The aim of the following work was a preliminary assessment of the possibility of producing a topical preparation containing dihydroxyacetone (DHA) and amino acids, encapsulated separately in polysaccharide capsules. The colour reaction between the DHA and amino acids would be possible after mixing of the two components triggered with mechanical disruption of the capsules on application. Such preparation could be used in patients suffering from vitiligo, by masking the symptoms of the disease. Several capsule compositions were evaluated in terms of appearance, dimensions and mechanical parameters. In addition, the reactions of various amino acids with DHA were carried out and obtained colours were analysed. The results indicate that formation of the dual-phase formulation of encapsulated DHA and amino acids is possible. It was also found that certain amino acids vary in colour resulting from reaction with DHA. This feature can be potentially utilized to better design the colour resulting from application of the prepared composition.

New analogues of deltorphin I (DT I), in which the phenylalanine residue in position 3 is substituted with amphiphilic α,α-disubstituted amino acid enantiomers, (R) and (S)-α-hydroxymethylnaphtylalanine, were synthesized and tested for μ and δ opioid receptor affinity and selectivity. Although both analogues have lower affinity to δ receptors than DT I, they both expressed specificity to δreceptors.

Opis:
The polymerization of amino acids under anhydrous prebiotic conditions was first studied several decades ago. Here we use a stochastic model stressing the relevant role of the polarity of amino acids in the formation of oligopeptides in a prebiotic milieu. Our goal is to outline the predominance of co-polypeptides over homo-polypeptides, resulting not only from the randomness, but also from polarity properties of amino acids. Our results conclude that there was a higher probability of the formation of co-polypeptides than of homo-polymers. Besides, we may hypothesize that the former would have a more ample spectrum of possible chemical functions than homo-polypeptides.

Introduction and objective: Back pain are the most common ailment within the human locomotor system. Because of their prevalence, they were classified as diseases of civilization. The aim of the study was to attempt to assess the concentration of selected amino acids in plasma and correlating the results of laboratory tests with the occurrence of backaches. Material and method: The study group included 188 patients presenting for CT scan administering as the cause of their symptoms low back pains. All of these patients gave the blood samples from which the concentration of free amino acids was estimated by ion exchange chromatography using an automated amino acid analyzer AAA 400 from INGOS Praha. The control group consisted of patients who underwent testing using computed tomography and there were no primary or secondary changes associated with degeneration in the lumbar spine. Results: Patients on the basis of research carried out by computed tomography were divided into five groups according to disease entity. Analyzing the average concentration of essential amino acids in the blood plasma of patients of each group, it was found that it is higher in the case of lysine for each considered disease entity. The average concentration of methionine in all disease entities does not deviate from the average values in the control group. Analysis of the average concentration of selected essential amino acids revealed that in the case presented disorders underwent their level of variation. Average concentrations of selected amino acids have proven to be very similar in both groups. Slightly higher values proved to be in the control group for proline and lysine. Conclusions: The concentration of amino acids varies with the severity of degenerative changes in the connections as well as in interbody joints. The highest increase in the concentrations of all tested amino acids are present in root bands. Decrease in the concentrations of all tested amino acids appears in cancer.

In view of the predicted increase in sulphur deficiency in crop production, a study was undertaken to assess the effect of fertilization with this nutrient on criteria determining the biological value of narrow-leafed lupin seeds (total and true protein, their ratio, nitrates and amino acid composition). The study was based on a three-year, three-factor field experiment, in which the Elf cultivar of narrow-leafed lupin (Lupine Angustifolius L.) was grown on light soil with low sulphur content. The experiment included foliar and soil application of sulphur and its elemental and ionic forms. The application doses were 0, 20, 40 and 60 kg S ha-1. In all years of the study, the sulphur application dose was found to significantly determine the total and true protein content as well as their ratios in the lupin seeds. Application of just 20 kg S ha-1 caused a significant increase in true protein in the lupin seeds and in the proportion of true protein in the total protein, as compared to the control. The CS index for all the essential amino acids showed that the first limiting amino acid in the lupin seed protein was sulphur-containing methionine. As the dose of applied sulphur increased, there was an overall increase in CSmeth. and EAAI. Given the positive effect of sulphur, particularly in the elemental form, on the protein content and amino acid composition of lupin seeds and in reducing the content of harmful nitrates, the use of this nutrient in fertilization of this species is recommended.

