Opis:
Lipopolysaccharides (LPS) of Gram-negative bacteria are composed of three regions: О-specific chain (OPS), the core oligosaccharide and lipid A. All three regions of Proteus LPS were studied. The differences in the structure of OPS serve as the basis for the serological classification of Proteus strains. The serological classification scheme of these bacteria currently consists of 76 serogroups. The structural diversity of the core region is characteristic for Proteus sp. and distinguishes this genus from other bacteria. In this paper the results of structural, immunochemical and serological studies of all three regions o f Proteus LPS, as well as a function of LPS as endotoxin and its role in the formation of urinary stones, swarming phenomenon and bacterial growth in biofilm are reported.

Opis:
A large body of evidence suggests that epidermal melanocytes are an integral part of the skin immune system and can be considered immunocompetent cells. Recently, it has been reported that human melanocytes constitutively express Toll-like receptors and may be involved in the induction of several inflammatory cytokines. In the study the secretion of IL-1β, IL-6 and TNF-α by cultured normal melanocytes was investigated after stimulation with lipopolysaccharide. LPS increased the secretion of IL-1β in a dose-dependent manner. IL-1β stimulated release of IL-6 and TNF-α by melanocytes, whereas LPS activated production of TNF-α, but not of IL-6. These observations indicate that LPS can participate in the regulation of cytokine activity in normal human melanocytes and suggest that cytokines released by melanocytes could affect melanocytes themselves or/and other cells of the epidermis.

To extend the knowledge on the fragments of Proteus penneri lipopolysaccharide core regions, which determine the cross-reactions with specific antibodies, serological studies were performed by use of P. penneri 7 core-specific antiserum and Proteus sp. lipopolysaccharides. Different reactivity of the tested antiserum with three groups of antigens suggested differences in their core regions' epitope specificity. Comparing the results of the serological investigations with the previously determined structures of the core regions of the tested P. penneri lipopolysaccharides allowed distinguishing two potential tri- and tetrasaccharide epitopes and a third fragment which could not be determined precisely.

Opis:
Lipopolysaccharide (LPS), a core part of gram-negative bacteria, is crucial for inducing an inflammatory response in living things. In the current study, we used LPS from Salmonella to stimulate chicken macrophages (HD11). Proteomics was used to investigate immune-related proteins and their roles further. Proteomics investigation revealed 31 differential expression proteins (DEPs) after 4 hours of LPS infection. 24 DEPs expressions were up-regulated, while seven were down-regulated. In this investigation, ten DEPs were mainly enriched in S. aureus infection, complement, and coagulation cascades, which were all implicated in the inflammatory response and clearance of foreign pathogens. Notably, complement C3 was shown to be up-regulated in all immune-related pathways, indicating that it is a potential protein in this study. This work contributes to a better understanding and clarification of the processes of Salmonella infection in chickens. It might bring up new possibilities for treating and breeding Salmonella-infected chickens.

Most Proteus lipopolysaccharides (LPSs) contain uronic acids or their amides with different amino acids, which together with other negatively charged components account for the acidic character of such LPS molecules. Previous studies have shown the significance of an amide of galacturonic acid with lysine [D-GalA(L-Lys)] for serological specificity of O-antigens from few P. mirabilis strains. In this work, the immunodominant role of GalALys was indicated for the P. penneri 42 LPS core region. The studies also showed the serological identity of core oligosaccharides from P. penneri 42 (O71), P. mirabilis 51/57 (O28) and R14/S1959 strains.

