Opis:
A rapid, sensitive and simple reversed-phase high-performance liquid chromatographic (HPLC) method has been developed and validated for the determination of acyclovir (ACV) in rabbit plasma. BDS C18 column was used to conduct analysis using ammonium dihydrogen phosphate buffer (50mM) and methanol as mobile phase (98:2), with pH adjusted to 2.5 using orthophosphoric acid. Flow rate was kept at 1 mL/min. Selective precipitation of plasma proteins were done by adding 5% perchloric acid. Precipitated plasma proteins were separated by centrifugation. ACV moves in a supernatant, which was snapped and passed through a syringe filtration assembly. Direct injection of supernatant was given into a BDS C18 column and ACV was detected at 256 nm. The limit of detection for ACV in plasma was estimated as 15ng/mL whereas the limit of quantitation was calculated as 25 ng/mL. Moreover, the developed method has been found to be selective and linear into concentration range of 25 – 2000 ng/mL. The present method could be successfully applied to samples from bioavailability and bioequivalence studies.

Opis:
Ionic liquids (IL) are new materials with a wide spectrum of use. Being a variable combination of an organic cation and an organic or inorganic anion, their have controllable properties. The effect of the ionic liquid added to the mobile phase on the separation of uracil derivatives is studied and demonstrated in this paper. The ionic liquid used was didecyldimethylammonium lactate, which has an advantageous effect on the separation of the analytes studied. Pyrimidine bases which were used as the test compounds are characterized by alkaline properties due to which so-called peak tailing is observed during separation. A methanol/water mixture was used as the mobile phase in the chromatographic analyses. Optimum separations were obtained for 8% levels of the organic modifier. The best separations of the test isomers were obtained for 0.01% IL added to the mobile phase. The findings obtained are evidence that the use of ionic liquids in HPLC is an efficient method for the separation of polar compounds: the duration of the analysis is shorter and the resulting peaks are sharp and symmetrical.

Oleuropein is a secoiridoide monoterpene. It is found primarily in all parts of the Olea europaea L. tree, mostly in its unripe fruit and leaves. It has a number of beneficial effects on the human body, being readily absorbed from the gastrointestinal tract due to its hydrophilic properties. The paper presents a simple, rapid and precise method for the assay of oleuropein in herbal formulations and the European Olive leaf extract. Chromatographic separation was performed using a C-18 column with an acetonitrile/water 20/80% vol. mixture as the mobile phase. The method has a wide range of linearity between 0.05 and 1.5 mg/mL, very good repeatability of retention time, and accuracies between 95.8 and 103.3%. Its limit of detection is 0.04 mg/mL. The developed method was tested on two herbal formulations which are commercially available in Poland.

Opis:
The effects of various technological steps and raw material inhomogeneity on the composition of pigments in paprika (Capsicum annuum) powders were studied by reversed-phase high-performance liquid chromatography (RP-HPLC), using gradient elution technique (various mixtures of water, acetonitrile and methanol) and diodearray detector. The standard deviation of the composition of pigments in the samples taken at different technological stages was lower than among the batches of raw material. This indicates that the technological steps have a negligible effect on the composition of pigments in paprika powder. It was further found that the raw material for the paprika powder manufacture could considerably be inhomogeneous even when harvested on the same field.

This study aimed to develop a simple stability-indicating HPLC-DAD method for the assay of Tildipirosin (TPS), which is a 16-member ring semi-synthetic macrolide, in an injectable solution. TPS is commonly used in the USA for the treatment of animal's respiratory infection. The isocratic elution was set up using the Inertsil-ODS column with the flow rate set at 0.9 mL min-1 and the detection wavelength of 285 nm. The column temperature was ambient and the overall runtime was 5 minutes. Additionally, the mobile phase was water: acetonitrile: trifluoroacetic acid (53.45: 46.45: 0.10, by volume). The HPLC-forced degradation stability method for TPS in Egydipiro Injectable Solution was performed in different stress conditions, i.e. light/UV, heat, acid, base, and oxidation. The results showed the linearity range was 36.00 to 108.00 µg mL-1 for TPS. The correlation coefficient was higher than 0.999 and the accuracy was 98.78%. The degradation percentages were presented ranging from 31.54% in the photodegradation condition to 50.10% in the base degradation. The Eco-scale score was about 82 points indicating an excellent green chromatographic technique. Therefore, the novel stability method was unique in terms of simplicity and environment-friendly aspects. The new HPLC–DAD method was the first, green, stability-indicating chromatographic method for a TPS analysis with its degradation products. However, the major drawbacks of the new chromatographic method are the incomplete identification of degradation products and the inability to propose an appropriate mechanism for drug degradation. Overall, the method can be employed to evaluate TPS stability under various storage conditions.

