Opis:
Genetic analysis of two rye interline crosses and a set of wheat/rye chromosomal addition lines was performed to reveal the mechanism underlying wide variation range of alpha-amylase activity in sound grain. The long arm of chromosome 6R was found to be responsible for increased enzyme synthesis during late stages of triticale grain maturation. Only nuclear genes seemed to control alpha-amylase activity, as reciprocal crosses between rye lines showed no maternal effects. Low enzyme activity showed complete dominance over high level of its synthesis. Segregation ratios, observed in F₂ and BC₁ crosses, indicated that recessive alleles at two independent duplicative loci underlie intensive alpha-amylase production.

Opis:
Fluorescence in situ hybridization (FISH) is a very useful method for assessing chromosome rearrangements. When neither banding pattern nor clinical symptoms are sufficient to determine the origin of additional chromosomal fragment, FISH with multiple chromosome-specific libraries (chromosome painting), allows to solve this diagnostic problem rapidly. Three chromosomal additions, 7q+, 13p+ and 22q+, found in routine cytogenetic studies performed in children with phenotypic abnormalities were analysed using FISH. This technique documented the origin of the extra material to be derived from chromosome 16[der(7)t(7; 16)(q36.3;p 13.11)], 18[der(13)t(13; 18)(p12;q 12.2)] and 22[dup(22)(q11.2q13.1)], respectively. In two cases the abnormality arose de novo, while in the third case the product of translocation t(13;18) was maternal by origin. It was present in 30% of mother's lymphocytes, and in 70% of them a balanced Robertsonian translocation t(13q;15q) was found. In the presented cases the chromosome analysis with both traditional banding and chromosome painting techniques, allowed to establish final clinical diagnosis.

The paper concerns the influence of added rye chromosomes on physical properties of kernels of the addition lines T. eastivum Grana - S. cereale Dańkowskie Złote 1R", 2R", 3R", 3RS", 4R", 5R", 6R", and 6RL", as well as initial forms: wheat cv. Grana, rye cv. Dańkowskie Złote and octoploid triticale Grana × Dańkowskie Złote. Kernels from each form, were analysed for the following features: basic geometric dimensions of kernels, 100-kernel weight and kernel volume; surface roughness, index of kernel damage and mechanical strength (determined by maximum force, force within elasticity limits, maximum deformation, elastic deformation, energy causing deterioration of kernel structure). Wheat lines with added rye chromosomes differed from one another and from the initial form wheat cv. Grana in basic geometric dimensions. The lines 6R" and 6RL" had a distinctly higher 100-kernel weight and kernel volume than their initial forms. All the addition lines and octoploid triticale were characterised by markedly higher indices of external damages than the wheat cv. Grana. The mechanical strength of addition line kernels also varied considerably.

Opis:
Tetraploid rye was crossed with different tetraploid triticale lines. The F₁ generation of tetraploid rye × tetraploid triticale hybrids was backcrossed with 4x rye. After backcrossing, all BC₁-F₁ plants were subjected to open pollination, whereas in the BC₁-F₂ generations only plants with wheat chromosomes in their karyotypes were open-pollinated. Substitution, addition and addition-substitution lines of wheat chromosomes in tetraploid rye were isolated from the F₂ and F₃ of BC₁. In 60 plants of BC₁-F₂, 59 chromosomes from the A genome and 9 from the B genome of wheat were recovered. The wheat chromosomes were monosomic except for five plants which were disomic, viz. 1A and 5A in two plants each, and a translocated 3AS/5AL in one plant. In 235 BC₁-F₃ plants, 174 wheat addition and substitution chromosomes were found, 143 from the A genome and 31 from the B genome. All wheat chromosomes except 3A from the A genome and four chromosomes from the B genome - 2B, 3B, 5B and 7B were recovered. The number of substitutions ranged from one to four per plant, only two plants having four. In the group of addition plants the number of added wheat chromosomes ranged from one to two, and in the case of addition-substitution plants — from two to four. Wheat chromosomes occurred in monosomic form, except 10 plants. Six substitution plants were disomic for 1 A, 2A, 5A, 7A, 2B and 3B, respectively. One was disomic for 1A and 5A in two addition plants. Two addition-substitution plants were double disomic: 1A and 5A - in one, and 1A and 3B in the other. In the BC₁-F₃ generation, 23 different translocations were found, four of which occurred between wheat chromosomes and the remaining 19 - between wheat and rye chromosomes. Translocated chromosomes were monosomic, except four plants. Two of them were disomic for 3AS/4RL, one for 4AS/4RS and one for 7AS/7RS. The fertility of both addition and substitution plants ranged from 0 to 38.0 seeds/spike, regardless of the chromosome number, with a mean of 9.61 seeds/spike. Plants with 28 chromosomes showed singnificantly higher fertility than plants with 29 and more chromosomes, except additoion plants with chromosomes 5A and 5B. The analysis of the influence of particular wheat chromosomes on plant fertility showed that both substitution and addition plants with chromosome 6A had the highest average fertility, while plants with chromosome 2B in substitution lines as well as plants with chromosome 2A in addition and addition-substitution lines had the lowest fertility.

