Opis:
Tomato is an economically important vegetable crop which is attacked heavily by insect pests leading to reduction of yield and quality of the fruits. Field experiments were carried out to investigate the dissipation of methomyl (a common insecticide) used mainly on tomato fruits. LC-MS/MS coupled with the QuEChERS method were used for the determination of methomyl. The results showed that the recovery using matrix-matched standards ranged from 87.8 to 101.3%, with relative standard deviation of 2.5 to 7.5%. Kinetics equation, Log R = log R0 – 0.434 Kt, was used to calculate the rate of degradation in tomato, soil and water. Residue half-life calculated using kinetic rate ranged from 1.95 to 1.63 days in tomato and soil, respectively. From the results it was concluded that tomato fruits can be safely harvested for consumption after 15 days of application based on estimated preharvest interval (PHI). It is advisable to re-estimate the PHI regularly owing to data from the EU and Codex.

Opis:
The rapid analytical method was developed in response to increasing concern over the environmental impact of azoles (sterol biosynthesis inhibitors) and neonicotinoids (nicotinic acetylcholine receptor site). These chemicals are commonly used to protect fruit and vegetables crops against fungi and pests. Seven insecticides and twenty one fungicides commonly occurring in food industrial wastewater have been determined. For this purpose, active substances from two new pesticide classes were extracted and isolated by QuEChERS by addition of acetonitrile, buffering salts and chitin as a clean-up sorbent. The novelty of this procedure was one step sample preparation including extraction and removing of co-extracts in short time. Instrumental analysis was conducted by liquid chromatography coupled with mass spectrometry using multiple reaction monitoring. The limits of detection ranged from 0.002 to 0.005 μg·L-1 with satisfactory accuracy and precision The recoveries for the pesticides ranged from 81–103%, with high repeatability (n = 3, RSD ≤ 9%) and low LOQs (0.01 μg·L-1). Matrix effects calculated were less than 12% for all analyses. The method was applied to routine analysis of food industrial wastewater. Concerning the results, total pesticide levels in most cases were below 1 μg·L -1. The most significant pesticides in terms of concentration and frequency of detection were acetamiprid (0.07 μg· L-1); tebuconazole (1.2 μg· L-1) and thiacloprid (0.04 μg·L-1).

Introduction and objective. Acrylamide is a “probably human carcinogen” monomer that can form in heated starchy food as a result of a reaction between asparagine and reducing sugars via Maillard reaction. The main source of acrylamide in human diet are potato products, cereal products and coffee. Tobacco smoke may be another significant source of exposure to acrylamide. The aim of our study was to determine acrylamide content in cigarettes available on the Polish market and to estimate the exposure to acrylamide originating from tobacco smoke in smokers in Poland. Materials and methods. The material was cigarettes of the top five brands bought in Poland and tobacco from non-smoked cigarettes. Acrylamide content in cigarettes mainstream smoke was determined by LC-MS/MS. Exposure assessment was carried out using analytical data of acrylamide content in cigarettes and the mean quantity of cigarettes smoked daily by smokers in Poland, assuming body weight at 70 kg. Results. The mean content of acrylamide was 679.3 ng/cigarette (range: 455.0 – 822.5 ng/cigarette). The content of acrylamide was evidenced to correlate positively with total particulate matter (TPM) content in cigarettes. The estimated average exposure to acrylamide from tobacco smoke in adult smokers in Poland is 0.17 μg/kg b.w./day. Conclusions. Our results demonstrate that tobacco smoke is a significant source of acrylamide and total exposure to acrylamide in the population of smokers, on average, is higher by more than 50% in comparison with non-smokers. Our estimation of exposure to acrylamide from tobacco smoke is the first estimation taking into account the actual determined acrylamide content in the cigarettes available on the market.

