Opis:
Pedunculate oak is a species of major economic importance in Polish forests. This species seedlings constitute 18.9% of the nursery production in the country. One of the most widespread fungal disease these oaks face is the oak powdery mildew induced by Erysiphe alphitoides. Nursery production is on the lookout for environment−friendly means of plant protection as well as growth simulators capable of raising levels of productivity. Our objectives was to assess two preparation treatments regarded as biostimulators (registered as fertilisers under the names Apol−Humus and Apol−Hum Chelat) in the raising of seedlings of pedunculate oak in bare−root nursery conditions. The experiment included 6 variants with seedlings treated with the above agents, protected against mildew in the traditional way using Falcon 460 EC, treated with fertiliser and fungicide at the same time, or left as control specimens with no measures taken. All preparations were applied by means of foliar spraying 5 times in the course of the season. Survival of seedlings was then assessed at the season end, along with biometric features like root−collar diameter, length of main root and stem, dry mass of leaves and stem, and degree of oak powdery mildew attack. Results indicated curtailment of mildew infection of oak seedlings thanks to both of the biostimulators, albeit to a more limited extent than when fungicide was applied. No stimulation of seedling growth was achieved, however, though the most favourable biometric parameters characterised the oaks supplied with both biostimulator and fungicide. Significantly higher survival rate was noted for oaks offered effective protection against mildew, in the variants with the fungicide or with simultaneous application of Apol−Humus or Apol−Hum Chelat with Falcon. The research points to the two biostimulators under study (based on natural components) being suitable for use in the nursery production of pedunculate oak seedlings, with it being presumed that either or both can allow for reduction in amount of fungicide necessary to be used to safeguard seedlings against mildew.

Opis:
Scots pine stands in central Poland are now seen to be experiencing dynamic development of lower layers formed by oaks. The under−canopy natural renewal taking place in this way is sometimes characterised by good silvicultural quality, and could therefore be taken advantage of in the process of stand conversion. The study assess the quality of existing undergrowth layers of oak present in older stands of Scots pine, most especially from the point of view of their suitability for subjection to further silvicultural measures. The research material was collected in 10 objects where site type was classified as mesic mixed coniferous site type (BMśw) as well as in 11 ones with mesic mixed broadleaved forest (LMśw) site type. In each site type, 2 or 3 stands were selected to typify undergrowth layers aged 15, 20, 25 or 30 years. Assessment of oak undergrowth trees was made on the basis of 4−point scale in case of trunk or crown quality and vitality, and on a 3−point scale when vitality and the development trend were concerned. Next we selected the trees with future potential characterised by the highest quality from the point of view of all studied traits. A further determination concerned the influence on the traits in these best trees of the oak understorey that was exerted by biometric features of both the sheltering Scots pine stand and the under−growth itself. The studied undergrowth oaks are of either very good or good quality. Around 90% of these young trees could be assigned to 1st or 2nd quality class with the respect of all of analysed traits. Furthermore, fertility of the site type was found not to have any influence on the quality of the young oaks. However, where the number of trees identified as having future potential is concerned, influence was demonstrated for density of trees in both stand and the undergrowth layer, as well as their height, breast−height diameter and basal area. Oaks forming the lower layer beneath Scots pine stands are thus characterised by good quality and growth trends, to the extent that this suggests allowing further growth to take place, with inclusion within the main stand or next generation.

