Opis:
Human peripheral blood cells stimulated by phytohemagglutinin (which serve as a model of cancerous cells) and resting cells were incubated in dimethyl sulfoxide solutions of various phthalocyanines. In order to diminish the influence of atmospheric oxygen the cells were embedded in a polymer (polyvinyl alcohol) film. Fluorescence spectra of the samples were measured over two regions of excitation wavelengths: at 405 nm (predominant absorption of the cell material) and in the regions of strong absorption of phthalocyanines (at about 605 nm and 337 nm). The intrinsic emission of cell material became changed as a result both of cells' stimulation and of incubation of cells in dye solution. In most cases the stimulated cells when stained by dye exhibited higher long wavelength fluorescence intensity than resting cells. This suggests higher efficiency of dye incorporation into cancerous cells than into healthy cells. The absorption spectra of samples were also measured. The spectra of various phthalocyanines in incubation solvent, in polymer and in the cells embedded in polymer, were compared. The comparison of properties of the cells stimulated for different time periods enabled to establish the conditions of stimulation creating a population of cells incorporating a large number of sensitizing molecules.

The interactions of two metal-free phthalocyanines [(H2Pc) and Solar Pc (with four peripherical groups: SO2N(CH2CH2OH)2)] and of one metal substituted dye (CoPc) with resting and stimulated human peripheral blood mononuclear cells (PBMC) were compared. The absorption, fluorescence, photoacoustic and EPR spectra of both resting cells and cells stimulated by phytohaemagglutinin, incubated in dimethyl sulfoxide (DMSO) with very low or 95% water content and with or without dye addition, were measured. The fate of the light absorbed by the samples was investigated. It is known that singlet oxygen production is crucial for photodynamic action of dyes. Thermal deactivation and luminescence emission compete with this process, so investigation of these alternative paths of sensitizer deactivation provides information about photodynamic action. The incorporation of the investigated dyes into cells and the perturbation of the cell structure caused by the dyes, the incubation solvent and the activator were investigated by comparing the spectral properties of PBMC before and after stimulation and incubation. Incubation of the cells for 1 h in a solution of Solar Pc in 99.5% aqueous DMSO, resulted in an efficient dye incorporation which was highly selective. Solar Pc being introduced much more efficiently into stimulated cells than into resting cells.

During the last few decades, owing to their unique properties, gold nanoparticles (AuNPs) have found numerous biomedical applications. Studies on rodents prove that AuNPs entering an organism easily reach the bloodstream and undergo wide tissue distribution. The presence of nanoparticles inside blood and bone marrow cells of exposed animals may implicate its influence on hematopoesis and the functions of peripheral blood leukocytes. The aim of this study was to determine the effect of oral administration of commercial gold nanocolloid, recommended by the producer as a dietary supplement, on the percentage of lymphocyte populations and proliferative response, as well as the activity of phagocytes in the peripheral blood of mice. The colloid was given to the animals in three different doses (0.25, 2.5, 25 ppm), for three different time periods (7, 14, 28 days). Mice given nanoparticles showed increased activity of phagocytes and some changes in the lymphocyte phenotypes. The elevated activity of granulocytes and monocytes, in terms of both phagocytic and respiratory burst activity, was transient and noticed only after a short time of administration, which may indicate some adaptability of blood phagocytes to prolonged presence of gold nanoparticles in the body. However, phenotypic modifications among lymphocytes in the group of animals given the middle dose of colloid (i.e. increased percentage of B and CD4+CD8+ DP T cells) did not occur until after the 28-day administration, which in turn seems indicative of some immune dysregulation due to the prolonged contact with nanogold.

Opis:
Determining the reference values for B and T lymphocytes and their subpopulations in rabbits used in Poland for economic and laboratory purposes is not very common. The goal of this study was to evaluate values of chosen B and T cells and their subpopulations in Polish mixed-breed rabbits, taking season of the year and sex of the animals into the consideration. The study was performed with the use of flow cytometry using monoclonal antibodies (mouse anti-rabbit). The study revealed that, both in Polish mixed-breed rabbits and in Polish mixed-breed rabbits with an addition of blood of meat breeds, the season of the year and sex of the animals affectthe percentage of B- and T-cells and their subpopulations in peripheral blood. As far as season of the year is concerned, it was shown that it mainly affects CD5+ T cells in mixed-breed rabbits and CD25+ T cells in mixed-breed rabbits with an addition of blood of meat breed, while taking sex of the animals into account, more changes were recorded in females in both groups of rabbits. Due to the fact that this is the first paper on those values in Polish rabbits conducted on a large amount of animals, the values may be treated as reference.
Ocena wartości referencyjnych dla limfocytów B i T oraz ich subpopulacji u królików wykorzystywanych w Polsce w celach ekonomicznych i laboratoryjnych jest rzadkością. Celem pracy jest ocena parametrów wybranych limfocytów B i T oraz ich subpopulacji u polskich królików mieszańców z uwzględnieniem pory roku i płci tych zwierząt. Badania wykonano z wykorzystaniem cytometrii przepływowej i przeciwciał monoklonalnych (mouse anti-rabbit). Badania wykazały, że zarówno u królików mieszańców polskich, jak i mieszańców polskich z dolewem krwi ras mięsnych, pora roku i płeć wpływa na kształtowanie badanych parametrów, tj. procentu limfocytów B i T oraz ich subpopulacji. Co się tyczy pory roku wykazano, że zmiany dotyczą głównie limfocytów T CD5+ w przypadku królików mieszańców polskich i limfocytów T CD25+ w przypadku królików mieszańców polskich z dolewem krwi ras mięsnych. Tymczasem w przypadku płci zwierząt, więcej zmian w obu grupach zwierząt wykazano u samic. Ze względu na to, że jest to pierwsza praca z tego zakresu i badania dotyczą dużej grupy zwierząt, winny one być traktowane jako wartości referencyjne.

