In view of the predicted increase in sulphur deficiency in crop production, a study was undertaken to assess the effect of fertilization with this nutrient on criteria determining the biological value of narrow-leafed lupin seeds (total and true protein, their ratio, nitrates and amino acid composition). The study was based on a three-year, three-factor field experiment, in which the Elf cultivar of narrow-leafed lupin (Lupine Angustifolius L.) was grown on light soil with low sulphur content. The experiment included foliar and soil application of sulphur and its elemental and ionic forms. The application doses were 0, 20, 40 and 60 kg S ha-1. In all years of the study, the sulphur application dose was found to significantly determine the total and true protein content as well as their ratios in the lupin seeds. Application of just 20 kg S ha-1 caused a significant increase in true protein in the lupin seeds and in the proportion of true protein in the total protein, as compared to the control. The CS index for all the essential amino acids showed that the first limiting amino acid in the lupin seed protein was sulphur-containing methionine. As the dose of applied sulphur increased, there was an overall increase in CSmeth. and EAAI. Given the positive effect of sulphur, particularly in the elemental form, on the protein content and amino acid composition of lupin seeds and in reducing the content of harmful nitrates, the use of this nutrient in fertilization of this species is recommended.

Embryo development of Lupinus albus L. and L. mutabilis Sweet. after pollination with L. angustifolius L. was studied. Observations of ovules revealed non-typical embryo development. From the 5th day after pollination, high number of degenerated ovules was found; the highest number was observed on 15 and 20 days after pollination. The period between the 10th and 15th day after pollination was the most suitable for isolation of embryos in all the examined combinations, with exception L. albus x L. angustifolius where optimal isolation time was between 15th and 20 th day after pollination. Embryos were obtained in each cross-pollination. Some of them developed in vitro into plantlets and grew in an artificial medium for some time, but most of them died due to non-typical development of the root system, stem, or both. The hybrid character of the survived plants requires confirmation.

A characteristic trait of lupins is a production of alkaloids, which are a toxic and bitter taste compound ofseeds. Due to the lack of fast, sensitive and inexpensive screening techniques to identify and reject high alkaloidplant material, development of suitable tools is important challenges for lupins breeding and seed production.The aim of this study was to compare two alkaloid content estimation methods in Lupinus angustifoliusL. and Lupinus albus L.During the Wagner’s colorimetric test, which is recommended by the UPOV, seed halves were stained onfour colors depending on the alkaloid content but only the level of 0.5% – 0.6% showed clear color change.Gas chromatography allowed accurate quantification and qualification of alkaloid content.Since safe alkaloid content for consumption is 0.02% of seed dry weight, colorimetric method is less usefulfor dividing lupin cultivars into sweet and bitter, than gas chromatography but can be used as a screeningtechnique.

Łubin, w porównaniu z innymi roślinami strączkowymi, zawiera niewielkie ilości związków antyżywieniowych, w tym lektyn. Dowiedziono jednak, że w jego nasionach znajdują się substancje lektynopodobne, co może być czynnikiem ograniczającym zastosowanie łubinu w żywieniu człowieka. Celem przeprowadzonych badań była ocena lektynopodobnych właściwości globulin łubinu wąskolistnego poprzez sprawdzenie, czy białka te mogą stymulować proliferację limfocytów oraz powodować aglutynację erytrocytów człowieka. Wyizolowane erytrocyty poddawano działaniu wybranych frakcji białkowych. Wynik doświadczenia oceniano mikroskopowo. Limfocytom w hodowli podawano odpowiednio: frakcje globulinowe oraz mitogen. Na podstawie ilości inkorporowanej do DNA limfocytów znakowanej radioaktywnie tymidyny określano stopień ich proliferacji. Proliferację limfocytów oceniano również z wykorzystaniem cytometrii przepływowej. Wśród erytrocytów poddanych działaniu łubinowych globulin nie zaobserwowano zjawiska aglutynacji. Podobnie, w populacji limfocytów inkubowanych z badanymi białkami łubinu nie stwierdzono podwyższonego poziomu proliferacji komórek. Na podstawie uzyskanych danych stwierdzono, że badane frakcje globulin łubinu nie wykazują typowej dla lektyn zdolności aglutynowania erytrocytów. Nie indukują również proliferacji limfocytów. Otrzymane wyniki świadczą o tym, że białka te, spożyte z pokarmem, nie mogą spowodować wystąpienia reakcji odpornościowej, wywołanej zlepianiem i aktywacją limfocytów, czy hemaglutynacją.
In comparison to other legume plants, lupin contains small amounts of anti-nutritional compounds, including lectins. However, it has been proved that lupin seeds may contain some lectin-like substances, which can limit the use of lupin in human nutrition. The objective of the study was to evaluate lectin-like properties of narrow-leafed lupin seed globulins. To achieve this, their potential for stimulating lymphocyte proliferation as well as their hemagglutinating properties were analysed. The isolated erythrocytes were treated with some selected protein fractions. The result of the experiment was microscopically assessed. The cultured lymphocytes were treated with globulin fractions and a mitogen, respectively. The amount of proliferating cells was evaluated based on the amount of radioactive thymidine incorporated into the DNA of lymphocytes. The proliferation of lymphocytes was also assessed using a flow cytometry. No cell agglutination was found among the erythrocytes treated with the lupin globulins. Furthermore, the level of cell proliferation did not increase in the population of lymphocytes incubated with the lupin proteins studied, either. Based on the results achieved, it was found that the lupin globulin fractions analysed did not show the ability, appearing typical for lectins, to agglutinate erythrocytes. They did not induce the proliferation of lymphocytes, either. The results obtained prove that those proteins, if eaten with food products, cannot cause the immune reaction to occur owing to the agglutination and activation of lymphocytes or to hemagglutination.

