Background. During the last few decades, adiposity has become a relatively common phenomenon worldwide. The available data on the effects of pro-inflammatory factors in both depression and adiposity has been attracting great attention. Aim. We sought to assess the prevalence of -889C>T IL-1α, -31T>C and -511C>T IL-1β, -330T>G IL-2 and -174G>C IL-6 genes and their association with adiposity and depression in Polish subjects. Methods. A cohort study was conducted in 2013/2014, covering a sample of 297 individuals (217 female and 80 male). Anthropometric data was handled using the BIA analysis method, while for genotyping PCR-RFLP techniques were used. Results. A positive correlation between depression and anthropometric parameters: adipose tissue (in kg) and adipose tissue (in %) (R=0.135 and p=0.02, R=0.114 and p<0.05, respectively) was found. No association between studied polymorphisms and depression was observed. Conclusion. Although it was not possible to demonstrate any influence of the studied polymorphisms as the genetic modulator of depression, authors believe that the presented data are noticeable and may provide the basis for future studies on larger groups.

Opis:
This study investigated the effect of T6 heat treatment on the microstructure and scratch wear behavior of hypoeutectic Al-12wt.%Si alloy manufactured by extrusion. Microstructural observation identified spherical eutectic Si phases before and after the heat treatment of alloys (F, T6). Phase analysis confirmed Al matrix and Si phase as well as Al2Cu and Al3Ni, Mg2Si in both alloys. In particular, Al2Cu was finer and more evenly distributed in T6 alloy. This resulted in Vickers hardness of T6 alloy that was 2.3 times greater compared to F alloy. The scratch wear test was conducted using constant load scratch test (CLST) mode and multi-pass scratch test (MPST) mode. The scratch coefficient and worn out volume obtained by such were used to evaluate wear properties before and after heat treatment. In the case of T6 alloy, its scratch coefficient was lower than F alloy in all load ranges. After 15 repeated tests to measure worn out volume, F alloy and T6 alloy measured 1.2×10-1 mm3 and 7.8×10-2 mm3, respectively. In other words, the wear resistance of T6 alloy were confirmed to be better than those of F alloy. In addition, this study attempted to identify the microstructural factors that contribute to the better scratch wear resistance of T6 alloy and wear mechanism from surface and cross-section observations after the wear tests.

Opis:
Celem przeprowadzonych badań było opracowanie elektrochemicznych biosensorów do wykrywania obecności prozapalnej cytokiny, interleukiny 6 (IL-6), w roztworze soli fizjologicznej buforowanej fosforanami (phosphate bufferred saline PBS). Do tego celu wykorzystano nowe, modyfikowane powierzchniowo warstwy nanorurek ditlenku tytanu (TNT – titania nanotubes) wytworzone w procesie anodowania folii tytanu w roztworze glikolu etylenowego. Warstwy TNT poddano obróbce termicznej poprzez wyżarzanie w atmosferze argonu i funkcjonalizowano przez bezpośrednią immobilizację przeciwciał cytokiny IL-6 na ich powierzchni metodą nakraplania. Oznaczenie stężenia IL-6 w roztworze PBS badano za pomocą dwóch typów biosensorów elektrochemicznych: impedancyjnego i amperometrycznego, wykorzystując oddziaływania pomiędzy ludzkim przeciwciałem monoklonalnym IL-6 i antygenem IL-6. Działanie biosensora impedancyjnego polegało na określeniu zmian wartości pojemności elektrycznej warstwy powierzchniowej (a równocześnie kąta przesunięcia fazowego jej charakterystyki impedancyjnej) na skutek powstawania kompleksu antygen-przeciwciało IL-6. Do wykrywania antygenu IL-6 w biosensorze amperometrycznym zastosowano natomiast przeciwciało IL-6 skoniugowane z peroksydazą chrzanową (HRP). Limity detekcji IL-6 opracowanych biosensorów wynoszące 5 pg/ml są znacznie niższe od limitu detekcji w obecnie stosowanych testach immunoenzymatycznych ELISA (25 pg/ml). Zarówno korzystne właściwości adsorpcyjne i dobre przewodnictwo elektryczne materiału podłoża biosensorów, umożliwiającego zastosowanie bezpośredniej immobilizacji substancji biologicznych, jak i stosowane dwie metody elektrochemiczne: impedancyjna oraz amperometryczna sprawiają, że opracowana metoda detekcji cytokin jest szybka, tania, selektywna i czuła.
Sensitive electrochemical biosensors for determination of proinflammatory cytokine Interleukin-6 are elaborated. They are based on novel and surface modified layers of titania nanotubes (TNT) formed by anodization of titanium foil in ethylene glycol solution. The TNT layers were thermally improved by annealing in argon and functionalized for biosensing by direct immobilization of biological reagents on their surfaces with the simple drop technique. Strong antibody-antigen interaction was used in two types of electrochemical biosensors: the impedimetric and the amperometric to detect the IL-6 concentrations in phosphate buffered solutions (PBS). The impedimetric biosensor determines changes of capacitance values due to the formation of IL-6 antibody-antigen complex on biosensor platform. Horseradish peroxide (HRP) conjugated antibodies are used to detect IL-6 antigen in the amperometric biosensor. Two novel electrochemical methods allow to detect 5 pg/ml of IL-6 in PBS solution, which is below detection limit 25 pg/ml in currently used enzyme-linked immunosorbent assay (ELISA) tests. The favorable adsorption and electrical conductance properties of the platform material, which allow the direct immobilization of biological substances on its surface and give the possibility to apply two electrochemical techniques: the impedimetric and the amperometric one, ensure the elaborated cytokine detection methods to be fast, inexpensive, selective and sensitive.

