HSPA2 is a human counterpart of the testis-specific rodent Hst70/Hsp70.2 gene. In contrast to the latter, the expression of the human HSPA2 gene is not limited to the testis, and recent data show that human tumor cells can express this gene at significant levels. The characteristics of HSPA2 expression suggests that it can influence the phenotype and survival of cancer cells similarly as overexpression of major members of the HSP70 gene family. Until now, neither the structure of the transcription unit of the human HSPA2 gene has been established nor a functional analysis of its promoter performed. In this study we established that the human HSPA2 gene, in contrast to its rodent counterparts, is intronless and has a single transcription start site. We also show that the same type of HSPA2 transcripts are synthesized in the testes and in cancer cell lines. In order to perform a functional study of the HSPA2 promoter, we used a transient transfection assay and found that the 392 bp fragment upstream of the ATG codon was a minimal region required for efficient transcription, while a 150 bp deletion from the 5' end of this region dramatically reduced the promoter activity. Delineation of the minimal promoter is a basic step toward identifiying the cis and trans elements involved in the regulation of the HSPA2 gene expression in cancer cells.

The bovine kappa-casein (CASK) gene is known as a potential quantitative trait locus in dairy cattle breeding. However, the molecular basis of the effect of the CASK allele B on different milk properties remains unclear. In this report, a 214 bp fragment of the 5' untranslated region of the CASK gene containing 5 potential consensus sequenses for different transcription factors was PCR-amplified to find RFLPs. A Dde I RFLP was identified. In a population of 112 Bos taurus (86 cows and 26 bulls of Polish Black and White crossbred Holstein-Friesian) and 7 Bison bonasus individuals, 7 had no recognition sites for Dde I, 23 were heterozygous and 89 were cut completely into two fragments.

In the paper the detection of the SSCP polymorphism within the 5’ fragment of bovine beta-lactoglobulin (LGB) gene is described. The 5’ fragment of LGB gene (209 bp) was PCR-amplified and then subjected to electrophoresis allowing the detection of SSCP polymorphism. Among 124 animals (50 cows and 74 bulls) six SSCP patterns were identified and named Rl, R2, R3, R4, R5 and R6, which occured with the frequency of 0.32, 0.51, 0.09, 0.06, 0.01 and 0.01, respectively. The PCR-SSCP method is simple, fast, and relatively inexpensive. The SSCP polymorphism reported in the paper may be useful in looking for the associations between different SSCP patterns and LGB gene expression and milk properties.

Opis:
The breast cancer suppressor proteins BRCA1 and BRCA2 interact with RAD51, a protein essential for maintaining genomic stability by playing a central role in homology-dependent recombinational repair of the DNA double-strand breaks. Therefore, genetic variability in the RAD51 gene may contribute to the appearance and/or progression of breast cancer. A single nucleotide polymorphism in the 5'- untranslated region of RAD51 (a G to C substitution at position 135, the G/C polymorphism) is reported to modulate breast cancer risk. We investigated the distribution of genotypes and frequency of alleles of the G/C polymorphism in breast cancer. Tumor tissues were obtained from postmenopausal women with node-negative and node-positive breast carcinoma with uniform tumor size. Blood samples from age matched healthy women served as control. The G/C polymorphism was determined by PCR-based MvaI restriction fragment length polymorphism. The distribution of the genotypes of the G/C polymorphism did not differ significantly (P >0.05) from those predicted by the Hardy-Weinberg distribution. There were no differences in the genotype distribution and allele frequencies between node-positive and node-negative patients. There were no significant differences between distributions of the genotypes in subgroups assigned to histological grades according to Scarf-Bloom-Richardson criteria and the distribution predicted by Hardy-Weinberg equilibrium (P >0.05). Our study implies that the G/C polymorphism of the RAD51 gene may not be directly involved in the development and/or progression of breast cancer and so it may not be useful as an independent marker in this disease.

