It has been found that alpha-chymotrypsin inhibitor isolated from Ascaris lumbricoides suum acts embryotoxically on the Leghorn chicken embryo. An increase in embryo mortality with increasing dose of the inhibitor has been observed after its administration into the yolk sac on day 4, 8 or 13 of incubation. There is a linear interrelationship between the logarithm of the dose of alpha-chymotrypsin inhibitor and the mortality of chickens. The LD50 values foralpha-chymotrypsin inhibitor increased for injections performed at later stages of embryo development. A significant decrease of mean mass of chicks injected with alpha-chymotrypsin inhibitor in comparison with control groups was observed. There was a more frequent occurrence of developmental abnormalities and pathological changes in groups of hatched chicks which received the Ascaris inhibitor.

Opis:
Administration of the Ascaris tegumental homogenate (0.6-1.2 g of Ascaris proteins/kg/day) at a early stage of organogenesis (5-9 days of pregnancy) had a harmful effect upon the developing mouse fetuses. It has been found that injection of the homogenate did not delay or prevent implantation, but causes a high rate of intrauterine deaths. The Ascaris homogenate significantly decreased the number of live fetuses per litter, increased the frequency of litter resorption, produced a delay in bone formation and induced pathological changes of fetal organs and tissues. The congenital malformations were noted in fetuses after injection of higher doses of Ascaris homogenate (exencephaly, craniomeningocele and intemal hydrocephalus). No malformations were noted in control groups and after injection of minimum dose of the homogenate. The symptoms that occurred after administration of the tegumental homogenate to pregnant mice included: decreased body weight gain (p<0.001) as compared to controls, vaginal hemorrhage, intrauterine resorption of litter and mortality. These signs suggest that the Ascaris homogenate causes matemal toxicity.

Administration intraperitoneally of the Ascaris suum extract - ASE-(0.6-1.4 g of Ascaris proteins/kg/day) at a late stage of organogenesis (8-12 days of gestation) disturbed course of mouse pregnancy. lt has been found that injections of higher doses of ASE to pregnant mice caused the symptoms manifesting maternal toxicity (decreased body weight gain /p < 0.001/ as compared to control, intrauterine resorption of litter, vaginal hemorrhages, female mortality and altered behaviour). There is a linear interrelationship between the logarithm of the dose of ASE and mortality of pregnant mice. The DL₅₀ value of Ascaris proteins for pregnant mice was 1.02 g/kg/day (confidence interval 0.97-1.07 g/kg/day). ASE exerted embryotoxic effects: significantly decreased the number of surviving fetuses per litter and the mean body weight of fetuses, increased the number of fetal resorptions.

According to literature of the problem, the disturbances of reproductiveness of human in various parasitic infections are discussed. The mechanisms disturbing the proper functioning of the hosts's reproductive system are not sufficiently understood. They may remain in association not only with a direct or indirect effect of the parasite on the host's endocrinal glands and sexual organs, but also with the host's overall reaction to stress or toxic influences. Parasite's activity may lead to abortion or a significant decrease in fetal body weight. Embryo or fetus may be also damaged in an intrauterine infection.

In this study, maternal toxicity and developmental effects of exposure to Ascaris trypsin inhibitor were evaluated in mice. Pregnant BALB/c females were injected intraperitoneally by Ascaris inhibitor /AIT/ at 200, 300, and 400 mg/kg body weight/day, on days 12 to 15 of gestation (stage of fetal development). At day 19 of pregnancy, uterine contents were inspected for implantation sites, early resorptions (moles), living fetuses and dead fetuses. The living fetuses were weighed and examined for external, internal and skeletal abnormalities. The results showed that AIT induced maternal toxicity, evidenced by maternal deaths, abortions, bleeding from uterus and reduced body weight gain as compared to control (p <0.01). There were no differences between the control group and the rest of all groups investigated for total implantation sites and early resorptions. Fetotoxicity was observed as shown by the decrease in the number of living fetuses and mean fetal weight, a high rate of intrauterine fetal deaths, delayed skeletal ossification, occurrence of pathological changes of fetal organs and tissues. Only one type of congenital malformations (hydronephrosis) was noted in fetuses after injection of higher doses of AIT.

