Opis:
Comet assay under neutral conditions allows the detection of DNA double-strand breaks, considered to be the biologically relevant radiation-induced lesion. In this report we describe modifications of the neutral comet method, which simplify and facilitate its use for estimation of DNA double strand breaks in human lymphocytes irradiated with doses of 60Co gamma-rays (from 10 to 100 Gy). The analysis carried out according to this protocol takes less time than those published so far. Also, the use of lysis at 50°C is avoided; this is important in view of the presence of heat-labile sites in the DNA of irradiated cells, recently reported by Rydberg [12]. The comets have well defined, sharp limits, are suitable for computer image analysis and chromatin of the control cells remains condensed.

Opis:
The aim of this study was to investigate the effect of signalling inhibition on survival and double-strand break (DSB) rejoining in cells differing in sensitivity to inhibitors, X-rays and bleomycin. Human glioma M059 cells, K (relatively radioresistant) and J (radiosensitive, defective in DSB rejoining for lack of DNA-dependent protein kinase catalytic subunit, DNA-PKcs) were pretreated with signalling inhibitors: tyrphostin AG 1478, specific for epidermalgrowth- factor-receptor (EGFR) kinase or PD 98059, specific for kinase MEK 1/2 (mitogen-activated, extracellular signal-activated kinases 1 and 2). Subsequently, the cells were X-irradiated or treated with bleomycin. Cell survival was determined by clonogenicity test. DSB rejoining was monitored with the use of pulsed-field gel electrophoresis (PFGE). We found that in X-irradiated M059 K cells EGFR kinase activity was necessary for efficient DSB rejoining and the kinase inhibitor, tyrphostin AG 1478, acted as radiosensitizer in the dose range that reduced cell survival to 0.7-0.8. Inhibition of EGFR kinase, however, did not decrease survival or affect DSB rejoining in DNA-PKcs-deficient M059 J cells. These results indicated that the decrease in cell survival was due to a disturbed DSB rejoining by the DNA-PK dependent system. In contrast, inhibition of MEK 1/2 kinase on EGFR downstream signalling pathway by PD 98059 did not affect DSB rejoining in either cell line and exerted a radioprotective effect.

We compared the effects of bleomycin (BLM) and ionizing radiation on two sublines of murine lymphoma L5178Y (LY): LY-R, radiation resistant and LY-S, radiation sensitive. This radiosensitivity difference is related to the ability to rejoin DNA double strand breaks. LY-S cells were about two times more sensitive to BLM than LY-R, similarly as in the case of sensitivity to X rays. Since there was no difference in the P-glycoprotein-related drug transport system between the sublines, it could be expected that the enhanced sensitivity of LY-S cells to BLM was caused by the DNA repair defect. Growth disturbances in BLM treated cell populations were proportional to the lethal effect and their duration was observed until elimination of dead cells (3-6 days after 50 ěM BLM, 1 h at 37oC). There was no slow growth phase accompanied by normal viability, as previously described for X-irradiated LY-S cells. Initial DNA damage, estimated with the single cell gel electrophoresis method was linearly related to BLM dose in LY-S cells; in LY-R cells - in the low dose range (up to 10 ěM) - there was more damage than in LY-S cells, however, at higher doses the dose - effect curves became identical. The doseeffect relationship for ă rays was linear and identical in both cell sublines. DNA damage distribution in BLM treated cells was much less uniform as compared to that in irradiated cells and indicated the presence of cells with severely damaged DNA, a feature typical for BLM action in vitro.

Opis:
Adaptive response (AR), an enhanced resistance to a high dose of ionising radiation acquired after pretreatment with a very low dose, was estimated in normal human lymphocytes. The question posed was whether the extent of radioadaptation, assessed by micronucleus test, would be related to the rate of DNA double-strand break (DSB) rejoining. Phytohemagglutinin-stimulated G1-lymphocytes from 5 healthy male volunteers were pre-treated (or not) with an adaptive (5 cGy) dose of X-rays, followed by a higher (5 or 10 Gy) challenge dose after 20-22 h. DSB rejoining after the challenge dose was monitored with the use of two methods: neutral comet assay, modified to reduce the contribution of single-strand breaks (SSBs) and thermolabile sites, and pulse-field gel electrophoresis (PFGE), specific for DSBs. At the level of micronuclei, an AR was observed in lymphocytes of 3 of 5 donors. Up to 60 min, comet assay showed no statistically significant differences in DNA break rejoining between adapted and non-adapted lymphocytes, independently of AR appearance. PFGE gave similar results, although in three donors it revealed secondary increases in DSBs levels at 30 min and/or 60 min post-irradiation in the adapted vs. the non-adapted samples. Failure to demonstrate changes in DSBs rejoining rate in the adapted lymphocytes could be due to diversity of AR intensity/timing at the level of DNA repair in not fully homogenous cell populations. Also, “rare” DNA cuts characteristic of early apoptosis/necrosis could overlap the process of DNA break rejoining.

