The article comprises a critical review on practical applications of molecular technology in parasitological diagnostics in a broad sense, also as a diagnosis of species and a method of epidemiological analysis. Techniques of genome analysis at different levels, as specific nucleic acid probes, DNA restriction profiles (RFLP), hybridization techniques, pulse-field gel electrophoresis, in vitro nucleic acid amplification, and DNA fingerprint technique used in studies on Giardia and Cryptosporidium were discussed. The essential reservation as far as this technology is concerned refers to its usefulness in parasitological diagnostics; there is no sense in working out methods for recognizing parasites which could otherwise be identified by well trained parasitologists and simple microscopic methods. The improved diagnosis of parasites resulting from the application of molecular technology significantly contributed to the armentarium of parasitologists. Application of recent molecular technology in diagnosis of giardiosis and cryptosporidiosis may basically support clinical diagnosis which provides possibilities of early and selective treatment and makes possible epidemiological studies. These assays will permit not only a rapid diagnosis and exact differentiation but will also enable a better recognition of Giardia and Cryptosporidium genome organization. However, in spite of the wide availability of this new techniques they have not been fully applied - as yet – in diagnosis and in epidemiological studies on these parasites. The authors share the opinion of BUSCH (1991) on the need of proper recognition of high-quality and rigorous work in employing new molecular assays, because their wide availability and high sensitivity could cause "false-positive" results by contamination with amplified DNA sequences. They totally support the conclusion made by THOMPSON and MELONI (1993) that the molecular data must be fairly interpreted by collaborating molecular biologists, parasitologists, and epidemiologists - and clinicians as well - to avoid forming of a confusing picture of genetic diversity of the parasites with no practical application.

The authors present the actual review on several publications concerning the molecular characterizations of the viruses found in parasitic protozoa such as Giardia, Trichomonas, Leishmania and Entamoeba histolytica. All of the RNA viruses observed in parasitic protozoa showed several similarities and did not considerably differ from the viruses found in simple eukaryotic cells; they closely correspond to dsRNA viruses of yeast. The supposition that the protozoan symbionts detected in laboratories transfer to their hosts in natural conditions seemed to be rational, though, there are no evidences that these symbionts are potential pathogens. However, the opinion reiterates that intestinal protozoa (e.g. Entamoeba histolytica) may serve as vectors for HIV or cofactors of HIV infection. The authors point out that irrespective of the potential role of viruses as vectors in the transfection system for parasitic protozoa, the observed viral system constitutes an unusual experimental system to solve the problems of gene expression.

Phytoplankton samples collected from the Northern Indian Ocean (Bay of Bengal, northern Arabian Sea, and Dona Paula Bay Goa, west coast of India), were utilized to quantify changes in cell size, cell volume and carbon per cell of diatoms and dinoflagellates. The dataset from the Bay of Bengal also provides inter- and intra-annual variations (April 2008 to March 2010). The variations in cell size and volume were large in regions influenced by the riverine influx or terrigenous inputs. An interregional comparison of commonly available forms (8 species) points out that cell volumes are highest in the North Atlantic and lowest in the Mediterranean. The information provided will be useful in estimation of carbon biomass and biogeochemical studies.

Giardia duodenalis (syn. G. intestinalis, G. lamblia) is a cosmopolitan flagellate organism belonging to the most common intestinal protozoan parasites of humans and animals. Great genetic heterogeneity has been found within G. duodenalis, where only genotypes representing assemblages A and B have zoonotic potential. Fecal samples (447 specimens) obtained from 232 humans in West-central region of Poland were examined by microscopy and PCR. The total prevalence of Giardia in humans was 1.3%. DNA was extracted from three positive fecal samples and PCR products were obtained after amplification using the β-giardin primers G7 and G759. Sequence and phylogenetic analyses showed that G. duodenalis isolates from humans belonged to A and B genotypes. Moreover, three subgenotypes, including a cosmopolitan subgenotype A2 and two new subgenotypes A and B were detected.

Proterochampsians are a South American endemic group of non-archosaurian archosauriforms with morphological characteristics recollecting Recent crocodilians, and therefore have been proposed as aquatic species. However, this has not been based on careful examination of anatomical and histological features. We provide a review of the morphological and histological evidence present in the skeleton of proterochampsids and discuss its implications for inferring the lifestyles of these organisms. Anatomical features such as a secondary palate, marginal dentition, palatine teeth, morphology of the tail, limb modification, and dermal armor are reviewed, and details of histological structures are described based on bone thin sections. Histological examination reveals a predominance of fibrolamellar bone tissue, suggesting rapid periosteal osteogenesis and therefore overall fast bone growth. The existence of discontinuities (LAGs) demonstrates that these animals responded to changes in their environment. Ecomorphological features do not provide definitive evidence for the lifestyles of proterochampsids, but allow us to propose a terrestrial/amphibious condition. The same is true of the histological features, particularly compactness of the bone.

