Plant breeders have focused much attention on polyploid trees because of their resistance for forestry. To evaluate the impact of intraspecies genome duplication on NaHCO3 stress, a series of Betula platyphylla autotetraploids and diploids were generated from the same family. The growth, proline content and proline-associated gene expression of these autotetraploid individuals were compared with those diploid trees. Autotetraploids were superior in injury index and relative growth of height and base diameter compared to diploids. The proline content was higher in autotetraploid individuals compared to diploids. Gene expression data revealed autotetraploids were generally higher expression in BpP5CS1, BpP5CS2, Bp- P5CR1, BpP5CR2, BpP5CR3 and BpOAT and were lower expression in BpProDH and BpP5CDH compared to diploid trees. These results shed light on resistance variation in birch autotetraploidization and polyploidy breeding as a new approach for genetic improvement of birch trees.

We performed here a systematic ab initio calculations on neutral gas-phase L-proline. A total of 8 local minima were located by geometry optimization of the trial structures using density functional theory (DFT) with B3LYP three parameter hybrid potential coupled with the 6-31G)d( basis set. The absolute minimum obtained will be subject to a rigid potential energy surface (PES) scan by rotating its carboxylic group using the same method with more accurate basis set B3LYP/6-311++G(d,p), to get a deeper idea about its conformational stability. The main aim of the present work was the study of the rigidity of the L-proline structure and the puckering of its pyrrolidine ring.

Opis:
Monocultures of Norway spruce widely used for afforestation of drained peat soils often have low productivity and decline in vitality. The research aim was to elucidate: (1) imbalances in the nutrient status of soil and plants in relation to Norway spruce crown status and annual increment on drained peat soil in the sub-boreal zone; (2) the suitability of using proline accumulation as a biomarker for Norway spruce vitality and stress in nutrient imbalance conditions. The study was conducted at five forest (Oxalidosa turf. mel.) sites in Latvia, each containing trees with different crown condition status. Chemical analyses of soil and spruce needles, assessment of tree crown vitality and annual increment measurements were done. Our results revealed serious disturbances in the supply of nutrients, which is an important factor in the decline of Norway spruce in monoculture stands on drained peat soils. Deficiency of K, Fe, Cu, B, N and P was found in the current year needles for all trees, but in damaged trees (≥61% defoliation, ≥61% discoloration) Zn deficiency was also found. For 2-year-old needles, deficiency of K, P, S, Zn, and Cu was found in all trees, additionally, deficiency of Fe was found in healthy and medium damaged trees (26–60% defoliation, 21–60% discoloration), and N deficiency was found for medium damaged and damaged trees. Thus, K, Cu, B, Fe concentrations had significant negative correlations with crown status parameters – defoliation, discoloration, and density. K and B deficiency could play the most significant causal role in decreased stem increment. The results revealed that the stress amino-acid proline is a reliable biomarker, having a significant relationship with spruce crown condition parameters, annual increment and nutrient status in needles – low levels of K, Fe, S, Cu, B, N. The increase in proline content in spruce needles was pronounced in moderately damaged trees, indicating the potential for the use of proline accumulation as early stress indicator for spruce. Therefore, further studies on the identification of early stress and factors affecting nutrient uptake and accumulation in needles are particularly valuable for evaluation of stand management options before significant decline of spruce stands.

Seasonal changes in the levels of water-soluble carbohydrates, free proline and tolerance to freezing and water deficit were studied in leaves of cherry laurel (Prunus laurocerasus) and ivy (Hedera helix). Frost and water deficit tolerance was estimated based on the measurement of electrical conductivity of aqueous media containing leaf discs that were previously treated with frost (–7°C) or polyethylene glycol solution (PEG 600), respectively. Carbohydrate content in leaves of examined species changed differently throughout the measurement period. In laurel leaves the highest carbohydrate level was found in February, while it was lowest in June. In contrast, in ivy leaves the highest carbohydrate content was recorded during summer (June, July), while the lowest in February. However, a lack of correlation between soluble carbohydrate level and membrane injury index was shown in this study. Free proline content in leaves of investigated species was the highest in early spring, i.e. April. The lowest level of free proline in laurel leaves was found in July, September and October, but in ivy from July to February. A negative correlation between proline content and membrane injury index observed in frost and PEG treated leaves of both species suggest that this amino acid may have resulted on membrane protection.