This study was aimed to evaluate the effects of inulin used as prebiotic on the kidney in lipopolysaccharide (LPS)-induced endotoxemia model. Wistar Albino rats were divided into four groups: Control group, LPS (endotoxemia) group, Inulin + LPS group in which LPS (1.5 mg/kg, E. coli, Serotype 0111: B4) was treated after inulin (500 mg/kg) given by gavage for 21 days and Inulin group. The animals were sacrificed 24 h after the last LPS injection. Kidney samples were taken for biochemical and immunohistochemical analyses. Total antioxidant status (TAS), total oxidant status (TOS), oxidative stress index (OSI), malondialdehyde (MDA) and myeloperoxidase (MPO) values were determined. In addition, kidney sections were stained for inducible nitric oxide synthase (iNOS), tumor necrosis factor (TNF)-α and interleukine-6 (IL-6) expression, and leukocyte infiltration. LPS caused oxidative stress and inflammation. Inulin administration could prevent oxidative stress and lipid peroxidation. Moreover, inulin decreased iNOS, TNF-α and IL-6 expression. However, it did not change the distribution of leukocytes in kidney tissues. These results suggest to promising benefits of inulin as prebiotic in reducing the effects of endotoxemia. Further studies should be conducted to evaluate the capacity of prebiotics in endotoxemia.

Opis:
Human organism, constantly exposed to a large variety of pathogenic microorganisms and their products, such as lipopolysaccharide (LPS), developed innate immunity as a fi rst line of defence. One of the compartments of our organism well equipped with these defence mechanisms is the respiratory system. The cells lining the airways respond to the presence of virulent microorganisms by producing natural antimicrobial peptides, including the only member of the cathelicidins family found to date in humans, peptide LL-37. LL-37 is a small peptide of 37 amino acid residues. The peptide, in addition to its bactericidal effect, plays numerous roles in infl ammatory and tissue remodelling processes. It stimulates angiogenesis, induces proliferation of lung epithelial cells, accelerates wound closure of the airway epithelium, and provokes cytokine release (e.g. IL-8) and cell migration. LL-37 is also able to neutralize LPS, a heteropolymer associated with organic dust, produced by Gram-negative bacteria. LPS (commonly referred to as endotoxin) plays an important role in pathogenesis of many respiratory diseases caused by organic dust, including organic dust toxic syndrome and chronic illnesses such as chronic obstructive pulmonary disease (COPD), asthma or allergic alveolitis (hypersensitivity pneumonitis). LPS is a strong pro-infl ammatory stimulus, inducing in respiratory airways expression of antimicrobial peptides, including LL-37, which is in turn a potent LPS-neutralizing factor. The article discusses the complex interplay between endotoxin and the LPS-neutralizing, pleiotropic peptide LL-37 in pathogenic mechanisms of lung diseases, with regard to closer perspectives of using LL-37 and its derivatives as therapeutic agents.

Opis:
Commensal bacteria and their genes associated with host are known as microbiome. In recent years, microbial influence on host endocrine system has been under detailed investigation. The role of microbiome in the pathogenesis of insulin resistance and obesity, the function of hypothalamic-pituitary-adrenal axis and secretion of hormones regulating appetite is well described in world literature. In this article we discuss poorly reviewed issues: the microbiome role in modulation of non-peptide (sex and thyroid) and peptide (growth hormone and parathyroid hormone) functions. Understanding complex bidirectional relations between host endocrine system and bacteria is of fundamental importance to understanding microbial impact on host reproduction, risk of endocrine-related cancers, pathogenesis of non-thyroidal illness syndrome, growth failure in children and hormonal changes during chronic kidney disease. This article also highlights effects of dietary compounds on microbiome composition and bacterial enzymes activity, and thus host hormonal status.

Dioscin is a natural steroidal saponin mainly exacted from Dioscoreae Rhizoma, which usually used as traditional medicine in east asia area. Dioscin has various pharmacological actions. However, little is known concerning the role on Lipopolysaccharide (LPS)-induced endotoxemia. The aim of the present study was to investigate the effects and possible mechanisms of dioscin against LPS-induced endotoxemia in mice. The data showed dioscin significantly increased mouse survival when mice were treated with dioscin prior to or after LPS challenge and significantly attenuated TNF-α, IL-1β and IL-6 production in serum. Further studies revealed that dioscin could downregulate LPS-induced nuclear transcription factor - қB (NF-қB) signal transduction pathways. These observations indicated that dioscin modulated early cytokine responses by blocking NF-қB activation, and thus, increased mouse survival in murine endotoxemia induced by LPS.

