Effects of a high terbuthylazine concentration (3.3 mg/l) on Cyprinus carpio were studied using a commercial herbicide formulation Click 500 SC (terbuthylazine 500 g/l). The fish were exposed to the pesticide for 24 h and allowed to recover for 6 days. Biometric parameters, plasma biochemical parameters and biomarkers of oxidative stress as well as histopathological changes in selected tissues were assessed on day 1 and 7. After a 24-h exposure, there were significant alterations found in the activities of alanine aminotransferase (ALT) and aspartate aminotransferase (AST) as well as in the plasma concentrations of glucose, natrium, chlorides, calcium and phosphorus. Hepatosomatic index, plasma albumin and lactate reflected the treatment with a delay. Ion levels and ALT were found to be restored after a 6-day recovery period, which was too short for AST activity and glucose to diminish to the control levels. The histopathological examination revealed disorders in the gills of the exposed fish, however, the changes were not detected after a 6-day recovery period. The study shows high regeneration potential of the fish.

Opis:
Over the past 50 years, the complete histological study of carp fish gonads was first carried out at Zaporozhian Reservoir. In conditions of environmental transformation of the reservoir the regularities of oogenesis and gonadogenesis, passing sexual cycles were found and the ecology of spawning carp fish was investigated. The adaptive potential of the carp fish reproductive systems, which is expressed through the asynchrony of reproductive products development as well as the functional peculiarities of the fish spawning process, was brought to light. Both environmental assessment and faunal analysis of biodiversity and distribution of young fish of Zaporozhian Reservoir were carried out. The growth and development of the fish larvae dwelling within the industrial agglomerations of Dnipropetrovsk city are found to be under anthropogenic influence. The efficiency of the use of artificial spawning nests for the carp fish spawning was studied, and practical recommendations for their installation and operation in the waters of the reservoir were elaborated.

This study aims to determine the method of generating water flow inside a round container to increase the survival and growth of common carp fingerlings. The approach used in this research is an experimental method which applies a Completely Randomized Design (CRD) and consists of three and five replications. The treatments used are (A) No Water flow (control), (B) 0.1 m/s of water flow, (C) Water flow utilizing an additional venturi system. The test fish used were 450 majalaya common carp of size 3-5 cm. The containers used in this research are in the form of 15 gallons capacity with a diameter of 27 cm x 36 cm height x bottom diameter of 6 cm containing 19 liters of water. The density of common carp during the study was 30 carp per treatment container. The treatment period is 40 days. The food given is 5% of the bodyweight of fish. Water quality parameters (temperature, pH, dissolved oxygen, ammonia and nitrates) were observed every 10 days for 40 days. Other parameters are survival, daily growth rate, feed conversion ratio, and feed efficiency observed every 10 days for 40 days. The results showed that the use of water flows with the venturi system provides the best results - with a life growth rate of 1.5%, a feed conversion ratio of 3.18, feed efficiency of 31.73% and 75% survival.

The research was conducted at the Aquaculture Laboratory of the Faculty of Fisheries and Marine Science, Universitas Padjadjaran, Jatinangor. The purpose of this research is to determine the optimum temperature for the maintenance of Common carp strain Majalaya fingerlings based on resistance to temperature stress that is known through their blood profile and survival rate. Common carp strain Majalaya fingerlings originating from Ciparay, West Java. Total body length is 8-12 cm with a minimum weight of 10 g. The method used is experimental with a completely randomized design (CRD) consisting of three treatments and five replications. The fish are kept at 24 ºC (control without heater), 29 ºC and 34 ºC. Each experimental unit contains 10 fishes. The results obtained by the survival rate (SR) of Common carp strain Majalaya which were maintained at a treatment temperature of 24 ºC by 100%, the 29 ºC by 70%, the 34 ºC by only 42% remained on the 21st day. The number of erythrocytes of common carp strain Majalaya fingerlings before the treatment temperature was 2.77 × 106 cells/mm3. The average number of erythrocytes of the 24ºC treatment from 7th, 14th and 21st day were 2.82, 2.82 and 2.81 × 106 cells/mm3; the 29 ºC were 2.93, 2.68 and 2.42 × 106 cells/mm3; the 34 ºC were 3.08, 2.52 and 1.99 × 106 cells/mm3. While the number of leukocytes of common carp strain Majalaya fingerlings before the treatment temperature was 13.61 × 104 cells/mm3. The average number of leukocytes of the 24 ºC treatment from 7th, 14th and 21st day were 13.73, 13.71 and 13.88 × 104 cells/mm3; the 29 ºC were 18.40, 16.46 and 15.76 × 104 cells/mm3; the 34 ºC were 20.78, 18.46 and 16.81 × 104 cells/mm3.