The p53 tumour suppressor protein is one of the major factors responsible for cell cycle regulation and protection against cancer development. This is why it is often referred to as "the guardian of the genome". On the other hand, mutations in the p53 gene are connected with more than 50% of tumours of various types. The thirty-six years of extensive research on the p53 gene and its protein products have shown how sophisticated the p53-based cell system control is. An additional level of complexity of the p53 research is connected with at least twelve p53 isoforms which have been identified in the cell. Importantly, disturbance of the p53 isoforms' expression seems to play a key role in tumorigenesis, cell differentiation and cell response to pathogenic bacteria, and RNA and DNA viruses. Expression of various p53 isoforms results from the usage of different transcription promoters, alternative splicing events and translation initiation from alternative AUG codons. The importance of the 5'-terminal regions of different p53 mRNA transcripts in the multi-level regulation of the p53 gene has recently been documented. In this review we focus on the structural features of these regions and their specific role in the p53 translation initiation process.

Opis:
ABCC10 (MRP7) plays a role in cellular detoxification and resistance to anticancer drugs. Since ABCC10 gene transcription in human prostate cancer CWR22Rv1 cells was found dependent on E2F binding sequence motif, ABCC10 expression in G1 and S phases of the cell cycle of CWR22Rv1 cells, was analyzed. The cells were synchronized in G1 phase by double thymidine block and in S phase by thymidine/mimosine double block. ABCC10 mRNA level was found to be similar in S phase-synchronized and asynchronous cell populations. In G1 phase it decreased by 2.4- to 3-fold. It is thus inferred, that ABCC10 expression in CWR22Rv1 cells is not S phase-specific but is primarily associated with cell proliferation.

The objective of this study was to describe and compare the genetic structure (TNF-α-position 824) of dairy cattle herds infected and not infected with the bovine leukemia virus (BLV). The results of the present study indicate that BLV-positive herds were characterized by similar genetic structure (TNF-α-824A/G). The genetic equilibrium in these herds was preserved, but a tendency to increased frequency of G/G homozygotes was found. The genetic structure of the healthy herd differed considerably from that of leukemic herds.

Opis:
A total of 684 Ixodes ricinus ticks (321 nymphs, 184 males, and 179 females) were collected by fl agging lower vegetation in 6 forest districts located on the territory of Lublin province (eastern Poland). Ticks were examined by polymerase chain reaction (PCR) method for the presence of Anaplasma phagocytophilum DNA with two pairs of primers: EHR521/EHR747 for detecting 16S rRNA gene, and LA6/LA1 for detecting ankA gene. To study the relationship between infection in ticks and people occupationally exposed to tick bite, blood serum samples of 261 forestry workers employed in the same forest districts were examined by immunofl uorescence method for the presence of specifi c antibodies against A. phagocytophilum. A total of 70 ticks out of 684 examined (10.2%) showed the presence of A. phagocytophilum 16S rRNA gene. The prevalence of infection was signifi cantly dependent on tick’s stage (χ2=49.2, p<0.00001) and geographical locality (χ2=34.4, p<0.00001). The percentage of I. ricinus females infected with A. phagocytophilum (24.6%) was signifi cantly greater compared to males (6.5%) and nymphs (4.4%) (p<0.00001). Only 19 ticks out of 684 examined (2.8%) showed the presence of A. phagocytophilum ankA gene, signifi cantly less compared to 16S rRNA gene (p<0.00001). The prevalence of infection demonstrated by the presence of ankA gene was also signifi cantly dependent on tick’s stage (χ2=23.6, p<0.00001) but not on locality (χ2=9.8, p=0.082). A signifi cant correlation was found between the presence of A. phagocytophilum 16S rRNA gene in I. ricinus female ticks from the particular forest districts and the serologic response to A. phagocytophilum of forestry workers employed in the same districts (p<0.05). No signifi cant correlation was found between the presence of A. phagocytophilum ankA gene in I. ricinus ticks and serologic response of exposed workers. In conclusion, detection of A. phagocytophilum infection in ticks by PCR with the use of EHR521/EHR747 primers detecting 16S rRNA gene is signifi cantly more sensitive compared to LA6/LA1 primers detecting ankA gene.