BALB/c mice were given daily doses of 40-80 mg of Ascaris chymotrypsin inhibitor /AChl/ per kg body weight from 12" until 15" day of gestation (stage of fetal development). It has been found that injection of AChI disturbed the course of pregnancy (bleeding from uterus, abortions, decreased body weight gain as compared to control /p < 0.05/). AChI exhibited embryotoxic effects (a high rate of intrauterine deaths, decreased number of living fetuses and mean fetal weight, delayed skeletal ossification, induced pathological changes of fetal organs and tissues). Congenital malformation (hydronephrosis) was noted in fetuses after injection of higher doses of the chymotrypsin inhibitor from Ascaris.

It bas been found that trypsin and α-chymotrypsin inhibitors isolated from Ascaris suum act embryotoxically and teratogenically on White Leghorn clucken embryos. Mortatity rate for the chicken embryos on day 15 of incubation was 45.0 ± 3.5% after injection of trypsin inbibitor and 44.0 ± 3.5% after adminstration α-chymotrypsin inbibitor. Gross examination of surviving embryos and their dissection revealed pathological changes (abdominal dropsy, umbilical hernia, subcutaneous oedema, hemoperitoneum, hemopericardium), symptoms indicating retardation in growth (lack of down, retarded ossification of long bones, decreased mean body weight) as well as malformations (schistocelia, micrognathia, cyclopia, crossed beak, cranial deformities) after injection of inhibitors from Ascaris. The highest incidence of embryos with pathological changes and malformations was found after administralion of α-chymotrypsin inbibitor. The most commonly occurring abnormality was schistocelia (21.4 ± 3.88%). Growth malformations were not found in the control groups. The trypsin and α-chymotrypsin inhibitors present in Ascaris homogenate have a significant disturbing effect on the development of the chicken embryo.

Opis:
Recent literature data suggests that parasitic and fungal diseases, which pose a threat to both human and animal health, remain a clinical, diagnostic and therapeutic problem. Attention is increasingly paid to the role played by natural microbiota in maintaining homeostasis in humans. A particular emphasis is placed on the possibility of manipulating the human microbiota (permanent, transient, pathogenic) and macrobiota (e.g., Trichuris suis) to support the treatment of selected diseases such as Crohn’s disease, obesity, diabetes and cancer. Emphasis is placed on important medical species whose infections not only impair health but can also be life threatening, such as Plasmodium falciparum, Echinococcus multilocularis and Baylisascaris procyonis, which expand into areas which have so far been uninhabited. This article also presents the epidemiology, diagnosis and treatment of opportunistic parasitoses imported from the tropics, which spread across large groups of people through human-to-human transmission (Enterobius vermicularis, Sarcoptes scabiei). It also discusses the problem of environmentally-conditioned parasitoses, particularly their etiological factors associated with food contaminated with invasive forms (Trichinella sp., Toxoplasma gondii). The analysis also concerns the presence of developmental forms of geohelminths (Toxocara sp.) and ectoparasites (ticks), which are vectors of serious human diseases (Lyme borreliosis, anaplasmosis, babesiosis), in the environment. Mycological topics contains rare cases of mycoses environmentally conditioned (CNS aspergillosis) and transmissions of these pathogens in a population of hospitalized individuals, as well as seeking new methods used to treat mycoses.

Administration intraperitoneally of the Ascaris α-chymotrypsin inhibitor (40-300 mg/kg/day.) at a late stage of organogenesis (8-12 days of gestation) disturbed course of mouse pregnancy. The low doses of α-chymotrypsin inhibitor (40-80 mg/kg/day) significantly decreased the number of live fetuses per litter, increased the number of fetal resorptions. The symptoms of matemal toxicity that occurred after administration of the highest doses of the inhibitor (80-300 mg/kg/day) to pregnant mice included: decreased body weight gain as compared to control, vaginal hemorrhage, intrauterine resorption of litters, abortions, altered behaviour of animals immediately after injection and death. There is a linear interrelationship between the logarithm of the doses of the inhibitor and mortality of pregnant mice. The DL50 value of the inhibitor for female was 116 mg/kg /day (confidence interval: 95.5-140.0 mg/kg/day).

Opis:
It has been found that tegument homogenate of Ascaris lumhricoides suis and also trypsin inhibitor isolated from it induce the Leghorn chick embryos mortality when injected into their yolk sac on 4th, 8th or 13th day of incubation. The trypsin inhibitor is one of important components of Ascaris homogenate causing mortality. There is linear interrelationship between the logarithm of dose of homogenate or trypsin inhibitor and the mortality of chickens in %. A significant decrease of mean mass of chicks injected with Ascaris homogenate or trypsin inhibitor in comparison with control groups was observed. There was more frequent occurrence of developmental abnormalities and pathological changes in groups of hatched chicks which received Ascaris homogenate or inhibitor.