Bovine leukemia virus (BLV) is a deltaretrovirus which infects and induces proliferation of B-lymphocytes in the peripheral blood circulation and in lymphoid organs primarily of cattle, leading to leukemia/lymphoma. This study was carried out to investigate the presence of BLV in cattle, sheep and camels from the Chaharmahal va Bakhtiary and Isfahan provinces in Iran. A total of 874 blood samples collected from cattle, sheep and camels were used in this study to detect BLV using a nested- PCR. The results from this study indicated that 17.2% (n=874) of all blood samples collected were positive for BLV. The percentages of blood samples positive for BLV from cattle, sheep and camels were 22.1 (n=657), 5.3 (n=95) and 0 (n=122) respectively. The results from this study showed that BLV infected cattle and sheep. Camels seemed to be resistant to BLV infection. This study contributes to the nationwide effort to obtain baseline information on the prevalence of BLV, which will assist in planning the control strategy for the disease in Iran.

The influence of caprine arthritis-encephalitis (CAE) virus infection on the population of peripheral blood leukocytes in goats was evaluated. For this purpose two groups of adult dairy female goats were formed. The experimental group consisted of 17 goats, which had been naturally infected for many years. The control group comprised 29 non-infected goats, which originated from CAE-free herd. All goats were clinically healthy. Whole blood was collected and tested in hematological analyzer and light microscope to assess the total number of leukocytes and the percentage of four leukocyte populations - neutrophils, eosinophils, monocytes and lymphocytes. Then, flow cytometry with monoclonal antibodies against several surface antigens (namely CD14, CD2, B-B2, CD4, CD8h, TCR-N6, WC1-N2 and WC1-N3) was performed to assess the proportion of lymphocyte subpopulations. Statistically significant differences (α ≤ 0.01) were observed only in the subpopulations of T lymphocytes - percentage of all subpopulations were significantly higher in the group of seropositive goats. No statistically significant differences were revealed with respect to the total number of blood leukocytes, the average percentage of blood leukocyte populations and proportions of both T and B lymphocytes.

Silver nanoparticles, which have found a wide range of applications owing to their antimicrobial properties, are also recommended as dietary supplements in alternative medicine. Studies on rodents confirm that nanosilver is absorbed from the digestive tract into the bloodstream, which implies its possible interactions with leukocytes. The objective of the experiment discussed herein has been to determine the effect of 28-day oral administration of different doses (0.25, 2.5, 25 ppm) of commercial silver nanocolloid on hematological parameters, percentages of particular lymphocyte populations and activity of the peripheral blood leukocytes in mice. All the tested colloid doses decreased the counts of monocytes in the animals’ blood and induced phenotypic modifications among lymphocytes: an increase in CD4+/CD8+ T cell distribution, a decrease in NK and NKT cell distribution (doses of 0.25 and 2.5 ppm) and an increased CD4+:CD8+ ratio (25 ppm). Silver nanocolloid also affected the activity of cells, depressing the proliferation of lymphocytes (0.25 ppm) and stimulating phagocytosis as well as the respiratory burst of granulocytes and monocytes (all doses). The results verify the influence of orally administered silver colloid on the peripheral blood leukocytes, at the same time implying the potential risk of developing an inappropriate immune response of an organism exposed to prolonged administration of this substance.

The pathogenicity of RHDV (rabbit haemorrhagic disease virus) is mainly associated with its affinity to blood vessels, with causing disseminated intravascular coagulations (DIC), and with the stimulation of the host immune system. Moreover, there are implications suggesting that apoptosis may be a pivotal process in understanding the basis of viral haemorrhagic disease in rabbits - a serious infectious disease causing mortality to wild and domestic rabbits. The aim of this study is to evaluate, by means of flow cytometry, the dynamics of apoptosis in peripheral blood granulocytes and lymphocytes in rabbits experimentally infected with seven different strains of RHDV and so-called antigenic variants of RHDV denominated as RHDVa, i.e.: Hungarian 24V/89, 1447V/96, 72V/2003; Austrian 01-04, 237/04, V-412 and French 05-01. The results showed that all of the RHDV and RHDVa strains cause an increase in the number of apoptotic cells throughout the infection, which might indicate the need for further analysis of the importance of this process.

Studies identifying specific pathologically expressed genes have been performed on diseased myocardial tissue samples, however less invasive studies on gene expression of peripheral blood mononucleated cells give promising results. This study assessed transcriptomic data that may be used to evaluate Dachshunds with chronic mitral valve disease. Dachshunds with different stages of heart disease were compared to a control, healthy group. Microarray data analysis revealed clusters of patients with similar expression profiles. The clusters were compared to the clinical classification scheme. Unsupervised classification of the studied groups showed three clusters. Clinical and laboratory parameters of patients from the cluster 1 were in accordance with those found in patients without heart disease. Data obtained from patients from the cluster 3 were typical of advanced heart failure patients. Comparison of the cluster 1 and 3 groups revealed 1133 differentially expressed probes, 7 significantly regulated process pathways and 2 significantly regulated Ariadne Metabolic Pathways. This study may serve as a guideline for directing future research on gene expression in chronic mitral valve disease.

Sildenafil is a selective type 5 phosphodiesterase (PDE5) inhibitor, commonly used in the treatment of erectile dysfunction (ED) and pulmonary arterial hypertension (PAH). The results of recent studies suggest that this drug modulates function of the immune system. However, only few studies were performed in humans. Therefore, the aim of this study was to investigate the ability of sildenafil to regulate the production of pro-inflammatory cytokines including tumor necrosis factor alpha (TNF-α), interleukin (IL)-1β and IL-6 in peripheral blood mononuclear cells (PBMCs) from healthy men. Phorbol myristate acetate (PMA) - stimulated and unstimulated PBMCs from 16 participants were cultured in the absence or presence of sildenafil (400 ng/ml and 4 µg/ml). TNF-α, IL-1β and IL-6 concentrations in culture supernatants were measured by enzyme-linked immunosorbent assay (ELISA). Cell viability was assessed by trypan blue staining. Sildenafil at a concentration of 400 ng/ml significantly increased TNF-α production in stimulated PBMCs (p<0.05) but had no effects on IL-1β and IL-6. The drug did not affect PBMCs viability. This is the first report describing such effects of sildenafil in humans.

