Opis:
Inhibition of heme oxygenase-1 (HO-1, encoded by HMOX1), a cytoprotective, anti-apoptotic and anti-inflammatory enzyme, may serve as a valuable therapy in various pathophysiological processes, including tumorigenesis. We compared the effect of chemical inhibitors - metalloporphyrins, with genetic tools - shRNA and CRISPR/Cas9 systems, to knock-down (KD)/knock-out (KO) HO-1 expression/activity. 293T cells were incubated with metalloporphyrins, tin and zinc protoporphyrins (SnPPIX and ZnPPIX, respectively) or were either transduced with lentiviral vectors encoding different shRNA sequences against HO-1 or were modified by CRISPR/Cas9 system targeting HMOX1. Metalloporphyrins decreased HO activity but concomitantly strongly induced HO-1 mRNA and protein in 293T cells. On the other hand, only slight basal HO-1 inhibition in shRNA KD 293T cell lines was confirmed on mRNA and protein level with no significant effect on enzyme activity. Nevertheless, silencing effect was much stronger when CRISPR/Cas9-mediated knock-out was performed. Most of the clones harboring mutations within HMOX1 locus did not express HO-1 protein and failed to increase bilirubin concentration after hemin stimulation. Furthermore, CRISPR/Cas9-mediated HO-1 depletion decreased 293T viability, growth, clonogenic potential and increased sensitivity to H2O2 treatment. In summary, we have shown that not all technologies can be used for inhibition of HO activity in vitro with the same efficiency. In our hands, the most potent and comprehensible results can be obtained using genetic tools, especially CRISPR/Cas9 approach.

The aim of this article is to show the functions which music can perform in radio report-age. The analysis is based on documentaries created over the past decade by famous Polish reporters, such as Katarzyna Michalak, Cezary Galek, Julia Prus and Hanna Bo-gorya-Zakrzewska together with Ernest Zozuń. Fragments of their works were used as the exemplification of the different roles which soundtrack can perform, such as: to illus-trate, to describe, as well as to complete narration, composition or the characteristics of a situation, people, etc.

Despite the fact that radio documentaries broadcast mainly strong emotions, the news valu of this genre is also an essential feature. Therefore, it is necessary to describe the potential of such works to expand knowledge about the surrounding world which could be significant for the listeners. The aim of this article is to present theses put forward by scholars with regard to radio documentaries. Researchers’ analyses are related to the genre’s potential to broaden people’s minds and influence the recipient’s perception.

The article presents the form of accompanying musical programming in commercial radio broadcasters in Poland. It examines the questions of the place of entertainment in this kind of production, based on examples from a variety of stations, such as Radio Zet, RMF FM, Radio Eska, Radio Złote Przeboje or Vox FM. The text also refers to hybrid forms of media entertainment such as edutainment or democratainment. In the conclusion, this conglomerate radio genre is treated as a polyphonic, mosaic form, distinguished, though, by its dynamism and the nature of the programmes broadcast at the morning and afternoon peak hours of radio audiences.

The media environment is an extremely variable universe where every now and again we can observe the emergence of new phenomena. Many of those form through blending of often rather different and distant areas. As a result, there emerge hybrid forms, which are not entirely established or completely defined. This article is focussed on those kinds of mixed types based on media entertainment, e.g. infotainment, edutainment, politainment, politicotainment, docutainment etc., which can be observed in the means of mass communication. The goal of the study was to define their essence, their distinctive features, and to indicate the place of those hybrid forms within the media discourses being carried on today.

The article is an attempt to enter scientific reflection relating to radio reportage in the field of sound studies, especially in one of its original elements — the theory of soundscape created by Raymond Murray Schafer in the 1960s. It starts with the basic terminological solutions in the field of soundscape and the audial form of reportage, then juxtaposes them with each other and indicates the possibilities of their mutual penetration in both scientific and media fields. It presents the common features of a radio documentary and a soundscape, e.g. focus on reality, attention to sound, specific narration, the use of the oral history method etc. This paper also shows the difference between these two forms such as a greater level of creativity in the case of reportage in combination with the “raw” naturalness of the soundscape. It also highlights how the idea of acoustic ecology can use radio reportage to disseminate its assumptions. Finally, it postulates the inclusion of this audio genre as an element of the soundscape to a harmonious hi-fi space and considers as justified to set together the experiences of both the discussed phenomena on theoretical as well as practical grounds, and to continue the reflection as sketched out in this article.

Celem artykułu jest omówienie dokumentów radiowych dotyczących migrantów wojennych, które pojawiły się w polskich stacjach w pierwszych dwóch tygodniach konfliktu rosyjsko-ukraińskiego. Metodę badawczą stanowi analiza treści i artystycznych środków wyrazu dźwiękowego oraz analiza porównawcza. Dokumenty uchodźcze są dość jednorodne i kreślą polifoniczny portret migrantów jako społeczności, z której jednak można wyłonić indywidualne historie przesycone konkretną ludzką tragedią. Większość analizowanych prac skupia się na temacie, przywiązując nieco mniejszą wagę do formalnej strony, przez co tylko pojedyncze projekty można określić mianem reportaży artystycznych.
The goal of the following text is to discuss radio documentaries concerning war emigrants that appeared in Polish stations during the first two weeks of the conflict between Russia and Ukraine. The research method used in the article is the analysis of the content and artistic means of audio expression as well as comparative analysis. Refugee documentaries are quite homogeneous and paint a polyphonic portrait of emigrants as a community, from which, however, it is possible to emerge individual stories saturated with concrete human tragedy. Most of the analyzed works focus on the subject, while attaching a little less importance to the formal side of the program, as a result only individual projects can be described as artistic reportages.

