 0.01) was observed between Se level and tobacco plant tissue Se concentration. This correlation indicated that the tobacco plant tissues were not saturated within the concentration range tested. The pattern of total Se concentration in the tobacco plant tissues followed the order root > leaf > stem. The Se concentration in the roots was 3.17 and 7.57 times higher than that in the leaves and stems, respectively, after treatment with 24 mg kg−1 Se. In conclusion, the present study suggested that optimal Se dose (6 mg kg−1) improved the plant growth mainly by enhancing photosynthesis, stomatal conductance, carboxylation efficiency and Rubisco content in the flue-cured tobacco leaves. However, the inhibition of excess Se on tobacco growth might be due to high accumulation of Se in roots and the damage of photosynthesis in leaves.

Here we report on a simple and reproducible system of Agrobacterium-mediated transient gene expression assay that utilizes infiltration of young Nicotiana benthamiana leaves. Although some of the phenomena described in this paper have been already reported by other researchers, here we have further developed them. The highest level of transient gfp gene expression was detected in the youngest leaves of N. benthamiana infiltrated with A. tumefaciens strains AGL0 and EHA105 precultured in the presence of 450-600 µM acetosyringone. Although the maximum level of transient gfp gene expression was restricted presumably by RNA silencing, it was completely suppressed in the presence of the viral protein HC-Pro. The transient expression system described here can be used to identify new viral suppressors of RNA silencing, for detailed analysis of unidentified genes and for industrial production of proteins in plants as well.

Green Tobacco Sickness (GTS) has been one of the unexplored areas of occupational health safety. Green Tobacco Sickness is caused by the absorption of nicotine through the skin from wet tobacco plants who have direct contact with tobacco plants during cultivation and harvesting. The present review was carried out to discuss the etiology, symptoms, diagnosis, treatment and prevention of GTS. It is caused by the absorption of nicotine through the skin while the workers are engaged in handling the uncured tobacco leaves. The symptoms include nausea, vomiting, pallor, dizziness, headaches, increased perspiration, chills, abdominal pain, diarrhea, increased salivation, prostration, weakness, breathlessness, and occasional lowering of blood pressure. The use of personal protective equipment like water‑resistant clothing, chemical‑resistant gloves, plastic aprons, and rain‑suits with boots should be used by the tobacco farmers to prevent its occurrence. An international‑level awareness campaign has to be taken up and more stringent workers safety regulations have to be formulated.

The common trait of all auxins is a stimulation of cell elongation and also cell division in the presence of cytokinin; both are essential for callus induction and its multiplication. The response of plant tissues to various compounds with auxin activity may be quite different. In this study, the effectiveness of a synthetic auxin, 2,4-dichlorofenoxyacetic acid (2,4-D), instead of the generally applied natural auxin, indole-3-acetic acid (IAA), was tested for the proliferation of Nicotiana tabacum callus. The following concentrations of 2,4-D were tested: 0.1, 0.5, 1.0, 1.5, and 2.0 mg dm−3. Callus was derived from stem pith and its proliferation allowed on MS medium through five subcultures at 25°C and in darkness. After each passage, the fresh weight and morphological features of the callus were determined. The 0.5 mg dm−3 2,4-D treatment was the most favorable for producing the greatest increase in fresh weight in each of five subsequent subcultures as well as maintaining normal morphological features for proliferation. However, the 1.0 mg dm−3 2,4-D treatment in comparison with the lowest, 0.1 mg dm−3, was more beneficial when considering regular increases of fresh weight and a better cell cohesion for callus growth.
Do indukcji i namnażania kalusa niezbędne jest zastosowanie odpowiedniej auksyny, podanej samodzielnie lub z cytokininą. Spośród naturalnych auksyn IAA jest często wykorzystywana do stymulowania proliferacji kalusa Nicotiana tabacum. Zastosowanie IAA wymaga wyższych stężeń ze względu na nietrwałość, wynikającą z wrażliwości na światło i wysoką temperaturę. Takich ograniczeń nie posiada syntetyczna auksyna 2,4-D. Celem tej pracy była ocena przydatności 2,4-D do proliferacji kalusa Nicotiana tabacum z następującymi wariantami stężeń 0.1, 0.5, 1.0, 1.5 i 2 mg dm−3. Proliferację kalusa z rdzenia łodygi przeprowadzono na pożywce MS przez 5 pasaży bez dostępu światła i w temperaturze 25°C. Po każdym pasażu określono świeżą masę i cechy morfologiczne kalusa takie jak barwa, struktura powierzchni i stopień spójności komórek. Spośród badanych stężeń 2,4-D, najefektywniej stymulującym było 0.5 mg dm−3 ze względu na najwyższy przyrost świeżej masy w każdym z pięciu kolejnych pasaży oraz sprzyjające proliferacji cechy morfologiczne kalusa. Wariant stężenia 1.0 mg dm−3 2,4-D w porównaniu z 0.1 mg dm−3 okazał się korzystniejszy, ponieważ charakteryzował się regularnymi przyrostami i lepszą dla namnażania spójnością komórek kalusa.