In asymmetric synthesis of organic compounds more effective solutions are being looked for which will result in higher yield(s) of product(s) and their high enantioselectivity [1]. One of such solutions is an use of a multilevel and cheap catalyst. Proline used as a catalyst is a substance of natural origin which was synthetically obtained by Willstätter who was carrying out research on hygric acid (Scheme 1) [10]. The cells of many organisms have a suitable enzymatic system essential for proline biosynthesis [15]. So far, three proline biosynthesis pathways have been described: from glutamate (Scheme 3 and 4), ornithine (Scheme 5 and 6), and arginine (Scheme 7) [16–28]. Proline which is obtained as a result of biosynthesis or supplementation is a substrate for many proteins. Characteristic and significant content (about 23%) of this amino acid was observed in collage. In cells proline can play an important role of osmoregulator [31–35] – a protective substance regulating the activity of such enzymes as catalase and peroxidase [36]. Proline as a secondary amine shows exceptional nucleophilicity facilitating imine and enamine formation. Used as a catalyst in aldol reaction makes with substrates like imine or enamine transition state imitating the activity of naturally occurring enzymes for this type of reaction, that is aldolases. In their research Hajos and Parrish, and Eder, Sauer and Wiechert used proline in intramolecular aldol reaction obtaining proper enones (Scheme 9) [60–62]. The process of intramolecular aldol reaction was used for a separation of racemic mixture of diketones (Scheme 10) [63, 64], cyclization of ortho-substituted aromatic aldehydes and ketones (Scheme 11) [65], synthesis of cyclic diketones (Scheme 13) [68] and domino reaction to obtain substituted cyclohexanones from beta-diketones and unsaturated ketones (Scheme 14) [69].

The study aimed at demonstrating intracellular proline synthesis under osmotic stress conditions in selected soil fungi on the example of Trichoderma sp. and Trichotecium roseum. Effect of the increase of sodium chloride (NaCl) salinity, in concentrations of 1 to 1000 mmol dm–3 PDA medium, on proline content in the fresh matter of mycelium cultured on medium was examined. The increase of medium salinity affects production of osmoregulatory substances in the form of proline in selected soil fungi. Its content in mycelium depends on salt (NaCl) concentration in medium as well as on species. Intracellular proline synthesis increased starting with the least salinity. As osmotic stress increased, proline content in mycelium almost quadrupled in Trichoderma sp. and quintupled in Trichotecium roseum.
Celem badań było wykazanie syntezy wewnątrzkomórkowej proliny w warunkach stresu osmotycznego u wybranych grzybów glebowych na przykładzie Trichoderma sp. i Trichotecium roseum. Badano wpływ stresu osmotycznego w wyniku zasolenia chlorkiem sodu (NaCl), w stężeniu od 1 do 1000 mmol dm-3 pożywki PDA na zawartosć proliny w świeżej masie grzybni wyrosłej na podłożu. Wzrost zasolenia podłoża wpływa na wytwarzanie substancji osmoregulacyjnych w postaci praliny u wybranych grzybów glebowych. Jej zawartość w grzybni jest zależna od stężenia soli NaCl w podłożu oraz od gatunku. Synteza proliny wewnątrzkomórkowej zwiększała się począwszy od najmniejszego zasolenia. W miarę wzrostu stresu osmotycznego zawartość proliny w grzybni zwiększyła się prawie czterokrotnie u Trichoderma sp. i pięciokrotnie Trichotecium roseum.

The LlPRP2 gene coding for a proline-rich protein shows a high level of similarity to, as well as significant differences from the family of ENOD2 nodule-specific genes. Several sequence motifs with putative regulatory function were identified in the 5' and 3' noncoding regions of the LlPRP2 gene. Northern blot analysis revealed that the expression of the LlPRP2 gene begins 9 days after inoculation of yellow lupin roots with Bradyrhizobium sp. (Lupinus); the expression is restricted to symbiotic nodules and is not detected in other tissues or organs. Detailed hybridization analysis showed that, when expression is activated, the LlPRP2 transcript is modified so as to produce at least three bands and a continuous distribution of decay intermediates. The modification of the LlPRP2 transcript probably involves degradation from the 5'- and/or 3'-ends of the RNA molecules. Southern blot analysis indicates that only one gene is present in the yellow lupin genome. The presence of genes homologous to the LlPRP2 gene was confirmed for three cultivars of yellow lupin and for Lupinus angustifolius. However, LlPRP2 homologues were not detected in Lupinus albus cv. Bac, indicating that this plant may lack the ENOD2 sequence.