Introduction. Positron Emission Tomography (PET) is a non-invasive diagnostic tool that provides tomographic images and measures quantitative parameters of cell viability and metabolic activity of tissues. The most important used tracer in PET is an 18-F-Fluorodeoxy-glucose (FDG), the glucose molecule of which is labeled with a radiotracer and allows measurement and mapping of tissue glucose uptake. There are many studies in PET/CT which rely on some mammalian species, and recently on fish. Objective. The aim of this survey by using FDG-PET/CT are to optimize and determine FDG uptake in fish, in this case, common carp (Cyprinus carpio) using three treatments: Basic fish meal, Vita Pulvis and Probiotics s with two replications were used. Sphere Volume of interest (VOI) were drawn hand for the all organs, and standard uptake value (SUV) means were calculated. Results. The SUV mean for glucose uptake in the liver and gastrointestinal tract of fish were more similar to those of humans than rats or mice. SUV mean in fish fed by Probiotics ss in the major organ were less than those fed by Vita Pulvis and basic fish meal. The results present the opportunity to focus on studies of metabolism and screening for the effects of nutrients on body development.

Fish are aquatic animals, where they share the same ecosystem with the bacterial community they are associated with. Community structure is a concept that studies the composition of species and their abundance in a community. Information related to bacterial communities associated with their hosts is very important to support fisheries resource management efforts, both through nature conservation and aquaculture. This study of the bacterial community in the digestive tract of fish can be used to support further utilization of potential bacteria that can be used to improve the fish's immune system. This research was conducted to analyze the structure of the bacterial community found in the intestines of Common carp through a metagenomic approach using the NGS (Next-Generation Sequencing) method. The method used is a metagenomic approach, including DNA isolation from Common carp intestines. Then molecular identification was carried out using Illumina's NGS (Next-Generation Sequencing) method using the 16S rRNA gene marker. The results of the analysis using the NGS method showed as many as 19 types of bacterial phyla identified with the top three phyla, namely Fusobacteria (80%), Bacteriodetes (15%), and Firmicutes (3%). The phylum Fusobacteria is dominated by the genera Fusobacterium, Propionigenium, Ilyobacter, Leptotrichia, and Sebaldella. The phylum Bacteriodetes is dominated by the genera Porphyromonas, Odoribacter, Bacteroides, Sphingobacterium, and Barnesiella. The phylum Firmicutes is dominated by the genera Clostridium, Lactococcus, and Sarcina, Bacillus, and Lactobacillus.

The depletion of natural resources such as freshwater and cropland makes it necessary to find a new solution for sustainable food production. Aquaponic systems seem to be a great alternative to traditional agriculture, however, there are still many unknowns that need to be explored. It is already known how fish stocking affects water quality in aquaponic systems, but not how it affects the plants’ growth, and especially on chlorophyll fluorescence. In this study, we examined how the density of 0, 2, 4, 8, and 16 stocking fish in five aquaria affects lettuce growth. The first tank was only a hydroponic system with plants but without fish (control). In the remaining four aquaria - 2, 4, 8 and 12 specimens of common carp fry with an average weight of 20 grams (average 8.5-33.2 g) were placed in the aquaponic growing system. Physicochemical analysis of water was conducted to determine the levels of pH, electrical conductivity (EC), N-NO3, N-NO2, N-NH4, P-PO4, O2 and physiological parameters of plants (nitrogen balance index - NBI, chlorophyll content index - CCI, quantum yield - QY, flavonoid content - Flv) were analysed. The results showed that fish stocking density has different effects on plant physiological parameters, but in most cases, was insignificant. It seems that the greater number of fishes and higher density indirectly causes growth inhibition (lower photosynthetic efficiency) due to the increase of N-NO3 and a decrease of O2 in the water.

This study aims to determine the best density of Bacillus sp. which is given through feed to increase the body resistance of carp that is infected by pathogenic bacteria, Aeromonas hydrophila. The research was conducted at the Aquaculture Laboratory and Laboratory of Microbiology and Biotechnology at FPIK UNPAD. This study used an experimental method with a completely randomized design consisting of 4 treatments and 3 replications, namely treatment A (control), treatment B (Bacillus sp. 106 cfu/mL feed), treatment C (Bacillus sp. 108 cfu/mL feed) and treatment D (Bacillus sp. 1010 cfu/mL feed) for two weeks later in Aeromonas hydrophila infection with an intramuscular density of 108 cfu/mL for seven days. Observation parameters were clinical symptoms, fish response to feed, fish response to shock and survival. The results of this study showed that treatment C (Bacillus sp. 108 cfu/mL feed) resulted in 89% survival and reduced clinical symptoms.