A rapid, accurate and precise LC-MS method is described for the quantitative determination of pramipexole in human plasma matrix using ropinirole as internal standard. Pramipexole and ropinirole were extracted from plasma by liquid-liquid extraction technique. The method was validated over the concentration range of 100-2514 pg/mL. The method was found to have acceptable accuracy, precision, linearity and selectivity. The mean extraction recovery from spiked plasma samples was in the range of 79.415-87.00 %. The intra-day accuracy of the assay ranged from 98.924 to 112.236 % and intra-day precision ranged from 3.489 to 6.756 %. Inter-day accuracy and precision results for quality control samples ranged between 100.340 and 107.443% of nominal and precision is observed to be 3.970-5.714 %. The pramipexole was found to be stable after several stability studies. The proposed method yielded a quick, simple and reliable protocol for estimating pramipexole concentrations in human plasma.

Betacyanins are natural, red-violet betalain pigments which can be found in plenty of plants of the Cactaceae family. The Mammillaria is the widest genus of the Cactaceae family of which fruit was examined for betacyanins contents. As a result of the investigation, a new pigment structure, mammillarinin, was identified. Heretofore, no betacyanins of Mammillaria coronata flowers has been extensively studied. In this report, the results of betacyanin analysis by LC-DAD-ESI-MS/MS in the flower extract are presented.

Background: Lopinavir, saquinavir, and ritonavir are viral protease inhibitors (PIs) developed for and widely used in the therapy of human immunodeficiency virus (HIV)-related disease. These compounds are also active in vitro against the pathogens causing tuberculosis, malaria and coronavirus infections. PIs have been regarded as a platform for the design of inhibitors targeting severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2)-encoded proteases. This study aimed to develop a liquid chromatography/mass spectrometry (LC/MS) procedure for accurate simultaneous determination of concentrations of these three PIs in the plasma. Methods: Samples of human plasma were protein precipitated with 0.3 M zinc sulfate in a water/methanol solution (30:70, v/v). The extracts were analyzed with reversed-phase chromatography coupled with the electrospray ionization (ESI) source of the ion trap mass detector operating in mEass spectrometry (MS) and tandem mass spectrometry (MS/MS) modes. Results: Calibration curves demonstrated good linearity from 0.01 to 10 µg/mL and acceptable reproducibilities and recoveries. Conclusions: The described procedure proves that a very basic ion-trap LC/MS system could be applied for selective, rapid, and precise determination of antiviral protease inhibitors.

Background Cytotoxic antineoplastic drugs (ADs), widely used in treating cancer, are considered hazardous in the workplace and thus require safe handling practices. An analytical protocol for environmental and biological AD monitoring in the healthcare environment has been developed, since Europe lacks clear guidelines and regulations for cytostatic preparation and handling. Material and Methods Liquid chromatography-tandem mass spectrometry (LC-MS/MS) was used for measuring contemporaneously 20 multi-class cytostatic compounds and urinary α-fluoro-β-alanine, whereas platinum was detected by inductively coupled plasma mass spectrometry (ICP-MS). Sampling procedures and analytical conditions were optimized and the assays were validated. Environmental AD monitoring data, collected in 2009–2017, for a total of 3749 wipe tests and 57 720 determinations, was evaluated. Results The proportion of positive samples gradually decreased from 11.7% in 2010 to 1% in 2017, however, 2266 determinations were positive. No urine sample had detectable concentrations of any of the 4 drugs considered (0/398 samples). Conclusions These improvements are so large that the key role played by the new, more stringent rules for preparing and administering ADs is evident. Hence, the analytical method involving multi-element determinations allows for a more thorough and complete investigation into the AD contamination of work environments. Med Pr 2018;69(6):589–604

Beta vulgaris L. is an important source of bioactive saponins – a group of secondary metabolites – that have spurred a growing interest due to their health-promoting properties. This study aimed to gain information on triterpene saponin profile of the peel and flesh of white, yellow and red beet of six cultivars – Snow Ball, Boldor, Ceryl, Chrobry, Forono and Tytus – harvested in Poland, in the same region. Twenty four saponins with oleanolic acid, hederagenin, akebonoic acid and gypsogenin as aglycons were identified and quantified by liquid chromatography/tandem mass spectrometry (LC-ESI-MS/MS). Among them, betavulgaroside I, II, III and IV were the major compounds, but the quantitative profile of saponins was found to be dependent on beet cultivar and root part, respectively. The highest content of saponins was found in the peel of yellow B. vulgaris Boldor (20812 mg/kg fresh weight, fw), while the lowest saponin content was determined in the flesh of white B. vulgaris Snow Ball (497 mg/kg fw). In addition, the total saponin content in peel and flesh in yellow beet (26054 mg/kg fw) was much higher than the total content in peel and flesh in red beet Tytus (8364 mg/kg fw) and white beet Snow Ball (1204 mg/kg fw). This is the first report on the profile of saponins in white and yellow beets.

