Opis:
A three-dimensional (3D) time-domain method is developed to predict ship motions in waves. To evaluate the Froude- Krylov (F-K) forces and hydrostatic forces under the instantaneous incident wave profile, an adaptive mesh technique based on a quad-tree subdivision is adopted to generate instantaneous wet meshes for ship. For quadrilateral panels under both mean free surface and instantaneous incident wave profiles, Froude-Krylov forces and hydrostatic forces are computed by analytical exact pressure integration expressions, allowing for considerably coarse meshes without loss of accuracy. And for quadrilateral panels interacting with the wave profile, F-K and hydrostatic forces are evaluated following a quad-tree subdivision. The transient free surface Green function (TFSGF) is essential to evaluate radiation and diffraction forces based on linear theory. To reduce the numerical error due to unclear partition, a precise integration method is applied to solve the TFSGF in the partition computation time domain. Computations are carried out for a Wigley hull form and S175 container ship, and the results show good agreement with both experimental results and published results.

Opis:
The Craig-Bampton modal synthesis method was used to establish the dynamic model of marine four-stroke diesel engine body and crankshaft. Based on the Greenwood / Tripp microlong contact theory considering the surface roughness and the generalized Reynolds equation considering the oil filling rate, the elastohydrodynamic lubrication model of the main bearing of the four - stroke diesel engine is found. At the rated speed, the lubrication performance of the main bearing is simulated and analyzed by the maximum dynamic pressure, the minimum oil film thickness and the friction power. The results show that the oil pressure of 4 # main bearing is the largest and the maximum oil film pressure is in the 4 # main bearing position. The friction load of 4 # main bearing is the largest. The average oil film thickness of 4 # main bearing is the smallest and the minimum oil film The thickness also occurred in the 4 # main bearing position; it can be seen 4 # bearing the most bad lubrication conditions.

A study has been carried out to investigate the formation mechanism and development of streamwise vortices induced by leading edge tubercles in a high speed compressor cascade. The preliminary assessment of the cascade performance in terms of the total pressure loss coefficient shows that the loss reduction is achieved at high incidence angles. A smaller wavelength leads to higher additional losses at the design point, but gives rise to a greater loss reduction at high incidence angles. The modified cascade with a tubercle wavelength of 4% chord achieves the maximum loss reduction of 36.1% at i = 10◦ , as well as the stall angle improvement of 27.6%. The formation mechanism of streamwise vortices is elaborated on the basis of the streamwise vorticity equation, in which the streamwise turning terms may be responsible for the generation of streamwise vortices. Slices of streamwise vorticity at various streamwise locations, combined with vorticity strength distributions, have been presented to study the development of streamwise vortices. The counter-rotating vortices are divided into the crest-induced streamwise vortices (CSVs) and trough-induced streamwise vortices (TSVs). A streamwise vortex pair formed from a part of the CSV sheets behind troughs, is gradually entrained by the TSV pair along the streamwise direction. In addition, the tubercles with a smaller wavelength result in higher streamwise vorticity strength with which the streamwise vortices interact with the flow separation more sufficiently and delay the separation to a greater extent.

Pinellia ternata is one of the most important herbs in traditional Chinese medicine. Recently, its yield is hard to meet the market demand, therefore the system of in vitro micro-tubers was developed for its propagation and endogenous GA was revealed probably function negatively during micro-tubers induction. However, the mechanism is still unknown. In this study, 70 mg/L chlormequat chloride (CCC) was successfully used to inhibit the endogenous GA content and promote the micro-tubers induction. Subsequently, suppression subtractive hybridization (SSH) was performed to identify GA-related genes associated with in vitro micro-tubers formation in P. ternata. The cDNAs of micro-tubers induced with and without CCC were used as the “tester”, and “driver”, respectively. SSH library sequencing yielded 300 expressed sequence tags (ESTs). Finally, 226 ESTs were retained after screening, 84 of which had no significant homology to any of previously identified genes and 39 of the remaining 142 ESTs represented singletons. Real-time quantitative RT-PCR analysis of the expression patterns showed that all 5 transcripts showed signal alteration during the process of in vitro micro-tubers formation. The sequences appeared to be highly homologous with 60S ribosomal protein, 26S ribosomal RNA gene, zinc transporter protein, 12kD storage protein and malate dehydrogenase, respectively. These results would facilitate the functional characterization of the GA-related genes associated with in vitro micro-tubers development and subsequent in vitro manipulation.

Opis:
A proof of concept for using paper test as a suitable method in the production of monoclonal antibodies (MAbs) is reported. The paper test which detects antibodies against porcine circovirus type 2 (PCV2) using colloidal gold-labelled capsid protein as the antigen probe was applied exclusively in the screening of anti-PCV2 MAbs. It allowed the detection of 118 single cell clones within 30 min using naked eyes. MAbs with specific binding to authentic epitopes on the virus were selected using a blocking strategy in which the antibody was pre-incubated with PCV2 viral sample before applying to the test paper. Five hybridomas secreting MAbs against the capsid protein were obtained, with only three of them capable of binding to PCV2. The results were validated and confirmed using enzyme-linked immunosorbent assay and immunofluorescence assay. The paper test is simple, rapid, and independent on professional technicians and proves to be an excellent approach for the screening of MAbs against specific targets.