The simple protein production method developed in the present study enables the production of large quantities of active recombinant common-carp growth hormone proteins in a few brief steps. With this method, the recombinant protein is obtained in a soluble form without the need for refolding. This method can be utilised for determining if the two common-carp growth hormones have similar functions in vitro and in vivo and what role each plays in growth regulation. If differences in activity exist, this method can also be used to produce intermediate forms of the two common carp growth hormones rapidly in order to determine which of the six amino acids that differ between the two proteins are responsible for the altered activity. Common carp has the same tetraploid ancestor as goldfish, and the products of the growth hormone paralogues in goldfish also differ. It would be interesting to understand how the same duplicated orthologues have evolved, and the methods elaborated in this study could be used for such comparative investigations.

Cyanobacteria (Cyanophyta, Cyanoprocaryota, Cyanobacteria) (blue-green algae) are procaryotic phototrophic microorganisms playing an important ecological role in the freshwater and marine environment as primary producers. However, as a consequence of water eutrophication observed in many reservoirs in different parts of the world, these microorganisms form massive scums, known as water blooms, releasing cyanotoxins hazardous to fish and other aquatic organisms. Cyanotoxins are cyanobacterial secondary metabolites of various chemical structures harmful to humans, terrestial and aquatic animals such as fish. The most abundant cyanotoxins are microcystins and hepatotoxins inducing toxic changes in fish liver, kidney, gills, digestive tract and immune system. Very little is known on the effects of alkaloid neurotoxic anatoxin-a on fish and their immunity. The aim of this study was to assess the in vitro influence of anatoxin-a on immune cells isolated from the common carp (Cyprinus carpio L.). The leukocyte intracellular level of ATP was reduced only at the highest concentration of anatoxin-a. Apoptotic and necrotic leukocytes were observed at the lower and the highest concentrations of anatoxin-a, respectively. Elevated activity of caspases 3/7 after 2 hours and a concentration-dependent decrease in the proliferative ability of T and B lymphocytes was also observed. The results suggest that anatoxin-a could be a possible immunotoxic agent in the aquatic environment and may increase the susceptibility of fish to infectious and neoplastic diseases. Therefore, constant monitoring of anatoxin-a and its producers in lakes and fish ponds should be performed.

Annual changes in hematological parameters of common carp juveniles under laboratory conditions. The aim of the study was to evaluate the changes of the values of hematological parameters of carp juveniles in annual cycle under stable laboratory conditions. Some parameters showed distinct rhythms of changes, e.g. 2 peaks of hematocrit occurred in II and VIII, of hemoglobin concentration in I and VI, while erythrocyte count showed maximum in II. The largest erythrocytes were observed in VIII, and the smallest in XII. Leukocyte count showed two peaks in XII and III. Maximum lymphocyte frequency occurred in III and minimum in XI, while percentage of neutrophils showed the reverse pattern. Oxidative metabolic activity of phagocytes peaked in III, while minimum occurred in XI–XII and VI–VII. Thrombocyte count was highest in XII, and lowest in VII. The obtained results revealed that the values of hematological parameters in carp considerably changed during the year despite little alterations in environmental factors. Some of these changes, e.g. increase in oxidative activity of phagocytes in spring and increase in hemoglobin level in summer were similar to those that occur in fi sh under natural conditions. Another changes, such as increase in erythrocyte size or decrease in leukocyte count suggest long-term adjustment to the laboratory environment.