Opis:
Secondary isotope effects for carbonyl addition reactions of methyl thioacetate, acetone and acetaldehyde have been calculated by ab initio quantum mechanical methods in an effort to interpret measured beta-deuterium isotope effects on acetylcholinesterase- catalyzed hydrolysis of acetylthiocholine. The calculated beta-deuterium isotope effect for equilibrium addition of methanol to methyl thioacetate is D3Keq = 0.965, and the corresponding effect for addition of methoxide ion to methyl thioacetate wherein three waters are hydrogen bonded to the carbonyl oxyanion is D3Keq = 1.086. Neither of these calculated isotope effects is as inverse as the experimental beta-deuterium isotope effect for acetylcholinesterase-catalyzed hydrolysis of acetylthiocholine, D3kE = 0.90š0.03. Structural comparisons show that the water-solvated methoxide adduct of methyl thioacetate is more expanded than is the neutral methanol addition adduct, and suggest that the degree to which the isotope effect is inverse (i.e. less than) is inversely correlated to the degree of expansion of the adduct. A similar correlation of beta-deuterium and beta-deuterium secondary isotope effects with the degree of expansion of the adducts is found for equilibrium additions of methanol and methoxide ion to acetaldehyde. These computational results suggest that the markedly inverse beta-deuterium isotope effect for the acetylcholinesterase reaction arises from enzymic compression of the transition state.

Balanced Pattern Specification Language (BPSL)

Opis:
Design patterns help us to respond to the challenges faced while developing Distributed Object Computing (DOC) applications by shifting developers' focus to high-level design concerns, rather than platform specific details. However, due to the inherent ambiguity of the existing textual and graphical descriptions of the design patterns, users are faced with difficulties in understanding when and how to use them. Since design patterns are seldom used in isolation but are usually combined to solve complex problems, the above-mentioned difficulties have even worsened. The formal specification of design patterns and their combination is not meant to replace the existing means of describing patterns, but to complement them in order to achieve accuracy and to allow rigorous reasoning about them. The main problem of the existing formal specification languages for design patterns is the lack of completeness. This is mainly because they tend to focus on specifying either the structural or behavioral aspects of design patterns but not both of them. Moreover, none of them even ventured in specifying DOC patterns and pattern combinations. We propose a simple yet Balanced Pattern Specification Language (BPSL) aimed to achieve equilibrium by specifying both the aspects of design patterns. The language combines two subsets of logic: one from the First-Order Logic (FOL) and the other from the Temporal Logic of Actions (TLA).

The aim of paper was evaluation of composition of high molecular weight (HMW) and low molecular weight (LMW) glutenin subunits in wheat-rye addition lines cv. Grana - cv. Dańkowskie Złote, substitution line 1B/1R and initial forms, as well as, localization of Sec genes on chromosomes ‘Dańkowskie Złote’ rye. Analysis were performed using SDS-PAGE electrophoresis. The composition of HMW glutenin for all addition lines and ‘Grana’ wheat was: N - 6+8 - 2+12. Composition of LMW glutenin was estimated only for 2R and 6RL addition lines, wheat, and octoploid triticale. In both addition lines the subunits N-6+9-15 were identified, in wheat cv. Grana subunits 1-6+9-15 were present, whereas in octoploid triticale polymorphism was observed (1-6+9-15 or 1-11+13-N). Rye storage proteins (secalins) was found in addition lines: 1R, 2R and substitution line 1B/1R. It can be concluded that genes responsible for secalin biosynthesis in rye cv. Dańkowskie Złote are localized on 1R and 2R chromosomes.

The toleration of poor soil and high efficiency of mineral nutrients use of rye has been successfully transferred to triticale, but only one rye chromosome (1RS) has been used in wheat breeding. We started studies on identification of other rye chromosomes potentially useful in improvement of wheat, with special emphasis on root characters. We analyzed dry matter, length and number of roots (seminal and adventitious roots separately), together with plant dry matter, in two sets of disomic wheat-rye addition lines (CS-‘Blanco’ and CS-‘Imperial’). Plants were grown in hydroponic culture. In relation to the ‘Chinese Spring’ (CS) wheat, all the addition lines showed decrease of plant and root size parameters. The chromosomes 5R and 7R were best tolerated in wheat, but they caused a decrease of root proportion in plant. The 4R addition was the least viable one, but the root/plant ratio was higher than in wheat. The results were influenced mainly by interaction between homoeology group and cultivar of origin of rye chromosomes. The highest interactions of this kind were found in the 5R and 6R additions.