Opis:
Rothmannia wittii, which is found in Binh Chau-Phuoc Buu Nature Reserve, Bung Rieng ward, Xuyen Moc District, Ba Ria-Vung Tau Province, Vietnam, is a rare species of Rubiaceae family. The present study aimed to determine the phytochemical composition and antimicrobial activity of the ethanolic extract of the R. wittii trunk. A total of 10 compounds were identified by liquid chromatography-mass spectrophotometry (LC-MS), including 1) benzene, 1-(1,5-dimethyl-4- hexenyl)-4-methyl, 2) diphenylsulfone, 3) 6β-hydroxy-10-O-acetylgenipin, 4) cinnamyl-cinnamate, 5) genipin, 6) megastigmatrienone, 7) ethyl p-methoxy-trans-cinnamate, 8) rothalazepane, 9) 1,3,6-trimethylpyrano[2,3-d ]imidazole-2,5(1H,3H)-dione, and 10) shanzhiol. The extract also exhibited an antibacterial activity against six bacterial strains: Bacillus cereus, Pseudomonas aeruginosa, Escherichia coli, Staphylococcus aureus, Salmonella typhimurium and Salmonella enteritidis, as determined via disk diffusion agar method.

This study was focused on the identification and quantification of betaxanthins using high-performance liquid chromatography with a diode array detector coupled to electrospray ionisation mass spectrometry (HPLC-DAD-ESI-MS) in leaves of various cultivars of Beta vulgaris (beet) and Gymnocalycium mihanovichii grafted cacti. In G. mihanovichii grafted cacti, four betaxanthins, namely histidine-Bx, histamine-Bx, serine-Bx, and proline-Bx, were tentatively identified in the yellow, orange, pink, and red varieties, with contents ranging from 0.09 to 1.55 mg/kg fresh weight (FW). Betaxanthins were not detected in the green cactus. Histidine-Bx was the prevailing betaxanthin compound in the majority of cultivars. Fifteen betaxanthins were successfully identified in the leaves of five B. vulgaris cultivars (cv.): Snow Ball, Boldor, Cylindra, Rhubarb, and Round Dark Red. Leaves of yellow beet (cv. Boldor) had the highest total betaxanthin content (20.4 mg/kg FW), while white beet (cv. Snow Ball) had the lowest one (3.43 mg/kg FW). The leaves of red cultivars had comparable betaxanthin contents, ranging from 13.4 to 18.8 mg/kg FW, similary to the yellow cultivar, indicating their potential as valuable sources of betaxanthins. There was no single dominant betaxanthin in B. vulgaris leaves. The leaves of B. vulgaris were found to be a richer source of betaxanthin than the grafted cactus G. mihanovichii, which opens up new prospects for research into the potential utilization of these compounds in the industry.

The text presents the results of laboratory analyses conducted on vegetal intoxicating substances identified on the walls of selected pottery forms discovered at Polish sites attributed to the south-eastern group of the FBC. The samples taken from miniature vessels and collared flasks were examined using the GC-MS/MS method (triple quadrupole) and then the reference method LC-MS/MS (linear ion trap). As a result of the research, psychotropic substances were identified in four samples: papaverine, scopolamine and atropine.

The study investigates the relationship between soilsthat vary for different basic physicochemical parameters (pH,phosphorus and potassium content, granulometric compositionand soil organic matter content) and the free amino acids content of Stellaria media. According to some research, plant aminoacid profile is a good indicator of the condition of the soils onwhich plants are grown. In this experiment, a rapid and sensitiveliquid chromatography coupled with tandem mass spectrometry(LC-MS/MS) method was used for the identification and quantification of nineteen proteinogenic amino acids in Stellaria mediasamples, which is a very common weed found worldwide. Significant linkage was found between the soil parameters and Stellariamedia amino acid content. Garden soil is the most favourable soilfor amino acid content. Glutamine, glutamic and aspartic acid arethe most abundant amino acids in Stellaria media grown on allthree tested soils.