Subject to analysis in this article are the poetic and the metapoetic statements made by Henryk Sienkiewicz. Even though the latter category is now and then analysed in lingustic research – as part of the analyses of Sienkiewicz’s legacy as a columnist and a reviewer – the former category remains terra incognita for language researchers. The first part of the article analyses the key concepts of Sienkiewicz’s metapoetic statements (e.g. talent [gift, talent] or prostota [simplicity]) as well as the stylistic qualities of these statements (e.g. poetic nature, emotionality). The second part of the article features a linguistic analysis of the idiostylistic qualities of Sienkiewicz’s poetic works: grammar of rhymes, selection of stylistic means (e.g. parallels and epithets) as well as his choice of the lexical stock.

The article’s goal is to present the consequences of Polish membership in the European Union (EU) for the functioning of the Polish Sejm and Senate at the national and international level. Polish accession to the European Union resulted in changes to the scope of competences of certain organs of the Republic of Poland. This was related to the transfer of specific state competences to an international organization and it applied, in particular, to the Polish Parliament (the Sejm and Senate) which has lost its primary role as legislator of laws applicable in Poland. In order to avoid the marginalization of national parliaments in the EU decision-making process, appropriate legal measures have been introduced at the national and international level. Additionally, after the Treaty of Lisbon entered into force, national parliaments have received new competences which allowed them to exist on the forum of the European Union. The article puts forward the thesis that the Polish Sejm and Senate have lost their position as main legislative bodies at the national level but, at the same time, they have acquired a new European function which can neither be qualified as traditionally understood legislative nor controlling functions.

Będące przedmiotem analizy Godzinki moskalofilskie. 300 dni łaski carskiej za każdorazowe odmówienie tych godzinek z pokorą i skruszonym sercem T. Chełchowskiego to tekst odpowiadający poetyce polonijnych sporów politycznych przełomu XIX i XX wieku, a przy tym czerpiący z polskiej tradycji literackiej. Za sprawą nawiązania do modlitewnego wzorca – godzinek, i uwzględniając ich genezę, Godzinki moskalofilskie uznać można za trawestację (w ujęciu Nycza) lub okaz (w ujęciu Wojtak) wzorca modlitewnego. Nawiązanie to przejawia się w siedmioczęściowej budowie (jutrznia, pryma, tercja, seksta, nona, nieszpory i kompleta) z zaczęciem, hymnem, wersetami i responsoriami, modlitwą; konsekwentnie stosowany w wezwaniach i hymnach wzorzec metryczny; apostroficzne otwarcie hymnów; wersety i responsoria następujące po hymnie i/lub modlitwie; nawiązania stylistyczne i semantyczne (wezwania do cara i bohaterów hymnów na wzór apostrof do Boga i Maryi; parafrazy kolokacji biblijnych i modlitewnych). Odstępstwem od wzorca kanonicznego są: zredukowana forma ofiarowania (tylko antyfona), brak modlitwy, modyfikowane w każdej z godzin zaczęcie.
Subject to analysis in this article is T. Chełchowski’s Godzinki moskalofilskie. 300 dni łaski carskiej za każdorazowe odmówienie tych godzinek z pokorą i skruszonym sercem [Moscophile Hours: 300 Days of Grace for Each Saying of the Hours with Humility and Contrite Heart]. The Hours is a text that fits the general poetics of the political disputes typical of the Polish émigré milieus at the turn of the 20th century. At the same time, the text retains a lot from the Polish literary tradition. Taking into account the fact that the text exploits a prayer as a textual genre (Canonical Hours), and acknowledging the roots of the genre, the Hours can be classified as a travesty (trawestacja in Nycz’s approach) or as an idiosyncratic token (okaz in Wojtak’s terms) of the prayer genre. The generic linkage is realized by constructing the text of the Hours in seven parts (Matins, Prime, Terce, Sext, Nones, Vespers and Compline) with its introduction, hymn, verses, responsories and prayers; by consecutive use of metre in invocations and hymns; by an apostrophic opening of the hymns; by positioning verses and responsories after hymns or prayers; as well as by stylistic and semantic references (invocations addressed to the Tsar and hymn protagonists, modelled on apostrophes to God and Mary; paraphrases of biblical and prayer collocations). Departures from the canonical text-type include: the reduced form of the final antiphon, lack of prayer and an introduction that is modified per each hour.