Opis:
The aim of presented work was to identify of PCR amplifiedDNAfragments differentiating aluminium tolerant and sensitive forms of rye and to locate the markers on rye chromosomes. For identification of markers, the PCR system with semi-specific primers targeting intron-exon sequences of plant genes was applied. The modified method of bulked segregant analysis was used. The pooled DNAs of two or three F2 segregating populations were screened together with DNA of their parental inbred lines. Potential marker of tolerance gene was located on rye chromosomes using wheat/rye (Chinese Spring/Blanco) additional lines. The specific probes obtained from DNA fragments differentiating sensitive and tolerant forms of rye were hybridized to PCR amplified DNA fragments of sensitive and tolerant forms of rye and the set of wheat/rye addition lines. Independently of the method of digoxygenin labelling (primer extension or Taq polymerase reaction), the probes obtained showed similar hybridization patterns. The results of hybridisation of 21 probes prepared from 12 DNA fragments confirmed connection of selected DNA fragments with to aluminium tolerance or sensitivity. Most of these DNA fragments originated from tolerant forms of rye. Using this method it was possible to locate eight DNA fragments on rye chromosomes. Three DNA fragments hybridised to chromosome 4R, two DNA fragments to chromosome 6R and single DNA fragments to chromosomes 1R, 2R and 3R. Four DNA fragments indicating clear relationship with character studied were not located on particular chromosomes using this set of wheat/rye addition lines. Hybridisation of probes prepared from four DNA fragments revealed length polymorphism. Probes prepared from two DNA fragments were characterised as dominant markers. In other cases the type of marker (dominant/codominat) was not fully documented.

Among different methods used in surface engineering such as CVD, PVD, ion implantation etc., the techniques using high intensity ion or plasma beams are relatively new ones. The results reported thus far show that the treatment of steel surface with high intensity plasma pulses can lead to changes of its morphology and mechanical properties. Stainless steels have very good corrosion resistance, but they have low hardness and poor tribological properties. The intense pulsed plasma beams were used for modification of alloyed steels especially austenitic (1.4301 and 1.4401) and ferritic (1.4016) stainless steels with various content of alloying elements. Samples were irradiated with 2, 5 or 10 short (μs scale) intense (density of energy was about 5 J/cm2) pulses. Heating and cooling processes were of non-equilibrium type. In all samples the near surface layer of the thickness in μm range was melted and simultaneously doped with cerium and lanthanum. The aim of this work was to investigate the changes of stainless steel surface morphology after melting, rare earth elements (REE) addition and rapid solidification after interaction with intense pulsed plasma beams. The surface morphology was analyzed using a scanning electron microscopy (SEM) technique. Changes of surface roughness were determined by profilometric measurements. The efficiency of REE addition process was also determined.

Materiał glebowy pobrano z 2 gleb uprawnych (podtyp: czarna ziemia właściwa) Próbki, o strukturze naruszonej i nie naruszonej, przeznaczone do badań , pobrano z założonego (w warunkach kontrolowanych) doświadczenia wazonowego.
The paper presents results of investigations which aimed to test the effect of varying doses of effective microorganisms (EM) on basic physical and water properties of arable-humus horizons of mineral soils of different texture composition. The soil material for experiments was collected from two arable soils (subtype: proper black earth). Samples of impaired and intact structure intended for investigations were collected from pot experiments established under controlled conditions. The following parameters were determined in the examined soils: texture composition, soil density, solid phase density, total and drainage porosity, filtration coefficient, soil water binding potentials and useful retention. It was found that various EM doses exerted different influence on soils characterised by different texture. In general, such effect is advantageous for the soil but it always changes nearly all its physical and water properties (primarily: porosity, filtration coefficient, binding potential and water availability).

Opis:
Bones, even after their growth has been completed, react to both external and internal factors which include feeding. The process of bone mineralization, including limb bones, is a very important index of the effectiveness and quality of feeding procedures, aswell as a measure of the organism’s health status. Antibiotics, which used to be very popular growth stimulators in feeding, have recently been replaced with natural feed additives, namely herbs. Although the usefulness of herbs has been known for a long time, little information has been found so far concerning their use in animal feeding and effect on the physical parameters and the chemical composition of the bones chickens. The aim of the present work was to determine a potential influence of herbs added to chickens’ diets on the body weight, chosen physical parameters of tibia bones: their weight, length and the content of the basic mineral components of the bone. The experiment was conducted on 210 broiler chickens of the Ross 308 line, divided into seven feeding groups: the control group (I – a mixture containing an antibiotic) and six experimental ones (II – hops, III – lime, IV – lemon balm, V – pansy, VI – peppermint, VII – nettle). In the bone samples, their weight, length, and the chemical composition of ash: Ca, Mg and P were determined. The results obtained in the present study reveal that an addition of nettle or lemon balm to the mixtures led to an increase in the body weight of broiler chickens. An addition of hops (II) to the mixtures led to a decrease in the length of the bones. An addition of lemon balm, pansy, peppermint or nettle to the mixtures had a negative effect on the level of Mg in the bones.