In this study, the effect of different alkaline concentrations (0, 10, 20, 30, 40, 50, 60 mM) on germination and biochemical characteristics of the two sesame (Sesamum indicum L.) cultivares (Dashtestan and GL-13) which are registered cultivars of Iran were investigated. The experiment was carried out in a completely randomized design with three replications. Results showed that , germination percentage, germination rate, shoot length and dry weight, root length and dry weight and K+ content decreased, whereas, malondialdehyde (MDA), proline, total soluble sugars and Na+ contents increased with increasing alkalinity stress. GL-13 cultivar had the least root and shoot length, proline and K+ content than Dashtestan.

Accumulation of proline is a defense mechanism against external stress conditions, preventing damage to the structure and function of cells and improving plant development processes, such as germination. The purpose of this study was to investigate proline treatment as a means of improving the germination and development of Norway spruce seedlings. The effect of exogenous proline has been studied in three stages of initial plant development. The collected seeds were soaked in water or 8 mM proline solution and placed on the germinators. The germination capacity and the mean germination time were determined. Seedlings with radicles >10 mm were transferred to the sand-peat substrate at a constant temperature of 20 °C. Seedlings at 3 subsequent developmental stages (S1 – germinated seeds with radicles > 3 mm; S2 – seedlings with radicles >10 mm; S3 – established seedlings grown for 90 days) were examined for the oxygen consumption rate, total antioxidant capacity, hydrogen peroxide level, malondialdehyde level and intracellular proline content. Proline treatment was conducive to lowering the levels of hydrogen peroxide and malondialdehyde at stage S1. At the subsequent stages of development, the levels of hydrogen peroxide and malondialdehyde increased, and at the S3 stage, there was also a marked increase in total antioxidant capacity. At stage S3, the seedlings of the proline treatment were characterized by a lower total mass, and the response to exogenous proline was stronger in the root tissues than in the leaves. The oxygen consumption rate was higher for the proline treatment at all stages of development. Seedlings at the analyzed stages of establishment differed in response to proline treatment. Exogenous proline had some beneficial effects during the first phase of germination by reducing the level of hydrogen peroxide and improving the condition of lipid membranes. In the subsequent stages of seedling development, in response to the same concentration of proline solution, undesirable effects, such as an increase in hydrogen peroxide levels and damage to cytoplasmic membranes, were observed. Optimal concentrations of exogenous proline should be determined prior to commercial use of proline treatment to improve plant stress tolerance.