Opis:
Basing on 43 lectures and reports from the scope of current parasitological and mycological issues presented during the 50th Jubilee Clinical Day of Medical Parasitology (Lodz, 19–20 May 2011), the increasing importance of parasitic diseases and mycoses in medicine was presented. Difficulties in diagnosis and treatment of both imported parasitoses (malaria, intestinal amoebiosis, mansonelliasis) and native parasitoses (toxoplasmosis, toxocariasis, CNS cysticercosis), as well as parasitic invasions coexisting with HIV infection (microsporidiosis) have been emphasized. The possibility of human parasites transmission by vertical route and transfusion has been discussed. The important issue of diagnostic problems in intestinal parasitoses has been addressed, noting the increasing use of immunoenzymatic methods which frequently give false positive results. It was highlighted that coproscopic study is still the reference method for detecting parasitic intestinal infections. The mechanism of the immune reaction induced by intestinal nematodes resulting in, among others, inhibition of the host innate and acquired response was presented. Mycological topics included characteristics of various clinical forms of mycoses (central nervous system, oral cavity and pharynx, paranasal sinuses, nails and skin), still existing problem of antimicrobial susceptibility of fungal strains, diagnostic and therapeutic difficulties of zoonotic mycoses and the importance of environmental factors in pathogenesis of mycosis.

Recent literature data reveals the most common etiological agents of congenital parasitoses to be Toxoplasma gondii, Trypanosoma cruzi, Leishmania donovani and Plasmodium falciparum. An analysis of clinical data indicates that parasitic congenital infections are often asymptomatic, whereas symptomatic newborns usually display nonspecific symptoms, which greatly hinders correct diagnosis. The long-term consequences of prenatal infections are serious clinical problems. This article presents the possible routes of vertical transmissions (mother-to-child) of pathogens including prenatal, perinatal, as well as postnatal routes. It highlights the role of factors involved in protozoa transmission and development of congenital parasitic diseases, such as parasite genotypes, the relationship between the timing of maternal infection and the probability of passage of the parasite through the placental barrier, and the immunological features of pregnant women. Acquired and congenital babesioses in human and experimental animals are presented. It emphasises that the mechanisms by which parasites infect the placenta and cross from mother to fetus are still poorly understood. It also describes the cellular mechanisms of infection by T. gondii, such as tachyzoites crossing biological barriers, the expression of Toll-Like Receptors (TLR) family on trophoblasts and syncytiotrophoblasts as an immune response to intrauterine infection and cases of congenital and acquired toxoplasmosis, as well as the long-term consequences of congenital invasion with T. gondii, episodes of reactivation of latent toxoplasmosis and T. gondii reinvasions. Mycological topics include a rare case of in utero fungal infection of offspring by a mother with vaginal candidosis, and the fungal contamination of ward facilities and medical equipment as potential sources of exogenous infections of newborn children.

Most important infectious diseases which pose a risk to human health and life are associated with parasites transmitted by a variety of arthropod vectors, or from animal to man. Some of these (malaria, toxoplasmosis, leishmaniosis, dirofilariosis, alveococcosis, cystic echinococcosis) still represent a serious public health problem in many regions in the world. This review describes the epidemiological and clinical aspects of important parasitoses and fungal infections from a medical point of view. It should be emphasized that the development of invasive disease depends on both host (susceptibility/resistance) and parasite factors (pathogenicity/virulence); an immunocompromised state can favour opportunistic parasitic infections: toxoplasmosis, cryptosporidiosis, giardiosis, cyclosporidiosis, blastocystosis and strongyloidosis. This article highlights the role of free-living amoebae in the pathogenesis and transmission of human diseases, the high pathogenicity of Echinococcus multilocularis, and the growing importance of ticks as a reservoir and vector for numerous dangerous pathogens (e.g., Borrelia burgdorferi, Anaplasma phagocytophilum, Babesia microti). It also discusses the diagnostic problems of toxoplasmosis including cross-reactions in serological tests and reviews the search for new drugs and vaccines against toxoplasmosis. Attention is increasingly paid to the role played by the human microbiome in maintaining homeostasis and in the development of fungal infections. This review also presents the most common human superficial fungal infections and the role of Candida albicans infection in the pathogenesis of irritable bowel syndrome.