Betacyanins are conjugates of betalamic acid and glycosylated cyclo-DOPA which bring about the creation of a specific chromophoric system, the 1,7-dia- zaheptamethin chain. Betanin, a betacyanin pigment from red beet (Beta vulga- ris L.), as well as its decarboxylated derivatives were subjected to reduction with the use of NaBH4. The pigment structures contain a vinyl linker which is postu- lated to be protonated during reaction with NaBH4. In this study, 17-decarboxy- betanin and 2,17-bidecarboxy-betanin were obtained from betanin, which had been previously isolated from red beet root (Beta vulgaris L.). Purified pigments were tested in the presence of the reduction reagent while looking for the prod- ucts of the reaction identified by LC-MS.

Apple processing into juice generates vast amounts of a by-product, namely fruit pomace, which poses a serious problem for the processing industry. At the same time, fruit pomace features a high health potential. The aim of the present study was to develop recipes of 8 variants of cookies with wheat flour substituted by apple pomace (0, 10, 30 and 50% of flour weight), sweetened with sucrose and erythritol. The cookies were analyzed for their nutritional value; antioxidant capacity (ABTS•+ scavenging activity and oxygen radical absorption capacity – ORAC); the ability to inhibit α-amylase, α-glucosidase and pancreatic lipase; and consumer acceptability. In total, 13 phenolic compounds were identified in the cookies with pomace. Cookies with 50% addition of apple pomace had an approximately 8-fold higher content of dietary fibre than traditional products (without the apple pomace) and simultaneously reduced energy value (by 32.6 and 40.5 kcal/100 g of cookies sweetened with sucrose and erythritol, respectively). The antioxidant capacity of cookies was 0.032–0.316 mmol TE/100 g in the ABTS assay and 1.153–2.070 mmol TE/100 g when ORAC was determined. The IC50 enabling α-amylase and α-glucosidase inhibition ranged from 138.1 to 221.8 mg/mL and from 976.4 to 1374.9 mg/mL, respectively. The anti-lipase activity of cookies with the addition of 50% apple pomace and erythritol was the highest (IC50 of 7.3 mg/mL). Both antioxidant capacity and antidiabetic potential increased significantly with the increasing proportion of pomace in cookies. Replacing sucrose with erythritol favorably influenced the consumer assessment. The study results show that the proposed products can be a perfect alternative to traditional sweet snack products, especially for consumers with diet-related diseases. The feasibility of using waste raw materials, which are a challenge to the food industry, has been proven as well.

Celem pracy przedstawionej w artykule była analiza stosowanych metod oznaczania mikotoksyn, takich jak ochratoksyna A i zearalenon w żywności oraz w paszach.
Due to the necessity for determining mycotoxins in accordance with the guidelines of the European Commission (EC) and the growing interest of the problem of mycotoxins due to the changing climate in Poland and in the world, many scientific institutions and companies to develop new methods for their determination. The aim of the study was to discuss the most commonly used methods for the determination of mycotoxins, such as ochratoxin A and zearalenone in food and feed.

In this study, a chromatographic fractionation of betacyanin pigments from extract of purple Gomphrena globosa petals was performed by preparative high performance liquid chromatography (prep-HPLC). The particular betacyanins in each collected fractions were tentatively identified by chromatography with optical amd mass spetrometric detection (LC-DAD-ESI-MS). Betacyanins are natural pigments, which are confirmed to have an antioxidant activity. It was reported that betacyanins can prevent civilization diseases, because of an ability of free radicals scavenging. Betacyanins are present in tissue of plants from Amaranthaceae family, e.g. in petals of Gomphrena globosa. Preparative separation of these particular pigments is difficult, because they are structurally very similar to each other and tend to coelute. However, in this study, fractions, containing a dominant amount of the principal pigments of Gomphrena globosa inflorescences, were obtained successfully. In the first fractions, two isomeric pigments assigned to gomphrenin I and isogomphrenin I were detected. The fractions of significant amounts of gomphrenin III and isogomphrenin III were obtained separately with sufficient purity. The presence of other, minor gomphrenin-type betacyanins were also confirmed as: gomphrenin II, cis-isomer of gomphrenin II and sinapoyl-gomphrenin I as well as their 15S-diastereomers. Moreover, in other fractions, various unknown pigments were detected.