Membrane fluidity measurements were performed for synthetic DPPC liposomes sonicated in aqueous solution and doped by Plesiomonas shigelloides strain CNCTC 144/92 (serotype O74:H5) lipopolysaccharide (LPS) extracted from the phenol (LPS_{PhOH}) and water (LPS_{H₂O}) phases. Concentrations of LPS in relation to DPPC ranged from 0 to 1.4% (molar ratio). The EPR spin probe method was used to describe physicochemical properties of different regions of the lipid bilayer. Since TEMPO spin probe dissolves both in the hydrophobic region of the membrane and in an aquatic environment it is possible to determine the spectroscopic partition parameter F, indicating the changes that occur in the water-lipid interface. The 16-DOXYL probe distributed in the middle of the lipid bilayer makes it possible to obtain the rotational correlation time τ parameter, which provides information about fluidity changes in the liposome membrane. Here we report that increasing concentrations (mainly in the range of 0.4-0.8%) of investigated LPS_{PhOH} and LPS_{H₂O} significantly influence spectroscopic parameters (F and τ). The surface area of the DPPC liposomes membranes was affected predominantly by LPS_{H₂O} while the lipid bilayer was most influenced by LPS_{PhOH}.

The unique feature of some Proteus O-polysaccharides is occurrence of an amide of galacturonic acid with Nε-[(S/R)-1-Carboxyethyl]-l-lysine, GalA6(2S,8S/R-AlaLys). The results of the serological studies presented here, with reference to known O-antigens structures suggest that GalA6(2S,8S/R-AlaLys) or 2S,8R-AlaLys contribute to cross-reactions of O13 Proteus antisera, and Proteeae LPSs. It was also revealed that the Proteus mirabilis RMS 203 strain can be classified into the O13 serogroup, represented so far by two strains: Proteus mirabilis 26/57 and Proteus vulgaris 8344. The O13 LPS is a serologically important antigen with a fragment common to LPSs of different species in the Proteeae tribe.

Introduction and aim. Violation in the salivary glands will inevitably cause changes in periodontium, and periodontitis can disrupt the functioning of the salivary glands. The purpose of the work is to evaluate changes in NO-synthase and arginase activities, pro- and antioxidant balance in rat salivary glands and soft periodontal tissues during administration of bacterial lipopolysaccharide (LPS) and water avoidance stress (WAS) modeling. Material and methods. The experiment was performed on 24 rats. The animals were divided into 4 groups: control, WAS, animals injected intraperitoneally with 0.4 μg/kg of bacterial LPS of Salmonella typhi, WAS+LPS. Results. Water avoidance stress led to decrease of inducible NO-synthase (iNOS) activity in salivary glands by 1.63 times, but decreased arginase activity by 1.15 times, superoxide production increased by 1.53 times, catalase activity decreased by 1.2 times, and malonic dialdehyde (MDA) increased by 1.19 times compared to the control. Lipopolysaccharide led to increase of constitutive NO-synthases (cNOS) activity in salivary glands by 1.48 times, but decreased arginase activity by 6.15 times, catalase activity increased by 2.6 times and superoxide dismutase activity decreased by 2.74 times, and MDA increased by 6.84 times compared to the control. Water avoidance stress + LPS in salivary glands led to decrease of cNOS and arginase activity by 1.09 and 1.19 times, respectively, superoxide production increased by 1.88 times, catalase and superoxide dismutase activity decreased by 1.06 times and 1.34 times, respectively, and MDA increased by 2.44 times compared to the control. Water avoidance stress led to increase of iNOS activity in periodontium by 1.44 times and arginase activity decreased by 1.37 times, superoxide production increased 1.32 times, catalase activity and superoxide dismutase activities decreased by 1.27 times and by 1.53 times, respectively, and MDA increased by 1.31 times compared to the control. Lipopolysaccharide led to increase of iNOS activity in the periodontium by 3.88 times, arginase activity decreased by 2.69 times, superoxide production increased 1.64 times, catalase activity increased by 4.32 times, and MDA increased by 4.51 times compared to the control. Water avoidance stress + LPS in periodontium led to increase of iNOS and cNOS activities by 1.95 times and 1.53, respectively, arginase activity decreased by 1.39 times, superoxide production increased 1.66 times, catalase activity increased by 1.11 times, and MDA increased by 1.53 times compared to the control. Conclusion. The combination of LPS and WAS leads to changes in NO production and oxidative stress in salivary glands and the periodontium.