Celem artykułu jest pokazanie, jak o zdrowiu psychicznym mam pisze psycholog Aleksandra Sileńska, znana w sieci jako Mama Terapeutka. Przedmiotem badania stały się jej wpisy z serwisu Facebook, wykorzystano analizę zawartości oraz wybrane narzędzia z zakresu retoryki – kategorię toposu oraz triadę etos, patos, logos. Zaobserwowano, że autorka korzysta w swoich publikacjach głównie ze strategii patosowych oraz etosowych, a propagowany przez nią model Mamy Zaopiekowanej stanowi – w wielu aspektach – opozycję dla toposu Matki Polki, w którym pomijano kwestię zdrowia psychicznego mam.
This article aims to explore how psychologist Aleksandra Sileńska, known online as Mom Therapist, addresses the mental health of mothers in her Facebook posts. Through content analysis and rhetorical tools, including the categories of topos and the triad of ethos, pathos, and logos, the study examines Sileńska’s communication strategies. The findings reveal that she predominantly employs pathos and ethos in her posts. Additionally, her promotion of the Cared-for Mom model contrasts sharply with the traditional Polish Mother topos, which often overlooks the mental health of mothers.

Celem badania jest nakreślenie obrazu uchodźców wyłaniającego się z reportaży radiowych stworzonych w pierwszych sześciu tygodniach wojny w Ukrainie, tj. w okresie od 24 lutego do 6 kwietnia 2022 r. Materiał do analizy objął 96 reportaży radiowych zrealizowanych przez polskie rozgłośnie radiowe: publiczne, komercyjne i internetowe. W badaniu wykorzystano metodę analizy zawartości do wyłonienia reportaży uchodźczych z grona wszystkich dokumentów radiowych odnoszących się do wojny w Ukrainie. Następnie dokonano analizy porównawczej wybranych audycji. Ustalono, że najczęściej portretowanymi migrantami były zatroskane o los najbliższych, walczące o lepsze jutro kobiety, matki. Czasem pojawiały się także dzieci, choć najczęściej nie na pierwszym planie. Wśród wojennych migrantów zdarzały się również zwierzęta i mężczyźni – starsi lub ojcowie, członkowie wielodzietnych rodzin. Wojenni migranci pokazywani byli z różnych perspektyw, a sposób ich obrazowania różnił się w zależności od czasu realizacji reportażu.
The aim of the study is to outline the picture of refugees emerging from radio documentaries created in the first six weeks of the war in Ukraine, i.e. between February 24 and April 6, 2022. The material for the analysis included 96 radio documentaries made by Polish radio stations – public, commercial and Internet. The study used the content analysis method to select refugee documentaries from among all radio documentaries related to the war in Ukraine. Then, a textual analysis and comparative analysis of selected programmes was carried out. It was established that the most frequently portrayed migrants were women, mothers concerned about the fate of their loved ones, fighting for a better tomorrow. Sometimes there were also children, although most often not in the foreground. Among the war migrants there were also animals and men – elderly or fathers, members of large families. War migrants were shown from different perspectives, and the way they were portrayed differed depending on the time of reportage.

Commercial radio stations – from a marketing point of view – consider delivering entertainment to listeners to be their main aim. In order to carry this out, they use not only programmes broadcast daily, but also opportunities provided by the phenomenon of convergence. The subject of this article is the types of convergence seen in entertainment created by radio ma kers. These are discussed based on t he example of Radio Eska – a formatted radio station, targeted at young listeners and directing its message to their needs.

Reportaż radiowy to gatunek ksenochtoniczny, przeniesiony z gruntu literatury, przystosowany do możliwości medium audialnego. Wielokrotnie szereg dyskusji budziła kwestia jego statusu i pogranicznego charakteru. Badacze próbowali rozstrzygnąć, czy reportaż jest tworem czysto dziennikarskim, publicystycznym, czy też należałoby go zaliczyć do dzieł sztuki. Podejmuje on bowiem najczęściej ważne społecznie kwestie, ale niejednokrotnie przedstawia je z wykorzystaniem środków artystycznego wyrazu. Artykuł stanowi próbę zabrania głosu w tej sprawie ‒ uwzględnia nie tylko stanowiska badaczy, lecz także opinie odbiorców. Pokazuje historyczne, społeczne i kulturowe aspekty ewolucji tego gatunku, a także wyniki badań ankietowych, przeprowadzonych na reprezentatywnej grupie słuchaczy, dotyczące procesu percepcji reportaży radiowych – jego przebiegu i skutków oraz wpływu na kształtowanie kompetencji kulturowych, społecznych czy komunikacyjnych.
The documentary is originally a literary genre. It has been adapted to the possibilities and the specificity of the acoustic media. Frequently, it has evocated discussions connected with its status and the character of its boundaries. Researchers tried to decide if it is a typically journalistic or artistic creation. The documentary usually takes up socially important issues but often shows them using artistic ways of presentation. This article points at different aspects of this case using researchers‟ opinions and radio listeners as well as the historic, social and cultural background of the development of this genre. The perception of radio documentaries is also mentioned in order to show its influence on cultural, social and communicative competence.

The situation on the contemporary media market, including radio, has led to the building of a radio network’s value becoming more than just the result of an interesting programme offer that meets listeners’ needs. Listeners also desire other elements that strengthen their attachment to a particular brand. For this reason, radio broadcasters quite often prepare for listeners different forms of entertainment that take place outside the radio studio; examples include summer concert tours, trips to well-known health resorts during winter and summer holidays, New Year’s Eve parties, special-occasion concerts, football matches, etc. This article is a review of entertainment offers that are part of a strategy of interaction with the audience outside of the radio studio and an attempt to evaluate their influence on building the market position of a radio brand.
