Concentration of glycogen, maltose, and trehalose as well as the activity of hydrolases catabolising them: «-amylases, glucoamylase, maltase, and trehalase were studied during a 48 h infection of Galleria mellonella (Lepidoptera) instar larvae infected with infective Heterorhabditis zealandica (20 nematodes/insect). The content of trehalose in the infected insects during the first day of infection was higher (p < 0.05) than in the controls. The concentration of maltose and glycogen were similar in both groups despite the fact that the activity of all examined o-glycosidases was generally much higher in the infected insects than in the controls.

Cancers are the leading cause of deaths all over the world. Available anticancer agents used in clinics exhibit low therapeutic index and usually high toxicity. Wide spreading drug resistance of cancer cells induce a demanding need to search for new drug targets. Currently, many on-going studies on novel compounds with potent anticancer activity, high selectivity as well as new modes of action are conducted. In this work, we describe in details three enzyme groups, which are at present of extensive interest to medical researchers and pharmaceutical companies. These include receptor tyrosine kinases (e.g. EGFR enzymes) and non-receptor tyrosine kinases (Src enzymes), type A, B and C Aurora kinases and aminopeptidases, especially leucine aminopeptidase. We discuss classification of these enzymes, biochemistry as well as their role in the cell cycle under normal conditions and during cancerogenesis. Further on, the work describes enzyme inhibitors that are under in vitro, preclinical, clinical studies as well as drugs available on the market. Both, chemical structures of discovered inhibitors and the role of chemical moieties in novel drug design are discussed. Described enzymes play essential role in cell cycle, especially in mitosis (Aurora kinases), cell differentiation, growth and apoptosis (tyrosine kinases) as well as G1/S transition (leucine aminopeptidase). In cancer cells, they are overexpressed and only their inhibition may stop tumor progression. This review presents the clinical outcomes of selected inhibitors and argues the safety of drug usage in human volunteers. Clinical studies of EGFR and Src kinase inhibitors in different tumors clearly show the need for molecular selection of patients (to those with mutations in genes coding EGFR and Src) to achieve positive clinical response. Current data indicates the great necessity for new anticancer treatment and actions to limit off-target activity.
Nowotwory stanowią jedną z głównych przyczyn zgonów na świecie. Dostępne w lecznictwie substancje przeciwnowotworowe charakteryzują się niskim indeksem terapeutycznym jak i wysoką toksycznością. Rozwijająca się oporność komórek nowotworowych na dostępne w terapii leki przyczynia się do konieczności poszukiwania nowych punktów uchwytu/miejsc docelowych (z ang. targets) dla potencjalnych substancji przeciwnowotworowych. Obecnie prowadzonych jest również wiele prac nad nowymi związkami przeciwnowotworowymi o wysokim potencjale terapeutycznym, nowym mechanizmie działania i/bądź wyższym indeksie selektywności. W pracy, autorzy skupili uwagę na trzech grupach enzymów, będących obecnie w obszarze zainteresowań współczesnej medycyny. Omówione zostały kinazy tyrozynowe na przykładzie enzymów EGFR i Src, kinazy Aurora typu A, B i C, a także aminopeptydazy na przykładzie aminopeptydazy leucynowej. Scharakteryzowano klasyfikację enzymów, ich rolę w cyklu komórkowym w warunkach fizjologicznych i procesie nowotworowym. Opisano również inhibitory enzymów, substancje będące w trakcie badań in vitro, przedklinicznych i klinicznych jak i leki wprowadzone na rynek farmaceutyczny. Zwrócono uwagę na budowę chemiczną inhibitorów enzymów i tym samym na kierunek poszukiwań nowych leków przeciwnowotworowych. Omówione enzymy w warunkach fizjologicznych odgrywają ważną rolę w cyklu komórkowym, zwłaszcza na etapie podziału mitotycznego. Jednakże w procesie nowotworowym dochodzi do ich nadekspresji. Zjawisko to można zahamować poprzez inhibicję aktywności enzymu. Autorzy omówili wpływ inhibitorów kinaz tyrozynowych, kinaz Aurora czy aminopeptydaz leucynowych na cykl komórkowy i bezpieczeństwo stosowania tych potencjalnych leków u ludzi. Dotychczasowe badania przedkliniczne i kliniczne inhibitorów kinazy tyrozynowej typu EGFR czy Src potwierdziły konieczność selekcji pacjentów, na tych z mutacją w genie kodującym dany enzym. Badania prowadzone na wybranej grupie chorych przynosiły oczekiwany pozytywny wynik. Wiele aspektów dotyczących nowych punktów uchwytu w terapii przeciwnowotworowej wciąż wymaga dalszych prac, aczkolwiek daje również nadzieję na odkrycie skutecznych i selektywnych leków.