The objective of this study was to investigate the effects of inoculum concentration, plant post-inoculation incubation temperature and exogenous hydrogen peroxide (H2O2) on the induction of the hypersensitive response (HR) in Nicotiana tabacum against Xanthomonas perforans. Inoculation of leaves with X. perforans at a concentration of 108 CFU · ml–1 and incubation of plants at 30°C resulted in the strongest HR elicitation. Furthermore, an exogenous supply of H2O2 accelerated X. perforans-induced HR, whereas in planta H2O2 removal by application of catalase led to a delay in HR development. Our data suggest that H2O2 has an important role in HR of N. tabacum against X. perforans.

Przeprowadzono badania zdolności pobierania i gromadzenia kadmu przez 59 dzikich gatunków Nicotiana i 6 odmian botanicznych N. tabcum i N. rustica. Badania prowadzono na glebie o naturalnej zawartości kadmu 0,15 ppm i glebie z dodawanym kadmem w ilości 8 ppm. W wyniku analizy chemicznej stwierdzono zróżnicowaną zawartość kadmu w liściach i korzeniach w zależności od badanych genotypów. Zdecydowanie największymi zdolnościami do gromadzenia szkodliwego pierwiastka w liściach z podłoża o naturalnej jego zawartości odznaczały się gatunki: N. raimondii, N. bigelovii i odmiany botaniczne N. tabacum, natomiast w korzeniach N. rosulata. W przypadku podłoża, do którego dodano 8 mg czystego składnika najwyższą zawartość kadmu w liściach stwierdzono u N. langsdorfii, N. plumbaginifolia, N. setchelii, N. debneyi i N. velutina. Zawartość kadmu w korzeniach w tej kombinacji najwyższa była u N. miersii, N. clevelandii i N. nudicaulis. Zdecydowanie najmniej kadmu pobierały w obydwu badanych kombinacjach: N. benavidesii i N. umbratica. Różnice istotnie statystycznie wyliczone dla poszczególnych sekcji wykazano najczęściej w kombinacjach z dodawanym kadmem, rzadko w obiektach kontrolnych. Nie wykazano istotnych różnic zawartości kadmu w przypadku odmian botanicznych tego samego gatunku.
Fifty nine wild species of Nicotiana and six botanical varieties of N. tabacum and N.rustica were examined for their ability to take up and accumulate cadmium in their leaves and roots. The plants were grown on two soils: cadmium-enriched (8 ppm Cd), and untreated, with native cadmium content of 0.15 ppm as control. The leaf and root Cd content varied with the genotype and with the level of soil Cd. N. raimondii, N. bigelovii, botanical varieties of N tabacum and N. rosulata were found to be the most potent cadmium absorbers from the untreated soil. N. langsdorfii, N. plumbaginifolia, N. setchelii, N. debneyi and N. velutina took up and accumulated in their leaves large amount of cadmium from the cadmium-enriched soil. The root accumulation of cadmium from cadmium-enriched soil was high in N. miersii, N. clevelandii, and N nudicaulis. N. benavidesii and N. umbratica were found to be the weakest cadmium absorbers from both, cadmium-enriched and untreated soils. In individual sections statistically significant differences were most frequently demonstrated for the treatments with cadmium supplement and rarely for untreated-controls. No significant differences in cadmium content were found among botanical varieties within the same species.
