Ganoderma sinense, a well-known medicinal macrofungus of Basidiomycetes, is widely used in traditional medicine for promoting health and longevity in East Asia. The fruiting bodies of G. sinense contain polysaccharides, ergosterol, and coumarin, which have antitumor, antioxidant, and anticytopenia activities. Mushroom cultivation requires suitable conditions for the formation of fruiting bodies and yield. However, little is known about the optimal culture conditions for mycelial growth and cultivation of G. sinense. In this study, the successful cultivation of a G. sinense strain collected from the wild was reported. The optimal culture conditions were identified by examining one factor at a time. The results of this study revealed that the nutritional requirements for the optimal mycelial growth of G. sinense were fructose (15 g/l) as the carbon source and yeast extract (1 g/l) as the nitrogen source. The optimal pH and temperature for G. sinense were 7 and 25–30EC, respectively. The mycelia grew fastest in treatment II (69% rice grains + 30% sawdust + 1% calcium carbonate). G. sinense produced fruiting bodies under all tested conditions and showed the highest biological efficiency (2.95%) in treatment B (96% sawdust, 1% wheat bran, 1% lime). In summary, under optimal culture conditions, G. sinense strain GA21 showed satisfactory yield and a high potential for commercial cultivation.

Opis:
Humans have used mushrooms from the beginning of their history. However, during the last few decades, the market demand for these fruiting bodies has increased significantly owing to the spread in the capabilities of culinary and pharmacological exploitation. Natural mushroom resources have become insufficient to meet the support needs. Therefore, traditional methods of extensive cultivation as well as modern technologies have been exploited to develop effective growing recommendations for dozens of economically important mushroom species. Mushrooms can decompose a wide range of organic materials, including organic waste. They play a fundamental role in nutrient cycling and exchange in the environment. The challenge is a proper substrate composition, including bio-fortified essential elements, and the application of growing conditions to enable a continuous supply of fruiting bodies of market quality and stabilized chemical composition. Many mushroom species are used for food preparation. Moreover, they are treated as functional foods, because they have health benefits beyond their nutritional value, and are used as natural medicines in many countries. Owing to the rapid development of mushroom farming, we reviewed the growing technologies used worldwide for mushroom species developed for food, processing, and pharmacological industries.

Opis:
Ganoderma (Basidiomycota, Polyporales) is a cosmopolitan genus with the greatest diversity in the tropics. It causes white rot of a wide range of woody plants all over the world. In Europe, 7 species of the genus Ganoderma grow: Ganoderma adspersum, G. applanatum, G. carnosum, G. lucidum, G. pfeifferi, G. resinaceum and G. valesiacum. All of them also occur in Slovakia. The aim of the present study was to identify the presence of Ganoderma species in the urban areas of Slovakia and to find out their host preferences and topology of the basidiocarps on the colonised trees. A total 263 findings of Ganoderma obtained by own field research and processing of records from herbarium items located in natural museums in Bratislava (Slovakia) and Prague (Czech Republic) were analysed to detect their ecological characteristics. The occurrence of all 7 Ganoderma species was  recorded in the urban areas of Slovakia. The most common species is G. adspersum (39% of all records) with the widest range of host woody plants (24 species). G. applanatum is second most widespread species in the urban areas of Slovakia growing mostly on a decomposing wood of unknown trees (31% of all findings) and also on beeches (9% of all findings). G. resinaceum is third most widespread in the urban areas and it grew mainly on Quercus. G. lucidum was most often found on unknown trees (65% of all findings) and also on oaks (16% of all findings). The rarest Ganoderma species are G. valesiacum (only 1 record on Larix sp.), G. pfeifferi (4 records, mainly on Fagus sylvatica) and G. carnosum (4% of all records, mainly on a decomposing wood of unknown trees).
Lakownica – Ganoderma P. Karst. (Basidiomycota, Polyporales) to kosmopolityczny rodzaj o największej różnorodności w tropikach. Lakownice powodują białe zgnilizny wielu różnych drzewostanów na całym świecie. W Europie, w tym także na Słowacji, występuje siedem gatunków lakownic: Ganoderma adspersum (l. europejska), G. applanatum (l. spłaszczona), G. carnosum (l. brązowoczarna), G. lucidum (l. zółtawa), G. pfeifferi (l. czerwonawa), G. resinaceum (l. jasnomiąższowa) oraz G. valesiacum (l. walezyjska). Celem niniejszych badań było zidentyfikowanie obecności poszczególnych gatunków lakownic w zurbanizowanym środowisku Słowacji oraz określenie ich preferencji żywicielskich, a także topologii bazydiokarpów na skolonizowanych drzewach. W badaniu wykorzystano 263 okazy lakownic zebranych w trakcie badań terenowych oraz oznaczonych rekordów, pochodzących ze zbiorów zielnikowych z muzeów przyrodniczych w Bratysławie (Słowacja) oraz w Pradze (Czechy). W zurbanizowanym środowisku Słowacji odnotowano występowanie wszystkich 7 gatunków lakownic. Najczęściej notowywanym gatunkiem jest G. adspersum (39% wszystkich rekordów), z najszerszym spektrum drzew żywicielskich (24 gatunki). G. applanatum jest drugą najbardziej rozpowszechnioną lakownicą w zurbanizowanycm środowisku na Słowacji. Rośnie głównie na rozkładającym się drewnie nieidentyfikowalnych gatunków drzew (31% wszystkich rekordów), a także na bukach (9% wszystkich rekordów). G. resinaceum jest trzecim najbardziej rozpowszechnionym gatunkiem i rośnie głównie na dębach. Natomiast G. lucidum zazwyczaj występowała na nieidentyfikowalnym drewnie (65% wszystkich rekordów) oraz na dębach (16% wszystkich rekordów). Najrzadsze lakownice to: G. valesiacum (tylko jedno znalezisko na Larix sp.), G. pfeifferi (4 rekordy, głównie na Fagus sylvatica) oraz G. carnosum (4% wszystkich rekordów, zwłaszcza na nieidentyfikowalnym gatunkowo rozkładającym się drewnie).