Platelets play a crucial role in hemostasis. Their activation has not yet been evaluated in healthy dogs with a normal and low platelet count. The aim of this study was to determine the influence of activators on platelet activation in dogs with a normal platelet count and asymptomatic thrombocytopenia. 72 clinically healthy dogs were enrolled. Patients were allocated into three groups. Group 1 consisted of 30 dogs with a normal platelet count, group 2 included 22 dogs with a platelet count between 100 and 200×109/l and group 3 consisted of 20 dogs with a platelet count lower than 100×109/l. Platelet rich-plasma (PRP) was obtained from peripheral blood samples using tripotassium ethylenediaminetetraacetic acid (K3-EDTA) as anticoagulant. Next, platelets were stimulated using phorbol-12-myristate-13-acetate or thrombin, stabilized using procaine or left unstimulated. The expression of CD51 and CD41/CD61 was evaluated. Co-expression of CD41/CD61 and Annexin V served as a marker of platelet activation. The expression of CD41/CD61 and CD51 did not differ between the 3 groups. Thrombin-stimulated platelets had a significantly higher activity in dogs with a normal platelet count than in dogs with asymptomatic thrombocytopenia. Procaine inhibited platelet activity in all groups. In conclusion, activation of platelets of healthy dogs in vitro varied depending on the platelet count and platelet activator.

The aim our study was qualitative and quantitative analysis of white blood morphometric elements of peripheral blood (determining the quantity, blood cells dimension and several hematological values) in White Stork chicks. One of the aims was to indicate whether the sex relevantly influences the variety of the examined white blood indicators. White blood cells parameters of 53 White Stork chicks, with molecularly marked sex, was examined. Blood samples were collected in southern Poland (around Krapkowice town, near Opole city). Lymphocytes of White Storks (mean 37% for females and males) were identified as round cells with dark purple non-lobed, eccentrically positioned nucleus. Among the whole population we differentiated small lymphocytes: with diameters of 5.31 ± 0.65 μm in males and 5.57 ± 0.59 μm in females, and large lymphocytes with a diameter of 8.10 ± 0.66 μm and 8.28 ± 0.74 μm, respectively in females and males. Monocytes (mean 8% for female and males) were the largest leukocytes found in the blood film of White Stork, measuring 13.40 ± 0.97 μm for males and 13.09 ± 1.05 μm for females in diameter. The cytoplasm was abundant and it stained blue-gray, very often contained vacuoles. Heterophils (mean 42.7%) were the largest in granular leukocytes group. They were round and 11.14 ± 0.65 μm for males and 11.01 ± 0.48 μm for females in diameter. The nucleus of heterophils was lobed, usually with two or three lobes. The cytoplasm contained brick-red, elongated granules. Eosinophils (mean 9.44%) were round cells, with a diameter of 10.72 ± 0.49 μm and 10.97 ± 0.53 μm, respectively in males and females. The nucleus was lobed and mostly stained clear blue and contained red-orange, round or rod-shaped granules. Basophils (mean 1.84%) were round and contained dark blue granules, with average of 9.56 ± 0.78 and 9.13 ± 0.84 μm in diameter, for males and females. The nucleus was usually non-lobed. The H/L ratio was 4/4 for both sexes. No significant differences in levels and types of leukocytes between male and female juvenile storks have been observed.
Celem naszego badania była analiza jakościowa i ilościowa morfometrycznych elementów krwi obwodowej (określenie ilości, wymiarów krwinek i kilku wartości hematologicznych) u piskląt bociana białego. Jednym z celów było wskazanie, czy płeć ma istotny wpływ na różnorodność badanych wskaźników krwi białej. Zbadano parametry białokrwinkowe 53 piskląt bociana białego, z molekularnie oznaczoną płcią. Próbki krwi pobierano w południowej Polsce (okolice miasta Krapkowice, w pobliżu miasta Opola. Limfocyty bocianów białych (średnio 37% dla samic i samców) zostały zidentyfikowane, jako okrągłe komórki z ciemnofioletowym, ekscentrycznie usytuowanym jądrem. Spośród całej populacji wyodrębniliśmy małe limfocyty: o średnicy 5,31 ± 0,65 μm u samców i 5,57± 0,59 μm u samic oraz limfocyty duże o średnicy 8,10 ± 0,66 μm i 8,28 ± 0,74 μm, odpowiednio u samic i samców. Monocyty (średnio 8% dla samic i samców) były największymi leukocytami znalezionymi we krwi obwodowej bociana białego, o średnicy 13,40 ± 0,97 μm u samców i 13,09 ± 1,05 μm u samic. Cytoplazma była obfita i zabarwiona na niebiesko-szaro, często zawierała wakuole. Heterofile (średnio 42,7%) były największymi krwinkami wśród granulocytów. Były okrągłe i miały średnicę 11,14 ± 0,65 μm u samców i 11,01 ± 0,48 μm u samic. Jądro heterofilów było płatowate, zwykle z dwoma lub trzema płatami. Cytoplazma zawierała ceglasto-czerwone, wydłużone ziarnistości. Eozynofile ( średnio 9,44%) były okrągłymi komórkami o średnicy 10,72 ± 0,49 μm i 10,97 ± 0,53 μm, odpowiednio u samców i samic. Jądro było płatowate i przeważnie wybarwione na jasnoniebiesko, cytoplazma zawierała czerwono-pomarańczowe, okrągłe ziarnistości. Bazofile (średnio 1,84%) były okrągłe i zawierały ciemnoniebieskie ziarnistości o średniej średnicy 9,56 ± 0,78 μm i 9,13 ± 0,84 μm u samców i samic. Współczynnik H/L wynosił 4/4 dla obu płci. Nie zaobserwowano istotnych różnic w liczbie jak i frekwencji poszczególnych rodzajów krwinek białych między młodymi bocianami płci męskiej i żeńskiej.

