Opis:
The content of organic matter affects the quality and productivity of agricultural soils. Due to the shortage of manure as the basic source of organic matter for agricultural soils, al- ternative and commonly available exogenous sources of organic matter, such as sewage sludge and bottom sediments, are taken into account. The aim of the study was to evaluate the effect of the addition of various types of manure, municipal sewage sludge and bottom sediments on the enzymatic activity of soils (dehydrogenases, acidic and alkaline phosphatases). Manure and sludge were applied to the soil in two doses, corresponding to 20 and 40 tons of dry matter of material per ha. The control was soil fertilized with ammonium nitrate in the amount corresponding to 170 kg N per ha. The experiment included 2 different variants of the same fertilization combinations: without plants and with a test plant (Sinapis alba L.). The greenhouse experiment showed the stimulating effect of one of the municipal sewage sludge and poultry manure. On the other hand, the introduction of bottom sediments to the soil limited its enzymatic activity.

In these studies we attempted to evaluate the lipolytic, proteolytic and cellulolytic activity of bacterial strains isolated from water and the bottom sediments of Turawa Lake. The following bacterial genera prevailed among the isolated strains: Bacillus, Pseudomonas, Enterobacter, Cellulomonas and Cytophaga. The lipolytic activity was determined using a titrimetric method, the proteolytic activity - using a modified Anson method, and the cellulolytic activity - on the basis of mass decrement of a cellulose disk after 14 days of bacterial culture. The cultures were maintained at 28°C, pH 7.0 with the following substrates: olive oil, albumin and cellulose disk. Among the analysed microorganisms, Bacillus and Pseudomonas strains showed the highest lipolytic and proteolytic activity. In the cellulolytic assay Cytophaga bacteria showed about twofold higher activity than that of Celulomonas.

Opis:
A pot experiment was performed to determine the effect of soil contamination with nickel, applied at a dose of 100, 200, 300 and 400 mg kg-1, on the activity of dehydrogenases, urease, acid phosphatase and alkaline phosphatase. The impact of nickel on the enzymatic activity of soil was studied on samples of heavy loamy sand and light silty loam. The experiment was conducted in two series: in the first one soil was cropped to yellow lupine, and in the second one it was left uncropped. Soil samples were analyzed on day 14, 28, 42 and 56. It was found that soil contamination with nickel reduced the activity of all the enzymes. This negative influence was most noticeable in the case of dehydrogenase. The activity of urease and alkaline phosphatase was higher in light silty loam, while the activity of dehydrogenases and acid phosphatase was higher in heavy loamy sand. The activity of dehydrogenases and urease was higher in soil cropped to yellow lupine, whereas the activity of acid phosphatase and alkaline phosphatase was higher in uncropped soil. Yellow lupine was sensitive to excessive amounts of nickel in the soil, and partly alleviated the adverse impact of this heavy metal on urease activity, but did not reduce its inhibitory effect on the other enzymes.
Próbki glebowe analizowano: w 14., 28., 42. i 56. dniu. Stwierdzono, że zanieczyszczenie gleby niklem spowodowało zmniejszenie aktywności wszystkich badanych enzymów. Najbardziej niekorzystnie nikiel wpływał na aktywność dehydrogenaz. Wyższą aktywność ureazy i fosfatazy alkalicznej stwierdzono w glinie lekkiej pylastej niż w piasku gliniastym mocnym, natomiast dehydrogenaz i fosfatazy kwaśnej - w piasku gliniastym mocnym. Wyższą aktywność dehydrogenaz i ureazy odnotowano w glebie obsianej łubinem żółtym, natomiast fosfatazy kwaśnej i fosfatazy alkalicznej - w glebie nie obsianej.

This paper assesses the dissipation of napropamide and its impact on the activity of dehydrogenases, alkaline phosphatase, acid phosphatase, and urease in sandy clay loam. The experiment was carried out on soil samples with organic carbon content of 12.08 g·kg-1, total nitrogen content of 0.97 g·kg-1, and pH 5.24 with the following variable factors: (a) dose of Devrinol 450 SC formation (containing 450 g of naprop-amide in dm3): 0 (control), 0.5, 1, 2, 4, 8, and 16-fold hold of field dose; (b) day of experiment: 1, 7, 14, 28, 56, and 112. The half-life of napropamide ranged from 33.50 to 71.42 days. The use of napropamide at the dose recommended by the manufacturer and at the dose reduced by half appeared to exhibit low toxicity in relation to enzymes determined. In contrast, the application of elevated napropamide doses decreased the values of biochemical parameters of the soil in most cases. The Pearson’s correlation coefficients showed statistically significant negative correlation between the content of napropamide residues and the enzymatic activity of the soil.

