The method for determination of the starch reducing power after dissolution in water at 121°C and for determination of the quanti ty of reducing end-groups by the Park and Johnson ferricyanide modified method, was elaborated.
Większość metod oznaczania właściwości redukcyjnych skrobi polega na jej rozpuszczeniu w roztworze wodorotlenku i określeniu ilości końcowych grup aldehydowych w łańcuchach skrobiowych jako substancji redukujących. Wyniki redukcyjności skrobi uzyskiwane tymi metodami są na ogół wyższe niż wynikałoby to z masy molowej skrobi oznaczonej metodami fizycznymi. Powodowane jest to procesami częściowej degradacji skrobi i powstawania różnych związków redukujących, zachodzącymi przypuszczalnie podczas wykonywania oznaczenia. Celem pracy było znalezienie warunków oznaczania redukcyjności skrobi zapewniających możliwie małą jej degradację. W opracowanej metodzie skrobię rozpuszczano w wodzie w podwyższonej temperaturze i ciśnieniu, a substancje redukujące oznaczano zmodyfikowaną metodą Parka i Johnsona. Otrzymane wyniki porównywano z rezultatami oznaczeń wykonanych metodą z kwasem 3,5-dwunitrosalicylowym. Wartości redukcyjności skrobi uzyskane przy zastosowaniu opracowanej metody były istotnie niższe od wyników oznaczeń metodą z kwasem dwunitrosalicylowym. Opracowana metoda jest powtarzalna, prosta i szybka w wykonaniu.

Iron-reducing bacteria (IRB) seek to unravel iron corrosion for oil and gas steel pipeline failure. IRB continued to be dominating the microbiological corrosion of iron structures in steel by deteriorating steel surface via Fe(III) reduction. The mechanisms by IRB mediate Fe(III) reduction into Fe(II) for bacterial respiration to contribute to iron steel corrosion. However, the complexity of corrosion is not fully comprehended. It remains controversial due to the corrosion mechanisms proposed by IRB that may induce or inhibit corrosion when engaged with microbial biofilm. In this brief review, understanding microbiological corrosion mechanisms associated with IRB interactions may better understand microbiological corrosion and derive corrosion control.

Steel production processes generate the coke wastewaters contaminated predominantly by highly toxic phenol. Numbers of physical, chemical and physicochemical methods have been developed for the removal of phenol from coke wastewaters. Biological methods are eco-friendly and present appropriate alternative of conventional processes. Various microorganisms are able to degrade phenolic compounds including sulphate-reducing bacteria (SRB). In this work, we study the adaptation of SRB isolated from natural source to phenol and consequently the application of the adapted bacterial cultures for the biodegradation of phenol from model solutions. Two types of bacterial culture were used (monoculture containing SRB genera Desulfovibrio and mixed bacterial consortium containing SRB genera Desulfovibrio). In experiments the modified Postgate’s medium C was used - sodium lactate as the original energy and carbon source was replaced by phenol with concentration 10 mg/dm3 for adaptation and 50 mg/dm3 for biodegradation. The mixed bacterial consortium has been shown as more capable to be adapted and grow on phenol and it has the better potential for phenol biodegradation as the bacterial monoculture of SRB genera Desulfovibrio.

Opis:
Phosphatized biomorphs, resembling modern and ancient bacteria, were identified for the first time in phosphate nodules, present at the base of the Bílá Hora Formation (uppermost Cenomanian - lower Turonian), and in phosphate coprolites at the base of the Teplice Formation (upper Turonian) in the Bohemian Cretaceous Basin. They are present in colonies as filaments, coccoids, strings, rods and outgrowths, associated with the phosphate as part of the rock constituents and display the characteristics of fossilized bacteria. Two types of bacteria were identified: chemotrophic, sulphur-reducing bacteria in the phosphate nodules and phototrophic cyanobacteria in the phosphate coprolites. Microanalysis of some of the fossil bacteria revealed a fluoride-rich calcium phosphate composition, compatible with the composition of bulk samples, in which carbonate-fluorapatite is the main mineral in the phosphate nodules and coprolites. The environmental indications of these fossil bacteria support the interpretation of an anoxic environment of phosphogenesis in the latest Cenomanian - earliest Turonian and variable redox conditions of coprolite phosphatization in the late Turonian. The potential microbial role in phosphogenesis in the former may have involved the suboxic breakdown of P-rich organic matter by sulphur-reducing bacteria and the release of phosphorus in the pore water, leading to the biochemical precipitation of phosphate. The latter involved initial P-storage by phototrophic bacteria in an oxic environment, followed by P-release below the sediment-water interface under suboxic conditions and subsequent phosphatization of the coprolites.