Opis:
Black alder is an important forest−forming species in Poland. Its wood is decomposed by many species of fungi. Ganoderma lucidum (Curtis) P. Karst. is a species of Basidiomycetes which belongs to family Ganodermataceae, order Polyporales. This fungus causes white rot decay of wood in dead and sometimes living alder trees. G. lucidum has been under partial protection in Poland since 2014. It is also red−listed as a rare species (category R – taxa with small populations) on the ‘Red list of the macrofungi in Poland’. The fungus was cut out with a sizeable fragment of wood from the alder stump in Łuków Forest District (eastern Poland) and transported to the laboratory of the Department of Mycology and Forest Phytopathology, Warsaw University of Life Sciences−SGGW. Pure culture of G. lucidum mycelium was obtained from a fragment of fruitbody. The aim of this study was to investigate, through laboratory decay tests, the ability of G. lucidum to degrade alder wood. Sterilization of wood samples (30×20×20 mm) consisted of placing the material in an accelerator and irradiating it with high−energy electrons at a dose of 30 kGy at the Institute of Nuclear Chemistry and Technology in Warsaw. On the next day, wood samples were put into 200 ml flasks with mycelium of G. lucidum on agar−wort medium (2 samples per flask). The flasks were placed in a Heraeus BK 600 incubator for 180 days, with a constant temperature of 22°C and humidity of 80 ±5%. Every 30 days 10 flasks were randomly selected, i.e. 20 samples of alder wood from this experiment. After each incubation period the samples were removed from the flasks, cleaned to remove mycelia and dried at a temperature of 105°C until they reached constant weight. By comparing mass of the samples at the start and the end of experiment in an absolutely dry state, the relative wood mass loss was calculated. After exposure times 180 days, the relative weight loss of alder wood as a result of decay by G. lucidum mycelium was on average 10.88%. The rate of the decay was almost constant during the study period. On some alder wood samples structures resembling fruitbodies of G. lucidum were noticed.

Demand for Ganoderma lucidum basidiocarps on the global market is increased due to its numerous health benefits. As they are rare in nature and traditional cultivation on logs is not ecologically and economically justified method, current trend is finding of good alternative substrate for production. Whether wheat straw, the most abundant crop residue in Europe, could be a novel substrate for G. lucidum cultivation was the question which led to the definition of the goals. Two wild and one commercial strains were objects of the study. Despite some morphological differences among basidiocarps, all strains belong to G. lucidum sensu stricto, which was confirmed by analyses of ITS, tef1-a and rpb2 gene sequences. Wheat straw showed as a good substrate, namely the periods required for the complete colonisation of wheat straw by mycelium as well as the formation of primordia and basidiocarps were relatively short. The totally fresh and dry yield and biological efficiency were also significant. If it is taken into consideration that untreated wheat straw was used, validities of its introduction in industrial-scale G. lucidum cultivation can be demonstrated.