Six rhizobium (Rhizobium leguminosarum bv. Trifolii TA1, Sinorhizobium meliloti 1021, Mesorhizobium huakuii IFO 15243T, Ochrobactrum lupini LUP 21T, Bradyrhizobium japonicum USDA110 and B. elkanii USDA 76) and two Escherichia coli strains (E. coli ATCC 25922 and E. coli HB 101) were compared in respect to polymyxin B and EDTA resistance, as well as bacterial outer membrane (OM) permeability to a fluorescent hydrophobic agent (N-phenyl-1-naphthylamine - NPN). TEM (Transmission Electron Microscopy) and a microbial test demonstrated that all the rhizobia were much more resistant to polymyxin B in comparison with E. coli strains. EDTA and polymyxin B enhance permeability of B. japonicum and O. lupini OM. Other rhizobia incorporated NPN independently of the presence of membrane-deteriorating agents; however, the level of fluorescence (measured as NPN absorption) was strain dependent.

Specific adhesion to host tissue cells is an essential virulence factor of most bacterial pathogens. Adherence is often an essential step in bacterial pathogenesis or infection, required for colonizing a new host. To effectively adhere to host surfaces, many bacteria produce multiple adherence factors called adhesins. There are two types of adhesins: fimbrial and nonfimbrial adhesins. Lipopolysacharide and outer membrane proteins belong to non fimbrial adhesins. The role of LPS in adherence of Gram-negative organisms to host cells has been evaluated for several bacterial species. The O-specific chain of bacteria can lead mostly to an increased tendency for this organism to bind to mammalian cells. The attachment of bacteria could be inhibited by purified LPS. A few receptors responsible for recognizing LPS have been identified: CD 14, scavenger receptor, Toll-like receptors, integrins, selectins. LPS receptors transduce signals from the membrane to the cytosol. Interaction between adhesins and their receptor can lead to invasion to host cells. Adhesins are attractive vaccine candidates because they are often essential to infection and are surface-located, making them readily accessible to antibodies.

Intravenous lipopolysaccharide (LPS) leads to acute lung injury (ALI) in rats. The purpose of this study was to examine the anti-inflammatory and antioxidant efficacy of ketamine, propofol, and ketofol in a rat model of ALI. We induced ALI in rats via intravenous injection of LPS (15 mg kg-1). The animals were randomly separated into five groups: control, LPS only, LPS + ketamine (10 mg·kg-1·h-1), LPS + propofol (10 mg·kg-1·h-1), LPS + ketofol (5 mg·kg-1·h-1 ketamine + 5 mg·kg-1·h-1 propofol). LPS resulted in an increase in the release of pro-inflammatory cytokines, mRNA expression related with inflammation, production of nitric oxide, and lipid peroxidation. Ketamine prevented the increase in markers of oxidative stress and inflammation mediators, both in plasma and lung tissue. Propofol decreased the levels of cytokines in plasma and lung tissue, whereas it had no effect on the IL-1-beta level in lung tissue. Ketamine downregulated mediators of lung tissue inflammation and reduced the level of circulating cytokines and protected lung tissue against lipid peroxidation. Ketofol decreased the level of TNF-α and IL-1β in plasma, as well as expression of cyclooxygenase-2 mRNA and the nitrate/nitrite level in lung tissue. The results of this investigation support the hypothesis that ketamine may be effective in preventing ALI.