The aim of the study was to assess the dynamics of cadmium and nickel accumulation in the carp, Cyprinius carpio L., during the rapid growth period between 3rd and 6th month of life, including seasonal changes. Chemical assays were performed on samples of gills, anterior and mid-posterior part of the alimentary tract, liver, kidneys, skin and muscles. Prior to the assay, 1-g tissue samples were digested with 3 ml of concentrated HNO3 in a CEM MDS 2000 microwave oven. The samples prepared this way were assayed for Cd and Ni. Cadmium level was determined with flameless graphite furnace atomic absorption spectrometry (GF-AAS) in a ZL 4110 Perkin Elmer. Nickel was determined inductively coupling plasma atomic emission spectrometry (ICP-AES) in a JY-24 Jobin Yvon apparatus. The results indicated that in the period of growth between 3rd and 6th month of life, cadmium and nickel concentrations in the examined carps fluctuated. Average cadmium content in examined organs ranged between 0.053 ÷ 0.004 µg g-1 wet weight. Average nickel content in examined organs ranged between 0.326 ÷ 0.023 µg g-1 wet weight. The observed fluctuations are supposed to be a result of intensive growth and accompanying accumulation of metals from water.
Oceniano dynamikę kumulacji kadmu i niklu w różnych narządach i tkankach karpi (Cyprinus carpio L.) w okresie intensywnego ich wzrostu, czyli między 3. a 6. miesiącem życia. Badania przeprowadzono na 240 karpiach hodowanych w wodach pochłodniczych. Z każdej ryby do analiz chemicznych pobierano próbki: nerek, wątroby, przewodu pokarmowego, listków skrzelowych, mięśni grzbietowych i skóry. Próbki narządów o masie 1 g zmineralizowano na mokro, w obecności 3 cm3 HNO3, w piecu mikrofalowym CEM MDS 2000. Kadm oznaczono metodą bezpłomieniowej absorpcyjnej spektrometrii atomowej GF-AAS, nikiel z użyciem emisyjnej spektrometrii atomowej w plazmie indukcyjnie sprzężonej (ICP-AES). Stwierdzono, że u przebadanych karpi między 3. a 6. miesiącem życia wystąpily wahania zawartości Cd i Ni. Średnie zawartości kadmu w badanych narządach wynosiły 0.053 ÷ 0.004 µg g-1 mokrej masy, natomiast średnie zawartości niklu - 0.326 ÷ 0.023 µg g-1 mokrej masy. Zaobserwowane niewielkie zmiany poziomu kadmu i niklu są wynikiem intensywnego wzrostu karpi i towarzyszącej temu kumulacji metali ze środowiska wodnego.

Trichodinids are probably the most commonly encountered protozoan parasites on fishes in marine and freshwater environments. They are typically found on the gills, skin and fins of fish. The aim of this study was to evaluate the prevalence and mean intensity of Trichodina sp. in freshwater fish from cyprinid aquaculture facilities in Macedonia. This study was carried out by seasons, in a period of three years. A total of 578 specimens of common carp (Cyprinus carpio, Linnaeus 1758), 76 specimens of grass carp (Ctenopharyngodon idella, Valenciennes 1844), 47 specimens of silver carp (Hypophthalmichthys molitrix, Valenciennes, 1844) and 53 specimens of bighead carp (Hypophthalmichthys nobilis, Richardson, 1845) from the most significant and larger cyprinid aquaculture facilities in Macedonia, including fish farms and reservoirs, were examined for parasitological investigations. Infestation with Trichodina sp. was found in 206 specimens of common carp in all aquaculture facilities examined, in all seasons, as well as, in 9 specimens of grass carp from two fish farms, only in autumn season. Total, the prevalence with Trichodina sp. was 11.84% (grass carp) and 35.67% (common carp), while the, mean intensity was 7.56 (grass carp) and 4.56 (common carp). The prevalence with Trichodina sp. in common carp by season was as following: spring – 11.82%, summer – 3.44%, autumn – 7.85%, winter – 7.05%, while the mean intensity was: spring – 4.55; summer – 4.67; autumn – 4.40; winter – 4.72. Our findings of Trichodina sp. in grass carp in the present study is considered as the first record in Macedonia. Also, this fish species is regarded as new host for Trichodina sp. in Macedonian waters.
Trichodinidae są prawdopodobnie najczęściej spotykanymi pierwotniakami pasożytującymi na rybach słodkowodnych i morskich. Zazwyczaj spotykane są na skrzelach, skórze i płetwach ryb. Celem artykułu jest określenie częstotliwości i intensywności zarażenia ryb karpiowatych przez Trichodina sp. Badania trwały trzy lata. Łącznie przebadano pod kątem parazytologicznym 578 osobników karpia (Cyprinus carpio, Linnaeus 1758), 76 amurów białych (Ctenopharyngodon idella, Valenciennes 1844), 47 tołpyg białych (Hypophthalmichthys molitrix, Valenciennes, 1844) i 53 tołpyg pstrych (Hypophthalmichthys nobilis, Richardson, 1845). Zakażenie Trichodina sp. stwierdzono u 206 osobników karpia we wszystkich badanych stawach i okresach, oraz w okresie jesiennym u 9 amurów białych z dwóch stawów hodow-lanych. Łącznie zarażonych Trichodina sp. było 11.84% amurów białych oraz 35.67% karpi, średnia intensywność zarażenia amurów białych wynosiła 7.56, a w przypadku karpia – 4.56. W okresie wiosennym zarażonych było 11.82% karpi, latem – 3.44%, jesienią – 7.85%, a zimą – 7.05%. Odpowiednio średnia intensywność zarażenia wynosiła wiosną – 4.55; latem – 4.67; jesienią – 4.40 I zimą – 4.72. Stwierdzenie Trichodina sp. pasożytującego na amurze białym jest pierwszym przypadkiem zaobserwowanym w Macedonii. W związku z tym gatunek ten jest nowym gospodarzem dla Trichodina sp. w wodach Macedonii.

