Opis:
Human peripheral blood cells stimulated by phytohemagglutinin (which serve as a model of cancerous cells) and resting cells were incubated in dimethyl sulfoxide solutions of various phthalocyanines. In order to diminish the influence of atmospheric oxygen the cells were embedded in a polymer (polyvinyl alcohol) film. Fluorescence spectra of the samples were measured over two regions of excitation wavelengths: at 405 nm (predominant absorption of the cell material) and in the regions of strong absorption of phthalocyanines (at about 605 nm and 337 nm). The intrinsic emission of cell material became changed as a result both of cells' stimulation and of incubation of cells in dye solution. In most cases the stimulated cells when stained by dye exhibited higher long wavelength fluorescence intensity than resting cells. This suggests higher efficiency of dye incorporation into cancerous cells than into healthy cells. The absorption spectra of samples were also measured. The spectra of various phthalocyanines in incubation solvent, in polymer and in the cells embedded in polymer, were compared. The comparison of properties of the cells stimulated for different time periods enabled to establish the conditions of stimulation creating a population of cells incorporating a large number of sensitizing molecules.

The interactions of two metal-free phthalocyanines [(H2Pc) and Solar Pc (with four peripherical groups: SO2N(CH2CH2OH)2)] and of one metal substituted dye (CoPc) with resting and stimulated human peripheral blood mononuclear cells (PBMC) were compared. The absorption, fluorescence, photoacoustic and EPR spectra of both resting cells and cells stimulated by phytohaemagglutinin, incubated in dimethyl sulfoxide (DMSO) with very low or 95% water content and with or without dye addition, were measured. The fate of the light absorbed by the samples was investigated. It is known that singlet oxygen production is crucial for photodynamic action of dyes. Thermal deactivation and luminescence emission compete with this process, so investigation of these alternative paths of sensitizer deactivation provides information about photodynamic action. The incorporation of the investigated dyes into cells and the perturbation of the cell structure caused by the dyes, the incubation solvent and the activator were investigated by comparing the spectral properties of PBMC before and after stimulation and incubation. Incubation of the cells for 1 h in a solution of Solar Pc in 99.5% aqueous DMSO, resulted in an efficient dye incorporation which was highly selective. Solar Pc being introduced much more efficiently into stimulated cells than into resting cells.

During the last few decades, owing to their unique properties, gold nanoparticles (AuNPs) have found numerous biomedical applications. Studies on rodents prove that AuNPs entering an organism easily reach the bloodstream and undergo wide tissue distribution. The presence of nanoparticles inside blood and bone marrow cells of exposed animals may implicate its influence on hematopoesis and the functions of peripheral blood leukocytes. The aim of this study was to determine the effect of oral administration of commercial gold nanocolloid, recommended by the producer as a dietary supplement, on the percentage of lymphocyte populations and proliferative response, as well as the activity of phagocytes in the peripheral blood of mice. The colloid was given to the animals in three different doses (0.25, 2.5, 25 ppm), for three different time periods (7, 14, 28 days). Mice given nanoparticles showed increased activity of phagocytes and some changes in the lymphocyte phenotypes. The elevated activity of granulocytes and monocytes, in terms of both phagocytic and respiratory burst activity, was transient and noticed only after a short time of administration, which may indicate some adaptability of blood phagocytes to prolonged presence of gold nanoparticles in the body. However, phenotypic modifications among lymphocytes in the group of animals given the middle dose of colloid (i.e. increased percentage of B and CD4+CD8+ DP T cells) did not occur until after the 28-day administration, which in turn seems indicative of some immune dysregulation due to the prolonged contact with nanogold.