Opis:
A possibility of side lobes level reducing in plane antenna array by creation of aperture comb edge in arrays with rectangular and circular aperture shape together with usage of different types of reducing to the array edges amplitude distributions is investigated by computer analysis. It is shown that usage of the comb structure in combination with the reducing amplitude distribution enables one to reach the side lobes level reducing in the array to -27 ÷ -29 dB.

Magnetizing roasting is an important pre-treatment technique for beneficiation of hematite and limonite ores. Reduction mechanisms of these iron minerals have been fully studied while the mechanism of reduction of chlorite is not well understood. In this study, a reduction roasting study examining the phase change of chlorite with roasting temperature was undertaken. The major finding from this study was that chlorite partially was reduced to magnetite by carbon at 750 ⁰C, and the newly formed magnetite was finely disseminated within quartz and olivine. It was demonstrated that these locked magnetite particles would be reported to iron concentrate by low intensity magnetic separation resulting in high impurity content, especially SiO2 and Al2O3 contents in the concentrate.

Risk-reducing mastectomy is a recognized prophylactic treatment for women at high and very high risk of breast cancer development in many countries. In surgical treatment, mastectomy with simultaneous reconstruction is preferred. The most common method is simultaneous reconstruction with the use of implants, but an equivalent method with a comparable or lower complication rate is simultaneous reconstruction with own tissues, taking into account free tissue flaps. The patient should be informed about the high risk of complications, possible deterioration of the life quality and high rate of corrective reoperations. In order for this form of the most effective and financially measurable health prophylaxis to be actually implemented, it is necessary to rationally evaluate the procedure and differentiate its costs depending on the type of reconstruction performed.

The descriptions found in the literature, of cases of even relatively strong non-linearity of the water supply systems, as well as their mathematical treatment, are rare and incomplete. They refer mainly to the non-linearity with respect to smooth flow control components (for instance, to the operation of pressure reducing valves (PRV), flow reducing valves (FRV) and pumps with motors equipped with smooth rotational speed control - variable frequency drives (VFD)). The article proposes a formally correct method of a comprehensive solution of the above non-linearity cases based on successive simulations with some dynamic analysis of the sensitivity of the system to the operation of the smooth control components mentioned. An appropriate algorithm controlling the effectiveness of the calculations secures correct description of the behaviour of the system for an arbitrary, in practice, method of control of flows, even if all the proposed smooth control methods are used simultaneously. The effects of the use of the solutions presented in mathematical modelling are illustrated by selected results of simulation carried out for an existing water supply system.

