The aim of the dissertation was to determine (select) the best growth medium for a basket willow (Salix viminalis L.) which contains optimal concentration of kinetin. The plant material taken from three varieties of willows: Bjor, Jorr and Tora, which grew in the experimental field, was sterilized and placed on a complete MS (Murashige and Skoog, 1962) growth medium. The propagated plants were used in an experiment, where MS substrate was enriched by growth regulator – kinetin, in various concentrations (0.5 mg∙dm-3, 1 mg∙dm-3, 2 mg∙dm-3 and 3 mg∙dm-3). The substrate which contained 0.5 KIN was considered to be the best for willow’s propagation, because plants cultivated with usage of this medium have shown the largest mass, the highest number of shoots and leaves, and also the highest quantitative and elongation growth of the roots. Among examined varieties of Salix, the best one was Bjor variety, where evaluated parameters were distinctive in comparison to two remaining varieties.

The aim of the study was to determine an optimal composition of growth media for Drosera rotundifolia L. cultured in vitro. Adventitious shoots of Drosera rotundifolia originating from an established in vitro culture were placed on complete Murashige and Skoog (MS) (1962) medium, MS medium with nutrient content reduced to ¾ MS, ½ MS, ¼ MS, 1/6 MS and MS medium with selected macronutrient (KNO3, KNO3 + NH4NO3, KH2PO4 or CaCl2) content reduced by ½. The medium containing ¼ of standard MS nutrients was found the most effective, as plants in this variant had the greatest weight and developed the greatest number of adventitious shoots (37.1). It was also conducive to root growth and development. Flowering was only observed on complete MS, ¾ MS and ½ MS media and MS with reduced content of KNO3 or KNO3 + NH4NO3. Reducing mineral content to ¼ MS or more resulted in red coloration of glandular tentacles.

Opis:
Successful in vitro propagation through ex vitro rooting mechanism has been achieved in Piper longum by nodal shoot segment cultures. Shoot tip explants were less proliferative compared to the nodal meristems. The explants were sterilized using 0.1% HgCl2 and cultured on Murashige and Skoog (MS) medium with various concentrations of growth regulators. MS medium supplemented with 1.0 mg/L 6-benzylaminopurine (BAP) was found suitable for bud breaking within four weeks. Kinetin (Kin) was not reported impressive compared to BAP in culture induction response. The shoots were multiplied and elongated on MS medium supplemented with 0.5 mg/L each of BAP and Kin and 0.1 mg/L indole-3 acetic acid (IAA), where the shoots were elongated up to 5.8 cm length. Rooting and acclimatization was achieved by ex vitro rooting methods using 300 mg/L indole-3 butyric acid (IBA) for 5 min, and 5.3 roots with 3.2 cm average length were observed. The rooted shoots were transferred to the greenhouse for acclimatization. The hardened plantlets were transferred to earthen pots and maintained in the greenhouse. Normal flowering was observed in micropropagated plants of P. longum.

In vitro propagation methods using nodal segments of young tissues of Hibiscus subdariffa plant have been established. The nodal shoot segments responded better than apical shoot tip explants. Murashige and Skoog (MS) semi-solid medium supplemented with 1.0 mg/L 6-benzylaminopurine (BAP) induced 4.2 ±0.24 shoots per node with 2.40 ±0.18 cm in length after four weeks of inoculation. About 92% explants responded on this medium combination. The shoots were further multiplied (8.2 ±1.37 shoots with 5.9 ±1.05 cm length) by repeated subculture of mother explants and freshly induced shoot clumps on MS medium with the reduced concentrations of BAP and Kinetin (0.5 mg/L each) with additives within 4 weeks. Half strength MS medium augmented with 2.0 mg/L indole-3 butyric acid (IBA) was found most favorable for root induction with 98% response. On this medium 7.5 ±0.61 roots with 4.8 ±0.23 cm length of roots were recorded. The regenerated plantlets were successfully hardened and field transferred.

An effective protocol has been developed for the adventitious root culture of Cannon Ball tree (Couroupita guianensis Aubl.) from the leaves of in vitro germinated 30 days’ old seedlings. Seeds germinated on cytokinins and auxins supplemented with Murashige and Skoog (MS) medium. Half strength MS medium supplemented with 2.0 mg/L indole-3-butyric acid (IBA) was found the most suitable for the induction of adventitious roots (92%) from the midrib region of the leaves than the other auxins. The roots were proliferated on half strength MS liquid medium supplemented with IBA 0.5 mg/L. After eight weeks of inoculation the roots in the medium yielded 5.933 ±0.135 gm fresh and 2.731 ±0.010 gm dry biomass of adventitious roots. The growth rate was increased by 8-9 folds in this investigation.

Plants have been an important source of medicine for thousands of years. Medicinal plants are the most important source of life saving drugs. Even today, the WHO estimates that up to 80 per cent of people still rely mainly on traditional remedies such as herbs for their medicines. Medicinal plants are resources of new drugs. Approximately one quarter of prescribed drugs contain plant extracts or active ingredients obtained from plant substances. Medicinal plants are the most important source of life saving drugs for the majority of the world’s population. The biotechnological tools are important to select, multiply and conserve the critical genotypes of medicinal plants. In-vitro regeneration holds tremendous potential for the production of high-quality plant-based medicine. An efficient protocol was developed for high frequency and rapid regeneration was achieved in Withania somnifera (L.) Dunal. Invitro cultures were initiated by meristematic explants on M.S–B5 medium with various growth hormones individually as well as combinations. Highest shoot multiplication rates were observed when explants grown in the medium supplemented with 2.0 mg/l of BAP. M.S–B5 medium fortified with 1.0 mg/l BAP was found suitable for optimum growth and elongation of shoots. The best rooting response were observed when the medium containing 1.0 mg/l of IBA. The rooted plantlets were successfully established in soil.