Introduction. White blood cell (WBC) count constitutes a part of routine peripheral blood examination (FBC, full blood count). Precise analysis of leukocytes’ parameters in blood smear is usually performed only when leukopenia or leukocytosis is found. We aimed to assess the usefulness of leukocytes’ smear test in predicting in-hospital death of patients undergoing high-risk gastrointestinal (GI) surgery. Materials & Methods. We prospectively enrolled 101 subjects undergoing high-risk GI surgery from 01.01.2017 till 31.12.2017. Blood tests were performed preoperatively, with analysis focused on the assessment of total WBC count, as well as the count of neutrophils (NEUT), lymphocytes (LYM), monocytes (MONO), eosinophils (EOS) and basophils (BASO). Indices of NEUT/LYM (NLR) and MONO/LYM (MLR) were calculated. In-hospital mortality was considered the outcome. Results. Mortality reached 5%. There was no significant difference in WBC count between survivors and the deceased (p=0.2) and WBC failed to predict the outcome (AUC=0.69; p=0.3). MONO (p=0.009) and BASO (p=0.02) counts, as well as MLR (p=0.007) were significantly higher in patients who died. MONO count and MLR index predicted in-hospital death with good accuracy, respectively: AUC[MONO]=0.85 (p<0.001) and AUC[MLR]=0.86 (p<0.001). Other investigated parameters played no significant role in outcome prediction. Conclusion. Routine peripheral blood smear evaluation should be considered in all patients undergoing high-risk GI surgery, because the number of monocytes can be a valuable predictor of in-hospital death.

The aim of the study was to evaluate the effects of long-term green tea extract (GTE) supplementation on white blood cells (WBC) in CrossFit-trained and untrained men. Twenty-eight men were recruited for this study including CrossFit-trained (CF-TR) and untrained (CF-UNT) men. The study was carried out in two phases. Phase 1 involved non-supplemented untrained and trained men. Phase 2 involved the same groups but after 6-week supplementation with 2 GTE capsules once daily. Body weight, body composition and the number and percentage of peripheral blood leukocyte populations were assessed in each phase. The results revealed that the long-term green tea supplementation did not have a significant effect on body weight, body composition and the total white blood cell count of the study participants. However, the percentage of peripheral blood eosinophils increased while monocyte and neutrophil counts decreased. The changes in white blood cells were less pronounced in CrossFit-trained compared to control participants. Summing up, it should be noted that long-term GTE supplementation in CrossFit-trained men did not result in significant changes in body weight and composition and the total white blood cell count. However, the changes in leukocyte subsets were less pronounced in CrossFit-trained compared to untrained participants.

Opis:
Rowing is a sport discipline, which requires extreme physical strength and endurance and appropriate aerobic and anaerobic capacity as well. However, when the workout intensity and load is very high, exercise is associated with temporary changes in cellular metabolism and the immune system. The study included one male rower aged 28 years - the highly-skilled and experienced athlete. We determined basic cardiorespiratory fitness measures, complete blood count, and 24 clinical chemistry parameters including relevant biochemical and haematological parameters and matrix metaloproteinases activities. Maximal exercise on the rowing ergometer induced 2-fold increase in absolute counts of all leukocytes subsets. There was observed an increase in C-reactive protein concentrations as well. MMP-9 activity increased 1,3-fold compared to the baseline value. Exhaustive exercise caused significant changes in creatinine and urea serum levels, but the most prominent changes were found in total and direct bilirubin concentrations. Maximal exercise induced also a decrease in the iron and magnesium levels. No changes in ALT, GGT and ALP activity were observed, while increase in CK, AST and LDH activity in post-exercise time and the decrease during the recovery was found. Therefore acute specialized movement on the rowing ergometer is not the cause of muscular damage, but rather indicate efficient adaptation to the physical exercise. Moreover, it seems that maximal exercise induces an inflammatory response characterized by greater count of all subpopulations of leukocytes, elevated levels of CRP and MMP-9 serum activity.

Nowadays, increasing attention has been focused on relation between increased D-dimer levels and cancer among patients without detectable thrombosis. The aim of the study was to measure plasma D-dimer levels in portal and peripheral blood in pancreatic cancer patients with absence of venous thromboembolism. Material and methods. Fifteen consecutive patients hospitalized in the Department of General and Transplant Surgery of Medical University in Łódź, from January to March 2012 who underwent surgery due to a pancreatic cancer were enrolled. At laparotomy, portal and peripheral blood were sampled concurrently. D-dimer and fibrinogen levels were measured. Moreover, to investigate overall coagulation function prothrombin time (PT), prothrombin index (PI), international normalized ratio (INR), thrombin time (TT), activated partial thromboplastin time (APTT), TT and APTT index were evaluated. Results. Peripheral plasma D-dimmer levels above normal range were found in 10/15 patients (66,67%), whereas D-dimer above normal values were confirmed in all portal blood samples. Mean D-dimer values were higher in portal than in peripheral blood (3279.37 vs 824.64, by 297%, p=0,025). These discrepancies were accompanied by normal limits of portal and peripheral levels of fibrinogen and comparable coagulation function indexes. Conclusion. Our preliminary study showed the close relation between activation of hemostasis, reflected by elevated D-dimers in portal blood and presence of pancreatic cancer. These data suggest that measurement of portal blood D-dimer levels may be a potentially useful technique for screening the pancreatic cancer.

The aim of this study was to use a two-marker assay for the detection of breast cancer cells circulating in patients' blood. We have applied a PCR-based methodology to follow up the possibility of the development of metastatic disease in stage I and II patients who had undergone curative surgery. Since the number of circulating cancer cells in peripheral blood is very low, the technique for their detection needs to be not only highly sensitive, but also very specific. The reverse transcriptase-polymerase chain reaction (RT-PCR) technique may improve the sensitivity of breast cancer cell detection up to only a few cells per one million. The principle of the RT-PCR assay is to amplify a messenger RNA characteristic for breast epithelial cells in a blood sample. Since we do not expect such cells to be circulating in peripheral blood of healthy subjects, detection of the characteristic mRNA should indicate the presence of circulating breast cancer cells. We analyzed the usefulness of three mRNA markers: cytokeratin 19 (CK19), mammaglobin (hMAM) and β subunit of human chorionic gonadotropin (β-hCG) for this test. Blood samples (112) were obtained from 55 patients, in stages I and II, with or without metastasis to regional lymph nodes (N0 or N1). We found that a two-marker assay increases the sensitivity of detection of breast cancer cells in comparison with a single-marker one. Combination of two tumor-specific mRNA markers, hMAM/CK19 or β-hCG/CK19, allowed the detection of circulating breast cancer cells in 65% of N1 patients and 38% of N0 patients. By comparison, the combination hMAM/β-hCG allowed the detection of circulating breast cancer cells in the blood of 68% of N1 patients and 46% of N0 patients. Addition of the third marker did not significantly increase the detection sensitivity.

