The mannose-binding agglutinin from bulbs of Lycoris aurea (LAA) agglutinates rabbit but not human erythrocytes. The molecular mass of the monomer in SDS/PAGE is 12 kDa while the apparent molecular mass in gel filtration is 48 kDa, indicating that LAA is a homotetramer. The full-length cDNA of LAA contains 683 bp with an open reading frame encoding a protomer of 162 amino-acid residues. Hydrophobic Cluster Analysis and molecular modeling of the 109-residue mature polypeptide suggested a similar secondary and tertiary structure to those of Narcissus pseudonarcissus agglutinin (NPA). Molecular docking revealed that, besides the three mannose-binding sites common among Amaryllidaceae lectins, LAA also contains a fourth unique mannose-binding site formed by a tryptophan cluster. The existence of four mannose-binding sites in each monomer of LAA is very unusual and has only been reported for NPA earlier.

Opis:
Copper, an essential transient element, can be toxic to cells when present in excess. Altered copper homeostasis is involved in pathological events of many diseases. Human CUTA isoform2 is a member of cation tolerance protein (CutA1) family. In this study, we examined the effect of CUTA isoform2 overexpression on copper toxicity. It was shown that overexpressed CUTA isoform2 sensitized HeLa cells to copper toxicity by promoting copper-induced apoptosis. The inhibition effect of excessive copper on cell proliferation was also enhanced by overexpressed CUTA isoform2. So CUTA isoform2 was implicated to be involved in the cytotoxicity of copper.

Opis:
For one kind of high current diodes composed of a ceramic-metal welding vacuum interface, the electrical design was presented. For compactness, a radial type insulator and a cone-column anode crust were adopted. The shielding methods around cathode and anode region were applied to mitigate the influence of welding solder to vacuum flashover. Finite Element Analysis (FEA) simulation results indicated that by adjusting the anode outline and shielding shape, the electric fields along the ceramic were well distributed. High voltage test was conducted on a long-pulse accelerator and experimental results confirm the theoretic design: the diode can stably hold on 400 kV and 200 ns voltage pulse.

Microstructure and electrical properties of hydrogenated nanocrystalline silicon (nc-Si:H) film deposited on glass substrate at low temperature were characterized by average grain size, crystallinity, and dark electrical conductivity data obtained from the Raman and electrical conductivity spectroscopy, respectively. The average grain size, crystallinity and electrical conductivity have a similar change with substrate temperature. A threshold substrate temperature determined by silane concentration appears in their corresponding spectroscopy vs. substrate temperature. The dependence of crystallinity, average grain size and electrical conductivity on substrate temperature were accounted for by surface diffusion model and heterojunction quantum dot model, respectively.

Opis:
A novel type of coaxial vircator with 3 anodes is proposed. The advantages on high power microwave generation is tested by particle-in-cell simulation and theoretical analysis model is set up as well. The tri-anode coaxial vircator behaves a very stable frequency in the pulse duration. In the tri-anode coaxial vircator, the injecting electron beam is well premodulated in the first anode cavity and forms an inerratic bunch-dispersion mechanism in the second anode cavity; the energy-loss electrons penetrate through the third anode and absorbed by the dumping cavity locates in the centre electrode, which helps keeping the bunch-dispersion cycle undisturbed. A 3.8 GHz tri-anode coaxial vircator is theoretically designed and optimized by particle-in-cell simulation, which gives beam-wave conversion efficiency of over 20% with a stable predominant frequency, in accordance with the particle-in-cell simulation and analytic model.

Opis:
The life and reliability of machined components or elements are affected greatly by the surface integrity. Machined surfaces by conventional manufacturing processes such as turning and milling have inherent irregularities and defects like tool marks and scratches that cause energy dissipation (friction) and surface damage (wear). Burnishing is a kind of chip-less processing which improves the surface integrity of machined components. To promote the application of this process, reliability assessment of burnishing is investigated theoretically and experimentally in this paper. The methodology was developed based on probabilistic modeling and experimental results. The reliabilities of two burnishing processes (rigid burnishing and elastic burnishing) are analyzed and compared with the methodology proposed.
Na trwałość i niezawodność obrabianych komponentów i elementów wielki wpływ wywiera stan warstwy wierzchniej. Powierzchnie obrabiane w tradycyjnych procesach wytwórczych, takich jak toczenie czy frezowanie nieodłącznie charakteryzują się nierównościami i defektami w postaci śladów po narzędziach i zadrapań, które powodują rozpraszanie energii (tarcie) oraz niszczenie powierzchni (zużycie). Nagniatanie jest rodzajem obróbki bezwiórowej, która poprawia stan warstwy wierzchniej obrabianych komponentów. Chcąc propagować stosowanie tego procesu, przedstawiamy w niniejszym artykule badania teoretyczne i eksperymentalne oceny niezawodności nagniatania. Metodologię stworzono w oparciu o modelowanie probabilistyczne i wyniki eksperymentów. Przeanalizowano niezawodność dwóch procesów nagniatania (nagniatania z dociskiem sztywnym i nagniatania z dociskiem sprężystym) i dokonano ich porównania za pomocą proponowanej metodologii.