This study aims to see the condition of hematocrit levels and differential leukosit after immersion in LAB isolated and after in vivo, using Aeromonas hydrophila bacterium. LAB isolated were obtained from the organs of the common carp intestine. The study used a Completely Randomized Design (RAL) consisting of four treatments and three replications. The treatment used was immersion of test fish for 24 hours in several isolates of LAB with a density of 108, namely A: without LAB isolate (control), B: isolate CcB7, C: isolate CcB8, D: isolate CcB15. Immersion was done three times, with a frequency of seven days. Parameters observed included hematocrit levels, leukocyte differentiation and survival rate. The results showed that immersion in LAB isolate CcB15 will enhance the differential leukocytes and hematocrit levels that serve as indicators of carp body resistance, which is shown by high levels of white and red blood cells.

Before being put out onto the market many fish species sold around the world need to be processed, which may result in the subsequent removal of characteristics used for their classification (head, fins, internal organs). The biochemical characterization of fish species could be achieved using proteins or DNA sequences as species-specific markers. However, since different fish products undergo different processes, the method of analysis has to be chosen according to the modifications undergone by fish constituents during processing. As DNA molecules are more resistant than proteins to various processes (including thermal treatment), DNA analysis appears to be a promising method for fish species identification. For the species identification of the Common Carp (Cyprinus carpio) among 15 different freshwater fish species a specific predesigned molecular - genetic marker of Common Carp (Cyprinus carpio) was used, which comes from the mtDNA control D - loop area. Next we analyzed the presence of mtDNA in DNA isolates of the 15 kinds of freshwater fish and compared them with the Common Carp markers by using the following two PCR identification methods. The isolates were diluted to 10 % concentration, using the TaqMan Real-Time PCR method and the SYBR® Green Real-Time PCR method. The results of using the optimized the SYBR® Green Real-Time PCR method for species identification of the Common Carp (C. carpio) pointed to its suitability. We were able to create an analysis of the monitored standard curve which represented the PCR positive control (C. carpio), containing the characteristic melting peak (up to the melting point 80.72 °C). A single peak indicated a single product (C. carpio) which can be verified upon characterization of the PCR product by agarose gel electrophoresis. The TaqMan Real-Time PCR method with a TaqMan probe is a very sensitive and reliable method of authentication used on food of animal origin. The suitability of this method, which we used for species identification of the Common Carp (C. carpio), was confirmed. Thanks to using this method, already in the 17th cycle of the PCR amplification procedure, the presence of the Common Carp gene (C. carpio) was detected in the positive control and not detected in the rest of the fish samples.
Przed wprowadzeniem do obrotu ryby są wstępnie przetwarzane, co może spowodować usunięcie tych części anatomicznych (tj. głowy, płetwy, organy wewnętrzne), na podstawie których identyfikuje się gatunek. Do identyfikacji ryb można wtedy zastosować charakterystykę biochemiczną, którą dla danego gatunku mogą być specyficzne białka lub sekwencja DNA – specyficzne markery danego gatunku ryb. Jednak ryby podlegają różnym procesom przetwórczym, w związku z czym musi być opracowana taka metoda ich biochemicznej identyfikacji, która byłaby zgodna ze zmianami składników tkanki ryb, zachodzącymi podczas tego przetwarzania. Cząsteczki DNA są stosunkowo odporne na czynniki przetwórcze (włącznie z obróbką termiczną), dlatego analiza sekwencji DNA może być przydatną metodą do identyfikacji gatunkowej ryb. W celu gatunkowej identyfikacji karpia zwyczajnego, spośród 15 innych gatunków ryb słodkowodnych, opracowano specyficzny molekularno-genetyczny marker karpia zwyczajnego, który pochodził z mDNA (z obszaru pętli kontrolnej D). Następnie analizowano izobaty mDNA z 15 różnych gatunków ryb słodkowodnych, porównując je do markera karpia za pomocą dwóch metod identyfikacji PCR. Izolaty były rozcieńczane do 10 % stężenia w obu stosowanych metodach oznaczeń tj. TaqMan Real-Time PCR i SYBR® Green Real-Time PCR. Na podstawie wyników badań karpia zwyczajnego (Cyprinus carpio), uzyskanych zoptymalizowaną metodą SYBR® Green Real-Time PCR, wykazano jej przydatność do identyfikacji gatunkowej. Monitorowano krzywą standardową topnienia PCR (z maksimum w temp. 80,72 °C) świadczącą o pozytywnej weryfikację Cyprinus carpio oraz krzywe topnienia pozostałych próbek ryb. Maksima topnienia poszczególnych (15) próbek ryb były następnie weryfikowane metodą elektroforezy żelowej na agarozie. Druga z zastosowanych metod TaqMan Real-Time PCR, z wykorzystaniem próbnika TaqMan, jest bardzo dokładną i wrażliwą metodą identyfikacji (potwierdzania autentyczności) karpia zwyczajnego (Cyprinus carpio). Dzięki zastosowanie metody amplifikacji już w 17. cyklu potwierdzono obecność, poprzez PCR, genu Cyprinus carpio w próbce kontrolnej i brak obecności tego genu w pozostałych próbkach ryb.