Species of the genus Ganoderma are an example of some of the most thoroughly studied representatives of Basidiomycota both in terms of chemical composition and biological activity. Among the compounds found in this kind, the therapeutic effect is primarily associated with the polysaccharides that are heteroglycans or β-D-glucans and terpenoids represented mainly by triterpenes. Triterpene compounds have a structure composed of 30 carbon atoms, usually forming a system of five six-membered rings. Characteristic of these structures are functional groups (hydroxyl, carboxyl or ketone) and double bonds. Mycochemical studies have led to the isolation of numerous triterpenes of the lanostane type (ganoderic acids, aldehydes, alcohols, esters), lucidenic acids and others from various species of the Ganoderma genus. The broad spectrum of biological activity determined by triterpene compounds includes anti- tumor, anti-inflammatory, antioxidant, hepatoprotective, antidiabetic, and antiviral effects. This work describes biologically active triterpenes in selected species of the genus Ganoderma: Ganoderma lucidum, Ganoderma applanatum, Ganoderma adspersum, Ganoderma pfeifferi, Ganoderma colossum and others. These species are sources of natural compounds valued for thousands of years in the traditional medicine of the Far East, while ongoing research has confirmed their medicinal properties nowadays.

Timber is one of the longest-used building materials for various types of structures, and has been used in the construction of both historical and modern structures. Timer decay is caused by primarily enzymatic activities of microorganisms. The eco-friendly management of timber degrading fungi is tried by using plant extracts, oils and gels. In most of the fungi 25% methanolic extract was more effective than 5 and 10% concentrations. Lenzites sterioides was completely inhibited by 5% leaf extract of P. Juliflora and 10% leaf extracts of Prosopis, Cymbopogon and Datura at 25% concentration. Oils and gels of Cymbopogon citrates, Anacardium occidentale L., Gossypium barbadensis L., Linum usitatissimum L., Aloe vera L., and Aloe ferox Mill. were used to control the timber degrading fungi. Out of four oils cashew nut shell oil was most effective followed by cotton seed oil. Of the two Aloe gels tried the A. ferox gel showed better results than A. vera. For the first time the biocontrol of L. sterioides T. pini and S. commune, by botanical pesticides was reported. For the first time the biocontrol of L. sterioides T. pini S. commune, G. lucidum, and S. hirsutum by Oils and gels was reported.

The evaluation of glycosyl composition is an essential step to guide future research designs applied in bioactivity. In the same way, the unexplored potential bioactivity of exopolysaccharide from Ganoderma lucidum is huge. Therefore, this study investigated the glycosyl composition of the exopolysaccharide isolated from submerged fermentation of G. lucidum to serve as guide for future studies on bioactivity. Glycosyl content and composition were evaluated by combined GC/ MS of the TMS derivatives of the monosaccharide methyl glycosides produced from the sample by acidic methanolysis. Glycosyl composition analysis showed that the dominant carbohydrate component in all samples of exopolysaccharide isolated from submerged fermentation of G. lucidum CG 144 was glucose (58.1%), mannose (26.6%) and galactose (12.5%) which can be referred to as heteroglycan. These results suggest that this Ganoderma exopolysaccharide may be a new immunomodulatory agent.

Celem pracy było opracowanie metody pozyskiwania i oczyszczania zewnątrzkomórkowych polisacharydów ze szczepu Ganoderma applanatum z wykorzystaniem metod chromatograficznych. Surowy preparat polisacharydu poddawano analizie chromatograficznej z wykorzystaniem nośników Sepharose 6B lub Sepharose CI-4B uzyskując trzy symetryczne piki elucji. Frakcje polisacharydowe nie wykazywały absorbancji przy długości fali 280 i 260 nm co świadczy o braku białek i kwasów nukleinowych w badanych substancjach.
The aim of this work was to study the production of exopolysaccharides from Ganoderma applanatum and the description of chromatographic methoc purification of fungal polysaccharides. Crude polysaccharides were fractionated using preparative Sepharose 6B or Sepharose CI-4B chromatography obtain three fractions, which were selected on the total carbohydrate elution profile. Polysaccharides fractions had no absorbance at 280 and 260 nm UV spectrum, indicating the absence of proteins and nucleic acids.