There are many methods for detection of Cryptosporidium oocysts. Most of them (more than 20) enable the microscopic detection of Cryptosporidium oocysts in faecal smears. Such a great variability of diagnostic methods may lead to confusion as far as the choice of an appropriate technique by a given laboratory is concerned. This study evaluated the diagnostic usefulness of Cryptosporidium oocysts and coproantigen detection methods in the diagnosis of cryptosporidiosis in human (266 stool specimen) and animals (205 from cattle, 160 from sheep, 30 from horses, 80 from cats, 227 from dogs and 11 from wild animals). The total number of human and animal stool specimens processed was 266 and 713, respectively. In this study the usefulness of several diagnostic methods was compared. The following techniques were taken into account: wet mounts, hematoxylin staining, four different specific methods (modified Zeihl-Neelsen, Kinyoun's, safraninmethylene blue, as well as carbol-methyl violet and tartrazyne) and commercially available kit based on enzyme-linked immunoassay (ProspecT(r) Cryptosporidium Microplate Assay). The final number of positive specimens was 123. Out of them 77 were positive in all specific methods. The oocysts found in stool specimens were measured. Humans were infected with C. parvum and animals with C. parvum, C. andersoni or C. felis. The statistical analysis has shown that EIA test was a better than microscopy method for identification of Cryptosporidium in faecal samples in human and wild animal. Sensitivity and specificity are important factors for the choice of a proper diagnostic method for Cryptosporidium detection, however other factors such as cost, simplicity and ease of interpretation of results are also important considerations.

As part of a comprehensive examination of all radiodontans from Cambrian localities in the USA, Pates et al. (2017a, b) and Pates and Daley (2017) revised the taxonomic affinities of several described specimens. This included the reinterpretation of two putative lobopodians, one from the Wheeler Formation (Utah, USA) and one from the Valdemiedes Formation (Spain), as frontal appendages of the radiodontan genera Stanleycaris and Caryosyntrips respectively. In their comment, Gámez Vintaned and Zhuravlev (2018) disagree with these conclusions and raise three topics for discussion: (i) anatomical features they suggest support a lobopodian affinity for “Mureropodia”; (ii) the identity of Caryosyntrips as a radiodontan, and the assignment of certain specimens to this genus; and (iii) the nomenclatural status of Stanleycaris hirpex as an invalid taxon. For (i), we dispute that the anatomical features put forward by Gámez Vintaned and Zhuravlev (2018) are biological and conclude that a lobopodian affinity for Mureropodia is untenable. In response to (ii), we provide further evidence supporting a radiodontan affinity for Caryosyntrips, and those specimens ascribed to this genus. Finally, we concur with (iii) Stanleycaris as an invalid taxon according to the International Code on Zoological Nomenclature (ICZN), and have rectified the situation by providing a valid systematic description.

Opis:
Effects of temperature and glycerol concentration on rheological properties of potato starch solutions were investi- gated. The flow behaviour (shear stress against shear rate) was fitted to various models: power law, Herschel-Bulkley, Bingham, modified Bingham and Casson models. However, it was found that the Herschel-Bulkley model describes better the flow behaviour observed at various temperatures and glycerol concentrations, for flow behaviour index values between 0.44 and 0.78, typical of pseudoplastic solutions. The effect of glycerol concentration on each of the fitting parameters for Herschel-Bulkley model was well modelled by a second-degree polynomial at various temperatures. The simultaneous influence of glycerol concentration and tempe- rature on shear stress could be represented empirically by a second- degree polynomial function that includes linear coupling between concentration and temperature. Finally, the variation of the con- sistency coefficient with both temperature and glycerol concentra- tion was well described by an exponential expression, with an acti- vation energy value of 2.78 kJ mol -1 . The results indicate that both glycerol content and temperature have the effect of diluting potato starch solutions.