Inflammation plays an important role in the development of many obesity-related diseases. This study aimed to investigate the effect of ezetimibe on inflammation and myocardial remodeling in obese rats. A rat model of obesity was established, and myocardial damage was examined by transmission electron microscopy and Masson staining. Twenty obese rats were divided into two groups (n=10): obese group and ezetimibe group. Ten SD rats were used as controls. Western blot was performed to monitor the expression of P-p38MAPK and interleukin (IL)-6. Immunohistochemical staining was used to monitor the expression of intercellular adhesion molecule-1 and vascular cell adhesion molecule-1. In the obese rats group, we observed increased inflammatory factors and myocardial hypertrophy. In contrast, the ezetimibe group exhibited decreased expression of inflammatory factors and an improvement in myocardial remodeling compared to the obese group. Mechanistically, we found that ezetimibe decreased P-p38MAPK, IL-6, intercellular adhesion molecule-1, and vascular cell adhesion molecule-1 levels in the hearts of the obese rats. Taken together, these results indicate that ezetimibe may improve myocardial remodeling in obese rats by inhibiting inflammation.

Opis:
The results of study of Pb influence on high temperature magnetic and thermal properties of the chalcogenides $(Pb_ySn_{1-y})_2P_2S_6$ are presented. The increasing Pb content shifts phase transition to the ferroelectric state at about 337 K towards lower temperatures while magnetic field till 3 T has no influence on this transition. The measured susceptibility and magnetisation data are discussed.

Come giustamente ha rilevato Moreschini (Gregorio di Nissa, Omelie sul Cantico dei Cantici, a cura di C. Moreschini, Roma 1996, Intr. p. 9), tutto il complesso delle Homiliae in Canticum Canticorum di Gregorio «è costruito sullo schema dell’epektasis» e non è esagerato affermare che «esse vogliono rappresentare una serie di esperienze successive dell’anima la quale, dopo avere avuto un contatto, sia pure parziale, con lo sposo divino, approfondisce sempre di più il suo rapporto spirituale con lui». Ma soprattutto a proposito dell'esegesi di Cant. 1, 5-6 (Sono nera e bella, o figlie di Gerusalemme, come le tende di Kedar, come le pelli di Salomone. Non guardatemi perché sono stata fatta nera, se il sole mi guardò di traverso...) l’epéktasis è inserita in un contesto di ampio respiro e assume un significato compiuto in una prospettiva soteriologica. Dunque, con il nostro intervento, si vuole portare l’attenzione specificatamente sulle Homiliae 2 e 4 che sono dedicate all’interpretazione di Cant. 1, 5-6 e si vogliono mettere a fuoco le varie argomentazioni con le quali il Nisseno, ormai alla fine della vita, attribuì il progresso spirituale dell’anima all’azione salvifica di Dio, in linea con quanto aveva sostenuto molti anni prima nel De virginitate 12,2 («non è opera nostra… divenire simili alla Divinità, ma è il risultato della munificenza di Dio, che fin dalla sua prima origine ha fatto grazia della somiglianza con Lui alla nostra natura»).
As Moreschini rightly pointed out (Gregorio di Nissa, Omelie sul Cantico dei Cantici, a cura di C. Moreschini, Roma 1996, Intr. p. 9), Gregory of Nyssa’s Homilies on the Song of Songs «sono costruite sullo schema dell’epektasis», and it is no exaggeration to say that «esse vogliono rappresentare una serie di esperienze successive dell’anima la quale, dopo avere avuto un contatto, sia pure parziale, con lo sposo divino, approfondisce sempre di più il suo rapporto spirituale con lui». But above all about the exegesis of Ct. 1: 5-6 (I am dark, but lovely, You daughters of Jerusalem, Like Kedar’s tents, Like Solomon’s curtains. Don’t stare at me because I am dark, Because the sun has scorched me…) the epektasis is placed in a wide-ranging context and takes on a new meaning in a soteriological perspective. Hence the purpose of the present study is to analyze specifically the Homilies 2 and 4, concerning the exegesis of Ct. 1: 5-6, and to focus on all the theological topics (the gratuity of grace, the free acceptance of the gift of grace) with which Nyssen, now at the end of life, attributed the spiritual progress of the soul to the saving action of God, in accordance with what he had claimed, many years earlier, in De virginitate 12 («In fact this likeness to the divine is not our work at all…; it is the great gift of God bestowed upon our nature at the very moment of our birth»).

Interleukin 1 (IL-1) and interleukin 6 (IL-6) are typical examples of multfunctonal pro-infammatory cytokines involved in the regulaton of the immune response, hematopoiesis, and infammaton. Both peripheral and intraventricular administraton of these cytokines causes acute phase symptoms, e.g. fever, actvaton of the hypothalamic-pituitary-adrenal axis and psychological depression. The amygdala belongs to the structures of the limbic system involved in the regulaton of the immune response. Increased actvity of immune system may lead to changes in the role of amygdala, medial prefrontal cortex, anterior cingulate cortex or insula. The aim of the study was to present the mutual interactons between the amygdala and pro-infammatory cytokines such as interleukin-1β (IL-1 beta) and interleukin 6 (IL-6). Most of the data included in this review comes from animal studies.
