Since 1901, the Nobel Prize has been awarded to scientists who have made the most important discoveries for the benefit of humanity. The 2017 Nobel Prize in Physiology or Medicine was awarded jointly to Jeffrey C. Hall, Michael Rosbash and Michael W. Young “for their discoveries of molecular mechanisms controlling the circadian rhythm.” It may be surprising to learn that those three scientists dedicated their entire careers to research on the fruit fly, Drosophila melanogaster. However, as their studies progressed, it became increasingly clear that the mechanism of the biological clock that they discovered in Drosophila is very similar to a timekeeping mechanism present in mammals, including humans. Through interdisciplinary work between scientists performing basic research on model organisms and doctors working in medical schools, we have learned over time that daily rhythms support human health while disruption of these rhythms is associated with a range of pathological disorders such as cardiovascular problems, metabolic, neurological, and many other diseases. This short review will highlight critical milestones on the way to understanding biological clocks, focusing on the roles played by the three Nobel Prize winners.

The growth and saponin accumulation were measured in two lines of transgenic hairy roots of Platycodon grandiflorum, Pl 6 and Pl 17, cultured for 8 weeks in 250-ml shake flasks containing 50 ml of hormone-free woody plant medium supplemented with 40 g/l sucrose and in the Pl 17 line cultured for 12 weeks in a 5-l mist bioreactor containing 1.5 l of the same medium. With both methods, the growth of transgenic hairy roots was assessed as both fresh and dry weight and the biomass growth was correlated with the conductivity and sucrose uptake. The accumulation of saponins was measured and compared with that in roots derived from the field cultivation. The saponin concentrations were significantly higher in the two hairy root lines cultured in shake flasks [6.92 g/100 g d.w. (g%) and 5.82 g% in Pl 6 and Pl 17, respectively] and the line cultured in the bioreactor (5.93 g%) than in the roots derived from the field cultivation (4.02 g%). The results suggest that cultures of P. grandiflorum hairy roots may be a valuable source for obtaining saponins.

Mössbauer spectroscopy is not routinely used for the determination of the concentration of iron. However, as this method does not need any pre-treatment of samples before measurements, it may be of extreme importance for the assessment of iron in samples, which can then be used for further investigations. Biological samples are a good example, however, as the concentrations of iron are very low in these, it is important to exclude possible artefacts from the background spectrum related to iron present in the counter and cryostat windows. The aim of this study was to compare two methods of determination of the amounts of iron in investigated sample: one, in which the background spectrum was subtracted from the sample spectrum measured, and the other, in which the obtained non-elaborated spectrum was fitted with two doublets – a doublet for the measured sample and a doublet for the background spectrum. Three samples containing known amounts of natural iron (400, 800 and 1600 µg) and a sample of lyophilized human brain tissue obtained from globus pallidus were assessed. Both methods led to the creation of a very good calibration curve with a correlation coefficient of 0.99. Although both methods gave similar results for the concentration of iron in the sample, the subtraction of the background spectrum had a significantly lower error of the final result.

We describe experiments which study static and dynamic aspects of Mn spin organization in magnetic semiconductor superlattices and quantum wells. Neutron diffraction studies of ZnSe/MnSe and ZnTe/MnSe superlattices show how static Mn spin organization in these antiferromagnetic layers may be modified by strain and lowered dimensionality. We also use a novel form of femtosecond magnetic spectroscopy to examine magnetic polaron dynamics in diluted magnetic semiconductor quantum wells, and show how the organization of Mn spins by a spin-polarized carrier population may be directly viewed in the time domain.