This work presents a comparative analysis of the phenolic composition (UHPLC-PDA-ESI-MS 3 , HPLC-PDA- fingerprint, UV-spectrophotometric methods) and antioxidant activity (DPPH, FRAP) of leaf samples from two vegetation seasons of a medicinal and dietary plant Sorbus domestica growing in its natural habitat (Croatia, C) and cultivated in Poland (P). The samples from both sources were rich in structurally diverse polyphenols (44 analytes; P: 73.4–76.6 and C: 98.3–106.7 mg GAE/g dry leaves) including the dominating flavan-3-ols and flavonoids. The greatest qualitative and quantitative differences were observed for flavonoids (P: 14.3–20.3%; C: 27.5–34.1% of polyphenols) – in the Polish samples flavonoid diglycosides predominated, in the Croatian samples the contents of both monoglycosides and diglycosides were similar. In the case of dry methanolic extracts, despite the higher extraction efficiency obtained for the Croatian samples (32–36% vs 23–24%), the quality of the extracts was comparable, both in terms of the total phenolic content (P: 269.4–280.0; C: 297.6–304.4; mg GAE/g dry extract) and antioxidant activity parameters (DPPH, EC 50 , μg/mL. P: 10.5–10.9, C: 10.0–10.3; and FRAP, mmol Fe 2+/g, P: 6.64–7.13, C: 7.06–7.11). As a result, the study confirmed the influence of environmental conditions on the phenolic profile and antioxidant capacity of S. domestica leaves, as well as showed that despite some differences, plant materials from both Poland and Croatia might be suitable for production of natural health products.

The report demonstrates that plenty of methylated flavones of the plant, as the end product of the flavone biosynthesis pathway, are concentrated in the root bark, which largely remains in the waste that makes up 7% w/w of the commercial root and is discarded. The study of the waste extract showed that the last enriched with monomethylated flavones wogonin and oroxylin A, along with some polymethylated ones represent an end of the biosynthesis pathway of flavones in the plant. In addition to wogonin, 7 known methylated flavones were found for the first time in the root bark. Due to a high content of wogonin and oroxylin A in the extract, these flavones were concentrated using preparative liquid chromatography. As a result, the obtained complex of these two target flavones turned out to be 0.8% w/w of the waste. Such morphologically specific accumulation of methylated flavones at the root boundary raises the question of their role in plant life.

Opis:
Natural organic dyes are group of substances that belong to various types of chemical compounds. The most commonly used in paintings and dyeing textiles were naturally occurring dyestuffs from group of anthraquinones, flavones and indigoid dyes. Identification of coloring substances present in historical artistic paints provides relevant information for a wide range of specialists dealing with works of art and in the field of conservation science. This research presents a review of analytical techniques (mainly chromatographic and spectroscopic) used for identification of dyes. It also describes the methods of colorants isolation from historical artistic paints and textiles. Furthermore, the article presents a brief characteristics of fragmentation reactions of selected groups of dyestuffs, which is useful in their identification.

Introduction. Functional food plays an important role in the prevention, management and treatment of chronic diseases. One of the most interesting techniques of functional food production is extrusion-cooking. Functional foods may include such items as puffed cereals, breads and beverages that are fortified with vitamins, some nutraceuticals and herbs. Due to its pharmacological activity, chamomile flowers are the most popular components added to functional food. Objective. Quantitative analysis of polyphenolic antioxidants, as well as comparison of various methods for the extraction of phenolic compounds from corn puffed cereals, puffed cereals with an addition of chamomile (3, 5, 10 and 20%) and from Chamomillae anthodium. Materials and Methods. Two modern extraction methods – ultrasound assisted extraction (UAE) at 40 °C and 60 °C, as well as accelerated solvent extraction (ASE) at 100 °C and 120 °C were used for the isolation of polyphenols from functional food. Analysis of flavonoids and phenolic acids was carried out using reversed-phase high-performance liquid chromatography and electrospray ionization mass spectrometry (LC-ESI-MS/MS). Results and Conclusions. For most of the analyzed compounds, the highest yields were obtained by ultrasound assisted extraction. The highest temperature during the ultrasonification process (60 °C) increased the efficiency of extraction, without degradation of polyphenols. UAE easily arrives at extraction equilibrium and therefore permits shorter periods of time, reducing the energy input. Furthermore, UAE meets the requirements of ‘Green Chemistry’.

