Opis:
Plants must continually calibrate their growth in response to the environment throughout their whole life cycle. Revealing the regularity of plant early growth and development is of great significance to plant genetic modification. It was previously demonstrated that loss of two key light signaling transcription factors, FHY3 and FAR1, can cause a stunted stature in the plant adult stage, and numerous defense response genes can be continuously activated. In this study, we performed a time-course transcriptome analysis of the early 4 weeks of leaf samples from wild plants and their fhy3 and far1 transcription factors. By comparative transcriptome analysis, we found that during the early 4 weeks of plant growth, plants primarily promoted morphogenesis by organizing their microtubules in the second week. In the third week, plants began to trigger large- scale defense responses to resist various external stresses. In the fourth week, increased photosynthetic efficiency promoted rapid biomass accumulation. Weighted gene coexpression network analysis of FHY3 and FAR1 revealed that the two light signaling transcription factors may be originally involved in the regulation of genes during embryonic development, and in the later growth stage, they might regulate gene expression of some defense-related genes to balance plant growth and immunity. Remarkably, our yeast two-hybrid and bimolecular fluorescence complementation experiments showed that FAR1 interacts with the immune signaling factor EDS1. Taken together, this study demonstrates the major biological processes occurring during the early 4 weeks of plant growth. The light signaling transcription factors, FHY3 and FAR1, may integrate light signals with immune signals to widely regulate plant growth by directly interacting with EDS1.

Somatic cell nuclear transfer (SCNT) is currently the most efficient and precise method to generate genetically tailored pig models for both agricultural and biomedical research. However, its efficiency is crucially dependent on the source of nuclear donor cells. In this study, we compared the cloning efficiency by using three lines of donor cells that are derived from fetal, newborn and adult fibroblasts of Chinese indigenous Meishan pig. We showed that cleavage rate and blastocyst formation rate of the reconstructed embryos were not significantly different between the fetal (80.7% and 15.6%) and newborn ear skin (77.5% and 12.3%) fibroblast groups (p>0.05), but in both groups these indices were significantly higher than that found in the adult ear skin (70.5% and 8.8%; p<0.05). Reconstructed embryos derived from fetal, newborn, and adult ear skin fibroblasts were transferred to four surrogates, respectively. For the fetal, newborn, and adult ear skin fibroblasts, the number of pregnancies were two (50.0%), two (50.0%), and one (25.0%), respectively, and the number of deliveries were two (50.0%), one (25.0%), and zero (0.0%), respectively. Seven and two cloned piglets were obtained from the fetal and newborn ear skin fibroblasts respectively, while no piglets were obtained from the adult ear skin fibroblasts. Two cloned piglets from the newborn ear skin fibroblasts died shortly after birth because of neonatal asphyxia caused by dystocia. The birth weights of the piglets derived from the fetal and newborn ear skin fibroblasts were 1230.5 and 1310.0g, respectively, which were statistically insignificant (p>0.05), but both were significantly higher than that of the control groups (p<0.05). Microsatellite analyses demonstrated that the genotypes of all cloned piglets were identical to their donor cells. Therefore, cloned pigs were successfully produced using two sources of donor cells isolated from the fetal and newborn ear skin fibroblasts of Meishan piglet, and indicating a better cloning efficiency than that obtained from adult fibroblasts. We concluded that the nuclear donor cell lines have significant impact on the developmental competence of cloned embryos as well as on the cloning efficiency of Meishan pig.

$Fe_{68}Zr_{20}B_{12}$ amorphous alloy prepared by mechanical alloying technique of 60 h of duration was annealed at different temperatures (420-720°C) during 1 h. Microstructure and magnetic property of as-milled and annealed alloy were investigated by X-ray diffraction, transmission electron microscopy, the Mössbauer spectroscopy and vibrating sample magnetometer techniques. $Fe_{68}Zr_{20}B_{12}$ powders milled during 60 h exhibit amorphous character. After annealing at 420°C, α-Fe phase precipitates from amorphous matrix. Transmission electron microscopy analysis shows the nucleation and growth of α-Fe grains. The Mössbauer results of the annealed alloy at 670°C show that the $Fe_3B$ phase precipitates already and there is still remaining amorphous phase. Coercivity increases with increasing annealing temperature, which is closely related to the microstructures after annealing.

Thermal properties, microstructure and magnetic properties of $Fe_{79}Zr_{9}B_{12},$ $Fe_{76}Zr_{9}B_{15},$ $Fe_{39.5}Co_{39.5}Zr_{9}B_{12}$ and $Fe_{38}Co_{38}Zr_{9}B_{15}$ alloys were investigated by differential thermal analysis, X-ray diffraction and vibrating sample magnetometer. High Co addition has a significant impact on the properties of alloys. High Co addition increases the main crystallization peak temperature and the crystallization activation energy. Moreover, high Co addition changes the crystallization products and influences the crystallization processes of $Fe_{79}Zr_{9}B_{12}$ and $Fe_{76}Zr_{9}B_{15}$ alloys. The specific saturation magnetization $(M_{s})$ increases and coercivity $(H_{c})$ decreases by high Co additions.

Thermal behavior and microstructure of $Fe_{81 - x}Co_{x}Zr_7Nb_2B_{10}$ (x = 2, 4, 6) alloys were investigated by differential thermal analysis and X-ray diffraction. Both the supercooled liguid region Δ $T_{x}$ and the first crystallization peak temperature $T_{p1}$ are the lowest and the span Δ $T_{p}$ between $T_{p1}$ and $T_{p2}$ is the highest when 4 at.% Fe is substituted by Co. The crystallization activation energy $E_{p1}$ for $Fe_{75}Co_6Zr_7Nb_2B_{10}$ alloy is the minimum for the heating rates ν = 5-20 K/min. $E_{p1}$ for $Fe_{77}Co_4Zr_7Nb_2B_{10}$ alloy is the minimum for the heating rates ν = 20 - 50 K/min. The crystallization processes of $Fe_{81 - x}Co_{x}Zr_7Nb_2B_{10}$ (x = 2, 4, 6) amorphous alloys are similar on the whole, which is as follows: amorphous → amorphous + α-Fe(Co) → α-Fe(Co) + $Fe_3Zr$ + $Fe_2Nb_{0.4}Zr_{0.6}$.

The main mining coal seams 71 and 72 of Huaibei mine area show a relation between splitting and merging. Under the slicing mining circumstance of the splitting area, it is the key problem to face the mining area to ensure the safety of the lower 72 coal mining. Based on face 7225 of Xutuan Coal Mine the influence of 71 coal seam mining on the coal seam 72 was analyzed by the FLAC3D. The law of stress propagation in the floor, failure extent and movement during the mining of coal seam 71 and coal seam 72 were studied by the numerical simulation; it provides the basis for roof management and support selection of 72 coal seam mining.
Główne pokłady węglowe nr 71 i 72 kopalni Huaibei wykazują cechy zarówno rozszczepienia, jak i scalania. Podczas wybierania warstwowego na obszarze rozszczepienia głównym problemem jest zapewnienie bezpieczeństwa dolnego pokładu nr 72. Na przykładzie przodka 7225 kopalni Xutuan przeanalizowano wpływ wydobycia w pokładzie nr 71 na pokład nr 72 przy użyciu programu FLAC3D. Propagację fal naprężeń w spągu, zakres pęknięcia i przesunięcia podczas wybierania pokładu węgla nr 71 i 72 zbadano za pomocą symulacji numerycznej, co daje podstawy do lepszej kontroli stropu i do wyboru obudowy pod wybieranie pokładu węgla nr 72.