Opis:
Determining the reference values for B and T lymphocytes and their subpopulations in rabbits used in Poland for economic and laboratory purposes is not very common. The goal of this study was to evaluate values of chosen B and T cells and their subpopulations in Polish mixed-breed rabbits, taking season of the year and sex of the animals into the consideration. The study was performed with the use of flow cytometry using monoclonal antibodies (mouse anti-rabbit). The study revealed that, both in Polish mixed-breed rabbits and in Polish mixed-breed rabbits with an addition of blood of meat breeds, the season of the year and sex of the animals affectthe percentage of B- and T-cells and their subpopulations in peripheral blood. As far as season of the year is concerned, it was shown that it mainly affects CD5+ T cells in mixed-breed rabbits and CD25+ T cells in mixed-breed rabbits with an addition of blood of meat breed, while taking sex of the animals into account, more changes were recorded in females in both groups of rabbits. Due to the fact that this is the first paper on those values in Polish rabbits conducted on a large amount of animals, the values may be treated as reference.
Ocena wartości referencyjnych dla limfocytów B i T oraz ich subpopulacji u królików wykorzystywanych w Polsce w celach ekonomicznych i laboratoryjnych jest rzadkością. Celem pracy jest ocena parametrów wybranych limfocytów B i T oraz ich subpopulacji u polskich królików mieszańców z uwzględnieniem pory roku i płci tych zwierząt. Badania wykonano z wykorzystaniem cytometrii przepływowej i przeciwciał monoklonalnych (mouse anti-rabbit). Badania wykazały, że zarówno u królików mieszańców polskich, jak i mieszańców polskich z dolewem krwi ras mięsnych, pora roku i płeć wpływa na kształtowanie badanych parametrów, tj. procentu limfocytów B i T oraz ich subpopulacji. Co się tyczy pory roku wykazano, że zmiany dotyczą głównie limfocytów T CD5+ w przypadku królików mieszańców polskich i limfocytów T CD25+ w przypadku królików mieszańców polskich z dolewem krwi ras mięsnych. Tymczasem w przypadku płci zwierząt, więcej zmian w obu grupach zwierząt wykazano u samic. Ze względu na to, że jest to pierwsza praca z tego zakresu i badania dotyczą dużej grupy zwierząt, winny one być traktowane jako wartości referencyjne.

Bovine leukemia virus (BLV) is a deltaretrovirus which infects and induces proliferation of B-lymphocytes in the peripheral blood circulation and in lymphoid organs primarily of cattle, leading to leukemia/lymphoma. This study was carried out to investigate the presence of BLV in cattle, sheep and camels from the Chaharmahal va Bakhtiary and Isfahan provinces in Iran. A total of 874 blood samples collected from cattle, sheep and camels were used in this study to detect BLV using a nested- PCR. The results from this study indicated that 17.2% (n=874) of all blood samples collected were positive for BLV. The percentages of blood samples positive for BLV from cattle, sheep and camels were 22.1 (n=657), 5.3 (n=95) and 0 (n=122) respectively. The results from this study showed that BLV infected cattle and sheep. Camels seemed to be resistant to BLV infection. This study contributes to the nationwide effort to obtain baseline information on the prevalence of BLV, which will assist in planning the control strategy for the disease in Iran.

The influence of caprine arthritis-encephalitis (CAE) virus infection on the population of peripheral blood leukocytes in goats was evaluated. For this purpose two groups of adult dairy female goats were formed. The experimental group consisted of 17 goats, which had been naturally infected for many years. The control group comprised 29 non-infected goats, which originated from CAE-free herd. All goats were clinically healthy. Whole blood was collected and tested in hematological analyzer and light microscope to assess the total number of leukocytes and the percentage of four leukocyte populations - neutrophils, eosinophils, monocytes and lymphocytes. Then, flow cytometry with monoclonal antibodies against several surface antigens (namely CD14, CD2, B-B2, CD4, CD8h, TCR-N6, WC1-N2 and WC1-N3) was performed to assess the proportion of lymphocyte subpopulations. Statistically significant differences (α ≤ 0.01) were observed only in the subpopulations of T lymphocytes - percentage of all subpopulations were significantly higher in the group of seropositive goats. No statistically significant differences were revealed with respect to the total number of blood leukocytes, the average percentage of blood leukocyte populations and proportions of both T and B lymphocytes.