In this in vitro study, the influence of tin ions at concentrations of 1–1,000 ppm on the development and enzymatic activity of four entomopathogenic fungi (Beauveria bassiana, B. brongniartii, Isaria fumosorosea, and Metarhizium robertsii), that are commonly used in biological plant protection, are examined. Each of the fungal species tested reacted differently to contact with the Sn2+ ions at the tested concentrations. Exposure to Sn2+ ions affected the rate of development, morphology, and enzymatic activity of fungi. Of the four fungal species studied, M. robertsii was the most resistant and showed complete growth inhibition at the highest Sn2+ concentration tested (1,000 ppm). For the other entomopathogenic fungi, the fungicidal effect of Sn2+ ions was noted at the concentration of 750 ppm. Exposure to Sn2+ ions (up to 500 ppm) resulted in enhanced biochemical activity; and all entomopathogens that were tested showed increased production of N-acetyl-β-glucosaminidase (NAG) as well as several proteases. Moreover, B. brongniartii and M. roberstii showed increased lipases synthesis. These changes may increase the pathogenicity of the fungi, thereby making them more effective in limiting the population of pest insects. The exposure of the entomopathogenic fungi to a medium containing Sn2+ ions, at concentrations that were appropriate for each species, induced hyperproduction of hydrolases, which might be involved in aiding the survival of entomopathogenic fungi in the presence of heavy metals. This study shows that the fungistatic effect of Sn2+ on entomopathogenic fungi did not restrict their pathogenicity, as evidenced by the stimulation of the production of enzymes that are involved in the infection of insects.

Initial experiment was made in order to test wood-decomposing fungus Phlebiopsis gigantea for enzymatic activity. Laccase, peroxidase, cellulase, phosphatase, dehydrogenase were marked using different methods for P. gigantea isolates growing on Scots pine wood.
Najwyższą aktywnością dehydrogenazy, obok izolatu H. annosum, odznaczał się izolat P.g.5, a najmniejszą zaś izolat P.g.6 pozyskany z owocnika.

A strain of Haloarcula hispanica isolated from Tuzkoy salt mine, Turkey exhibited extracellular lipolytic activity. Important parameters such as carbon sources and salt concentration for lipase production were investigated. Optimal conditions for the enzyme production from Haloarcula hispanica 2TK2 were determined. It was observed that the lipolytic activity of Haloarcula hispanica was stimulated by some of the carbon sources. The high lipase acitivity values were obtained in the presence of 2% (v/v) walnut oil (6.16 U/ml), 1% (v/v) fish oil (5.07 U/ml), 1% (v/v) olive oil (4.52 U/ml) and 1% (w/v) stearic acid (4.88 U/ml) at 4M NaCl concentration. Lipase was partially purified by ammonium sulfate precipitation and ultrafiltration. Optimal temperature and pH values were determined as 45oC and 8.0, respectively. Lipase activity decreased with the increasing salt concentration, but 85% activity of the enzyme was maintained at 5M NaCl concentration. The enzyme preserved 41% of its relative activity at 90oC. The partially purified lipase maintained its activity in the presence of surfactants such as Triton X-100 and SDS. Therefore, the lipase which is an extremozyme may have potential applications especially in detergent industry.

The enzymatic activity of soil was studied in a three-year pot experiment, where different amounts of zinc were introduced: 0, 200, 400, and 600 mg of Zn kg-1 of soil, as well as organic materials (control object, bovine manure, laying hens’ litter, spent mushroom substrate), used separately with the soil application dose of 2 g Corg kg-1 soil. Zinc and organic materials were used once, only in the first year of research, before sowing the test plant. In each year of the experiment, the test plant was orchard grass (Dactylis glomerata L.), harvested after 4 cuts. The activity of urease, dehydrogenases, as well as acid and alkaline phosphatase were determined in the soil collected after the last grass cutting, in each year of the study. Application of zinc to soil, regardless of its dose, resulted in a decrease in the activity of urease, dehydrogenase and alkaline phosphatase, although the significance of differences in relation to the control object was demonstrated only at higher doses, such as 400 and 600 mg Zn kg-1 soil. The study showed no negative effect of zinc on acid phosphatase activity. Organic fertilizers generally resulted in an increase in the enzymatic activity of the analyzed soil, and simultaneously limited the negative effects of higher zinc doses (400 and 600 mg Zn kg-1 of soil) on urease and dehydrogenase activity. During the consecutive years, urease and alkaline phosphatase activity decreased, while acid phosphatase activity increased. Dehydrogenase activity did not change significantly in the subsequent years of the study.

