Opis:
Cadmium is a dangerous occupational and environmental toxin. It accumulates in the human organism mainly in liver and kidneys. Cadmium half-life is about 10 years, so the symptoms of cadmium intoxication may occur several years after the exposure. Until now in treating intoxication with this metal chelating compounds have been used, burdened with numerous undesirable symptoms. In our investigations anthocyanins from Aronia melanocarpa were used to reduce the harmful results caused by cadmium. Administering anthocyanins with cadmium chloride resulted in a statistically significant decrease of aspartate aminotransferase (AST) and alanine aminotransferase (ALT) activity, concentration of bilirubin and urea in blood serum and decreased cadmium cumulation in liver and kidneys in relation to animals receiving cadmium chloride only.

ObjectivesThe aim of this study was to determine the physiological and biochemical changes among bakers, induced by heat exposure at the workplace.Material and MethodsInformation was collected by means of a self-administered questionnaire. Vital signs were measured and recorded by a trained nurse before and after each work shift. A venous blood sample was drawn at the end of each work shift. The mean wet-bulb globe temperature (WBGT) index in the bakeries and offices was measured.ResultsThis was a cross-sectional study involving 137 bakers working in 20 bakeries, and 107 control subjects comparable in terms of age, race, marital status, years of service, income, and cigarette smoking. There was a significant weight loss and increase in the respiratory rate, the heart rate, and oral body temperature among the bakers compared to the control group. Sodium levels were significantly lower, while blood urea nitrogen levels were significantly higher, among the bakers than in the control group. There were unfavorably hot working conditions in the bakeries: the WBGT index in the bakeries was 37.4°C while the average WBGT for the offices was 25.5°C.ConclusionsThe changes in the physiological and biochemical parameters among the bakers were found to be associated with exposure to high environmental heat in the bakeries, as judged by the WBGT index. Preventive measures should be aimed at reducing the adverse effects of heat exposure among bakers and should be directed towards the man-machine-environment triad.

Opis:
Infections caused by nematodes of the subfamily Cyathostominae affect nearly 100% of pastured horses. Despite of an absence of pronounced symptoms, cyathostominosis can have very serious health consequences. The aim of this study was to monitor changes in total protein levels and concentrations of selected microelements and macroelements in the blood of horses before and after ivermectin treatment. In healthy horses infected by the studied parasites, total blood protein levels were below the physiological norm, but iron (Fe), magnesium (Mg), calcium (Ca) and phosphorus (P) concentrations were within normal limits. Ivermectin treatment reduced the number of excreted parasite eggs (FEC) by 100%, and dead parasites were observed in feces. Decreased iron (Fe) concentrations and an insignificant increase in total blood protein levels were reported. A progressive decline in iron levels was observed when parasite eggs reappeared in feces 60 days after treatment. Iron loss takes place as a result of bleeding from the large intestine when adult nematodes affected by the drug are removed from intestine and fourth-stage larvae leave parasitic nodules in the intestinal wall. A drop in iron levels could be an indirect indicator of the severity of cyathostominosis.

Opis:
Zearalenone (ZEA) is a mycoestrogen frequently found in food and animal feed materials all over the world. Despite its hydrophobic character, ZEA is also found in surface and ground waters which suggests an environmental risk for aquatic animals. Knowledge concerning mycotoxin-related mechanisms of toxicity is still incomplete, e.g. little is known about the influence of ZEA exposure on fish. The aim of this study was to investigate the effect of ZEA on selected biochemical parameters in juvenile rainbow trout after 24, 72, and 168 h of intraperitoneal exposure (10 mg/kg of body weight). The analysis showed a slight tendency towards prolonged blood clotting time and significant iron deficiency in the liver and ovary of exposed animals. However, no differences in aminotransferase (AlaAT, AspAT) activity or glucose levels in fish plasma was observed. The results of this study suggest that although trout exposed to ZEA did not exhibit any distinct symptoms of liver damage, the mycotoxin tested was able interfere with blood coagulation and iron-storage processes.