The increasing interest in enzymatic growth promoters prompted this investigation of the effect of an enzymatic stimulating complex on selected hematological and serum biochemical parameters in boars. The enzymatic complex [five proteases (proteinases-endopeptidases) and two peptidases (exopeptidases) obtained by fermentation from Streptomyces fradiae] was added to diets for three months, at initial doses of 90 (group El) and 120 (group E2) g/ton feed for the first seven days followed by 40 and 60 g/ton, respectively, in the subsequent weeks. The evaluation was based on selected hematological (RBC, WBC, HCT (Ht), HGB (Hb), MCV, MCH, and MCHC) and biochemical (AST, ALT, AP, Ca, Mg, urea, cholesterol, creatinine, inorganic phosphorus, and oxyglucose) parameters determined before and after the experimental period. The reported values were within physiological norms. Statistical differences were found between experimental groups with respect to WBC, HCT (Ht), MCV, MCH, AP, Ca, Mg, creatinine, and inorganic phosphorus. The study results indicate that the complex of proteolytic enzymes administered to boars caused no negative changes in their metabolic profiles. The statistically different hematological and serum biochemical parameters, while within normal limits, point to an early stage of microcytic anemia and heightened agitation that could be the result of intensified nitrogen metabolism.

Genetic variability of a selected population of Dreissena polymorpha, an invasive species playing a significant role in aquatic ecosystems, was studied. Starch gel electrophoresis was used to analyse 8 enzymatic loci in 200 individuals collected from 20 sites in a lake. The population was found to have 75.0% of polymorphic loci, 2.8 alleles per locus, 3.5 alleles per polymorphic locus, 0.393 coefficient of expected heterozygosity, and 149 genotypes. Zebra mussel clumps were strongly polymorphic; almost every individual had a different genotype. The high polymorphism observed in the D. polymorpha clumps had most likely resulted from external cross-fertilisation and the presence of free-swimming veliger larvae as well as from a considerable heterozygosity of individual bivalves. Genetic variability of the population studied was found to be similar to that of populations inhabiting other Western Pomeranian lakes, including both highly polluted ones and those formed as recently as about 40 years ago. This provides evidence for a mass colonisation of freshwater reservoirs effected by very polymorphic parent populations of D. polymorpha. The literature data on North American zebra mussel populations which invaded that continent about 10 years ago show them to be polymorphic, too, but not as much as European ones.

Enzyme activities in soils are influenced by soil characteristics and pollutants and could be indicator of soil quality. This study was undertaken to determine the relationship between physicochemical characteristics, heavy metals contents and enzyme activities (EA) in agricultural soils in the Saiss plain (Morocco). The analysis of six agricultural soil samples (sites 1–6), collected from urban and periurban agricultural soils of Fez region, showed that soils are generally alkaline with high CaCO3 that rich 46%, EC (525–703 μS/cm), rich in organic matter (3.14–5.80%). The Cr, Cu, Ni, Pb and Zn concentrations in the studied area are generally greater than the Upper Continental Crust, with a deceasing order: Zn> Cr> Pb> Cu> Ni. Geo-accumulation index showed that soils are unpolluted to moderately polluted except site 5 and site 6 that are moderately to strongly polluted by respectively Cu, Pb, Zn and Pb. Potential ecological rіsk factors were below 40, which means low potential ecological rіsk except site 5 that has moderate potential ecological risk by Cu and Pb. Soil potential ecological risk indices were found <150 indicating low ecological risk. According to this index, Cu caused more serious pollution than the other elements. The enzyme actіvities of the sіx soil samples showed almost the similar ranges of values. These EA showed the highest values in site 5.The sensіtivity of soil enzyme to heavy metals were observed UREA>PHOS>GALA. Pearson’s correlation showed significant posіtive correlatіon between studied soil EA and between these EA and heavy metals (Cu, Pb and Zn), and significant negative correlatіon between EA and heavy metals (Cr and Ni). The results of this study enrich and provide data base of the impact of heavy metals on soil enzyme actіvities in agrіcultural soils in the Saiss plain.

Hydroxyapatite is used as a matrix for immobilization of protease from Aspergillus oryzae by a process of adsorption. The matrix obtained has the surface area of 26 m2/g and particles in the shape of flakes of diameters no greater than 650 nm. The efficiency of the proposed method was confirmed by the Fourier transform infrared spectroscopy, elemental analysis and by analysis of parameters of the pore structure of matrix and products after immobilization. On the basis of the Bradford method it was found that the greatest amount of enzyme (132 mg/g) was immobilized from a solution of initial enzyme concentration of 7 mg/cm3 after 24 h of the process.