In this paper, a novel switching-capacitor DC-DC voltage converter with higher efficiency will be presented. This circuit was designed by modifying the output stage of the conventional static charge-transfer-switch (CTS) charge pump in a cross-coupled configuration. In this design, a control scheme to overcome both the reverse charge sharing and the threshold drops in the CTS pump was also employed. In this study, the capacitances for all the pumping capacitors were selected the same as 0.1 μF. With this design, our circuit can operate with a clock rate up to 1 MHz. The performance of this circuit was first evaluated by simulation by HSPICE with the 0.35-μm technology of TSMC. The results showed that this circuit can pump the low input of 1.5V nearly 5 times at the output. The conversion gain can be around 95%. The performance of the real chip manufactured by TSMC was measured and will be compared with the simulation results.

Objective: To study the effects of velvet antler polypeptides (VAPs) on osteoarthritic chondrocytes (OCs) in rabbits. Methods: An osteoarthritic rabbit model was established according to Hulth's method. OCs were isolated and cultured for observation of the cell cycle. Cell proliferation was detected by MTT assay and the cell cycle was monitored by flow cytometry. The phenotype was determined by toluidine blue staining as well as immunohistochemical staining for collagen type II. The expression of MMP-1, MMP-3, MMP-13, TIMP-1, and collagen I and X mRNA by chondrocytes was assayed by RT-PCR. Results: The VAPs had no obvious proliferative effect on OCs and did not affect the cell cycle. However, they significantly reduced the proportion of early apoptotic cells in a dose-dependent manner. Further, VAPs inhibited the expression of collagen I and X mRNA and induced abnormal expression of MMP-1 and MMP-13 mRNA. VAPs had no significant effect on MMP-3 and TIMP-1 mRNA levels. The toluidine blue and collagen type II immunohistochemical staining intensities of VAP-treated chondrocytes were positively correlated with the concentration of VAPs used. Conclusion: VAPs had no significant effect on OC proliferation and the cell cycle, but did increase the glycosaminoglycan (GAG) and collagen type II expression levels in the extracellular matrix, and down-regulated collagen I and X mRNA expression. Treatment of cartilage cells with VAPs maintained their normal phenotype, inhibited matrix metalloproteinases (MMPs) secretion, kept the balance of cartilage matrix metabolism, and sustained an external environment where the cartilage cells could survive. Moreover, VAPs reduced the proportion of early apoptotic cells, suggesting that they may block the apoptotic pathway in OCs.

Objective. To explore the effect of dexamethasone (DEX) on monocyte adhesion function and its underlying mechanism. Methods. The effects of DEX and fasudil on adhesion of cultured U937 monocytes to human umbilical vein endothelial cells (HUVEC) following stimulation with phorbol myristate acetate (PMA) were studied; Changes in the Rho-associated coiled-coil protein kinase 1 (ROCK1) protein content and activity were evaluated. Results. DEX and fasudil significantly inhibited U937 cell adhesion rates under PMA stimulation and inhibited ROCK1 activity. Mifepristone (RU-486) and cycloheximide (CHX) did not alter these effects of DEX. Conclusions. DEX interferes with the adhesion function of U937 cells through the inhibition of ROCK1 activity.

The extracellular polymeric substance (EPS) produced from Rhizobium radiobacter F2, designated as EPSF2 , was investigated as a biosorbent for the removal of Pb(II) and Zn(II) from aqueous solution. The optimum biosorption pH values were 5.0 for Pb(II) and 6.0 for Zn(II). Kinetics study revealed that the biosorption followed pseudo-first-order model well, and the equilibrium data fit the Langmuir model better. The adsorbed metal ions could be effectively desorbed by HCl. Desrobed EPSF2 regained 80% of the initial biosorption capacity after five cycles of biosorption-desorption-elution. These results demonstrated that EPSF2 could be a promising alternative for Pb(II) and Zn(II) removal from aqueous solution.