A field trial was conducted to evaluate the residues of Bispyribac sodium 10 % SC on rice crop during Kharif season 2013 at Kandikai in Thiruvallur District, Tamilnadu, India. Randomized block design was followed withthree treatments forthree replicates. Bispyribac sodium 10 % SC 200 g a.i./ha, 500 g a.i./ha and control (water spray) was sprayed using hand operated Maax battery sprayer with a spray volume of 300 litres per hectare at 15 days after transplantation of rice crop (ADT 45). At harvest, samples of grain, straw and soil were collected replicate wise from each treatment along with the control. These samples were stored in icebox and transfer to the laboratory under cooled condition for analysis.All the residues samples were analyzed for Bispyribac sodium content by a validated HPLC method at the minimum detectable concentration of 0.01 ppm. The result revealed that no detectable level of Bispyribac sodium in straw, grain and soil at harvest. Thepost treatmentand pre-harvest intervalwas 60 days after transplantation.

This work reports on the process of phenol electrooxidation, which is carried-out through continuous electrolysis of synthetic, sodium sulphate-based wastewater. Phenol electrodegradation is examined by means of a laboratory size (ca. 700 cm3  of working volume), poly (methyl methacrylate)-made electrolyser unit for various, carbon fibre and graphite-based anode configurations, and stainless steel cathodes, two different current-densities and concentrations of phenol in synthetically prepared wastewater solution. Proper monitoring of phenol degradation (including quantitative identification of reaction products and calculation of specific energy consumption) in wastewater is performed by means of instrumental, combined HPLC and MS technique in function of electrolysis time.

Method for determination of carbendazim residues in fruits, vegetables and cereals was described. The compound was extracted with methanol-hydrochloric acid mixture, and after liquid-liquid partition step with dichloromethane, was determined by high performance liquid chromatography (HPLC) with column switching and ultraviolet (UV) detection. The average recoveries of carbendazim from fortified sample were from 68.7% ± 4.3% to 92.6% ± 4.5%, the coefficients of variation were from 2.9% to 6.3%, and the limits of quantification at λ = 279 nm were from 0.02 mg/kg to 0.2 mg/kg.
Przedstawiono procedurę analityczną oznaczania pozostałości karbendazymu w materiale roślinnym techniką wysokosprawnej chromatografii cieczowej HPLC z wykorzystaniem przełączania kolumn. Procedura ta opracowana została w celu wykorzystania w badaniach monitoringowych płodów rolnych i ogrodniczych traktowanych preparatami zawierającymi substancje czynne z grupy fungicydów benzimidazolowych: benomyl, karbendazym, tiofanat metylu. Średni odzysk karbendazymu z próbek fortyfikowanych mieścił się w granicach od 68,7% ± 4,3% do 92,6% ± 4,5%, współczynnik zmienności wynosił od 2,9% do 6,3%, a granica oznaczalności przy długości fali λ = 279 nm była od 0,02 mg/kg do 0,2 mg/kg.

Sulfacetamide sodium (SFS), in combination with prednisolone sodium phosphate (PSP), is indicated for ophthalmic infections. The present study describes a reverse phase-high performance liquid chromatography method for simultaneous estimation of these two drugs using diode-array detector. Isocratic elution with solvent system acetate buffer (0.01 M, pH 7): acetonitrile: methanol 75:20:5 (v/v/v), at a flow rate of 0.8 mL min-1 gave optimum resolution. The detection limits for SFS and PSP were calculated as 1.15 µg mL-1 and 1.45µg mL-1, while quantification limits were 3.83 µg mL-1 and 4.84 µg mL-1 respectively. Accuracy was found to be 99-101% with relative standard deviation (RSD) values less than 2%. The proposed method is accurate, precise, sensitive, reproducible and applicable for normal usage in quality control laboratories. It would prove advantageous in utilizing less time and resources as previously there is no reported single assay for simultaneous quantification of the aforementioned drugs.