Betanin and neobetanin (14,15-dehydrogenated betanin), natural betacyanin pigments were subjected to a gentle reduction by 2-methylpyridine borane. During each reduction reaction, the visible spectra were collected. The results were tentatively confirmed by LC-DAD-ESI-MS. In comparison to a previous study with sodium borohydride, various types of products were obtained. Depending on pH, one or two double bonds are reduced. Interestingly, a preliminary decarboxylation of the pigments is also noticed. Betacyanins are plant red-violet pigments applied as colorants in food and pharmaceutical industry which are proven to have an antioxidant activity. They are confirmed to be beneficial for human health as free radical scavengers. Therefore, betacyanins can prevent from many diseases which would be induced by an excessive accumulation of free radicals in the human body. There are more and more reports of their pro-health properties. However, the mechanism of betacyanins oxidation remains not completely elucidated. It is postulated that the rearrangement in the chromophoric system is a significant part of this mechanism. The products obtained as a result of reduction of betanin and neobetanin can be useful in oxidation investigations. Betacyanins with partially destroyed chromophoric system by reduction of the double bonds would by perfect models for further studies on their oxidation.

Izoprostany to duża grupa związków, które budową przypominają prostaglandyny. Powstają jako produkty wolnorodnikowej reakcji utleniania wielonienasyconych kwasów tłuszczowych i występują powszechnie we wszystkich tkankach oraz płynach biologicznych ciała w możliwym do oznaczenia stężeniu. Od 2018 r. oznaczanie izoprostanów techniką wysokosprawnej chromatografii cieczowej lub gazowej sprzężonej ze spektrometrią mas jest uznawane za złoty standard wśród oznaczeń markerów stresu oksydacyjnego w kontekście oksydacyjnych uszkodzeń lipidów. Publikacja jest syntetycznym przeglądem najnowszej literatury dotyczącej wykorzystania izoprostanów jako markera peroksydacji lipidów z użyciem techniki wysokosprawnej chromatografii cieczowej sprzężonej z tandemową spektrometrią mas. Przedstawiono w niej wyniki badań wykorzystujące oznaczanie izoprostanów w pracy klinicystów i wskazano potencjał zastosowania tych oznaczeń do monitoringu osób pracujących w narażeniu na szkodliwe substancje oraz ich przydatność w badaniach dotyczących stylu życia. Isoprostanes are a large group of compounds formed as products of free radical oxidation of polyunsaturated fatty acids, which are isomers of prostaglandin. They are present in all body tissues and biological fluids in quantifiable concentrations. Since 2018, the determination of isoprostanes by chromatographic technique with mass spectrometry is the golden standard of the oxidative stress markers determination in relation to oxidative damage to lipids. The publication is a synthetic review of recently published articles on the use of isoprostanes as a marker of lipid peroxidation determined with the liquid chromatography with tandem mass spectrometry technique. It presents the results of research using isoprostanes as a marker in medicine, in monitoring people working in exposure to harmful substances and in lifestyle research. Med Pr. 2023;74(2)