BACKGROUND Desulfovibrio desulfuricans belong to the heterogeneous group of anaerobic, sulphate-reducing bacteria (SRB), widely distributed in various environments. As a result of dissimilatory sulphate reduction, they release hydrogen sulphide (H2S), which has a cytotoxic effect in human and animal organisms. It has been shown by many authors, that Desulfovibrio was the genus predominating in patients with ulcerative colitis. Some of these bacteria can act as opportunistic pathogens associated with primary bacteremia and abdominal infections such as abscesses. MATERIAL AND METHODS Fifteen (soil and intestinal) strains of Desulfovibrio desulfuricans species were cultured in modified sulphate-free Postgate’s liquid medium with pyruvate for 10 days. Bacterial DNA was extracted by using a commercially available kit and DNA was used as a template for amplification of the full-length 16S, 23S rDNA and the intergenic spacer region. Digested with restriction enzymes (AluI, EcoRI, HaeIII, HindIII, HinfI, MboI and PstI) PCR amplicons were resolved by electrophoresis on 2% agarose gels. RESULTS Digestion of rrn operon of Desulfovibrio desulfuricans by seven restriction enzymes allowed to obtain the characteristic restriction profiles for all 15 investigated strains. The results allow us to suggest three of used enzymes: HinfI, AluI and HaeIII as a useful for confirmation of the similarity within of rrn operon of isolates belonging to this species. Considering the restriction profiles received with HindIII, and EcoR1 enzymes it seems that their application is insufficient, but PstI enzyme is not acceptable for the analysis of rrn operon of these bacteria. CONCLUSIONS The obtained data have shown that ARDRA can be used for establishment of phylogenetic relations among isolates of Desulfovibrio desulfuricans species, providing the appropriate restriction enzyme is used.
WSTĘP: Bakterie Desulfovibrio desulfuricans należą do szerokiej grupy beztlenowych bakterii redukujących siarczany (BRS), rozpowszechnionej w różnych środowiskach. Jako rezultat dysymilacyjnej redukcji siarczanów uwalniają do środowiska siarkowodór (H2S), który wpływa cytotoksycznie na organizm ludzi i zwierząt. Wielu autorów wykazało, iż rodzaj Desulfovibrio jest dominujący u osób cierpiących z powodu wrzodziejącego zapalenia okrężnicy. Ponadto niektóre gatunki tego rodzaju odgrywają role oportunistycznych patogenów wywołując bakteriemię, infekcje w obrębie jamy brzusznej oraz wrzody. MATERIAŁY I METODY: Piętnaście szczepów Desulfovibrio desulfuricans (glebowych i jelitowych) hodowanych było na modyfikowanym podłożu Postgata z dodatkiem pirogronianu przez 10 dni. Po wyizolowaniu genomowego DNA tych bakterii przeprowadzono reakcje PCR w celu namnożenia fragmentu operonu rrn obejmującego geny 16S, 23S oraz odcinek zmienny pomiędzy nimi. Otrzymane amplikony poddane zostały trawieniu enzymami restrykcyjnymi (AluI, EcoRI, HaeIII, HindIII, HinfI, MboI and PstI), a otrzymane fragmenty rozdzielono w 2% żelu agarozowym. REZULTATY: Przeprowadzona analiza restrykcyjna operonu rrn bakterii Desulfovibrio desulfuricans pozwoliła na otrzymanie charakterystycznych profili restrykcyjnych dla wszystkich piętnastu badanych szczepów. Uzyskane rezultaty pozwoliły uznać trzy z zastosowanych enzymów (HinfI, AluI oraz HaeIII) za odpowiednie do potwierdzenia podobieństw w obrębie operonu rrn bakterii tego gatunku. Biorąc pod uwagę profile otrzymane po trawieniu enzymami HindIII, EcoR1, możemy stwierdzić ich małą użyteczność w analizach operonu rrn gatunku Desulfovibrio desulfuricans, zaś enzym PstI w ogóle nie nadaje się do tego celu.

The work was an attempt to determine and describe certain quality characteristics of edible potato on sale in Siedlce and Międzyrzec Podlaski. Potato tubers for examination were purchased in 3 kinds of retail outlets (ten shops of each kind) in east-central Poland. A total of 90 samples were collected to determine tuber size, protein and vitamin C content, reducing sugars and sucrose content. Potatoes purchased in all kinds of outlets met the standards set for tuber size. Potatoes bought in supermarkets had a better chemical composition than tubers purchased in groceries or fruit and vegetable shops. All the potato tubers were a good staple.

The paper presents empirical methodology of reducing various kinds of observations in geodetic network. A special case of reducing the observation concerns cartographic mapping. For numerical illustration and comparison of methods an application of the conformal Gauss-Krüger mapping was used. Empirical methods are an alternative to the classic differential and multi-stages methods. Numerical benefits concern in particular very long geodesics, created for example by GNSS vectors. In conventional methods the numerical errors of reduction values are significantly dependent on the length of the geodesic. The proposed empirical methods do not have this unfavorable characteristics. Reduction value is determined as a difference (or especially scaled difference) of the corresponding measures of geometric elements (distances, angles), wherein these measures are approximated independently in two spaces based on the known and corresponding approximate coordinates of the network points. Since in the iterative process of the network adjustment, coordinates of the points are systematically improved, approximated reductions also converge to certain optimal values.