Mitracarpus scaber, an endemic medicinal plant to Nigeria, Africa, has medicinal value. In the present study, Mitracarpus scaber leaves extract (MSLE) was fed to common carp, Cyprinus carpio to evaluate its effect on growth performance, nutrient utilization, gut morphometry, and innate immunity parameters. Four isonitrogenous diets (32% crude protein) containing 0.0, 5, 10, or 15 g MSLE/kg diet were fed to fish (7.52 ±0.23 g) for 12 weeks. After the feeding trial, fish were exposed to pathogenic bacteria (Aeromonas hydrophila) for 14 days. Growth performance, nutrient utilization, and feed intake were significantly improved with increasing MSLE levels up to 10 g/kg diet. Similarly, fish fed MSLE diets increased significantly intestinal villi length/width, and absorption area. Furthermore, activities of respiratory burst, lysozyme, catalase, and superoxide dismutase were significantly higher in fish fed diets containing MSLE levels, and their highest values were obtained at fish fed 15 g MSLE/kg diet. After bacterial challenge, fish mortality was lowest (8.45 ±1.30%) in fish fed 15 g MSLE/kg diet, whereas highest mortality (52.50 ±4.56%) was observed with fish fed the control diet. The present study conjured that MSLE inclusion in fish diets with optimum level of 10 g/kg diet stimulated significantly the performance, nutrient utilization, modified gut morphometry, and innate immune response of common carp. Also, its inclusion protected fish against pathogenic bacteria, A. hydrophila infection.
Mitracarpus scaber, endemiczna roślina lecznicza z Nigerii w Afryce, ma wartość leczniczą. W niniejszym badaniu ekstrakt z liści Mitracarpus scaber (MSLE) podawano karpiowi, Cyprinus carpio, aby ocenić jego wpływ na szybkość wzrostu, wykorzystanie składników odżywczych, morfometrię jelit i parametry odporności wrodzonej. Cztery izoazotowe diety (32% surowego białka) zawierające 0,0; 5, 10 lub 15 g diety MSLE/kg podawano młodym rybom (7,52 ±0,23 g) przez 12 tygodni. Po tym okresie ryby były narażone na bakterie chorobo-twórcze (Aeromonas hydrophila) przez 14 dni. Szybkość wzrostu, wykorzystanie składników odżywczych i spożycie paszy zwiększały się wraz ze wzrostem poziomu MSLE do wartości 10g/kg diety. Ryby ze wzbogaconą dietą znacznie zwiększyły stosunek długości/szerokości kosmków jelitowych i obszar wchłaniania. Ponadto wielkość wybuchu tlenowego, oraz aktyw-ność lizozymu, katalazy i dysmutazy ponadtlenkowej były znacznie wyższe u ryb karmionych pokarmem wzbogaconym o MSLE. Najwyższe wartości wyżej wspomnianych parametrów uzyskano u ryb karmionych pokarmem wzbogaconym o 15 g MSLE/kg. Po wprowadzeniu bakterii śmiertelność ryb była najniższa (8,45 ±1,30%) u ryb karmionych 15 g diety MSLE/ kg, podczas gdy najwyższą śmiertelność (52,50 ±4,56%) obserwowano u ryb karmionych dietą kontrolną. W niniejszym badaniu wykazano, że włączenie MSLE w ilości 10 g/kg diety zna-cząco stymuluje wydajność, wykorzystanie składników odżywczych, modyfikuje morfometrię jelit i wrodzoną odpowiedź immunologiczną karpia. Włączenie do diety MSLE chroniło ryby przed infekcją bakteriami chorobotwórczymi, A. hydrophila.