The work presents a characteristic of some elements of the protein conformation of albumin and globulin fractions isolated from rapeseed meal and rapeseed protein concentrate (see Part I). Investigations were based on spectrophotometry in the ultraviolet and infrared radiation, and on circular dichroism. The proportion of alpha, beta and gamma structures was determined in rapeseed and concentrate albumin and globulin fractions.
W pracy scharakteryzowano niektóre elementy konformacji białek frakcji albumin i globulin wyizolowanych z mączki i koncentratu białkowego uzyskanych z nasion rzepaku (patrz cz. I). Badania przeprowadzono przy wykorzystaniu spektrofotometrii w ultrafiolecie i podczerwieni oraz dichroizmu kołowego. W celu stwierdzenia obecności w frakcjach albumin i globulin koncentratu i nasion rzepaku kwasów nukleinowych poddano je hydrolizie kwasem mrówkowych, a następnie badaniom spektrofotometrycznym w UV. Szczególnie interesujące okazało się widmo frakcji albumin nasion rzepaku, które charakteryzowało się pasmem absorpcji przy 320 nm. Pojawienie się tego pasma można wytłumaczyć obecnością kwasów fenolowych związanych z tą frakcją. Zaobserwowany przy 260 i 280 nm wzrost absorbancji w przypadku frakcji globulin nasion rzepaku po ogrzewaniu z kwasem mrówkowym można wytłumaczyć obecnością związanych z tą frakcją kwasów nukleinowych, Analiza widm w podczerwieni frakcji albumin i globulin nasion rzepaku i koncentratu pozwoliła na wyznaczenie procentowej zawartości β struktury, która w obu frakcjach globulin wynosiła 140/o, a w albuminach odpowiednio 18 i 17%. Analiza tych widm sugerowała również obecność w badanych frakcjach składników niebiałkowych, jak stwierdzone wcześniej kwasy fenolowe i nukleinowe. Wyniki badań tych samych frakcji za pomocą dichroizmu kołowego pozwoliły na określenie procentowych zawartości poszczególnych struktur α, β i γ. Wartości te wynosiły odpowiednio dla: albumin koncentratu 37,8, 21,7 i 40,7%, globulin nasion rzepaku 57,1, 14,3 i 28,6%, globulin koncentratu 50,2, 12,1 i 37,6%.

Legumes should be considered as an important part of a healthy diet, because of both their nutritional value (high protein and low fat content) and functional properties (Trichopoulou et al. 2014). The functional components include carbohydrates, soluble fiber, vitamins and polyphenols. Epidemiological studies have shown a positive correlation between the consumption of legumes and a reduced risk of cardiovascular diseases, diabetes and certain types of cancer. In the past few years, phenolic compounds have attracted much attention owing to a large variety of biological actions, including anti-inflammatory effects, which they can produce (García-Lafuente et al. 2014). Chronic inflammation is a cause of many serious diseases, including cardiac disorders, allergy, DM 1, Alzheimer’s disease. In the anti-inflammatory food pyramid, pulses were placed at the base (Viveky et al. 2013). The aim of this study was to determine the content of amino acids, fatty acids, carbohydrates, macro- and micro-elements in the common legumes. The total protein content varied from 17.9 to 22.5 g 100 g-1. The mean content of fat varied from 1.1 to 2.0 g 100 g-1 and the soluble sugar content ranged from 4.3 to 7.9 g 100 g-1. The dominant amino acids were glutamic and aspartic acids. Methionine and cysteine composed the lowest percentage of all amino acids. Pulses were characterized by the presence of such fatty acids as a-linoleic, linoleic, oleic and palmitic acid. The mean content of iron varied from 3.3 to 5.3 g 100 g-1. The highest amount of this element was found in small bean, which was also characterized by the highest copper content. The highest amounts of zinc, calcium and magnesium were contained in dark red kidney bean. The analyzed pulses were distinguishd by high nutritive value. The identifed amino acids profile indicates sufficient quantity of lysine and shortage of sulphuric acids. The pulses also had a beneficial composition of fatty acids. The omega-6 to omega-3 ratio was between 1:2 and 4:1.