In the soil environment, there are many enzymeswhose origin is related to microorganisms. Enzymes participatein the synthesis of proteins and nucleic acids, they are also elementsof the carbon, nitrogen and phosphorus cycles. They alsoplay a role in less known cycles, such as the sulphur cycle orcellulose decomposition. Determination of soil enzymes activityis very popular in microbiology, biochemistry and agriculturalsciences. Their activity may be a measure of soil fertility. Thepresent report describes the importance of selected soil enzymesand the methods of their determination. However, it is importantto remember that a single enzyme cannot be an indicator of theenvironment. The enzymatic activity is sensitive to many naturaland anthropogenic factors affecting soil microorganisms. Onlycomprehensive researchers on the activity of many, different enzymescan provide reliable information on the state of the soilenvironment.

Soil microorganisms play a key role in many biochemical processes essential for the environment and for the ecological and production functions of soils, hence they are very important quality indicators. The study aimed at evaluating the effect of 1 and 2% additions of wheat and miscanthus straw biochars on soil enzymatic activity (dehydrogenases, urease, phosphatases) and the number of microorganisms (bacteria, fungi, actinobacteria). Pot experiments were carried out on soil with a loamy sand texture. The geometric mean of enzyme activities, biological index of fertility, and the integrated total enzyme activity index were used to explore the relationships between soil enzyme activities and the microbiological and chemical properties of soil. The addition of 1% miscanthus straw biochar had the most beneficial effect on the number of bacteria and fungi (increase by 380 and 26%, respectively), and 1% wheat straw biochar on the number of actinomycetes (increase by 273%). The correlation analysis between the number of actinomycetes and the chemical parameters of the soil showed the significant effect of the content of Ntotal (r = 0.76) and Ctotal (r = 0.85). The values of biological index of fertility, total enzyme activity index and geometric mean of enzyme activities indexes showed that the best quality of soil was produced by a 2% addition of wheat straw biochar.

Opis:
The variety of technological conditions and raw materials from which biochar is produced is the reason why its soil application may have different effects on soil properties and plant growth. The aim of this study was to evaluate the effect of the addition of wheat straw and Miscanthus giganteus straw (5 t DM ha-1) and biochar obtained from this materials in doses of 2.25 and 5 t DM ha-1 on soil enzymatic activity, soil ecotoxicity, and plant yield (perennial grass mixture with red clover). The research was carried out under field conditions on soil with the granulometric composition of loamy sand. No significant effect of biochar amendment on soil enzymatic activity was observed. The biochar-amended soil was toxic to Vibrio fischeri and exhibited low toxicity to Heterocypris incongruens. Application of wheat straw biochar and M. giganteus straw biochar in a dose of 5 t DM ha-1 contributed to an increase in plant biomass production by 2 and 14%, respectively, compared to the soil with mineral fertilisation. Biochars had a more adverse effect on soil enzymatic activity and soil ecotoxicity to H. incongruens and V. fischeri than non-converted wheat straw and M. giganteus straw, but significantly increased the grass crop yield.

Opis:
The aim of the study was to evaluate microbial functional diversity based on community level physiological profiling and b-glucosidase activity changes in soil degraded by sulphur mining and subjected to reclamation with various waste. The experiment was set up in the area of the former ‘Jeziórko’ Sulphur Mine (Poland), on a soilless substrate with a particle size distribution of slightly loamy sand. The experimental variants included the application of post-flotation lime, sewage sludge and mineral wool. The analyses of soil samples included the assessment of the following microbiological indices: b-glucosidase activity and functional diversity average well color development and richness). The results indicate that sewage sludge did not exert a significant impact on the functional diversity of microorganisms present in the reclaimed soil. In turn, the application of other types of waste contributed to a significant increase in the parameters of total metabolic activity and functional diversity of the reclaimed soil. However, the temporal analysis of the metabolic profile of soil microorganisms demonstrated that a single application of waste did not yield a durable, stable metabolic profile in the reclaimed soil. Still, there was an increase in β-glucosidase activity, especially in objects treated with sewage sludge.