The effects of rearing conditions on hematology and susceptibility of common carp to experimental manipulation stress. The aim of present study was to compare hematological values in pond-reared and hatchery-reared common carp, and their hematological alterations following experimental manipulation. Two groups of carp: pond, and hatchery-reared juveniles were subjected to experimental manipulation – transfer of each fi sh for three hours to separate, aerated but confi ning aquaria. Blood was sampled and hematological parameters were evaluated (erythrocyte and leukocyte counts, hemoglobin concentration, hematocrit, metabolic activity of phagocytes), and differential erythrocyte and leukocyte counts were calculated in the smears. Prior to stress, pond fi sh showed higher erythrocyte count and hemoglobin concentration, lower erythrocyte volume, and higher leukocyte count and phagocyte activity comparing to the hatchery fi sh. Reaction of both groups of fi sh to manipulation also differed. Pond fi sh showed erythrocyte swelling, and strong leukopenia (lymphopenia and neutropenia), and a decrease in phagocyte activity, while in hatchery fi sh increase in erythrocyte count and phagocyte activity took place. Rearing conditions signifi cantly affected hematological parameters of fi sh, pond-reared carp showing higher oxygen transport and immunological capacities comparing to the hatchery-reared ones. Pond fi sh showed also higher susceptibility to stress.

In contrast to terrestrial animals, fish can ingest minerals from food or directly from water. Although micro- and macro-elements are needed in small quantities, they play a key role in many metabolic processes. Trace mineral and macromineral deficiencies may go unnoticed due to an absence of clear clinical symptoms in fish. Absorption processes are determined by various factors, mostly mineral concentrations in water but also other water parameters. The required dietary supplementation of macronutrients and micronutrients is very difficult to determine, and the amount of nutrients absorbed by fish from water is equally difficult to measure. Interactions between elements should also be taken into consideration. Many authors emphasize that phosphates may reduce the absorption of most micronutrients. Also, the current parameters of the water can affect the bioavailability. Some elements such as calcium, chlorine and sodium can be absorbed from ambient water in a quantity sufficient to meet the demand for this element. Other elements, however, require supplementation in a diet. For example, studies indicate the need for supplementation of phosphorus, zinc, copper and manganese. Most research concentrates on feedstuff as a source of micro- and macronutrients. Meanwhile, information concerning bioavailability of minerals directly from water is scarce. The aim of this study was to analyse literature from a different perspective, and concentrate on water as a source of minerals in fish nutrition. Measurements of water parameters such as temperature, pH, nitrate and nitrite levels and the amount of dissolved oxygen are a regular component of environmental control in fish farming. Determination of micro- and macro-element levels, however, remains uncommon in aquaculture. Measurements of these parameters could suggest which elements need to be supplemented and which are found in water in amounts that satisfy the needs of the fish.

The paper evaluates the effect of culture conditions and culture site on levels of certain microelements (Zn, Cu, Fe) and macroelements (Mg, Ca) in three species of freshwater fish: rainbow trout (Oncorhynchus mykiss Walbaum), common carp (Cyprinus carpio L.), and Siberian sturgeon (Acipenser baeri Brandt). The study involved 90 individuals of freshwater fish aged from 6 to 12 months. Samples of blood, liver, kidney, gills, skin and muscles were collected from each fish and subjected to chemical assay of Mg, Ca, Zn, Cu, Fe with inductively coupled plasma-atomic emission spectrometry in a JY-24 Jobin Yvon apparatus. The study revealed that culture site had statistically significant impact on levels of the examined elements among the three fish species. Mg content in kidney and skin was significantly higher in carp than in sturgeon. Similar regularities were observed for Ca content in skin and Zn content in kidney. Liver and kidney levels of Fe and Cu were significantly lower in carp than in rainbow trout. Of all the three examined fish species, rainbow trout had the highest skin levels of Ca and Mg, and the highest blood level of Fe. The results indicate that culture site and culture conditions exerted significant influence on levels of macro- and microelements in freshwater fish.
Celem pracy była ocena wpływu warunków i miejsca prowadzenia hodowli na zawartość mikroelementów (Zn, Cu, Fe) i makroelementów (Mg, Ca) w organizmach ryb słodkowodnych. Ocenie poddano trzy wybrane gatunki ryb: pstrąga tęczowego (Oncorhynchus mykiss Walbaum), karpia (Cyprinus carpio L.) i jesiotra syberyjskiego (Acipenser baeri Brandt). Badaniu poddano 90 ryb słodkowodnych w okresie od 6. do 12. miesiąca życia. Z każdej ryby do analiz chemicznych pobrano próbki: krwi, wątroby, nerek, skrzeli, skóry i mięśni. Próbki poddano analizie na zawartość Mg, Ca, Zn, Cu, Fe, z użyciem emisyjnej spektrometrii atomowej w plazmie indukcyjnie sprzężonej (ICP-AES), w aparacie Jobin Yvon typ JY-24. Wykazano, że miejsce hodowli ma statystycznie istotny wpływ na zawartości badanych pierwiastków u poszczególnych gatunków ryb słodkowodnych. Zawartość Mg w nerce i skórze karpi była istotnie wyższa niż u jesiotra. Podobne spostrzeżenia dotyczyły poziomu Ca w skórze i Zn w nerce. W przypadku Fe i Cu obserwowano niższą zawartość w wątrobie i nerkach karpi niż w tych samych narządach u pstrąga. Badania wykazały, że poziom Zn i Cu we wszystkich narządach przebadanych gatunków kształtował się na najniższym poziomie. Spośród badanych gatunków u pstrąga stwierdzono najwyższą zawartość Ca i Mg w skórze, a Fe we krwi. Otrzymane wyniki pozwalają stwierdził, że miejsce i warunki prowadzenia hodowli mają istotny wpływ na zawartość badanych makro- i mikroelementów w organizmach ryb słodkowodnych.