As far as the detection of metal genotoxicity in fish is concerned, micronucleus (MN) test is considered an extremely suitable measure. In this study, frequencies of micronucleated erythrocytes were scored in peripheral blood of catfish, Heteropneustes fossilis (bloch) after acute in-vivo exposure of zinc at different concentrations (5, 10 and 30 ppm) in the laboratory condition. These three concentrations of zinc were tested at different durations such as 24h, 48h, 72h and 96h respectively. Highly significant (P < 0.001) increased values were obtained for MN frequencies in the peripheral erythrocytes of exposed fishes compared to control groups of fishes. These results confirm that dose- and time-dependent micronucleation in the peripheral erythrocytes of fish after short-term exposure to zinc could provide valuable information regarding zinc containing effluent quality and also help in genetic biomonitoring with this test model. In this context safe concentration of zinc vis-a-vis genotoxicity range could be evaluated for future studies.

The objective of the study was to determine values of apoptosis for peripheral blood lymphocytes and granulocytes, including dependency on the season of the year, in Polish mixed-breed rabbits and in mixed-breed rabbits with the addition of blood of meat-breed rabbits. The study was carried out in four seasons of the year (spring, summer, autumn, winter) involving 120 Polish mixed-breed rabbits – group I, and 120 Polish mixed-breed rabbits with addition of meat-breed rabbit blood – group II. Assessment of apoptosis of lymphocytes and granulocytes was performed using as FACScan cytometer by Becton Dickinson with FACSDiva software (USA), using as ApoFluor®Green Caspase reagent kit (MP Biomedicals, USA) to detect the activity of the total caspase pool 1, 3, 4, 5, 6, 7, 8, 9 in granulocytes and lymphocytes of rabbit peripheral blood. The results for apoptosis of lymphocytes and granulocytes in peripheral blood in the animals investigated (group I and II) were subjected to statistical analysis with the t’Student test at p=0.05. It was noted that, in rabbits from group I, the values for apoptosis of lymphocytes were the highest in winter and autumn (36.02% and 31.24%, respectively), and the lowest in spring and summer (26.73% and 22.72%, respectively), whereas in the case of granulocytes the highest values were in summer and spring (14.69% and 12.95%, respectively), and the lowest in winter and autumn (8.16% and 8.57%, respectively). In mixed-breed rabbits with the addition of meat-breed blood (group II), the values for apoptosis of lympocytes were the highest in spring (29.13%), and the lowest in summer (25.43%); whereas in the case of granulocytes the highest values were in summer and spring (14.0% and 11.15%, respectively), and the lowest in autumn and winter (7.46% and 7.64%, respectively).

The objective of this study was to determine the effect of Biolex-MB40 on the phagocytic activity and oxidative metabolism of peripheral blood granulocytes and monocytes in lambs. The experimental material comprised 32 lambs aged 30 ± 3 days, divided into two equal groups: control and experimental. Experimental group animals were fed a diet supplemented with the Biolex-MB40 (Saccharomyces cerevisiae) in the amount of 3 g/kg of the concentrate. At the beginning of the experiment (day 0) and on experimental days 15, 30 and 60, blood was sampled from the jugular vein to determine and compare the phagocytic activity (PHAGOTEST) and oxidative metabolism (BURSTTEST) of peripheral blood granulocytes and monocytes by flow cytometry. Based on the results of an analysis of granulocyte and monocyte phagocytic activity, statistically higher levels of phagocytic activity were observed in the group of lambs administered Biolex-MB40 than in the control animals, expressed in terms of the percentage of phagocytic cells as well as mean fluorescence intensity. Biolex-MB40 also had a positive effect on the oxidative metabolism of both granulocytes and monocytes after stimulation with Escherichia coli bacteria and with PMA (4-phorbol-12-β-myristate-13-acetate), expressed in terms of the percentage of oxidative metabolism as well as mean fluorescence intensity.

Opis:
In Poland, rabbit is a highly valued animal, due to dietetic and flavour values of its meat, but above all, rabbits tend to be commonly used laboratory animals. The aim of the study was developing standards for counts of B-cells with CD19+ receptor, T-cells with CD5+ receptor, and their subpopulations, namely T-cells with CD4+, CD8+ and CD25+ receptor in the peripheral blood of mixed-breed Polish rabbits with addition of blood of meet breeds, including the assessment of the impact of four seasons of the year and animal sex on the values of the immunological parameters determined. The results showed that the counts of B- and T-cells and their subpopulations in peripheral blood remain within the following ranges: for CD19+ B-cells: 1.05 - 3.05%, for CD5+ T-cells: 34.00 - 43.07%, CD4+ T-cells: 23.52 - 33.23%, CD8+ T-cells: 12.55 - 17.30%, whereas for CD25+ T-cells: 0.72 - 2.81%. As it comes to the season of the year, it was observed that it principally affects the values of CD25+ T-cells, while in the case of rabbit sex, more changes were found in females.