This study proposes a method for the production of ethanol from wheat straw lignocellulose where the raw material is chemically processed before hydrolysis and fermentation. The usefulness of wheat straw delignification was evaluated with the use of a 4:1 mixture of 95% ethanol and 65% HNO3 (V). Chemically processed lignocellulose was subjected to enzymatic hydrolysis to produce reducing sugars, which were converted to ethanol in the process of alcoholic fermentation. Chemical processing damages the molecular structure of wheat straw, thus improving ethanol yield. The removal of lignin from straw improves fermentation by eliminating lignin’s negative influence on the growth and viability of yeast cells. Straw pretreatment facilitates enzymatic hydrolysis by increasing the content of reducing sugars and ethanol per g in comparison with untreated wheat straw.

Phosphotransacetylase activity and the kinetic properties of the enzyme from intestinal sulfate-reducing bacteria Desulfovibrio piger and Desulfomicrobium sp. has never been well-characterized and has not been studied yet. In this paper, the specific activity of phosphotransacetylase and the kinetic properties of the enzyme in cell-free extracts of both D. piger Vib-7 and Desulfomicrobium sp. Rod-9 intestinal bacterial strains were presented at the first time. The microbiological, biochemical, biophysical and statistical methods in this work were used. The optimal temperature and pH for enzyme reaction was determined. Analysis of the kinetic properties of the studied enzyme was carried out. Initial (instantaneous) reaction velocity (V0), maximum amount of the product of reaction (Pmax), the reaction time (half saturation period, τ) and maximum velocity of the phosphotransacetylase reaction (Vmax) were defined. Michaelis constants (Km) of the enzyme reaction (3.36 ± 0.35 mM for D. piger Vib-7, 5.97 ± 0.62 mM for Desulfomicrobium sp. Rod-9) were calculated. The studies of the phosphotransacetylase in the process of dissimilatory sulfate reduction and kinetic properties of this enzyme in intestinal sulfate-reducing bacteria, their production of acetate in detail can be perspective for clarification of their etiological role in the development of the humans and animals bowel diseases. These studies might help in predicting the development of diseases of the gastrointestinal tract, by providing further details on the etiology of bowel diseases which are very important for the clinical diagnosis of these disease types.

Over the recent years, ship traffic in the polar areas has increased. There is reason to believe that this traffic, and especially the cruise traffic, will increase further as the ice retracts towards the poles. There is also reason to believe that with the continued focus and exposure of the Polar Region, the cruise tourism to the region will grow. The increased presence in the polar areas will create positive repercussions for several actors, both on sea and land. There will, however, also be challenges associated with the growing presence in the polar areas. Vessels will be operating at long distances to other vessels and land infrastructures. These vessels will also be operating in climate and conditions that will put extra pressure on both vessel and crew. These challenges need to be solved in order for the ship industry to operate safely in the Polar Region. To ensure that companies operating in these areas identify and manage these challenges, the International Maritime Organization (IMO) developed the Polar Code (2017) with the intent of increasing the safety for vessels operating in polar waters, and to reduce the impact on humans and environment in this remote, vulnerable and harsh area. This code defines a number of requirements, with which the vessels should operate in accordance with. In this paper, we reveal which challenges the vessel and its crew need to deal with when navigating in polar waters. The challenges will be analysed and assessed through the use of a preliminary qualitative risk analysis to determine the potential hazards the vessel is exposed to under operations in polar waters, and to find out what level of risk the different hazards represents for the vessel and its crew. The main objective of the paper is to find out how the risk levels can be reduced, with particular focus on the use of simulator training as a risk reducing measure. The final goal is to measure the risk towards acceptance criteria, which have been determined prior to conducting the analysis.