Metakroleina oraz keton metylowo-winylowy są bardzo reaktywnymi zawiązkami karbonylowymi, które odgrywają istotną rolę w procesach atmosferycznych, w tym – w tworzeniu pyłów zwieszonych w powietrzu. Obydwa związki są produktami utleniania izoprenu – węglowodoru emitowanego do atmosfery w ogromnych ilościach. W pracy przedyskutowano mechanizmy chemiczne zaniku metakroleiny oraz ketonu metylowo-winylowego w wyniku addycji anionu wodorosiarczanowego w rozcieńczonych roztworach wodnych, odzwierciedlające procesy zachodzące w kroplach wód atmosferycznych w rejonach o znaczącym stężeniu ditlenku siarki. W pracy pokazano, że reakcje addycji anionu wodorosiarczynowego do badanych związków karbonylowych prowadzą do tworzenia pierwotnych i wtórnych hydroksysulfonianów, które mogą uczestniczyć w tworzeniu aerozoli atmosferycznych. Wyznaczono stałe szybkości wszystkich reakcji w mechanizmie addycji. Pierwotna addycja anionów wodorosiarczynowych do metakroleiny okazała się znacznie szybsza niż do ketonu metylowo-winylowego (stałe szybkości kMAC1f = 8 i kMVK1f = 0.18 mol–1 dm3 s–1 w temperaturach pokojowych). Stała szybkości addycji do cząsteczki metakroleiny była dziesięciokrotnie większa od stałej opublikowanej w literaturze. Analiza kinetyczna i badania produktów reakcji za pomocą spektrometrii mas wykazały, że w przypadku każdego z badanych związków karbonylowych dominującym produktem reakcji był C4 alfa-hydroksysulfonian (addukt pierwotny), natomiast C4 hydroksydwusulfonian (addukt wtórny, diaddukt) powstawał w ilościach śladowych. Pierwotna addycja anionów wodorosiarczynowych do metakroleiny i ketonu metylowo-winylowego może mieć znaczenie w konwersji reaktywnych związków karbonylowych w atmosferze i powinna być uwzględniana w badaniach dotyczących rejonów o znaczącej obecności ditlenku siarki, gwarantującej wysokie stężenia anionów wodorosiarczynowych w wodach atmosferycznych.
Methacrolein and methyl vinyl ketone are highly reactive carbonyls that play a pivotal role in the formation of secondary organic aerosols in the Earth’s atmosphere. Both carbonyls are the major products of isoprene oxidation. We show that among the atmospheric sinks of methacrolein and methyl vinyl ketone, the aqueous-phase addition of bisulfite anions to their molecules can be relevant under polluted conditions with the increased presence of sulfur dioxide. We demonstrate that aqueous-phase reactions of methyl vinyl ketone and methacrolein with bisulfite anions lead to the formation of primary and secondary organic hydroxysulfonates which currently are not included in the atmospheric chemistry modelling, but can be relevant in mechanisms explaining the formation and growth of the secondary organic aerosols from atmospheric carbonyls. The rate constants for all aqueous-phase reactions involved were determined. The primary addition of bisulfite anions to methacrolein was found significantly faster than that to methyl vinyl ketone, with rate constants kMAC1f = 8 and kMVK1f = 0.18 mol–1 dm3 s–1 at room temperatures, respectively. The rate constant for the bisulfite addition to methacrolein was ten times faster than reported in the literature. The kinetic and mass spectrometric analyses revealed that in both cases, the dominating product was the C4 alpha-hydroxysulfonate (a primary adduct), while the C4 alpha-hydroxy disulfonate (a secondary adduct or diadduct) was produced only in trace quantities. The primary addition of bisufite anions to methacrolein and methyl vinyl ketone should be considered in atmospheric studies relevant to areas with enhanced presence of sulfur dioxide providing sufficiently high concentrations of bisulfite ions in atmospheric waters.

Opis:
Renal cell carcinoma (RCC) and bladder cancer (BC) are among the most frequently diagnosed urinary system cancers worldwide. They are characterized by high mortality and recurrence rates. In response to the rising incidence and mortality rates, scientists are exploring innovative diagnostic and therapeutic methods. Metabolomics, which analyzes metabolite levels, may enable early diagnosis and monitoring of therapy progress. Compared to other omics technologies, it focuses on the outcomes of metabolite activity, providing a unique perspective on processes occurring in cancer cells. Metabolomic analyses utilize techniques such as mass spectrometry. These methods allow the identification of biomarkers and precise determination of the chemical composition of biological samples. However, the most commonly used method is liquid chromatography-mass spectrometry (LC-MS), which enables the most comprehensive screening of cancer metabolomes. Recent studies show significant progress in recognizing characteristic metabolites associated with urological cancers, although this area remains partially unexplored. Research on circulating metabolites, especially those present in easily accessible samples like blood or urine, demonstrates promising potential in clinical practice. Study results reveal differences in metabolic profiles between various stages of cancer development, which may have clinical significance. The future of this field involves an increasing number of clinical cohorts, standardization of sample preparation, and further improvements in instrument sensitivity and speed. LC-MS-based metabolomics has the potential to contribute to the improvement of diagnostics, therapy, and the quality of life of patients with some urological cancers. However, challenges, such as the lack of uniform methodologies and understanding of metabolite determinants, require further research and innovation.

