Opis:
The global prevalence of diabetes mellitus (DM) is escalating, posing a significant challenge to human healthcare systems. In the present study, mineral and monosaccharide compositions, and antidiabetic potential of polysaccharide fractions separated from Centella asiatica (L.) leaves (CAL) were assessed. Initial single-factor experiments identified key extraction parameters, further optimized through response surface methodology (RSM) with optimal conditions determined as a liquid-to-solid ratio of 24.43:1 (mL/g), extraction time of 60.76 min, and a temperature of 83.31℃. Alcohol insoluble residue (AIR) extraction yielded three crude polysaccharide fractions (P50, P70, and P90). P50 was obtained with the highest yield (14.31%) and exhibited the highest content of total sugars and uronic acids (68.01 and 24.28 g/100 g, respectively). Calcium, magnesium, and manganese were dominant minerals in P50 and P70. Monosaccharide composition analysis via high-performance liquid chromatography (HPLC) demonstrated enrichment of galacturonic acid, galactose, and arabinose in P50 and P70. Furthermore, both fractions (P50 and P70) exhibited dose-dependent inhibitory effects on α-amylase and α-glucosidase. At 5 mg/mL, P50 caused 68.3% α-amylase inhibition and 62.3% α-glucosidase inhibition, while P70 triggered 46.8% and 34.1% inhibition, respectively. In conclusion, this study is the first to provide valuable insights into optimizing CAL extraction conditions. Potential utilization of polysaccharide fractions obtained from CAL as components for formulating functional foods can be explored in the future.

To develop a sensitive, specific, and rapid approach for the detection Getah virus (GETV), a set of primers targeting the conserved region of the E1 gene was created. The TaqMan-based real-time PCR method for GETV detection was developed by optimizing the reaction conditions. The method demonstrated excellent specificity, and amplification did not occur with the causative agents of all prevalent swine viral infections (CSFV, PRRSV, PRV, PEDV, PTV, and JEV), except GETV. Additionally, upon assessing the sensitivity of the method, the minimum detection limit for GETV was found to be 5.94 copies/μL, which is 10 times higher than that of the traditional PCR approach. Further, the intra- and inter-assay variation coefficients were less than 1%, demonstrating good repeatability. Moreover, GETV was found in 10 of the 20 field serum samples using real-time PCR but only in three of the samples using traditional PCR. Consequently, the first GETV TaqMan-based real-time PCR approach based on the E1 gene was developed for GETV pathogenic diagnoses, and this exhibited high specificity, sensitivity, and repeatability. This assay is practical for the pathogenic diagnosis and epidemiology of GETV.

Opis:
Fibril formation of proteins and peptides is associated with a large group of major human diseases, including Alzheimer’s disease, prion disorders, amyotrophic lateral sclerosis, type 2 diabetes, etc. Therefore, understanding the key factors that govern this process is of paramount importance. The fibrillogenesis of polypeptide chains depends on their intrinsic properties as well as on the external conditions. In this mini-review we discuss the relationship between fibril formation kinetics and the sequence, aromaticity, hydrophobicity, charge and population of the so called fibril-prone conformation in a monomer state. The higher the population, the faster is the fibril elongation and this dependence may be described by a single exponential function. This observation opens up a new way to understand the fibrillogenesis of bio-molecules at the monomer level. We will also discuss the influence of the environment with focus on the recently observed dual effect of crowders on the aggregation rat es of polypeptide chains.

Opis:
A proof of concept for using paper test as a suitable method in the production of monoclonal antibodies (MAbs) is reported. The paper test which detects antibodies against porcine circovirus type 2 (PCV2) using colloidal gold-labelled capsid protein as the antigen probe was applied exclusively in the screening of anti-PCV2 MAbs. It allowed the detection of 118 single cell clones within 30 min using naked eyes. MAbs with specific binding to authentic epitopes on the virus were selected using a blocking strategy in which the antibody was pre-incubated with PCV2 viral sample before applying to the test paper. Five hybridomas secreting MAbs against the capsid protein were obtained, with only three of them capable of binding to PCV2. The results were validated and confirmed using enzyme-linked immunosorbent assay and immunofluorescence assay. The paper test is simple, rapid, and independent on professional technicians and proves to be an excellent approach for the screening of MAbs against specific targets.