Opis:
Quantitative determination of polysaccharides in Ganoderma lucidum fruit bodies from different sawdust cultivation substrates and their antibacterial activity was done. Thirty six samples were analyzed. Four strains of Ganoderma lucidum (GL01, GL02, GL03 and GL04) were cultivated on the growth substrates of three different sawdust types: birch (Bo), maple (Kl) or alder (Ol) amended with wheat bran in three different concentrations: 10, 20 and 30% (w/w). Even though the richest in polysaccharides was GL01 strain, the highest yields of the polysaccharides were determined in GL04Kl3 sample and was 112.82 mg/g of dry weight. The antibacterial activity of polysaccharides was determined in vitro using micro-dilution broth method. The panel of eight reference bacterial strains was used. All the polysaccharide samples tested showed the broad spectrum and the moderate antibacterial activity. Micrococcus luteus ATCC 10240 strain was the most sensitive with MIC (minimal inhibitory concentration) = 0.63 − 1.25 mg/mL.

Opis:
Mushrooms are cultivated on a wide range of materials of organic origin. Textile industry wastes seem to be interesting in this regard. In experiment, mycelium growth of the following eight mushroom species: Pholiota nameko (Ito) Ito et Imai, Flammulina velutipes (Curt. ex Fr.) Sing., Lyophyllum ulmarium (Bull. ex Fr.) Kumm., Marasmius oreades (Bolt.: Fr.) Fr., Hericium erinaceus (Bull: Fr.) Pers., Agrocybe aegerita (Brig.) Sing., Lentinula edodes (Berk.) Pegl. and Ganoderma lucidum (Curt: Fr.) Karst. cultivated on alder sawdust, rye straw, hemp and flax shive substrates was investigated. Significant variability in mycelium growth was observed depending on the mushroom species and the type of the applied substrate. The best mycelium growth of the examined mushroom species was recorded on flax shive and alder sawdust. In the majority of the experimental mushroom species, the worst growth of mycelia was recorded on the hemp shive substrate.
W uprawie grzybów wykorzystywane są różnego rodzaju materiały pochodzenia organicznego. Odpady przemysłu włókienniczego wydają się pod tym względem interesujące. Badano wzrost grzybni ośmiu gatunków grzybów tj. Pholiota nameko (Ito) Ito et Imai, Flammulina velutipes (Curt. ex Fr.) Sing., Lyophyllum ulmarium (Bull. ex Fr.) Kumm., Marasmius oreades (Bolt.: Fr.) Fr., Hericium erinaceus (Bull: Fr.) Pers., Agrocybe aegerita (Brig.) Sing., Lentinula edodes (Berk.) Pegl. i Ganoderma lucidum (Curt: Fr.) Karst. na podłożach z trocin olszowych, słomy żytniej oraz paździerzy konopnych i lnianych. Stwierdzono duże zróżnicowanie wzrostu grzybni w zależności od gatunku grzyba i rodzaju podłoża. Najlepszy wzrost grzybni badanych gatunków grzybów zanotowano na podłożu z paździerzy lnianych i trocin olszy. Na podłożu z paździerzy konopnych grzybnia rosła najsłabiej u większości badanych gatunków grzybów.

According to a recent study, Ganoderma may be the third genus, after Alternaria and Cladosporium, whose spores cause symptoms of allergy and whose levels are directly related to meteorological factors. There are only few articles from different parts of the world about the relationships between Ganoderma spore count and meteorological factors. The aim of the study was to review all available publications about airborne Ganoderma spores and to compare the results in a short useful form.

Opis:
The effect of Ganoderma lucidum extract on glucose uptake was studied in L6 rat skeletal muscle cells. G. lucidum extract increased glucose uptake about 2-fold compared to control. The extract stimulated the activity of phosphatidylinositol (PI) 3-kinase which is a major regulatory molecule in the glucose uptake pathway. About 7-fold increased activity of a PI 3-kinase was observed after treatment with G. lucidum extract, whereas PI 3-kinase inhibitor, LY294002, blocked the G. lucidum extract-stimulated PI 3-kinase activity in L6 skeletal muscle cells. Protein kinase B, a downstream mediator of PI 3-kinase, was also activated by G. lucidum extract. We then assessed the activity of AMP-activated protein kinase (AMPK), another regulatory molecule in the glucose uptake pathway. G. lucidum extract increased the phosphorylation level of both AMPK α1 and α2. Activity of p38 MAPK, a downstream mediator of AMPK, was also increased by G. lucidum extract. Taken together, these results suggest that G. lucidum extract may stimulate glucose uptake, through both PI 3-kinase and AMPK in L6 skeletal muscle cells thereby contributing to glucose homeostasis.