Copper is an essential trace element in living organisms, but its excess interferes with metabolic transformations in plant, animal and human cells. The uptake of copper from contaminated soils can be regulated, for example, by soil liming or application of other substances which enhance soil adsorption capacity and Cu binding in the substrate. The purpose of this study has been to assess the response of maize to soil contamination with copper depending on the applied neutralizing substances. The tested factors were increasing concentrations of Cu in soil: 0, 200, 400 and 600 mg Cu kg ?1 dm, and soil application of mineral (lime, loam and zeolite) and organic (manure, peat and bark) neutralizing substances. Soil contamination with copper within the range of 200 to 600 mg kg ?1 of soil caused reduction in maize yields. Positive influence such as alleviation of the harmful effect of copper contamination was demonstrated by lime and manure, which enhanced yields of maize, especially in the series polluted with 200 and 400 mg Cu kg ?1 . The other soil amending substances, especially peat added in amounts of 400 and 600 mg kg ?1 of soil, caused aconsiderable depression in maize yields. A linear dependence has been demonstrated between the concentration of Cu in maize plants and the content of Cu in the substrate, with the root content of Cu being on average six-fold higher than in aerial organs. Mineral soil amendments significantly decreased the BTC index in maize compared to organic substances, and lime as well as pine bark decreased the BCF index to 2.33 and 1.67 versus the value of 4.21 found in the control treatment without any neutralizing substances. The uptake of copper depended on the volume of yield and ? to a lesser degree ? on the concentration of Cu in plants. The uptake of copper by plants was the highest in treatmentscontaminated with a rate of 200 mg Cu kg ?1 , which was the consequence of higher yields from that treatment than from the plots polluted with 400 or 600 mg Cu kg ?1 of soil.

Opis:
A new sauropod titanosaur from the Upper Cretaceous Anacleto Formation is described. Narambuenatitan palomoi gen. et sp. nov., is diagnosed by cranial and axial autapomorphies. The holotype, which represent a subadult individual, consists of the left premaxilla and maxilla, braincase, both quadrates, one cervical vertebrae, one dorsal vertebra, fragments of cervical and dorsal ribs, seventeen caudal vertebrae, caudal transverse processes, fragments of haemal arches, left sternal plate, right coracoid, left humerus, left ulnae, both pubes, iliac pedicel, proximal fragment of right ischia, and an incomplete left femur. The phylogenetic analysis indicates that Narambuenatitan is a non−eutitanosaurian lithostrotian, and that it shares with Epachthosaurus a neural spine in middle caudal vertebrae which are laminar and posteriorly elongated.

In this study, [Beta]-galactosidase enzyme from Kluyveromyces fragilis was immobilised on a commercial polyethersulfone membrane surface, 10 kDa cut-off. An integrated process, concerning the simultaneous hydrolysis–ultrafiltration of whey lactose was studied and working conditions have been fixed at 55[degrees]C and pH 6.9, the same conditions that are used for the industrial process of protein concentration. For the immobilisation, best results were obtained using 5% (v/v) of glutaraldehyde solution and 0.03 M galactose; the total activity recovery coefficient (TARC) was 44.2%. The amount of immobilised enzyme was 12.49 mg with a total activity of 86.3 LAU at 37[degrees]C, using 5% (w/v) lactose solution in phosphate buffer (100 mM pH 6.9). The stability of the immobilised enzyme was approximately 585 fold higher in comparison with the stability of free enzyme. Multipoint covalent immobilisation improves the stability of the enzyme, thereby enhancing the decision to use the membrane as a filtering element and support for the enzyme immobilisation.

Seeds of sesame (Sesamum indicum L), genotypeTMV3 were treated with various concentrations of EMS and Colchicine like 0.2, 0.4, 0.6, 0.8, 1.0, 1.2, 1.4, 1.6, 1.8 and 2.0 % and the LD50 values observed at 1.0 % of EMS and 0.6 % of Colchicine. Based on their LD50 values appreciable concentrations only selected for further study and their effects on various morphological characters such as plant height per plant, number of branches per plant, number of leaves per plant, days to first flowering, number of capsules per plant, number of seeds per capsule and seed yield per plant (g) were measured quantitatively and the results showed that all the characters were consequently or significantly reduced when compared to control and also EMS showed more effective than colchicine.

Developmental mode and proximal structures are commonly accepted as the best for the recognition of high−level taxonomic categories within the Graptoloidea. The petalolithids and retiolitids are unique in possessing a virgellar ancora and in the latter, distal ancora development. The ancora structures are considered homologous, and the ancorate petalolithids are considered to be the direct ancestors to the retiolitids. The Retiolitidae are unique among the diplograptoids in possessing (1) outer, lateral, ancora sleeve walls (derived from distal extension of the ancora), and (2) a skeletal framework of bandaged lists between which are a succession of very thin and rarely preserved fusellar layers. Retiolitids possess different kinds of thecal profiles and two types of micro−ornamentation on the lists, and these have served to distinguish between the subfamilies Retiolitinae and Plectograptinae. Complete retiolitid morphological terminology is clarified and explained. Cladistic analysis of the retiolitids provides some measure of a better understanding of retiolitid evolution, but adds only modest support for the retention of the two subfamily categories.