Opis:
Reverse flotation of collophanite at natural pH could significantly decrease the cost of pH regulators. In this study, isooctyl polyoxyethylene ether phosphate (AEP) was tested as a new surfactant in the reverse flotation of collophanite. Micro-flotation tests were conducted, and the adsorption mechanism of the new collector was analysed using X-ray photoelectron spectroscopy (XPS) and zeta potential analyses. The results of the flotation tests demonstrated that AEP could enable dolomite to float under natural pH (pH=7.2) and showed profound selectivity towards dolomite as opposed to fluorapatite. Based on the zeta potential and XPS results, the adsorption phenomena are mainly attributed to calcium active sites on both mineral surfaces. Dolomite possesses more magnesium active sites than fluorapatite, which tend to reinforce the interaction effect between AEP and dolomite. Furthermore, when compared to CO32- ions on the dolomite surface, PO43- ions on the fluorapatite surface tend to exhibit a stronger hindrance to the adsorption of AEP on the fluorapatite surface. This is attributed to their larger volumes and more charges on their surfaces, thereby causing a floatability difference between the two minerals.

The root system architecture (RSA) displays complex morphological characteristics because of diverse root growth behaviors. Recent studies have revealed that swarming behavior among roots is particularly important for RSA to adapt to environmental stimuli. However, few models are proposed to simulate RSA based on swarming behavior of roots. To analyze plasticity of RSA affected by swarming behavior, we propose viewing it as a swarm of single roots. A swarming behavior model is proposed by considering repulsion, alignment, and preference of individual single roots. Then, the swarming behavior model is integrated into a simple and generic RSA model (called ArchiSimple). Lastly, characteristics of RSA affected by swarming behavior model and non-swarming behavior model are compared and analyzed under three different virtual soil sets. The characteristics of RSA (such as primary root length, lateral root length, lateral roots, and resource uptake) are significantly promoted by swarming behavior. Root system distributions can also be greatly affected by swarming behavior. These results show that root foraging and exploration in soil can be regarded as collective behavior of individual single root.

Opis:
Conservation farming practices using the least soil disturbance and straw-return benefits the crop agronomic attributes and soil nutrient accumulation. The four-year (2016-2019) research was conducted under randomized complete blocks design to explore the agronomic benefit of conservation tillage practices on wheat yield performance and on soil fertility parameters. The two straw treatments consisted of wheat straw-return to the notilled soil and straw incorporation into the conventionally tilled soil. The two tillage treatments were the no-tillage and conventional tillage control. These conservative tillage treatments were compared with the conventional tillage control. In comparison with conventional tillage, the conservation management practices of no-tilled soil, conventionally tilled soil, and no-tillage notably increased the yield by an average of 33, 26, and 18% respectively. Moreover, conservative tillage practices improved the soil nitratenitrogen, ammonium nitrogen and carbon contents in the 0-30 cm soil layer by 12, 9, and 15% respectively over conventional tillage, averaged across conventionally tilled soil, no-tilled soil, and no-tillage. The overall distribution of soil nitrate-nitrogen, ammonium nitrogen, and carbon in the 0-30 cm soil layer with regard to conventionally tilled, no-tilled soil, and no-tillage was greater than conventional tillage, based on Principal Component Analysis. We concluded that conservation tillage practices could replace conventionally tilled practice with respect to productivity, soil mineral nitrogen, and carbon accumulation benefits.