Celem pracy było określenie możliwości zastosowania bakterii potencjalnie probiotycznych Lactobacillus johnsonii K4 lub Lactobacillus casei O12 do produkcji napoju miodowego akceptowanego sensorycznie i zawierającego żywe mikroorganizmy. Materiałem do badań były napoje miodowe przygotowane z miodu wielokwiatowego i wody wodociągowej (12,05 g/100 ml). Napoje poddano 2-, 3-, 4- i 5-dniowej fermentacji monokulturą bakterii (po inokulacji ok. 7 log jtk/cm³) w temp. 30 i 37 ºC. Średnia liczb bakterii po fermentacji zawierała się w przedziale 6,75 ÷ 8,14 log jtk/cm³. Wzrost bakterii był zależny od szczepu, czasu i temperatury fermentacji (p < 0,05). Te same czynniki istotnie wpłynęły na średnie wartości pH we wszystkich próbach w kolejnych dniach fermentacji i mieściły się w granicach 4,6 ÷ 4.8 (p < 0,05). Czas fermentacji był czynnikiem statystycznie istotnie wpływającym na ocenę sensoryczną (p < 0,05). Najwyższe noty w ocenie sensorycznej przyznano próbkom fermentowanym szczepem Lb. casei O12 przez 2 dni w temp. 37 ºC. Wybrane bakterie z rodzaju Lactobacillus można zastosować do wytwarzania innowacyjnego napoju o pożądanych cechach sensorycznych. Na tym etapie badań produkt nie może być nazwany probiotycznym, jednak jakość sensoryczna i zakres przeżywalności szczepów predysponuje do dalszej oceny ich właściwości probiotycznych.
The objective of the research study was to determine the possibility of using potentially probiotic strains of Lactobacillus johnsonii K4 and Lactobacillus casei O12 to manufacture a honey beverage having a appropriate sensory quality and containing viable microorganisms. The research material consisted of honey beverages made of multifloral honey and tap water (12.05 g/100 ml). The beverages were fermented for 2, 3, 4 and 5 days with a bacterial monoculture (after inoculation of approx. 7 log cfu/ml) at a temperature of 30 and 37 ºC. The average count of bacteria ranged from 6.75 to 8.14 log cfu/ml. The growth of bacteria depended on the strain, the time and the temperature of fermentation (p < 0.05). The same factors significantly impacted the mean pH values of all the samples on the subsequent days of fermentation; those pH values ranged between 4.6 and 4.8 (p < 0.05). The fermentation time was a factor to statistically significantly impact the sensory evaluation (p < 0.05). The samples fermented with a Lb. casei O12 strain for 2 days at 37 ºC were given the highest scores in the sensory evaluation. The selected bacteria of the Lactobacillus species can be used to manufacture an innovative beverage having desirable sensory qualities. At this stage of the research, the product cannot be called probiotic; however, the sensory quality and the range of viability of the strains predispose them to continue evaluation of their probiotic properties.

Opis:
In most cases, mechanical site preparation (MSP) is a precondition if high−quality (natural or artificial) regeneration of forests is to be achieved. However, the measures involved here may differ in terms of the level of intervention of the soil environment, at both the surface and deeper down. The choice of MSP methods should be based on climatic conditions, site type and the species whose renewal is sought, while the effect should improve conditions for the emergence and growth of the young generation of trees, with the influence on the soil environment being limited as far as possible. The research sought to compare physical and chemical features of the soils of the microhabitats created in clear−cut areas as a result of MSP using either an LPz double mould−board forest plough (furrow and ridge), an active plough (furrow and ridge) and a forest mill (belt and beyond the belt) as compared with unscarified soil. Investigated physical features included grain size (content of sand, silt and clay), bulk density and actual moisture, while the chemical ones: pHH2O and pHKCl, exchangeable acidity, total content of N and C and the C/N ratio, P2O5 content and base cations Mg2+, Ca2+, K+ and Na+. The most invasive MSP method for the soil environment is the active plough, which leaves the ridge exposed to processes of the decomposition of organic matter (and the highest content of N, C, Mg2+, Ca2+ and K+). Equally, this method may pose the greatest threat of mineral components of the soil being leached, with impoverishment of the habitat ensuing. The forest mill in turn offers the least invasive MSP. Most of the physical and chemical features of soil (other than exchangeable acidity and pHH2O and pHKCl) both in the belt areas and beyond them differed little from the those characterising non−scarified soil. Also the analysed features determined for the furrows ploughed by the active or LPz ploughs or the belt prepared using the forest mill did not differ significantly.

Opis:
Nanoparticles are gaining ever−wider application in plant production (for both agriculture and forestry), in the role of pesticides, as well as stimulators of plant growth and resistance. We sought to determine the efficacy of silver and copper nanoparticles (AgNPs and CuNPs respectively), used as seed dressings or subjected to foliar application, in affording protection from parasitic damping−off disease among soil−grown seedlings of Scots pine in a forest nursery. Experiments also assessed the influence of the nanoparticles on the growth of the plants potentially safeguarded in this way. Nanoparticles were used in seed soaking and foliar spraying at 50 ppm concentration. Pines treated with fungicides (Zaprawa Nasienna T 75 DS, Acrobat MZ 69 WG, Topsin M 500 SC, Gwarant 500 SC, Thiram Granuflo 80 WG and Signum 33 WG) or unprotected at all were used for comparison. In each treatment (AgNPs, CuNPs, fungicides or unprotected), seedlings were inventoried 6 weeks after the sowing and at the end of the growing season, while all individuals on 80 1−m−long segments of seed row were counted. At the end of the growing season, shoot length, root−collar diameter, root length and dry mass of shoots and roots were determined. The seedlings treated with nanoparticles had longer root systems of greater dry mass, but also only more weakly−developed above−ground parts (both height and dry mass being limited) in comparison with young Scots pines that had been fungicide−treated or were unprotected. It resulted in a significantly more favourable ratio between shoot and root masses, where nanoparticle treatment had been applied. The effectiveness of the protection extended to the germination and first−growth stages up to 6 weeks from the time of sowing was furthermore shown to be greatest where AgNPs had been applied, while at the season end there were comparable results among pines treated with either nanoparticles or fungicides. Our results thus indicate that nanoparticles limit damping−off disease in pine seedlings (AgNPs more effectively than CuNPs), with that effectiveness also proving comparable with that noted for the fungicides applied traditionally.