Opis:
Introduction. Nicotine is an alkaloid that affects the functioning of the central nervous system and produces dependence. In low doses, it acts as a stimulant and relaxant. Nicotine was reported to have pro-cognitive effects in humans and animals. However, high doses of nicotine are harmful for many organs. The aim of the study was to check whether a 30-day exposure to transdermal nicotine affects memory and biochemical parameters in mice. Materials and method. A total of 32 mice (16 males and 16 females) were used in the experiment. Mice were divided into 4 groups of 8 animals each: I control-females receiving placebo patches for 30 days, II females receiving nicotine patches for 30 days, III control-males receiving placebo patches, IV males receiving nicotine patches. Spontaneous alternation and locomotor activity were examined weekly in a Y-maze. Body mass was recorded daily. On day 30, venous blood samples were obtained and the animals were anaesthetized with CO2. Their blood was used to measure alanine transaminase (ALT), asparagine transaminase (AST), cholesterol, creatinine and glycosylated haemoglobin (HbA1C). Results. Nicotine significantly improved memory in male mice on day 8. It increased ALT and AST activities in males and females, as well as the concentration of cholesterol in their blood sera. Conclusions. In conclusion, transdermal nicotine may produce transient improvement in fresh spatial memory in male mice, but it is not a long-term effect and therefore nicotine does not seem to be appropriate for use in the treatment of neurodegenerative disorders. It elevates blood cholesterol level and thus may increase the risk of atherosclerosis and cardiovascular events; moreover, it negatively affects liver enzymes. Nicotine use is therefore not recommended.

Selected qualitative and biochemical parameters were determined in cryopreserved semen used for artificial insemination, sampled from 120 bulls reared at the Animal Breeding and Insemination Center in Bydgoszcz. The total average motility of the analyzed sperm samples was determined at 62.51%. The percentage of motile spermatozoa displaying progressive forward motility was 21.65%. Analyzed samples were characterized by a high percentage of sperm cells with a intact plasma membrane (71.21%) and active mitochondria (71.32%). High efficiency of the enzymatic antioxidant system of the evaluated sperm cells was demonstrated by high activity of CAT, GPx and SOD (494.37, 2847.83 and 5.31U/1x109 spermatozoa, respectively) values and low values of the DNA Fragmentation Index (9.32). The results of the study, obtained with the involvement of advanced analytical methods, indicate a high fertilizing capability of the analyzed sperm samples.

Opis:
The increasing interest in enzymatic growth promoters prompted this investigation of the effect of an enzymatic stimulating complex on selected hematological and serum biochemical parameters in boars. The enzymatic complex [five proteases (proteinases-endopeptidases) and two peptidases (exopeptidases) obtained by fermentation from Streptomyces fradiae] was added to diets for three months, at initial doses of 90 (group El) and 120 (group E2) g/ton feed for the first seven days followed by 40 and 60 g/ton, respectively, in the subsequent weeks. The evaluation was based on selected hematological (RBC, WBC, HCT (Ht), HGB (Hb), MCV, MCH, and MCHC) and biochemical (AST, ALT, AP, Ca, Mg, urea, cholesterol, creatinine, inorganic phosphorus, and oxyglucose) parameters determined before and after the experimental period. The reported values were within physiological norms. Statistical differences were found between experimental groups with respect to WBC, HCT (Ht), MCV, MCH, AP, Ca, Mg, creatinine, and inorganic phosphorus. The study results indicate that the complex of proteolytic enzymes administered to boars caused no negative changes in their metabolic profiles. The statistically different hematological and serum biochemical parameters, while within normal limits, point to an early stage of microcytic anemia and heightened agitation that could be the result of intensified nitrogen metabolism.