The estimation of the protein content and amino acid composition under the influence of metal nanoparticles (Mn, Cu, Zn, Ag) for seven species of aquatic macrophytes: Limnobium laevigatum (Humb. & Bonpl.ExWilld.), Pistia stratiotes L., Salvinia natans L., Elodea canadensis Michx., Najas guadelupensis (Spreng.) Magnus, Vallisneria spiralis L. and Riccia fluitans L. was conducted. The plants were exposed during 7 days on the experimental solutions of metal nanoparticles at the rate of 1 g of plant per 100 ml of the mixture of stock colloidal solutions of metal nanoparticles (Mn – 0.75 mg/l, Cu – 0.37 mg/l, Zn – 0.44 mg/l, Ag+, Ag2O – 0.75 mg/l) diluted 200 times. In the five investigated species, reduction of the protein content was observed. However, this indicator remained stable only in P. stratiotes (52 mg/ml) and, conversely, increased in V. spiralis (46 mg/ml to 51 mg/ml). The content of the studied amino acids in N. guadelupensis decreased by 46% (from 112.05 mmol/g to 60.15 mmol/g), in R. fluitans – by 44% (from 104.06 mmol/g to 58.25 mmol/g), in S. natans – by 23% (from 90.08 mmol/g to 69.59 mmol/g), in E. canadensis – by 10% (from 143.92 mmol/g to 129.4 mmol/g), and in P. stratiotes as well as in L. laevigatum – by 8% (from 210.65 mmol /g to 193.77 mmol/g and with 155.0 mmol/g to 142.60 mmol/g), but in V. spiralis, on the contrary, increased by 7% (from 91.31 mmol/g to 97.59 mmol/g). Changes in the composition and content of amino acids for each species of aquatic plant were analyzed. It was suggested that the studied plants, which belong to different families, have different defense mechanisms, according to which the amino acid composition of plants varies.

Considerable progress has been made in the past few years with industrial use of essential key intermediates for chemical and pharmaceutical industry. The increasing demand for obtaining chiral drugs in enantiomerically pure form makes it necessary to search for novel biocatalysts useful in the synthesis of amino acids, chiral amines, amino sugars and alcohols. According to the reaction mechanism, aminotransferases (ATs) have useful applications because of their capability of transfer of an amino group from a donor substrate to an acceptor, thus resulting in the synthesis of a wide variety of building blocks. This article reviews current biocatalytic approaches using microbial ATs in the synthesis of optically active products. Focus is also put on the engineering of ATs and their limitations in the industrial applications. Moreover this review covers biocatalytic approaches using ATs isolated from extreme environments.

In accordance with the second law of thermodynamics, the Universe as a whole tends to higher entropy. However, the sequence of far-from-equilibrium events that led to the emergence of life on Earth could have imposed order and complexity during the course of chemical reactions in the so-called primordial soup of life. Hence, we may expect to find characteristic profiles or biases in the prebiotic product mixtures, as for instance among the first amino acids. Seeking to shed light on this hypothesis, we have designed a high performance computer program that simulates the spontaneous formation of the amino acid monomers in closed environments. The program was designed in reference to a prebiotic scenario proposed by Sydney W. Fox. The amino acid abundances and their polarities as the two principal biases were also taken into consideration. We regarded the computational model as exhaustive since 200 000 amino acid dimers were formed by simulation, subsequently expressed in a vector and compared with the corresponding amino acid dimers that were experimentally obtained by Fox. We found a very high similarity between the experimental results and our simulations.

Pasta is a great vehicle for the incorporation of vegetable-derived ingredients to increase the consumption of the health-beneficial components originating from vegetables. Notably, by-products of vegetable processing can also serve as a rich source of phytochemicals. An important step in pasta processing is drying which can affect the content of bioactive compounds in pasta. This study aimed to evaluate the effect of drying on the nutritional quality and cooking properties of durum wheat pasta fortified with broccoli leaves. Pasta enriched with broccoli leaf powder (BLP) at 2.5% (B2.5) and 5% (B5), and control pasta without BLP (C), which differed in drying conditions: fresh pasta without drying (F), pasta dried at 50oC for 8 h (L), and pasta dried at 80oC for 3 h (H) were formulated. The obtained pasta products were analysed for the cooking properties (optimal cooking time, cooking loss, water absorption and swelling capacity); colour parameters; proximate composition; and contents of free amino acids (FAA), fatty acids and sugars. BLP significantly improved the contents of ash by up to 35 g/100 g, FAA and fatty acids to up to 1298 nmol/g dry matter (DM) and 16741 µg/g DM, respectively, without compromising the cooking quality of pasta. Drying had a significant effect on fatty acids, which content in pasta processed at the highest temperature tested decreased. From the nutritional point of view, the low-temperature drying seems to be an interesting method for pasta preparation, with the highest content of FAA, fatty acids, especially unsaturated ones, and the lowest content of sugar. However, at the same time, the dried pasta products were characterised by greater cooking loss approximating 10%.