The main aim of this study was to compare the reaction of quiescent and proliferating, i.e. phytohemagglutinin (PHA)-stimulated, human peripheral blood mononuclear cells (PBMCs) to γ-radiation, and analyse changes of proteins related to repair of DNA damage and apoptosis, such as γH2A.X, p53, p53 phosphorylation at serines-15 and -392, and p21 and their dose dependence. Freshly isolated PBMCs in peripheral blood are predominantly quiescent, in G0 phase, and with very low amounts of proteins p53 and p21. Using confocal microscopy we detected dose dependent (0.5-5 Gy) induction of foci containing γH2A.X (1 h after γ-ray exposure), which are formed around radiation-induced double strand breaks of DNA. Apoptosis was detected from 24 h after irradiation by the dose of 4 Gy onwards by Annexin V binding and lamin B cleavage. Seventy two hours after irradiation 70% of CD3+ lymphocytes were A+. Neither increase in p53 nor its phosphorylation on serine-392 after irradiation was detected in these cells. However, massive increase in p21 (cyclin-dependent kinase inhibitor 1A) was detected after irradiation, which can be responsible for late occurrence of apoptosis in these quiescent cells. PHA-stimulation itself (72 h) caused an increase in early apoptosis (A+PI-) in comparison to non-stimulated PBMCs (38% A+ resp. 13.4%). After PHA-stimulation also the amount of γH2A.X, p53, and p21 increased, but no phosphorylation of p53 on serine-392 or -15 was detected. Reaction to γ-radiation was different in PHA-stimulated lymphocytes: the p53 pathway was activated and p53 was phosphorylated on serines-15 and -392 4 h after irradiation by the dose of 4 Gy. Phosphorylation of p53 at serine-15 increased in a dose-dependent manner in the studied dose range 0.2-7.5 Gy. Also the amount of p21 increased after irradiation. Seventy two hours after irradiation of PHA-stimulated CD3+ T lymphocytes by the dose of 4 Gy 65% of cells were A+.

The degree of activation of cells involved in cellular immune re-sponse against tumor antigens (cytotoxic lymphocytes Te) as well as efficiency of the mechanisms which promote immunosuppression (Treg — regulatory cells CD4±CD25±Foxp3±) may determine the course of the neoplastic disease. The aim of this study was to assess the function of autologous peripheral blood mononuclear cells (PBMCs) involved in the immunological processes on the basis of expression of Foxp3 and RORgamma t molecules as well as analysis of the relationships with clinical and morphological features of the tumor (pT and pN stage, G feature, degree of invasiveness according to the TFG classification) in laryngeal carcinoma. Materia! and methods: The analysis included a group of 59 patients with verified squamous celt carcinoma of the larynx. In the pathologic evaluation pTNM classification criteria, depth of invasion and degree of histological dif-ferentiation were used. Expression levels of mRNA for Foxp3 and RORgamma t in peripheral blood mononuclear cells by quantitative analysis of the amplified product in real time (real-time RT2-PCR) were evaluated. The level of Foxp3 and RORgamma t protein expression by Western blot analysis was determined. Results: In squamous celt carcinomas of the larynx, with the highest tumor aggressiveness the significantly highest level of mRNA and protein expres-sion for Foxp3 molecule were observed. The severity of Foxp3 expression at both gene and protein level were positively linearly correlated with the degree of local extent of the tumor (pT3-4), depth of invasion (invasion of cartilage) and the degree of histological differentiation (low-differentiated tumors G3). In the study group of laryngeal cancers significantly lower level of RORgamma t expression in carcinomas with less invasive changes (pT1-2, high-differentiated tumors G1, carcinomas with microinvasion without evi-dence of invasion beyond the lamina propria) was also noted. Conclusions: The study results indicate the important role of immune celt activity as indicators of advancement of clinical and morphological changes in squamous celt carcinoma of the larynx.

It is a well known, that the physical activity promotes mental health. Physically active people relatively rarely suffer from psychosomatic and depressive disorders. It is possible that the differences in gene activity in peripheral blood leukocytes may be associated with depression, especially genes participating in inflammatory response. Thus, the aim of our study was to investigate the levels of IL6 and IL10 mRNA and IL10/IL6 ratio in peripheral blood leukocytes in healthy, physically active individuals. One hundred healthy young men (20-23 years old) participated in this study. All of them declared regular physical activity. Participants were non-smokers, and consumed alcohol occasionally. To access genes expression, 2 ml of venous blood was collected. RNA isolation was performed and then the relative expression of IL6 and IL10 was calculated using quantitative real-time polymerase chain reaction (Q-RT-PCR). Pro and anti-inflammatory balance was calculated as 2^relative of IL10/ 2^relative expression of IL6. Low expression of tested genes was found in healthy young men. Mean expression for IL6 was 2^0.051 (n=90) and for IL10 2^0.08 (n=98). Mean ratio IL10/IL6 was 2^1.58. Higher expression of IL10 compared to IL6 may be essential not only for the physical performance but also for the mental health. Diverse reports in the literature may be associated with choosing various control groups, i.e. sedentary or older individuals. It is possible, that measuring the expression of IL6 and IL10 (especially the ratio IL10/IL6) in peripheral blood leukocytes may be useful in the assessment of depressive disorders. Thus, molecular study of active young men can confirm the need for physical activity among people suffering from depression, but further studies are needed, particularly among people with psychosomatic disorders.