Opis:
Mechanical site preparation is recommended to create optimum conditions for seed germination and seedling growth in a clear−cut area. Lack of soil scarification or less−intense methods may be more suitable from an environmental point of view, but can result in a failure to obtain natural regeneration. This study compared the effects of three mechanical site preparation methods (SPM) characterized by different levels of soil scarification (i.e. double mould−board forest plough (LPz), active plough (PA) or forest mill (FL)), as well as a control variant without mechanical site preparation, on height, density, spatial distribution and survival among naturally regenerating Scots pines (Pinus sylvestris L.) in the first 2 years of growth in a clear−cut area of NE Poland. The experiment was based around a four−block randomized block design involving the four variants. Seedling height was measured at the end of the first and second growing seasons, with measurements performed on 8 meter−wide transects extending across the entire width of the cleat−cut. We found no effect of SPM on seedling height, though the density of seedlings regenerating naturally was found to be dependent. After two years, the highest density was achieved in the PA variant (8.1 seedlings/m2) and the FL variant (7.0 seedlings/m2). A significantly lower density characterized the LPz variant (5.1 seedlings/m2) and the lowest of all the variant without soil scarification (3.2 seedlings/m2). The most homogeneous natural regeneration was achieved following the use of PA (2.6% of 1 m2 plots without seedlings after two growing seasons), while the least homogeneous was the variant without soil scarification (22.4% of 1 m2 plots). Survival of pines after the second growing seasons was quite high (over 80%), and did not differ significantly depending on the SPM. Our research therefore confirmed that, in the absence of mechanical site preparation, natural regeneration of sufficient density and homogeneity cannot be obtained. The best natural regeneration was obtained on sites prepared with PA, and FL scarification being only slightly less favorable. The low density and uneven distribution of seedlings on the soil prepared with LPz was a surprising result. The probable reason for this lay in the high precipitation in the first growing season. On one hand, this provided very good moisture conditions for seed germination and seedling growth on a substrate with high porosity in the PA and FL variants, but on the other the high bulk density of mineral soil in the LPz variant led to oxygen deficit.

Since its initial discovery in 1945, pitch canker caused by Gibberella circinata (anamorph Fusarium circinatum) has become one of the most important pine diseases in the world. It infests 57 pine species (including Pinus sylvestris) and Douglas−fir. F. circinatum has been reported in both nurseries and forests on all continents except Australia and Oceania. Both vegetative and reproductive parts of the host plant (i.e. stem, branches, roots, flowers, cones and seeds) can be infected by F. circinatum. For mature trees, the main symptoms include bleeding resinous cankers on the stem and branches as well as tree death. In nurseries, F. circinatum causes pre− and post−emergence damping off as well as mortality of older seedlings. Dispersal of fungal spores occurs through air, water splash, soil, insect vectors and translocation of the infected plant material. Currently there are no effective means of controlling pitch canker, although there is ongoing research into using chemical, physical and biological methods, quarantine and breeding for resistance to control this disease. The occurrence of the disease depends mainly on the presence and density of the host species, favorable climatic conditions (especially high humidity), presence of suitable vectoring and wounding agents and various stress factors. Taking into account the above−mentioned factors, threat to Scots pine in Poland should be considered marginal, but it cannot be ruled out completely. KEY WORDS

