Opis:
Two cytotypes of Chenopodium album, diploid (2n=2x=18) and hexaploid (2n=6x=54), were analysed using flow cytometry and a FISH experiment. The genome size was indicated as 1.795 pg for the diploid and 3.845 pg for the hexaploid plants which suggested genome downsizing in the evolution of hexaploid cytotype. Double FISH with 25S rDNA and 5S rDNA allowed three to five homologue chromosome pairs to be distinguished depending on the cytotype. The Variation in size and number of rDNA sites between the polyploid C. album and its putative diploid ancestor indicated that rDNA loci underwent rearrangements after polyploidization. Flow cytometry measurements of the relative nuclear DNA content in the somatic tissue of C. album revealed extensive endopolyploidization resulting in tissues comprising a mixture of cells with a different DNA content (from 2C to 32C) in varying proportions. The pattern of endopolyploidy was characteristic for the developmental stage of the plant and for the individual organ. Polysomaty was not observed in the embryo tissues however endopolyploidization had taken place in most tested organs of seedlings. The endopolyploidy in diploid and hexaploid C. album was compared to find any relationship between the pattern of polysomaty and polyploidy level in this species. This revealed that polyploid plants showed a decline in the number of endocycles as well as in the frequency of endopolyploidy cells compared to diploid plants.

We analyzed DNA damage, mitotic activity and polyploidization in Crepis capillaris callus cells during short- and long-term in vitro culture, and the influence of plant growth regulators on these processes. Changes in the concentration of growth regulators altered the stability of callus. The level of DNA damage was highly dependent on the growth regulator composition of the medium. Cytokinin at high concentrations damaged DNA in the absence of auxin. Short- and long-term callus differed in sensitivity to growth regulators. Mitotic activity changed when callus was transferred to medium with modified growth regulators. Callus cell nuclear DNA content increased with age and in response to plant growth regulators. Hormones played a role in the genetic changes in C. capillaris callus culture. We demonstrated the usefulness of C. capillaris callus culture as a model for analyzing the effect of culture conditions, including plant growth regulators, on genetic stability.

The physicochemical and dispersive characterizations were conducted on the selected commercial titanium dioxides produced by, Z. Ch. POLICE, S. A. The dispersive properties were defined in detail by an analysis of particle size distribution and polydispersity index. Moreover, the microscope studies were executed to evaluate the surface morphology of the studied TiO2 forms. The profiles of titanium dioxides sedimentation in water were determined and the specific surface areas were defined by the BET method.

Opis:
The aim of this study was to effectively combine synthetic magnesium silicate with the lignin-derived biopolymer calcium lignosulfonate, using the in situ sol-gel route. Magnesium ethoxide and tetraethoxysilane were used as precursors of MgO and SiO2. The synthesis was carried out in alcoholic solution with the addition of ammonia solution as a promoter of hydrolysis. Calcium lignosulfonate was introduced to the reaction medium prior to the synthesis of magnesium silicate. The resulting hybrid powder material was thoroughly characterized, including morphology and particle sizes (SEM microscopy and the DLS technique), porous structure parameters (the BET method and BJH model), thermal stability (TG analysis) and electrokinetic stability (LDV measurements). FTIR spectral analysis was carried out to confirm the effectiveness of the proposed synthesis methodology. Based on the results, a mechanism is proposed for the MgSiO3/lignosulfonate interactions. The resulting novel type of hybrid material combines the multifunctional nature of the biopolymer (diversity of functional groups) with the well-developed porous structure of synthetic magnesium silicate. Its physicochemical parameters were found to depend significantly on the quantity of lignosulfonate used in the synthesis.

Opis:
Studies were conducted on the modification of titanium white surface using selected silane coupling agents. The effect of the concentration of the organic coupling agents was examined (0.5; 1 or 3 weight parts per 100 weight parts of TYTANPOL R-003, TYTANPOL R-211 or TYTANPOL R-213 preparation of titanium white). The dispersive properties were determined by an estimation of the particle size distribution curves and of the polydispersity index. Moreover, microscopic observations were conducted permitting to evaluate the surface morphology of the modified TiO2 particles. The profiles of sedimentation in water were also determined for the titanium whites and the BET specific surface areas were determined. Selected samples of the modified and unmodified titanium whites were subjected to elemental analysis.

Opis:
Hydroxyapatite is used as a matrix for immobilization of protease from Aspergillus oryzae by a process of adsorption. The matrix obtained has the surface area of 26 m2/g and particles in the shape of flakes of diameters no greater than 650 nm. The efficiency of the proposed method was confirmed by the Fourier transform infrared spectroscopy, elemental analysis and by analysis of parameters of the pore structure of matrix and products after immobilization. On the basis of the Bradford method it was found that the greatest amount of enzyme (132 mg/g) was immobilized from a solution of initial enzyme concentration of 7 mg/cm3 after 24 h of the process.

Opis:
The effects of microwave radiation on viability of fungal and actinomycetal spores growing on agar (medium optimal for growth) as well as on wooden panel and drywall (common building construction/fi nishing materials) were studied. All materials were incubated at high (97-99%) and low (32-33%) relative humidity to mimic “wet” and “dry” environmental conditions. Two microwave power densities (10 and 60 mW/cm2) and three times of exposure (5, 30, and 60 min) were tested to fi nd the most effective parameters of radiation which could be applied to non-invasive reduction or cleaning of building materials from microbial contaminants. Additionally, a control of the surface temperature during the experiments allowed differentiation between thermal and microwave effect of such radiation. The results showed that the viability of studied microorganisms differed depending on their strains, growth conditions, power density of microwave radiation, time of exposure, and varied according to the applied combination of the two latter elements. The effect of radiation resulting in a decrease of spore viability on “wet” wooden panel and drywall was generally observed at 60 min exposure. Shorter exposure times decreased the viability of fungal spores only, while in actinomycetes colonizing the studied building materials, such radiation caused an opposite (supporting growth) effect.