Introduction: In the present thesis the association between the variants of C421A polymorphism of ABCG2 gene and the risk of gastric cancer development were searched. The occurrence of polymorphic variant 421 C > A leads to the weakening of the activity of BCRP protein which is encoded by the ABCG2 gene. Due to the presence of BCRP protein in the gastrointestinal tract and its protective role the reduction of the amount of BCRP protein which is the result of the tested polymorphism may lead to the increased predisposition to cancer development. The main aim of this study was to compare the frequency of C421A polymorphism genotypes between the examined and the control groups. Material and methods: The material for the research consisted of DNA isolated from 19 scraps of the tumor tissue taken from patients with gastric cancer and 44 samples of peripheral blood collected from healthy individuals. DNA was extracted from the tested materials by microcolumn-method. Then the obtained samples of DNA were subjected to polymerase chain reaction (PCR) to amplify the ABCG2 gene fragment. To determine C421A polymorphism genotypes ABCG2 gene fragments amplified by PCR the were analyzed by RFLP method using an digestive enzyme MseI. Results: All the examined patients irrespectively of gender, age and clinical stage showed the presence of genotype C/C, which is homozygous wild for C421A polymorphism. All 44 persons from the control group also showed the presence of genotype C/C. Conclusions: No differences in the occurrence of C421A genotypes between the study group and the control group were found. In both the groups 100% of the results for the C/C genotype were obtained. Due to the presence of only one genotype (C/C) for the C421A polymorphism in all the patients no relationship between the occurrence of C421A genotypes and tumor stage, age and gender of the examined patients was evaluated. Although no correlation between the C421A polymorphism and the susceptibility to the development of gastric cancer was found the exclusion of the participation of BCRP protein in the pathogenesis of this disease is possible only after the increase of the number of analyses on larger groups of respondents.
Wstęp: W niniejszej pracy poszukiwano związku między występowaniem wariantów dla polimorfizmu C421A w obrębie genu ABCG2, a zwiększonym ryzykiem rozwoju raka żołądka. Występowanie wariantu polimorficznego 421 C>A prowadzi do zmniejszonej aktywności białka BCRP, kodowanego przez gen ABCG2. Ze względu na obecność białka BCRP w obrębie przewodu pokarmowego i związaną z tym rolę ochronną, zmniejszenie ilości białka BCRP będące efektem badanego polimorfizmu może zwiększać predyspozycję do rozwoju raka. Głównym celem pracy było porównanie częstości występowania genotypów dla polimorfizmu C421A pomiędzy grupą badaną a kontrolną. Materiał i metody: Materiał do badań stanowiło DNA wyizolowane z 19 mrożonych skrawków tkankowych pobranych od pacjentów z rakiem żołądka i z 44 prób krwi obwodowej pobranych od osób zdrowych. Izolację DNA z badanych materiałów biologicznych przeprowadzono wykorzystując metodę kolumienkową. Następnie, uzyskane próby DNA poddawano reakcji łańcuchowej polimerazy, by powielić badany fragmentu genu ABCG2. Celem określenia genotypów dla polimorfizmu C421A na mnożony w reakcji PCR fragment genu ABCG2 poddawano analizie metodą RFLP z użyciem enzymu MseI. Wyniki: U wszystkich badanych, bez względu na płeć, wiek oraz stopień zaawansowania klinicznego raka żołądka, wykazano obecność genotypu C/C, czyli homozygoty dzikiej dla polimorfizmu C421A. Wśród 44 osób z grupy kontrolnej również u wszystkich stwierdzono genotyp C/C. Wnioski: Nie wykazano różnic w częstości występowania genotypów dla polimorfizmu C421A między grupą badaną i kontrolną. W obydwu grupach zaobserwowano tylko jeden z możliwych genotypów dla badanego polimorfizmu - genotyp C/C. Z uwagi na stwierdzenie u wszystkich badanych osób z obu grup tylko genotypu C/C dla polimorfizmu C421A nie oceniano zależności między występowaniem genotypów C421A a stopniem zaawansowania nowotworu, wiekiem i płcią badanych osób. Mimo, że nie wykazano korelacji między polimorfizmem C421A a podatnością na rozwój raka żołądka, jednoznaczne wykluczenie udziału białka BCRP w patogenezie tej jednostki chorobowej jest możliwe dopiero po przeprowadzeniu większej ilości analiz na większych grupach badanych.