Bulked segregant analysis combined with specific length amplified fragment sequencing techniques have been applied to determine the fine genetic mapping of fruit flesh thickness-related genes in cucumber. Herein, the Csa2M058670.1 gene was subjected to real time fluorescence quantitative PCR (qRT-PCR) and sequence analysis, indicating a strong correlation with cucumber fruit flesh thickness. Expression and characterisation of the Csa2M058670.1 gene were performed based on previous studies. The results of the fluorescence-based quantitative PCR showed that Csa2M058670.1 was expressed in all organs, but levels were highest in fruit peel, fruit flesh, and female flowers. Furthermore, Csa2M058670.1 expression was induced by abiotic stresses including drought, low temperature, and high salt. Domain analysis revealed that the protein encoded by Csa2M058670.1 possesses an SET (Su(var), Enhancern of zeste CE(z), and Trithorax) domain that may control cell division and differentiation. Therefore, we speculated that Csa2M058670.1 might affect fruit flesh thickness in cucumbers by influencing cell division.

In this study, the flotation separation of rutile from almandine using cationic surfactant Octadecyl amine polyoxyethylene ether (AC1815) as a new collector was investigated. The adsorption mechanism of AC1815 on rutile was illustrated through zeta potential measurement, infrared spectrum and XPS analyses. The flotation experiments demonstrated that AC1815 exhibited an excellent collecting ability and selectivity for rutile. The results of zeta potential measurements and XPS analysis indicated that more AC1815 was adsorbed on rutile surface instead of almandine. The adsorption mechanism of AC1815on rutile was mainly attributed to the electrostatic interaction between the positively charged molecules of AC1815 and the negatively charged rutile surface, and the hydrogen bonding between the protonated $≡NH^+$, $≡N$ group of AC1815 and $Ti-OH$ on rutile surface.

The enzyme strictosidine synthase (STR, EC: 4.3.3.2) plays a key role in the biosynthetic pathway of terpenoid indole alkaloid (TIA). It catalyzes the condensation of the tryptamine and secologanin to form 3α(S)-strictosidine, which is the common precursor of all TIAs. In this paper, a STR gene designated as DoSTR (GenBank: KX068707) was first cloned and characterized from Dendrobium officinale with rapid amplified cDNA ends method (RACE). DoSTR has a length of 1380bp with 1179bp open reading frame encoding 392 amino acids. BlastP analyses showed that its amino acid sequence was classified into Str_synth superfamily. qRT-PCR showed that DoSTR was expressed in all tissues tested, with a significantly higher level in flower and the lowest in stem. Four different treatments with MeJA, SA, ABA and AgNO₃, respectively, could induce the DoSTR expression to a different extent. And the effect of MeJA was the most obvious and transcript level of DoSTR induced by MeJA was 20.7 times greater than that of control at 48 hours after treatment. Furthermore, it was found that DoSTR was localized in vacuole through transient expression in tobacco. The characterization and expression of DoSTR can help in further studying the role of DoSTR in the biosynthesis of TIAs in D. officinale. This study may throw light on the alkaloid biosynthesis pathway of D. officinale.

In this research, sterile soil columns with different contents of biochar made from apple-tree residues (0, 1 and 5% w/w) at three levels of water filled pore space (40, 60, and 80%) were set up in the laboratory to study nitrous oxide diffusion and binding processes. The results indicated that nitrous oxide emission can be effectively mitigated at 5% biochar regardless of soil water content. However, 1% biochar stimulated nitrous oxide diffusion compared to the other biochar treatments, which was opposite to expectations due to the stronger aeration than adsorption effect, while 0% had a suppression effect between 1 and 5%. Nitrous oxide emissions increased with increasing water filled pore space due to concomitantly decreasing biochar tortuousity at high water content. The increase of nitrogen from 1.11 to 1.50% on the biochar surface in the 5% treatment, and from 1.11 to 1.46% in the 100% biochar treatment, suggested that the main abiotic mechanisms for mitigation of nitrous oxide emission is adsorption and subsequent reactions with C = C bonds on appletree biochar surfaces since C = O and C-O bonds both increased and C=C/C-C/C-H declined.