Spormann score
Opis:
For many years, there has been a search for a set of biochemical parameters that could facilitate the assessment of severity, prognosis, and administration of early and appropriate treatment in acute pancreatitis. Administration of treatment within the first 48 hours since admission is associated with many problems of distinguishing patients with a mild form of acute pancreatitis (AP) from those with a severe form of acute pancreatitis. Study aim: To assess the relationship between the extent of change in the concentration of 10 selected biochemical indicators: amylase, lipase, total bilirubin, creatinine, uric acid, aspartate transaminase, alanine transaminase, glucose, magnesium, and iron and histopathological lesions in the pancreas within 2 and 6 hours since induction of AP. The selected time periods correspond to the first and the second day of the disease in people, respectively. Material and methods: The experiments were conducted in 110 male Wistar rats weighing from 250 to 300 g. Experimental animals were divided into three groups: Z – a group in which the ranges of the studied factors and histological structure were established; K – a group of animals operated on which were injected with 0.9% NaCl into the biliary-pancreatic duct; E – a group of animals operated on in which acute pancreatitis was induced by an injection of 5% sodium taurocholate into the biliary-pancreatic duct. Animals from the K and E groups were randomly assigned to one of five subgroups from which the material for biochemical and histological examinations was collected at 2 h and 6 h since the induction of AP. Whole pancreases were dissected for histological examinations, and the samples were dyed with hematoxylin and saturated alcoholic eosin solution. The degree of pancreatic lesions was assessed according to the Spormann score. Quantitative variables were characterized by arithmetic means, standard deviations, medians, minimum and maximum values, and 95% CIs. Results: In histological preparations from rats from the E group, after 2 hours, edematous lesions, neutrophilic infiltrations in the pancreatic parenchyma, together with single petechiae started to appear and were observed. After 6 hours, the lesions became more intense, and minor foci of coagulation necrosis and minor foci of purulent inflammation in the fatty tissue appeared. Within 2 hours, statistically significant differences in the amount of four markers: creatinine, ALT, amylase, and magnesium were observed. After six hours, statistically significant differences in the amount of two markers: AST and glucose were seen. The correlations between histological assessments according to the Spormann scale and biochemical indicators were investigated, and it was observed that within 2 hours the intensity of pancreatitis increased together with an increase in AST. In group K, within 6 hours, the intensity of inflammatory infiltration increased together with an increase in creatinine concentration (correlation coefficient 0.95; p=0.0138). In group E, in the period of 2 hours, lesion intensity in the form of inflammatory infiltration increased together with an increase in the AST level (correlation coefficient 0.90; p=0.0063) and an increase in the iron level (correlation coefficient 0.78; p=0.0399). In the same group and in the same period, an increase in the AST level (correlation coefficient 0.79; p=0.0343) was associated with an increase in lesion intensity in the form of ecchymoses. Inflammatory infiltration increased (correlation coefficient -0.87; p=0.0117) within 6 hours, whereas the creatinine level decreased. Interesting results were obtained with the use of regression analysis – forward stepwise regression. In the period of 2 hours, if the creatinine level increased by 1, the intensity of lesions in acute pancreatitis decreased by 9.02, according to the Spormann score, while the other variables remained at a stable level. However, if ALT level increased by 1, the intensity of lesions in acute pancreatitis increased by 0.02, according to the Spormann score; and if the amylase level increased by 1, the intensity of lesions in acute pancreatitis increased by 0.01, according to the Spormann score, while the other variables remained at a stable level. Conclusions: Histopathological lesions occurred prior to changes in laboratory test results, whereas significant correlations with Spormann scores were seen in the case of changes in AST and creatinine levels. The study results confirm the fact that diagnostics in acute pancreatitis is very difficult and requires monitoring of many laboratory parameters.

This study examined the impact of oral supplementation with a new organic selenium preparation (selenitetriglycerides) on the health of camels (Camelus dromedarius). The experiment was conducted on 7 female camels, which were given 240 mg per animal per day of selenitetriglycerides with an oesophageal probe and subsequently monitored for 28 days. Blood samples for tests were collected 10 times (on day 1, 2, 3, 4, 5, 6, 7, 8, 14 and 28) and the following parameters were determined: the level of selenium, vitamin E, the activity of aspartate transaminase (AST), gamma-glutamyl transpeptidase (GGTP), lactate dehydrogenase (LDH), creatine kinase (CK) and the concentration of total protein, glucose, urea and creatinine. The following hematological parameters were determined in whole blood: white blood cell count (WBC), red blood cell count (RBC), hemoglobin level (HGB), platelet count (PLT) and packed cell volume (PCV). Selenium serum level increased significantly on the second day (from 40.18 µg l-1 to 198.79 µg l-1), peaked on day 7 (514.76 µg l-1) and remained at a statistically significant level relative to the baseline to the end of the experiment. Clinical observations and analysis of the hematological parameters, liver and kidney function parameters did not show any negative effect of selenitetriglycerides on camel health. The results indicate that selenitetriglycerides can be a safe and effective method of selenium supplementation in camels.

The activity of aspartate aminotransferase, alanine aminotransferase and lactate dehydrogenase, total protein, albumin and cholesterol levels were determined in the blood serum of Japanese quails infected with coccidia and treated with Baycox (active ingredient: toltrazuril). Lower levels of AST and ALT activity were noted in treated birds regardless of the applied Baycox dose. The biochemical changes observed in the blood serum of Japanese quails point to coccidia-induced damage of digestive system tissues despite an absence of pronounced clinical symptoms. Significantly lower levels of AST activity and higher levels of LDH activity in treated birds indicate that coccidiosis treatment with toltrazuril contributed to the regeneration of digestive system tissues. An insignificant increase in cholesterol levels was noted, whereas the other serum biochemical parameters remained within the reference ranges.