In recent years there has been a dynamic development of asymmetric synthesis. Groups of researchers, particularly the one led by Benjamin List and Carlos Barbas, carried out a number of reactions and showed the effectiveness of the use of small organic molecules such as proline as catalysts. Michael addition catalyzed with proline is a particularly interesting reaction because it can be carried out in two aminocatalytic pathways. The analysis of Michael reaction reveals potential for both forms of aminocatalysis: enamine and iminium catalysis (Scheme 1) [1–14]. Presumably Michael reaction proceeds mainly according to enamine mechanism. The use of proline in Michael reaction with imine activated acceptor is slightly effective. So far the researches have shown that the modification of proline molecule or addition of other catalyst is necessary for condensation to appear. Enamine catalysis concerns the activation of carbonyl compound in situ being a donor. There is no need for enolase anion to be created earlier [2, 15–17]. When, as a result of the reaction of a,b-unsaturated carbonyl compound with proline, Michael acceptor activation appears it means that it is enamine mechanism reaction (Scheme 1) [2, 24]. One of the first examples of direct Michael reaction proceeding through enamine transition state is the reaction of cyclopentanone with nitrostyrene (Scheme 6) [20–23]. Other examples of Michael addition of ketone with nitro olefin catalysed by proline are shown in table 2 and 3 [10, 23, 30]. Nitroketones obtained in that way are useful as precursors for different organic compounds [33], also pyrrolidines [34]. Pyrrolidines are pharmacologically active and they selectively block presynaptic dopamine receptors [34] (Scheme 7). Except for Michael intermolecular reaction, intramolecular condensation adducts were also obtained. Michael intramolecular proline-catalyzed condensation in which inactive ketones transform into α,β-unsaturated carbonyl compounds was described (Scheme 9) [35, 36]. These reactions require a stoichiometric amount of a catalyst and a long time of reaction and they give as a result a little enantiomeric excess [11, 24, 35]. In 1991, Yamaguchi and co-workers carried out malonates Michael addition to α, β-unsaturated aldehydes catalyzed by L-proline [24, 39]. The reaction proceeded according to enamine mechanism, for example dimethyl malonate was reacted with hex- 2-enal in the presence of proline to give Michael adduct in 44% yield. To improve the yield an attempt of a slight modification of a proline molecule was made transforming it into proper salt. Proline lithium salt enabled to obtain the condensation product in 93% yield (Tab. 4). Regardless of a used catalyst the products in the form of racemates were obtained. In order to improve enantioselective properties of a catalyst, Michael addition of diisopropyl malonate to cycloheptenone was carried out in chloroform in the presence of different proline salts. Optimal enantioselectivity and yield was obtained by using rubidium salt (Tab. 5–7) [40, 41]. Rubidium prolinate-catalyzed Michael additions are used in industry e.g. for enantioselective synthesis of the selective serotonine reuptake inhibitior (SSRI) (–)-paroxetine (antidepressant) (Scheme 12) [24].

Mannich reaction occuring among ketone, aldehyde, and amine is one of the ways of a synthesis of biologically active compounds. Reactions of this type were carried out in the presence of different catalysts [3–10], however in recent years a lot of attention has been paid to enantioselective Mannich reaction catalyzed with proline. Such reactions were carried out with the use of different compounds containing carbonyl group and the most frequently used amine was p-anisidine. The advantage of the use of p-anisidine is a possibility of conducting the direct Mannich reaction (Scheme 3). In this way β-amino ketones (Tab. 1, 2, 4) [15, 18–20, 23, 24], α-hydroxy-β-amino ketones (Tab. 3) [15, 22], and β-amino alcohols (Tab. 5, 6) [25, 26] were obtained. A possibility of syntheses of β-amino sugars and α-amino acids with their derivatives (Tab. 7) [28, 29] is worth noticing. In a great number of described reactions, the products were obtained with satisfactory yield and enantiomeric excess. Taking into consideration the difficulty of a removal of p-hydroxyphenyl group which protects amine group in the resulting products, the attempts of using different amine compounds in Mannich reactions catalyzed with proline were undertaken. The use of amines blocked by tert-butoxycarbonyl group (Boc) enabled to obtain the products with high yield and ee values (Tab. 12–15) [35–38]. However in the case of the use of Boc the reaction must be carried out in an indirect way (it is necessary to prepare imine blocked by Boc earlier).

Opis:
The effect of soil drought on leaf water content, proline content, and the activity of guaiacol (GuPX) and ascorbate (APX) peroxidases as well as the level of lipid peroxidation were investigated in leaves of drought resistant red fescue (Festuca rubra) and drought sensitive perennial ryegrass (Lolium perenne). Plants were grown under glasshouse conditions in soil pot culture. 26 day-old grasses were exposed to drought by withholding irrigation for 18 days. Water content in leaves of perennial ryegrass decreased more than in red fescue throughout the experimental period. On the other hand, proline content (PC) was higher in red fescue. The activity of APX and GuPX increased in leaves of red fescue, while it did not change in perennial ryegrass. Our data demonstrate that both red fescue and perennial ryegrass were able to survive applied drought, as shown by a lack of stress-induced lipid peroxidation and hence no evidence of oxidative damage. We speculate, that the observed drought stress tolerance at cellular level was associated with the ability to accumulate proline, and to maintain high activity of APX and GuPX, resulting in protection against oxidative damage and lipid peroxidation. It seems that this mechanism works better in red fescue.