Opis:
The first symptoms caused by heavy metals poisoning are usually non-specific, therefore their diagnosis requires specialized knowledge and experience. Incorrect diagnosis can lead to various disorders and irreversible changes in patient’s health. Lead poisoning is one of the heavy metals poisoning which is associated with non-specific symptoms and may cause a broad range of biochemical, physiological and behavioral disfunctions. Lead is commonly found in industry and manufacturing and in 2017, lead poisoning caused over a million deaths worldwide. Due to non-specific symptoms, lead poisoning is often diagnosed too late or incorrectly. Early symptoms are headache and stomachache, which potentiate with metal concentration. Lead poisoning increasing risk of anemia due to inhibition the ability to produce hemoglobin by interfering the heme biosynthesis pathway and decreasing red blood cell survival. Because of regularly occurred disorders of hematopoiesis and the presence of atypical cell in smears, diagnosis of lead poisoning that gives relevant information is bone marrow and peripheral blood smear study. The manual smear test made by qualified technician involves the evaluation of the preparation by using an optical microscope. Manual microscopic examination of blood cells is time-consuming and subjective. Automation of this process would enable to reduce the risk of human failure and solve the problem with lack of professional staff. Traditional machine learning is the most popular approach to automating microscopic smear testing. This method consists of the following stages: microscopic image acquisition, preprocessing, cell segmentation, feature extraction, followed by classification into types, artifacts and atypical cells. This work presents the recent methods proposed to automate the analysis of peripheral blood and bone marrow smears using traditional machine learning methods. A review of different machine learning methods was carried out, focusing on the presentation of an algorithm for the construction of automatic blood cell classifiers.
Pierwsze objawy pacjenta po zatruciu metalami ciężkimi są najczęściej nieswoiste, dlatego ich diagnostyka jest zadaniem wymagającym specjalistycznej wiedzy i doświadczenia. Nieprawidłowa diagnoza może prowadzić do nieodwracalnych zmian w stanie zdrowia pacjenta. Jednym z metali ciężkich, którym zatrucie wiąże się z nieswoistymi objawami, a może prowadzić do poważnych zaburzeń neurologicznych i hematologicznych jest ołów. Ołów jest metalem ciężkim powszechnie występującym w przemyśle i produkcji. W 2017 roku zatrucie tym metalem było przyczyną ponad miliona zgonów na świecie. Z powodu nieswoistych objawów zatrucie ołowiem często jest diagnozowane nieprawidłowo i zbyt późno. Zatrucie metalami ciężkimi może prowadzić do zaburzeń hematopoezy i pojawienia się w rozmazach krwi i szpiku kostnego komórek atypowych, w związku z czym badaniem niosącym istotną informację w diagnostyce zatrucia jest ocena komórek w rozmazach. Analiza rozmazu manualnego polega na ocenie preparatu przy użyciu mikroskopu optycznego oraz szczegółowej interpretacji obrazu mikroskopowego wykonywanej przez wykwalifikowany personel. Automatyzacja tego procesu pozwoliłaby na ograniczenie ryzyka pomyłki spowodowanej błędem ludzkim oraz rozwiązanie problemu braku doświadczonego personelu. Najpopularniejszym podejściem wykorzystywanym w automatyzacji mikroskopowego badania rozmazu jest tradycyjne uczenie maszynowe. Metoda ta, składa się z kilku etapów: zbieranie obrazów mikroskopowych i ich wstępna obróbka, wyodrębnienie komórek oraz ich charakterystycznych cech, a następnie klasyfikacja na rodzaje, artefakty i komórki atypowe. W pracy opisano aktualnie proponowane sposoby automatyzacji badania rozmazu krwi obwodowej i szpiku kostnego, wykorzystujące tradycyjne metody uczenia maszynowego. Przeprowadzono przegląd metod uczenia maszynowego, skupiając się na przedstawieniu algorytmu postępowania w konstrukcji automatycznych klasyfikatorów komórek krwi.

Ionizing radiation affects the expression of adhesive and co-stimulatory molecules in lymphocytes. The physiological function of cellular isoform of prion protein (PrPc) is little known. Evidences indicate a link between lymphocytes activation and PrPc expression on their surface; however, no direct effect of radiation on PrPc level in these cells was investigated. The objective of this study was to determinate the effect of low doses of ionizing radiation on the expression of PrPc on the surface peripheral blood lymphocytes in the women operating X-ray equipment. In 36 female workers and 30 persons of the control group the PrPc expression on CD3 (T lymphocytes), CD4 (T helper), CD8 (T cytotoxic) and CD19 (B lymphocytes), as well as the percentage of lymphocytes with PrPc on their surface, were tested. Subgroups with respect to age and length of employment were selected. A signifi cant increase was observed in PrPc expression on CD3 and CD4 with lowered PrPc level on CD8 and percentage of CD8 cells with PrPc in workers compared to control. The PrPc level did not show signifi cant changes in subgroups in relation to age (below and over 40 years old) both in the investigated and control groups, whereas a lower percentage of PrPc expressing CD19 cells showed in employed women below 40 years of age. A signifi cant decrease was found in PrPc expression on the surface of CD3, CD4 and CD8 cells in the subgroup employed for over 10 years than in the subgroup with less than 10 years of employment.

Megachemotherapy with autologous peripheral blood stem cell transplantation (auto-PBSCT) is a standard treatment option in patients below 70 years of age with multiple myeloma (MM) as well as with relapsed and refractory lymphomas. Recombinant granulocyte colony-stimulating factors (G-CSF) are commonly used to accelerate bone marrow recovery after chemotherapy and reduce the duration of severe neutropenia. Lipegfilgrastim is a glicopegylated G-CSF with prolonged action registered for adult patients with malignant neoplasms in order to reduce the duration of neutropenia and the incidence of febrile neutropenia (FN). So far, there is not enough data to confirm the effectiveness and safety of this drug in patients with hematological malignancies including those undergoing auto-PBSCT. The aim of this study was to determine the effect of lipegfilgrastim on hematopoietic regeneration and supportive care after auto-PBSCT in patients with lymphoproliferative malignancies. The study population consisted of 30 patients (12 female and 18 male; median age: 50 years ± 13), including 13 patients with MM, 5 with Hodgkin’s lymphoma (HL) and 12 with non-Hodgkin’s lymphoma (nHL). The median number of transplanted CD34+ cells was 3.96 ± 1.56 × 10^6/kg of body mass. On day +1 after auto-PBSCT, the patients received lipegfilgrastim in a single 6 mg subcutaneous injection. The control group consisted of 32 patients (13 female and 19 male; median age: 50 years ± 6.4), including 13 with MM, 8 with HL and 11 with nHL, who received subcutaneous filgrastim in a dose of 5 μg/kg/day from day +1 after transplantation and continued to an absolute neutrophil count (ANC) > 1.5 × 10^9/L. There was no significant difference in the time of regeneration ANC > 0.5 × 10^9/L which was 10.65 ± 1.00 vs. 11.51 ± 2.29 days respectively in the study and control group. Similar observations were noted regarding the duration of febrile neutropenia (2.16 ± 2.22 vs. 1.70 ± 4.17 days; p = 0.998), regeneration of platelets (PLT) > 20 × 10^9/L (12.41 ± 2.41 vs. 13.82 ± 4.48 days; p = 0.233) and demand for transfusion of red blood cells (0.76 ± 1.07 vs. 1.33 ± 2.33 units; p = 0.414) and platelets (11.5 ± 6.9 vs. 19.2 ± 17.7 units; p = 0.08). Different results were observed for the length of hospitalization, which was significantly shorter in the lipegfilgrastim group (16.14 ± 14 vs. 24.46 ± 6.79 days; p = 0.000). Lipegfilgrastim is as effective as filgrastim with regards to the regeneration of the hematopoietic system, duration of febrile neutropenia, demand for transfusion of blood products and significantly reduces hospitalization in patients with lymphoproliferative malignancies after auto-PBSCT.

