Opis:
The present study was aimed to evaluate the activity of Achyranthes aspera against allergy caused by heavy metal potassium dichromate in albino mice. The potassium dichloromate as induced at a concentration of 20 mg/kg. body weight dissolved in distilled water for 10 days continuously. On 11th day, blood was drawn from mice for serum analysis of IgM and IgG to confirm allergy induction in mice. After allergy induction the mice received petroleum ether, chloroform and methanolic extracts of Achyranthes aspera given 3 doses at concentration of – 200 (low), 400 (medium) and 600 (high) mg/kg to mice by oral route for 7 days after allergy induction. On the 8th day, blood was drawn from mice for serum analysis of IgM and IgG in determine the effect of extracts on serum Titre of mice blood.

The present study reports potential activities like cytotoxic, genotoxic and apoptotic activity of phenolic compound 4-(4-(3,4-dimethoxyphenyl)hexahydrofuro[3,4-c]furan-1-yl)-2-methoxyphenyl acetate isolated from methanolic extract of Pandanus odorattissimus. The compound showed a significant cytotoxic effect on Human Leukemia 60 (HL-60) cell line. Exposure of the compound reduced the viability of HL-60 cells after 12, 24 and 48 hours, the compound exerted a significant cytotoxic effect on HL-60 cells. The compound also induced significant DNA damage. The results of comet assay with pattern of the HL-60 cells has shown an intact head and complete absence of DNA fragments in the form of tail suggesting that the doses are not genototoxic. The phenolic compound at concentration of 20 µg/mL, showed an increase in percentage tail of DNA upto 6.21 units when compared to control 5.35. Although all of the compound induced significant DNA damage it induced apoptotic in the middle level and led to significant level. Apoptotic effect of compound on HL-60 cells after 72 h increased. Furthermore, the compound induced slight necrosis in HL-60 cells. Although the compound induced significant DNA damage, it induced apoptotic at the middle level. Apoptotic effect of compound on HL cells after 72 hrs increased level, furthermore the compound induced slight necrosis in HL-60 cells.

Bioassay guided chromatographic purification of methanolic extract of Pandanus odoratissium leaves afforded new phenolic compound. The isolated compound was identified as 4-(4-(3,4-dimethoxyphenyl) hexahyddrofuro[3,4-c]furan-l-yl)-2-methoxyphenyl acetate by the spectroscope methods. The puropose of this investigation was to check the effect of the phenolic compound 4-(4-(3, 4-dimethoxyphenyl)hexahyddrofuro[3,4-c]furan-l-yl)-2-methoxyphenyl acetate isolated pure compound, the human leukemia (HL-60) cell line as model to evaluate the cytoxicity of compound on the MTT assay. The results obtained from the parameter indicates the compound has significantly reduced the viability of HL-60 cells shows LD50 values of 21.24 ± 0.5 μg/mL, 10.25 ± 0.3 μg/mL and 5.2 ± 0.6 μg/mL, upon 12, 24 and 48 hours of exposure respective indicates a dose and dependent response relationship. The present study indicate the compound is highly cytotoxic to human leukemia (HL-60) cells supports it as an effective therapeutic agent against leukemia.

The presence of phytochemical compounds in the plant Achyranthes aspera was evaluated in root using different solvents such as Petroleum ether, Chloroform, Methanol In the present study, the preliminary phytochemical screening of leaf and flower extracts of Achyranthes aspera showed the presence of phytochemicals such as alkaloids, carbohydrates, flavonoids, proteins, aminoacids, tannins, phenols, steroids, glycosides and saponins.