The history of annual meetings of parasitologists under the name of Clinical Days of Medical Parasitology began on 24th February 1962 at the initiative of Prof. Józef Wacław Grott, MD, PhD, the Head of the 1st Internal Diseases Department, Medical Academy of Lodz. The purpose of the meeting was promotion of parasitological issues among practicing physicians and enabling them to meet experienced researchers in the field of parasitology. In subsequent years the organization of the Conference has been continued by the staff of the Chair and Department of Biology and Medical Parasitology, Medical Academy/Medical University of Lodz. In the course of over 50 annual sessions of Clinical Days of Medical Parasitology, a total of 1678 lectures, papers and reports on parasitological and mycological issues were presented, including those on therapy and diagnostics, as well as on environmental determinants of parasitic diseases and fungal infections.

The 50% trichomonocidal concentration of 7 examined ethers (T. vaginalis strain N° 1/86, Roiron medium, safranine staining, 30 min.) was estimated by means of dose-response curve. The minimal mycostatic concentration (Candida albicans L-45, Geotrichum candidum; 3% Sabouraud agar, 37°C, 24 hr) was calculated with the aid of regression equation. All examined ethers show in vitro a marked trichomonocidal effect comparable with activity of ornidazole or phenol. Mycostatic activity of new compounds is many times higher than activity of phenol, but considerably lower than that of klotrimazole. The strongest complex trichomonoddal and mycostatic effect-comparable to ornidazole or klotrimazole - shows the new derivative N,N-diethylaminoethyl oxime of 1-tioflavone (compound II), which is fairly toxic for mammal.

Celem pracy było określenie poziomu lęku jako stanu i lęku jako cechy u płetwonurków amatorów. Grupę badawczą stanowiło 30 płetwonurków (w tym 13 kobiet) zrzeszonych w jednej z bydgoskich szkół nurkowania. Narzędziem badawczym był kwestionariusz ISCL (polska wersja kwestionariusza STAI Spielbergera). U większości (90%) badanych lęk jako cecha kształtował się na poziomie średnim i niskim. 46% badanych przejawiało lęk jako stan na poziomie wysokim. Kobiety istotnie silniej odczuwały lęk jako stan w porównaniu z mężczyznami (p<0,001). Nie stwierdzono istotnych korelacji pomiędzy wiekiem badanych a poziomem odczuwanych przez nich lęków.
The aim of this study was to evaluate the level of trait and state anxiety in recreational scuba divers. 30 divers (including 13 female) aged 14-44 were examined with Spielberger's STAI Questionnaire. Results revealed that majority of subjects (90%) presented low and intermediate level of trait anxiety. Anyway, 46% of them presented high level of state anxiety. State anxiety differed significantly between male and female divers (p<0.001). There were no significant differences according to the age of the subjects.

The pathogenicity of fungi depends on, inter alia, the secretion of hydrolytic enzymes. The aim of this study was to determine the enzymatic activity of yeasts and yeast-like fungi isolated from children’s recreation areas, and compare the results with literature data of strains obtained from patients with mycoses. The enzymatic activity of 96 strains was assessed using an API ZYM kit (bioMerieux, France) and their biotypes were established. The fungal species were found to produce from 16 to 19 hydrolases: the most active were: leucine arylamidase (e5), acid phosphatase (e10), alkaline phosphatase (e1), naphthol-AS-BI-phosphohydrolase (e11), esterase – C4 (e2), β-galac - tosidase (e13) and β-glucosidase (e16). In addition, 13 biotypes characteristic of particular species of fungi were defined. Most strains could be categorized as biotypes C2 – 39.5% and A – 26%. The examined fungal strains isolated from recreational areas have selected biochemical characteristics i.e. production of hydrolases, which demonstrate their pathogenicity. They produce a number of enzymes which are also present in strains isolated from patients with mycoses, including: leucine arylamidase (e5), acid phosphatase (e10), naphthol-AS-BI-phosphohydrolase (e11) and alkaline phosphatase (e1). The biotypes identified in the course of this study (A, B3, B4, C1, C6 and D3) have been also reported in cases of fungal infection. Therefore, the fungi present in the sand and soil of recreational have pathogenic properties and are possible factors of fungal infection among children.