The aim of the studies was to establish in vitro conditions for the culture of pea and lupin embryos as the first step in the development of an in vitro assisted single seed descent technique for the attainment of homozygous populations. Materials for the study included of pea, and narrow-leafed and yellow lupin cultivars. Embryos dissected from mature but still-green seeds were cultured in vitro on two modified MS media and under three temperature regimes. Shoot and root lengths of regenerated plants were measured after 7, 14 and 21 days of culture. For pea plants full-strength MS medium with 4 g l−1 agar and temperature 22/ 20°C (day/night) appeared to be the most conducive to shoot and root development, whereas for lupin plants lower temperatures were more propitious: 12°C in the dark for narrow-leafed lupin and 16/ 12°C (day/night) for yellow lupin. Almost all the cultured embryos developed into plants, but not all the regenerated plants survived acclimation to ex vitro conditions.

The aim of the research was to determine changes in the chemical composition in lupin seeds during seed germination in a solution containing selected nitrogen compounds. Blue lupin seeds of the Neptun cultivar were germinated for 3 or 4 days in darkness, at 24°C in 1% solutions containing the following nitrogen compounds: urea, nitric acid, ammonium sulphate, methionine or yeast extract. The control consisted of seeds germinated in water. The presence of all nitrogen sources has resulted in a poorer growth of the sprouts as compared to seeds germinated in water. There was a significant increase in the true protein content as well as a significant decrease in the content of crude ash and alkaloids in the germinated seeds. The seeds germinated for 4 days had significantly higher total protein content and significantly lower content of nitrogen-free extract compounds and oligosaccharides as compared to the seeds germinated for 3 days. A significant influence of the nitrogen source on the concentration of some amino acids was found. Cystine was found to be absent in seeds germinated in the nitric acid solution, while there occurred a twofold increase in the cystine content and a fivefold increase in the methionine content in seeds germinated in the aqueous solution of methionine and an 50% increase in the cystine concentration was observed in seeds germinated in the yeast extract and ammonium sulphate solution. Methionine and cystine were the limiting amino acids in all the samples, accept of seeds germinated in the methionine solution were it was valine. Aqueous solutions of ammonium sulphate and yeast extract were found to have the most advantageous influence on the chemical composition of lupin germination products.

The pollen grain germination and the growth of pollen tubes after intra- and interspecific reciprocal pollination of Lupinus albus L., L. mutabilis Sweet, and L. angustifolius L. were examined. Results of microscopic observations of pollen grain germination and pollen tube growth enable to deny the existence of certain barriers that prevent fertilization after pollination of L. albus flowers with pollen of L. angustifolius and vice versa, as well as after pollination of L. mutabilis flowers with pollen of L. angustifolius and vice versa. Some delay in pollen grain germination and a slower growth of pollen tubes were observed in all combinations, as related to intraspecific ones, though in the case of each interspecific combination pollen tubes reached the ovary. Moreover, a different level of pollination efficiency (the number of growing pollen tubes) and various rates of pollen grain germination and pollen tube growth, depending on the direction of pollination, were found. The noted pollination efficiency may generally be considered to be low.