The aim of this study was to determine the mycorrhizal status of nursery seedlings of the wild service tree (Sorbus torminalis), which belongs to the Rosaceae family. Its mycorrhizal associations are still fragmentarily known, and data from the few existing studies indicate that it forms ectomycorrhizal symbiosis (ECM). We analyzed the degree of mycorrhizal colonization of thirty 2-year-old container-grown S. torminalis nursery seedlings, which belonged to three single-tree progenies. The roots were dominated by arbuscular mycorrhizae (AM), with the morphology of the Arum-type containing arbuscules, vesicles and hyphae; however, no ECM structures were found. The degree of root colonization of the analyzed seedlings by AM fungi was 83.6% and did not differ significantly between the three single-tree progenies. In addition to AM, structures of dark septate endophytes (0.7%) and sporangia of Olpidium spp. (1.1%) were found in wild service tree roots. In agreement with previous studies, we confirmed arbuscular mycorrhizae for S. torminalis. Moreover, this is the first report that roots of this Sorbus species show the Arum-type morphology of AM and are associated with Olpidium species.

Opis:
Different methods of logging slash management (LSM) and methods of soil preparation (MSP) can create different conditions for the growth of forest trees. This occurs through variation in soil aeration, tree nutrition or microbial conditions. These factors are modified by climate, soil fertility, treats from insects or fungal pathogens and renewed species. The aim of this study was to compare effects of three LSM methods: 1) crushing and mixing with the soil by Mery Crusher, 2) chopping and leaving on the soil surface, and 3) removal from the clear−cut, and three MSP: 1) double−mouldboard forest plough LPz−75, 2) active plough U−162, and 3) forest mill FAO−FAR FV 4088 on biometric parameters, density and survival of six−years old Scots pine seedlings. Field research was conducted in Narol Forest District (south−eastern Poland). The soil in the study area was classified as brown podzolic, formed on loose sands with typical mor humus.The experiment was established by planting 10 000 one−year−old pine seedlings per hectare, after clear cut and followed by a two−factor block design consisting of nine variants distinguished on the basis of LSM and MSP randomly assigned to each of the three blocks. Tree height and root collar diameter were measured three times, at the end of 1st, 3rd and 6th growing season. The results showed no statistically significant differences in the size of the analyzed parameters of planted seedlings. Seedlings in the variant with site preparation by crusher and forest mill were slightly higher and thicker. This indicates the better trophic conditions in that variant. LSM did not affect either seedlings density or their survival. A slightly higher survival rate and tree density occurred in variants with crusher regardless of soil preparation method. It follows that, all presented experimental variants give similar results. Therefore, selection of the most appropriate methods of site and soil preparation should primarily respect ecological and economic indicators, and such local factors as microclimate, mainly precipitation, site fertility and threats from pests and fungal diseases.