This paper attempts to study the dynamics of apoptosis of granulocytes and lymphocytes in peripheral blood in rabbits infected with haemagglutinating (Vt97, Triptis, Hartmannsdorf) and non-haemagglutinating (Pv97, 9905 RHDVa) antigenic variants of the RHD virus. The pathogenicity of those antigenic variants was also assessed by recording the mortality of the infected animals. The animals were infected with antigenic variants and blood was sampled at hour 0,4,8,12,24,36 p.i. and the percentage of apoptotic granulocytes and lymphocytes was measured with the use of flow cytometry. The results of the study showed that apoptosis is included during RHDV infection, as the number of apoptotic granulocytes and lymphocytes increases throughout the experiment; depending on the antigenic variant, apoptosis joins in at 4-8-12 h p.i. and lasts until 24-36 h p.i. Furthermore, the mortality of rabbits infected with the examined strains of RHD virus varied from 30% to 100%. This study performed for the first time in this manner, indicates the importance of apoptosis during infection with the RHD virus.

The present study was undertaken to contribute to the characterization of the degree of variability in baseline damage in white blood cells from control population, and to investigate how this variability is associated with external and internal factors. Altogether 170 healthy volunteers, randomly selected from the general population of the Republic of Croatia, participated in the study. Two sensitive tests: the alkaline comet assay and the chromosome aberration test were applied to study the background levels of DNA damage in their white blood cells. The results point to inter-individual differences, indicating different genome sensitivity. As revealed by both assays, the background levels of DNA damage were mostly influenced by smoking habit as well as medical exposure (especially to diagnostic X-rays). Sex and age of subjects did not significantly influence the values of DNA damage recorded in the white blood cells. Although higher levels of DNA damage were recorded in blood samples collected during winter and autumn, they were mostly influenced by medicinal exposure and smoking habit. Statistical evaluation of the data confirmed that a positive correlation exists between DNA migration and the number of long-tailed nuclei found with the comet assay and the total number of chromosome aberrations. The data obtained can serve as control values in forthcoming biomonitoring studies.

The aim of the study was to determine whether treatment with recommended doses of meloxicam or flunixin had an effect on the apoptosis of peripheral blood T lymphocytes in calves. The study was carried out on 4-5 months old calves (n = 24, 8 per group). Experimental animals were injected subcutaneously with a single dose of 0.5 mg . kg-1 of meloxicam or intravenously with 3 doses of 2.2 mg . kg-1 day-1 of flunixin. The non-treatment animals served as control. Blood samples were taken at day 0 and at days 1, 2, 3, 5, 7 and 14 after the first NSAIDs injection. Apoptosis was determined by flow cytometry using Annexin V-PE/7-AAD staining. The kinetic analysis of apoptosis in the total lymphocyte population, as well as in the CD4+ and CD8+ subsets did not reveal significant differences in the frequency of early apoptotic cells between control and experimental groups throughout the period studied. Although, 24 h after administration of the first dose of NSAIDs, late-stage apoptosis/necrosis was significantly increased in the total lymphocyte population (the meloxicam group), as well as in the CD4+ (the meloxicam group and the flunixin group) and CD8+ (the flunixin group) subsets of T cells. However, this disturbance was transient, relatively poorly expressed and, thus, unlikely to be of clinical significance. Our results indicate that the use of meloxicam or flunixin in accordance with the recommended dosage regimen in cattle do not have a clinically significant influence on apoptosis of peripheral blood T cells.

The aim of this study was to investigate changes in selected parameters of cellular immune response in the conditions of endotoxin fever and pyrogenic tolerance in pigeons. On the first day of observation the experimental birds (n=18) were intravenously injected with Escherichia coli LPS at a dose of 10 μg/kg b.w., while the control animals (n=6) received apyrogenic physiological saline also in the form of injection. On the second and the third day of the experiment LPS was injected additionally at 24 h intervals. Four and a half hours after the saline and pyrogen administration blood samples were collected from the control and experimental pigeons. The following immunological assays were performed: WBC, leucogram and immunophenotyping of lymphocyte subsets in peripheral blood, i.e. CD 3+ (T lymphocytes), CD 4+ (T helper lymphocytes) and CD 8+ (T suppressor/ cytotoxic lymphocytes) cells. In the conditions of endotoxin fever (i.e. after the first LPS injection) leucopenia, monocytopenia, heterophilia and eosinophilia were observed. Additionally, the immunophenotyping of peripheral blood lymphocytes indicated an increase in percentage of CD 3+, CD 4+ and CD 8+ cells in response to the single injection of LPS. In contrast, the consecutive injections of LPS, which created a pyrogenic tolerance effect, caused a decrease in WBC value, heteropenia, eosinopenia and lymphocytosis. Moreover, during this state an increase in percentage of CD 3+ and CD 8+ cells was demonstrated in contrast to the percentage of CD 4+ lymphocytes. The general tendencies in cellular immune response of the affected pigeons in the conditions of endotoxin fever and pyrogenic tolerance aim at activation of defence mechanisms against LPS for its prompt elimination from the animal’s organism.

Exposure to phenol causes ultrastructural disorders in supporting and receptor cells in the peripheral segment of the olfactory organ of yellowfin (Cottocomephorus grewingkii) and in cells in peripheral blood of stone sculpins (Paracottus knerii), yellowfin and perch (Perca fluviatilis). Identical responses to phenol exposure in the blood cells were observed in endemic Lake Baikal fish and in nonendemic fish. Differences were found only in the rate and intensity of the recovery response. Fast activation of immune processes were found in the coastal nonendemic perch, but they were slow in the stone sculpin, a coastal endemic species.