Angiotensin I-converting enzyme (ACE), which plays an important role in blood pressure regulation, and methylenetetrahydrofolate reductase (MTHFR) involved in homocysteine metabolism belong to a large group of polypeptides which may be potential risk factors for atherosclerosis and coronary artery disease (CAD). To assess whether polymorphisms of the genes encoding these peptides are associated with CAD in Silesian we conducted a study among 68 individuals suffering from CAD (including 52 cases after myocardial infarction), 51 subjects with positive family history of CAD and 111 controls. We analysed the distribution of genotypes and allele frequencies of the insertion/deletion (I/D) polymorphism in the ACE gene using PCR amplification, and the C677→T polymorphism in the MTHFR gene using PCR-RFLP analysis. We found that D allele frequency was significantly higher in CAD patients (61%) than in controls (43%) (P = 0.001, OR = 2.06). The D allele carriers (DD + ID genotypes) were more frequent in the CAD patients (85%) compared to control group (65%) (P = 0.003, OR = 3.14), whereas the familial CAD risk group shows the highest frequency of the ID genotype (57% vs 43% in controls). In contrast, the MTHFR polymorphism does not seem to be associated with the disease. Our data indicate that in Silesian CAD patients the disease is strongly associated with carrier-state of the ACE D allele, but not with the C677→T transition in the MTHFR gene.

Metale ciężkie, w tym także cynk, gdy dostaną się do gleby w nadmiernych ilościach, stanowią dość trwałe zanieczyszczenie, w przeciwieństwie do różnego rodzaju zanieczyszczeń organicznych, podatnych w większym lub mniejszym stopniu na degradację mikrobiologiczną. Mimo iż akumulacja metali ciężkich przez drobnoustroje jest procesem powszechnym, to nie do końca poznanym. Granice tolerancji nie są jednakowe i zależą od ich bioróżnorodności i czasu trwania imisji, a także właściwości fizykochemicznych gleby, jej struktury, zawartości materii organicznej, pH i pojemności kompleksu sorpcyjnego. Coraz częściej do oceny stanu zanieczyszczenia gleby wykorzystuje się metody biologiczne, w tym aktywność enzymów glebowych, i w nich należy upatrywać szybkiej diagnozy jakości gleb. Wskaźniki te mogą być jednak wykorzystywane tylko do oceny gleb zanieczyszczonych metalami ciężkimi, a są mało przydatne w ocenie jakości gleb zanieczyszczonych związkami organicznymi.
Heavy metals, e.g. zinc, present in soil in excessive quantities become quite persistent contaminants. Thus, they differ from organic pollutants, which can be degraded, to a various degree, by microorganisms. Although the accumulation of heavy metals by microorganisms is widespread, this phenomenon has not been thoroughly investigated. Tolerance limits are not uniform and depend on the biodiversity and duration of ambient concentration, as well as on the physicochemical properties of soil, soil structure, organic matter content, pH and sorptive complex capacity. Soil contamination is more and more frequently assessed with biological methods, by testing for example the activity of soil enzymes. Such assays, however, can be used to evaluate the quality of soils contaminated with heavy metals, but are less useful for testing soils polluted with organic compounds.

This research aims to investigate the effect of adding potassium diformate to commercial feed on the increase of absolute growth rate and the activity of protease enzyme. The research was conducted from July to October 2019 in the Aquaculture Laboratory Faculty of Fisheries and Marine Sciences, Padjadjaran University. The method applied in this research was an experimental method using a Completely Randomized Design (CRD), which consists of four treatments and four replications. The treatments were: (A) without addition of Potassium diformate (control), (B) addition of Potassium diformate by 0.3%, (C) addition of Potassium diformate by 0.5%, and (D) addition Potassium diformate by 0.8%. The test fish were 300 giant gouramis with 4-6 cm in length. The containers used in this research were 16 rearing aquaria with a size of 40 30 40 cm3. The density of studied giant gourami fingerlings was 10 fish per aquarium. The rearing period was 40 days. The feeding rate was 3% from biomass. Water quality parameters (temperature, pH, and dissolved oxygen), the absolute growth rate, feed conversation ratio and survival rate were observed every 10 days. The protease enzyme activities were observed at the end of the research. Data on the absolute growth rate, feeding conversion ratio, the characteristics of protease enzyme and survival rate were analyzed using the Analysis of Variance (ANNOVA) continued with Duncan’s Multiple Range Test at the level of 95%, while the water quality was analyzed descriptively. The results show that the addition of potassium diformate by 0.3% gave the best result with the absolute growth rate of 1.50%, feed conversion ratio of 2.70, protease enzyme activity by 634.2 μ/mL and survival rate of 100%.

Opis:
The effects of externally applied sodium nitroprusside on superoxide dismutase (SOD), glutathione peroxidase (GPx), ascorbate peroxidase (APX), and catalase (CAT) antioxidant enzyme activities, nitric oxide (NO) levels have been investigated in salt stress resistant and sensitive sunflower plants. NaCl treatments and SNP treatments simultaneously with salt application (NaCl+SNP) were performed. SOD, GPx, APX and CAT antioxidant enzyme activities and NO levels showed differences in leaf tissues treated with 100 µM SNP, different concentrations of NaCl, and NaCl+SNP. SOD, GPx and APX enzyme activities were generally increased in sensitive variety, but decreased in tolerant variety. However, while general increase in CAT enzyme activity was determined in tolerant type, a reduction was established in sensitive type. An increase was determined in both types in NO levels. It is evident from these results that administration of NO donor SNP can cope with reactive oxygen species in both varieties. This study indicates that negative effects of salt stress on different sun flower varieties can be recovered by nitric oxide.
