Objectives: This study aims at exploring associations of general self-efficacy (GSE), workplace violence and doctors' work-related attitudes. Material and Methods: In this study a cross-sectional survey design was applied. Questionnaires were administrated to 758 doctors working in 9 hospitals of Zhengzhou, Henan province, China, between June and October 2010. General information on age, gender, and years of working was collected, and the doctors' experience and witnessing workplace violence, job satisfaction, job initiative, occupational stress as well as GSE were measured. General linear regression analysis was performed in association analyses. Results: Both experiencing and witnessing workplace violence were significantly positively correlated with the level of occupational stress but significantly negatively correlated with job satisfaction, job initiative, and GSE. General self-efficacy significantly modified relationships between both experiencing and witnessing workplace violence with occupational stress (β = 0.49 for experiencing violence; β = 0.43 for witnessing violence; p < 0.001) and with job satisfaction (β = -0.35 and -0.34, respectively; p < 0.05). However, it did not modify the relationships between both experiencing and witnessing workplace violence with job initiative (p > 0.05). The levels of occupational stress declined significantly with the increase of GSE, while job satisfaction increased significantly along with its increase. The effects of GSE on occupational stress and job satisfaction weakened as the frequency of violence increased. Conclusions: The findings suggest that GSE can modify effects of workplace violence on health care workers' stress and job satisfaction. Enhancing GSE in combination with stress reduction may lead to facilitating health care workers' recovery from workplace violence, and thereby improving their work-related attitudes.

Hypericum perforatum is well known for its antidepressant and anti-inflammatory activities, for which hypericin and its derivatives are indicated to be the most active compounds. Hypericin synthase (Hyp-1) is the only protein proven to catalyze the synthesis of hypericin. In this study, the full-length cDNA of Hyp-1 was chemically synthesized according to the Hyp-1 sequence in GenBank (accession no. AY148090) and then cloned into the plasmid pET22b. Hyp-1 was expressed in Escherichia coli BL21 (DE3) and purified with a Ni-NTA column. The purified protein was used to immunize New Zealand white rabbits, from which an antiserum was purified by protein G affinity chromatography. The polyclonal antibody against Hyp-1 provides a valuable tool for the study of hypericin biosynthesis in H. perforatum. Expression of Hyp-1 and the cellular distribution of hypericin were analyzed in different organs of red-pigmented H. perforatum plantlets. The black glands were not the only site of hypericin accumulation and the results indicated that hypericin might be synthesized in mesophyll cells or in tissues of the root and/or stem and then transported to the glands. This work provides a foundation for further investigation of the regulatory mechanism of hypericin synthesis during the development of H. perforatum.

The role of Survivin in the pathogenesis of leukemia was explored in order to discover the effective avenues for gene therapy. Most primary leukemia cells isolated from patients as well as three leukemia cell lines (HL-60, K562, and U937) all expressed Survivin gene. To investigate the relationship between Survivin and chemotherapeutic resistance, HL-60 cells were treated with daunorubicin (DNR), mitoxantrone (MIT) or arsenious oxide (As2O3), and it was found that after 24 h the level of Survivin mRNA was decreased by 9.7%, 41.0% and 27.5%, respectively. At 72 h, the level of Survivin mRNA was increased by 21.2% and 65.2% in HL-60 cells treated with DNR or MIT, but decreased by 33.2% in those treated with As2O3 as compared with that in the cells treated for 24 h. These results showed that DNR and MIT could initally decrease the expression of Survivin and then increase it, but As2O3 could decrease the Survivin expression continually. Furthermore, shRNA plasmids targeting the Survivin gene (pEGFP-Survivin), which can silence the expression of Survivin with a high specificity, were constructed. pEGFP-Survivin and pEGFP-H1 were transfected into HL-60 cells via electroporation and selected by G418, and HL-60/Survivin and HL-60/EGFP cells were obtained. After treatment with DNR, the cell survival rate and IC50 of DNR in HL-60/Survivin cells were decreased substantially as compared with those of HL-60/EGFP and HL-60 cells (IC50 of DNR: 18.3 ± 2.45 vs 40.8 ± 6.37 and 39.2 ± 5.91 ng/ml, respectively), and the apoptosis rate was elevated ((84.3 ± 19.7)% vs (45.8 ± 13.8)% and (50.9 ± 12.4)%, respectively). These results suggest that shRNA can down-regulate the expression of Survivin in HL-60 cells substantially and improve their sensitivity to DNR. They also further explain the pathogenesis of leukemia drug resistance and provide new theory in the design of clinical therapies.