Opis:
Spektrometria mas sprzężona z chromatografią cieczową (LC/MS) lub chromatografią gazową oraz spektroskopia magnetycznego rezonansu jądrowego (NMR) są trzema technikami analitycznymi, które umożliwiają i w największym stopniu przyczyniają się obecnie do rozwoju badań metabolomicznych. Wśród technik LC/MS szczególną rolę odgrywają te zapewniające dokładny pomiar masy. W badaniach metabolomicznych najczęściej wykorzystywane są wysokorozdzielcze spektrometry mas (HRMS) typu kwadrupol-analizator czasu przelotu jonów (QTOF) oraz Orbitrap. W pracy przedstawiono przykłady zastosowań LC/QTOF w analizach metabolomicznych, krótko omówiono rolę optymalizacji metody analitycznej do badań metabolomicznych, wskazano zalety i ograniczenia techniki oraz wyzwania i perspektywy jej rozwoju w tym obszarze badawczym.

This paper concerns the results of dehydrogenation of gomphrenin pigments isolated from the purple flowers of Gomphrena globosa L. depending on pH and Cu2+ activity. Betalains are the plant dyes divided into red-violet betacyanins and yellow-orange betaxanthins. These compounds show prohealth properties proven by numerous studies [1]. Isolation of gomphrenins from the plant material was performed in the following stages: preparative extraction, filtration on a silica layer, preliminary purification and concentration on a bed of strong anion exchanger, preparative chromatographic fractionation and sample liofilization. The dehydrogenation studies of gomphrenin I and II were carried out in aqueous solutions (pH 3, 5 and 7) catalyzed by Cu2+ ions addition. The resulting product mixtures of dehydrogenated and decarboxylated gomphrenin derivatives were monitored by high-performance liquid chromatography with diode-array and mass spectrometric detection (LC-DAD-ESI-MS). Based on the results, it can be stated that decarboxylated and dehydrogenated gomphrenin II derivatives are more stable than gomphrenin I derivatives, presumably due to acylation of the glucose moiety. The highest levels of gomphrenin I derivatives were observed at pH 7, however, they were mainly monodecarboxylated ones. At pH 3, significant fraction was represented by tridecarboxylated and dehydrogenated derivatives. The lowest yield of the degradation products was indicated at pH 5.

In this report, the influence of ammonia on dehydrogenation of gomphrenin pigment, isolated from purple inflorescences of Gomphrena globosa L., in selected organic solvents (methanol, ethanol, acetone and acetonitrile) at 40°C is presented. Betacyanins are water-soluble, vacuolar plant pigments. Due to their numerous pro-health properties, they can be applied in the pharmaceutical industry and as food colorants. However, as most of natural products, they are less stable than synthetic dyes, therefore, further studies need to be carried out on their stability, but also on determination of their degradation products as well as degradation mechanisms. For the experiments of the ammonia treatment, prolonged isolation of gomphrenin from the plant material including extraction, preliminary purification, preparative HPLC fractionation, concentration and liofilization were performed. The tested pigment dissolved in organic solvents was treated by ammonia and the samples for analysis were taken during and after reaction. For identification of obtained derivatives of gomphrenin, high-performance liquid chromatography with diode-array and mass spectrometric detection (LC-DAD-ESI-MS) was performed. As a result of heat processing, the mixture of decarboxylated and dehydrogenated gomphrenin derivatives was detected. Based on the experimental results, it can be stated that gomphrenin is characterized by the greatest stability in methanolic solutions while it is the most labile in the acetonitrilic ones.