Opis:
The aim of the study was to estimate the effect of culture conditions and culture site on magnesium (Mg) and zinc (Zn) concentrations in freshwater fish. The study encompassed dorsal muscles in five fish species: common carp (Cyprinus carpio L.), rainbow trout (Oncorhynchus mykiss Walbaum) and Siberian sturgeon (Acipenser baeri Brandt), northern pike (Esox lucius L.) and grass carp (Ctenopharyngodon idella Valenciennes). A total of 125 fish comprised 25 individuals of each species, aged from 6, 9, and 12 months. The fish were cultured in privately owned fish breeding ponds (Western Pomerania, Poland). For chemical and biochemical assays, samples of dorsal muscles were taken from each fish. Tissue samples were wet mineralised in concentrated HNO3 in a CEM MDS 2000 microwave oven. Magnesium and zinc concentrations were determined by inductively coupled plasma atomic emission spectrometry (ICP-MS) in a Jobin Yvon type JY-24 apparatus. The pursuit of the research we had an approval of the Polish Local Ethics Committee nr 9/05. The magnesium concentration in the dorsal muscles ranged from 95.3÷347.6 mg kg–1 w.w. The highest Mg concentration was found in rainbow trout (347.6±32.2 mg kg–1 w.w.), and the lowest in grass carp (95.3±11.3 mg kg–1 w.w.). Zinc concentration varied from 6.7÷98.8 mg kg–1 w.w. The highest was found in the muscles of Siberian sturgeon (98.8±0.4 mg kg–1 w.w.), and the lowest in rainbow trout (6.7±0.7 mg kg–1 w.w.). It was found that the breeding place significantly affected the Zn and Mg concentrations in the muscle tissue among individual freshwater fish species. The magnesium and zinc concentrations were also significantly affected by the type of feed.
Celem pracy była ocena wpływu warunków i miejsca hodowli na zawartość magnezu (Mg) i cynku (Zn) w organizmach ryb słodkowodnych. Ocenie poddano mięśnie grzbietowe pobrane od pięciu wybranych gatunków ryb słodkowodnych: pstrąga tęczowego (Oncorhynchus mykiss Walbaum), karpia (Cyprinus carpio L.), jesiotra syberyjskiego (Acipenser baeri Brandt), szczupaka pospolitego (Esox lucius L.) i amura białego (Ctenopharyngodon idella Valenciennes). Badaniami objęto ogółem 125 ryb słodkowodnych, po 25 osobników z każdego gatunku. Materiał do badań pobrano w 6., 9. i 12. miesiącu życia ryb. Z każdej ryby do analiz chemicznych pobrano próbki mięśni grzbietowych. Pobrany materiał zmineralizowano na mokro w stężonym HNO3 w piecu mikrofalowym CEM MDS 2000. Próbki poddano analizie na zawartość Mg i Zn z użyciem emisyjnej spektrometrii atomowej w plazmie indukcyjnie sprzężonej (ICP-AES) w aparacie Jobin Yvon typu JY-24. Na powyższe badania uzyskano zgodę Lokalnej Komisji Etycznej nr 9/05. Zawartość Mg w mięśniach mieściła się w przedziale 95.3÷347.6 mg kg–1 m.m. Najwyższy poziom Mg odnotowano u pstrąga (347.6±32.2 mg kg–1 m.m.), a najniższy u amura (95.3±11.3 mg kg–1 m.m.). Zawartość Zn mieściła się w przedziale 6.7÷98.8 mg kg–1 m.m. Najwyższy poziom Zn odnotowano w mięśniach jesiotra syberyjskiego (98.8±0.4 mg kg–1 m.m.), a najniższy u pstrąga (6.7±0.7 mg kg–1 m.m.). Wykazano, że miejsce hodowli ma statystycznie istotny wpływ na zawartości magnezu i cynku w tkance mięśniowej poszczególnych gatunków ryb słodkowodnych. Na poziom magnezu i cynku w tkance mięśniowej istotne znaczenie miał również wpływ rodzaj spożywanego pokarmu.