In recent years, various models based on convolutional neural networks (CNN) have been proposed to solve the cardiac arrhythmia detection problem and achieved saturated accuracy. However, these models are often viewed as “blackbox” and lack of interpretability, which hinders the understanding of cardiologists, and ultimately hinders the clinical use of intelligent terminals. At the same time, most of these approaches are supervised learning and require label data. It is a time-consuming and expensive process to obtain label data. Furthermore, in human visual cortex, the importance of lateral connection is same as feed-forward connection. Until now, CNN based on lateral connection have not been studied thus far. Consequently, in this paper, we combines CNNs, lateral connection and autoencoder (AE) to propose the building blocks of lateral connection convolutional autoencoder neural networks (LCAN) for cardiac arrhythmia detection, which learn representations in an unsupervised manner. Concretely, the LCAN contains a convolution layer, a lateral connection layer, an AE layer, and a pooling layer. The LCAN detects salient wave features through the lateral connection layer. The AE layer and competitive learning is used to update the filters of the convolution network—an unsupervised process that ensures similar weight distribution for all adjacent filters in each convolution layer and realizes the neurons’ semantic arrangement in the LCAN. To evaluate the performances of the proposed model, we have implemented the experiments on the well-known MIT–BIH Arrhythmia Database. The proposed model yields total accuracies and kappa coefficients of 98% and 0.95, respectively. The experiment results show that the LCAN is not only effective, but also a useful tool for arrhythmia detection.

This paper proposes theoretical and numerical approaches to scrutinize the free vibration of orthogonal stiffened cylindrical shells. According to K´arman-Donnell shell theory, the total energy of the stiffened cylindrical shells is derived. Based on the principle of minimum potential energy, the eigenfunction related to the frequency is established and solved by developing a Matlab program. Analytical solutions of the natural frequency for free vibraion of the stiffened cylindrical shells are calculated and are verified against the finite element results from ABAQUS software. On account of the observations from the parametric study, an optimization scheme of the stiffeners is proposed.

Opis:
Human STAU1 is one member of the family of double-stranded RNA (dsRNA)-binding proteins. It is thought to function in transporting mRNA, controlling translation and eliciting mRNA decay in neurons, and to function in infection of influenza virus and human immunodeficiency virus type 1 (HIV-1). Four transcripts coding two isoforms have been identified before. In this study, we have isolated a novel transcript of STAU1, coding a novel isoform that has six amino acids more (SFPLKQ) than isoform a. In order to examine the tissue distribution of this novel isoform, we have performed RT-PCR experiments and the analysis showed that it was highly expressed in heart, liver, kidney and pancreas.

In gold ore, quartz plays an important role in mineral formation by acting as the follower. Understanding counterion release, transport, and deposition in alkali solution is a prerequisite for evaluating the potential role of gold separate from quartz deposits in pretreatment. In this work, the aggregation, retention, and release of counterion in alkali solution media were investigated by kinetic research and pure mineral experiments, the correlation and mechanism of these processes were revealed by combining geochemical theory, interaction energy calculation, and quantum chemistry. The results showed that the retention and release of counterion were closely related to the dissolution and corrosion rate of quartz. The NH4+ and Fe2+ with higher mineral affinity reduced the quartz stability, and the dispersion stability and mobility of the quartz were greatly improved by an alkaline substance due to the enhancement of steric hindrance effects. Quantum chemical calculation results show that ammonium ion promotes the dissolution of quartz stronger than ferrous ion, which is mainly reflected in reducing the activation energy required for the formation of transition state (TS1), which can be verified by kinetic calculation. These findings provide essential insight into the extraction of gold coated by quartz as well as a vital reference for the experiment of gold-loaded quartz leaching in mineral processing.

African swine fever is an acute, febrile and highly virulent porcine disease causing serious economic losses worldwide. The pK205R protein of the African swine fever virus (ASFV) is largely expressed in the early stages of infection, which has given the K205R gene extensive attention. In this study, the ASFV K205R was cloned and expressed in Escherichia coli BL21 (DE3). Expression of histidine-tagged pK205R with a molecular mass of 44 kDa was determined by 12% sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and Western blot analysis. Optimisation of culture conditions allowed induction of the recombinant protein with 0.4 mM Isopropyl β-D-thiogalactoside (IPTG) at 37oC for 2 h. The protein existed in cellular supernatant and was purified using a Ni-NTA resin column. The purified protein was used to immunize rabbits four times to enable the production of polyclonal antibodies, and the antiserum titre was detected by ELISA. The results showed that the purified pK205R can react with ASFV positive serum specifically by Western blotting. The pK205R had high antigenicity, which indicated that pK205R could be used as an antigen for detection of ASFV-specific antibodies in ELISA testing, and the recombinant protein could contribute to further research of the action and structure of pK205R.