Rutin, as a common flavonol glycoside, exists abundantly in many plants and exhibits various nutritional and pharmacological function. Current study focused on the dose-effect relationship of rutin on the anti-inflammatory and antioxidant activities in vitro against LPS-stimulated RAW 264.7 cells and free radicals or ferric-ion. This study evaluated the anti-inflammatory activity by observation cell morphological and determination of the content of NO, TNF-α, IL-1β and IL-6, and the phagocytic activity in LPS induced RAW 264.7 cells; and revealed the antioxidant activities via assay of DPPH radical scavenging capacity, ABTS radical scavenging capacity, and reducing power. The study indicated that 5, 10, 20, 50 and 100 μM of rutin all exhibited a better anti-inflammatory activity, in especial 50 and 100 μM. Meanwhile, it exerted a stronger antioxidant activities than BHT, and equivalent to VC at the same concentration. Combined, the results suggested that rutin will be a potential and important adjuvant in treatment of inflammatory diseases and oxidative stress.

Chronic inflammation is characterized by excessive production of cytokines and eicosanoids and is associated with unsufficient resolution. Supplementation with n-3 fatty acids may result in a lower incidence of many inflammatory diseases. The aim of this study was to determine the effect of α – linolenic acid (ALA) on the fatty acids profile of cell membranes and on the pro-inflammatory proteins cyclooxygenase – 2 (COX-2), prostaglandin E2 synthase (cPGES) and prostaglandin F2α receptor (FP) expression in murine RAW 264.7 macrophages, activated with lipopolysaccharide (LPS). It has been shown that COX-2, cPGES as well as FP receptor expression was highest in cells activated by LPS. In macrophages supplemented with ALA and activated with LPS a pro-inflammatory protein levels were significantly reduced, suggesting anti-inflammatory activity of α-linolenic acid. There were also statistically significant changes in the fatty acid profile after incubation of the RAW 264.7 cells for 48 hours with ALA. A deficiency or excess of specific fatty acids affect the cellular membrane fluidity, can also cause changes in cell morphology. Therefore it is appropriate to carry out further research on the ALA properties.