Opis:
The natural mineral resources (hard coal, sands, dolomites, lead and zinc ores) found in the Silesia and the excavation of them led to significant transformation or even degradation of the environment. The landscape of Upper Silesia was dominated by heaps created as a result of the accumulation of post-mining coal waste. These postindustrial sites are characterised by difficult conditions for the development of plant communities. Nevertheless, the heaps are spontaneously overgrowing and over time, a separate ecosystem can be observed (for heaps). The article analyzes the enzymatic activity of the substrate in relation to the selected dominant grass (Monocots) and herbaceous (Dicots) plant species. The aim of this study was to compare the activity of particular enzymes in soil substratum of the vegetation patches dominated by grass and herbaceous plants.

Opis:
Introduction: The secretion of hydrolytic enzymes is a factor facilitating pathogenic fungi invasion into the tissues. Purpose: To assess hydrolytic activity and biotypes of Candida strains isolated from samples collected from the surfaces of mobile phones and the hands of their owners. Materials and methods: The study included 175 mobile telephones and hands. The API ZYM test was used to assess enzymatic activity; biotyping was performed according to Williamson’s classification. Results: Among the strains isolated from hand surfaces, the highest activity was shown for C. albicans (acid phosphatase, esterase), C. glabrata (leucine arylamidase, acid phosphatase, esterase), and C. krusei (acid phosphatase). Of the strains isolated from phone surfaces, the highest activity was shown for C. albicans (leucine arylamidase, acid phosphatase), C. glabrata (esterase, leucine arylamidase, esterase lipase), and C. krusei (acid phosphatase). Biotypes G, B and F were dominant for all types of fungi, both for strains isolated from phones and hand surfaces. Additionally, biotype A was dominant for C. krusei. Conclusions: C. albicans, C. glabrata, and C. krusei showed activity for all hydrolytic enzymes. The strongest correlation between the hydrolytic activity of fungi isolated from hand and phone surfaces was shown for C. albicans.

Opis:
The aim of this study was to determine the effect of the soil tillage system on soil enzymatic activity. The performed investigations, employing two soil tillage systems: classical (ploughing) and simplified (no-tillage), were carried out on Luvisols and Arenosols differing typologically, with regard to their kind and species. The activity of the following five enzymes was determined in soil samples: dehydrogenases, acid phosphatase, alkaline phosphatase, urease and protease. The applied enzymes tests turned out to be good indicators differentiating the examined soil objects depending on the employed tillage system. The employment of the simplified tillage system stimulated significantly the activity of the analysed enzymes irrespective of the soil type. This effect was particularly apparent in the top layer (0-10 cm) of the soil. An exceptionally wide range of activity was obtained for dehydrogenases indicating the usefulness of this group of enzymes for the evaluation of changes in the soil environment under the influence of the soil tillage system. The observed activity stimulation of the examined enzymes was accompanied by advantageous changes in soil chemical conditions.

Sterlux), podłoże do wysiewu i pikowania (prod. Hollas), torf ogrodniczy odkwaszony (prod. Nowy Chwalim). W podłożach oznaczono aktywność czterech enzymów z klasy oksydoreduktaz (dehydrogenaz, katalazy, peroksydaz i reduktazy azotanowej), czterech enzymów z klasy hydrolaz (fosfatazy kwaśnej, fosfatazy alkalicznej, proteaz i ureazy), odczyn oraz zawartość materii organicznej.
The aim of the study was to compare the activity of enzymes in selected horticultural growing media. Analysed media included: vegetable and flower growing medium enriched in microorganisms (Evolution Group), universal growing medium (Hollas), universal growing medium (Sterlux), growing medium for sowing and transplanting (Hollas); horticultural acid peat (Kronen) and horticultural de-acidified peat (Nowy Chwalim). The activity of four enzymes from the class of oxidoreductases: dehydrogenase, catalase, peroxidase, nitrate reductase and four enzymes from the class of hydrolases: acid phosphatase, alkaline phosphatase, urease and proteases, was determined in growing media. In addition, pH and the organic matter content were analysed. Enzymatic activity varied across horticultural growing media and the observed differences depended on the type of growing medium, pH and the type of the enzyme. The highest activity of enzymes, especially oxidoreductases, was observed in vegetable and flower growing medium enriched in microorganisms. The biggest differences among the substrates were found in the activity of phosphatases, whereas no significant differences in the activity of peroxidase and urease were observed. It could, therefore, be concluded that the medium enriched in microorganisms is most useful in flower and vegetable crops.