Opis:
Fruit biomechanics is needed for quality determination, multiscale modelling and engineering design of fruit processes and equipments. However, these determined fruit biomechanics data often have obvious differences for the same fruit or tissue. In order to investigate it, the fruit biomechanics based on anatomy was reviewed in this paper. First, the anatomical characteristics of fruit biomaterials were described at the macroscopic ‘tissue’ level and microscopic ‘cellular’ level. Subsequently, the factors affecting fruit biomechanics based on anatomy and the relationships between fruit biomechanics, texture and mechanical damage were summarised according to the published literature. Fruit biomechanics is mainly affected by size, number and arrangement of cells, quantity and volume of intracellular spaces, structure, thickness, chemical composition and permeability of cell walls, and pectin degradation level and turgor pressure within cells based on microanatomy. Four test methods and partial determined results of fruit biomechanics were listed and reviewed. The determined mechanical properties data of fruit are only approximate values by using the existing four test methods, owing to the fruit biomaterials being non-homogeneous and living. Lastly, further aspects for research on fruit biomechanics were proposed for the future.

This paper is devoted to studying the globally exponential stability of impulsive high-order Hopfield-type neural networks with time-varying delays. In the process of impulsive effect, nonlinear and delayed factors are simultaneously considered. A new impulsive differential inequality is derived based on the Lyapunov–Razumikhin method and some novel stability criteria are then given. These conditions, ensuring the global exponential stability, are simpler and less conservative than some of the previous results. Finally, two numerical examples are given to illustrate the advantages of the obtained results.

Opis:
A single crystal of iron (II) carbohydrazide perchlorate [FeII (CHZ)3](ClO4)2 (FeCP), a novel, lead-free, energetic coordination compound, was synthesized and its structure determined by X-ray single crystal diffraction for the frst time. The crystal belongs to the monoclinic system P2(1)/n space group, with a = 1.0066(2) nm, b = 0.8458(2) nm, c = 2.1194(4) nm, β = 100.693(3)° and Z = 4. The central Fe(II) ion is coordinated to three bidentate carbohydrazide units through the carbonyl oxygen atom and an amino nitrogen atom, forming a six-coordinated, non-centrosymmetric complex cation. The thermal analyses by differential scanning calorimetry and thermogravimetry show that the onset temperature of thermal decomposition (152.7 °C) and the critical temperature of thermal explosion of FeCP (161.2 °C) are both much lower than those of other transition metal carbohydrazide perchlorate compounds, and also those of some other primary explosives in service. FeCP has a high enthalpy of combustion, as measured by oxygen bomb calorimetry. The impact, friction and fame sensitivity tests indicate that FeCP is extremely sensitive and hazardous. Unexpected explosions occurred even during the operational processes. In order to explore the intrinsic cause of these explosions, theoretical calculations of the orbital energies were performed based on DTF. These results reveal that the impact sensitivity is positively correlated with the energy gap between HOMO and LUMO: the smaller energy gap results in the higher impact sensitivity.

Tri(8-quinolinolato)iron complex immobilized onto SBA-15 catalyst has been synthesized through a stepwise procedure. The characterization results indicated that the BET surface area, total pore volume and average pore width decrease after stepwise modification of SBA-15, while the structure keeps intact. Catalytic tests showed that FeQ3-SBA-15 catalyzes the oxidation reaction well with 34.8% conversion of benzyl alcohol and 74.7% selectivity to benzaldehyde when water is used as the solvent after 1 h reaction. In addition, homogeneous catalyst tri(8-quinolinolato)iron exhibits very bad catalytic behavior using water as the solvent.

The electrohydraulic servo valve (EHSV) is widely used in many engineering fields. Its reliability is of great importance to the reliability and safety of entire servo control systems. With the aim of analyzing and evaluating reliability of EHSV, this paper firstly presents the physical structure and functional principle of EHSV. It is followed by the Failure Mode, Effects and Criticality Analysis (FMECA). From the analysis, the common cause failures (CCF) in the studied EHSV are identified. Lastly, a method that can quantitatively analyze reliability and failure rate of EHSV with considering the common cause failures is proposed. It is observed from the study that the failure rate of the EHSV with CCF is lower than the failure rate without considering CCF.
Serwozawory elektrohydrauliczne (EHSV) mają szerokie zastosowanie w wielu dziedzinach inżynierii. Ich niezawodność ma decydujące znaczenie dla niezawodności i bezpieczeństwa całych układów sterowania serwomechanizmami. W celu analizy i oceny niezawodności zaworów EHSV, w pracy przedstawiono najpierw ich budowę fizyczną i zasadę działania. Następnie przeprowadzono analizę przyczyn, skutków i krytyczności uszkodzeń (FMECA). Na podstawie tej analizy określono uszkodzenia zaworu EHSV spowodowane wspólną przyczyną (CCF). Wreszcie, zaproponowano metodę, za pomocą której można ilościowo analizować niezawodność i awaryjność EHSV z uwzględnieniem uszkodzeń spowodowanych wspólną przyczyną. Badania wykazały, że awaryjność EHSV przy uwzględnieniu CCF jest niższa niż w wypadku nieuwzględnienia CCF.