Objectives The aim of the study has been to investigate the effect of the Standard Reference Material of fine particulate matter (SRM 2786) on cytotoxicity and apoptosis in human lung bronchial epithelial cells (16HBE cells). Whether the lipopolysaccharide (LPS)-induced inflammation could further accelerate cell apoptosis induced by SRM 2786 stimulation has also been determined. Material and Methods 16HBE cells were exposed to various doses of SRM 2786 with or without LPS. The following parameters: cytotoxicity, apoptotic rate, Bax/Bcl-2 expression, nitric oxide (NO) production, and reactive oxygen species (ROS) generation were measured. Results The results have shown that SRM 2786 induces cell damage and apoptosis of 16HBE cells as demonstrated by significant decrease in expression of Bcl-2 and increase in expression of Bax. When compared with the control cells, the apoptotic rate of cells treated by 500 μg/ml of SRM 2786 increased from 2.43±0.21% to 43.96±2.95% (p < 0.01). Further, there was an elevated production of NO and ROS post SRM 2786 treatment. The level of NO in cells treated with 500 μg/ml of SRM 2786 was 18.33±1.02 μmol/l whereas that of control cells was 1.58±0.31 μmol/l (p < 0.01). When compared with the control group, the level of intracellular ROS increased by 24% after treatment with 500 μg/ml of SRM 2786 (p < 0.05). In addition, LPS pre-treatment may accelerate cell apoptosis by increasing generation of NO and ROS followed by SRM 2786 stimulation. When compared to cells treated with 125 μg/ml of SRM 2786 alone, the levels of NO and ROS in cells pretreated with LPS increased by 28% and 11.6%, respectively (p < 0.05), and the apoptotic rate increased from 34.62±4.44% to 54.11±3.34% (p < 0.01). Conclusions These findings have suggested that in vitro exposure to SRM 2786 could induce 16HBE cells apoptosis probably by means of the mechanism involving the generation of free radicals, while the degree of apoptosis would be further aggravated under inflammation condition. Sialic acids are important constituents of animal tissue glycoconjugates and are also present in the antigens of some bacterial strains. Capsular polysaccharides with sialic acid (NeuAc) have been extensively studied with regard to sensitivity to the bactericidal action of serum, whereas little is known in this regard about lipopolysaccharides (LPS) which contain NeuAc. Strains of Salmonella 048, able to infect animals and containing the same structures of LPS with NeuAc, were examined for their susceptibility to the bactericidal action of normal bovine serum (NBS). The strains showed varied sensitivity to the bactericidal action of NBS, which indicates that the expression of LPS containing NeuAc residues is not critical for the strains' resistance to the serum's activity. In this study the mechanisms of complement activation responsible for killing serum-sensitive Salmonella 048 rods by NBS were also established. Three such mechanisms were distinguished: activation of the classi- cal/lectin pathways, important (decisive) in the bactericidal mechanism of complement activation, parallel activation of the classical/lectin and alternative pathways, and independent activation of the classical and lectin or the alternative pathway.

Microorganisms synthesize several monomeric chemical structures that are not found elsewhere in nature, e.g. muramic acid (an amino sugar) and D-amino acids (D-alanine and D-glutamic acid) are ubiquitous in bacterial peptidoglycan (PG). Characteristic sugars (e.g. heptoses) and 3-hydroxylated fatty acids are found in the endotoxin (lipopolysaccharide, LPS) of gram-negative bacteria [1]. The best way to protect against environment contamination is microbial control. Methods in current use for monitoring microorganisms mainly include culture and direct microscopy. However, several factors, including samples collection, growth conditions, incubation temperature and interaction between different organisms all affected the culture results. Additionally, culture based methods can detect only viable organisms and they are also time consuming, sometimes taking days or weeks. However, since both living and dead microorganisms express irritating and toxic structures, they should all be taken into consideration. Muramic acid has been suggested for use as a chemical marker in gas chromatography-mass spectrometry (GC-MS) determination of bacterial peptidoglycan [2]. While 3-hydroxylated fatty acids are the best proposition for use as a chemical markers in gas chromatography-mass spectrometry determination of bacterial lipopolysaccharide (endotoxin) of gram-negative bacteria in both clinical and environmental samples [38]. Two derivatives have been applied, including the trimethylsilyl (TMS) and pentafluorobenzoyl (PFBO) derivatives [80]. Both derivatives (TMS and PFBO) have been proven suitable for use with GC-ion-trap tandem MS [3]. The aim of our proposition is trial of application of gas chromatography-mass spectrometry (GC-MS) method as an alternative or complement to culturing, microscopy and other assays for detection, characterization and monitoring of microbial contamination of environment (e.g. water, air, air-conditioning systems), contamination of biochemical and food production chain processes, packaging for foodstuffs etc. by analysis of bacterial 3-hydroxylated fatty acids as a biochemical markers. A method is described for the quantitation of methyl esters of 3-hydroxyacids, markers of bacterial lipopolysaccharide (endotoxin), as trimethylsilyl or pentafluorobenzoyl derivatives using GC/MS method. The described methods are quick and simple, can be applied for monitoring microbial contamination directly, without prior culturing, in complex environmental samples. This method can be also applied for testing processes of cleaning and disinfections on packaging materials or on both packaging materials/foodstuffs in order to decrease their microbial load and thus to ensure better shelf-life.