Opis:
Celem przeprowadzonych badań było określenie wpływu dodatku butylowanego hydroksytoluenu (BHT) do rozcieńczalnika mrożeniowego na jakość plemników knura oraz ich zdolność zapładniającą po procedurze zamrażania-rozmrażania. Nasienie pochodzące od 5 knurów (36 ejakulaty) kriokonserwowano w rozcieńczalniku żółtkowo-laktozowo-glicerolowym uzupełnionym różnymi stężeniami BHT: 0 mM (kontrola); 1,0 mM (R1); 1,5 mM (R2); 2,0 mM (R3). Jakość nasienia po rozmrożeniu oceniano na podstawie: ruchliwości plemników (CASA; ruch całkowity – TM, ruch postępowy – PM), translokacji fosfatydyloseryny (Annexin-V-FLuos Staining Kit) oraz stopnia fragmentacji DNA (TUNEL Assay). Jednocześnie kriokonserwowane nasienie użyto do inseminacji loszek, a zdolność zapładniającą plemników określano na podstawie skuteczności inseminacji oraz liczby prawidłowych morfologicznie zarodków ocenianych na podstawie stopnia fragmentacji DNA. Najwyższy odsetek plemników o ruchu postępowym oraz plemników żywych stwierdzono w rozcieńczalniku R3 (74,8 ±4,4% oraz 63,7 ±5,8%) w porównaniu z rozcieńczalnikiem kontrolnym (38,3 ±2,8% oraz 36,1 ±2,6%). Jednocześnie stwierdzono, że dodatek BHT do rozcieńczalnika mrożeniowego nie indukuje wzrostu odsetka plemników wczesnoapoptotycznych po rozmrożeniu, w porównaniu z rozcieńczalnikiem kontrolnym. Niezależnie od zastosowanego rozcieńczalnika mrożeniowego, procedura zamrażania-rozmrażania nie indukuje fragmentacji DNA w plemnikach knura. Najwyższą liczbę zarodków prawidłowych morfologicznie uzyskano od loszek inseminowanych nasieniem kriokonserwowanym w rozcieńczalniku z dodatkiem 1,5 mM BHT. Nie wykazano różnic w jakości zarodków, ocenianych na podstawie stopnia fragmentacji DNA, uzyskanych od loszek inseminowanych nasieniem mrożonym w badanych rozcieńczalnikach.
The objective of the study was to determine the effect of butylated hydroxytoluene (BHT) on the quality and fertilizing capacity of frozen-thawed (FT) boar semen. Semen from five boars (36 ejaculates) was resuspended in lactose-egg yolk-glycerol extender supplemented with 0 (control), 1.0 (R1), 1.5 (R2) or 2.0 mM BHT (R3). Sperm quality was assessed based on motility (CASA; TM: total motility; PM: progressive motility), phosphatidylserine (PS) translocation across the plasma membrane (Annexin-V-FLuos Staining Kit) and DNA fragmentation (TUNEL Assay). The FT semen was also used for intrauterine artificial insemination (AI) of synchronized gilts. The fertilizing capacity of the FT semen was assessed on the basis of the gilt insemination rate and the number of morphologically normal embryos. The quality of the preimplantation embryos was determined by observing a TUNEL-positive reaction. The highest percentage of progressive motile and viable spermatozoa was noted in extender R3 (74.8 ±4.4% and 63.7 ±5.8%), as compared with the control (38.3 ±2.8% and 36.1 ±2.6%). The addition of BHT to the extender did not increase early apoptotic changes in the frozen-thawed spermatozoa with respect to the control. Irrespective of the variant of the extender, cryopreservation and thawing did not induce fragmentation in the boar spermatozoa. The highest number of morphologically normal embryos from inseminated gilts was observed in the case of semen cryopreserved in extender supplemented with 1.5 mM BHT. No significant differences were observed in DNA fragmentation in the expanded blastocysts from gilts inseminated with FT semen cryopreserved in the extenders analysed.

This study evaluated the effect of supplementing the freezing extender with exogenous antioxidants on apoptotic-like changes in post-thaw boar spermatozoa. A total of 36 ejaculates were resuspended in standard lactose-egg yolk-glycerol extender supplemented with antioxidant to final concentrations of 0 (as control), 2.5mM GSH (group I), 5.0 mM GSH (group II), 150 IU/mL SOD (group III), 300 IU/mL SOD (group IV), 200 IU/mL CAT (group V), 400 IU/mL CAT (group VI), 150 IU/mL SOD+200 IU/mL CAT (group VII), 300 IU/mL SOD+400 IU/mL CAT (group VIII). Sperm motility and apoptotic-like changes were determined before and after freeze-thawing. The various markers of apoptotic-like changes were measured: plasma membrane permeability by YO-PRO-1/PI assay, phosphatidylserine (PS) translocation across the plasma membrane using fluorescein-labeled Annexin-V, mitochondrial transmembrane potential detected by JC-1, and DNA fragmentation evaluated by TUNEL assay. The highest percentage of progressive motile sperm was noticed in group II (PM% 64.2±15.4) compared with control (PM% 36.8±5.5). The supplementation of 400 IU/mL CAT (group VI) revealed significant (P<0.01) reduction of apoptotic-like changes (YO-PRO-1⁺/PI⁻: 13.1±7.5%, AnV⁺/PI⁻: 9.9±4.1%) in frozen-thawed spermatozoa compared with extendersupplemented with200 IU/mL CAT (groupV). Irrespectiveof the concentrationused, SOD and CAT in combination (group VII and group VIII) significantly (P<0.01) improved post-thaw sperm survival compared with the control. Evaluation by TUNEL assay revealed that cryopreservation and thawing did not induce DNA fragmentation in boar spermatozoa.

Opis:
The internal alpha particle beam of the heavy ion cyclotron operated by the Heavy Ion Laboratory (HIL) of the University of Warsaw has a maximum energy of 32 MeV and currently an intensity of up to 1 pμA. This beam is used by the HIL-University of Silesia collaboration for the production of research quantities of $\text{}^{211}At,$ $\text{}^{72}Se//\text{}^{72}As$ and $\text{}^{43,44}Sc$ radioisotopes. The produced activities are transported to the Institute of Nuclear Chemistry and Technology in Warsaw where research on therapeutic and imaging radiopharmaceuticals based on these radioisotopes is pursued.
