Opis:
The 85-m Brushy Basin Member of the Morrison Formation in western Colorado, USA, comprises dinosaur-bearing sandstones (architectural element CHR and CH), crevasse-splay deposits and minor levee deposits (architectural element CS), mudstones, marlstones, altered ash beds and minor limestones as well as caliche paleosols and noncalcareous paleosols (architectural element FF). Channel sandstones occur at five stratigraphic levels at Trail Through Time (TT), eleven levels at Fruita Paleontological Research Area (FP), and at five levels at Echo Canyon (EC). River-channel sandstones hosted by floodplain mudstones tend to have cut down to resistant caliche paleosols. Depositional facies and architectural element analysis show that the rivers were low gradient, mainly anastomosing, with perennial flow, but seasonal with “flashy” peaks in discharge. Dinosaur bone accumulations are found in some floodplain ponds. Isolated bones are present in anastomosing channel sandstones at TT and in channel sandstone 2 at EC. At FP, major accumulations of bones were rapidly buried in the deep pools at three bends in the meandering river resulting in the formation of channel sandstone 2. There is no evidence for a large lacustrine or playa system at the three localities.

Opis:
Study of the pedogenic features of the Upper Jurassic Morrison Formation in western Colorado, USA, shows a clear difference in the types of paleosols between the strata of the lower and upper Brushy Basin Member. Lower Brushy Basin paleosols are mostly calcareous Aridisols with Stage I through Stage III calcrete Bk horizons, abundant root traces, occasional vertic features, but only rarely with ochric epipedons. Upper Brushy Basin paleosols are mainly thicker and commonly display ochric epipedons and well-developed Bt and Bw horizons. We assign these paleosols to the order Inceptisol. Limestones occur in the Brushy Basin Member and include both uniformly micritic limestones and limestones with strongly brecciated textures. The former contain sparse body fossils and charophyte debris, while the latter are characterized by clotted-peloidal fabrics with circumgranular cracking and silica replacement. We interpret these limestones as the deposits of carbonate in small water bodies on a low-gradient flood plain, with the textures resulting from pedogenic reworking of the carbonate sediment. We find no evidence for the presence of extensive lacustrine or wetlands (Lake T’oo’dichi’) deposits in the study area. The paleoclimate suggested by all of these features is strongly seasonal, but subject to variations on orbital (precessional and higher) timescales causing intervals of semi-aridity during weaker monsoons, to alternate with sub-humid periods during stronger monsoons. The apparent long-term change in climate during Brushy Basin deposition potentially resulted from northward drift of North America.

Opis:
The aim of the study was to optimize conditions for producing Salmonella Enteritidis recombinant heat shock protein 60 (rHsp60). Seven Escherichia coli host strains (Rosetta, Turner, C41, C43, Origami, BL21pLys, Rosetta pLys) were transformed by a recombinant plasmid containing Hsp60 gene from Salmonella Enteritidis, and then cultured and induced by isopropyl-β-D-thiogalactopyranoside (IPTG). The highest S. Enteritidis rHsp60 yield was obtained using E. coli strain C41. Induction of this strain using IPTG allowed the yield 400 μg of S. Enteritidis Hsp60 protein/2L of culture, but by autoinduction the yield exceeded 800 μg/2L.