Fusarium wilt is the most harmful soil-borne disease of lupins in Russia and in the Ukraine. The aim of the studies was to find broad resistance to disease in collection accessions and to breed lupin forms resistant to Fusarium wilt for the future breeding program. Lupin cultivars and lines (547 accessions) from the genebank of the N.I. Vavilov All Russian Scientific Research Institute of Plant Industry, St. Petersburg, Russia, were tested for Fusarium resistance under different environmental conditions of three regions in Russia and in the Ukraine on plots with artificially infested soil. So, differences in the disease susceptibility of the same accessions were found in contrasting environments. Resistant forms from one region were crossed with accessions showing their resistance in two other regions to accumulate resistance genes in new genotypes. As a result of hybridization two transgressive forms were obtained in F₄ of the crosses cv. Frost × cv. Apendrilon (L. angustifolius) and line G-413 × line 85 (L. luleus). Their resistance in all three regions appeared to be higher than that of their parental forms. These two lines were found suitable for the breeding programme of Fusarium resistant forms in Russia and in the Ukraine.

W pracy zbadano wpływ czynników pogodowych na plonowanie odmiany Mirela łubinu wąskolistnego w latach 1990-2008 oraz dokonano prognozy plonów na lata 2050-2060, z wykorzystaniem modeli pogoda-plon, przy założeniu trzech scenariuszy zmian klimatu przeznaczonych dla Europy Środkowej: GISS model E, HadCM3 i GFDL. Materiał źródłowy stanowiły wyniki doświadczeń odmianowych oraz obserwacje meteorologiczne pochodzące ze stacji doświadczalnych COBORU, zlokalizowanych w północno-wschodniej Polsce. W analizach zastosowano metodę korelacji i regresji wielokrotnej krokowej postępującej. Dopasowanie modeli oceniono na podstawie współczynnika determinacji R², poprawionego współczynnika determinacji R² adj, błędu standardowego estymacji oraz współczynnika determinacji R² pred wyznaczonego przy użyciu procedury Cross Validation. Istotność równań regresji określono za pomocą testu F-Snedecora. Jedno z równań posłużyło do prognozy plonowania łubinu w warunkach podwojenia zawartości CO2 w atmosferze. Stwierdzono, że wpływ czynników meteorologicznych na plonowanie łubinu wąskolistnego był zróżnicowany w zależności od lokalizacji stacji. Na wysokość plonu najczęściej istotnie wpływała temperatura (maksymalna, średnia i minimalna) początkowych okresów wegetacji oraz opady w okresie koniec kwitnienia–dojrzałość techniczna. Badania wykazały, że przewidywane zmiany klimatu wpłyną korzystnie na plonowanie łubinu. Plony symulowane były wyższe od uzyskiwanych w latach 1990-2008, a najbardziej korzystny okazał się scenariusz HadCM3.
The study examined the impact of weather factors on yielding of blue lupine cultivar Mirela in 1990-2008 and forecasted the yields for 2050-2060 using the weather-yield models and three climate change scenarios for Central Europe: GISS model E, HadCM3 and GFDL. The source material was the results of cultivar experiments and meteorological observations obtained from the COBORU research stations, situated in North-Eastern Poland. The analyses included the correlation and multiple progressive stepwise regression methods. The fitting of the models was evaluated with the coefficient of determination R², adjusted coefficient of determination R² adj, standard estimation error and coefficient of determination R² pred calculated with the Cross Validation procedure. The significance of regression equations was determined by a F-Snedecor test. The selected equation was used to prediction the yielding of lupin in conditions of doubling CO2 content in the atmosphere. It was found that the impact of meteorological factors on the yielding of blue lupine was diversified depending on the location of the station. Most frequently, the yield was significantly influenced by temperature (maximum, average, minimum) in the beginning of growing seasons and also by precipitation in the period: the end of flowering – technical maturity. The studies demonstrated that the predicted climatic changes would exert a beneficial impact on lupin yielding. Simulated yields were higher than those observed in 1990-2008 and the most favourable scenario was HadCM3.

A great progress had been made in narrow-leafed lupin in a comparatively short period. Despite elimination of basic disadvantages (hard seeds, high content of alkaloids, pod shattering) disease-resistant, unbranched and thermoneutral cultivars have been bred. Investigations on the variation range and inheritance mode of commercially important characters were followed by breeding achievements. The paper contains a review of references for 35 genes controlling alkaloid content, growth rhythm, branching, vegetation length, shape and color of leaves, color of flowers and seeds, seed hardiness, pod characters and disease resistance. Authors of characters and gene symbols from different research centres and from different years are given on the basis of priority. The importance of these genes for breeding improvement is discussed. Type lines for characters/genes and genotype of more important cultivars collected in the Polish narrow-leafed lupin genebank are presented.
